Den foreliggende opfindelse angår de hidtil ukendte cholecalciferol-derivater, 26,26,26-trifluor-la,25R-dihydroxycholecalciferol i det væsentlige fri for 25S-epimeren, og 26,26,26-trifluor-lo,25S-dihy-droxycholecalciferol i det væsentlige fri for 25R-epimeren, disse 5 derivater til anvendelse som terapeutisk aktive substanser, især til behandling af mælkefeber hos drægtige drøvtyggere, til behandling af sygdomstilstande karakteriseret ved lavere end normale niveauer af endogent produceret le,25-dihydroxycholecalciferol, og som specifikke induceringsmidler for celledifferentiering og inhibitorer af celle-10 formering, en fremgangsmåde til fremstilling af disse, et farmaceutisk præparat på basis af disse samt anvendelse af disse ved fremstilling af et farmaceutisk præparat.
Fremgangsmåden ifølge opfindelsen er ejendommelig ved det i krav 3's kendetegnende del angivne.
15 Cholecalciferolderivaterne ifølge opfindelsen er 26,26,26-trifluor-la,25-dihydroxycholecalciferol i form af 25R-epimeren i det væsentlige fri for 25S-epimeren, og i form af 25S-epimeren i det væsentlige fri for 25R-epimeren. I overensstemmelse med opfindelsen fremstilles disse forbindelser ved, at en forbindelse med den almene formel IV
20 CH, tf, ($Π· 0 hvor Y er trialkyleret Si, omsættes med carbanionen af [3S-(3a,5yS,Z)]-2-[2-methylen-3,5-bis-[ (1,1 - dime thyle thyl) dimethyls ilyloxy] cyclohexy liden] ethyl diphenyl-25 phosphinoxid, og silylbeskyttelsesgrupperne fjernes.
Ovennævnte reaktion kan udføres ved reduceret temperatur, fx under-50eC, fortrinsvis ved ca. -78°C, under en inert atmosfære, fx argon, i et inert opløsningsmiddel såsom en cyclisk ether, fortrinsvis te-trahydrofuran (THF). Omdannelsen af phosphinoxidet til den tilsva 2

rende carbanion kan udføres ved indledningsvis at behandle phosphi-noxidet med et alkyllithium, fx n-butyllithium.
Fjernelse af trialkylsilylbeskyttelsesgruppen kan udføres ved, at reaktionsproduktet behandles med en kationbytterharpiks under omgivel-5 sesbetingelser i et opløsningsmiddel såsom en halogeneret alkan, fortrinsvis methylenchlorid. De resulterende produkter kan oprenses fx ved chromatografi på silicagel.
I nærværende beskrivelse betegner "lavere alkyl" en ligekædet eller forgrenet alkylgruppe med 1-8 carbonatomer, fx methyl, ethyl, n-10 propyl, i-propyl og tert.butyl. "Aryl" betegner phenyl, der eventuelt er substitueret med alkyl, halogen, nitro, cyano og trifluorme-thyl. Eksempler på aralkylgrupper er benzyl og phenethyl.
De i ovennævnte fremgangsmåde anvendte indenonudgangsmaterialer kan fremstilles på følgende måde: 15 [IR-[10, [aS*,ØS*] ,3aa,4aØ,7aØ]]-octahydro-0,7a-dimethyl-4-[(1,1-di-methylethy1)dimethyls ilyloxy]-a-etheny1-IH-inden-1-ethano1 omdannes til [ IR- [ 1/3 (R*) ,3aa,40,7a/3]]-l-(4- chlor -1 -methyl - 2 - buteny 1) oc tahydro -4-[(1,l-dimethylethyl)dimethylsilyloxy]-7a-methyl-lH-inden ved behandling med thionylchlorid efterfulgt af pyridin. Det resulterende 20 allyliske chlorid omsættes derefter med et arylsulfinsyresalt, fx benzensulfinsyrenatriumsalt, til dannelse af den tilsvarende aryl-sulfonylf orbindelse, fx [ IR- [ 10 (R*) , 3aa, 4)8,7a/3 ] ] -1 - [ (4 -phenylsulf o -ny1)-1-methyl-2-butenyl]octahydro-4-[(l,l-dimethylethyl)dimethyls ily-loxy]-7a- methyl-lH-inden. Den nævnte inden hydrogeneres derefter 25 katalytisk under anvendelse af en palladium-på-carbon(Pd/C)-katalysator til dannelse af den tilsvarende sidekædemættede forbindelse [1R-[10(R*) , 3aa,40,7aØ] ] -1- [4-(phenylsulfonyl) -1-methylbutyl] -octahydro-4-[(1,1-dimethylethyl)-dimethylsilyloxy]-7a-methyl-lH-inden. Fuldendelsen af sidekæden til fremstilling af en forbindelse med den 30 almene formel I herunder udføres ved, at carbanionen af førnævnte sulfonylforbindelse, der er dannet ved behandling med n-butyllithium eller lithiumdiisopropylamid, omsættes med trifluoracetone.

En forbindelse med den almene formel I, som en blanding af epimerer 10 ved sidekædens 4-stilling, kan dearylsulfonyleres ved hjælp af et alkalimetal eller et alkalimetalamalgam i nærværelse af et alkalime-talphosphat i et inert organisk opløsningsmiddel såsom en lavere al-kanol, en cyclisk ether eller fortrinsvis en blanding deraf. Foretrukne reaktanter er natriumamalgam og dikaliumhydrogenphosphat.

Efter oprensning, fx ved chromatografi på silicagel, kan reaktionsproduktet omsættes med et epoxideringsmiddel såsom en persyre, for-20 trinsvis trifluorpereddikesyre, ved reducerede temperaturer, fx ved 0°C. Reaktionen udføres hensigtsmæssigt under omgivelses-betingelser i et inert opløsningsmiddel såsom en halogeneret alkan, fx en chloralkan, fortrinsvis methylenchlorid. Herved fås ketonen med 5 den almene formel IV ovenfor, hvor Y er hydrogen.
Den vundne keton kan derefter uden isolering behandles med et reagens, der indfører en trimethylsilylbeskyttelsesgruppe på sidekædens hydroxygruppe, fx med trimethylsilylimidazol, ved omgivelsestempera-tur under en inert atmosfære. De resulterende beskyttede forbindelser 10 har den almene formel IV, hvor Y er trialkyleret Si.
25R- og 25S-26,26,26-trifluor-la,25-dihydroxycholecalciferol har vitamin D3-lignende virkning og udviser antiproliferative og differentiationsinducerende virkninger. Disse antiproliferative og differentieringsinducerende virkninger på HL-60-celler kan påvises in 15 vitro under anvendelse af kendte fremgangsmåder, fx som beskrevet i Progress in Cancer Research and Therapy, bind 23: "Maturation Factors and Cancer." Dataene i tabellen antyder, at 26,26,26-trifluor-la,25-dihydroxycho-lecalciferol, i 25R- eller 25S-form, begrænser formeringen af humane promyelocytiske tumorceller in vitro, selv om de ikke var toxiske over for cellerne. Endvidere blev celler dyrket i lave koncentratio-5 ner af disse forbindelser (10"^ til 10"® molær) induceret til at differentiere hen imod en mere moden celletype som påvist ved opnåelse af enzymaktivitet og cellefunktion. Det kan forventes, at de ovennævnte forbindelser er nyttige ved behandling af sygdomme såsom neoplastiske sygdomme, som til dels skyldes afvigende celleformering 10 og/eller differentiering.

Der kan være forskellige andre materialer til stede såsom overtræksmidler, fx 30 shellak, eller til på anden måde at modificere doseringsenhedens fysiske form.
De ovennævnte forbindelser kan også administreres til behandling af mælkefeber hos drægtige drøvtyggere før fødsel i doser i området 200-1000, fortrinsvis 25-200 pg/dag under anvendelse af konventio-35 nelle formuleringer. Fx kan sterile præparater til injektion og/eller

10 EKSEMPEL 1 a) En opløsning af 2,9 g (8,22 mmol) [IR- [ίβ, [aS*,j8S*] ,3aa,4aj8,7a/J] ] -octahydro-j9,7a-dimethyl-4- [ (1, l-dimethylethyl)dimethylsilyloxy] -a-ethenyl-IH-inden-1-ethanol i 100 ml vandfri ether blev afkølet til 0°C og under argon behandlet dråbevis med 2,76 ml (37,84 mmol) thio- 15 nylchlorid efterfulgt af 0,276 ml pyridin. Blandingen blev omrørt ved 0°C i 2 timer, hvorefter reaktionen blev standset ved tilsætning af 50 ml af en 2N natriumkaliumtartratopløsning. Etherfasen blev skilt fra, og den vandige fase blev ekstraheret med ethylacetat. De forenede organiske faser blev vasket med IN saltsyre, vand, 2N kalium-20 hydrogencarbonat og saltvand, tørret og inddampet. c) En opløsning af 1,00 g (2,10 mmol) af produktet fra lb) i 40 ml ethanol blev hydrogeneret ved stuetemperatur og under normaltryk over 5 350 mg 10%'s Pd/C. Efter 2 timer blev katalysatoren filtreret fra, og filtratet blev inddampet til tørhed, hvilket gav 1,00 g [IR- [1/3-(R*) , 3aa,4/?,7a/J] ] -1- [4-(phenylsulfonyl)-l-methylbutyl] -octahydro-4-[ (1, l-dimethylethyl)dimethylsilyloxy] -7a-methyl-lH-inden.

Efter omrøring i 15 minutter ved 0eC blev den resulterende opløsning afkølet ved -78°C og fortyndet med 10 ml vandfrit THF. Den blev derefter behandlet dråbevis med en opløsning af 1,00 g 15 (2,09 mmol) af sulfonen fra lc) i 15 ml THF og omrørt ved -78°C i 15 minutter og derefter ved 0°C i 30 minutter. Efter afkøling igen ved-78°C blev blandingen behandlet med 0,50 ml (5,59 mmol) 1,1,1-tri-fluoracetone og omrørt ved samme temperatur i 1,5 time. Reaktionen blev derefter standset ved tilsætning af 30 ml af en i:l-blanding af 20 2N natriumkaliumtartrat og 2N kaliumhydrogencarbonat, hvorefter reaktionsblandingen fik lov at varme op til stuetemperatur og blev ekstraheret med methylenchlorid. f) En opløsning af 0,260 ml (1,84 mmol) trifluorpereddikesyrearihydrid i 5 ml vandfrit methylenchlorid blev afkølet til 0°C, behandlet med 0,052 ml (1,89 mmol) 90%'s hydrogenperoxid og omrørt i 30 minutter.
Den resulterende opløsning blev sat til 400 mg af olefinen fra le) 10 (0,92 mmol) og 1,5 g dikaliumhydrogenphosphat i 5 ml methylenchlorid ved 0eC. Efter tilsætningen fik blandingen lov at varme op til stuetemperatur i 30 minutter, reaktionen blev standset med 5 ml af en 10%'s vandig opløsning af natriumsulfit, og blandingen blev fortyndet med en 10%'s vandig natriumhydrogencarbonatopløsning og ekstraheret 15 med methylenchlorid. De organiske ekstrakter blev vasket med saltvand, tørret og inddampet, hvilket gav 190 mg rent beskyttet produkt, [IR- [1)8(R*) ,3aa,4/J,7a/}] ] -octahydro:l-(6,6,6-trifluor-l,5-dimethyl-4,5-epoxyhexyl)-4-[(1,l-dimethylethyl)dimethylsilyloxy]-7a-methyl-lH-inden som en blanding af epimerer og 140 mg desilyleret produkt.
20 Efter oprensning ved flashchromatografi kan begge produkter anvendes i næste trin.
g) Til en suspension af 100 mg (2,63 mmol) lithiumaluminiumhydrid i 3 ml vandfri ether blev der ved stuetemperatur og under en argonatmosfære sat en opløsning af 190 mg (0,42 mmol) af det beskyttede pro- 25 dukt fra If) i 2 ml ether. Den resulterende blanding blev omrørt i 24 timer, hvorefter reaktionen blev standset ved tilsætning af 2N vandigt natriumkaliumtartrat, og blandingen blev ekstraheret med ether. h) En opløsning af 175 mg (0,39 mmol) af blandingen af epimerer fra lg) i 9 ml methanol blev behandlet med 2,8 g af en kat ionby tterhar- 35 piks (AG 50W-X4, 0,074-0,037 mm, Bio-Rad Laboratory, Richmond, CA,

11 USA) og omrørt ved stuetemperatur i 12 dage. Harpiksen blev fjernet ved filtrering og vasket med methanol. Filtraterne blev inddampet til tørhed. Inc., Clifton, NJ) med chloroform/ethylacetat, hvilket gav 65 mg krystallinsk 5R*-produkt [IR- [1)3(IR*, 5R*) , 3aa,Ufi, 7a/J] ] -octahydro-1-(6,6,6-trifluor-5-hydroxy-l,5-dimethylhexyl)-7a-methyl-lH-inden-4-ol, smeltepunkt 110-llleC, [a]§^ - +40,3® (c - 0,2 i chloroform) og 55 10 mg 5S*-produkt [IR-[1/8(1R*,5S*) ,3aa,4/8,7a/8] ]-octahydro-l-(6,6,6-tri-fluor-5-hydroxy-1,5-dimethylhexyl)-7a-methyl-lH-inden-4-ol, [o]g5 - +31, 6° (c - 0,2 i chloroform).
i) En opløsning af 50 mg (0,15 mmol) af 5R*-diolen fra lh) i 2 ml methylenchlorid blev sat til en opslæmning af 97 mg (0,45 mmol) py-15 ridiniumchlorchromat i 4 ml methylenchlorid. Den resulterende blanding blev omrørt ved stuetemperatur i 1,5 time og derefter fortyndet med 10 ml ether, omrørt i 15 minutter og filtreret gennem kieselgur. Remanensen fra filtreringen blev tritureret med ether. Triturerings-ekstrakterne blev forenet, filtreret og inddampet til tørhed. Rema-20 nensen blev oprenset ved chromatografi med hexan/ethylacetat. De 49 mg produkt blev opløst i 4 ml vandfrit methylenchlorid, behandlet med 0,13 ml (0,89 mmol) trimethylsilylimidazol og omrørt ved stuetemperatur under argon i 22 timer. Efter tilsætning af 0,5 ml vand blev blandingen omrørt i 20 minutter og derefter fortyndet med vand og 25 ekstraheret med ethylacetat. Ekstrakterne blev vasket med vand og saltvand, tørret og inddampet til tørhed. Remanensen blev oprenset ved chromatografi med hexan/ethylacetat, hvilket gav 51 mg [1R-[ 1/3(IR*,5R*) ,3aa,4/9,7a/3] ] -octahydro-1-(6,6,6 -trifluor-5-trimethyl-s i -lyloxy-l,5-dimethylhexyl)-7a-methyl-lH-inden-4-on som en tyk olie.
30 j) En opløsning af 141 mg (0,23 mmol) [3S-(3a,5/8,Z)]-2-[2-methylen-3,5-bis[(1,l-dimethylethyl)dimethylsilyloxy]cyclohexyliden]ethyldi-phenylphosphinoxid i 6 ml vandfrit THF blev afkølet til -78°C og under argon behandlet dråbevis med 0,137 ml (0,22 mmol) af en 1,6 molær opløsning af n-butyllithium i hexan. Den resulterende blanding blev omrørt ved -78°C i 1,5 time, derefter behandlet med 3 ml af en l:l-blanding af en 2N natrium-kaliumtartrat- og en 2N kaliumhydrogencarbonatopløsning, fik lov at varme op til stuetemperatur, blev fortyndet med vand og ekstraheret 5 med vand og ethylacetat. De forenede organiske faser blev vasket med saltvand, tørret og inddampet til tørhed. Remanensen blev oprenset ved filtrering gennem silicagel, elueret med hexan/ethylacetat, derefter opløst i 0,5 ml methylenchlorid og 5 ml methanol og omrørt ved stuetemperatur natten over sammen med 2 g af en kationbytterharpiks 10 (AG 50W-X4). Efter filtrering og afdampning af opløsningsmidlet blev remanensen oprenset ved chromatografi med hexan/ethylacetat, hvilket gav 23 mg 26,26,26-trifluor-le,25R-dihydroxycholecalciferol.
