Description
- Die vorliegende Erfindung bezieht sich auf neue 2-Amino-5- aryloxymethyl-oxazoline sowie auf ihre Salze mit pharmakologisch kompatiblen Säuren.
- Die Verbindungen der Erfindung haben die allgemeine Formel:
- in der Ar eine 2-Phenyl-phenyl, 3-Phenyl-phenyl oder 4-Phenyl-phenyl-Gruppe ist.
- Die Verbindungen der Erfindung werden durch Kondensation von 1-Aryloxy-2,3-epoxy-propanen der Struktur II
- in der Ar die gleichen Werte wie oben besitzt, mit Mono-Natriumcyanamid in einem organischen Lösungsmittel, das in Wasser lsöich ist, wie niedrige Alkohole von C&sub1; bis C&sub4; wie Methanol, Ethanol, Isopropanoloder Butanol oder Tetrahydrofuran oder Dimethylformamid bei einer Temperatur von 0 ºC bis 80 ºC hergestellt.
- Die Verbindungen der Struktur II sind bekannt und ihre Herstellungsverfahren sind in dein US Patent 2 181 085 der Firma Dow Chemical Company beschrieben.
- Die Anmelderin hat bereits in den Patenten FR 2 533 922 und FR 2 546 167 2-Amino-5-aminomethyl-oxazoline beschrieben.
- JARRY et al. haben in Bull. Soc. Pharm. Bordeaux, 1981, 120, 153-162 5-Aryloxymethyl-2-amino-oxazoline der Struktur III
- beschrieben, in der der Rest R eine Methylgruppe in Position 4, ein Chlor in Position 4, ein Methoxy in Position 4, eine Acetamido-Gruppe in Position 4 oder Allyl in Position 4 und Methoxy in Position 2 ist; der Rest R eine CH=CH-CH=CH-Gruppe in Position 2,3 ist und mit dem Benzolring eine 1-Naphthyl- Gruppe darstellt. Diese Verbindungen besitzen interessante blutdruckmindernde Eigenschaften.
- Die Firma DOW CHEMICAL hat im US Patent 3 637 726 (3,4-Dihalo-phenoxy)-5-methyl-2-amino-oxazoline der Struktur IV beschrieben, in der X ein Chlor-, Brom-Halogenatom darstellt. Diese Verbindungen besitzen interessante antimikrobielle Eigenschaften.
- Das Patent US-A- 3 818 028 beschreibt (2,4,6-Trihalo-phenoxy)-5-methyl-2-amino-oxazoline, die als Antidepressiva verwendbar sind.
- Die Erfindung wird in den folgenden Beispielen genauer beschrieben werden.
- Die Schmelzpunkte werden mit einem Kofler-Block gemessen, die chemischen Verschiebungen werden in ppm in bezug auf TMS, das als interner Standard verwendet wird, angegeben.
- 2-Amino-5-(2-phenyl-phenoxy-methyl)-oxazolin oder COR3264, ein Produkt der Struktur I mit Ar = 2-Phenyl-phenyl.
- In einen 500 cm³-Kolben, ausgerüstet mit einem Rührer und einem Kühler gibt man 34 g 1-(2-Phenyl-phenoxy)-2,3-epoxy- propan, 150 ml wasserfreies Methanol und 11 g Mono-Natriumcyanamid. Man rührt die Reaktionsmischung während 16 Stunden bei Umgebungstemperatur. Man filtert über Celite 535 die mineralischen Salze ab. Das Filtrat wird unter Vakuum eingedampft. Der Rückstand wird in 250 ml Chloroform aufgenommen. Die Chloroform-Lösung wird zweimal mit 20 ml Wasser gewaschen, über Natriumsulfat getrocknet und unter Vakuum eingedampft. Das erhaltene Produkt wird aus Heptan umkristallisiert. Man erhält 13 g COR3264 in Form eines farblosen Feststoffs. Ausbeute: 31 %; molare Masse: 268,32.

- 3,2-4,2 ppm, 4 Protonen, komlexes Signal, 2 CH&sub2;; 4,5-5,0 ppm, 3 Protonen, komplexes Signal, 1 CHO des Oxazolins + NH&sub2;, mit D&sub2;O austauschbar; 6,8-7,7 ppm, 9 Protonen, komplexes Signal, aromatische Protonen.
- 2-Amino-5-(4-phenyl-phenoxy-methyl)-oxazolin oder COR4408, Produkt der Struktur I mit Ar = 4-Phenyl-phenyl.
- In einen 1500 cm³-Kolben, ausgerüstet mit einem Rührer und einem Kühler gibt man 55 g 1-(4-Phenyl-phenoxy)-2,3-epoxypropan, gelöst in 800 ml einer Mischung aus Tetrahydrofuran/Methanol von 50:50, dann fügt man 32 g Mono-Natriumcyanamid zu. Man rührt die Reaktionsmischung während 48 Stunden bei Umgebungstemperatur. Man filtert über Celite 535 die mineralischen Salze ab. Das Filtrat wird unter Vakuum eingedampft. Der Rückstand wird in 250 ml Ethylether aufgenommen, der abgeschiedene Feststoff wird filtriert, dann mit 200 ml Wasser gewaschen. Der erhaltene Feststoff wird aus einer Mischung Toluol/Methanol umkristallisiert und unter Vakuum getrocknet. Man erhält 30,5 g COR4408 in Form eines farblosen Feststoffs. Ausbeute: 47 %; molare Masse: 268,32.
- Schmelzpunkt: 201 ºC
- NMR-Spektrum in DMSO D&sub6;
- 3,1-4,3 ppm, 4 Protonen, komplexes Signal, 2 CH&sub2;; 4,5-5,0 ppm, 1 Proton, Multiplett, 1 CHO des Oxazolins; 5,8 ppm, breiter Peak, 2 Protonen, NH&sub2;, mit D&sub2;O austauschbar; 6,9-7,7 ppm, 9 Protonen, komplexes Signal, aromatische Protonen.
- 2-Amino-5-(3-phenyl-phenoxy-methyl)-oxazolin oder COR4463, Produkt der Struktur 1 mit Ar = 3-Phenyl-phenyl.
- In einen 500 cm³-Kolben, ausgerüstet mit einem Rührer und einem Kühler gibt man 18 g Mono-Natriumcyanamid in 150 ml wasserfreiem Methanol, dann fügt man tropfenweise 28 g 1-(3- Phenyl-phenoxy)-2,3-epoxy-propan, gelöst in 50 ml wasserfreiein Methanol hinzu. Man rührt die Reaktionsmischung während 16 Stunden bei Umgebungstemperatur. Man filtert über Celite 535 die mineralischen Salze ab. Das Filtrat wird unter Vakuum eingedampft. Der Rückstand wird mit 250 ml Dichlormethan wiederaufgenommen. Die Lösung wird zweimal mit 20 ml Wasser gewaschen, über Natriumsulfat getrocknet und unter Vakuum eingedampft. Das erhaltene Produkt wird in Heptan umkristallisiert. Man erhält 15 g COR4463 in Form eines farblosen Feststoffs. Ausbeute: 48 %; inolare Masse: 268,32.
- Schmelzpunkt: 124 ºC
- NMR-Spektrum in CDCl&sub3;
- 3,3-4,3 ppm, 4 Protonen, komplexes Signal, 2 CH&sub2;; 4,6-5,3 ppm, 3 Protonen, komplexes Signal, 1 CHO des Oxazolins + NH&sub2;, austauschbar mit D&sub2;O; 6,7-7,7 ppm, 9 Protonen, komplexes Signal, aromatische Protonen.
- Die akute Toxizität von COR3264, COR4408 und COR4463 ist mit Mäusen durch orale Verabreichung bestimmt worden. Die DL&sub5;&sub0;- Werte für die drei Verbindungen sind nachstehemd angegeben:
- 1) COR3264 - DL&sub5;&sub0; = 273 mg/kg
- 2) COR4408 - DL&sub5;&sub0; = 800 mg/kg
- 3) COR4463 - DL&sub5;&sub0; = 389 mg/kg
- Die Verbindungen der Erfindung sind in einem pharmakologischen Screening getestet worden. Die verschiedenen, verwendeten Tests sind nachstehend aufgeführt.
- - Messung der motorischen Aktivität im Aktimeter von Boissier
- - Antagonismus gegenüber Hypothermie, hervorgerufen durch subkutane Injektion von 2,5 mg/kg Reserpin.
- - Antagonismus gegenüber Hypothermie, hervorgerufen durch subkutane Injektion von 16 mg/kg Apomorphin
- - Potenzierung der Toxizität durch Yohimbin
- - TST, Am-Schwanz-Aufhängungstest.
- Alle Produkte wurden oral verabreicht.
- Bei dem Aktimeter-Test zeigt das COR3264 überhaupt keine Aktivität bis zu einer Dosis von 64 mg/kg und bewirkt eine leichte Abnahme der spontanen motorischen Aktivität bei einer Dosis von 128 mg/kg; das COR4408 zeigt überhaupt keine Aktivität bis zu einer Dosis von 64 mg/kg und bewirkt eine Abnahme der spontanen motorischen Aktivität bei einer Dosis von 256 mg/kg; das COR4463 wirkt ab einer Dosis von 16 mg/kg sedativ.
- Die Aktivität gegenüber Hypothermie, induziert durch Reserpin, wurde bei Mäusen bestimmt; das COR3264 zeigte überhaupt keine Aktivität bis zu einer Dosis von 128 mg/kg, das COR4408 hat eine DE&sub5;&sub0; von 40 mg/kg, das COR4463 ist bis zu einer Dosis von 128 mg/kg inaktiv.
- Die Aktivität gegenüber Hypothermie, induziert durch die Injektion von 16 mg/kg Apomorphin, wurde bei Mäusen bestimmt; das COR3264 zeigte überhaupt keine Aktivität bis zu einer Dosis von 128 mg/kg, das COR4408 hat eine DE50 von 73 mg/kg, das COR4463 ist bis zu einer Dosis von 128 mg/kg inaktiv.
- In dem Test des Aufhängens am Schwanz wurde eine wirksame Dosis 25, DE25 für die drei Substanzen bestimmt; für das COR3264 beträgt sie 16 mg/kg, für das COR4408 130 mg/kg und für das COR4463 90 mg/kg.
- Die Potenzierung der Toxizität wurde durch das Yohimbin bestimmt. Die DL&sub5;&sub0;-Werte nach der Verabreichung von 30 mg/kg Yohimbin auf subkutanem Weg betragen für das COR3264 70 mg/kg, für das COR4408 16 mg/kg und für das COR4463 37 mg/kg.
- Die vorstehend beschriebenen pharmakologischen Ergebnisse weisen auf eine antidepressive Aktivität für die Verbindungen der Erfindung hin.
- In Anbetracht ihrer toxipharmakologischen Aktivitäten sind die Produkte gemäß der vorliegenden Erfindung also einzeln oder zusammen mit anderen Wirkstoffen für die Behandlung von depressiven Zuständen verwendbar.
- Die Dosen und die therapeutischen Schemata werden von der Person und der zu behandelnden Krankheit abhängen. Die Produkte könnten auf oralem Weg (z. B. in Form von Gelatinekapseln, Tabletten, Sirup, trinkbaren Tropfen) oder auf injizierbarem Weg (injizierbare Lösung auf intramuskulärem Weg oder auf intravenösem Weg, Lösung für die intravenöse Infusion) oder auf rectalem Weg (Suppositorien), auf lokalem Weg (Cremes, Pomaden, Gele) verabreicht werden. Gemäß den Indikationen kann die tägliche Dosis 10 bis 2000 mg pro Tag, eingenommen in einer bis zu vier Dosen, variieren.
Claims (8)
1. Neue Produkte der allgemeinen Formel
mit Ar = Phenyl-2-Phenyl, Phenyl-3-Phenyl oder Phenyl-4-
Phenyl
2. Verfahren zur Herstellung von Produkten nach Anspruch 1,
dadurch gekennzeichnet, daß man in einem in Wasser
löslichen organischen Lösungsmittel wie niedere Alkohole,
Tetrahydrofuran, Dimethylformamid und bei einer
Temperatur von 0 ºC bis 80 ºC das Mononatrium-Derivat des
Cyanamids mit einem Aryloxy-1-epoxy-2,3-propan
der
Formel
mit Ar = Phenyl-2-Phenyl, Phenyl-3-Phenyl oder Phenyl-4-
Phenyl kondensiert.
3. Neues Medikament, dadurch gekennzeichnet, daß die aktive
Substanz aus mindestens einem der Produkte nach Anspruch
1 besteht.
4. Neues Medikament nach Anspruch 3, dadurch
gekennzeichnet, daß die aktive Substanz aus 2-Amino-5-(2-phenyl-
phenoxy-methyl)-oxazolin oder COR3264 besteht.
5. Neues Medikament nach Anspruch 3, dadurch
gekennzeichnet, daß die aktive Substanz aus 2-Amino-5-(4-phenyl-
phenoxy-methyl)-oxazolin oder COR4408 besteht.
6. Neues Medikament nach Anspruch 3, dadurch
gekennzeichnet, daß die aktive Substanz aus 2-Amino-5-(3-phenyl-
phenoxy-methyl)-oxazolin oder COR4463 besteht.
7. Neues Medikament nach Anspruch 3, das für die Behandlung
von Depressionszuständen geeignet ist.
8. Pharmazeutische Zusammensetzung, dadurch gekennzeichnet,
daß sie als aktive Substanz mindestens ein Produkt nach
Anspruch 1, in Verbindung mit einem pharmazeutischen
Träger oder einer geeigneten Grundmasse enthält.
