Description
- Diese Erfindung betrifft ein 1,2-Ethandiol-Derivat und ein Salz desselben, ein Verfahren zur Herstellung desselben und ein die zerebrale Funktion verbesserndes Mittel, welches dasselbe umfaßt. Das die zerebrale Funktion verbessernde Mittel dieser Erfindung ist zur Behandlung von zerebrovaskulärer Demenz, seniler Demenz, Alzheimer-Demenz, Folgen von ischämischer Enzephalopathie und zerebraler Apoplexie nützlich.
- Als 1,2-Ethandiol-Derivate sind beispielsweise diejenigen bekannt, die im US-Patent Nr. 2,928,645; in J. Pharm. Sci., Band 50, Seiten 769 - 771(1961); Farmaco. Ed. Sci., Band 19, Seiten 1056 - 1065 (1964); usw. beschrieben sind.
- Diese Verbindungen sind als Lokalanästhetika in Verwendung. Jedoch ist nichts bezüglich ihrer Verwendung als ein die zerebrale Funktion verbesserndes Mittel, als antiamnestisches Mittel oder als nootropes Mittel bekannt.
- Die WO Nr. 88/8424 beschreibt, daß 1,2-Ethandiol-Derivate als Arznei für die Alzheimer-Krankheit und andere degenerative neurologische Störungen verwendet werden können. Jedoch ist weder eine spezielle Beschreibung noch ein spezielles Beispiel für diese Derivate in der Anmeldung gegeben.
- Die JP-A-61118304 offenbart 1,2-Ethandiol-Derivate, in denen eine 1,2-Ethandiol-Gruppe direkt mit einem der Kohlenstoffatome eines Pyrazolrings verknüpft ist.
- DE-A-3003323, US-A-4,237,138 und Campbell et al., J. Med. Chem., Band 31 (1988), Seiten 516 - 520 offenbaren Piperidin-Derivate, worin eine 1,2-Ethandiol-Einheit direkt mit einem der Kohlenstoffatome des Piperidinrings verknüpft ist.
- Vodicka et al., Carcinogenesis, Band 9, Nr. 9 (1988), Seiten 1657 - 1660 und Moschel et al., J. Org. Chem., Band 51 (1986), Seiten 2952 - 2955 offenbaren O&sup6;-(2-Hydroxy-2-phenylethyl)desoxyguanosin.
- Winterfeld et al., Archiv der Pharmazie, Band 296 (1963), Seiten 38 - 47 offenbaren 1-Phenyl-2-(piperidin-2-methyloxy)ethanol.
- Baker, J. Org. Chem., Band 50 (1985), Seiten 3942 - 3943 offenbart 1-Phenyl-2-(pyridin-2-yloxy)ethanol.
- Baiocchi et al., Rend. Accad. Nat. 40 (Quaranta) [4] 18 - 19 (1968), Seiten 269 - 280 offenbart 1-Phenyl-2-(1-benzyl-1H-indazol-3- yloxy)ethanol.
- Arzneistoffe wie Verstärker des zerebralen Metabolismus, Zerebrovasodilatatoren und dergleichen sind zur Zeit für die Behandlung verschiedener Demenzien, insbesondere der Demenz vom Alzheimer-Typ und der zerebrovaskulären Demenz, in Verwendung.
- Jedoch ist bis jetzt kein die zerebrale Funktion verbesserndes Mittel gefunden worden, das für die Behandlung zerebrovaskulärer Demenz, seniler Demenz, Alzheimer-Demenz, Folgen von ischämischer Enzephalopathie und zerebraler Apoplexie nützlich ist.
- Ein Ziel dieser Erfindung ist es, ein neues 1,2-Ethandiol-Derivat und ein Salz desselben bereitzustellen.
- Ein anderes Ziel dieser Erfindung ist es, ein Verfahren zur Herstellung eines neuen 1,2-Ethandiol-Derivats und eines Salzes desselben bereitzustellen.
- Noch ein anderes Ziel dieser Erfindung ist es, ein neues die zerebrale Funktion verbesserndes Mittel bereitzustellen, das für die Behandlung von zerebrovaskulärer Demenz, seniler Demenz, Alzheimer-Demenz, Folgen von ischämischer Enzephalopathie und zerebraler Apoplexie nützlich ist und dennoch wenige Nebenwirkungen aufweist.
- Die vorliegenden Erfinder haben Untersuchungen vorgenommen, um die oben erwähnten Probleme zu lösen. Als Ergebnis wurde gefunden, daß ein 1,2-Ethandiol-Derivat, das durch die folgende allgemeine Formel [I] dargestellt wird, oder ein Salz desselben eine ausgezeichnete antiamnestische Wirkung und eine ausgezeichnete antihypoxische Wirkung aufweist und als ein die zerebrale Funktion verbesserndes Mittel sehr nützlich ist:
- in der R¹ eine substituierte oder unsubstituierte Phenylgruppe darstellt; R² ein Wasserstoffatom, eine C&sub1;&submin;&sub6;-Alkylgruppe oder eine Hydroxyl- Schutzgruppe darstellt; R³ ein Wasserstoffatom oder eine C&sub1;&submin;&sub6;-Alkylgruppe darstellt; die nR&sup4; und nR&sup5; gleich oder verschieden voneinander sind und Wasserstoffatome oder C&sub1;&submin;&sub6;-Alkylgruppen darstellen; n eine ganze Zahl von 1 bis 6 darstellt; und R&sup6; eine substituierte oder unsubstituierte stickstoffhaltige heterocyclische Gruppe darstellt, wobei die stickstoffhaltige heterocyclische Gruppe am Kohlenstoffatom durch ein Kohlenstoffatom, das den heterocyclischen Ring bildet, angebracht ist und, wenn n 1 darstellt, aus der Gruppe ausgewählt ist, die aus Pyrrolyl-, Pyrrolidinyl-, Piperazinyl-, Imidazolyl-, Pyridyl-, Tetrahydropyridyl-, Pyrimidinyl-, Morpholinyl-, Thiomorpholinyl-, Chinolyl-, Chinolizinyl-, Tetrahydrochinolinyl-, Tetrahydroisochinolinyl-, Chinuclidinyl-, Thiazolyl-, Tetrazolyl-, Thiadiazolyl-, Pyrrolinyl-, Imidazolinyl-, Imidazolidinyl-, Pyrazolinyl-, Pyrazolidinyl-, Purinyl- und Indazolylgruppen besteht, oder, wenn n eine ganze Zahl von 2 bis 6 darstellt, aus der Gruppe ausgewählt ist, die aus Pyrrolyl-, Pyrrolidinyl-, Piperidyl-, Piperazinyl-, Imidazolyl-, Pyridyl-, Tetrahydropyridyl-, Pyrimidinyl-, Morpholinyl-, Thiomorpholinyl-, Chinolyl-, Chinolizinyl-, Tetrahydrochinolinyl-, Tetrahydroisochinolinyl-, Chinuclidinyl-, Thiazolyl-, Tetrazolyl-, Thiadiazolyl-, Pyrrolinyl-, Imidazolinyl-, Imidazolidinyl-, Pyrazolinyl-, Pyrazolidinyl-, Purinyl- und Indazolylgruppen besteht, worin der Substituent an R¹ aus der Gruppe ausgewählt ist, die aus Halogenatomen, substituierten oder unsubstituierten Amino-, C&sub1;&submin;&sub6;-Alkyl-, Aryl-, Ar-C&sub1;&submin;&sub4;-alkyl-, C&sub1;&submin;&sub6;-Alkoxy-, Ar-C&sub1;&submin;&sub4;-alkoxy-, Aryloxy-, Carbamoyloxy-, C&sub1;&submin;&sub6;-Alkylthio-, C&sub2;&submin;&sub6;-Alkenyl-, C&sub2;&submin;&sub6;-Alkenyloxy-, Ar-C&sub1;&submin;&sub4;-alkylthio-, Ar-C&sub1;&submin;&sub6;-alkylsulfonyl-, Arylsulfonyl-, C&sub1;&submin;&sub6;-Alkylsulfonylamino-, Arylsulfonylamino- und heterocyclischen Gruppen, geschützten Aminogruppen, geschützten oder ungeschützten Hydroxylgruppen, der Nitrogruppe, Oxogruppe und C&sub1;&submin;&sub4;-Alkylendioxygruppen besteht; die substituierte C&sub1;&submin;&sub6;-Alkyl-, Aryl-, Ar-C&sub1;&submin;&sub4;-alkyl-, C&sub1;&submin;&sub6;-Alkoxy-, Ar-C&sub1;&submin;&sub4;- alkoxy-, Aryloxy-, Carbamoyloxy-, C&sub1;&submin;&sub6;-Alkylthio-, C&sub2;&submin;&sub6;-Alkenyl-, C&sub2;&submin;&sub6;- Alkenyloxy-, Ar-C&sub1;&submin;&sub4;-alkylthio-, Ar-C&sub1;&submin;&sub6;-alkylsulfonyl-, Arylsulfonyl-, C&sub1;&submin;&sub6;- Alkylsulfonylamino-, Arylsulfonylamino- oder heterocyclische Gruppe als Substituent von R¹ und die substituierte stickstoffhaltige heterocyclische Gruppe als R&sup6; mindestens einen Substituenten aufweisen, der aus der Gruppe ausgewählt ist, die aus Halogenatomen, geschützten oder ungeschützten Hydroxylgruppen, geschützten oder ungeschützten Aminogruppen, geschützten oder ungeschützten Carboxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-Alkylgruppen, C&sub1;&submin;&sub6;-Alkylgruppen, die durch eine geschützte oder ungeschützte Hydroxylgruppe substituiert sind, unsubstituierten oder Halogensubstituierten Arylgruppen, unsubstituierten oder Halogen-substituierten Aroylgruppen, unsubstituierten C&sub1;&submin;&sub6;-Alkoxygruppen, C&sub1;&submin;&sub6;-Alkoxygruppen, die durch eine C&sub1;&submin;&sub6;-Alkoxygruppe substituiert sind, C&sub1;&submin;&sub6;-Acylgruppen, Ar-C&sub1;&submin;&sub4;- alkylgruppen, Ar-C&sub2;&submin;&sub6;-alkenylgruppen, heterocyclischen Gruppen, Heterocyclus-CO-Gruppen, Oxogruppen, C&sub1;&submin;&sub6;-Alkylsulfonylgruppen und Arylsulfonylgruppen besteht; und die substituierte Aminogruppe als Substituent von R¹ mindestens jeweils einen Substituenten aufweist, der aus der Gruppe ausgewählt ist, die aus geschützten oder ungeschützten Hydroxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-Alkylgruppen, C&sub1;&submin;&sub6;-Alkylgruppen, die durch eine geschützte oder ungeschützte Carboxyl- oder Hydroxylgruppe substituiert sind, Cycloalkylgruppen, Arylgruppen, C&sub1;&submin;&sub6;-Acylgruppen, Ar- C&sub1;&submin;&sub4;-alkylgruppen, heterocyclischen Gruppen, unsubstituierten oder Oxosubstituierten Heterocyclus-CO-Gruppen, der Adamantylgruppe, C&sub1;&submin;&sub6;- Alkylsulfonylgruppen und Arylsulfonylgruppen besteht; wobei alle obigen heterocyclischen Gruppen aus der Gruppe ausgewählt sind, die aus den in der Definition von R&sup6; erwähnten stickstoffhaltigen heterocyclischen Gruppen und Furyl-, Thienyl-, Benzothienyl-, Pyranyl-, Isobenzofuranyl-, Oxazolyl-, Benzofuranyl-, Indolyl-, Benzimidazolyl-, Benzoxazolyl-, Benzothiazolyl-, Chinoxalyl-, Dihydrochinoxalyl-, 2,3-Dihydrobenzothienyl-, 2,3-Dihydrobenzopyrrolyl-, 2,3-Dihydro-4H-1-thianaphthyl-, 2,3- Dihydrobenzofuranyl-, Benzo[b]dioxanyl-, Imidazo[2,3-a]pyridyl-, Benzo[b]piperazinyl-, Chromenyl-, Isothiazolyl-, Isoxazolyl-, Oxadiazolyl-, Pyridazinyl-, Isoindolyl- und Isochinolylgruppen besteht.
- Im übrigen bezieht sich das das hierin erwähnte die zerebrale Funktion verbessernde Mittel auf ein die zerebrale Funktion verbesserndes Mittel, das nicht nur die von herkömmlichen, die zerebrale Funktion verbessernden Mitteln besessene Wirkungen, beispielsweise bei Folgen von ischämischer Enzephalopathie und zerebraler Apoplexie, sondern auch therapeutische und prophylaktische Wirkungen für Amnesie und Demenzien (z.B. zerebrovaskuläre Demenz, senile Demenz und Alzheimer-Demenz) aufweist.
- In der vorliegenden Beschreibung haben die folgenden Ausdrücke die folgenden Definitionen, falls nicht anders angegeben.

- Die Schutzgruppen für die Hydroxylgruppe, Carboxylgruppe und Aminogruppe schließen die üblichen Schutzgruppen für die Hydroxylgruppe, Carboxylgruppe und Aminogruppe ein, die in Protective Groups in Organic Synthesis [Theodra W. Greene (1981), John Wiley & Sons, Inc.] beschrieben sind. Insbesondere schließt die Schutzgruppe für die Hydroxylgruppe beispielsweise Niederalkyl-, Niederacyl-, Tetrahydropyranyl- und Arniederalkylgruppen, wie substituiertes oder unsubstituiertes Benzyl, ein.
- Das Salz des 1,2-Ethandiol-Derivats, das durch die allgemeine Formel [I] dargestellt wird, kann irgendein pharmazeutisch annehmbares Salz sein. Es schließt beispielsweise Salze mit Mineralsäuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure und dergleichen; Salze mit Carbonsäuren, wie Ameisensäure, Essigsäure, Oxalsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Asparaginsäure und dergleichen; Salze mit Sulfonsäuren, wie Methansulfonsäure, Benzolsulfonsäure, p-Toluolsulfonsäure, Naphthalinsulfonsäure und dergleichen; und Salze mit Alkalimetallen, wie Natrium, Kalium und dergleichen, ein.
- Wenn das 1,2-Ethandiol-Derivat der allgemeinen Formel [I] oder dessen Salz Isomere aufweist (z.B. optische Isomere, geometrische Isomere, Tautomere), sind alle diese Isomere in dieser Erfindung eingeschlossen. Auch das Hydrat, Solvat und alle Kristallformen der Verbindung dieser Erfindung sind in dieser Erfindung eingeschlossen.
- Als nächstes wird ein Verfahren zur Herstellung eines 1,2-Ethandiol- Derivats der allgemeinen Formel [I] oder eines Salzes desselben beschrieben.
- Das 1,2-Ethandiol-Derivat der allgemeinen Formel [I] oder sein Salz kann durch ansich bekannte Verfahren oder deren geeignete Kombinationen, beispielsweise die folgenden Herstellungsverfahren, hergestellt werden. Herstellungsverfahren 1 oder dessen Salz
- In den obigen Reaktionsschemata haben R¹, R³, R&sup4;, R&sup5;, R&sup6; und n die gleichen Bedeutungen wie oben definiert; und m stellt eine ganze Zahl von 1 - 6 dar.
- Als Salze der Verbindungen der allgemeinen Formeln [III], [IIIa] und [Ia] können die gleichen Salze wie die Salze der Verbindung der allgemeinen Formel [I] erwähnt werden.
- Als nächstes wird jedes der obigen Herstellungsverfahren beschrieben.
- Eine Verbindung der allgemeinen Formel [II] wird mit einer Verbindung der allgemeinen Formel [III] oder deren Salz oder einer Verbindung der allgemeinen Formel [IIIa] oder deren Salz in Anwesenheit oder Abwesenheit einer Base umgesetzt, um eine Verbindung der allgemeinen Formel [Ia] oder deren Salz zu erhalten.
- Bei dem in dieser Reaktion zu verwendenden Lösungsmittel kann es sich um irgendein Lösungsmittel handeln, solange es die Reaktion nicht nachteilig beeinflußt. Beispielsweise können aromatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylol und dergleichen; Sulfoxide, wie Dimethylsulfoxid und dergleichen; Amide, wie N,N-Dimethylformamid und dergleichen; und Ether, wie Tetrahydrofuran, Dioxan und dergleichen, erwähnt werden. Diese Lösungsmittel können allein oder in Mischung von zwei oder mehr verwendet werden. Es ist auch möglich, die Verbindung der allgemeinen Formel [III] oder [IIIa] als das Lösungsmittel zu verwenden.
- Als gegebenenfalls zu verwendende Base können beispielsweise Natriumhydrid, metallisches Natrium und Kalium-tert-butanolat erwähnt werden.
- In der Reaktion wird die Verbindung der allgemeinen Formel [III] oder deren Salz oder die Verbindung der allgemeinen Formel [IIIa] oder deren Salz in einer Menge von 1 - 100 Mol, vorzugsweise 1 - 10 Mol, pro Mol der Verbindung der allgemeinen Formel [II] verwendet.
- Die Base als fakultative Komponente wird in einer Menge von 0,01 - 1,2 Mol pro Mol der Verbindung der allgemeinen Formel [II] verwendet.
- Diese Reaktion kann gewöhnlich bei 20 - 150ºC, vorzugsweise 70 - 90ºC, über 1 Minute bis 24 Stunden, vorzugsweise 5 Minuten bis 5 Stunden, durchgeführt werden.
- Bei dem obigen Herstellungsverfahren können, wenn die Verbindungen der allgemeinen Formeln [II], [III] und [IIIa] Isomere aufweisen (z.B. optische Isomere, geometrische Isomere, Tautomere), die Verbindungen in irgendeiner Isomerform verwendet werden. Weiter können die Verbindungen in Hydratform, Solvatform oder jeder Kristallform verwendet werden.
- Wenn die Verbindungen der allgemeinen Formeln [II], [III], [IIIa], [I] und [Ia] eine Hydroxylgruppe, eine Aminogruppe oder eine Carboxylgruppe aufweisen, ist es möglich, zuvor diese Gruppen mit einer üblichen Schutzgruppe zu schützen und nach der Reaktion die Schutzgruppe, falls benötigt, gemäß an sich bekannten Verfahren zu entfernen.
- Die so erhaltene Verbindung kann üblichen Isolierungs- und Reinigungsverfahren, wie Säulenchromatographie, Kristallisation, Destillation, Extraktion und dergleichen, unterzogen werden.
- Das 1,2-Ethandiol-Derivat der allgemeinen Formel [I] oder sein Salz kann in ein anderes 1,2-Ethandiol-Derivat der allgemeinen Formel [I] oder dessen Salz überführt werden, indem man die erstgenannte Verbindung einer geeigneten Kombination von an sich bekannten Reaktionen, wie Oxidationsreaktion, Reduktionsreaktion, Additionsreaktion, Acylierungsreaktion, Alkylierungsreaktion, Sulfonylierungsreaktion, Desacylierungsreaktion, Substitutionsreaktion, Dehydratisierungsreaktion, Hydrolysereaktion und dergleichen, unterzieht.
- Die Verbindung der allgemeinen Formel [II], die das Ausgangsmaterial für die Herstellung der Verbindung dieser Erfindung ist, kann durch an sich bekannte Verfahren hergestellt werden, beispielsweise durch das Verfahren, das in JACS, Band 87, S. 1353 (1965) beschrieben ist, und durch das Verfahren, das in Shin Jikken Kagaku Koza, Band 14, S. 579 (1977), Maruzen, beschrieben ist.
- Die Verbindung dieser Erfindung kann, wenn sie als Arzneistoff verwendet wird, geeignet mit Hilfsstoffen, wie einem Füllstoff, einem Trägerstoff, einem Verdünnungsmittel und dergleichen, gemischt werden und gemäß herkömmlichen Verfahren zu Tabletten, Kapseln, Pulvern, Granula, feinen Granula, Pillen, Suspensionen, Emulsionen, Flüssigkeiten, Sirupen, Injektionen usw. gebildet werden. Diese Arzneistoffe können oral oder parenteral verabreicht werden. Der Dosierungsweg, die Dosis und die Anzahl der Verabreichungen können abhängig vom Alter, Gewicht und Symptom des Patienten geeignet variiert werden, aber im Fall der oralen Verabreichung können einem erwachsenen Patienten im allgemeinen 0,01 - 500 mg der vorliegenden Verbindung täglich in einer bis mehreren Portionen verabreicht werden.
- Als nächstes werden die pharmakologischen Wirkungen von repräsentativen Verbindungen dieser Erfindung beschrieben.
- Die Nummern der Testverbindungen, die in den folgenden pharmakologischen Tests verwendet werden, beziehen sich auf die Nummern der Verbindungen, die in den nachstehend aufgeführten Herstellungsbeispielen angegeben sind.
- Eine Testverbindung, die in physiologischer Kochsalzlösung (100 mg/kg) gelöst war, wurde oral weiblichen ddY-Mäuse (5 - 6 Wochen alt, jede Gruppe bestand aus 10 Mäusen) verabreicht. Eine Stunde (oder 30 Minuten*) nach der Verabreichung wurde jede Maus in eine 300 ml-Glaskammer gegeben, und eine Gasmischung, die aus 4% Sauerstoff und 96% Stickstoff bestand, wurde mit einem Durchsatz von 5 Litern/Min. durch die Kammer geleitet. Die Zeit vom Beginn des Gasdurchleitens bis zum Tod jeder Maus wurde gemessen.
- Einer Kontroll-Mäusegruppe wurde nur physiologische Kochsalzlösung oral verabreicht.
- Die antihypoxische Wirkung der Testverbindung wurde aus der folgenden Formel berechnet:
- Mittlere Überlebenszeit von Mäusen der Verabreichungsgruppe / Mittlere Überlebenszeit von Mäusen der Kontrollgruppe x 100 (%)
- Die Ergebnisse sind in Tabelle 1 aufgeführt. Tabelle 1
- Fußnote: * Mäuse wurden 30 Minuten nach Verabreichung anstelle von 1 Std. nach Verabreichung in die Kammer gegeben.
- Eine in physiologischer Kochsalzlösung gelöste Testverbindung wurde intraperitoneal einer männlichen ddY-Maus (5 - 6 Wochen alt, jede Gruppe bestand aus 10 Mäusen) verabreicht. Der Versuch zum Erlernen einer Aufgabe des passiven Vermeidens wurde 1 Stunde nach der Verabreichung durchgeführt. Jede Maus wurde in das helle Abteil einer Apparatur mit zwei Abteilen zur passiven Vermeidung des Durchtretens, die aus einem hellen Abteil und einem dunklen Abteil bestand (MPA-100M, hergestellt von Muromachi Kikai) gesetzt. Als die Maus das dunkle Abteil betrat, wurde die Falltür des dunklen Abteils geschlossen; nach 0,5 Sekunden wurde ein unvermeidbarer Fußschock (1,6 mA, 3 Sekunden) verabreicht. Unmittelbar danach wurde durch die beiden Augen der Maus ein ECS (25 mA, 0,5 Sekunden) angewendet. Nach 24 Stunden wurde im Merkversuch die Maus wiederum in das helle Abteil gesetzt, und die Antwortlatenz der Maus, das dunkle Abteil zu betreten, wurde gemessen. Wenn die Maus dies länger als 300 Sekunden vermied, wurde ihr eine Höchstbewertung von 300 Sekunden zugerechnet.
- Bei einer Kontroll-Mäusegruppe, der nur physiologische Kochsalzlösung intraperitoneal verabreicht worden war, wurde die Antwortlatenz ebenfalls auf die gleiche Weise gemessen.
- Die antiamnestische Wirkung wurde als Mittelwert der Antwortlatenzen der 10 Mäuse genommen und durch die folgenden Symbole ausgedrückt:
- -: 0 - 60 Sekunden
- +: 61 - 100 Sekunden
- ++: 101 - 150 Sekunden
- +++: 151 - 300 Sekunden
- Die Ergebnisse sind in Tabelle 2 aufgeführt. Tabelle 2
- Es wurde von Yamazaki et al. [Drugs, Mind and Action, Band 3, Seiten 127 - 136 (1983)] berichtet, daß Cycloheximid den Zugriffsprozeß auf das Gedächtnis einer Maus stört. Demgemäß wurde der folgende Test durchgeführt.
- Ein Test wurde gemäß dem in Drugs, Mind and Action, Band 3, Seiten 127 - 136 (1983) und Folia Pharmacologica Japonica, Band 89, Seiten 243 - 252 (1987) beschriebenen Verfahren durchgeführt.
- Als Testapparatur wurde ein Trainingskasten vom Typ der passiven Vermeidung des Hinuntersteigens verwendet. Es handelte sich um einen schwarzen Acrylharzkasten mit 22 cm x 22 cm x 21 x cm (Höhe), dessen Bodenteil aus einem Gitter aus rostfreiem Stahl bestand und der in einer Ecke des Bodengitters eine Plattform mit 7 cm x 7 cm x 2 cm (Höhe) aufwies.
- Cycloheximid wurde in physiologischer Kochsalzlösung gelöst und subkutan mit 120 mg/kg einer männlichen ddY-Maus (5 - 6 Wochen alt, jede Gruppe bestand aus 10 Mäusen) injiziert. In einem Lernversuch wurde jede Maus 15 Minuten nach der Verabreichung auf die Plattform der obigen Testapparatur gesetzt. Sobald die Maus von der Plattform hinunterstieg, wurde 2 Sekunden lang ein 2 mA-Strom verabreicht, nach dem die Maus unmittelbar in ihren Heimkäfig zurückgebracht wurde. Ein Merktest wurde 24 Stunden nach dem Lernen durchgeführt. Jeder Maus, die mit Cycloheximid behandelt worden war, wurde oral eine in physiologischer Kochsalzlösung gelöste Testverbindung verabreicht; 30 Minuten nach der Verabreichung wurde die Maus wiederum auf die Plattform gesetzt, und die Antwortlatenz der Maus, hinunterzusteigen, wurde gemessen. Wenn die Maus dies länger als 300 Sekunden vermied, wurde ihr eine Höchstbewertung von 300 Sekunden zugerechnet.
- Eine Kontroll-Mäusegruppe, der nur physiologische Kochsalzlösung oral verabreicht worden war, wurde ebenfalls der Messung der Antwortlatenz auf die gleiche Weise unterzogen.
- Die antiamnestische Wirkung wurde als Mittelwert der Antwortlatenzen der 10 Mäuse genommen und durch die folgenden Symbole ausgedrückt:
- -: 0 - 60 Sekunden
- +: 61 - 100 Sekunden
- ++: 101 - 150 Sekunden
- +++: 151 - 300 Sekunden
- Die Ergebnisse sind in Tabelle 3 aufgeführt. Tabelle 3
- Ein Test wurde gemäß dem Verfahren von Ellman et al. [Biochem. Pharmacol., Band 7, Seiten 88 - 95, 1961] durchgeführt.
- Das heißt, Acetylcholin (als Substrat) wurde zu Phosphatpuffer- Lösung gegeben, die 5,5'-Dithiobis-(2-nitrobenzoesäure) (DTNB), eine Testverbindung und ein Mäusehimhomogenisat (als Acetylcholinesterase- Quelle) enthielt. Die resultierende Mischung wurde inkubiert, und die Menge an resultierender 5-Thio-2-nitrobenzoesäure wurde photometrisch bei 412 nm gemessen.
- Die Hemmwirkung der Testverbindung auf Acetylcholinesterase wurde durch die Hemmung (%) ausgedrückt, als die Endkonzentration der Testverbindung 10 µg/ml betrug.
- Die Ergebnisse sind in Tabelle 4 aufgeführt. Tabelle 4
- Eine in physiologischer Kochsalzlösung gelöste Testverbindung wurde intravenös einer Gruppe von 3 männlichen ddY-Mäusen (5 - 6 Wochen alt) verabreicht, um die akute Toxizität der Testverbindung zu überprüfen.
- Als Ergebnis ergaben die Testverbindungen Nr. 119, 120, 126, 132, 147, 151, 176, 219, 220 und 221 keinen Todesfall bei 50 mg/kg.
- Es wird aus den obigen Testergebnissen leicht erkannt, daß die Verbindung dieser Erfindung eine ausgezeichnete antihypoxische Wirkung, antiamnestische Wirkung und Hemmwirkung auf Acetylcholinesterase aufweist und eine niedrige Toxizität besitzt.
- Aus den obigen Ergebnissen wird auch leicht erkannt, daß das die zerebrale Funktion verbessernde Mittel der Erfindung für die Behandlung von zerebrovaskulärer Demenz, seniler Demenz, Alzheimer-Demenz, Folgen von ischämischer Enzephalopathie und zerebraler Apoplexie nützlich ist.
- Als nächstes wird das Verfahren zur Herstellung der Verbindung dieser Erfindung speziell mit Herstellungsbeispielen beschrieben.
- In den Herstellungsbeispielen ist das Mischungsverhältnis von Eluens in allen Fällen auf Volumen bezogen, und als Träger bei der Säulenchromatographie wurde ein Kieselgel (Kieselgel 60, Art. 7734), hergestellt von Merck Co., verwendet.
- Die in den Herstellungsbeispielen verwendeten Abkürzungen weisen die folgenden Bedeutungen auf:
- Me: Methyl, Et: Ethyl, i-Pr: Isopropyl, t-Bu: tert-Butyl, Ac: Acetyl, Ph: Phenyl, DPM: Diphenylmethyl, Bz: Benzyl, Tr: Trityl, IPA: Isopropylalkohol, IPE: Diisopropylether, PTS: p-Toluolsulfonsäure.
- In den folgenden Sätzen und Tabellen beziehen sich die Substanzen in [ ] auf Lösungsmittel, die bei der Umkristallisation verwendet wurden. Herstellungsbeispiel 1
- (1) Eine Mischung von 10,8 g (L)-Benzoylcystin, 1,6 g Lithiumborhydrid, 2,4 g tert-Butanol und 180 ml Tetrahydrofuran wurde 1 Stunde refluxiert und dann auf -60ºC abgekühlt. Dazu wurden 6,1 g 4- Benzyloxyphenacylbromid gegeben. Die Mischung wurde 30 Minuten bei der gleichen Temperatur und weiter 3 Stunden bei -40 bis -30ºC gerührt. Die Reaktionsmischung wurde zu einer Mischung von 100 ml Wasser und 200 ml Diethylether gegeben. Die organische Schicht wurde abgetrennt, mit Wasser, einer gesättigten wäßrigen Natriumhydrogencarbonat-Lösung und einer gesättigten wäßrigen Natriumchlorid-Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Toluol) gereinigt, wodurch man 2,8 g (S)-1-(4-Benzyloxyphenyl)-2-bromethanol erhielt.
- (2) 2,8 g (S)-1-(4-Benzyloxyphenyl)-2-bromethanol wurden in einer Lösungsmittelmischung von 20 ml Methanol und 10 ml Tetrahydrofuran gelöst. Zu der Lösung wurde unter Eiskühlung eine Lösung von 0,8 g Kaliumhydroxid, gelöst in 4 ml Wasser, gegeben. Die Mischung wurde 5 Minuten bei der gleichen Temperatur und weiter 10 Minuten bei Raumtemperatur gerührt. Die Reaktionsmischung wurde zu einer Mischung von 50 ml Diethylether und 50 ml Eiswasser gegeben. Die organische Schicht wurde abgetrennt. Die wäßrige Schicht wurde mit 25 ml Diethylether extrahiert. Der Extrakt wurde mit der zuvor abgetrennten organischen Schicht vereinigt. Die vereinigte organische Schicht wurde mit Wasser und einer gesättigten wäßrigen Natriumchlorid-Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man 1,4 g (S)-2-(4- Benzyloxyphenyl)oxiran erhielt.
- Schmelzpunkt: 56 - 61ºC
- Optische Drehung: [α]²&sup5;D = +5,2º (C = 2, CHCl&sub3;)
- Die folgenden Verbindungen wurden auf die gleiche Weise erhalten.
- - (S)-2-(3-Methylphenyl)oxiran
- [α]²&sup5;D = +15,7º (C 2, CHCl&sub3;)
- - (S)-2-(4-Phenoxyphenyl)oxiran
- [α]²&sup5;D = +12,5º (C = 4, CHCl&sub3;).
- (1) Eine Lösung 23 g (+)-Diisopinocamphenylchlorboran, gelöst in 30 ml Tetrahydrofuran, wurde auf -25ºC abgeköhlt. Dazu wurden 12 g 4- Benzyloxyphenacylbromid gegeben. Die resultierende Mischung wurde 4 Stunden bei -20 bis -15ºC gerührt. Die Reaktionsmischung wurde zu einer Mischung von 150 ml Diethylether und 100 ml Eiswasser gegeben. Die organische Schicht wurde abgetrennt, mit Wasser und einer gesättigen wäßrigen Natriumchlorid-Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Hexan:Toluol = 1:2) gereinigt, wodurch man 6,8 g (R)-1-(4-Benzyloxyphenyl)-2-bromethanol erhielt. (2) 6,0 g (R)-1-(4-Benzyloxyphenyl)-2-bromethanol wurden in einer Lösungsmittelmischung von 50 ml Methanol und 25 ml Tetrahydrofuran gelöst. Dazu wurde unter Eisköhlung eine Lösung von 1,5 g Kaliumhydroxid, gelöst in 5 ml Wasser, gegeben. Die resultierende Mischung wurde 5 Minuten bei der gleichen Temperatur und weiter 10 Minuten bei Raumtemperatur gerührt. Die Reaktionsmischung wurde zu einer Mischung von 100 ml Diethylether und 100 ml Eiswasser gegeben. Die organische Schicht wurde abgetrennt. Die wäßrige Schicht wurde mit 50 ml Diethylether extrahiert. Der Extrakt wurde mit der zuvor abgetrennten organischen Schicht vereinigt. Die vereinigte organische Schicht wurde mit Wasser und einer gesättigten wäßrigen Natriumchlorid-Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man 3,7 g (R)-2-(4- Benzyloxyphenyl)oxiran erhielt.
- Schmelzpunkt: 58 - 67ºC
- Optische Drehung: [α]²&sup6;D = -14,4º (C = 2, CHCl&sub3;)
- Die folgenden Verbindungen wurden auf die gleiche Weise erhalten.
- - (R)-2-(3-Methylphenyl)oxiran
- [α]²&sup7;D = -18,6º (C = 2, CHCl&sub3;)
- - (R)-2-(4-Phenoxyphenyl)oxiran
- [α]²&sup5;D = -11,3º (C = 2, CHCl&sub3;).
- 3,4 g Kalium-tert-butanolat wurden zu 31 ml 2-(N,N- Dimethylamino)ethanol gegeben. Die resultierende Mischung wurde auf 80ºC erwärmt. Dazu wurden tropfenweise über 40 Minuten 7,7 g 2-(3- Fluorphenyl)oxiran gegeben. Die Mischung wurde 3 Stunden bei der gleichen Temperatur gerührt. Die Reaktionsmischung wurde zu einer Mischung von 200 ml Eiswasser und 200 ml Ethylacetat gegeben. Die organische Schicht wurde abgetrennt. Zu der organischen Schicht wurden 50 ml Wasser gegeben. Die Mischung wurde mit 6 N Salzsäure auf pH 1 eingestellt. Die wäßrige Schicht wurde abgetrennt und mit 100 ml Chloroform gemischt. Die resultierende Mischung wurde mit einer 2 N wäßrigen Natriumhydroxid-Lösung auf pH 11 eingestellt. Die organische Schicht wurde abgetrennt, mit Wasser gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Ethanol = 5/1) gereinigt. Das resultierende ölige Produkt wurde in 25 ml Aceton gelöst. Chlorwasserstoffgas wurde in die Lösung eingeleitet. Die resultierenden Kristalle wurden durch Filtration gesammelt, mit Aceton gewaschen und getrocknet, wodurch man 3,1 g 2-[2-(N,N- Dimethylamino)ethoxy]-1-(3-fluorphenyl)ethanolhydrochlorid erhielt.
- Schmelzpunkt: 164 - 165ºC
- Die in Tabelle 5 aufgeführten Verbindungen wurden auf die gleiche Weise erhalten.
- In Tabelle 5 zeigen R¹, R², R³, R4a, R4b, R&sup6;, na und nb jeweils einen Substituenten oder eine ganze Zahl an, der bzw. die in der folgenden Formel verwendet werden. Tabelle 5
- Eine Mischung von 5,00 g 1-Benzyl-4-hydroxypiperidin, 1,97 g Kaliumtert-butanolat und 4 ml Dimethylsulfoxid wurde auf 80ºC erwärmt. Dazu wurden tropfenweise über 40 Minuten 2,10 g Styroloxid gegeben. Die resultierende Mischung wurde 3 Stunden bei der gleichen Temperatur gerührt. Die Reaktionsmischung wurde zu einer Mischung von 100 ml Eiswasser und 80 ml Ethylacetat gegeben. Die Mischung wurde mit 6 N Salzsäure auf pH 11,5 eingestellt. Die organische Schicht wurde abgetrennt, mit Wasser und einer gesättigten wäßrigen Natriumchlorid- Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Ethanol = 10/1) gereinigt. Das resultierende ölige Produkt wurde in 8 ml Ethanol gelöst. Zu der Lösung wurden 1 ml einer trockenen 6 N Chlorwassertoff-Ethanol-Lösung und 20 ml Diethylether gegeben. Die Mischung wurde 30 Minuten bei Raumtemperatur gerührt. Die resultierenden Kristalle wurden durch Filtration gesammelt, mit einer Mischung von 2 ml Ethanol und 2 ml Diethylether gewaschen und getrocknet, wodurch man 930 mg 2-(1-Benzylpiperidin-4-yloxy)-1- phenylethanolhydrochlorid erhielt.
- Schmelzpunkt: 193 - 195ºC
- Die in Tabelle 6 aufgeführten Verbindungen wurden auf die gleiche Weise erhalten.

- (1) Eine Mischung von 4,30 g 4-Benzyl-2-hydroxymethylmorpholin, 930 mg Kalium-tert-butanolat und 4 ml Dimethylsulfoxid wurde auf 80ºC erwärmt. Dazu wurden tropfenweise über 20 Minuten 2,50 g Styroloxid gegeben. Die resultierende Mischung wurde 2 Stunden bei der gleichen Temperatur gerührt. Die Reaktionsmischung wurde zu einer Mischung von 30 ml Diethylether und 30 ml Eiswasser gegeben. Die organische Schicht wurde abgetrennt und mit 10 ml Wasser gemischt. Die Mischung wurde mit 6 N Salzsäure auf pH 2,0 eingestellt. Die wäßrige Schicht wurde abgetrennt und mit 30 ml Diethylether gemischt. Die Mischung wurde mit einer 2 N wäßrigen Natriumhydroxid-Lösung auf pH 11 eingestellt. Die organische Schicht wurde abgetrennt, mit einer gesättigten wäßrigen Natriumchlorid-Lösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der Rückstand wurde durch Säulenchromatographie (Eluens: Toluol/Ethylacetat = 1/1) gereinigt, wodurch man 2,00 g öliges 1-Phenyl-2-[(4-benzylmorpholin-2- yl)methoxy]ethanol erhielt (Verbindung Nr. 144).
- Die in Tabelle 7 aufgeführten Verbindungen wurden auf die gleiche Weise erhalten.
- In Tabelle 7 zeigen R¹, R², R³, R&sup4;, R&sup6; und n jeweils einen Substituenten oder eine ganze Zahl an, der bzw. die in der folgenden Formel verwendet werden: Tabelle 7 Tabelle 7 (Forts.)
- (2) Eine Mischung von 1,00 g 1-Phenyl-2-[(4-benzylmorpholin-2- yl)methoxy]ethanol, 500 mg 5% Palladium-Kohle und 10 ml Methanol wurde 4 Stunden bei Raumtemperatur unter Atmosphärendruck einer Hydrierung unterzogen. Nach der Beendigung der Reaktion wurde Palladium-Kohle durch Filtration entfernt. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Methanol = 10/1) gereinigt, wodurch man 450 mg eines öligen Produktes erhielt. Das ölige Produkt wurde in 2,5 ml Isopropanol gelöst. Die Lösung wurde mit 220 mg Fumarsäure gemischt. Die Mischung wurde 1 Stunde bei Raumtemperatur gerührt. Die resultierenden Kristalle wurden durch Filtration gesammelt, mit 2 ml Isopropanol gewaschen und getrocknet, wodurch man 460 mg 1/2 Fumarat von 1-Phenyl-2-[(morpholin-2-yl)methoxy]ethanol erhielt (Verbindung Nr. 151).
- Schmelzpunkt: 146,5 - 147,0ºC [EtOH]
- Die in Tabelle 8 aufgeführten Verbindungen wurden auf die gleiche Weise erhalten.
- In Tabelle 8 zeigen R¹, R², R³, R&sup4;, R&sup6; und n jeweils einen Substituenten oder eine ganze Zahl an, der bzw. die in der folgenden Formel verwendet werden: Tabelle 8
- (3) 7 g 1-(3-Methoxyphenyl)-2-[(4-tritylmorpholin-2- yl)methoxy]ethanol wurden in 35 ml Aceton gelöst. Zu der Lösung wurden unter Eiskühlung 2,6 ml einer trockenen 5,9 N Chlorwasserstoff-Ethanol- Lösung gegeben. Die Mischung wurde 2 Stunden bei Raumtemperatur gerührt. Nach Beendigung der Reaktion wurde das Lösungsmittel durch Destillation unter reduziertem Druck entfernt. Zu dem so erhaltenen Rückstand wurden 50 ml Wasser und 30 ml Ethylacetat gegeben. Die wäßrige Schicht wurde abgetrennt und mit Ethylacetat gewaschen. Dazu wurden 50 ml Chloroform gegeben. Die resultierende Mischung wurde mit einer 1 N wäßrigen Natriumhydroxid-Lösung auf pH 11 eingestellt. Die organische Schicht wurde abgetrennt und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Methanol = 5/1) gereinigt, wodurch man 1 g eines öligen Produktes erhielt. Das ölige Produkt wurde in 3 ml Isopropanol gelöst. Dazu wurden 430 ml Fumarsäure gegeben. Die Mischung wurde 1 Stunde bei Raumtemperatur gerührt. Die resultierenden Kristalle wurden durch Filtration gesammelt und getrocknet, wodurch man 800 mg 1/2 Fumarat von 1-(3-Methoxyphenyl)-2-[(morpholin-2-yl)methoxy]ethanol erhielt (Verbindung Nr. 155).
- Schmelzpunkt: 122 - 123,5ºC
- Die in Tabelle 9 aufgeführten Verbindungen wurden auf die gleiche Weise erhalten.
- In Tabelle 9 zeigen R¹, R², R³, R&sup4;, R&sup6; und n jeweils einen Substituenten oder eine ganze Zahl an, der bzw. die in der folgenden Formel verwendet werden: Tabelle 9
- Eine Mischung von 2,0 g 1-(4-Benzyloxyphenyl)-2-[2-(N,N- dimethylamino)ethoxy]ethanolhydrochlorid, 500 mg 10% Palladium-Kohle und 10 ml Methanol wurde 2 Stunden bei Raumtemperatur unter atmosphärischern Druck einer Hydrierung unterzogen. Nach Beendigung der Reaktion wurde Palladium-Kohle durch Filtration entfernt. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man 1,4 g 1-(4- Hydroxyphenyl)-2-[2-(N,N-dimethylamino)ethoxy]ethanolhydrochlorid erhielt.
- Schmelzpunkt: 169,5 - 170,5ºC
- Die in Tabelle 10 erhaltenen Verbindungen wurden auf die gleiche Weise erhalten.
- In Tabelle 10 zeigen R¹, R², R³, R&sup4;, R&sup6; und n jeweils einen Substituenten oder eine ganze Zahl an, der bzw. die in der folgenden Formel verwendet werden: Tabelle 10
- 0,65 ml Pyridin und 0,99 ml Acetanhydrid wurden zu 1 g 1-(3- Methylphenyl)-2-[2-(morpholin-4-yl)ethoxy]ethanol gegeben. Die Mischung wurde 3 Stunden bei Raumtemperatur gerührt. Die Reaktionsmischung wurde einer Destillation unter reduziertem Druck unterzogen, um das Lösungsmittel zu entfernen. Zu dem Rückstand wurden 20 ml Ethylacetat und 10 ml Wasser gegeben. Die Mischung wurde mit Kaliumcarbonat auf pH 10 eingestellt. Die organische Schicht wurde abgetrennt, mit Wasser und einer gesättigten wäßrigen Natriumchlorid-Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Ethanol = 30/1) gereinigt, wodurch man 1 g öliges 1-Acetoxy-1-(3-methylphenyl)-2-[2-(morpholin-4- yl)ethoxy]ethan erhielt.
- Die in Tabelle 11 aufgeführte Verbindung wurde auf die gleiche Weise erhalten.
- In Tabelle 11 zeigen R¹, R², R³, R&sup4;, R&sup6; und n jeweils einen Substituenten oder eine ganze Zahl, der bzw. die in der folgenden Formel verwendet werden: Tabelle 11
- Eine Mischung von 500 mg 2-[(Imidazol-4-yl)methoxy]-1-phenylethanol, 1,1 ml Pyridin, 1,1 ml Triethylamin und 1,1 ml Acetanhydrid wurde 1 Stunde bei 100ºC gerührt. Die Reaktionsmischung wurde auf Raumtemperatur abgekühlt. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Zu dem Rückstand wurden 1,1 ml Methyliodid und 5 ml Acetonitril gegeben. Man ließ die Mischung 24 Stunden bei Raumtemperatur stehen. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Zu dem so erhaltenen Rückstand wurden 4 ml Ethanol und 6,8 ml einer 5%-igen wäßrigen Natriumhydroxid-Lösung gegeben. Die resultierende Mischung wurde 6 Stunden bei Raumtemperatur gerührt. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Zu dem so erhaltenen Rückstand wurden 30 ml Chloroform und 20 ml Wasser gegeben. Die organische Schicht wurde abgetrennt, mit Wasser gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Das resultierende ölige Produkt wurde durch Säulenchromatographie (Eluens: Chloroform/Ethanol = 10/1) gereinigt. Dusopropylether wurde zu den resultierenden weißen Kristallen dazugegeben, und die resultierende Mischung wurde filtriert, wodurch man 450 mg 2-[(1-Methylimidazol-5-yl)methoxy]-1-phenylethanol erhielt (Verbindung Nr. 214).
- Schmelzpunkt: 102 - 105ºC
- Die folgende Verbindung wurde auf die gleiche Weise erhalten.
- 1-(4-Benzyloxyphenyl)-2-[(1-methylimidazol-5-yl)methoxy]ethanol (Verbindung Nr. 215)
- Schmelzpunkt: 148,5 - 150,5ºC [IPA-AcOEt]
- (1) 5 g (S)-4-Benzyl-2-acetoxymethylmorpholin wurden in 10 ml Ethanol gelöst. Zu der Lösung wurde unter Eiskühlung eine Lösung von 1 g Natriumhydroxid, gelöst in 5 ml Wasser, gegeben. Die Mischung wurde 10 Minuten bei Raumtemperatur gerührt. Die Reaktionsmischung wurde zu 50 ml Eiswasser gegeben und mit 50 ml Chloroform extrahiert. Die organische Schicht wurde mit Wasser und einer gesättigten wäßrigen Natriumchlorid- Lösung in dieser Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde durch Säulenchromatographie (Eluens: Chloroform/Ethanol = 30/1) gereinigt, wodurch man 3,6 g öliges (S)-4-Benzyl-2-hydroxymethylmorpholin erhielt.
- (2) 3,5 g (S)-4-Benzyl-2-hydroxymethylmorpholin wurden in 5 ml Ethanol gelöst. Zu der Lösung wurden unter Eiskühlung 5 ml einer 5,9 N Chlorwasserstoff-Ethanol-Lösung gegeben. Die resultierende Mischung wurde 10 Minuten bei der gleichen Temperatur gerührt. Dazu wurde eine Mischung von 500 mg 5% Palladium-Kohle und 10 ml Methanol gegeben. Die resultierende Mischung wurde 3 Stunden bei 40ºC einer Hydrierung unterzogen. Nach Beendigung der Reaktion wurde Palladium-Kohle durch Filtration entfernt. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man ein gelbes öliges Produkt erhielt. Zu dem öligen Produkt wurden 20 ml trockenes Methylenchlorid und 4,7 ml Triethylamin gegeben. Die resultierende Mischung wurde 30 Minuten bei Raumtemperatur gerührt. Dazu wurde tropfenweise innerhalb von 20 Minuten unter Eiskühlung eine Lösung von 4,7 g Tritylchlorid, gelöst in 10 ml Methylenchlorid, gegeben. Die Mischung wurde 3 Stunden bei Raumtemperatur gerührt und zu 50 ml Eiswasser gegeben. Die organische Schicht wurde abgetrennt, und 20 ml Wasser wurden dazugegeben Die resultierende Mischung wurde mit einer 1 N wäßrigen Natriumhydroxid-Lösung auf pH 12 eingestellt. Die organische Schicht wurde abgetrennt und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man ein gelbes öliges Produkt erhielt. Das ölige Produkt wurde mit 10 ml Diisopropylether gemischt. Die resultierenden Kristalle wurden durch Filtration gesammelt, wodurch man 2,7 g (S)-4-Trityl-2-hydroxymethylmorpholin erhielt.
- Schmelzpunkt: 142,0 - 142,5ºC [AcOEt-IPE]
- Optische Drehung: [α]²&sup7;D = 10,90 (C = 1, CHCl&sub3;)
- Die (2R)-Form mit den folgenden Eigenschaften wurde auf die gleiche Weise erhalten.
- Schmelzpunkt: 142,0 - 142,5ºC [AcOEt-IPE]
- Optische Drehung: [α]²&sup7;D = +10,9º (C = 1, CHCl&sub3;)
- (3) Das gleiche Verfahren wie in (1) von Herstellungsbeispiel 5 wurde wiederholt, außer daß das Styroloxid und das 4-Benzyl-2- hydroxymethylmorpholin durch (S)-2-Phenyloxiran bzw. (S)-4-Trityl-2- hydroxymethylmorpholin ersetzt wurden, wodurch man öliges (15,2'S)-1- Phenyl-2-[(4-tritylmorpholin-2-yl)methoxy]ethanol erhielt (Verbindung Nr. 217).
- (4) In 20 ml Aceton wurde (15,2'S)-1-Phenyl-2-[(4-tritylmorpholin-2- yl)methoxylethanol gelöst. Zu dieser Lösung wurden unter Eiskühlung 7 ml einer trockenen 5,9 N Chlorwasserstoff-Ethanol-Lösung gegeben. Die resultierende Mischung wurde 30 Minuten bei Raumtemperatur gerührt, um eine Reaktion zu bewirken. Nach Beendigung der Reaktion wurde das Lösungsmittel durch Destillation unter reduziertem Druck entfernt. Der so erhaltene Rückstand wurde zu einer Mischung von 30 ml Eiswasser und 30 ml Ethylacetat gegeben. Die wäßrige Schicht wurde abgetrennt und mit 30 ml Ethylacetat gewaschen, wonach 50 ml Chloroform dazugegeben wurden. Die resultierende Mischung wurde mit einer 2 N wäßrigen Natriumhydroxid-Lösung auf pH 11 eingestellt. Die organische Schicht wurde abgetrennt, mit einer gesättigten wäßrigen Natriumchlorid-Lösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation unter reduziertem Druck entfernt, wodurch man 1,9 g eines gelben öligen Produktes erhielt. Das ölige Produkt wurde in 10 ml Ethanol gelöst. Zu der Lösung wurden 720 mg Oxalsäure gegeben. Die resultierende Mischung wurde erwärmt, wodurch man eine Lösung erhielt. Man ließ die Lösung über Nacht bei Raumtemperatur stehen. Die resultierenden Kristalle wurden durch Filtration gesammelt und getrocknet, wodurch man 1,8 g (15,2'S)-1-Phenyl- 2-[(morpholin-2-yl)methoxy]ethanoloxalat erhielt (Verbindung Nr. 218).
- Schmelzpunkt: 130 - 132ºC
- Optische Drehung: [α]²&sup4;D +19,9º (C = 1, CHCl&sub3;)
- Die folgenden Verbindungen wurden auf die gleiche Weise erhalten.
- (1R,2'S)-1-Phenyl-2-[(morpholin-2-yl)methoxy]ethanolmaleat (Verbindung Nr. 219)
- Schmelzpunkt: 136 - 136,5ºC
- Optische Drehung: [α]²&sup5;D = -21,7º (C = 1, CHCl&sub3;)
- (15,2'R)-1-Phenyl-2-[(morpholin-2-yl)methoxy]ethanolmaleat (Verbindung Nr. 220)
- Schmelzpunkt: 136 - 136,5ºC

- (1R,2'R)-1-Phenyl-2-[ (morpholin-2-yl)methoxylethanoloxalat (Verbindung Nr. 221)

- Als nächstes wird diese Erfindung speziell mittels Beispielen beschrieben. Jedoch ist die Erfindung keinesfalls durch diese Beispiele beschränkt.
- Tabletten, die jeweils 50 mg 1-(4-Benzyloxyphenyl)-2-[2-(N,N- dimethylamino)ethoxy]ethanolhydrochlorid (Verbindung Nr. 48) enthielten, wurden unter Verwendung des folgenden Rezepts gemäß dem folgenden Verfahren hergestellt: Pro Tablette:
- Die Komponenten (1) wurden mit einer wäßrigen Lösung geknetet, die 8% Polyvinylpyrrolidon K-90 enthielt. Das geknetete Produkt wurde bei 40ºC getrocknet und mit den Komponenten (2) gemischt. Die resultierende Mischung wurde zu runden Tabletten verarbeitet, von denen jede ein Gewicht von 175 mg aufwies und einen Durchmesser von 8 mm hatte.
- Kapseln, die jeweils 50 mg 2-[2-(N,N-Dimethylamino)ethoxy]-1-[4-(4- phenyloxy)phenyl]ethanolhydrochlorid (Verbindung Nr. 54) enthielten, wurden unter Verwendung des folgenden Rezepts gemäß dem folgenden Verfahren hergestellt: Pro Kapsel:
- Die Komponenten (1) wurden mit einer wäßrigen Lösung geknetet, die 8% Polyvinylpyrrolidon K-90 enthielt. Das geknetete Produkt wurde bei 40ºC getrocknet und mit den Komponenten (2) gemischt. Die resultierende Mischung wurde in einer Menge von 150 mg pro Kapsel in Gelatinekapseln Nr. 3 eingefüllt, wodurch man Kapseln erhielt.
- Eine Flüssigkeit, die 25 mg 2-[2-(N,N-Dimethylamino)ethoxy]-1-(3- trifluormethylphenyl)ethanolhydrochlorid (Verbindung Nr. 42) enthielt, wurde unter Verwendung des folgenden Rezepts gemäß dem folgendem Verfahren hergestellt: Pro Ampulle:
- Die obigen Komponenten wurden in physiologischer Kochsalzlösung gelöst, und das Gesamtvolumen der Lösung wurde auf 1 ml eingestellt. Die Lösung wurde aseptisch filtriert und in eine Ampulle gegossen, wodurch man eine Flüssigkeit erhielt.
- Eine Injektion, die 25 mg 2-[2-(N,N-Dimethylamino)ethoxy]-1-(3- fluorphenyl)ethanolhydrochlorid (Verbindung Nr. 1) enthielt, wurde unter Verwendung des folgenden Rezeptes gemäß dem folgenden Verfahren hergestellt:
- Die obigen Komponenten wurden in 1,5 ml destilliertem Wasser, hergestellt zur Injektion, gelöst. Die Lösung wurde aseptisch filtriert, in 3 ml-Miniampullen gegossen und gefriergetrocknet, wodurch man eine Injektion erhielt.
- Feine Granula, die jeweils 50 mg 2-(1-Benzylpiperidin-4-yloxy)-1- phenylethanolhydrochlorid (Verbindung Nr. 112) enthielten, wurden unter Verwendung des folgenden Rezeptes gemäß dem folgenden Verfahren hergestellt:
- Die Komponenten (1) wurden unter Hochgeschwindigkeitsrühren mit einer wäßrigen Lösung, die 8% Polyvinylpyrrolidon K-90 enthielt, einer Granulation unterzogen. Die erhaltenen Granula wurden durch ein Sieb mit 32 Mesh gesiebt und getrocknet, wodurch man feine Granula erhielt.
- 2-[(1-Methylimidazol-5-yl)methoxy]-1-(4-benzyloxyphenyl)ethanol (Verbindung Nr. 215) wurde wie in Beispiel 1 verarbeitet, wodurch man Tabletten erhielt, die jeweils 50 mg der obigen Verbindung enthielten.
- 2-[(1-Methylimidazol-5-yl)methoxy]-1-(4-benzyloxyphenyl)ethanol (Verbindung Nr. 215) wurde wie in Beispiel 2 verarbeitet, wodurch man Kapseln erhielt, die jeweils 50 mg der obigen Verbindung enthielten.

geschützten oder ungeschützten Hydroxylgruppen, der
Nitrogruppe, Oxogruppe und C&sub1;&submin;&sub4;-Alkylendioxygruppen
besteht; die substituierte C&sub1;&submin;&sub6;-Alkyl-, Aryl-, Ar-C&sub1;&submin;&sub4;-
alkyl-, C&sub1;&submin;&sub6;-Alkoxy-, Ar-C&sub1;&submin;&sub4;-alkoxy-, Aryloxy-,
carbamoyloxy-, C&sub1;&submin;&sub6;-Alkylthio-, C&sub2;&submin;&sub6;-Alkenyl-, C&sub2;&submin;&sub6;-
Alkenyloxy-, Ar-C&sub1;&submin;&sub4;-alkylthio-, Ar-C&sub1;&submin;&sub6;-alkylsulfonyl-,
Arylsulfonyl-, C&sub1;&submin;&sub6;-Alkylsulfonylamino-,
Arylsulfonylamino- oder heterocyclische Gruppe als Substituent von R¹ und die
substituierte stickstoffhaltige heterocyclische Gruppe
als R&sup6; mindestens einen Substituenten aufweisen, der aus
der Gruppe ausgewählt ist, die aus Halogenatomen,
geschützten oder ungeschützten Hydroxylgruppen,
geschützten oder ungeschützten Aminogruppen, geschützten
oder ungeschützten Carboxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-
Alkylgruppen, C&sub1;&submin;&sub6;-Alkylgruppen, die durch eine geschützte
oder ungeschützte Hydroxylgruppe substituiert sind,
unsubstituierten oder Halogen-substituierten Arylgruppen,
unsubstituierten oder Halogen-substituierten
Aroylgruppen, unsubstituierten C&sub1;&submin;&sub6;-Alkoxygruppen, C&sub1;&submin;&sub6;-
Alkoxygruppen, die durch eine C&sub1;&submin;&sub6;-Alkoxygruppe
substituiert sind, C&sub1;&submin;&sub6;-Acylgruppen, Ar-C&sub1;&submin;&sub4;-alkylgruppen,
Ar-C&sub2;&submin;&sub6;-alkenylgruppen, heterocyclischen Gruppen,
Heterozyklus-CO-Gruppen, Oxogruppen, C&sub1;&submin;&sub6;-
Alkylsulfonylgruppen und Arylsulfonylgruppen besteht; und
die substituierte Aminogruppe als Substituent von
mindestens jeweils einen Substituenten aufweist, der aus
der Gruppe ausgewählt ist, die aus geschützten oder
ungeschützten llydroxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-
Alkylgruppen, C&sub1;&submin;&sub6;-Alkylgruppen, die durch eine geschützte
oder ungeschützte Carboxyl- oder Hydroxylgruppe
substituiert sind, Cycloalkylgruppen, Arylgruppen, C&sub1;&submin;&sub6;-
Acylgruppen, Ar-C&sub1;&submin;&sub4;-alkylgruppen, heterocyclischen
Gruppen, unsubstituierten oder Oxo-substituierten
Heterozyklus-CO-Gruppen, der Adamantylgruppe, C&sub1;&submin;&sub6;-
Alkylsulfonylgruppen und Arylsulfonylgruppen besteht;
wobei alle obigen heterocyclischen Gruppen aus der Gruppe
ausgewählt sind, die aus den in der Definition von R&sup6;
erwähnten stickstoffhaltigen heterocyclischen Gruppen und
Furyl-, Thienyl-, Benzothienyl-, Pyranyl-,
Isobenzofuranyl-, Oxazolyl-, Benzofuranyl-, Indolyl-,
Benzimidazolyl-, Benzoxazolyl-, Benzothiazolyl-,
Chinoxalyl-, Dihydrochinoxalyl-,
2,3-Dihydrobenzothienyl-, 2,3-Dihydrobenzopyrrolyl-, 2,3-Dihydro-4H-1-
thianaphthyl-, 2,3-Dihydrobenzofuranyl-,
Benzo[b]dioxanyl-, Imidazo[2,3-a]pyridyl-, Benzo[b]piperazinyl-,
Chromenyl-, Isothiazolyl-, Isoxazolyl-, Oxadiazolyl-,
Pyridazinyl-, Isomdolyl- und Isochinolylgruppen besteht.
2. 1,2-Ethandiol-Derivat oder Salz desselben nach Anspruch
1, in dem R² ein Wasserstoffatom oder eine Hydroxyl-
Schutzgruppe darstellt; die nR&sup4;'s gleich oder verschieden
voneinander sind und Wasserstoffatome oder C&sub1;&submin;&sub6;-
Alkylgruppen darstellen; die nR&sup5;'s Wasserstoffatome
darstellen; und R&sup6; eine substituierte oder unsubstituierte
stickstoffhaltige heterocyclische Gruppe darstellt, wobei
die stickstoffhaltige heterocyclische Gruppe durch ein
Kohlenstoffatom, das den heterocyclischen Ring bildet, am
Kohlenstoffatom angebracht ist und aus der Gruppe
ausgewählt ist, die aus Pyrrolyl-, Piperazinyl-,
Imidazolyl-, Pyridyl-, Pyrimidinyl-, Chinolyl-,
Chinolizinyl-, Tetrahydrochinolinyl-, Chinuclidinyl-,
Thiazolyl- und Thiadiazolylgruppen besteht.
3. 1-(4-Benzyloxyphenyl)-2-[(1-methyl-5-
imidazolyl)methoxy]ethanol oder ein Salz desselben.
4. Verwendung eines 1,2-Ethandiol-Derivats, das durch die
folgende allgemeine Formel dargestellt wird, oder eines
Salzes desselben für die Herstellung eines Arzneimittels
zur Behandlung von zerebrovaskulärer Demenz, seniler
Demenz, Alzheimer-Demenz, Folgen von ischämischer
Enzephalopathie oder zerbraler Apoplexie bei einem
Patienten:
in der R¹ eine substituierte oder unsubstituierte
Phenylgruppe darstellt; R ein Wasserstoffatom, eine C&sub1;&submin;&sub6;-
Alkylgruppe oder eine Hydroxyl-Schutzgruppe darstellt; R³
ein Wasserstoffatom oder eine C&sub1;&submin;&sub6;-Alkylgruppe darstellt;
die nR&sup4;'s und nR&sup5;'s gleich oder verschieden voneinander
sind und Wasserstoffatome oder C&sub1;&submin;&sub6;-Alkylgruppen
darstellen; R&sup6; eine substituierte oder unsubstituierte
stickstoffhaltige heterocyclische Gruppe darstellt,
wobei, wenn n null ist, die stickstoffhaltige
heterocyclische Gruppe durch ein Kohlenstoffatom, das
den heterocyclischen Ring bildet, am Sauerstoffatom oder,
wenn n eine ganze Zahl von 1 bis 6 ist, am
Kohlenstoffatom angebracht ist und aus der Gruppe
ausgewählt ist, die aus Pyrrolyl-, Pyrrolidinyl-,
Piperidyl-, Piperazinyl-, Imidazolyl-, Pyridyl-,
Tetrahydropyridyl-, Pyrimidinyl-, Morpholinyl-,
Thiomorpholinyl-, Chinolyl-, Chinolizinyl-,
Tetrahydrochinolinyl-, Tetrahydroisochinolinyl-,
Chinuclidinyl-, Thiazolyl-, Tetrazolyl-, Thiadiazolyl-,
Pyrrolinyl-, Imidazolinyl-, Imidazolidinyl-,
Pyrazolinyl-, Pyrazolidinyl-, Purinyl- und
Indazolylgruppen besteht; und n 0 oder eine ganze Zahl
von 1 bis 6 darstellt, worin der Substituent an aus der
Gruppe ausgewählt ist, die aus Halogenatomen,
substituierten oder unsubstituierten Amino-, C&sub1;&submin;&sub6;-Alkyl-,
Aryl-, Ar-C&sub1;&submin;&sub4;-alkyl-, C&sub1;&submin;&sub6;-Alkoxy-, Ar-C&sub1;&submin;&sub4;-alkoxy-,
Aryloxy-, carbamoyloxy-, C&sub1;&submin;&sub6;-Alkylthio-, C&sub2;&submin;&sub6;-Alkenyl-,
C&sub2;&submin;&sub6;-Alkenyloxy-, Ar-C&sub1;&submin;&sub4;-alkylthio-, Ar-C&sub1;&submin;&sub6;-alkylsulfonyl-,
Arylsulfonyl-, C&sub1;&submin;&sub6;-Alkylsulfonylamino-,
Arylsulfonylaminound heterocyclischen Gruppen, geschützten Aminogruppen,
geschützten oder ungeschützten Hydroxylgruppen, der
Nitrogruppe, der Oxogruppe und C&sub1;&submin;&sub4;-Alkylendioxygruppen
besteht; die substituierte C&sub1;&submin;&sub6;-Alkyl-, Aryl-, Ar-C&sub1;&submin;&sub4;-
alkyl-, C&sub1;&submin;&sub6;-Alkoxy-, Ar-C&sub1;&submin;&sub4;-alkoxy-, Aryloxy-,
carbamoyloxy-, C&sub1;&submin;&sub6;-Alkylthio-, C&sub2;&submin;&sub6;-Alkenyl-, C&sub2;&submin;&sub6;-
Alkenyloxy-, Ar-C&sub1;&submin;&sub4;-alkylthio-, Ar-C&sub1;&submin;&sub6;-alkylsulfonyl-,
Arylsulfonyl-, C&sub1;&submin;&sub6;-Alkylsulfonylamino-, Arylsulfonylamino
oder heterocyclische Gruppe als der Substituent von R¹ und
die substituierte stickstoffhaltige heterocyclische
Gruppe als R&sup6; jeweils mindestens einen Substituenten
aufweisen, der aus der Gruppe ausgewählt ist, die aus
Halogenatomen, geschützten oder ungeschützten
Hydroxylgruppen, geschützten oder ungeschützten
Aminogruppen, geschützten oder ungeschützten
Carboxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-Alkylgruppen, C&sub1;&submin;&sub6;-
Alkylgruppen, die durch eine geschützte oder ungeschützte
Hydroxylgruppe substituiert sind, unsubstituierten oder
Halogen-substituierten Arylgruppen, unsubstituierten oder
Halogen-substituierten Aroylgruppen, unsubstituierten C&sub1;&submin;&sub6;-
Alkoxygruppen, C&sub1;&submin;&sub6;-Alkoxygruppen, die durch eine C&sub1;&submin;&sub6;-
Alkoxygruppe substituiert sind, C&sub1;&submin;&sub6;-Acylgruppen, Ar-C&sub1;&submin;&sub4;-
alkylgruppen, Ar-C&sub2;&submin;&sub6;-alkenylgruppen, heterocyclischen
Gruppen, Heterozyklus-CO-Gruppen, der Oxogruppe, C&sub1;&submin;&sub6;-
Alkylsulfonylgruppen und Arylsulfonylgruppen besteht; und
die substituierte Aminogruppe als der Substituent von
jeweils mindestens einen Substituenten aufweist, der aus
der Gruppe ausgewählt ist, die aus geschützten oder
ungeschützten Hydroxylgruppen, unsubstituierten C&sub1;&submin;&sub6;-
Alkylgruppen, C&sub1;&submin;&sub6;-Alkylgruppen, die durch eine geschützte
oder ungeschützte carboxyl- oder
substituiert sind, Cycloalkylgruppen, Arylgruppen, C&sub1;&submin;&sub6;-
Acylgruppen, Ar-C&sub1;&submin;&sub4;-alkylgruppen, heterocyclischen
Gruppen, unsubstituierten oder Oxo-substituierten
Heterozyklus-CO-Gruppen, der Adamantylgruppe, C&sub1;&submin;&sub6;-
Alkylsulfonylgruppen und Arylsulfonylgruppen besteht;
wobei alle obigen heterocyclischen Gruppen aus der Gruppe
ausgewählt sind, die aus den in der Definition von R&sup6;
erwähnten stickstoffhaltigen heterocyclischen Gruppen und
Furyl-, Thienyl-, Benzothienyl-, Pyranyl-,
Isobenzofuranyl-, Oxazolyl-, Benzofuranyl-, Indolyl-,
Benzimidazolyl-, Benzoxazolyl-, Benzothiazolyl-,
Chinoxalyl-, Dihydrochinoxalyl-,
2,3-Dihydrobenzothienyl-, 2,3-Dihydrobenzopyrrolyl-, 2,3-Dihydro-4H-1-
thianaphthyl-, 2,3-Dihydrobenzofuranyl-,
Benzo[b]dioxanyl-, Imidazo[2,3-a]pyridyl-,
Benzo[b]piperazinyl-, Chromenyl-, Isothiazolyl-,
Isoxazolyl-, Oxadiazolyl-, Pyridazinyl-, Isomdolyl- und
Isochinolylgruppen besteht.
5. Verwendung eines 1,2-Ethandiol-Derivats oder eines Salzes
desselben nach Anspruch 4, in dem R² ein Wasserstoffatom
oder eine Hydroxyl-Schutzgruppe darstellt; die nR&sup4;'s
gleich oder verschieden voneinander sind und
Wasserstoffatome oder C&sub1;&submin;&sub6;-Alkylgruppen darstellen; die
nR&sup5;'s Wasserstoffatome darstellen; und R eine
substituierte oder unsubstituierte stickstoffhaltige
heterocyclische Gruppe darstellt, wobei, wenn n null ist,
die stickstoffhaltige heterocyclische Gruppe durch ein
Kohlenstoffatom, das den heterocyclischen Ring bildet, am
Sauerstoffatom oder, wenn n eine ganze Zahl von 1 bis 6
ist, am Kohlenstoffatom angebracht ist und aus der Gruppe
ausgewählt ist, die aus Pyrrolyl-, Pyrrolidinyl-,
Piperidinyl-, Piperazinyl-, Imidazolyl-, Pyridyl-,
Pyrimidinyl-, Morpholinyl-, Chinolyl-, Chinolizinyl-,
Tetrahydrochinolinyl-, Chinuclidinyl-, Thiazolyl- und
Thiadiazolylgruppen besteht.
6. Verwendung eines 1,2-Ethandiol-Derivats oder eines Salzes
desselben nach Anspruch 4, in dem R&sup6; eine substituierte
oder unsubstituierte stickstoffhaltige heterocyclische
Gruppe darstellt, wobeil wenn n null ist, die
stickstoffhaltige heterocyclische Gruppe durch ein
Kohlenstoffatom, das den heterocyclischen Ring bildet, am
Sauerstoffatom oder, wenn n eine ganze Zahl von 1 bis 6
ist, am Kohlenstoffatom angebracht ist und aus der Gruppe
ausgewählt ist, die aus Pyrrolinyl, Piperidyl,
Piperazinyl, Imidazolyl, Pyridyl, Morpholinyl,
Thiomorpholinyl, Tetrahydropyridyl, Chinuclidinyl und
Tetrazolyl besteht, der Substituent an R aus der Gruppe
ausgewählt ist, die aus Halogenatomen; C&sub1;&submin;&sub6;-Alkylgruppen;
C&sub2;&submin;&sub6;-Alkenylgruppen; Phenylgruppen, die gegebenenfalls
durch ein Halogenatom oder eine C&sub1;&submin;&sub6;-Alkyl- oder C&sub1;&submin;&sub6;-
Alkoxygruppe substituiert sein können; Phenyl-C&sub1;&submin;&sub4;-
alkylgruppen; C&sub1;&submin;&sub6;-Alkoxygruppen, die gegebenenfalls durch
eine Pyridyl-, Furyl- oder Thienylgruppe substituiert
sein können; Phenyl-C&sub1;&submin;&sub4;-alkoxygruppen, die gegebenenfalls
durch ein Halogenatom oder eine C&sub1;&submin;&sub6;-Alkyl-, Hydroxyl- oder
C&sub1;&submin;&sub6;-Alkoxygruppe substituiert sein können; einer
Phenoxygruppe; Phenylaminocarbonyloxygruppen, die
gegebenenfalls durch ein Halogenatom substituiert sein
können; C&sub1;&submin;&sub6;-Alkylaminocarbonyloxygruppen; C&sub1;&submin;&sub6;-
Alkylthiogruppen; einer Phenylsulfonylaminogruppe;
Aminogruppen, die gegebenenfalls durch eine C&sub1;&submin;&sub6;-
Alkylgruppe substituiert sein können; einer
Hydroxylgruppe; einer Nitrogruppe; einer Oxogruppe;
Phenyl-C&sub1;&submin;&sub4;-alkylthiogruppen; Phenyl-C&sub1;&submin;&sub4;-
alkylsulfonylgruppen; einer Thienylgruppe; einer
Pyridylgruppe; und C&sub1;&submin;&sub4;-Alkylendioxygruppen besteht; und
der Substituent an R&sup6; aus der Gruppe ausgewählt ist, die
aus Halogenatomen; einer Hydroxylgruppe; C&sub1;&submin;&sub6;-Alkylgruppen,
die gegebenenfalls durch eine Hydroxylgruppe substituiert
sein können; einer Aminogruppe; C&sub1;&submin;&sub6;-Acylgruppen, die
gegebenenfalls durch eine Pyrrolidinylgruppe substituiert
sein können; Phenyl-C&sub1;&submin;&sub4;-alkylgruppen; Phenyl-C&sub2;&submin;&sub4;-
alkenylgruppen; Aroylgruppen, die durch ein Halogenatom
substituiert sind; einer Pyridylgruppe und einer
Oxogruppe besteht.
7. Verwendung eines 1,2-Ethandiol-Derivats oder eines Salzes
desselben nach Anspruch 6, in dem R¹ eine Phenylgruppe,
die gegebenenfalls durch ein Halogenatom substituiert
sein kann; eine Phenyl-C&sub1;&submin;&sub4;-alkoxygruppe; eine
Phenylgruppe; eine Thienylgruppe; eine Phenoxygruppe, die
gegebenenfalls durch ein Halogenatom oder eine C&sub1;&submin;&sub6;-
Alkoxygruppe substituiert sein kann; oder eine C&sub1;&submin;&sub6;-
Alkoxygruppe darstellt, die gegebenenfalls durch eine
Thienylgruppe substituiert sein kann; R² ein
Wasserstoffatom darstellt; R³ ein Wasserstoffatom
darstellt; die nR&sup4;'s und nR&sup5;'s Wasserstoffatome
darstellen; R&sup6; eine Morpholinylgruppe, eine Pyridylgruppe
oder ein Imidazolylgruppe darstellt, die gegebenenfalls
durch eine C&sub1;&submin;&sub6;-Alkylgruppe substituiert sein kann.
8. Verwendung eines 1,2-Ethandiol-Derivats oder eines Salzes
desselben nach Anspruch 4, in dem das Derivat oder das
Salz 1-(4-Benzyloxyphenyl)-2-[(1-methyl-5-
imidazolyl)methoxy]ethanol oder ein Salz desselben ist.
9. Ein die Zerebralfunktion verbesserndes Mittel, umfassend
ein 1,2-Ethandiol-Derivat, das durch die folgende
allgemeine Formel dargestellt wird, oder ein Salz
desselben:
in der R¹, R², R³, R&sup4;, R&sup5;, R&sup6; und n wie in Anspruch 4
definiert sind.
