Description
Die vorliegende Erfindung betrifft neue Verfahren zur Herstellung von
optisch aktiven 3-Benzylamino-1-arylbutanen und 3-Amino-1-arylbutanen der
allgemeinen Formeln Ia und Ib
in denen Ar für einen aromatischen Rest steht.
Aus der Arbeit von ervinka et al. (Coll. Czech. Chem. Commun., (1968)
Band 33, Seite 3551) und der DE-B 11 82 245 ist bekannt, daß man racemisches
3-Amino-1-phenylbutan mit Hilfe von Weinsäure bzw. Mandelsäure nach
dem Prinzip der Racemattrennung über die diastereomeren Salzpaare in seine
Antipoden zerlegen kann.
Beide Verfahren benötigen große Mengen an Lösungsmitteln, erfordern mehrfache
Umkristallisation und liefern nur geringe Ausbeuten an reinem
Produkt.
Der Erfindung lag die Aufgabe zugrunde, die optisch aktiven Verbindungen
Ia und Ib besser zugänglich zu machen.
Demgemäß wurde ein Verfahren zur Herstellung von optisch aktiven 3-Benzylamino-1-arylbutanen
der allgemeinen Formel Ia und Ib
in der Ar für einen aromatischen Rest steht, gefunden, welches dadurch
gekennzeichnet ist, daß man
- a) zur Herstellung der Verbindungen Ia racemisches Ia mit einer optisch aktiven Säure (II) in ein diastereomeres Salzpaar überführt, dieses durch fraktionierte Kristallisation in die beiden diastereomeren Salze trennt, von diesen die optisch aktive Säure (II) mittels einer starken Base wieder abtrennt und
- b) zur Herstellung der Verbindungen Ib die gemäß Schritt (a) erhältlichen, optisch aktiven Verbindungen Ia durch katalytische Hydrierung in die Verbindungen Ib und Toluol zerlegen.
Die racemischen Ausgangsverbindungen Ia sind bekannt oder in bekannter
Weise erhältlich, z. B. durch Umsetzung von Benzaldehyd mit einem 3-Amino-1-arylbutan
oder von einem 1-Arylbutan-3-on mit Benzylamin in Gegenwart
eines Reduktionsmittels (Houben Weyl, "Methoden der organischen Chemie",
(1957), Band 11/1, Seite 643 ff).
Im Hinblick auf die pharmakologischen Eigenschaften der aus Ia und Ib zugänglichen
Wirkstoffe sind solche Verbindungen Ia und Ib bevorzugt, in
denen Ar für den Phenylrest steht.
Daneben kommen aber auch weitere aromatische Gruppen wie die Naphthylgruppen
sowie substituierte Phenyl- und Naphthylgruppen in Betracht, wobei diese
Gruppen beispielsweise bis zu drei der folgenden Reste tragen können:
- - C₁-C₄-Alkyl, insbesondere Methyl, Ethyl und Isopropyl
- - C₁-C₄-Alkoxy, insbesondere Methoxy und Ethoxy
- - C₁-C₄-Alkoxycarbonyl, insbesondere Methoxycarbonyl und Ethoxycarbonyl
- - Carbonsäure-C₁-C₄-alkylester, insbesondere Carbonsäuremethylester
- - Carbonyloxy, insbesondere Acetyloxy und Propionyloxy
- - Halogen, insbesondere Fluor, Chlor und Brom
- - Hydroxy
- - Nitro
- - Cyano
Beispiele für solche Verbindungen sind:
3-Benzylamino-1-phenylbutan
3-Benzylamino-1-(4-hydroxy)phenylbutan
3-Benzylamino-1-(4-chlor)phenylbutan
3-Benzylamino-1-(2,4-dichlor)phenylbutan
3-Benzylamino-1-(4-tert.butyl)phenylbutan
3-Benzylamino-1-(4-hydroxy)phenylbutan
3-Benzylamino-1-(4-chlor)phenylbutan
3-Benzylamino-1-(2,4-dichlor)phenylbutan
3-Benzylamino-1-(4-tert.butyl)phenylbutan
Als optisch aktive Säuren (II) kommen Weinsäure und Milchsäure und
insbesondere Mandelsäure oder ihre optischen Antipoden in Betracht.
Man setzt die optisch aktive Säure (II) in der Regel in einem Molverhältnis
von 0,5 bis 2, vorzugsweise in etwa äquimolaren Mengen, bezogen auf
die racemischen Verbindungen Ia ein.
Als Lösungsmittel können alle Lösungsmittel und Lösungsmittelgemische in
denen die beiden Komponenten in der Wärme löslich sind, verwendet werden,
wobei Alkohole wie Methanol und Ethanol, Ketone wie Aceton und Wasser sowie
Wasser enthaltende Lösungsmittelgemische bevorzugt sind.
Die benötigte Menge an Lösungsmittel ist abhängig von der Art des Lösungsmittels
sowie von der Art der zu kristallisierenden diastereomeren Salze.
Geeignete Lösungsmittel sowie die benötigte Menge können durch einige Vorversuche
leicht ermittelt werden.
Die Freisetzung des Amins erfolgt unter Verwendung einer starken Base, wie
eines Alkalimetallhydroxides, wovon Natriumhydroxid bevorzugt ist. Die
Base wird vorzugsweise in äquimolaren Mengen oder in geringem Überschuß,
bezogen auf die Verbindungen Ia, eingesetzt.
Die Hydrogenolyse der Benzylgruppe kann man nach an sich bekannten
Methoden (Houben Weyl, "Methoden der organischen Chemie", (1957),
Band 11/1, Seite 960 ff) durchführen.
Die Abspaltung der Benzylgruppe erfolgt vorteilhaft durch katalytische
Hydrierung in Gegenwart von 0,1 bis 10 Gew.-%, bezogen auf die aktive
Masse Hydrierkatalysator pro Mol Ia. In der Regel werden Platinoxid,
Raney-Nickel oder Palladiumkatalysatoren verwendet, wovon die Palladiumkatalysatoren
bevorzugt sind. Vorzugsweise werden solche Palladiumkatalysatoren
eingesetzt, bei denen das Palladium auf einen Trägerstoff mit
großer Oberfläche aufgetragen ist. Geeignete Träger sind z. B. Aluminiumoxid,
Silicagel oder Kohle. Üblicherweise enthalten diese Katalysatoren
zwischen 0,1 und 10 Gew.-% Palladium.
Zweckmäßigerweise wird die Hydrierung bei erhöhtem Druck von etwa 5 bis
30 bar, vorzugsweise von 5 bis 20 bar, durchgeführt.
Es empfiehlt sich, die Reaktion bei Temperaturen von 0 bis 120°C, insbesondere
von 60 bis 100°C vorzunehmen.
Als Lösungsmittel für die Hydrierung, die in Mengen von etwa 0,5 bis 2,5,
vorzugsweise 0,8 bis 2 l pro mol Ia verwendet werden, kommen insbesondere
Alkohole wie Methanol, Ethanol, Propanol und Ether wie Diethylether,
Tetrahydrofuran und Dioxan in Betracht.
Die optisch aktiven Verbindungen Ib sind wichtige Ausgangsprodukte für die
Synthese von Arzneimitteln, beispielsweise von blutdrucksenkenden Mitteln
wie (-)-5-[(1R)-1-Hydroxy-2-[[(1R)-1-methyl-3-phenylpropyl]amino]ethyl]--salicylamid
(Dilevalol), indem man 3-Amino-1-phenylbutan mit
3-[(1R)-1-Hydroxy-2-bromethyl]-salicylsäureamid umsetzt.
In einzelnen Fällen kann es günstig sein, eine Substitution am Stickstoff,
zunächst mit den Verbindungen Ia durchzuführen und die Benzylgruppe erst
anschließend abzuspalten. Hierdurch kann man eine unerwünschte Zweifachsubstitution
am Stickstoffatom verhindern.
24 g (0,1 mol) racemisches N-Benzyl-3-amino-1-phenylbutan und 15,2 g
(0,1 mol) (-)-Mandelsäure wurden bei 80°C in einer Mischung aus 0,5 l
Wasser und 0,2 l Ethanol gelöst. Nach dem Abkühlen der Lösung
kristallisierte das (+)-N-Benzyl-3-amino-phenyl-butan-(-)-mandelat aus.
Nach üblicher Aufarbeitung und Freisetzung des Amins mit 0,1 m wäßriger
Natriumhydroxidlösung fielen das (+)-3-Benzylamino-1-phenylbutan und das
(-)-3-Benzylamino-1-phenylbutan in Ausbeuten von 10,5 g (87,5% d. Th.) bzw.
10,8 g (90% d. Th.) an.
275,7 g R-(+)-3-Benzylamino-1-phenylbutanhydrochlorid wurden in Gegenwart
von 15 g Pd/C (5 Gew.-% Pd) bei 80 bis 100°C und einem Wasserstoffdruck
von 6 bar 150 min hydriert. Anschließend wurde der Katalysator abgetrennt,
und das Lösungsmittel wurde abdestilliert. Nach der Umkristallisation aus
300 ml Essigester erhielt man 171 g R-(+)-3-Aminophenylbutan (92% d. Th.)

Claims (1)
- Verfahren zur Herstellung von optisch aktiven 3-Benzylamino-1-arylbutanen und 3-Amino-1-arylbutanen der allgemeinen Formeln Ia und Ib in denen Ar für einen aromatischen Rest steht, dadurch gekennzeichnet, daß man
- a) zur Herstellung der Verbindungen Ia racemisches Ia mit einer optisch aktiven Säure (II) in ein diastereomeres Salzpaar überführt, dieses durch fraktionierte Kristallisation in die beiden diastereomeren Salze trennt, von diesen die optisch aktive Säure (II) mittels einer starken Base wieder abtrennt und
- b) zur Herstellung der Verbindungen Ib die gemäß Schritt (a) erhältlichen, optisch aktiven Verbindungen Ia durch katalytische Hydrierung in die Verbindungen Ib und Toluol zerlegt.
