Description
- Castanospermin ist ein Alkaloid der folgen den Formel:
- Systematisch wird diese Verbindung als (1S,6S,7R,8R,8aR)-1,6,7,8- tetrahydroxyindolizidin bezeichnet.
- Die Isolierung dieser Verbindung aus Castanospermum australe wurde von L. D. Hohenshutz et al., Phytochemistry 20, 811 (1981) beschrieben. Das Verfahren umfaßt eine langwierige Extraktion und nachfolgendes Waschen mit Pyridin, und nur geringe Mengen des erwünschten Produkts werden erhalten. R. C. Bernotas et al., Tetrahedron Letters 25, 165 (1984) erhielten Castanospermin durch eine Totalsynthese und etablierten dadurch die absolute Konfiguration der Verbindung.
- Mehrere Veröffentlichungen erschienen, die Castanospermin als Inhibitor einer Reihe von Pflanzenenzymen beschreiben. Siehe Y. T. Pan et al., Biochemistry 22, 3975 (1983); R. Saul et al., Archives of Biochemistry and Biophysics 221, 593 (1983); R. Saul et al., Archiv es of Biochemistry and Biophysics 230, 668 (1984). Kürzlich zeigten sich nach Saul et al., Proc. Nat'l. Acad. Sci. USA 82, 93 (1985) Hinweise, daß Castanospermin intestinale Glykosidasen (d. h. Maltase und Saccharase) in Ratten inhibiert, wenn es durch Injektion verabreicht wird, und daß Diarrhoe und hohe Konzentrationen bakterieller Darmflora beobachtet wurden, wenn hohe Dosen verabreicht wurden. Sie wiesen jedoch nicht darauf hin, daß als Ergebnis dieser Tatsachen Castanospermin für irgendwelche Zwecke verwendbar ist.
- Es wurde auch gefunden, daß Castanospermin Verdauungsenzyme inhibiert, und somit als Diabetesmittel und als Inhibitor einer erhöhten Lipidbiosynthese verwendet werden kann. Werden Kohlenhydrate entweder als Glucose oder in Form von Maltose, Saccharose oder Stärke in Nahrung oder Getränken aufgenommen, so steigt der Glucosespiegel im Blut auf erhöhte Konzentrationen. In gesunden Personen normalisiert sich dieser hyperglykämische Zustand rasch wieder, wobei die Glucose im Blut schnell metabolisiert und gespeichert und/oder vom Organismus verwertet wird. Bei Diabetes mellitus ist jedoch die Glucosetoleranz des Patienten herabgesetzt, und ungewöhnlich hohe Blutzuckerspiegel, die sich entwickeln, bleiben für eine längere Zeitdauer erhöht. Eine ähnliche Antwort, wie die beim Menschen vorkommende, kann auch bei anderen Tieren, umfassend Vieh, Geflügel, Haustiere und Labortiere, beobachtet werden. Dieser Zustand kann als postprandiale Hyperglykämie beschrieben werden. Ein Verfahren zur Behandlung eines solchen Zustandes ist die Verabreichung einiger Substanzen, die die Umwandlung komplexer Zucker zu Glucose verhindern und somit die Entwicklung übermäßiger Glucosespiegel verhindern. In der vorliegenden Erfindung wurde gefunden, daß in dem Fall, in dem die hohen Glucosespiegel eine Folge der Hydrolyse komplexer Zucker sind, die Verabreichung von Castanospermin die anfängliche Bildung von Glucose im Blut inhibiert und somit eine Vermeidung der Probleme ermöglicht, die mit länger andauernden hohen Blutzuckerspiegeln verknüpft sind.
- Zu Castanospermin sind die Salze von Castanospermin mit pharmazeutisch verträglichen Säuren und im besonderen mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure und Phosphorsäure; und mit organischen Säuren, wie Essigsäure, Propionsäure, Methansulfonsäure und p-Toluolsulfonsäure für diese Zwecke äquivalent.
- Die vorliegende Erfindung betrifft ein neues Verfahren zur Isolierung von Castanospermin aus Castanospermum australe durch ein Verfahren, das das in dem vorstehend erwähnten veröffentlichten Verfahren beschriebene, langwierige Extraktionsverfahren und die Verwendung des unangenehmen Lösungsmittels Pyridin vermeidet, während es das erwünschte Produkt in deutlich verbesserten Gesamtausbeuten bereitstellt. Einzelheiten dieses Verfahrens sind in dem Beispiel aufgezeigt.
- Das folgende Beispiel wird zur Veranschaulichung der vorliegenden Erfindung gezeigt.
- 300 g (Naßgewicht) von im Handel erhältlichen reifen Samen von Castanospermum australe wurden gemahlen und 24 Stunden lang in einem Soxhlet-Apparat mit 1,0 l eines 3 : 7 (v/v) Gemisches auf Wasser und 2-Propanol kontinuierlich extrahiert. Das Extrakt wurde unter vermindertem Druck bis zu einem Endvolumen von 250 ml eingeengt und filtriert. Nach der Extraktion mit 10 · 100 ml Petrolether wurde das wäßrige Extrakt einer Säulenchromatographie mit Dowex 50W-X4 (H&spplus;)-Ionenaustauscherharz (2,5 cm · 32 cm) unterzogen. Die Säule wurde mit 200 ml destilliertem Wasser gewaschen, und das gewünschte Alkaloid wurde mit 400 ml 2 N wäßrigem Ammoniumhydroxid eluiert. Das basische Eluat wurde unter vermindertem Druck auf 125 ml eingeengt und auf eine weitere Säule mit Dowex 1-X4 (OH&supmin;)-Ionenaustauscherharz (2,5 cm · 20 cm) aufgetragen. Die Säule wurde mit 125 ml Wasser eluiert, und das Eluat wurde auf ein Endvolumen von 30 ml eingeengt. Durch das Verreiben des Konzentrats mit 300 ml Aceton entstanden 3,5 g Castanospermin als farblose Kristalle (1,2%, Berechnung bezogen auf das Gewicht frischer Samen). Die isolierten Kristalle wiesen folgende physikalische Eigenschaften auf:

Claims (2)
1. Verfahren zur Isolierung von Castanospermin, d. h. von
1S,6S,7R,8R,8aR-1,6,7,8-tetrahydroxyindolizidin der Formel
oder eines Salzes davon mit einer pharmazeutisch
verträglichen Säure aus den Samen von Castanospermum australe, das
umfaßt:
(a) Extrahieren der gemahlenen Samen mit einem Gemisch
aus Wasser und 2-Propanol;
(b) Waschen des Extrakts mit Petrolether;
(c) Chromatographieren der Lösung auf einem sauren Harz
unter Eluieren mit Ammoniumhydroxid;
(d) Erneutes Chromatographieren des Eluats auf einem
basischen Harz unter Eluieren mit Wasser; und
(e) Einengen des wäßrigen Eluats und anschließendes
Verreiben mit Aceton, wobei sich Castanospermin ergibt.
2. Verfahren nach Anspruch 1, wobei die Extraktion mit
einem 3 : 7 (v/v) Gemisch aus Wasser und 2-Propanol
durchgeführt wird.
