Description
- GEBIET DER ERFINDUNG
- Die vorliegende Erfindung betrifft Brenzcatechin-Hydrazon-Derivate, die die enzymatische Aktivität von Phosphodiesterase IV und dem Tumor-Nekrose-Faktor inhibieren. Diese Verbindungen können bei der Verhinderung oder Behandlung von Asthma bronchiale, Arthritis, Bronchitis, chronischem atretischem Luftweg, Psoriasis, allergischer Rhinitis, Dermatitis, AIDS, Crohn-Krankheit, Septikämie, septischem Schock, anderen entzündlichen Erkrankungen, wie Kachexie, mit TNF verbundenen Erkrankungen, etc., nützlich sein. Die vorliegende Erfindung betrifft auch ein Verfahren zur Herstellung der besagten Verbindungen und eine pharmazeutische Zusammensetzung, die die besagte Verbindung enthält.
- HINTERGRUND DER ERFINDUNG
- Phosphodiesterase IV ist ein Enzym, das spezifisch cAMP (zyklisches Adenosin-3',5'-monophosphat) in inaktives Adenosin-3',5'-monophosphat hydrolysiert. Von cAMP wurde gezeigt, dass es ein zweiter Bote ist, der die zellulären Antworten auf externe Reize vermittelt und entspannend oder anspannend auf Bronchialmuskeln wirkt.
- Die Inhibierung von Phosphodiesterase IV führt zur Verhinderung von Bronchospasmus, indem die Konzentration an cAMP aufrechterhalten wird, und induziert ebenfalls Entzündungshemmung. Daher sollten Verbindungen, die Phosphodie rase IV inhibieren, wirksam bei der Behandlung von Asthma und ähnlichen Erkrankungen sein.
- Es ist bekannt, dass der Tumor-Nekrose-Faktor (TNF) bei einer ansteckenden Erkrankung wie Kachexie und einer Autoimmunerkrankung beteiligt ist. TNF scheint auch als primärer Mittler für eine entzündliche Reaktion, wie Septikämie und septischen Schock, zu wirken.
- Daher wird erwartet, dass Verbindungen mit inhibitorischer Aktivität gegen Phosphodiesterase IV oder TNF pharmazeutisch wertvoll sind und es besteht immer ein Bedarf an der Entwicklung neuer Verbindungen, die Phosphodiesterase IV und TNF inhibieren.
- Viele Verbindungen sind als Inhibitoren für Phosphodiesterase IV und TNF vorgeschlagen worden. Beispielsweise beschreibt

- ZUSAMMENFASSUNG DER ERFINDUNG
- Die vorliegende Erfindung betriff Brenzcatechin-Hydrazon-Derivate der Formel oder pharmazeutisch verträgliche bzw. akzeptable Salze davon, wobei
R1 C3-7-Cycloalkyl ist;
R2 Wasserstoff, Hydroxy, C1-5-Alkyl oder -CH2CH2C(=O)NH2 ist;
R3 und R4 unabhängig voneinander Wasserstoff, C1-7-Alkyl, -C(=X)-R5, oder 2-, 3- oder 4-Pyridyl, Pyrimidyl oder Phenyl substitutiert mit einem oder zweien aus gewählt aus einer Gruppe bestehend aus Halogen, C1-6-Alkoxy, Nitro, Trifluormethyl, C1-6-Alkyl und Carboxyl sind, oder R3 und R4 durch C3-4-enthaltenden Sauerstoff, Schwefel oder Stickstoff unter Bildung eines heterozyklischen Rings direkt miteinander verbunden sind, X Sauerstoff, Schwefel oder NH ist, und R5 C1-7-Alkyl, -NHR6, CONH2 oder 2-, 3- oder 4-Pyridyl, Pyrimidyl oder Phenyl substituiert mit einem ausgewählt aus einer Gruppe bestehend aus Halogen, C1-6-Alkoxy, Nitril, Trifluormethyl, C1-6-Alkyl und Carboxyl ist, und R6 Wasserstoff, Hydroxy, NH2, C1-5-Alkoxy, C1-5-Alkyl, Pyridyl oder Phenyl ist. - GENAUE BESCHREIBUNG DER ERFINDUNG
- Die Verbindung der Formel I kann in Form der optischen Isomere oder Stereoisomere vorliegen. Somit schließt die vorliegende Erfindung derartige Isomere und Gemische davon ein.
- Die vorliegende Erfindung stellt eine pharmazeutische Zusammensetzung zur Inhibierung von Phosphodiesterase IV oder TNF zur Verfügung, die eine Verbindung der Formel I und einen pharmazeutisch verträglichen bzw. akzeptablen Träger umfasst.

- Einige Derivate wurden durch ein bekanntes Verfahren (J. Med. Chem., 1994, 37, 1969) synthetisiert. Hydrazin-Verbindungen wurden in einer Ausbeute von 60% bis 90% in Alkohollösungsmittel unter Verwendung eines Säurekatalysators (Tetrahedron Lett. 1994, 35, 3711) synthetisiert.
- Die Erfindung wird nun unter Bezugnahme auf die folgenden veranschaulichenden Beispiele beschrieben.

- VERGLEICHSBEISPIEL 1
- 3-Cyclopentyloxy-4-methoxybenzaldehyd
- Eine Lösung aus 100 g (0,66 Mol) Isovanillin, 136,2 g (0,99 Mol) wasserfreiem Kaliumcarbonat und 3 g Kaliumiodid in 650 ml wasserfreiem Dimethylformamid wurde bei 65°C gerührt. 127,3 g (0,85 Mol) Cyclopentylbromid wurden langsam tropfenweise über eine Stunde zu der Lösung hinzugefügt. Diese Lösung wurde einen Tag bei 65°C gerührt und dann wurde ihre Temperatur auf Raumtemperatur abgesenkt. Sie wurde mit 2,01 Toluol verdünnt und mit 1 M Natriumhydroxid (2 × 1,5 l) gewaschen. Die wässrige Schicht wurde mit 0,5 l Toluol extrahiert und die organische Schicht wurde mit destilliertem Wasser (2 × 1,5 l) gewaschen. Die organische Schicht wurde getrocknet und unter Erhalt von 117 g hellbrauner öliger Titelverbindung aufkonzentriert.
1H-NMR (CDCl3, d): 9,84(s, 1H) 7,42(m, 2H) 6,95(d, 1H, J = 9 Hz) 4,87(m, 1H) 3,93 (s, 3H) 2,1–1,6(m, 8H) - Eine katalytische Menge konzentrierter Schwefelsäure wurde einer Lösung von 0,44 g (2,0 mMol) Verbindung, die in Vergleichsbeispiel 1 hergestellt wurde, in 30 ml Ethanol zugefügt und das Gemisch wurde 10 Minuten bei Raumtemperatur gerührt. 0,33 g Isonikotinhydrazid wurde der Reaktionslösung zugegeben. Die Lösung wurde 4 Stunden bei 50°C gerührt und unter verringertem Druck aufkonzentriert. Der Rückstand wurde in Dichlormethan gelöst und zweimal mit 50 ml destilliertem Wasser gewaschen. Die abgetrennte organische Schicht wurde über wasserfreiem Magnesiumsulfat getrocknet und unter verringertem Druck destilliert. Das resultierende weiße Kristallisat wurde in Dichlormethan unter Erhalt von 0,67 g (88,45%) weißer Titelverbindung umkristallisiert. Smp. 170–171°C

Die Lösung wurde 4 Stunden bei 50°C gerührt und unter verringertem Druck aufkonzentriert. Der Rückstand wurde in Dichlormethan gelöst und zweimal mit 50 ml destilliertem Wasser gewaschen. Die abgetrennte organische Schicht wurde über wasserfreiem Magnesiumsulfat getrocknet und unter verringertem Druck destilliert. Das resultierende weiße Kristallisat wurde in Dichlormethan unter Erhalt von 1,25 g (89,87%) weißer Titelverbindung umkristallisiert. Smp. 146–147°C

Das resultierende hellgelbe Öl wurde durch eine Flash-Chromatographie (Kieselsäuregel bzw. Silicagel, 7,5% Methanol-Dichlormethan als Entwicklungslösung) unter Erhalt von 0,45 g (65,56%) weißer Titelverbindung aufgereinigt. Smp. 87–90°C

- Inhibierung der Aktivität von Phosphodiesterase IV
- Die in den Beispielen 1 bis 16 hergestellten Verbindungen und Rolipram als Kontrolle wurden auf die Inhibierung von Phosphodiesterase IV getestet.
- Teilweise aufgereinigte Phosphodiesterase IV aus Human-U937-Zellen, Testverbindung und 1,0 uM cAMP einschließlich 0,01 uM[3H]cAMP wurden 20 Minuten bei 30°C inkubiert. Die PDE-Reaktion zur Umwandlung von cAMP in AMP wurde durch 2-minütiges Kochen der Reaktionslösung vervollständigt. AMP wurde in Adenosin umgewandelt, indem Schlangengift-Nukleotidase zugefügt wurde und die Reaktionslösung 10 Minuten bei 30°C inkubiert wurde. Während nicht-hydrolysiertes cAMP an AG1-X2-Harz gebunden wurde, wurde das [3H]Adenosin in der wässrigen Lösung durch Szintillationszählung quantitativ bestimmt. Die Ergebnisse sind in Tabelle I unten gezeigt, in der die Werte die Inhibierung (%) der PDE IV für jede Testverbindung anzeigen.

- Verbindung der Formel I: oder pharmazeutisch verträgliche bzw. akzeptable Salze davon, wobei R1 C3-7-Cycloalkyl ist; R2 Wasserstoff Hydroxy, C1-5-Alkyl oder -CH2CH2C(=O)NH2 ist; R3 und R4 unabhängig voneinander Wasserstoff, C1-7Alkyl, -C(=X)-R5, oder 2-, 3- oder 4-Pyridyl, Pyrimidyl oder Phenyl substitutiert mit einem oder zweien ausgewählt aus einer Gruppe bestehend aus Halogen, C1-6-Alkoxy, Nitro, Trifluormethyl, C1-6-Alkyl und Carboxyl sind, oder R3 und R4 durch C3-a-enthaltenden Sauerstoff, Schwefel oder Stickstoff unter Bildung eines heterozyklischen Rings direkt miteinander verbunden sind, X Sauerstoff, Schwefel oder NH ist, und R5 C1-7Alkyl, -NHR6, CONH2, oder 2-, 3- oder 4-Pyridyl, Pyrimidyl oder Phenyl substituiert mit einem ausgewählt aus einer Gruppe bestehend aus Halogen, C1-6-Alkoxy, Nitril, Trifluormethyl, C1-6-Alkyl und Carboxyl ist, und R6 Wasserstoff, Hydroxy, NH2 C1-5-Alkoxy, C1-5-Alkyl, Pyridyl oder Phenyl ist.
- Verbindung nach Anspruch 1, wobei R1 Cyclopentyl ist, R2 Wasserstoff ist, R3 und R4 unabhängig voneinander C1-5-Alkyl, -C(=X)-R5, oder 2-, 3- oder 4-Pyridyl, Pyrimidyl oder Phenyl substituiert mit einem ausgewählt aus einer Gruppe bestehend aus Halogen, Nitro, Trifluormethyl und Carboxyl sind, oder R3 und R4 durch C3-4-enthaltenden Sauerstoff oder Stickstoff unter Bildung eines heterozyklischen Rings direkt miteinander verbunden sind, X Sauerstoff, Schwefel oder NH ist, und R5 -NHR6 oder 4-Pyridyl ist, und R6 Wasserstoff Hydroxy oder 4-Pyridyl ist.
- Verbindung nach Anspruch 1, ausgewählt aus einer Gruppe bestehend aus (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-isonikotin-hydrazon, (E)-Ethyl[(3-cyclopentyloxy-4-methoxyphenyl)methylen]hydrazinoformiat, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-phenylhydrazon, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-acethydrazon, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-7-chlorchinolon-4-ylhydrazon, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-2-imidazolinohydrazon, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazincarboxamid, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-2-nitrophenylhydrazon, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazincarbothioamid, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-4-chlorphenylhydrazon, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazincarbonylmethyl(trimethyl)-ammoniumchlorid, (E)-N-(1,4-Oxazin-4-yl)-3-cyclopentyloxy-4-methoxyphenylmethanimin, (E)-N-Piperidino-3-cyclopentyloxy-4-methoxyphenylmethanimin, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazincarboximidamid, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-2-pyridinylhydrazon, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-2-carboxyphenylhydrazon, (E)-3-Cyclopentyloxy-4-methoxybenzaldehyd-4-trifluormethylpyrimidin-2-ylhydrazon, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazincarbohydrazin, (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazindicarboxamid und (E)-2-[(3-Cyclopentyloxy-4-methoxyphenyl)methylen]hydrazinoessigsäure.
- Verfahren zur Herstellung einer Verbindung der Formel I: oder pharmazeutisch verträglichen bzw. akzeptablen Salzen davon, gemäß Anspruch 1, wobei das Verfahren das Umsetzen von C3-7-Cycloalkyl mit einer Verbindung der Formel II: unter Herstellung einer Verbindung der Formel III: wobei R1 die gleiche wie in Anspruch 1 definierte Bedeutung hat, und das Umsetzen der Verbindung der Formel III mit einer Verbindung der Formel IV: wobei R2, R3 und R4 die gleiche wie in Anspruch 1 definierte Bedeutung haben, umfasst.
- Pharmazeutische Zusammensetzung zur Inhibierung von Phosphodiesterase IV oder TNF, die eine pharmazeutisch wirksame Menge der Verbindung nach Anspruch 1 und einen pharmazeutisch verträglichen bzw. akzeptablen Träger umfasst.
