Den foreliggende opfindelse angår en særlig fremgangsmåde til fremstilling af orthohydroxyalkylerede phenoler ved indføring af I-hydroxyalkyl eller or-hydroxyara!kyl i orthostillingen i phenoler.
Det er kendt at fremstille o-hydroxyalkylerede phenoler ved 05 omsætning af en phenol, et aldehyd og borsyre eller borater under dannelse af en cyklisk borforbindelse, som derefter nedbrydes til det ønskede produkt, jvf. fx. fransk patentskrift nr. 1.331.630, danske patentskrifter nr. 106859 og nr. 126775, eller ved reducering af formyl eller carboxy i a-stillingen i phenoler til benzylalko-10 hol, Rec. Trav. Chim. 79, 825 (1960), japansk patentskrift nr.
39-17518 (1964) og britisk patentskrift nr. 774696. Disse fremgangsmåder er imidlertid ikke særlig velegnede, enten fordi de har et begrænset anvendelsesområde, fordi de kræver besværlige procedurer, giver det ønskede produkt i et kommercielt utilstrækkeligt udbytte 15 eller ikke tillader genvinding af udgangsmaterialer.
Eftersom orthohydroxyalkylerede phenoler i mange tilfælde udgør nyttige råmaterialer i den kemiske og farmaceutiske industri og desuden er mellemprodukter ved fremstilling af vigtige naturligt forekommende forbindelser, herunder saligenin, er der behov for en 20 fremgangsmåde, som er generelt anvendelig og billigere.
Ved fremgangsmåden ifølge den foreliggende opfindelse fremstilles o-hydroxyalkylerede phenoler, som har den almene formel:

Fremgangsmåden ifølge den foreliggende opfindelse beror på, at man omsætter en phenol med benzenborsyre og et aldehyd under tilstedeværelse af en katalytisk mængde af en organisk carboxylsyre, 20 og derefter nedbryder produktet, under indføring af 1-hydroxyalkyl eller α-hydroxyaralkyl i orthostillingen til den phenoliske hydroxy-gruppe i phenolen. De særlige fordele ved denne fremgangsmåde er, at den har et bredt anvendelsesområde med hensyn til o-hydroxyal-kylering af phenoler, giver et højt udbytte, sædvanligvis 90-100%, 25 med mulighed for genvinding af udgangsphenolen, og at det ønskede produkt let og på ren form kan udvindes fra reaktionsblandingen.
Et simpelt eksempel er vist i følgende reaktionsligning, hvor phenol benyttes som udgangsmateriale.

Substituenterne i den som udgangsmateriale anvendte phenol er sådanne, som ikke forstyrrer reaktionen væsentligt. Eksempler på 05 sådanne substituenter er alkyl (fx. methyl, ethyl, propyl, isopropyl, butyl, amyl), aryl (fx. phenyl, tolyl, xylyl, naphthyl), pyridyl, aralkyl (fx. benzyl, phenethyl), alkoxy (sådanne som indeholder førnævnte alkylgrupper), aryloxy (sådanne der indeholder de førnævnte arylgrupper) eller aralkoxy. Førnævnte alkyl, aryl, aralkyl, 10 alkoxy, aryloxy og aralkoxy kan yderligere være substitueret med alkyl, aryl, aralkyl, alkoxy, aryloxy, aralkoxy, halogen, carboxyl, alkoxycarbonyl, nitro eller cyano. Phenolerne kan være kondenserede med en benzenring; udgangsmaterialerne bliver så naphtholer. Naph-tholerne kan være 1-naphtholer eller 2-naphtholer, og førnævnte 15 substituenter kan sidde på den anden ring end den, hvor hydroxy-gruppen sidder. Både ved phenolerne og naphtholerne kan førnævnte substituenter sidde hvorsomhelst, idet dog den ene af orthostillinger-ne til den phenolske hydroxygruppe naturligvis udelukkes.
Benzenborsyren, der omsættes med ovennævnte forbindelser, 20 kan være den fri benzenborsyre eller syreanhydridet.
De anvendte aldehyder vil være aliphatiske eller aromatiske aldehyder, fx. formaldehyd, acetaldehyd, propionaldehyd, butyraldehyd, benzaldehyd og phenylacetaldehyd. Disse forbindelser kan være substituerede med ikke-reaktive substituenter som nævnt ovenfor eller 25 kan være derivater som fx. paraldehyder.
Den organiske carboxylsyre, der sættes til reaktionsmediet, virker som en slags katalysator, og en hvilken som helst type syre kan benyttes, når blot den indeholder en carboxylgruppe. Foretrukne eksempler på almindeligvis anvendte syrer er lavere alkansyrer 30 såsom eddikesyre, propionsyre og smørsyre. Om nødvendigt kan der anvendes en stærk syre såsom trichloreddikesyre.
Reaktionen udføres i et vandfrit opløsningsmiddel indeholdende 0,1-1,0 mol af den organiske carboxylsyre, under opvarmning og samtidig fjernelse af det dannede vand, under anvendelse af benzen-35 borsyren og aldehydet i et lille overskud i forhold til phenolen eller naphtholen. Opløsningsmidlet der benyttes under reaktionen, er et aprot, vandfrit opløsningsmiddel, især benzen, toluen eller xylen, 4 143332 således at man azeotropisk kan fjerne dannet vand, efterhånden som reaktionen skrider frem. Med henblik pi effektiv fjernelse af vand kan man også anvende et tørremiddel, fx. silicagel, molekylesi (handelsnavn Linde Co.) vandfri calciumsulfat, vandfri calciumchlorid 05 eller vandfri magnesiumsulfat. Reaktionstiden afhænger af aktiviteten af udgangsphenol, aldehyd og organisk carboxylsyre, reaktionstemperatur og anvendt opløsningsmiddel, og der kræves sædvanligvis adskillige timer eller døgn for fuldstændig omdannelse.
De fremkomne mellemprodukter er cykliske estere mellem den 10 ønskede o-hydroxybenzylalkohol og benzenborsyre, og disse estere behøver ikke nødvendigvis at oprenses, men kan nedbrydes direkte. Nedbrydningen udføres ved hydrolyse eller alkoholyse, fortrinsvis under alkaliske eller svagt sure betingelser, men forløber også glat under neutrale betingelser for visse mellemprodukter. Alkoholysen 15 kan udføres med methanol, ethanol, propanol og lignende. Til alkoholysen anvendes dog fortrinsvis polyalkoholer såsom ethylenglycol, propylenglycol, trimethylenglycol eller glycerin, fordi benzenborsyre og glycoler danner stabile cykliske estere. Nedbrydningen under sure betingelser ledsages let af sidereaktioner og bør derfor udføres 20 I et kun svagt surt medium; som syre kan man anvende organiske syrer såsom eddikesyre eller propionsyre.
Alle reaktionstrin går fint og selektivt under dannelse af de ønkede o-hydroxyalkylerede phenoler i ca. 90% udbytte og i nogle tilfælde i kvantitativt udbytte.
25 Som nævnt ovenfor udgør de o-hydroxyalkylerede phenoler en lang række industrielt vigtige forbindelser, herunder saligenin, og kan især anvendes som mellemprodukter ved fremstilling af o-hydroxy-methylphenyleddikesyrer, der er nyttige plantevækstregulerende midler eller protoberberinalkaloider, der er nyttige som lægemidler 30 for dyr og mennesker. Den foreliggende opfindelse tilvejebringer således en hidtil ukendt industrielt anvendelig fremgangsmåde til fremstilling af industrielt nyttige o-hydroxyalkylerede phenoler i højt udbytte.
Fremgangsmåden ifølge opfindelsen belyses i de følgende eksemp- 35 ler.
Eksempel 1
Til en opløsning af 1,882 (0,02 mol) phenol, 2,439 g (0,02 mol) 5 143332 benzenborsyre og 148 mg (2 mmol) propionsyre i 50 ml vandfri benzen sættes 1,0 g paraformaidehyd under omrøring og tilbagesvaling, idet man fjerner vandet azeotropisk. Med 2 timers mellemrum sættes 0,5 g paraformaidehyd til, og efter 4 timer tilsættes yderligere 05 0,488 g (4 mmol) benzenborsyre, idet den azeotropiske fjernelse af vand fortsættes. Der går ialt 10 timer, før phenolen er forsvundet (ifølge TLC). Reaktionsblandingen hældes i en blanding af methylen-chlorid og vand, det uomdannede paraformaidehyd filtreres fra og filtratet ekstraheres med methylenchlorid. Ekstrakten vaskes med 10 vand, tørres og inddampes, hvorved fås 4,114 (97,76% udbytte) af den cykliske ester mellem benzenborsyre og o-hydroxybenzylalkohol.
Denne omkrystalliseres fra petroleumsether, hvorved fås rene krystaller med et smeltepunkt på 36-38°C. NMR (CDCIg) 6: 5,15 (2H, singlet), 7,07 (4H, multiplet) 7,43 (3H, multiplet), 8,00 (2H, multiplet) ppm.

Ovennævnte cykliske ester (4,20 g, 0,02 mol) opløses i 150 ml trimethylenglycol sammen med 120 mg eddikesyre, og blandingen omrøres ved stuetemperatur i 6 timer. Reaktionsblandingen ekstraheres 20 med ether, og ekstrakten vaskes omhyggeligt med vand, tørres og inddampes til tørhed. Remanensen omkrystalliseres fra petroleumsether, hvorved fås 2,107 g (84,86% udbytte) sallgenol (smeltepunkt 86-87°C).
Eksempel 2 25 11,0 g 3-hydroxy-4-methoxyphenyleddikesyre opløses i 550 ml
benzen og koges under tilbagesvaling for azeotropisk fjernelse af vand. Derefter sættes 14,0 g benzenborsyre til, og der koges under tilbagesvaling i ca. én time, idet det dannede vand fjernes. (I dette eksempel virker phenyleddikesyren, der anvendes som udgangsmate-30 riale, som carboxylsyrekatalysatoren). Ved 2-3 timers mellemrum sættes 2-3 g paraformaidehyd til, idet fjernelsen af vand fortsættes, og efter 9 timer sættes yderligere 1,0 g benzenborsyre til. Reaktionen er færdig efter 20 timer, og opløsningsmidlet fjernes fuldstændigt ved destillation under reduceret tryk. Den krystallinske remanens 35 anbringes efter tilsætning af 150 ml vand i et oliebad ved 90-l00°C
i 1,5 time under omrøring. Reaktionsblandingen afkøles til stuetemperatur, og de udfældede krystaller filtreres fra og omkrystalliseres 6 143332 fra acetone-ether, hvorved fås 9,70 g (93% udbytte) 3-hydroxy-2- hydroxy-methyl-4-methoxyphenyleddikesyre i form af δ-lactonen med et smeltepunkt pi 183-185°C. IR yCHCI3 3540, 1740, 1600, 1277, 1030, max 05 893 cm \ NMR (dg-DMSO) δ 3,80 (3,80 (3H, singlet), 3,65 (2H, singlet, 5,32 (2H, singlet), 6,70 , 6,90 (AB kvartet, J = 8,0-Hz), 8,07 (1H, singlet) ppm. Analyse: beregnet for C^qH^qO^: C, 61,85; H, 5,19. Fundet: 10 C, 61,70; H, 5,17.
Eksempel 3
Til en opløsning af 1,286 g 4-chlorphenol og 1,219 g benzenborsyre i 30 ml vandfri benzen sættes 0,36 g propionsyre, og blanding-15 en koges under tilbagesvaling og omrøring. Derefter sættes 1,0 g paraformaldehyd til, og kogningen under tilbagesvaling fortsættes, idet det dannede vand fjernes azeotropisk. Med 2-3 timers mellemrum sættes 0,5 g paraformaldehyd til, og efter 9 timer sættes yderligere 0,244 g benzenborsyre til. Kogningen under tilbagesvaling fortsættes 20 i ialt 15 timer. Reaktionsblandingen hældes i en blanding af methylen-chlorid og vand, og det udfældede paraformaldehyd fjernes ved filtrering. Filtratet ekstraheres med methylenchlorid og ekstrakten vaskes med vand, tørres og inddampes til tørhed. Remanensen omkrystalliseres fra petroleumsether, hvorved fås 2,209 g (90,35% udbytte) af 25 den cykliske ester mellem 2-hydroxy-5-chlorbenzylalkohol og benzenborsyre med et smeltepunkt på 101-102°C. NMR (CDCIg) δ 5,13 (2H, singlet) 7,01 (3H, multiplet) 7,40 (3H, multiplet), 7,97 (2H, multiplet) ppm.
Denne cykliske ester nedbrydes i ethylenglycol, hvorved fås 30 2-hydroxy-5chlorbenzylalkohol (smeltepunkt 92-93°C) i kvantitativt udbytte.
Eksempler 4-15 Følgende udføres på samme måde som i eksempel 1. De nævnte 35 udbytter refererer til den cykliske ester af benzenborsyre; udbyttet ved nedbrydningstrinet er næsten kvantitativt (90% eller mere). Medmindre andet nævnes anvendes paraformaldehyd som aldehyd, propionsyre som carboxylsyre og benzen som opløsningsmiddel, og reaktionstiden er på 3-30 timer.

Eksempel 6 4- methylphenol (p-cresol) giver esteren i 97% udbytte, og ved nedbrydning af denne fis 2-hydroxy-5-methylbenzylalkohol; smp.

Eksempel 7 2-chlorphenol giver esteren i 38% udbytte (faktisk i 90% udbytte eller mere fordi udgangsphenolen genvindes kvantitativt), og ved nedbrydning af denne fis 2-hydroxy-3-chlorbenzylalkohol; smp.
20 115°C. 2-chlorphenol genvindes kvantitativt i en mængde pi 60% af udgangsmængden ved destillation af ekstrakten af den cykliske ester fra reaktionsblandingen med methylenchlorid.
Eksempel 8 25 3-methoxyphenol giv,er esteren i 99% udbytte, og ved nedbryd ning af denne fås en 1:1 blanding af 2-hydroxy-4-methoxybenzylalkohol og 2-hydroxy-6-methoxybenzylalkohol; kp. °C: 100-105/0,3-0,5 mm Hg.
