Description
Die vorliegende Erfindung betrifft neue asymmetrisch sub
stituierte bis-Naphthalimide, deren Herstellung sowie deren
Verwendung bei der Bekämpfung von Krankheiten.
Aus der US-PS 4.841.052 sind Verbindungen bekannt, bei denen zwei
Naphthalsäureimidreste über eine einen aromatischen Rest ent
haltende Brücke miteinander verbunden sind. Diese Verbindungen
dienen als Sensibilisatoren in elektrophotographischen Tonern. In
der US-PS 4.874.863 sind bis-Naphthalimide beschrieben, die sich
als Cancerostatica eignen. Analoges gilt für die Substanzen der
US-PS 5.086.059, die an ihren Naphthalinresten zusätzlich
Ethylenbrücken enthalten.
Gegenstand der Erfindung sind asymmetrisch substituierte bis-
Naphthalimide der Formel I
worin
A, B und D gleich oder verschieden sind und gerade- oder ver zweigtkettige Alkylidengruppen, Cycloalkyli den- oder Phenylenreste bedeuten,
R1 und R2 gleich oder verschieden sind und Wasserstoffatome oder C1-6-Alkyl-, oder Aryl- oder Benzylgruppen, die durch C1-6-Alkylgruppen, Halogenatome, CF3-, Nitro-, Hydroxy-, NR3 2-, OR4-, COOR5-, CONR6 2-, CONHR7-, NHCOR8-, SO2NHR9- oder SOmR10-Gruppe sub stituiert sein können, bedeuten, oder gemeinsam eine -CH2-, -C2H4- oder -C3H6-Gruppe darstellen,
X, X′, Y und Y′ gleich oder verschieden sind und Wasserstoff oder Halogenatome, Nitro-, Amino-, C1-6-Alkyl-, CF3-, Hydroxy-, NR3 2-, OR4-, COOR5-, CHO-, CONR6 2-, CONHR7-, NHCOR8-, SO2NHR9-, SOmR10, COR11-, Ureyl-, C1-6-Alkylureyl, N=C=S- oder N=C=O-Gruppen darstel len,

m die Zahl 0, 1 oder 2 ist und

sowie deren Salze mit physiologisch verträglichen Säuren.
Für A, B und D sind als Alkylidengruppen solche mit 1-8 und
insbesondere solche mit 1-4 C-Atomen bevorzugt. Als Cyclo
alkylidenreste sind solche mit 3-7 C-Atomen bevorzugt.
Als Arylgruppen sind für R1-R8 besonders Phenylgruppen zu nen
nen. Letztere sind vorzugsweise durch 1-2 der genannten Reste
substituiert. Bevorzugte Cycloalkylreste für R3 - R11 sind solche
mit 3-7 C-Atomen. X und Y sind vorzugsweise Nitro-, Amino- oder
Methylgruppen oder Chloratome und X′ und Y′ sind vorzugsweise
Wasserstoff.
Die neuen Verbindungen lassen sich nach folgendem Reaktionsschema
herstellen:
Naphthalsäureanhydride II werden mit einem Äquivalent oder mit
einem bis zu 50%igem Überschuß eines Polyamins III in einem or
ganischen Lösungsmittel wie Dioxan, Ethanol oder Dimethylformamid
bei 20 bis 150°C umgesetzt. Die so erhaltene Lösung wird eingeengt
und der Rückstand beispielsweise durch Umkristallisieren oder
Chromatographie gereinigt. Das so erhaltene Monoimid wird an
schließend mit dem Anhydrid V wie oben beschrieben umgesetzt und
gereinigt.
Die neuen Verbindungen aktivieren nicht-spezifische Immunzellen,
die Tumorzellen abtöten. Sie lassen sich daher bei Krebs
erkrankungen einsetzen. Ihre Applikationsdosis beträgt etwa
1-500 mg/kg Körpergewicht.
Die neuen Verbindungen haben den Vorteil einer guten Löslichkeit
in Wasser. Sie lassen sich daher viel leichter anwenden als die
bekannten Verbindungen ähnlicher Struktur.
Die so erhaltenen Verbindungen können anschließend gewünschten
falls in ihre Salze mit physiologisch verträglichen Säuren über
führt werden. Hierzu werden die Basen beispielsweise in Ethanol,
Dichlormethan oder Ether aufgenommen und das Salz mit der Säure
ausgefällt. Als Säuren eignen sich Zitronensäure, Weinsäure,
Milchsäure, Phosphorsäure, Alkansulfonsäuren, insbesondere
Methansulfonsäure, Essigsäure, Ameisensäure, Maleinsäure, Fumar
säure, Äpfelsäure, Bernsteinsäure, Malonsäure, Schwefelsäure,
Toluolsulfonsäure, L-Glutaminsäure, L-Asparaginsäure, Brenz
traubensäure, Schleimsäure, Benzoesäure, Glucuronsäure, Oxal
säure, Ascorbinsäure und insbesondere Salzsäure.
Die Ausgangsstoffe der Formel V mit einer Einfachbindung im Fünf
ring lassen sich nach in der Literatur bekannten Verfahren her
stellen (J. Amer. Chem. Soc. 93, 737 (1971)). Die entsprechenden
Verbindungen mit einer Doppelbindung im Fünfring erhält man aus
denen mit einer Einfachbindung nach dem in J. Amer. Chem.
Soc. 91, 917 (1969) für Pyracyclochinone beschriebenen Verfahren.
Die Polyamine III sind käuflich erhältlich oder lassen sich nach
bekannten Verfahren herstellen.
Folgende Beispiele sollen die Erfindung näher erläutern. Angaben
in Teilen bzw. Prozente beziehen sich jeweils auf das Gewicht.
- a) (3-Aminopropyl) [3-(1,8-naphthalimido)propyl]methylamin
2,2 g (0,015 Mol) bis-(3-Aminopropyl)methylamin wurden in 250 ml Ethanol gelöst und 2,4 g (0, 012 Mol) Naphtha lin-1,8-dicarbonsäureanhydrid langsam über einen Zeitraum von 4 h hinzugegeben. Die Mischung wurde anschließend noch 1 h bei Raumtemperatur gerührt. Das organische Lö sungsmittel wurde im Vakuum entfernt. Es blieb ein Öl zu rück, welches durch Flash-Chromatographie (CH2Cl2/MeOH/NH4OH = 4/6/1) gereinigt wurde. Man erhielt 1,7 g (50%) an Produkt [Fp. = 266°C (Methanol)], welches direkt für die weitere Umsetzung benutzt wurde.
Analog Aa wurden hergestellt:
- b) N-(2-Aminoethyl)-N′-[2-(1.8-naphthalimido)ethyl]-1,2-dia minoethan, Ausbeute 30%, Fp. 137°C (Ethylacetat).
- c) N-(2-Aminoethyl)-N′-[2-(4-chlor-1,8-naphthal imido)ethyl]-1,3-diaminopropan, Ausbeute 53%, Öl [Hydrochlorid, Fp. 253°C].
- d) N-(3-Aminopropyl)-N′-[3-(3-hydroxy-1,8-naphthal imido)-propyl]-1,4-di(ethylamino)butan, Ausbeute 44%, Öl.
- e) N-[3-(3-Amino-1,8-naphthalimido)propyl] (3-amino propyl)amin, Ausbeute 33%, Fp. 190°C.
- f) N-(2-Aminoethyl) [2-(4-chlor-1,8-naphthalimido)ethyl]amin, Ausbeute 24%, Fp. 280°C (Methanol).
- g) N-[3-(3-Amino-1,8-naphthalimido)propyl]-N′ (3-amino propyl)-1,4-diaminobutane, Ausbeute 25%, Fp. 260°C.
- h) N-(3-Aminopropyl) [3-(4-chloro-1,8-naphthalimido) propyl]methylamine, Ausbeute 37%, Öl.
- i) N-[3-(3-Amino-1,8-naphthalimido)propyl] (3-aminopropyl) methylamine, Ausbeute 32%, Fp. 330°C (Zersetzung).
0,7 g (0,0021 Mol) N-[3-(3-Amino-1,8-naphthalimido)propyl]
(3-aminopropyl)amin (aus Beispiel Ae) und 0,45 g (0,002 Mol)
5,6-Acenaphthendicarbonsäureanhydrid wurden in 50 ml Ethanol
2 h unter Rückfluß gerührt. Nach dem Abkühlen wurde der Nie
derschlag abfiltriert, mit Ethanol gewaschen und aus Xylol
umkristallisiert. Es wurden 0,4 g (38%) reine Substanz er
halten, Fp. 205°C.
Analog Beispiel 1 wurden die Substanzen der folgenden Tabelle
hergestellt:
Claims (3)
1. Asymmetrisch substituierte bis-Naphthalimide der Formel I
worin
A, B und D gleich oder verschieden sind und gerade- oder ver zweigtkettige Alkylidengruppen, Gyclo alkyliden- oder Phenylenreste bedeuten,
R1 und R2 gleich oder verschieden sind und Wasserstoffatome oder C1-6-Alkyl-, oder Aryl- oder Benzylgruppen, die durch C1-6-Alkylgruppen, Halogenatome, CF3-, Nitro-, Hydroxy-, NR3 2-, OR4-, COOR5-, CONR6 2-, CONHR7-, NHCOR8-, SO2NHR9- oder SOmR10-Gruppen sub stituiert sein können, bedeuten, oder gemeinsam eine -CH2-, -C2H4- oder -C3H6-Gruppe darstellen,
X, X′, Y und Y′ gleich oder verschieden sind und Wasserstoff oder Halogenatome, Nitro-, Amino-, C1-6-Alkyl-, CF3-, Hydroxy-, NR3 2-, OR4-, COOR5-, CHO-, CONR6 2-, CONHR7-, NHCOR8-, SO2NHR9-, SOmR10-, COR11-, Ureyl-, C1-6-Alkylureyl, N=G=S- oder N=C=O-Gruppen darstel len,
R3-R11 gleich oder verschieden sind und C1-6-Alkyl-, Gycloalkyl- oder Arylgruppen bedeuten,
m die Zahl 0, 1 oder 2 ist und
Ç| eine Einfach- oder Doppelbindung darstellt,
sowie deren Salze mit physiologisch verträglichen Säuren.

X, X′, Y und Y′ gleich oder verschieden sind und Wasserstoff oder Halogenatome, Nitro-, Amino-, C1-6-Alkyl-, CF3-, Hydroxy-, NR3 2-, OR4-, COOR5-, CHO-, CONR6 2-, CONHR7-, NHCOR8-, SO2NHR9-, SOmR10-, COR11-, Ureyl-, C1-6-Alkylureyl, N=G=S- oder N=C=O-Gruppen darstel len,
R3-R11 gleich oder verschieden sind und C1-6-Alkyl-, Gycloalkyl- oder Arylgruppen bedeuten,
m die Zahl 0, 1 oder 2 ist und
Ç| eine Einfach- oder Doppelbindung darstellt,
sowie deren Salze mit physiologisch verträglichen Säuren.
2. Verbindungen der Formel I gemäß Anspruch 1 zur Verwendung bei
der Bekämpfung von Krankheiten.
3. Arzneimittel enthaltend eine Verbindung der Formel I gemäß
Anspruch 1.
