Description
- Diese Erfindung betrifft Technetiumthioetherkomplexe. Der Thioether ist entweder ein Kronen-Thioether, der drei bis sechs Donor-Schwefelatome aufweist, die durch C2- oder C3- Gruppen in einen makrozyklischen Ring eingebunden sind oder ein tripodaler Ligand, der mehr als drei Donor-Schwefelatome aufweist, die durch C2- oder C3-Gruppen verbunden sind.
- Technetium-99m ist ein γ-Emitter mit einer Halbwertszeit von sechs Stunden, der sehr verbreitet zur Körper-Abbildung verwendet wird. Es wird erwartet, daß erfindungsgemäße Komplexe, in denen Technetium Tc99m ist, interessante biologische Verteilungseigenschaften aufweisen und nützliche Körper-Abbildungsagenzien darstellen.
- Tripodale Liganden sind üblicherweise 1,1,1-substituierte Ethane, worin jeder der drei Substituenten wenigstens ein Schwefelatom enthält. Die drei substituierenden Gruppen können verschieden sein, sind aber im allgemeinen gleich und enthalten zwischen sich mehr als drei Schwefelatome.
- Beispiele für Kronen-Thioether und tripodale Liganden werden in Abbildung 1 gezeigt.
- In den Kronen-Thioetherbezeichnungen stellt die erste Ziffer die Anzahl der Atome und die letzte Ziffer die Anzahl der Schwefelatome in dem makrozyklischen Ring dar. Somit: -
- 9S3 = 1,4,7-Trithiacyclononan;
- 12S3 = 1,5,9-Trithiacyclododecan;
- 14S4 = 1,4,8,11-Tetrathiacyclotetradecan;

- Der Komplex kann in Verbindung mit jedem Tc-Atom umfassen: ein oder zwei Thioetherliganden; 0, 1, 2 oder 3 Oxogruppen und 0, 1, 2 oder 3 Halogenid- oder Pseudohalogenidgruppen (z.B. F, Cl, Br, I oder NCS). Das folgende Diagramm zeigt die Tc-Thioetherkomplexe, die chemisch plausibel sind. Klammern umschließen die Ladungen von Verbindungen, die weniger wahrscheinlich existieren. X bedeutet Halogenid oder Pseudohalogenid. Oxidationsstufe:
- Der Ligand L wurde zum ersten Mal 1986 im anorganischchemischen Labor an der Oxford-Universität synthetisiert.¹ Diese und die nachfolgende Arbeit² zeigten die Wirksamkeit von L als ein Ligand für Co(II) und Ni(II). Die nachfolgende Arbeit hat gezeigt, daß er auch Ru(II) stark bindet, um das hexakoordinierte [Ru(L)]²&spplus;-Kation zu bilden.³
- Kronen-Thioetherliganden wurden über Synthesewege hergestellt, die jetzt in der Literatur erschienen sind.4,5 Im allgemeinen beinhalten diese eine Reaktion von Thioetherdithiolen mit Oligo(thioether)dichloriden in Dimethylformamid, das Cs&sub2;CO&sub3; als Base enthält. Die Synthesen von 9S3 (Gleichung 1), 1856 (Gleichungen 2a-2c) und dem supertripodalen Liganden L (Gleichungen 3a-b) sind nachfolgend dargestellt.

- Kronen-Thioether wurden vor mehr als 50 Jahren erstmals dargestellt &sup6;, aber ihre Koordinationschemie wurden bis zu den frühen 1960ern nicht untersucht und der größte Teil ihrer Koordinationschemie ist in den letzten 10 Jahren untersucht worden. Zum Beispiel wurde der Ligand 9S3 1977 hergestellt und seine Koordination geht nur auf 1982 zurück. In jener Zeit wurden Kronen-Thioetherkomplexe von einer Anzahl von Übergangsmetallen, einschließlich insbesondere Ru (mit 14S47, 8 und 9S39, 10), hergestellt.
- Angesichts dieser früheren Arbeiten war es unklar, welche Oxidationsstufe des Tc sich in Komplexen ergeben würde, die gebildet werden könnten, wenn der Ligand mit Pertechnetat umgesetzt würde, und die biologischen Verteilungseigenschaften von sich ergebenden Komplexen waren ebenfalls unklar.
- In Analogie mit früheren Arbeiten sollten die Kronen- Thioether Tc-Chelate bilden, die sich im Redoxpotential (für die Hexakis(thioether)-Reihe [Tc(L)&sub2;]²&spplus;, L = 9S3, 12S3, 18S6) und in der Koordinationszahl (für [Tc(14S4)Xm]n+, worin m 1 oder 2 sein kann und [Tc(15S5)X]n+) unterscheiden. Die Variation des Redoxpotentials stellt ein wichtiges Mittel zur Abstimmung der Ladung am Kation zur Verfügung und dadurch zur Beeinflussung seiner biologischen Verteilung. Zum Beispiel würde sich ein Chelat für das das Tc(III/II)-Potential in den physiologischen Bereich fällt, nicht sauber in einem Gewebe lokalisieren. Die geeignete Wahl eines anderen Liganden könnte dazu verwendet werden, das Potential aus dem physiologischen Bereich herauszuverschieben und dadurch diesem Problem zu begegnen.
- Die Verfügbarkeit von Koordinationsstellen an dem Tc-Ion beeinflußt ebenfalls die biologische Verteilung. Leicht austauschbare X-Liganden (im Gegensatz zu dem Kronen- Thioether, welcher schwieriger auszutauschen ist) erlauben dem biologischen Milieu, mit Tc-Chelaten (z.B. solchen, die zur Knochenabbildung verwendet werden) wechselzuwirken. Diese Wechselwirkung (zum Beispiel mit Proteinthiolatgruppen) kann die biologische Verteilung stark beeinflussen. Die Kronen- Thioether sind ausgewählt worden, um entweder die Tc- Koordinationssphäre abzusättigen (9S3, 12S3, 18S6) oder dieses nicht zu tun (14S4, 15S5), um beide Arten des Verhaltens zu untersuchen.
- Die Reaktion von n-Bu&sub4;NTcO&sub4; (85 mg, 0,21 mmol) mit dem Kronen-Thioether 9S3 (160 mg, 0,9 mmol) in MeCN (5 ml) und hinzugefügtem HBF&sub4; (0,4 ml von einer 54 % Lösung in Et&sub2;O) ergab eine dunkelbraune Lösung. Nach 1 h Rühren bildete sich ein brauner mikrokristalliner Niederschlag, der mit Et&sub2;O gewaschen wurde, um das Produkt zu ergeben (115 mg, 79 %). Die Elementaranalyse weist auf die Formel [Tc(9S3)&sub2;](BF&sub4;)&sub2;.2H&sub2;0.1/2MeCN hin, eine vormals nicht beschriebene Verbindung. (Anal. ber. (gefunden) für C&sub1;&sub3;H29,5N0,5O&sub2;S&sub6;B&sub2;F&sub8;Tc: C 22,76 (22,63), H 5,16 (4,34), N 0,99 (1,01)).
- Die Zugabe von gesättigter wässriger Lösung von NH&sub4;PF&sub6; zu dem in 5 ml H&sub2;O gelösten BF&sub4;&supmin;-Salz fällte einen goldbraunen Feststoff aus, der filtriert, mit H&sub2;O und Et&sub2;O gewaschen und an Luft getrocknet wurde. Die Elementaranalyse weist auf die Formel [Tc(9S3)&sub2;](PF&sub6;)&sub2;.H&sub2;O hin, ebenfalls eine vormals nicht beschriebene Verbindung. (Anal. ber. (gefunden) für C&sub1;&sub2;H&sub2;&sub6;0S&sub6;P&sub2;F&sub1;&sub2;Tc: C 18,75 (18,78); H 3,12 (3,41)).
- Bei langsamer Diffusion von Et&sub2;O in Lösungen der 8F4&supmin;- oder PF&sub6;&supmin;-Salze in CH&sub3;CN oder CH&sub3;NO&sub2; bildeten sich Kristalle. Während der Kristallisation wurden diese manchmal blaßrot, was einem Produkt entsprechen mag, das sich entweder durch Solvolyse (insbesondere Hydrolyse durch zusätzliches Wasser) oder langsame Oxidation durch Luft ergibt.
- Die elektrochemische Charakterisierung von [Tc(9S3)&sub2;]²&spplus; durch cyclische Voltammetrie (MECN, Pt-Elektrode, 0,1 M Bu&sub4;NPF&sub6; Elektrolyt) zeigt eine quasi-reversible Welle (wahrscheinlich dem Tc(II/I)-Paar entsprechend) bei +0,05 V vs die gesättigte Kalomelelektrode (GKE); -0,38 V vs Ferrocenium/Ferrocen (Fc&spplus;/Fc); +0,02 V vs Standardwasserstoffelektrode (SWE). Sie zeigt ferner eine irreversible Ein-Elektronenoxidation bei +1,3 V vs GKE (+0,87 V vs Fc&spplus;/Fc, +1,27 V vs SWE).
- Ein anderes Redoxpaar ergibt eine irreversible Reduktion bei -1,6 V vs GKE (-2,03 V vs Fc&spplus;/Fc, -1,63 V vs SWE). Aufgrund seines viel kleineren Peak-Stroms als die der oben beschriebenen Wellen rührt dieser Redoxprozeß wahrscheinlich entweder von einer Verunreinigung oder von einem Reaktionsprodukt von [Tc(9S3)&sub2;]²&spplus; unter den experimentellen Bedingungen her.
- Zusätzlich erscheint ein quasi-reversibler Prozeß bei -0,66 V vs GKE (-1,09 V vs Fc&spplus;/Fc, -0,69 V vs SWE). Im ersten Scan (100 mv/s) hat diese Welle ein Viertel des Stroms der Welle bei +0,05 V vs GKE. Während nachfolgender Scans wuchs der mit dieser Welle bei -0,66 V vs GKE verbundene Strom auf Kosten derjenigen bei +0,05 V vs GKE; bei dem zehnten Scan erreicht das System entweder ein Gleichgewicht oder einen stationären Zustand. Diese Ergebnisse legen nahe, daß bei Reduktion von [Tc(9S3)&sub2;]²&spplus; zu [Tc(9S3)&sub2;]&spplus; (der Prozeß bei +0,05 V) das elektroerzeugte [Tc(9S3)&sub2;]&spplus; (vielleicht) mit CH&sub3;CN reagiert, um ein Produkt zu ergeben, das für sich selbst redox-aktiv ist (und welches die Welle bei -0,66 V hervorruft).
- Bei cyclischer Voltammetrie in wässriger Lösung wurde 0,1 M NaCl als unterstützendes Elektrolyt verwendet; die anderen Bedingungen waren wie oben beschrieben. Es treten zwei quasireversible Redoxprozesse auf, einer bei -0,20 V vs GKE (+0,02 V vs NHE) und ein anderer bei -0,60 V vs GKE (-0,37 V vs NHE). Der Peak-Strom der zweiten Welle hängt von der Anzahl der Scans ab. Bei aufeinanderfolgenden Scans wuchs er zu einem Gleichgewichts- oder stationären Zustandswert von ungefähr der Hälfte desjenigen der Welle bei -0,20 V.
- Parallele synthetische Verfahren mit der supertripodalen
- a Verbindung L ergaben den Beweis für den analogen Tc(II)- w Komplex. Unter den oben beschriebenen Bedingungen reagiert L mit TcO&sub4;&supmin;, um eine in ihrer Erscheinung mit der des 9S3- Homologen identische Lösung zu ergeben. Folglich wird er als [Tc(L)]²&spplus; formuliert, welcher ebenfalls vormals nicht beschrieben war.
- Die Analogie mit der bekannten Koordinationschemie dieser Liganden legt für sie die in Abbildung 2 gezeigten Strukturen nahe, nämlich [Tc(9S3)&sub2;]²&spplus; bzw. [TcL]²&spplus;.
- [&sup9;&sup9;mTc(9S3)&sub2;]+ wurde durch SnCl&sub2;-Reduktion von [&sup9;&sup9;mTc(9S3)&sub2;]²&spplus; hergestellt. Die als mittlere %ID/g (S.D.) (N = 2 außer wie angezeigt) gegebene biologische Verteilung wurde in weiblichen CD-Mäusen 1 h nach Injektion bestimmt. Claims (7)
1. Technetiumkomplex eines Kronen-Thioether, der drei bis
sechs Donor-Schwefelatome aufweist, die durch C2- oder C3-
Gruppen in einen makrozyklischen Ring eingebunden sind.
2. Komplex nach Anspruch 1, worin der Kronen-Thioether
1,4,7-Trithiacyclononan (9S3) ist.
3. Komplex nach Anspruch 2, der die Formel [TcL&sub2;]&spplus; oder
[TcL&sub2;]²&spplus; oder [TcL&sub2;]³&spplus; aufweist, wobei L der Kronen-Thioether
ist.
4. Technetiumkomplex von einem tripodalen Liganden, der mehr
als drei Donor-Schwefelatome aufweist, die durch C2- oder C3-
Gruppen verbunden sind.
5. Komplex nach Anspruch 4, worin der Ligand 1,1,1-Tris(((2-
methylthio)ethylthio)-methyl)ethan ist.
6. Komplex nach Anspruch 5, der die Formel [TcL]&spplus; oder
[TcL]²+ oder [TcL]³&spplus; aufweist, wobei L der Ligand ist.
7. Komplex nach einem der Ansprüche 1 bis 6, worin das
Technetium Technetium-99m ist.
