Description
- Gebiet der Erfindung
- Die Erfindung bezieht sich auf kosmetische und pharmazeutische Zusammensetzungen, die ein Retinoid enthalten. Speziell bezieht sich die Erfindung auf wasserhaltige Zusammensetzungen, die stabilisiert sind, um einen Abbau des Retinoids zu verhindern.
- Hintergrund der Erfindung
- In der Vergangenheit sind Kosmetika über das Konzept einer bloßen Verschönerung für das Gesicht hinaus entwickelt worden. Konsumenten fordern nun mehr von ihrem Make-up als bloß Farbe, Abdeckung oder Feuchtigkeitsbildung: Es ist nun bevorzugt, daß Kosmetika einige Vorteile für die Haut bereitstellt, als sie bloß zu verschönern oder sie sanfter zu machen. Dieser Verbrauchervorzug führte zur häufigen Verwendung von biologisch wirksamen Bestandteilen in vielen kosmetischen Produkten. Angesichts der nun stark anerkannten schädigenden Wirkungen der Sonneneinstrahlung auf die Haut sind besonders beliebte wirksame Komponenten jene, die diesen Wirkungen entgegenwirken oder sie verhindern. Diese Komponenten schließen zum Beispiel Sonnenschutzmittel, Antioxidantien und Antifaltenmittel ein.
- Eines der Hauptschwierigkeiten bei der Anwendung von Wirkstoffen in einer Formulierung ist die potentielle Instabilität des Wirkstoffs, sobald er eingebracht ist. Der eigentliche Grund für ihre Verwendung in der Formulierung, d. h. ihre biologische Wirksamkeit, bedeutet, daß sie nicht inert sind, und deshalb potentiell einer Verminderung oder einem vollständigen Verlust der Potenz unterliegen, wenn sie nicht mit einem geeigneten Vehikel kombiniert sind. Eine Reihe von gewöhnlich zusammenkommenden Faktoren können eine biologisch wirksame Verbindung in einer Formulierung ohne weiteres inaktivieren, bevor sie überhaupt den Verbraucher erreicht. Solche Faktoren beinhalten zum Beispiel Sauerstoff, extreme Temperaturen, UV-Licht, Wasser und lipide Oxidaten. Es ist besonders schwierig, die Wirkungen von Sauerstoff und UV-Licht zu verhindern, die natürlich so gut wie überall in der Natur vorkommen. Obgleich der Abbau aufgrund von Wasser technisch durch wasserfreie Formulierungen verhindert werden kann, ist eine wasserhaltige Formel im allgemeinen ein bevorzugtes Abgabesystem, da sie der Haut eine Hydratation verleiht. Somit gibt es weiterhin ein Bedürfnis zur Entwicklung eines kosmetisch akzeptablen Vehikels, welches die hydratisierenden Vorzüge von Wasser liefern kann, aber auch die im Vehikel enthaltenen Wirkstoffe vor abbauenden Wirkungen von Umweltfaktoren einschließlich Wasser, die ihnen ihre biologische Wirksamkeit rauben, schützt. Die vorliegende Erfindung stellt eine Lösung für dieses fortlaufende Problem bereit.
- Zusammenfassung der Erfindung
- Die vorliegende Erfindung bezieht sich auf eine stabile kosmetische oder pharmazeutische Öl-in-Wasser-Emulsionszusammensetzung, umfassend ein biologisch wirksames Retinoid und ein Sauerstoffeinfang-Antioxidanssystem auf Enzymbasis, wobei das Antioxidanssystem wirksame Mengen einer Oxidase, eines Substrats für die Oxidase, einer Peroxidase und mindestens einer Spezies, die durch Wasserstoffperoxid oxidierbar ist, umfaßt.
- Ein besonders nützliches Antioxidanssystem umfaßt Jodid-Anionen und Thiocyanat-Anionen in einem Gewichts : Gewichts-Verhältnis von 0.1 : 1 bis 50 : 1 und mit einer kombinierten Anionen-Gewichtskonzentration von mindestens 5 mg/kg, D-Glucose in einer Gewichtskonzentration von mindestens 0,2 g/kg, und eine wirksame Menge der Glucoseoxidase und einer Lactoperoxidase.
- Detaillierte Beschreibung der Erfindung
- Antioxidantien werden aus einer Reihe von Gründen häufig in Formulierungen verwendet. Eine primäre Funktion von jeglichem Antioxidans ist es natürlich, den oxidativen Abbau der es enthaltenden Formulierung zu verhindern, indem Sauerstoffradikale der Umgebung bzw. Umwelt eingefangen werden. Ein wirksames Antioxidans kann auch zur Verhinderung oder Verminderung des mikrobiellen Wachstums wirken und deshalb den Verderb des Vehikels verhindern. Diese beiden Wirksamkeiten können zur Gesamtstabilität der Formulierung beitragen. Die Aufgabe ist jedoch schwieriger, wenn Wasser in der Formulierung vorliegt, und noch mehr, wenn es ein biologisch wirksames Material in der Formulierung gibt. Die Kombination der hydrolytischen Wirksamkeit von Wasser mit der Gegenwart eines biologisch reaktiven Moleküls, ohne gewisse schützende Maßnahmen, führt unweigerlich zum Abbau und Verlust der Wirksamkeit des biologischen Wirkstoffs. In einem solchen Fall ist ein Antioxidans alleine, welches potentiell in der Lage ist, das Vehikel insgesamt vor Abbau zu schützen, kaum in der Lage, eine adäquate Erhaltung der biologischen Wirksamkeit bereitzustellen. Das Problem ist in einem Öl-in-Wasser-Vehikel – dem in Hautpflegezusammensetzungen am weitest gebräuchlichen Verwendeten – besonders verschärft. In einem solchen System gibt es anscheinend eine beachtliche Diffusion von Sauerstoff bei der Wasser/Öl-Grenzfläche, was potentiell zu einem ernsthaften Abbauproblem selbst für empfindliche Wirkstoffe, die in der Ölphase eingeschlossen sind, führt.
- Es ist nun überraschenderweise gefunden worden, daß eine spezielle Art einer Antioxidanssystems, welches Sauerstoffmoleküle im Medium wirksam einfängt, zur Verhinderung des Verlusts der Wirksamkeit von biologischen Materialien selbst in der schwierigen Umgebung einer Öl-in-Wasser-Emulsion hoch wirksam sein kann. Diese Art von Antioxidans unterscheidet sich von gewöhnlicheren Antioxidantien, zum Beispiel BHT, BHA, Propylgallat, Tocopherol, Ascorbinsäure und dergleichen, die die schädigenden Sauerstoffradikale nicht wirksam aus dem Medium, in dem sie enthalten sind, eliminieren können. Um die Sauerstoffeinfangwirksamkeit zu erzielen, ist das Antioxidanssystem Enzym-basiert. Ein bevorzugtes System ist ein solches, das als seine Grundelemente eine Oxidase, eine Peroxidase und ein Substrat für die Oxidase umfaßt. Diese Komponenten sind Elemente von natürlich vorkommenden, physiologischen Stoffwechselwegen im Körper, und deshalb sorgt das Antioxidansystem für den zusätzlich Vorteil, daß es ein wirklich natürliches Schutzmittel ist.
- Die Oxidase im System ist eine, die die Produktion von H2O2 durch die Oxidation eines Substrats in Gegenwart von Wasser und Sauerstoff katalysiert. Beispiele von nützlichen Oxidasen für diesen Zweck sind Glucoseoxidase oder Galactoseoxidase. Geeignete Substrate für diese Enzyme sind jeweils D-Glucose oder Galactose oder Vorläufer dieser Verbindungen, zum Beispiel Oligomere oder Polymere, die in kleinere Zuckereinheiten zerfallen können. Die Menge der verwendeten Oxidase beträgt vorzugsweise mindestens etwa 25 U/kg der Gesamtzusammensetzung, wobei eine Einheit hier als die Menge an Enzymen definiert ist, die erforderlich ist, die Umwandlung von 1,0 μmol von Substrat pro Minute bei 25°C unter optimalen Bedingungen zu katalysieren. Weiter bevorzugt beträgt die Menge an Oxidase mindestens etwa 75 U/kg und vor allem etwa 150 U/kg. Das Substrat für die Oxidase wird vorzugsweise in einer Menge von mindestens etwa 0,5 g/kg, vorzugsweise mindestens 1 g/kg und weiter bevorzugt mindestens 2 g/kg bereitgestellt.
- Die andere Enzymkomponente des Antioxidanssystems ist eine Peroxidase, d. h. ein Enzym, welches die Oxidation einer leicht oxidierbaren Spezies durch Wasserstoffperoxid im Medium katalysiert. Die Peroxidase kann irgendeine Peroxidase sein, z. B. Lactoperoxidase, Myeloperoxidase oder Meerrettichperoxidase. Die Menge an Peroxidase beträgt vorzugsweise mindestens etwa 10 U/kg. Es ist bevorzugt, daß die leicht oxidierbaren Spezies als Teil des Antioxidanssystems bereitgestellt werden. Beispiele von nützlichen oxidierbaren Spezies sind Jodid-, Thiocyanat- oder Bromid-Anionen. Besonders bevorzugte oxidierbare Spezies sind Jodid- oder Thiocyanat-Anionen oder beide. Über die Bereitstellung eines relativ unschädlichen Reservoirs für potentiell schädigenden Sauerstoff hinaus besitzen die oxidierten Spezies eine starke antimikrobielle Wirksamkeit, die das Wachstum von Bakterien im Vehikel verhindert. Die Anionen werden im allgemeinen in das System in Form von Salzen, wie Kalium- und Natrium-Jodidsalzen oder Mischungen davon, sowie Kalium-, Natrium-, Ammonium-, Eisen- und Kupfer-Salzen von Thiocyanat sowie Mischungen davon eingeschlossen. Eine bevorzugte Gewichtskonzentration der Jodid-Ionen ist mindestens 5 mg/kg, und für das Thiocyanat mindestens 2 mg/kg.
- Die Komponenten des Antioxidans können zu der wasserhaltigen Zusammensetzung einzeln zugegeben werden oder als Teil einer getrennten Antioxidansmischung. Eine besonders nützliche Mischung von Komponenten ist kommerziell unter dem Namen Myavert (Boots Company PLC) verfügbar. Das System ist zum Beispiel im US-Patent Nr. 5,607,681 offenbart. Wie dort beschrieben, liegen Jodid- und Thiocyanat-Anionen in der Zusammensetzung in einem Gewichtsverhältnis in einem Bereich von 0,1 : 1 bis 50 : 1 vor und besitzen eine kombinierte Anionengewichtskonzentration von mindestens 5 mg/kg, vorzugsweise mindestens 10 mg/kg; D-Glucose in einer Gewichtskonstellation von mindestens 0,2 g/kg; Glucoseoxidase in einer Menge von mindestens 150 U/kg, und mindestens 10 U/kg Lactoseperoxidase.
- Die kombinierten Komponenten des Antioxidanssystems sorgen für einen beträchtlichen Grad an Schutz in einem wasserenthaltenden System für solche, leicht durch Sauerstoff abgebaute biologische Wirkstoffe wie Retinoide. Selbst bei Einschluß in die Ölphase einer Zusammensetzung zeigen diese Verbindungen in Gegenwart von Wasser einen schnellen Abbau, was zu einem beträchtlichen Verlust der biologischen Wirksamkeit führt. In Gegenwart der oben beschriebenen Sauerstoffeinfangkomponenten der Erfindung gibt es jedoch eine deutliche Verbesserung bei der Aufrechterhaltung der Wirksamkeit des Retinoids, wodurch die Potenz des Systems demonstriert wird.
- In der Zusammensetzung der Erfindung umfaßt die Zusammensetzung ein Retinoid als dem biologisch wirksamen Mittel. Im vorliegenden Kontext ist durch Retinoid Vitamin A (Retinol) sowie irgendwelche natürliche oder synthetische Analoga von Vitamin A, die qualitativ die gleiche Art der Aktivität wie Vitamin A gegenüber der Haut ausüben, gemeint. Beispiele von Retinoiden zusätzlich zu Retinol beinhalten Vitamin-A-Säure, Vitamin-A-Aldehyd und Vitamin-A-Ester, sind jedoch nicht darauf beschränkt. Die Retinoide besitzen eine große Vielfalt von dermatologischen Anwendungen, einschließlich der allgemeinen Verzögerung der Alterungswirkungen – sowohl der normalen als auch der Fotoalterung. Speziellere Anwendungen beinhalten die Verhinderung und Verminderung von Falten, der Hautathrophie, der Hyperpigmentierung sowie abnormaler epidermischer Wucherungen. Retinoide haben sich auch bei der Behandlung der Akne als nützlich erwiesen. Gemäß der vorliegenden Erfindung werden die Antioxidantien zur Stabilisierung eines Retinoids verwendet.
- In solchen Zusammensetzungen wird das wirksame Retinoidmittel in eine Menge von etwa 0,001–5 Gew.-%, vorzugsweise etwa 0,001–2 Gew.-% der Gesamtzusammensetzung verwendet, wobei die Menge und die Anwendungsführung der Applikation von der in Betracht gezogenen Verwendung des Retinoids abhängen.
- Die Wasser-Zusammensetzungen der Erfindung können in irgendeiner Form vorliegen, insbesondere als Wasser-in-Öl oder Öl-in-Wasser-Emulsionen. Ein besonderer Vorteil wird jedoch erhalten, wenn die Zusammensetzung eine Öl-in-Wasser-Emulsion ist, die wie oben erläutert sogar noch stärkere Schwierigkeiten gegenüber oxidativem Abbau zeigt als eine Wasser-in-Öl-Emulsion. Deshalb ist in einer bevorzugten Ausführungsform die Zusammensetzung eine Öl-in-Wasser-Emulsion, die das wirksame Mittel in der Ölphase enthält.
- Die wässrige Phase in der Zusammensetzung kann irgendein kosmetisch akzeptables, wasserbasiertes Material sein, wie deionisiertes Wasser oder Blütenwasser. Die Ölphase kann irgendein kosmetisch oder pharmazeutisch akzeptables Öl sein, wie ein Öl, welches für den vorliegenden Zweck als irgendein pharmazeutisch oder kosmetisch akzeptables Material definiert ist, welches in Wasser im wesentlichen unlöslich ist. Die Öle können flüchtig oder nicht-flüchtig sein oder eine Mischung von beidem. Zum Beispiel beinhalten geeignete flüchtige Öle sowohl cyclische als auch lineare Silikone wie Cyclomethicon, Octamethylcyclotetrasiloxan und Decamethylcyclopentasiloxan; oder gradkettige oder verzweigtkettige Kohlenwasserstoffe mit 8–20 Kohlenstoffatomen wie Dekan, Dodekan, Tridekan, Tetradekan und C8-20-Isoparaffinen sind jedoch nicht darauf beschränkt.
- Nicht-flüchtige Öle beinhalten Pflanzenöle wie Kokosöl, Jojobaöl, Getreideöl, Sonnenblumenöl, Palmöl, Sojabohnenöl; Carboxylsäureester wie Isostearyl-Neopentanoat, Cetyloctanoat, Cetylrecinoleat, Octylpalmitat, Dioctylmalat, Coco-Dicaprylat/-caprat, Decylisostearat, Myristylmyristat; tierische Öle wie Lanolin und Lanolin-Derivate, Talgöl, Nerzöl oder Cholesterin; Glycerylester wie Glycerylstearat, Glyceryldioleat, Glyceryldistearat, Glyceryllinoleat, Glycerylmyristat; nicht-flüchtige Silikone wie Dimethikom, Dimethiconol, Dime thicon-Copolyol, Phenyl-Trimethicon, Methicon, Simethicon; und nicht-flüchtige Kohlenwasserstoffe wie Isoparaffine, Squalen oder Petrolatum, sind jedoch nicht darauf beschränkt.
- Die Emulsionen können andere optionale Komponenten je nach beabsichtigter Endverbindung umfassen. Diese beinhalten Wasserlösliche Färbemittel (wie FD&C Blue#1); Öl-lösliche Farbstoffe (wie D&C Green#6), Wasser-lösliche Sonnenschutzstoffe (wie Eusolex 232); Öl-lösliche Sonnenschutzstoffe (wie Octyl-Methoxicinnamat); Teilchen-Sonnenschutzstoffe (wie Zinkoxid); Antioxidantien (wie BHT); Chelatisiermittel (wie Dinatrium-EDTA); Emulsionsstabilisatoren (wie Carbomer); Konservierungsstoffe (wie Methylparaben); Duftstoffe (wie Pinen); Geschmacksstoffe (wie Sorbitol); Befeuchter (wie Glycerin); Wasserschutzmittel (wie PVP/Eicosen-Copolymer); Wasser-lösliche Filmbildner (wie Hydroxypropyl-Methylcellulose); Öl-lösliche Filmbildner (wie hydrogeniertes C-9-Harz); kationische Polymere (wie Polyquaternium 10); anionische Polymere (wie Xantangummi); Vitamine (wie Tocopherol), sind jedoch nicht darauf beschränkt.
- Die Erfindung wird durch die folgenden Beispiele näher veranschaulicht.

- Eine Zusammensetzung gemäß der vorliegenden Erfindung wird wie folgt hergestellt:
Material Gewichts-% Phase I Deionisiertes Wasser 51,00 Weizenprotein-Stearat (Bioetica) 0,70 Dinatrium-EDTA 0,10 Sucrose 2,00 Coffein-Pulver 0,20 Phase II BHT 0,10 Isostearyl-Neopentanoat 3,00 Squalan 3,00 Cetearyl-Glucosid 5,00 Shea-Butter 5,90 Phase III Cyclomethicon 7,00 Phase IV Carbomer 0,50 Deionisiertes Wasser 9,50 Phase V Glycerol 2,00 Grüntee-Extrakt 1,00 1,3-Butylenglykol 1,50 Deionisiertes Wasser 1,50 Phase VI Triethanolamin 0,05 Deionisiertes Wasser 1,95 Phase VII Magnesiumascobylphosphat 0,35 Deionisiertes Wasser 2,04 Phase VIII Vitamin E 0,20 Phase IX MyavertTM-Substrat (Boots) 0,72 MyavertTM-Enzym (Boots) 0,04 Phase X Grüntee-Ölextrat (LipoChemical) 0,15 Retinol (10%-Lösung in Soyabohnenöl) 0,50 - Zum Herstellen der Zusammensetzung werden Phase I-Komponten im Hauptbehälter kombiniert und auf 80°C erwärmt. Die Phase II-Bestandteile werden in einem Hilfsbehälter kombiniert und auf 82°C erwärmt. Die Phase I- und Phase II-Bestandteile werden dann unter Homomischung und Seitenschlagbewegung kombiniert. Phase III-Bestandteile werden dem Hauptbehälter bei 65°C zugefügt. Phase V- und Phase IV-Bestandteile werden nacheinander dem Hauptbehälter bei 50°C zugesetzt, und jeweils bis zur Gleichförmigkeit vermischt. Phase VI-Bestandteile werden zum Hauptbehälter bei 40°C zugegeben und bis zur Gleichförmigkeit vermischt. Phase VII-Bestandteile werden bei 38°C zugegeben und bis zur Gleichförmigkeit vermischt. Phase VIII-Bestandteile werden bei 35°C zugegeben und bis zur Gleichförmigkeit vermischt, und dann werden die Phasen IX und X bei 30°C zugegeben und bis zur Gleichförmigkeit vermischt.
- Beispiel II
- Eine Retinol-Creme, die Retinol und MyavertTM wie im Beispiel I beschrieben, enthält, wird in Bezug auf Stabilität im Vergleich zu einer identischen Formulierung ohne MyavertTM getestet. Die Cremes werden bezüglich der Aufrechterhaltung der Retinolwirksamkeit über eine Dauer von 8 Wochen bei Temperaturen von 4°C, 25°C, 37°C und 45°C getestet. Nach Abschluß der 8 Wochen zeigte die Kontrollcreme ohne dem Antioxidanssystem eine im wesentlichen volle Aufrechterhaltung der Wirksamkeit bei 4°C und 25°C, etwa 86% Aufrechterhaltung der Aktivität bei 37°C und nur 74% Aktivität bei 45°C. Im Gegensatz dazu hielt die Antioxidanssystem-enthaltende Creme eine 100%-Wirksamkeit bei 4°C und 25°C und eine 94%-Wirksamkeit bei 37°C und 45°C, wodurch eine höhere Stabilität des Wirkstoffs gezeigt wird, wenn in Gegenwart der Glucoseoxidase und Lactoperoxidase formuliert wurde.
Claims (10)
- Stabile kosmetische oder pharmazeutische Öl-in-Wasser-Emulsionszusammensetzung, umfassend ein biologisch wirksames Retinoid und ein Sauerstoffeinfang-Antioxidanssystem auf Enzymbasis, wobei das Antioxidanssystem wirksame Mengen einer Oxidase, eines Substrats für die Oxidase, einer Peroxidase und mindestens einer Spezies, die durch Wasserstoffperoxid oxidierbar ist, umfaßt.
- Zusammensetzung von Anspruch 1, wobei das Antioxidanssy- stem eine Glucoseoxidase, Glucose und eine Lactoperoxidase umfaßt.
- Zusammensetzung von Anspruch 1 oder 2, wobei die oxidierbare Spezies ausgewählt ist aus der Gruppe, die aus Jodid-Ionen, Thiocyanat-Ionen sowie einer Kombination davon besteht.
- Zusammensetzung von Anspruch 3, die sowohl Jodid als auch Thiocyanat-Ionen umfaßt.
- Zusammensetzung irgendeines der Ansprüche 1 bis 4, wobei das Retinoid Retinol ist.
- Zusammensetzung von Anspruch 1, wobei das Antioxidanssystem Glucoseoxidase, Glucose, Lactoperoxidase und Jodid und/oder Thiocyanat-Anionen umfaßt.
- Zusammensetzung von Anspruch 1, wobei das Retinoid Retinol ist und das Antioxidanssystem Glucoseoxidase, Glucose, Lactoperoxidase und Jodid- und/oder Thiocyanat-Anionen umfaßt.
- Zusammensetzung irgendeines der Ansprüche 1 bis 7, die Jodid-Ionen und Thiocyanat-Ionen in einem Gewichts : Gewichts-Verhältnis von 0,1 : 1 bis 50 : 1 und einer kombinierten Anionen-Gewichtskonzentration von mindestens etwa 5 mg/kg, D-Glucose in einer Gewichtskonzentration von mindestens 0,2 g/kg und eine wirksame Menge von Glucoseoxidase und einer Lactoperoxidase umfaßt.
- Zusammensetzung irgendeines der Ansprüche 2, 6, 7 oder 8, oder von 3 bis 5 bei Abhängigkeit von Anspruch 2, wobei die Menge der Glucoseoxidase mindestens etwa 25 U/kg beträgt und die Menge der Lactoperoxidase mindestens etwa 10 U/kg beträgt.
- Zusammensetzung von Anspruch 9, bei der die Menge der Glucoseoxidase mindestens etwa 150 U/kg beträgt.
