Claims (13)
- Fremgangsmåde til formering af humane hepatocytafledte celler Patentkrav1. Skalerbar fremgangsmåde til formering af celler fra en human hepatocytcellelinje omfattende: a) Podning af celler fra en human hepatocytcellelinje ind i et alginatmatrixdannende middel og dannelse af alginatkugler omfattende matrixindkapslede celler og b) Prækultur af de matrixindkapslede celler i et dyrkningsmedium i et kammer med et fluidiseret leje, så de danner sammenhængende sfæroider i et celleformeringsstadium, hvori de matrixindkapslede celler holdes i kontinuerlig suspension i dyrkningsmediet i 8 til 12 dage og dyrkes til ydeevnekompetence, således at de opretholder en næsten-kubisk cellearkitektur, har en tæt celle til celle- og celle til matrix-organisation og afsondrer ekstracellulære matrixproteiner og et repertoire af leverspecifikke afsondrede proteiner.
- 2. Skalerbar fremgangsmåde ifølge krav 1, hvori den humane hepatocytcellelinje omfatter Hep G2-celler.
- 3. Skalerbar fremgangsmåde ifølge krav 1, hvori alginatkugleme polymeriseres.
- 4. Skalerbar fremgangsmåde ifølge krav 3, hvori alginatkugleme polymeriseres i 10- 60 minutter i calciumchloridopløsning indeholdende et eller flere overfladeaktive midler.
- 5. Skalerbar fremgangsmåde ifølge krav 1, hvori alginatkugleme omfatter en ikke-coatet alginat.
- 6. Skalerbar fremgangsmåde ifølge et hvilket som helst af kravene 3 til 5, hvori alginatkugleme har en middeldiameter på mellem 300-1200 pm.
- 7. Skalerbar fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvori de indkapslede celler endvidere omfatter et densitetskontrollerende middel.

Fortætning af det alginatmatrixdannende middel med en densitetsmodifikator, således at kugler af de matrixindkapsiede celler vil opnå kontinuerlig suspension, når de perfunderes i dyrkningsmediet eller i humant plasma; iv. Passage af de humane hepatocytafledte celler gennem en formet dyse, så der dannes en strøm; v. Udskæring af strømmen i kugler; og vi. Polymerisering af kuglerne.
- 12. Fremgangsmåde ifølge krav 11 omfattende 0,5xl06 til 3,0xl06 celler pr. ml alginat.
- 13. Fremgangsmåde ifølge krav 11, hvori den formede dyse er cirkulær i tværsnit og har en diameter på fra 250-450 pm, og strømmen udskæres med ståltråds skiver ved en hastighed på fra 3000 til 4000 rpm.
