Description
- Die vorliegende Erfindung betrifft neue Pyridinderivate, welche als Medikamente zur Behandlung von Osteoporose verwendbar sind und Arzneimittel, die diese Derivate enthalten.
- Substituierte Triphenylmethane, die einen Carbamoylsubstituenten aufweisen, sind als zur Behandlung von Osteoporose brauchbar bekannt; US-A-5,112,867, nicht aber carbamoylsubstituierte Pyridinverbindungen.
- In Übereinstimmung mit der vorliegenden Erfindung werden neue Pyridinderivate zur Behandlung von Osteoporose bereitgestellt. Dies sind Verbindungen der Formel (I) (im folgenden benannt als Verbindung(en) (I); eine ähnliche Bezeichnung wird im Falle der anderen Formeln benutzt):

- R¹ und R² unabhängig voneinander Wasserstoffatome oder C&sub1;-C&sub8;-Alkylreste darstellen; R³ und R&sup4; einzeln für sich betrachtet jeweils ein Wasserstoffatom, einen Niederalkylrest, monocyclischen Alkylrest, polycyclischen Alkylrest der Formel
- (wobei Q ein Wasserstoffatom oder einen C&sub1;-C&sub8;-Alkylrest darstellt, m 0 oder 1 und n 0 oder eine ganze Zahl zwischen 1 und 5 ist), einen Phenyl-, Naphthyl- oder substituierten Phenyl- oder Naphthylrest mit 1 bis 3 Substituenten, unabhängig ausgewählt aus C&sub1;-C&sub8;-Alkyl-, Hydroxy-, C&sub1;-C&sub8;-Alkoxy-, C&sub1;-C&sub8;-Alkylthio, Halogen-, Nitro-, Amino-, C&sub1;-C&sub5;-Alkanoyl-, Benzoyl-, Naphthoyl-, Carboxyl-, (C&sub1;-C&sub8;-Alkoxy)carbonyl- und Trifluormethylgruppen, oder einen heterocyclischen Rest bedeuten, ausgewählt aus Pyridyl-, Chinolyl-, Thiazolyl-, Pyrrolidinyl-, Piperidyl-, Piperidino-, Piperazinyl-, Morpholino- und Thiomorpholinogruppen, wobei die heterocyclischen Reste gegebenenfalls 1 oder 2 Substituenten aufweisen, unabhängig voneinander ausgewählt aus C&sub1;-C&sub8;-Alkyl-, C&sub1;-C&sub8;-Alkoxy-, Halogen-, Phenyl-, Benzyl-, Pyridyl-, Pyrimidyl- und substituierten Phenylresten, wobei der substituierte Phenylrest wie vorstehend definiert ist;
- oder in der R³ und R&sup4; zusammen mit dem Stickstoffatom, an das sie gebunden sind, eine Pyrrolidinyl-, Piperidyl-, Piperidino-, Piperazinyl-, Morpholino- oder eine Thiomorpholinogruppe, oder eine solche Gruppe, die 1 oder 2 Substituenten aufweist, unabhängig ausgewählt aus C&sub1;-C&sub8;-Alkyl-, C&sub1;-C&sub8;-Alkoxy-, Halogen-, Phenyl-, Benzyl-, Pyridyl-, Pyrimidyl- und substituierten Phenylresten, bedeutet, wobei der substituierte Phenylrest wie vorstehend definiert ist;

- Bei der Definition der einzelnen Gruppen der Formel (I) bedeutet der Niederalkylrest einen unverzweigten oder verzweigten Alkylrest mit 1 bis 8 Kohlenstoffatomen, z.B. eine Methyl-, Ethyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, sec-Butyl-, tert-Butyl-, Pentyl-, Neopentyl-, Hexyl-, Heptyl-, Octylgruppe usw. Der monocyclische Alkylrest bedeutet einen Cycloalkylrest mit 3 bis 8 Kohlenstoffatomen, z.B. eine Cyclopropyl-, Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclooctylgruppe usw.
- Sind R³ oder R&sup4; substituierte Phenyl- oder substituierte Naphthylreste, dann sind typische Alkylsubstituenten diejenigen, die vorstehend als Niederalkylreste aufgelistet sind, und dies trifft ebenso auf Substituenten wie Niederalkoxy-, Niederalkylthio- und Niederalkoxycarbonylreste zu. Typische Alkanoylsubstituenten schließen z.B. Formyl-, Acetyl-, Propionyl-, Butyryl-, Isobutyryl-, Valeryl-, Pivaloylreste usw. ein. Halogen steht für Fluor, Chlor, Brom oder Iod.
- Verwendbare pharmazeutisch verträgliche Salze der Verbindung (I) beinhalten Säureadditionssalze, wie Hydrochlorid, Hydrobromid, Hydroiodid, Nitrat, Phosphat, Formiat, Acetat, Benzoat, Maleat, Fumarat, Succinat, Tartrat, Citrat, Oxalat, Glyoxylat, Aspartat, Methansulfonat, Ethansulfonat und Benzolsulfonat. Die Verbindung (I) kann stufenweise gemäß dem folgenden Reaktionsschema hergestellt werden.
- wobei R1a ud R2a durch R¹ und R² benannte Reste, nicht aber Wasserstoff sind; Z ein Chlorid- oder Bromidatom ist; und R¹, R², R³ und R&sup4; haben die gleiche Bedeutung wie vorstehend erwähnt haben.
- Man erhält Verbindung (II) durch Reaktion von Isonicotinsäure mit einem Halogenierungsmittel, wie Thionylchlorid, Phosphorpentachlorid, Phosphortrichlorid oder Phosphortribromid, wobei ein Säurehalogenid erhalten wird, und Kondensieren des Säurehalogenids mit Diisopropylamin. Verbindung (IV) erhält man durch Reaktion von Verbindung (II) mit 0,5 bis 5 Äquivalenten einer metallorganischen Verbindung wie n-Butyllithium oder Lithiumdiisopropylamid in einem Zeitraum von 30 Minuten bis 5 Stunden in einem inerten Lösungsmittel, usw. z.B. einem Ether wie Diethylether, Tetrahydrofuran bei einer Temperatur von -78ºC bis 0ºC, und anschließende Umsetzung des Reaktionsprodukts mit Verbindung (III) bei einer Temperatur von -78ºC bis Raumtemperatur in einem Zeitraum von 30 Minuten bis 5 Stunden. Die Reaktion wird vorzugsweise unter Ausschluß von Feuchtigkeit unter Schutzgas, wie Stickstoff, Argon oder Helium durchgeführt.
- Man erhält Verbindung (V) aus Verbindung (IV) durch katalytische Reduktion mit Palladium-Kohle oder ähnlichen Katalysatoren.
- Die Reaktion ist üblicherweise in einem Zeitraum von 10 Minuten bis 48 Stunden in z.B. einer organischen Säure, wie Essigsäure, Ameisensäure, usw. bei einer Temperatur von Raumtemperatur bis zum Siedepunkt des Lösungsmittels abgeschlossen.
- Wenn nötig, wird die Reaktion in einem Lösungsmittelgemisch aus einer organischen Säure und einem Lösungsmittel, wie einem aromatischen Kohlenwasserstoff wie Benzol, Toluol, Xylol usw., einem Alkohol, wie Methanol, Ethanol, Isopropanol usw., einem Ether, wie Diethylether, Dioxan, Tetrahydrofuran, einem Amid, wie Formamid, Dimethylformamid, Acetonitril, Dimethylsulfoxid, Wasser oder dgl. durchgeführt.
- Man erhält Verbindung (VI) durch Umsetzung von Verbindung (V) mit einem Halogenierungsmittel, wobei ein Säurehalogenid erhalten wird. Als Halogenierungsmittel verwendet man Thionylchlorid, Phosphorpentachlorid, Phosphortrichlorid, Phosphortribromid usw. Das Halogenierungsmittel wird dabei in einer Menge von 1 bis 20 Äquivalenten bezüglich der Verbindung (V) oder in einem großen Überschuß für den Fall, daß es auch als Lösungsmittel dient, verwendet. Jegliches Lösungsmittel kann verwendet werden, solange es nicht an der Reaktion beteiligt ist, und halogenierte Kohlenwasserstoffe, wie Dichlormethan, Chloroform, Dichlorethan, Tetrachlorkohlenstoff usw. können verwendet werden. Die Reaktion ist üblicherweise in einem Zeitraum von 10 Minuten bis 24 Stunden bei einer Temperatur von 0ºC bis zum Siedepunkt des Lösungsmittels beendet.
- Verbindung (Ia), welche Verbindung (I) entspricht, in der R¹ und R² Niederalkylreste sind, wird durch Reaktion von Verbindung (VI) mit Verbindung (VII) erhalten. Verbindung (VII) wird dabei in einer Menge von 0,1 bis 5 Äquivalenten, vorzugsweise 0,5 bis 2 Äquivalenten bezüglich Verbindung (VI) eingesetzt. Irgendein Lösungsmittel wird allein oder als Gemisch eingesetzt, solang es nicht an der Reaktion beteiligt ist. Das Lösungsmittel schließt z.B. einen halogenierten Kohlenwasserstoff, wie Dichlormethan, Chloroform, Dichlorethan und Tetrachlorkohlenstoff, einen aromatischen Kohlenwasserstoff wie Benzol, Toluol und Xylol, ein Keton wie Aceton und Methylethylketon, einen Alkohol wie Methanol, Ethanol und Isopropanol, einen Ether wie Diethylether, Dioxan und Tetrahydrofuran, ein Amid wie Formamid und Dimethylformamid, Acetonitril, Ethylacetat, Dimethylsulfoxid, Pyridin, Wasser oder dgl. ein. In manchen Fällen ist es zur Beschleunigung der Reaktion bevorzugt, diese in Gegenwart einer anorganischen Base, wie Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat, Caesiumcarbonat, Natriumbicarbonat oder Silberoxid, durchgeführt oder einer organischen Base, wie Triethylamin, N,N-Diisopropylethylamin, N-Methylmorpholin, Pyridin oder Dimethylaminopyridin, durchzuführen. Die Reaktion ist normalerweise in einem Zeitraum von 10 Minuten bis 24 Stunden bei einer Temperatur von 0ºC bis zum Siedepunkt des Lösungsmittels beendet.
- Will man Verbindung (Ib) erhalten, welche der Verbindung (I) entspricht, in der R¹ und R² Wasserstoffatome sind, wird Verbindung (Ia) in Gegenwart z.B. einer Lewissäure, wie Bortribromid dealkyliert. Ein brauchbares Lösungsmittel hierfür schließt halogenierte Kohlenwasserstoffe wie Dichlormethan, Chloroform, Dichlorethan, Tetrachlorkohlenstoff oder dgl. ein. Die Reaktion ist üblicherweise in einem Zeitraum von 30 Minuten bis 5 Stunden bei einer Temperatur von -78ºC bis Raumtemperatur beendet.
- Die Zwischenprodukt eund Endprodukte, die nach den vorstehenden Verfahren hergestellt wurden, können isoliert und durch irgendwelche in der organischen Synthesechemie üblichen Methoden gereinigt werden, wie durch Filtration, Extraktion, Waschvorgänge, Trocknen, Einengen, Umkristallisieren, verschiedene Chromatographiemethoden usw. Es ist möglich, die Zwischenprodukte in einem späteren Reaktionsschritt einzusetzen, ohne diese auf irgendeine Art gereinigt zu haben.
- Soll ein Salz der Verbindung (I) erhalten werden, kann dieses als solches, wenn es in Salzform vorliegt, gereinigt werden, wohingegen, wenn die Verbindung in freier Form vorliegt, diese in einem geeigneten Lösungsmittel aufgelöst oder suspendiert wird und man anschließend eine geeignete Säure zur Salzbildung zusetzt und man das Salz in mehreren Stufen isoliert.
- Verbindung (I) und pharmazeutisch verträgliche Salze davon können auch als Addukte mit Wasser oder unterschiedlichen Lösungsmitteln vorliegen. Die vorliegende Erfindung schließt diese Addukte mit ein.

- Die Wirkung der Verbindungen der Erfindung auf die Knochenresorptionshemmung wird durch das folgende Experiment verdeutlicht.
- Die Schädeldecke von 5- bis 6-Tage alten dd-Mäusen wurden unter aseptischen Bedingungen entfernt, mit Dulbecco's modifizierter phosphatgepufferter Salzlösung, die kein Calcium und Magnesium enthielt (Produkt von Gibco Oriental Co.) gewaschen und entlang der Sagitalnaht in eine linke und rechte Hälfte zerschnitten. Jede Hälfte der Schädeldecke wurde in 1,5 ml einer Nährlösung kultiviert, die aus Dulbecco's modifiziertes Eagle-Medium (hochangereicherte Glucose) (Produkt von Gibco Oriental Co.) bestand, ergänzt mit 15 % hitzeinkaktiviertes (56ºC für 20 Minuten) Pferdeserum, 2,5 % hitzeinaktiviertes fetales Rinderserum und 1 % einer antibiotisch-antimykotischen Flüssigkeit. Die Testverbindung wurde in Dimethylsulfoxid gelöst und eine 10 µl- Probe dieser Lösung der Kultur bis zu Endkonzentrationen von 3 x 10&supmin;&sup6; M, 1 x 10&supmin;&sup5; M und 3 x 10&supmin;&sup5; M zugesetzt. PTH (Parathyroidhormon) wurde in 0,15 M Kochsalzlösung gelöst (pH 3) und eine 3 µl-Probe dieser Lösung der Kultur bis zu einer Endkonzentration von 1 x 10&supmin;&sup8; M zugesetzt. Die Kulturen wurden bei 37ºC in einer wassergesättigten Atmosphäre, die 5 % CO&sub2; und 95 % Luft enthielt, bebrütet. Üblicherweise wurden die Kulturen für 96 Stunden hierin belassen und das Medium nach 48 Stunden ausgetauscht. Die Testverbindung und PTH, welches nach der gleichen Methode, die vorstehend beschrieben wurde, zubereitet wurden, wurden dazu gegeben. Um die Wirkung der Testsubstanz auf die PTH-induzierte Calciumfreisetzung aus Knochen (d.h. Knochenresorption) zu untersuchen, wurden 3 Gruppen gebildet: eine Kontrollgruppe, eine mit PTH behandelte Gruppe (1 x 10&supmin;&sup8; M) und eine mit der Testverbindung und PTH behandelte Gruppe (3 x 10&supmin;&sup6; M, 1 x 10&supmin;&sup5; M, 3 x 10&supmin;&sup5; M). Die Knochenresorption wurde bestimmt, indem man die Accumulation von Calcium in der Nährlösung, die während 96 Stunden der Kultivierung gesammelt wurde, mißt. Die gesamten Calciumkonzentrationen wurden mit einem Calcium C-Test Wako (Produkt von Wako) bestimmt. Die Inhibierungsrate wurde daraus gemäß der unten angegebenen Formel berechnet und die Konzentration bestimmt, bei der 50 % Inhibierung auftreten (IC&sub5;&sub0;). Die Ergebnisse sind in Tabelle 2 wiedergegeben.

- Eine Testverbindung wurde drei männlichen Mäusen vom dd-Stamm mit einem Körpergewicht von 20 ± 1 g oral verabreicht. Die minimale tödliche Dosis (MLD) wurde anhand der Sterblichkeit binnen 7 Tagen nach der Verabreichung bestimmt.

- Verbindung (I) und pharmazeutisch verträgliche Salze davon können in jegliche gebräuchliche Darreichungsform, wie Tabletten, Kapseln, Sirup, Injektionen, Tropfen, Suppositorien usw. überführt werden und entweder oral oder parenteral einschließlich z.B. durch intramuskuläre Injektion, intravenöse Injektion, Tropfen und intrarektale Gabe von Suppositorien verabreicht werden. Derartige Darreichungsformen für orale oder parenterale Anwendungen werden nach gängigen Methoden hergestellt und können z.B. unterschiedliche Experimente, Gleitmittel, Bindemittel, Zerfallsbeschleuniger, isotonische Zusätze, Emulgatoren usw. enthalten.
- Als für derartige Darreichungsformen zu verwendende Träger können Wasser, destilliertes Wasser zur Injektion, physiologische Kochsalzlösung, Glucose, Fructose, Saccharose, Mannit, Lactose, Stärke, Cellulose, Methylcellulose, Carboxymethylcellulose, Hydroxypropylcellulose, Alginsäure, Talg, Natriumcitrat, Calciumcarbonat, Calciumhydrogenphosphat, Magnesiumstearat, Harnstoff, Silikonharze, Sorbitan- Fettsäureester, Glycerin-Fettsäureester usw. erwähnt werden.
- Die effektive Dosis und die Anzahl der Verabreichungen der Verbindung (I) oder pharmazeutisch verträgliche Salze davon, können in Abhängigkeit von der Art der Anwendungsform, nach Alter und Körpergewicht, nach Zustand, usw. des Patienten variieren, aber es wird im allgemeinen bevorzugt, die Verbindung (I) in einer Dosis von 0,1 bis 10 mg/kg, unterteilt in ein bis 4 Gaben zu verabreichen.
- Anhand der folgenden Beispiele und Vergleichsbeispiele werden Ausführungsformen der vorliegenden Erfindung verdeutlicht.
- 10 g der Verbindung b, erhalten in Bezugsbeispiel 2, wurden in 25 ml Methylenchlorid und 25 ml Thionylchlorid gelöst und die Lösung 30 Minuten unter Rückfluß erhitzt. Die Reaktionslösung wird eingeengt und man erhält 20 g 3-Bis(4- methoxyphenyl)methyl-4-pyridincarbonylchlorid-hydrochlorid (Verbindung c) als Rohprodukt.
- Zu 10 ml einer 1,2-Dichlorethanlösung, die 2 g 3-Aminotricyclo [3.3.1.03,7]nonan-hydrochlorid und 10 ml Triethylamin enthält, wurden bei Raumtemperatur 4,64 g Verbindung c zugegeben. Dann wurde das Gemisch 7 Stunden gerührt; anschließend wird Wasser zugesetzt, um die organische Phase zu trennen. Die wäßrige Phase wurde mit Chloroform extrahiert, dann mit der organischen Schicht vereinigt, mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Nach dem Einengen im Vakuum erhielt man 2,5 g der gewünschten Verbindung als Feststoff.

- 2,5 g der Verbindung 1, die nach Beispiel 1 erhalten wurde, wurden in 40 ml Methylenchlorid gelöst und die Lösung auf -5 bis -15ºC abgekühlt. Dann wurden man 20 ml einer Hexanlösung, die 1 M Bortribromid enthielt, zugegeben. Die Lösung wurde 2 Stunden und 30 Minuten bei Raumtemperatur gerührt. Die Reaktionslösung wurde in eine gesättigte wäßrige Natriumbicarbonatlösung gegossen und anschließend mit Ethylacetat extrahiert. Der Extrakt wurde über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde im Vakuum abgedampft und der Rückstand säulenchromatographisch an Kieselgel gereinigt. Man erhielt 0,63 g der gewünschten Verbindung als Feststoff.

- 81,5 ml Diisopropylamin wurden in 480 ml trockenem Tetrahydrofuran unter Stickstoffatmosphäre gelöst und die Lösung auf -70ºC abgekühlt. Zur Herstellung von Lithiumdiisopropylamid wurden 375 ml einer 1,6 N n-Butyllithiumlösung zugetropft, wobei die Temperatur der Lösung nicht über -65ºC ansteigen soll. Die Reaktionslösung wurde 1 Stunde bei -70ºC gehalten und hierzu eine Lösung von 43,97 g N,N-Diisopropylisonicotinamid, gelöst in 150 ml trockenem Tetrahydrofuran zugegeben, so daß die Temperatur der Mischung nicht über -65ºC anstieg. Das Gemisch wurde 1 Stunde und 15 Minuten gerührt und zum Reaktionsgemisch eine Lösung von 46,94 g 4,4'- Dimethoxybenzophenon, gelöst in 945 ml trockenem Tetrahydrofuran zugetropft, so daß die Temperatur der Lösung nicht über -65ºC anstieg. Anschließend wurde 16 Minuten gerührt. Danach wurde auf Raumtemperatur aufgetaut und Wasser zugefügt. Nach der Extraktion mit Ethylacetat wurde über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der erhaltene Rückstand säulenchromatographisch an Kieselgel gereinigt, wobei 63,69 g der gewünschten Verbindung als Feststoff erhalten wurden.
- NMR (CDCl&sub3;) δ (ppm): 8,53 (1H, d), 8,14 (1H, s), 7,28 - 7,05 (5H, m), 6,80 (4H, d), 6,14 (1H, d), 3,79 (6H, s), 3,59 (1H, m), 3,35 (1H, m), 1,43 (3H, d), 1,17 (3H, d), 1,16 (3H, d), 0,85 (3H, d)
- 103,38 g der in Bezugsbeispiel 1 erhaltenen Verbindung a wurden in 1000 ml Essigsäure gelöst und zu der Lösung wurden 15 g 10%ige Palladium-Kohle suspendiert, in 200 ml Wasser zugegeben. Das Gemisch wurde unter Wasserstoffatmosphäre bei 70ºC 8 Stunden und 15 Minuten gerührt. Das Reaktionsgemisch wurde mit einer Filtrierhilfe gefiltert, eingeengt und aus Wasser umkristallisiert, wobei 45,19 g der gewünschten Verbindung erhalten wurden.

Claims (9)
1. Verbindung der Formel
in der
R¹ und R² unabhängig voneinander Wasserstoffatome oder
C&sub1;-C&sub8;-Alkylreste darstellen; R³ und R&sup4; einzeln für sich
betrachtet jeweils ein Wasserstoffatom, einen
C&sub1;-C&sub8;-Alkylrest, monocyclischen Alkylrest mit 3-8
Kohlenstoffatomen, polycyclischen Alkylrest der Formel
(wobei Q ein Wasserstoffatom oder einen C&sub1;-C&sub8;-Alkylrest
darstellt, m 0 oder 1 und n 0 oder eine ganze Zahl
zwischen 1 und 5 ist), einen Phenyl-, Naphthyl- oder
substituierten Phenyl- oder Naphthylrest mit 1 bis 3
Substituenten, unabhängig ausgewählt aus C&sub1;-C&sub8;-Alkyl-,
Hydroxy-, C&sub1;-C&sub8;-Alkoxy-, C&sub1;-C&sub8;-Alkylthio- Halogen-,
Nitro-, Amino-, C&sub1;-C&sub5;-Alkanoyl-, Benzoyl-, Naphthoyl-,
Carboxyl-, (C&sub1;-C&sub8;-Alkoxy)carbonyl- und
Trifluormethylgruppen, oder einen heterocyclischen Rest bedeuten,
ausgewählt aus Pyridyl-, Chinolyl-, Thiazolyl-,
Pyrrolidinyl-,
Piperidyl-, Piperidino-, Piperazinyl-,
Morpholino- und Thiomorpholinogruppen, wobei die heterocyclischen
Reste gegebenenfalls 1 oder 2 voneinander unabhängige
Substituenten aufweisen, unabhängig voneinander
ausgewählt aus C&sub1;-C&sub8;-Alkyl-, C&sub1;-C&sub8;-Alkoxy-, Halogen-,
Phenyl-, Benzyl-, Pyridyl-, Pyrimidyl- und substituierten
Phenylresten, wobei der substituierte Phenylrest wie
vorstehend definiert ist;
oder in der R³ und R&sup4; zusammen mit dem Stickstoffatom,
an das sie- gebunden sind, eine Pyrrolidinyl-,
Piperidyl-, Piperidino-, Piperazinyl-, Morpholino- oder eine
Thiomorpholinogruppe, oder eine solche Gruppe, die 1
oder 2 Substituenten aufweist, unabhängig ausgewählt aus
C&sub1;-C&sub8;-Alkyl-, C&sub1;-C&sub8;-Alkoxy-, Halogen-, Phenyl-, Benzyl-,
Pyridyl-, Pyrimidyl- und substituierten Phenylresten,
bedeutet, wobei der substituierte Phenylrest wie
vorstehend definiert ist;
und deren pharmazeutisch verträgliche Salze.
2. Verbindung gemäß Anspruch 1, in der R¹ und R² jeweils
ein Wasserstoffatom oder eine Methylgruppe bedeuten.
3. Verbindung gemäß Anspruch 1, in der R¹ und R² jeweils
ein Wasserstoffatom bedeuten und R³ und R&sup4; zusammen eine
substituierte oder unsubstituierte Piperazinylgruppe
bilden.
4. Verbindung gemäß Anspruch 3, in der die Anzahl der
Substituenten in der Piperazinylgruppe 1 oder 2 ist.
5. Verbindung gemäß Anspruch 4, in der die Substituenten
(eine) Benzyl-, substituierte oder unsubstituierte
Phenyl-, Pyridyl- oder Pyrimidylgruppe(n) sind.
6. Verbindung gemäß Anspruch 4, in der die Substituenten
(eine) substituierte Phenylgruppe(n) sind, wobei die
Substituenten in der substituierten Phenylgruppe C&sub1;-C&sub8;-
Alkyl-, Hydroxy-, Alkoxy-, Halogen- oder
Trifluormethylgruppen sind.
7. Verbindung gemäß Anspruch 1, nämlich die Verbindung: 1-
(2-Chlorphenyl)-4-{[3-bis(4-hydroxyphenyl)methyl]-isonicotinyl}piperazin und deren pharmazeutisch verträgliche
Salze.
8. Arzneimittel umfassend eine Verbindung oder ein Salz,
nach einem der Ansprüche 1 bis 7, im Gemisch mit einem
pharmazeutisch verträglichen Träger oder
Verdünnungsmittel.
9. Verwendung einer Verbindung oder Salzes nach einem der
Ansprüche 1 bis 7 zur Herstellung eines Medikaments zur
Behandlung von Osteoporose.
