Abstract
Un compuesto para usar en el tratamiento de esquizofrenia en un animal de sangre caliente que lo necesita, que tiene la siguiente estructura (I):**Fórmula** o una de sus sales, estereoisómeros o solvatos farmacéuticamente aceptables, en donde: m, n y p son, de modo individual, 0 ó 1; x es 1, 2 ó 3; A es un heterociclo opcionalmente sustituido; B es -O-, -NR6- o -S(O)z- donde z es 0, 1 ó 2; R es hidrógeno u oxo; R1 está ausente o representa 1, 2 ó 3 sustituyentes que son iguales o diferentes y son, de modo independiente, halógeno, alquilo C1-6, -CHF2, -CF3, -CH2NH2, -CH2NH(alquilo C1-6) o -CH2N(alquilo C1-6)2; R2 es, en cada aparición, igual o diferente y es, de modo independiente, hidrógeno, alquilo C1-6, - C(>=O)(alquilo C1-6), bencilo, -CH2CONH2, -CHF2, -CF3 o dos grupos R2 cualesquiera se pueden tomar juntos para formar un alcandiílo C1-6; y R3, R4, R5 y R6 son iguales o diferentes y son, de modo independiente, hidrógeno o alquilo C1-6.

Los compuestos de la fórmula (13), donde Y = metilo (Me), etilo (Et), etc. se pueden obtener en comercios o se pueden preparar por medio de métodos estándar. Los compuestos de la fórmula (13) en etanol se pueden hacer reaccionar con un compuesto de hidrazina, como (6), para obtener los compuestos de la fórmula (14). Los compuestos de la fórmula (14) se pueden hacer reaccionar luego con una variedad de derivados de benzaldehído, como (2), en disolventes alcohólicos, como etanol y en presencia de una cantidad catalítica de ácido, tales como ácido acético y a reflujo para proporcionar los compuestos de la estructura (VI).
Los compuestos de la presente invención se pueden utilizar en general como el ácido libre o la base libre. De modo alternativo, los compuestos de esta invención se pueden usar en la forma de sales por adición de ácidos o bases. Las sales por adición de ácidos de los compuestos amino libres de la presente invención se pueden preparar por medio de métodos bien conocidos en la técnica y se pueden formar a partir de ácidos orgánicos y ácidos inorgánicos. Los ácidos orgánicos apropiados incluyen ácido maleico, fumárico, benzoico, ascórbico, succínico, metansulfónico, acético, trifluoroacético, oxálico, propiónico, tartárico, salicílico, cítrico, glucónico, láctico, mandélico, cinnámico, aspártico, esteárico, palmítico, glicólico, glutámico y bencensulfónico. Los ácidos inorgánicos apropiados incluyen ácido clorhídrico, bromhídrico, sulfúrico, fosfórico y nítrico. Las sales por adición de bases incluyen aquellas sales que se forman con el anión del carboxilato e incluyen sales formadas con cationes orgánicos e inorgánicos tales como aquellos seleccionados de metales alcalinos y metales alcalinotérreos (por ejemplo, litio, sodio, potasio, magnesio, bario y calcio), así como el ion amonio y sus derivados sustituidos (por ejemplo, dibencilamonio, bencilamonio, 2-hidroxietilamonio, y similares). De esta manera, la expresión “sal farmacéuticamente aceptable” de las estructuras (I) a (VI) pretende comprender todas y cada una de las formas salinas aceptables.
Respecto de los estereoisómeros, los compuestos de las estructuras (I) a (VI) pueden tener centros quirales y se pueden producir como racematos, mezclas racémicas y como enantiómeros o diastereómeros individuales. Todas estas formas isoméricas están incluidas dentro de la presente invención, que incluye sus mezclas. Por otra parte, algunas de las formas cristalinas de los compuestos de las estructuras (I) a (VI) pueden existir como polimorfos, que están incluidos en la presente invención. Además, algunos de los compuestos de las estructuras (I) a (VI) también pueden formar solvatos con agua u otros solventes orgánicos. Estos solvatos se incluyen de modo similar dentro del alcance de esta invención.
En otra realización de la invención, se describen las composiciones farmacéuticas que contienen uno o varios compuestos de las estructuras (I) a (VI). Con fines de administración, los compuestos de la presente invención se pueden formular como composiciones farmacéuticas. Las composiciones farmacéuticas de la presente invención comprenden uno o varios compuestos de la presente invención y un portador y/o diluyente farmacéuticamente aceptable. El inhibidor de PDE10 está presente en la composición en una cantidad que es efectiva para tratar un trastorno particular —es decir, en una cantidad suficiente para lograr la inhibición deseada de PDE10 y, con preferencia, con toxicidad aceptable para el animal de sangre caliente. Normalmente, las composiciones farmacéuticas de la presente invención pueden incluir un inhibidor de PDE10 en una cantidad de 0,1 mg a 250 mg por dosis según la vía de administración y, más normalmente, de 1 mg a 60 mg. Las concentraciones y las dosis apropiadas pueden ser determinadas con facilidad por el experto en la técnica.
En términos generales, una dosis diaria típica puede variar de aproximadamente 1 µg/kg a 100 mg/kg, con preferencia, de 0,01-100 mg/kg, con mayor preferencia, de 0,1-70 mg/kg, según el tipo y la gravedad de la enfermedad, por ejemplo, por una o varias administraciones separadas. Para administraciones repetidas durante varios días o más tiempo, según la condición, el tratamiento se sostiene hasta que se produzca una supresión deseada de los síntomas de la enfermedad. Sin embargo, pueden ser útiles otros regímenes posológicos. El progreso de esta terapia se puede controlar por medio de técnicas y ensayos estándar. Etapa 4B: Preparación de 2-(1-bencil-1H-benzo[d]imidazol-2-iltio)acetato de etilo 4b
A una solución agitada de 2-(1H-benzo[d]imidazol-2-iltio)acetato de etilo 4a (11,2 g, 47,6 mmol, 1,0 eq.) en 10:1 éter dietílico:dimetilformamida (450 mL) a 0 °C, se añadió hidruro de sodio al 60% en aceite mineral (2,09 g, 52,3 mmol, 5 1,1 eq.) y la reacción se agitó a 0 °C durante 20 minutos. Bromuro de bencilo (9,78 g, 57,1 mmol, 1,2 eq.) se añadió a 0 °C y la reacción se agitó con calentamiento hasta temperatura ambiente durante 16 horas. La mezcla de reacción se neutralizó con metanol (100 mL) y se concentró al vacío. El residuo se extrajo en cloroformo caliente (400 mL) y se filtró. El filtrado se lavó con agua (200 mL), HCl 1 M (200 mL) y salmuera (200 mL). La capa orgánica se secó sobre sulfato de magnesio y se concentró al vacío. La purificación por cromatografía flash (10% de
10 EtOAc:90% éter de petróleo) dio 4b en forma de un sólido de color amarillo pálido (4,82 g, 30%).
Etapa 4C: Preparación de 2-(1-bencil-1H-benzo[d]imidazol-2-iltio)acetohidrazida 4c
A una solución agitada de 2-(1-bencil-1H-benzo[d]imidazol-2-iltio)acetato de etilo 4b (200 mg, 0,61 mmol, 1,0 eq.) en etanol (5 mL), se añadió monohidrato de hidrazina (61 mg, 1,23 mmol, 2,0 eq.) y la mezcla de reacción se calentó
15 hasta reflujo y se agitó a esa temperatura durante 16 horas. La mezcla de reacción se enfrió hasta temperatura ambiente y se concentró al vacío. La mezcla se disolvió en acetato de etilo (20 mL) y se lavó con agua (20 mL) y salmuera (20 mL). La capa orgánica se secó sobre sulfato demagnesio y se concentró al vacío para dar 4c en forma de un sólido de color amarillo pálido que luego se usó sin ulterior purificación (200mg, 100%).
Etapa 4D Preparación de (E)-2-(1-bencil-1H-benzo[d]limidazol-2-iltio)-N’-(4-etoxi-3-metoxibenciliden)acetohidrazida 20 4-1
A una solución agitada de 2-(1-bencil-1H-benzo[d]imidazol-2-iltio)acetohidrazida 4c (200 mg, 0,61 mmol, 1,0 eq.) en etanol (5 mL), se añadió 3-metoxi-4-etoxibenzaldehído (110 mg, 0,61 mmol, 1,0 eq.) y ácido acético (3 gotas). La mezcla se calentó hasta reflujo y se agitó a esa temperaturadurante 16 horas. La mezcla de reacción se enfrió hasta
25 temperatura ambiente y el precipitado que se formó enfriando se filtró y se lavó con agua (10 mL), etanol (10 mL) y éter dietílico (10 mL). El sólido se secó por alto vacío durante la noche para dar 4-1 en forma de un sólido blanco (196mg, 67%).
Ejemplo 5 CLORHIDRATO DE (E)-N’-(3,4-DIMETOXIBENCILIDEN)-2-(2-METILQUINOLIN-4-ILTIO)ACETOHIDRAZIDA Etapa 5A Preparación de 2-metilquinolina-4-tiol 5a
5 Una suspensión agitada de 2-metilquinolin-4-ol (5,10 g, 32,0 mmol, 1,0 eq.) en tetrahidrofurano (51 mL) se calentó hasta 50 °C y se añadieron 6,5 g de reactivo de Lawesson (6,5 g, 16,0mmol, 0,5 eq.) en una porción. La reacción se calentó hasta 80 °C y se agitó a esa temperatura durante 6 horas. La reacción se vertió en una solución bifásica caliente de acetato de etilo (200 mL) y agua (200 mL) y se agitó vigorosamente. Un gel anaranjado viscoso se precipitó de la solución. La solución de agua/acetato de etilo se decantó en un embudo de separación y la capa

Etapa 5B Preparación de 2-(2-metilquinolin-4-iltio)acetato demetilo 5b
2-metilquinolina-4-tiol 5a (1,00 g, 5,71 mmol, 1,00 eq.) se disolvió en dimetilformamida (10 mL) a temperatura ambiente. Se añadió carbonato de potasio (0,87 g, 6,28 mmol, 1,10 eq.) y la mezcla se agitó durante 15 minutos. Se añadió bromoacetato de metilo (921 mg, 6,02 mmol, 1,05 eq.) gota a gota a la solución y la reacción se agitó a temperatura ambiente durante 2,5 horas. Se añadió agua (20 mL) a la mezcla de reacción y la capa acuosa se
20 extrajo con acetato de etilo (30 mL). La capa acuosa luego se extrajo con acetato de etilo (3 x 50 mL) y las capas orgánicas combinadas se secaron sobre sulfato de magnesio, se filtraron y se concentraron al vacío. El residuo resultante se secó a alto vacío para obtener 5b en forma de un sólido marrón (1,25 g, 88%).
