[0001] Bekæmpelsen af smitsomme sygdomme er stadig en af de største udfordringer inden for både human og veterinærmedicin. Situationen er særligt kompleks med virusinfektioner, der i modsætning til bakterielle infektioner generelt ikke kan bekæmpes med bredspektret medicin og forårsage stor økonomisk skade. Udviklingen af nye virkningsfulde vaccinationsstrategier mod virussygdomme er derfor af afgørende betydning.
[0002] Traditionelt anvendes der til forebyggende og også til terapeutisk vaccination mod virussygdomme, "dæmpet", dvs. ved mutagenese modificerede vira med betydeligt reduceret eller fraværende virulens ("levende vaccine") eller inaktiverede vira ("død vaccine"). I nyere tid er såkaldte "subunitvacciner" eller "subunit markørvacciner" i stigende grad blevet etableret, hvorved definerede genteknologisk fremstillede "hovedantigener" af patogenet anvendes til vaccination. Udtrykket "markørvaccine" indebærer, at vaccinerede individer klart kan differentieres fra naturligt inficerede individer ved efterfølgende diagnostisk analyse ("DIVA", "differentiate infected and vaccinated animals"). Større antigener er for eksempel proteiner fra viruskappen eller virus-capsiderne, som kan foranledige en humoral og/eller cellulær immunreaktion i værten i fravær af en komplet viruspartikel, som følge heraf kan en viral infektion bekæmpes forebyggende eller terapeutisk. En "subunitvaccination" kræver, at hovedantigenerne er karakteriseret. Fremstliingen (ekspression) og immunogen formulering af sådanne proteiner ("antigenformulering") til vaccinationsprocessen spiller en central rolle, især da virus kappeproteiner normalt ikke er vandopløselige og kun sjældent kan fremstilles rekombinant i bakterielle ekspressionssystemer. Fremgangsmåder til opnåelse af "subunitvacciner" i en oprenset form og for at sikre deres holdbahed er tilsvarende omkostningskrævende.
[0003] Gær som ekspressionssystemer til rekombinante proteiner kombinerer de økonomiske fordele ved bakterielle systemer den for højere celler typiske organisationsform. De er kompartmenteret via intracellulære membran systemer, der fundamentalt adskiller dem fra bakterielle værtssystemer og muliggør ekspression af membranforankrede virale kapsel proteiner eller endda hele viruskapsler. Derudover har beta-glucanerne, glucosepolymererne, hvorfra gærcellevæggen er bygget, en kendt immunstimulerende virkning. Det var derfor opgaven at udvikle en fremgangsmåde, hvor komplette gærceller kan anvendes til vaccination. Problemet blev løst ved hjælp af genteknologiske fremgangsmåder at indføre gener for immunogene determinanter i genomet af ikke-patogene gærarter og udtrykke dem, og disse rekombinante gærceller anvendes direkte til oral/mucosal vaccination.
Kluyveromyces lactis.
[0004] Kluyveromyces lactis tilhører de såkaldte "food grade" gærarter med GRAS-status ("generally regarded as safe"). Ligesom bagergær, der er blevet testet og bevist over årtusinder som et fødevaretilsætningsstof, anses gæren K. lactis, der ofte er repræsenteret i mejeriprodukter, som sikker af fødevareindustrien.
Oral/slimhindevaccination [0005] Vacciner/antigener kan indgives parenteralt eller oralt/mucosalt. Mens antigenerne i det første tilfælde kommer ind i blodbanen, hovedsagelig ved injektion, og omgår fordøjelseskanalen, finder optagelse i tilfælde af oral/mucosal vaccination sted via den mukosale tarmepitel efter passage gennem maven. Tarmslimhinden er permanent påtogen-eksponerede og udgør i sig selv en væsentlig barriere for optagelse af infektiva. Derudover er der et mucosale immunsystem, som er opdelt i de såkaldte "inductive sites" og "effector sites". Optagelsen af antigen finder sted i de "inductive sites" via M-celler i "Peyer's patches". MALT ("mucosa-associated lymphoid tissue") immunceller associeret med "Peyer's patches" fremkalder immunresponset. Når immunresponset er aftaget, dannes der specielle hukommelsesimmunceller i hele organismen såvel som i slimhindernes "effector sites", der som regel formidler langvarig beskyttelse mod det oprindelige anti-gen/patogen.
[0006] Fordele ved oral/mucosal vaccination. Sammenlignet med parenteral immunisering kræver oral/mucosal immunisering anvendelse af betydeligt større mængder antigen. Ved oral/mucosal vaccination er det i modsætning til parenteral vaccination imidlertid, i tillæg til det systemiske immunrespons, muligt at inducere et lokalt immunrespons ved slimhindens "effector sites". Især for patogener (såsom bovin virusdiarré-virus, BVDV, og klassisk svinepestvirus, CSFV; se også nedenfor), som (også) overføres via slimhinde, har en oral/mucosal vaccination, der har potentialet til at skabe en aktiv og langvarig immunitet. Andre vigtige fordele ved oral vaccination er god accept og økonomi. I bedste fald kan vaccinerne produceres til lave omkostninger og indgives ukompliceret med kosten.
[0007] Den orale indgivelse af gærstammer, der udtrykker virusantigener, er derfor ikke kun sikker, men kan have yderligere gavnlige og adjuvante virkninger [0008] Det var nu opgaven at udvikle et ekspressionssystem, som muliggør målrettet integration af fremmede gener i Kluyveromyces lactis' genom og følgelig muliggør den intracellulære syntese af antigener. Et andet formål var at anvende en sådan rekombinant K. lactis-stamme, som indeholder et specifikt virusantigen, til en oral/mucosalvaccine.
[0009] Ifølge opfindelsen blev der udviklet et vært/vektor-system, der via genteknologiske fremgangsmåder muliggør integration af fremmede gener målrettet K. lactis-genomets LAC4-locus, uden at der skal indføres yderligere DNA-sekvenser (eller lignende selektionsmarkører). Fremmedgen-ekspression er under styring af LAC4-promotoren og kan induceres ved doseret lactose eller galactose i dyrkningsmediet eller konstitutivt aktiveres efter slukning af regulatorgenet KIGAL80. De rekombinante gærstammer er stabile uden selektionstryk og kan dyrkes under fermenteringsbetingelser til høje densiteter. Den fremmede proteinekspression kan kvantificeres indirekte via ekspressionen af et endogent reportergen.
[0010] En række rekombinante K. lactis-vaccinestammer blev genereret. Generelt udtrykker disse stammer inducerbart betydelige mængder af et protein eller domæner af dette protein eller domæner af dette protein fusioneret til fremmede proteindomæner. De relaterede fremmede proteindomæner tjener adjuvante formål eller den målrettede kompartmentering af det udtrykte fremmede protein i gærcellen. Ud over adjuvansvirkninger er kompartmenteringen af det udtrykte fremmede protein vigtig for optimering af ekspressionen eller formulering af ekspressionsprodukterne. En rekombinant K. lactis-stamme (se udførelsesformer) blev med succes anvendt til oral/mucosal vaccination.
[0011] Formålet med opfindelsen opnås ved at tilvejebringe en rekombinant stamme af gæren Kluyveromyces lactis ifølge krav 20 og en fremgangsmåde til oral/mucosal vaccination ifølge krav 1, hvilken fremgangsmåde omfatter den rekombinante stamme ifølge opfindelsen af gæren Kluyveromyces lactis. Opfindelsen angår også anvendelsen af den rekombinante stamme af gæren Kluyveromyces lactis for fremmed protein ekspression såvel som i vaccinationen, navnlig mod BVD-virus eller CSF-virus, ifølge kravene 21 til 23.
EKSEMPLER 1. Fremstilling af K. lactis stamme VAK367-D4 (Metura3 Iac4 :: ScURA3).
[0012] Udgangsstammen VAK367 til heterolog ekspression af fremmede proteiner har følgende funktioner: Den muliggør dyrkning ved høj celledensitet, uden de intracellulære proteiner påviseligt frigives. I denne henseende adskiller denne stamme sig fra mange nært beslægtede K. /act/'s-stammer. Stammen VAK367 blev afledt ved to omgange mutagenese af stammen CBS 2359 (Centraalbureau voor Schimmelcultures http://www.fungalbiodiversitycentre.com) og er auxotrof for aminosyren methionin og nukleobasen uracil. Stammen VAK367-D4 (deponeret hos den tyske samling af mikroorganismer og cellekulturer Ltd (DSMZ) i Braunschweig) blev afledt af stamme VAK367 ved anvendelse af genteknologi, hvor sekvensen +358 til +1181 af L4C4-genet blev erstattet af ScURA3-genet ved hjælp af plasmidet pD4-2. Stammen VAK367-D4 muliggør nu integration af fremmede gener på LAC4-locus'et uden yderligere markører, ved at selektere på lactose-vækst. Ved anvendelse af en egnet integrationsvektor, f.eks. Klp3 (se nedenfor) erstattes disruptionskassetten ved homolog rekombination, således at et intakt L4C4-gen rekonstitueres med tab af Scl/ÆÆ3-markøren. (Fig. 1). 2. Generering af en integrationsvektor, der muliggør den inducerbare ekspression af fremmede gener.
Vektor; Klo3 (figur 2) [0013] Vektoren Klp3 er en E. coli-vektor baseret på YRp7, som ikke kan replikere autonomt i gær, fordi ARSl-sekvensen er blevet slettet. Klp3 indeholder K. lactis-sekvenser, som muliggør integration ved L4C4-locus ved homolog rekombination (opstrømsregion af LAC4 og 5'-enden af MC4-læserammen). Mellem MC4-promotoren og transkriptionsstart blev der indsat et DNA-segment indeholdende TEF1 -terminatoren og /CIGAZ_80-promotoren. Således er MC4-læserammen under styring af KIGAL80-promotoren, som er co-reguleret af transskriptionsfaktoren KIGal4 med L4C4-promotoren (Zenke et al, 1993, Molecular and Cellular Biology, 13:. 7566-7576). Denne konstruktion gør det muligt at følge induktionen af fremmed genekspression ved at måle den LAC4-kodede β-galactosidase. Klp3 muliggør integrationen af det fremmede gen mellem LAC4-promotoren og TEF1 -terminatoren via den unikke Sa/I-grænseflade. Til integration af ekspressionskassetten fordøjes Klp3 med Hpal eller EcoRl og transformeres ind i K. lactis VAK367-D4. Derved adskilles ekspressionskassetten fra E. co//-vektorgruppen, således at de resulterende stammer ikke indeholder nogen bakterielle sekvenser.
Plasmid KIp3-E2-l (9437 bp) [0014] BVDV-gensegmentet, som bl.a. indeholdt den BVDV E2 kodende region (BVDV E2 ORF) kodende, blev indsat som et Sa/I-XboI-fragment i Sa/I-stedet mellem MC4-promotor og TEF1 -terminator. Nedstrøms er KIGAL80-promotoren fusioneret til 5'-enden af LAC4-ORF’en. Restriktion af plasmidet med Hpal giver to fragmenter, hvoraf det største bærer ekspressionskassetten med BVDV E2-ORF. 3. Formulering af E2-hovedantigenerne af BVDV og CSFV.
Et hovedantigen er karakteriseret ved BVDV (bovin virus diarré virus), patogenet til bovin virus diarré og Mucosal Disease (BVD/MD), samt også ved CSFV (klassisk svinepestvirus), patogenet til klassisk svinepest (CSF). Dette er "envelope"- (viruskappe-integreret) protein E2. E2 inducerer også et massivt humoralt immunrespons, selv i fravær af en viruspartikel, dvs. dannelsen af virkningsfuldt virus-neutraliserende antistoffer. Ved genteknologisk formulering af E2 formulering kunne proteinets immunogene potentiale styrkes yderligere og kunne også generere et cellulært immunrespons (Bruschke et al., 1999; Nobiron et al., 2003). Det specifikke og eksklusive immunrespons mod enkelte, under visse omstændigheder, genteknologisk formulerede proteinområder af E2 muliggør skelnen af vaccinerede dyr fra feltvirusinficerede dyr, f.eks. ved ELISA-metode. 4. Konstruktion af en K. lactis-stamme, der udtrykker BVDV E2-proteinet [0015] Ved hjælp af teknologien ifølge opfindelsen blev K. /acf/s-stamme, VAK367-E2-1, fremstillet. Til dette formål blev plasmidet Klp3-E2-1 skåret med Hpal, og det større Hpal-fragment kodende for BVDV E2-ORF, kromosomalt integreret via homolog rekombination i udgangsstammen VAK367-D4 (DSM 23097). I VAK367-D4 (DSM 23097) ødelægges LAC4-genet ved indsættelse af URA3-genet (Iac4::URA3). Kromosomal integration af dette fragment erstattede locus lac4::URA3 og rekonstituerede det intakte MC4-gen (Figur 1). Udvælgelsen af K. lactis-ceIler indeholdende BVDV-gensekvensen blev udført ved dyrkning på lactosemedium. Tabet af URA3-genet blev bekræftet af Ura-minus-fænotypen. Sekvensen af det tilsvarende locus blev bekræftet ved DNA-sekventering. (Sekvensliste 1).
[0016] I den resulterende stamme, VAK367-E2-1, er et segment af BVDV-genomet af BVDV-stamme CP7 integreret i gærgenomet. Det tilsvarende BVDV-gensegment indeholdt området, som koder for E2-proteinet såvel som for dele af de tilstødende El- og p7-regioner.
El- og p7-regionerne indeholder signalsekvenserne, der er nødvendige til den korrekte forarbejdning af E2-proteinet (sekvensliste 2). Den korrekte forarbejdning (modning) af BVDV E2-proteinet finder sted via signalaser, som er kodet af gærcellen.
[0017] Ved hjælp af en specielt udviklet fremgangsmåde til immunfluorescensdetektion kan ekspressionen af BVDV E2 i celler fra K. lactis-stamme VAK367-E2-1 bestemmes. En ELISA-fremgangsmåde, der er specielt udviklet til BVDV E2-detektion, muliggør detektion og kvantificering af heterologt udtrykt antigen. Den analoge ELISA-metode kunne anvendes til den nøjagtige kvantificering af antistoftiteren af immuniserede dyr. Fremgangsmåder til virusneutralisering og til karakterisering af antistoffer og T-celler er blevet anvendt som rutinefremgangsmåder.
[0018] En ny qRT-PCR-metode gør det muligt at detektere og kvantificere BVDV-RNA-genomet fra serum og cellekultursupernatanter. 5. Demonstration af virkningen af K. lactis-stamme VAK367-E2-1 i orale/mucosale immuniseringsundersøgelser
Undersøgelse 1.
[0019] I dyreforsøg blev et betydeligt antal eksperimentelle mus indgivet en emulsion af naiv K. lactis under standardiserede betingelser. Der blev anvendt forskellige immuniseringsskemaer, hovedkriterierne var forskellige mængder af tilsat gær (3 - maks. 8% andel af det daglige fødeindtag) og forskellige "booster-intervaller".
Resultater: [0020] (i) En generel tolerance for K. lactis blev påvist: Oral indgivelse af gæremulsionerne forårsagede ikke nogen synlige ændringer i dyrets tilstand. (ii) Den orale indgivelse af K. lactis resulterede i et klart detekterbart humoralt immunrespons mod visse gærproteiner. Ved Western blot-metode fandt man et signifikant og specifikt antistofrespons på gærproteiner hos dyrene fodret med K. lactis sammenlignet med kontroldyr. Gærproteiner har altså i sig selv en immunogen virkning. Ud over det "proof of principle" at den orale indgivelse af K. lactis kan anvendes til at generere et immunrespons, er det blevet indikeret, at oral indgivelse af gær i kombination med et rekombinant antigen kan tilvejebringe en yderligere adjuvansvirkning.
Undersøgelse 2 [0021] I yderligere dyreforsøg med et betydeligt antal mus blev der vaccineret oralt med et optimeret immuniseringsskema (se undersøgelse 1) og med rekombinant K. lactis af stammen 367-E2-1.
Resultater: [0022] (i) Ved anvendelse af den specielle ELISA-metode var det muligt at detektere dannelsen af anti-BVDV E2-antistoffer i musene vaccineret med K. lactis-stamme 367-E2-1 sammenlignet med kontrolmus (immuniseret med naiv K. lactis). ii) I en neutraliseringstest med BVDV på bovine kulturceller kunne en neutraliserende virkning af antisera fra mus immuniseret med K. lactis-stammen 367-E2-1 igen etableres sammenlignes med serum fra kontrolmus. iii) Ved anvendelse af adjuvanser, såsom CpG-ODN og QuilA, blev en forbedring af immunresponset opnået. iv) Ved mucosal/oral immunisering med den rekombinante K. lactis stamme VAK367-E2-1 kunne en virkningsguld beskyttelse opnås. Dette blev demonstreret i challenge-forsøg med rekombinante vacciniavira, der udtrykker BVDV E2-proteinet.
Resultaterne af undersøgelserne viser, at ifølge opfindelsen ved hiælp af frembringelse af en K. lactis-stamme og varianter af denne stamme, kan fremmede gener uden yderligere selektionsmarkører specifikt integreres i disse. Det resulterende rekombinante afkom fra den pågældende K. lactis-stamme kan anvendes til ekspressionsformål til den efterfølgende biokemiske fremstilling af et heterologt protein. Cellerne fra de tilsvarende rekombinante K. lactis-stammer kan anvendes direkte til mucosal/oral eller parenteral immunisering.

Claims (21)
1. Fremgangsmåde til oral/mucosal vaccination, omfattende indgivelse afen genteknologisk genereret rekombinant gærstamme af gæren Kluyveromyces lactis kendeteg net ved, at denne rekombinante stamme indeholder en genomisk integreret ekspressionskassette, der er sammensat af den lactose-inducerbare L4C4-promotor, et fremmed gen, der koder for et immunogent protein, en transkriptionsterminator, og MC4-genet, som under styring af GAZ_80-promotoren fra K. lactis (KIGAL80-P) koder for β-galactosidase-enzymet, hvorved den målrettede integration af fremmede gener finder sted ved gærgenomets L4C4-locus, uden indføring af yderligere DNA-sekvenser, og ekspressionen af fremmede gener induceres doseret af lactose eller galactose eller aktiveres konstitutivt efter slukning af regulatorgenet KIGAL80.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at de rekombinante K. lactis-stammer er genetisk stabile, når de dyrkes under ikke-selektive betingelser.
3. Fremgangsmåde ifølge krav 1, kendetegnet ved, at de rekombinante stammer af gæren Kluyveromyces lactis anvendes som ekspressionssystem og til vaccination.
4. Fremgangsmåde ifølge krav 1 eller 2, kendetegnet ved, at de rekombinante stammer af Kluyveromyces lactis muliggør lactose-inducerbar ekspression af fremmede gener.
5. Fremgangsmåde ifølge ét eller flere af de ovennævnte krav 1 til 3, k e n d e t e g-n e t ved, at det fremmede gen integreres ved den rekombinante Kluyveromyces lactis-stammes LAC4-locus.
6. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at den rekombinante Kluyveromyces-stamme foruden det fremmede gen ikke indeholder nogen yderligere vektorsekvenser eller selektionsmarkører.
7. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at den fremmede genekspression er konstitutiv efter slukning af KIGAL80-ger\et
8. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at den fremmede genekspression kan kvantificeres indirekte via ekspression af et endogent reportergen.
9. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at det fremmede gen muliggør ekspression af virale proteiner med antigene egenskaber.
10. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at det fremmede gen muliggør ekspression af virale strukturelle proteiner.
11. Fremgangsmåde ifølge ét eller flere af de ovennævnte krav, kendetegnet ved, at det fremmede gen muliggør ekspression af virale strukturelle proteiner af familierne Flaviviridae, Orthomyxoviridae og Birnaviridae.
12. Fremgangsmåde ifølge ét eller flere af de ovennævnte krav, kendetegnet ved, at det fremmede gen muliggør ekspression af strukturelle proteiner af virus fra bovin virusdiarré (BVDV).
13. Fremgangsmåde ifølge ét eller flere af de ovennævnte krav, kendetegnet ved, at de rekombinante stammer af Kluyveromyces lactis-gær, der inducerbart eller kon-stitutivt udtrykker betydelige mængder af et protein, eller af domæner af dette protein, eller af domæner af dette protein fusioneret med fremmede proteindomæner.
14. Fremgangsmåde ifølge ét eller flere af de foregående krav, kendetegnet ved, at den biokemiske fremstilling fremmede proteiner er muliggjort.
15. Fremgangsmåde ifølge ét eller flere af de ovennævnte krav, kendetegnet ved, at der ved en vaccination ved hjælp af indgivelse af komplette gærceller af en rekom-binant stamme af gæren Kluyveromyces lactis genereres en specifik immunisering mod udtrykt fremmed protein.
16. Fremgangsmåde ifølge krav 15 kendetegnet ved, at der ved en vaccination ved hjælp af inhalering, bronkial, oral, nasal indgivelse af komplette gærceller af en rekombinant stamme af gæren Kluyveromyces lactis genereres en specifik immunisering mod udtrykt fremmed protein.
17. Fremgangsmåde ifølge krav 16, kendetegnet ved, at biokemisk fremstillede fremmede proteiner muliggør en specifik immunisering.
18. Rekombinant stamme af gæren Kluyveromyces lactis, kendetegnet ved, at denne rekombinante stamme indeholder en genomisk integreret ekspressionskassette, der er sammensat af den lactose-inducerbare L4C4-promotor, et fremmed gen kodende for et immunogent protein, en transkriptionsterminator og det for enzymet β-galactosidase kodende L4C4-gen under styring af G/4/.SO-promotoren fra K. lactis (KIGAL80-P), hvor de fremmede gener integreres specifikt ved gærgenomets LAC4-locus uden indføring af yderligere DNA-sekvenser, og ekspressionen af fremmede gener induceres doseret af lactose eller galactose eller aktiveres konstitutivt efter slukning af regulatorgenet KIGAL80.
19. Anvendelse af den rekombinante stamme af gæren Kluyveromyces lactis ifølge krav 18 til fremmed proteinekspression.
20. Rekombinant stamme af gæren Kluyveromyces lactis ifølge krav 18 til anvendelse ved vaccination.
21. Rekombinant stamme af gæren Kluyveromyces lactis ifølge krav 18 til anvendelse ved oral/mucosal vaccination mod BVD-viruset eller CSF-viruset.
