Description
- Die Erfindung betrifft neue Benzochinonverbindungen und deren Verwendung. Die neuen Verbindungen der vorliegenden Erfindung sind Inhibitoren sowohl für Cyclooxygenase-(CO)- als auch Lipoxygenase-(LO)-Enzyme und sind zur Verwendung bei der Behandlung oder Linderung von allergischen oder entzündlichen Zuständen bei Säugern, einschließlich Menschen, vorgesehen.
- Arachidonsäure ist als biologische Vorstufe verschiedener Gruppen endogener Metaboliten, Prostaglandinen, einschließlich Prostacyclinen, Thomboxanen und Leukotrienen, bekannt. Der erste Schritt des Arachidonsäuremetabolismus ist die Freisetzung veresterter Arachidonsäure und verwandter ungesättigter Fettsäuren aus Membranphospholipiden über die Wirkung von Phospholipase. Freie Fettsäuren werden dann entweder durch Cyclooxygenase unter Herstellung der Prostaglandine und Thromboxane oder durch Lipoxygenase unter Erzeugung von Hydroperoxyfettsäuren, die weiter zu Leukotrienen umgewandelt werden können, metabolisiert. Die Prostaglandine zeigen verschiedene physiologische Wirkungen in Abhängigkeit von ihrer Struktur. Zum Beispiel inhibieren PGE und PGA die Magensaftsekretion sowie auch niederen arteriellen Blutdruck. Thromboxan, insbesondere Thromboxan A&sub2;, ist ein starker Vasokonstriktor und eine Thrombozyten-aggregierende Substanz. Die Leukotriene sind die biologische Quelle für die langsam reagierende Substanz der Anaphylaxie (SRS-A), einem chemischen Mediator bei allergischem bronchialem Asthma.
- Aspirin und die meisten anderen nichtsteroiden entzündungshemmenden Arzneistoffe inhibieren das Cyclooxygenaseenzym. Sowohl entzündungshemmende Wirkung als auch analgetische Wirkung, die mit diesen Arzneistoffen verbunden sind, werden hinsichtlich ihrer Inhibierung der Wirkung von Cyclooxygenase erklärt. Die Lipoxygenase-hemmende Wirkung eines Mittels, AA861 2,3,5-Trimethyl-6-(12-hydroxy-5,10-cyclodecadiinyl)-1,4-benzochinon, wurde mitgeteilt (siehe Yoshimoto et al. Biochem. et Biophys. 713, 470-473 (1982)). CGS- 5391B (C. E. Hock et al., Prostaglandins, 28, 557-571 (1984)) wurde kürzlich bekannt als eine Kombination aus einem Cyclooxygenase- und Lipoxygenaseinhibitor.
- Die PCT-Patentanmeldung PCT/JP84/00452 (WO 85/01289) und die Japanische Patentveröffentlichung Nr. 107958/1988 beschreiben und beanspruchen daneben eine Reihe von Benzoxazolon- und Benzothiazolonderivaten, die zur Behandlung von Entzündungszuständen und Thrombose geeignet sind.
- Musso und Döpp in Chemische Berichte 1966, 99, 1470- 1478 offenbaren die Synthese und die Lichtabsorption von 5[2,4-Dimethoxyanilino]-2,3-dimethylbenzochinon, während Petersen et al. in Z. Krebsforschung 1969, 72, 162-175, verschiedene Derivate von 2-Amino-1,4-naphthochinonen mit carcinostatischer Aktivität beschreiben.
- Die erfindungsgemäßen Verbindungen haben die Formel
- oder die pharmazeutisch verträglichen Salze davon, worin:
- R¹ Wasserstoff, Alkyl mit 1 bis 5 Kohlenstoffatomen, Cycloalkyl mit 3 bis 7 Kohlenstoffatomen, Phenylalkyl mit 7 bis 9 Kohlenstoffatomen, -COR&sup4;, wobei R&sup4; Alkyl mit 1 bis 5 Kohlenstoffatomen, Phenyl, Hydroxy, Amino, Alkoxy mit 1 bis 5 Kohlenstoffatomen, ω-Carboxyalkyl mit 2 bis 5 Kohlenstoffatoinen oder ω-Alkoxycarbonylalkyl mit 3 bis 8 Kohlenstoffatomen oder -CH(OZ)R&sup8; bedeutet, wobei R&sup8; Wasserstoff, Alkyl mit 1 bis 3 Kohlenstoffatomen, ω-Carboxyalkyl mit 2 bis 5 Kohlenstoffatomen oder ω-Alkoxycarbonylalkyl mit 3 bis 8 Kohlenstoffatomen bedeutet und Z Wasserstoff oder Methoxyethoxymethyl darstellt;
- R² Wasserstoff oder Phenyl bedeutet;
- R³ (a) Cycloalkyl mit 3 bis 7 Kohlenstoffatomen, (b) Cycloalkylalkyl mit 5 bis 7 Kohlenstoffatomen, gegebenenfalls mit einem Sauerstoff-, Schwefel- oder Stickstoffatom als Ringglied, (c) einen Rest der Formel
- worin m eine ganze Zahl von 0 bis 3 darstellt und R&sup5; und R&sup6; jeweils Wasserstoff, Halogen, Carboxy, Carbamoyl, Carboalkoxy mit 2 bis 4 Kohlenstoffatomen, Alkoxycarbonylmethyl mit 3 bis 5 Kohlenstoffatomen, oder Sulfamoyl bedeuten oder (d) eine substituierte Alkylgruppe der Formel R&sup7;-Alkyl bedeutet, worin die Alkylgruppe 5 bis 12 Kohlenstoffatome enthält, und R&sup7; Hydroxy, Carboxy, Carboalkoxy mit 2 bis 4 Kohlenstoffatomen oder Hydroxyalkoxy mit 2 bis 4 Kohlenstoffatomen bedeutet; oder R² und R³ zusammengenommen mit dem Stickstoffatom, an das sie gebunden sind, eine Struktur der Formel
- bilden.
- Eine bevorzugte Gruppe von Verbindungen sind jene, worin R¹ und R² jeweils Wasserstoff bedeuten.
- Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R³ einen Rest der Formel
- darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils Wasserstoff darstellen.
- Eine zweite Gruppe bevorzugter Verbindungen sind jene, worin R¹ -COR&sup4; oder -CH(OZ)R&sup8; darstellt, wobei R&sup4; Alkyl mit 1 bis 5 Kohlenstoffatomen bedeutet, R&sup8; Alkyl mit 1 bis 3 Kohlenstoffatomen oder Wasserstoff bedeutet, Z Wasserstoff darstellt und R² Wasserstoff bedeutet.
- Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R¹ -COR&sup4; darstellt, wobei R&sup4; Methyl bedeutet und R³ einen Rest der Formel
- darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils Wasserstoff darstellen und worin R¹ -CH(OZ)R&sup8; ist, wobei R&sup8; Wasserstoff oder Methyl bedeutet und R³ einen Rest der Formel
- bedeutet, worin m gleich 0 ist und R&sup5; und R&sup6; jeweils Wasserstoff bedeuten.
- Eine dritte Gruppe bevorzugter Verbindungen sind jene, worin R² und R³ zusammengenommen mit dem Stickstoffatom, an das sie gebunden sind, die Struktur der Formel
- bilden.
- Besonders bevorzugt innerhalb dieser Gruppe ist die Verbindung, worin R¹ Wasserstoff bedeutet und R² und R³ zusammengenommen mit dem Stickstoffatom, an das sie gebunden sind, die Struktur der Formel
- bilden.
- Die vorliegende Erfindung schließt ebenfalls ein Arzneimittel zur Verabreichung an einen Menschen, umfassend eine Verbindung der Formel (I) und ein pharmazeutisch verträgliches Träger- oder Verdünnungsmittel, ein.
- Der Ausdruck "Halogen" schließt Fluor, Chlor, Brom und Jod ein.
- Die pharmazeutisch verträglichen Salze der Verbindungen der Formel (I) sind jene, die von Säuren gebildet werden, die nichttoxische Säureadditionssalze bilden, beispielsweise die Hydrochlorid-, Hydrobromid-, Sulfat- oder Bisulfat-, Phosphat-, Acetat-, Citrat-, Fumarat-, Gluconat-, Lactat-, Maleat-, Succinat-, Tartrat-, Methansulfonat-, Benzolsulfonat-, Toluolsulfonat- und Formiatsalze. Wenn R¹ oder R³ eine Carboxylgruppe enthalten, ist es möglich, Salze mit organischen und anorganischen Basen zu bilden. Diese schließen solche organischen Basen ein, wie Triethylamin, Ethanolamin und Triethanolamin und solche anorganischen Basen, wie Alkalimetallhydroxide und Erdalkalimetallhydroxide.
- Es ist bevorzugt, daß alle diese Salze pharmazeutisch verträglich sind.
- Die Verbindungen der Formel (I) können auf verschiedenen Wegen hergestellt werden. In einer Ausführungsform werden sie aus 2-substituiertem oder 2-nichtsubstituiertem 5,6- Dimethylbenzochinon (II) und einem substituierten Amin gemäß nachstehender Reaktion hergestellt:
- In den vorstehenden Formeln sind R¹, R² und R³ wie vorstehend definiert. Die Aminsubstituenten R² und R³ sind vorzugsweise H, eine Alkyl- oder Aralkylgruppe. Die Reaktion wird vorzugsweise bei Umgebungstemperatur in Gegenwart eines Oxidationsmittels ausgeführt. Das Oxidationsmittel kann Benzochinon (II) selbst sein. Erhöhte Temperaturen können ohne bedeutenden Nachteil angewendet werden. Geeignete Lösungsmittel, die nicht mit den Reaktanten und/oder Produkten reagieren, sind beispielsweise Ethanol, Methanol, Dichlormethan und Chloroform. Wenn eine bevorzugte Temperatur verwendet wird, ist die Umsetzung im wesentlichen innerhalb weniger Stunden beendet. Nach Beendigung kann Produkt (I) isoliert und/oder gereinigt werden auf übliche Weise, beispielsweise durch Umkristallisieren oder Chromatographie.
- In einer anderen Ausführungsform werden die Verbindungen der Formel (I) durch nachstehende Umsetzung hergestellt unter Verwendung von 2-substituiertem oder unsubstituiertem 5,6-Dimethylhydrochinon (III) und einem substituierten Amin: Oxidationsmittel
- In den vorstehenden Formeln sind R¹ und R² wie vorstehend definiert. Der Aminsubstituent R² ist vorzugsweise eine Arylgruppe. Die Reaktion wird vorzugsweise bei Umgebungstemperatur in Gegenwart eines Oxidationsmittels ausgeführt. Das Oxidationsmittel ist vorzugsweise Natriumjodat, obwohl Natriumperjodat, Natriumchlorat oder Mangandioxid ebenfalls angewendet werden können. Geeignete Lösungsmittel, die nicht mit den Reaktanten und/oder Produkten reagieren, sind beispielsweise wässeriges Methanol. Wenn die bevorzugte Temperatur verwendet wird, erfordert die Umsetzung im wesentlichen einige Stunden bis einige Tage. Nach Beendigung kann Produkt (I) durch übliche Standardverfahren, beispielsweise durch Umkristallisieren oder Chromatographie, isoliert und/oder gereinigt werden.
- In einer weiteren Ausführungsform können die Verbindungen der Formel (I) (R³ bedeutet Alkyl, Aralkyl oder R&sup7; Alkyl) ebenfalls aus den Verbindungen der Formel (I) hergestellt werden (R³ ist Wasserstoff) durch nachstehendes Verfahren:
- Die Aminsubstituenten, R² ist vorzugsweise Aryl und RCH&sub2; ist Aralkyl oder R&sup7; Alkyl. In der vorstehenden Formel sind R¹, R² und R&sup7; wie vorstehend definiert. Der erste Schritt umfaßt die Behandlung einer Gruppe von Verbindungen (I) (R³=H) mit einem Reduktionsmittel. Das bevorzugte Reduktionsmittel ist Natriumhydrogensulfit, obwohl katalytisches Hydrieren, beispielsweise in Gegenwart von Palladium-auf- Kohle, ebenfalls angewendet werden kann. Die Reaktion wird vorzugsweise bei Umgebungstemperatur ausgeführt. Geeignete Lösungsmittel, die nicht mit den Reaktanten und/oder Produkten reagieren, sind beispielsweise Diethylether/Wasser oder Essigsäureethylester/Wasser. Wenn die bevorzugte Temperatur verwendet wird, ist die Umsetzung im wesentlichen innerhalb weniger Stunden beendet. Nach Beendigung kann Produkt (IV) isoliert und/oder gereinigt werden auf übliche Weise, beispielsweise durch Umkristallisieren. Es ist zweckmäßiger, dieses Produkt nicht zu isolieren, sondern es den Reaktionsbedingungen des zweiten Schrittes zu unterwerfen.
- Der zweite Schritt bezieht eine Entwässerung von einem Aldehyd in Gegenwart von Säure ein. Die Säure kann entweder eine protische oder eine aprotische Säure, wie p-Toluolsulfonsäure, Camphersulfonsäure oder Bortrifluoretherat sein. Die bevorzugte Temperatur für diese Reaktion ist von 0 bis 110ºC. Reinigung des Produktes (V) kann durch übliche Maßnahmen, wie Umkristallisieren oder Chromatographie, erfolgen. Der dritte Schritt umfaßt die Reduktion der CN-Bindung durch die Reaktion mit einer geeigneten Wasserstoffquelle. Beispielsweise kann die Verbindung (V) unter Verwendung von Metallhydriden reduziert werden. Das Hydridmittel, das bei dieser Reaktion geeigneterweise zur Anwendung kommt, umfaßt Natriumborhydrid, Natriumcyanoborhydrid oder Lithiumcyanoborhydrid. Diese Umsetzung wird mit einem Überschuß eines Hydridmittels, beispielsweise in Essigsäure bei Umgebungstemperatur, ausgeführt. Eine bevorzugte Temperatur für diese Reaktion beträgt 15 bis 50ºC. Die Reduktion ist gewöhnlich innerhalb einer Stunde vollständig. Nach Ablauf kann Produkt (VI) isoliert und/oder gereinigt werden, beispielsweise durch Umkristallisieren. Es ist zweckmäßiger, dieses Produkt nicht zu isolieren, sondern es den Reaktionsbedingungen des vierten Schritts zu unterwerfen. Der vierte Schritt schließt die Oxidation des Hydrochinonrestes (VI) mit einem geeigneten Oxidationsmittel ein. Beispielsweise kann Verbindung (VI) mit Eisen(III)chlorid, Mangandioxid, Natriumperjodat oder Natriumjodat oxidiert werden. Die Reaktion wird vorzugsweise bei Umgebungstemperatur ausgeführt. Geeignete Lösungsmittel, die nicht mit den Reaktanten und/oder Produkten reagieren, sind beispielsweise Wasser und/oder Methanol. Das Produkt von Formel (I) wird durch Standardverfahren isoliert und durch übliche Maßnahmen, wie Umkristallisieren oder Chromatographie, gereinigt.
- Die Ausgangsmaterialien (II) (R¹=H) oder (III) (R¹= COCH&sub3;) sind entweder bekannte Verbindungen oder können durch Verfahren, die im Stand der Technik mitgeteilt wurden, hergestellt werden, vergleiche beispielsweise Etsuro Kurosawa, Bull. Chem. Soc. of Japan, 34, 300 (1961). Die anderen Ausgangsmaterialien können mit dem üblichen Verfahren hergestellt werden.
- In den vorstehenden Formeln kann COR&sup9; R¹ wie vorstehend definiert entsprechen oder kann eine funktionelle Gruppe darstellen, die in R¹ umgewandelt werden kann. Der erste Schritt umfaßt die Behandlung einer Gruppe von Verbindungen (VII) mit einem Metallisierungsmittel, gefolgt von Kohlendioxidbehandlung. Die bevorzugten Metallisierungsmittel sind Alkyllithium oder Magnesium. Geeignete Lösungsmittel, die nicht mit den Reaktanten und/oder Produkten reagieren, sind beispielsweise Diethylether, Tetrahydrofuran und niedere Kohlenwasserstoffe. Nach Ablauf der Umsetzung kann Produkt (VII) isoliert und/oder in üblicher Weise gereinigt werden, beispielsweise durch Umkristallisieren. Der zweite Schritt schließt die Veresterung oder Amidierung ein. Diese Reaktionsbedingungen können durch Standardverfahren ausgeführt werden, beispielsweise in Bezug auf "Comprehensive Organic Chemistry" von Sir Deric Barton, F R S und W. David Ollis, F R S Herausg., Pergamon Press, 1979, Bd. 2, Seiten 869 und 957.
- Der dritte Schritt umfaßt die Behandlung einer Gruppe von Verbindungen (IX) mit einem Oxidationsmittel. Das bevorzugte Oxidationsmittel ist beispielsweise Cerammonnitrat. Die Oxidation ist gewöhnlich innerhalb weniger Sekunden bei Raumtemperatur vollständig. Das Produkt der Formel X wird durch Standardverfahren isoliert und durch übliche Maßnahmen, wie Umkristallisieren oder Chromatographie, gereinigt.
- Ausgangsmaterial kann ebenfalls durch nachstehende Reaktionsfolge hergestellt werden. Lewissäure
- In den vorstehenden Formeln kann COR¹&sup0; zu R¹, wie bereits definiert, entsprechen oder kann eine funktionelle Gruppe darstellen, die in R¹ umgewandelt werden kann. Bei dieser besonderen Reaktion können akzeptable Lewissäurekatalysatoren oder Lösungsmittel jene sein, die im Stand der Technik mitgeteilt werden, wie "Organic Chemistry", Jerry March, Herausg., John Wiley & Sons, Inc., 1985, Seite 484 ff. Das acylierte Produkt (IX) kann mit einem Oxidationsmittel behandelt werden. Das bevorzugte Oxidationsmittel ist beispielsweise Cerammonnitrat. Die Oxidation ist gewöhnlich innerhalb weniger Sekunden bei Raumtemperatur vollständig. Das Produkt der Formel X wird durch Standardverfahren isoliert und durch übliche Maßnahmen, wie Umkristallisieren oder Chromatographie, gereinigt.
- Die pharmazeutisch verträglichen Salze der neuen Verbindungen der Formel (I) sind leicht durch Inkontaktbringen der Verbindung mit einer stöchiometrischen Menge einer geeigneten Mineral- oder organischen Säure oder Base entweder in wässeriger Lösung oder in einem geeigneten organischen Lösungsmittel herstellbar. Das Salz kann dann durch Fällung oder durch Abdampfen des Lösungsmittels erhalten werden. Unter diesen Salzen, die vorher aufgezählt wurden, ist ein besonders bevorzugtes Salz das Hydrochlorid oder Natriumsalz.
- Die Verbindungen der Formel (I) weisen inhibierende Aktivität auf die Wirkung der Cyclooxygenase sowie auf die Wirkung der Lipoxygenase auf. Diese Aktivität wurde demonstriert durch Zellkulturbestimmung unter Verwendung von Zellen aus der Bauchhöhle von Ratten, womit die Wirkung der Verbindungen auf den Metabolismus von Arachidonsäure bestimmt wird.
- Die Fähigkeit der Verbindungen der Formel (I), beide Enzyme zu inhibieren, macht sie geeignet zur Behandlung der Symptome, die durch bei einem Säuger auftretende endogene Metaboliten von Arachidonsäure induziert werden. Die Verbindungen sind daher wertvoll bei der Verhinderung und Behandlung solcher Krankheitszustände, bei denen die Akkumulation des Arachidonsäuremetaboliten der ursächliche Faktor ist, beispielsweise allergischem bronchialem Asthma, Hauterkrankungen, rheumatischer Arthritis, Osteoarthritis und Thrombose.
- Da übliche nichtsteroide entzündungsbekämpfende Mittel, wie Aspirin, lediglich Cyclooxygenase inhibieren, unterdrücken sie Entzündungszustände sowie neigen dazu, nachteilige gastrointestinale Reaktion aufgrund der Enzyminhibierung hervorzurufen. Verbindungen der vorliegenden Erfindung sind jedoch darüberhinaus gastrointestinal cytoschützend, indem sie antiallergische und entzündungshemmende Wirkungen aufweisen. Somit zeigen sie weniger nachteilige Wirkungen und sind für die Verwendung als sicherer Arzneistoff geeignet.
- Wenn eine Verbindung der Formel (I) oder ein pharmazeutisch verträgliches Salz davon entweder als antiallergisches Mittel oder als entzündungshemmendes Mittel verwendet werden soll, kann es an einen Menschen entweder einzeln oder vorzugsweise in Kombination mit pharmazeutisch verträglichen Träger- oder Verdünnungsmitteln in einem Arzneimittel im Einklang mit der üblichen pharmazeutischen Praxis verabreicht werden. Eine Verbindung kann auf zahlreichen üblichen Wegen der Verabreichung, einschließlich oralem, parenteralem Weg und durch Inhalation verabreicht werden. Wenn die Verbindungen oral verabreicht werden, wird der Dosierungsbereich etwa 0,1 bis 20 mg/kg Körpergewicht des zu behandelnden Patienten pro Tag in einzelnen oder verteilten Dosierungen betragen. Wenn parenterale Verabreichung erwünscht ist, kann die wirksame Dosis 0,1 bis 1,0 mg/kg Körpergewicht des zu behandelnden Patienten pro Tag betragen. In einigen Fällen mag es erforderlich sein, Dosierungen außerhalb dieser Grenzen zu verwenden, da die Dosierungen notwendigerweise gemäß dem Alter, Gewicht und der Reaktion des einzelnen Patienten sowie der Schwere der Symptome des Patienten und der Wirkung der einzelnen zu verabreichenden Verbindung schwanken wird.
- Zur oralen Verabreichung können die Verbindungen der Formel (I), beispielsweise in Form von Tabletten, Pulvern, Pastillen, Sirupen oder Kapseln oder als wässerige Lösung oder Suspension verabreicht werden. Im Falle von Tabletten zur oralen Verwendung sind üblicherweise verwendete Träger Lactose und Maisstärke und Schmiermittel, wie Magnesiumstearat, werden üblicherweise zugegeben. Im Falle von Kapseln sind geeignete Verdünnungsmittel Lactose und getrocknete Maisstärke. Wenn wässerige Suspensionen zur oralen Verwendung erwünscht sind, wird der Wirkstoff mit emulgierenden und suspendierenden Mitteln verdünnt. Falls erwünscht, können bestimmte Süßungs- und/oder Aromatisierungsmittel zugegeben werden. Für intramuskuläre, intraperitoneale, subcutane und intravenöse Verwendung werden sterile Lösungen des Wirkstoffs gewöhnlich hergestellt und der pH-Wert der Lösung sollte geeigneterweise eingestellt und gepuffert werden.
- Die vorliegende Erfindung wird durch die nachstehenden Beispiele erläutert. Selbstverständlich sind die Beispiele jedoch nur erläuternd und die Erfindung ist nicht auf die speziellen Einzelheiten dieser Beispiele begrenzt. Kernmagnetische Protonen-Resonanzspektroskopie (NMR) wurde bei 270 MHz gemessen, sofern nicht anders ausgewiesen, für Lösungen in Deuterochloroform und die Peakorte werden in ppm (parts per million) feldabwärts von Tetramethylsilan ausgedrückt. Die Peakformen werden wie nachstehend bezeichnet: s, Singulett; d, Dublett; t, Triplett; q, Quartett; m, Multiplett; b, breit.
- Zu einer gerührten Lösung von 2,3-Dimethylhydrochinon (13,8 g, 0,1 Mol) und Anilin (18,6 g, 0,2 Mol) in Methanol (600 ml) wurde Natriumjodat (59,4 g, 0,3 Mol) in Wasser (600 ml) bei Raumtemperatur gegeben. Das Gemisch wurde für 17 Stunden gerührt. Das Gemisch wurde eingeengt (Entfernung von 600 ml Lösungsmittel) und mit Chloroform (700 ml x 3) extrahiert. Die vereinigten organischen Phasen wurden getrocknet und zu einem dunkelrot violetten Öl eingeengt. Dieses Öl wurde über Nacht stehen gelassen. Der Rückstand war verfestigt und wurde in Hexan suspendiert. Die erhaltenen feuchten Niederschläge wurden durch Saugfiltration gesammelt und die Niederschläge wurden in Diisopropylether suspendiert. Der Niederschlag wurde durch Saugfiltration gesammelt. Der Feststoff wurde mit Diisopropylether unter Erhalt eines dunkelbraunen Feststoffs gewaschen (11,7 g). Dieser (1,05 g) wurde in Methanol (40 ml) gelöst und Aktivkohle (290 mg) wurde zugegeben. Das Gemisch wurde bei 50ºC für 10 Minuten gerührt und durch Kieselgur filtriert. Der Rückstand wurde mit Methanol gewaschen. Das vereinigte Filtrat wurde im Vakuum eingedampft. Umkristallisieren aus Methanol ergab rostbraune Kristallschuppen der Titelverbindung (500 mg, 47,6 %). Diese wurden bei 50ºC für 3 Stunden getrocknet unter Erhalt des Produktes, Fp. 115,0-115,7ºC/Zers.. Ein weiterer Teil war ebenfalls aus der Mutterlauge erhältlich (350 mg, 33%).
- ¹H NMR (270 MHz, CDCl&sub3;) δ 7,42-7,33 (3H, m,; Ph-H x 2+NH), 7,22-7,12 (3H, m, Ph-H), 6,15 (1H, s, H-3), 2,06-2,03 (6H, m, CH&sub3; x 2), IR (KBr) 3300, 1670, 1645.
- In ähnlicher Weise wurden die nachstehenden Verbindungen hergestellt. Beispiel
- a)¹H NMR-Spektrum dieser Probe wurde in DMSO-d&sub6; gemessen
- Zu einer Lösung von 2,3-Dimethyl-1,4-benzochinon (2,72 g, 20 mMol) in Methanol (50 ml) wurde Benzylamin (2,14 g, 12 mMol) bei Raumtemperatur gegeben. Nach Rühren für 1 Stunde wurde das Lösungsmittel entfernt und der Rückstand wurde durch Kieselgelsäulenchromatographie (250 g, eluiert mit Hexan/Essigsäureethylester) gereinigt. Umkristallisieren aus Essigsäureethylester und Diisopropylether ergab kastanienbraune Nadeln der Titelverbindung (0,82 g, 34%) Fp. 110- 111ºC.
- ¹H NMR (270 MHz, CDCl&sub3;) δ 7,36-7,26 (5H, m; Ph-H x 5), 5,93 (1H, br s; NH), 5,48 (1H, s; H-3), 4,28 (2H, d, J=5,9 Hz, CH&sub2;), 2,03 (3H, br s, CH&sub3;), 2,00 (3H, br s, CH&sub3;).
- IR (KBr) 3300, 1670, 1650.
- In ähnlicher Weise wurden die nachstehenden Verbindungen hergestellt. Beispiel Beispiel Beispiel 26
- Eine gerührte Lösung von 2-Anilino-5,6-dimethylbenzochinon (1) (2,24 g, 0,01 Mol) in Diethylether (100 ml) wurde zu einer gerührten Lösung von Natriumhydrogensulfit (6,96 g, 0,04 Mol) in Wasser (20 ml) gegeben. Das Gemisch wurde unter Stickstoffatmosphäre für 2,5 Stunden heftig gerührt. Die organische Schicht wurde mit Salzlösung ausgewaschen und über Magnesiumsulfat getrocknet. Die Lösung wurde zu einem dunkellila Öl eingeengt (2). Zu dem Gemisch wurde Benzol (32 ml), Benzaldehyd (1,1 ml, 10,8 mMol) und eine katalytische Menge Camphersulfonsäure gegeben. Das Gemisch wurde für 2 Stunden unter Rückfluß und Stickstoffatmosphäre gerührt. Das Gemisch wurde mit Natriumbicarbonatlösung und anschließend mit Salzlösung gewaschen. Die Lösung wurde getrocknet und eingeengt unter Erhalt eines dunkelrostfarbenen amorphen Feststoffs. Dieser wurde aus Ethanol umkristallisiert und ergab einen schwach violetten Feststoff (3). (1,7 g, 76 % Ausbeute), Fp. 148,5-149,0ºC.
- ¹H NMR (270 MHz, CDCl&sub3;) δ 7,52-7,57 (m, 2H), 7,35- 7,42 (m, 3H), 7,20-7,28 (m, 2H), 7,07 (br d, 2H, J=7,6Hz), 6,97 (br t, 1H, J=7,6Hz), 6,71 (s, 2H), 6,55 (s, 1H), 4,33 (s, 1H, OH), 2,11 (s, 3H), 2,10 (s, 3H).
- IR (KBr) 1600, 1513.
- Zu einer gerührten Lösung des Acetals (3) (1 g, 3 mMol) in Essigsäure (4 ml) wurde Natriumcyanoborhydrid (300 mg, 4,7 mMol) bei Raumtemperatur gegeben. Das Gemisch wurde für 20 Minuten gerührt und anschließend mit Ether extrahiert. Der Extrakt wurde mit Salzlösung gewaschen und über Magnesiumsulfat getrocknet. Die Lösung wurde im Vakuum eingeengt unter Erhalt eines schwach rostfarbenen Öls (4) (1 g). Dieses Hydrochinon wurde in Methanol (10 ml) gelöst und anschließend Natriumjodat (1 g, 5 mMol) in Wasser (10 ml) zugegeben. Das Gemisch wurde für 2 Stunden gerührt. Zu diesem Gemisch wurde Natriumperjodat (600 mg, 2,8 mMol) gegeben, um die Oxidation zur Vollständigkeit zu bringen. Das Gemisch wurde für 1 Minute gerührt und anschließend das Gemisch mit Wasser (50 ml) und Essigsäureethylester (100 ml) verdünnt. Die abgetrennte organische Phase wurde mit Salzlösung gewaschen und über Magnesiumsulfat getrocknet. Die Lösung wurde im Vakuum eingeengt und ergab ein rostfarbenes Öl (etwa 500 mg). Kieselgelsäulenchromatographie, eluiert mit Hexan/Essigsäureethylester (20/1 - 15/1), gefolgt von Kristallisieren aus Hexan/- Diisopropylether ergab ein rostfarbenes Pulver (100 mg, 12 % Ausbeute) Fp. 119,4-119,8ºC.
- ¹H NMR (270 MHz, CDCl&sub3;) δ 7,37-7,15 (8H, m), 7,08 (2H, br. d, J=7,6 Hz), 5,83 (1H, s), 4,94 (2H, s), 1,98 (3H, br. s), 1,92 (3H, br. s).

- b) Das IR-Spektrum dieser Probe wurde als reine Flüssigkeit gemessen. Beispiel 29
- Zu einer gerührten Suspension von 2-Acetyl-5,6-dimethylhydrochinon (1,8 g, 10 mMol) in Methanol-Wasser (1:2, 396 ml) wurde Anilin (2,48 g, 26,6 mMol) in Methanol (33 ml) gegeben. Das Gemisch wurde hauptsächlich zu einer gelben klaren Lösung. Zu dem Gemisch wurde Natriumjodat in heißem Wasser (67 ml) gegeben und für 24 Stunden gerührt. Der erhaltene Niederschlag wurde durch Saugfiltration gesammelt und mit Essigsäureethylester und Wasser extrahiert. Das Filtrat wurde eingeengt bis 300 ml und anschließend mit Essigsäureethylester (200 ml x 2) extrahiert. Der Extrakt wurde mit Salzlösung gewaschen. Die vereinigten Extrakte wurden über Magnesiumsulfat getrocknet und anschließend eingeengt. Chromatographie an Kieselgel, eluiert mit Hexan-Essigsäureethylester (20:1 bis 10:1) ergab die gewünschte Fraktion. Diese wurde im Vakuum eingeengt unter Erhalt eines braun-rostbraunen Pulvers (2,8 g). Dieses wurde aus Ethanol unter Erhalt braun-rostbrauner Nadeln umkristallisiert (2,0 g, 74,3 %). Fp. 115,4-116,3ºC.
- ¹H NMR (270 MHz, CDCl&sub3;) δ 12,97 (br s, 1H), 7,22-7,41 (m, 3H), 7,06 (d, 2H, J=6,8Hz), 2,63 (s, 3H), 2,10 (br s, 3H), 1,96 (br s, 3H).
- IR (KBr) 1675, 1625, 1605.
- Zu einer gerührten Lösung von 3-Acetyl-2-anilino-5,6- dimethyl-1,4-benzochinon (700 mg, 2,6 mMol) in Tetrahydrofuran-Wasser (5:4, 12,5 ml) wurde Natriumborhydrid (197 mg, 5,2 mMol) bei Raumtemperatur gegeben. Das Gemisch wurde bei Raumtemperatur für 10 Minuten gerührt. 1N Salzsäure (1 ml) und Essigsäureethylester (50 ml) wurden zu dem Gemisch gegeben und für 10 Minuten gerührt. Die organische Phase wurde mit Natriumbicarbonatlösung und Salzlösung gewaschen. Die Lösung wurde über Magnesiumsulfat unter Vakuum getrocknet und unter Erhalt eines schwach roten Öls eingeengt. Das Produkt wurde in Methanol (7 ml) gelöst. Zu der Lösung wurde Natriumperjodat in Wasser bei Raumtemperatur gegeben. Das Gemisch wurde für 5 Minuten gerührt und dann mit Essigsäureethylester (50 ml) extrahiert und mit Salzlösung gewaschen (10 ml x 4). Die Lösung wurde über Magnesiumsulfat getrocknet und im Vakuum eingeengt unter Erhalt eines dunkelroten Öls. Dieses wurde Kieselgelsäulenchromatographie unterzogen, eluiert mit Essigsäureethylester/Hexan (1:3,5). Die gewünschte rotfarbene Fraktion wurde aus Hexan-Diisopropylether umkristallisiert unter Erhalt eines roten Pulvers der Titelverbindung (380 mg, 54 % Ausbeute).
- Fp. 101,2-101,6ºC.
- ¹H NMR (270 MHz, CDCl&sub3;) δ 7,07-7,41 (m, 6H), 4,40- 4,53 (m, 1H), 3,77 (d, 1H, J=11Hz), 2,05 (s, 3H), 2,04 (s, 3H), 1,29 (d, 3H, J=6,6Hz).

jeweils Wasserstoff bedeuten.
3. Verbindung nach Anspruch 2, wobei R³ einen Rest
der Formel
darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils
Wasserstoff darstellen.
4. Verbindung nach Anspruch 1, wobei R¹ -COR&sup4; oder
-CH(OZ)R&sup8; darstellt, wobei R&sup4; Alkyl mit 1 bis 5
Kohlenstoffatomen bedeutet, R&sup8; Wasserstoff oder Alkyl mit 1 bis 3
Kohlenstoffatomen bedeutet, Z Wasserstoff darstellt und R²
Wasserstoff bedeutet.
5. Verbindung nach Anspruch 4, wobei R¹ -COR&sup4;
darstellt, wobei R&sup4; Methyl ist, und R³ einen Rest der Formel
darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils
Wasserstoff darstellen.
6. Verbindung nach Anspruch 4, wobei R¹ -CH(OZ)R&sup8;
darstellt, wobei R&sup8; Methyl bedeutet und R³ einen Rest der
Formel
darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils
Wasserstoff darstellen.
7. Verbindung nach Anspruch 4, wobei R¹ -CH(OZ)R&sup8;
darstellt, wobei R&sup8; Wasserstoff bedeutet und R³ einen Rest
der Formel
darstellt, worin m 0 bedeutet und R&sup5; und R&sup6; jeweils
Wasserstoff darstellen.
8. Verbindung nach Anspruch 1, wobei R² und R³
zusammengenommen mit dem Stickstoffatom, an das sie gebunden sind,
die Struktur der Formel
bilden.
9. Arzneimittel, umfassend eine Verbindung der Formel
(I) nach einem der Ansprüche 1 bis 9 gemeinsam mit einem
pharmazeutisch verträglichen Träger- oder Verdünnungsmittel.
