Description
Beschreibung
Die Erfindung betrifft neue mehrfach ungesättigte Cyclopenten-Derivate
der allgemeinen Formel I,
worin R, eine niedere Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, Rp eine Hydroxymethyl-, Acetoxymethyl- oder Formylgruppe,
die geschlängelte Linie am C-Atom 1 epimere Formen und die gestrichelt gezeichnete Linie eine fakultative Bindung
bedeuten.^
Die Erfindung betrifft zudem die Verwendung von Verbindungen der allgemeinen Formel I als Riechstoffe und somit auch Riechstoffkompositionen,
gekennzeichnet durch einen Gehalt an einer Verbindung der allgemeinen Formel I sowie die Herstellung solcher
Verbindungen.
Es wurde gefunden, daß die neuen mehrfach substituierten Cyclopenten-Derivate
der allgemeinen Formel I wertvolle und stabile Riechstoffe sind. Insbesondere weisen die in der Seitenkette
gesättigten oder ungesättigten Derivate mit R1 = Methyl, Äthyl
oder n-Propyl sowie R2 = Hydroxymethyl ausgeprägte Sandelholznoten
auf.
Das in der Parfümerie vielfach eingesetzte ostindische Sandelholzöl
enthält hauptsächlich die Sesquiterpene α- und ß-Santalol
(A, bzw. B), die auch den charakteristischen mild-holzigen Geruch des Öles bewirken.

Die begrenzte Verfügbarkeit und der hohe Preis des Sandel^-
holzöles veranlaßten die Entwicklung von chemischen Synthesen für A, B, die jedoch industriell nicht ausgewertet werden
konnten. Als synthetische Ersatzprodukte werden zur Zeit hauptsächlich Terpenylcyclohexanole verwendet (Übersicht in:
I. S. Aulchenko und L.A. Kheifits, American Perfumer and Cosmetic
1970/ 37), die durch säurekatalysierte Umsetzung von Camphen
mit Phenol-Derivaten und nachfolgende katalytische Hydrierung erhalten werden. Hierbei entstehen komplexe Gemische, die nur
zum geringen Teil aus wirksamen Riechstoffen vom Sandelholz-Typ bestehen und häufig durch Terpenylphenole verunreinigt
sind.
Es wäre grundsätzlich von der Überlegung auszugehen, daß die Erzielung einer möglichst natürlichen Sandelholznote von Verbindungen zu erwarten ist, die den oben gezeigten Santalolen
A und B strukturell ähnlich sind und eine vergleichbare Molekülgröße bzw. Kohlenstoffzahl aufweisen. Dies bedingt allerdings
den Aufbau eines verhältnismäßig komplizierten Ringsystems, was, wie oben gezeigt, komplizierte Verfahren und Reinigungsmaßnahmen oder die Einführung verhältnismäßig langer Seitenketten
mit entsprechender Partialstruktur an ein einfacheres Grundgerüst erfordert. Bei den zuletzt genannten Verfahren waren
die Ergebnisse hinsichtlich der Geruchsnuancen wenig erfolgreich (Übersicht in G. Ohloff, Fortschr. Chem. Forsch., Bd. 12/2,
Seite 212). So ist in der US-PS 4 052 341 neben höhermolekularen

bzw. längerkettigen Verbindungen auch die Verbindung 3-Methyl-5-(2,2,3-trimethylcyclopent-3-en-l-yl)-pentan-2-ol
gezeigt, also eine Verbindung, bei welcher an das einfache Trimethylcyclopenten-Ringgerüst
eine längere Seitenkette, eine sekundäre Hydroxylgruppe enthaltend, gebunden ist. Das für die technische Darstellung dieser
Substanz angegebene Verfahren ergibt ein Isomerengemisch/ das in größerem Anteil 6-(trimethyl-cyclopent-3-en-l-yl)-hexan-3-ol
enthält, so daß Geruchs-Intensität und -Qualität des Produkts weniger befriedigend sind. Die genannte Patentschrift sagt aus,
daß man längere Ketten und sekundäre Hydroxylgruppen einführen muß, um die gewünschten Geruchsnoten zu erzielen. Es wurde nun
aber überraschenderweise gefunden, daß die Partialstruktur mit primärer Hydroxylgruppe (1-Hydroxy-2-methyl-alkenyl) der Santalole
auch in Kombination mit dem Trimethylcyclopentengerüst ausgezeichnete
Geruchsnote ergibt, wenn man kürzere Seitenketten einführt, also insgesamt die Kohlenstoffzahl der Verbindung verringert.
Damit kann man als Ausgangsmaterial Campholenaldehyd (1) verwenden,
der einen Aufbau geeigneter Seitenketten ermöglicht und nach bekannter Vorschrift (z.B. L.C. King und H. Farber, J. Org.
Chera. 26.t 326 [1961]) durch Umlagerung von a-Pinen-epoxid auch
in großen Mengen erhalten werden kann. Aus den natürlich vorkommeurlen
optischen Antipoden des a-Pinens resultieren die enantiomeren Formen des Campholenaldehyds, die einzeln oder
als Geraisch eingesetzt werden können. Der Aufbau der Seitenketten
wird nach an sich bekannten Methoden durchgeführt (Schemata 1-3) und ergibt im Gegensatz zur US-PS 4052 341 einheitliche Produkte.

Durch Wittig-Reaktion von 1 mit a-Brom-alkansäure-estern mit
3 bis 5 Kohlenstoffatomen im Alkansäureteil (einschließlich der Carboxylgruppe), vorzugsweise den Äthylestern, nach an sich
bekannten Verfahren (HeIv. Chim. Acta 4JD, 1242 [1957]) können die
ungesättigten Verbindungen der Formel I leicht hergestellt werden. So wurden 2a, b und c des nachfolgenden Schemas auf diese Weise
hergestellt und aus diesen Verbindungen durch katalytische Hydrierung 3a, b und c erhalten. Reduktion der Ester, beispielsweise
mit Alkali- oder Erdalkalimetallen in Alkoholen oder mit gemischten Hydriden von Metallen der 1. und 3. Hauptgruppe, ergab
die Carbinole 4a, b und c bzw. 5a, b und c, die einen intensiven Sandelholzgeruch aufweisen. Die erwähnten Reduktionsmethoden sowie
die katalytische Hydrierung sind an sich bekannt und bedürften keiner näheren Erläuterung. Sie werden aber auch in den später
folgenden Herstellungsbeispielen noch gezeigt.

Ein weiterer Syntheseweg geht aus von der basenkatalysierten Aldolkondensation von 1 mit aliphatischen C^ bis C^ Aldehyden
(Schema 2). Die auf diese Weise erhaltenen ungesättigten Aldehyde 6a - c wurden zu den Ällylalkoholen 4a - c reduziert,
vorzugsweise unter Verwendung von Natrxumborhydrid, bzw. durch
katalytische Hydrierung zu 7a - c und nachfolgende Reduktion in die gesättigten Carbinole 5a - c überführt. Auch diese
Reaktionen sind an sich bekannt.
Die Aldehyde 6a - c sind auch durch Umsetzung von 1 mit substituierten
Vinylalkyläthern unter BF3-Katalyse zugänglich
(analog S. Satsumabayshi, K. Nakajo, R. Soneda und S. Motoki, Bull. Chem. Soc. Japan 4_3, 1586 [1970].

Die Verbindungen der allgemeinen Formel I zeichnen sich durch
besondere Riechstoffeigenschaften aus, insbesondere durch mildholzige, grün-holzige, auch etwas leicht harzige fruchtige,
esterartige Geruchsnoten. Vor allem die Alkohole 4a - c und 5a - c zeichnen sich durch zum Teil intensive Sandelholznoten aus;
so übertrifft 5a das strukturanaloge 3-Methyl-5-(2,2,3-trimethylcyclopent-3-en-1-yl)-penten-2-ol
(US-PS 4 052 341) in Geruchsintensität und somit auch in der Ergiebigkeit in Kompositionen.
Verbindungen der allgemeinen Formel I können demgemäß als Riechstoffe und als Bestandteil von Riechstoffkompositionen wie Parfüms
und Parfumbasen bzw. zur Parfümierung kosmetischer und technischer Produkte aller Art verwendet werden.
Die folgenden Beispiele zeigen die Herstellung der erfindungsgemäßen

1-(3-Carbäthoxy-but-2-enyl)-2,2,S-trimethyl-cyclopeht-B-en . (2a)
150 g {1 Mol) Campholenaldehyd (1) und 470 g (2-Carbäthoxyäthyliden)-triphenylphosphoran
in 4,5 1 Benzol wurden unter N„ 6 Stunden bei Siedetemperatur gerührt. Nach Abkühlung,
Waschen mit Wasser und Abdestillieren des Benzols wurde in Petroläther aufgenommen und filtriert. Aus dem nach Abdestillieren
des Lösungsmittels verbleibenden Rohprodukt (21 ο g gelbes Öl) wurde durch fraktionierte Destillation 176 g (75 %) 2a vom
Kp. 8 105 - 108°C gewonnen.
1-(3-Hydroxymethyl-but-2-enyl)-2,2,3-trimethyl-cyclopent-3-en (4a)
50 g (0,21 Mol) 2a in 200 ml absol. Äther wurden bei 15°C zu
einer Aufschlämmung von 9 g LiAlH. in 250 ml absol Äther getropft.
Nach 4 Stunden Rühren bei Raumtemperatur wurden 20 ml Eisessig zugegeben.
Das Reaktionsgemisch wurde auf ca. 300 g Eis gegossen, aufgearbeitet
und fraktioniert. Erhalten wurden 32 g (83 %) 4a vom

1- (3-Carbäthoxy-butyl)-2,2,3-trimethyl-cyclopent-3-en (3a)
Eine Lösung von 100 g (0,42 Mol) 2a in 300 ml Methanol wurde mit 5 g Raney-Nickel unter Normalbedingungen hydriert (ca.

10 Stunden). Filtration, Abdestillieren des Lösungsmittels und
Fraktionierung ergaben 94 g (94 %) 3a vom Kp.Q _, 92 - 95 C.
1-(3-Hydroxymethyl-butyl-2,2,3-trimethyl-cyclopent-3-en (5a)
Aus 50 g 3a wurden nach dem im Herstellungsbeispiel gegebenen Vorschrift 35 g (85 %) 5a erhalten.
1-(3-Formyl-2-alkenyl)-2,2,3-trimethyl-cyclopent-3-ene (6a - c)
Bei ca 00C wurden Suspensionen von 17g Natriummethylat in
450 ml Methanol 368 g (2,4 Mol) Campholenaldehyd (1) zugegeben. Innerhalb von 1 Stunde wurden jeweils ca. 5 Mol des Aldehyds
(Propion-, Butyr- und n-Valeraldehyd) zugetropft (Innentemperatur
^ 20 C) Nach 1 Stunde Rühren bei Raumtemperatur Ansäuern mit jeweils 50 ml Eisessig, Abdestillieren des Methanols,
Aufarbeitung und fraktionierter Destillation verblieben 60 bis 80 % des entsprechenden Aldehyds (8a - c).
1-(3-Hydroxymethyl-2-alkenyl)-2,2,3-trimethylcyclopent-3-ene (4a - c)
Zu Lösungen von jeweils 150 g der Aldehyde 6a - c (aus Herstellungsbeispiel
5) in 100 ml Äthanol werden bei 35 - 40°C jeweils Lösungen von 38 g Natriumborhydrid und 0,6 g NaOH in 75 ml Wasser innerhalb
von 30 Minuten zugetropft. Nach 3 Stunden Rühren bei Raum-

temperatur Aufarbeitung und Destillation lagen die Alkohole 4a - c in Ausbeuten von 75 - 85 % vor.
1-(3-Formyl-2-alkyl)-2,2/3-trimethyl-cyclopent-ene (7a - c)
Jeweils 100 g der Aldehyde 6a - c (aus Herstellungsbeispiel 5) in 200 ml Methanol wurden mit 5 g Raney-Nickel 3 Stunden unter
Normalbedingungen hydriert. Filtration und Abdestillieren des Methanols erbrachten 80 bis 90 % der Aldehyde 7a - c.
t-(3-Hydroxymethyl-2-alkyl)-2,2,3-trimethyl-cyclopent-3-ene (5a - c)
Nach der im Herstellungsbeispiel 6 gegebenen Vorschrift wurden
aus den Aldehyden 7a - c jeweils in ca. 80 % Ausbeute die Alkohole 5a - c erhalten.
Nachstehende Beispiele zeigen die Verwendung der erfindungsgemäßen
Verbindungen.

Verfahren zur Herstellung von Verbindungen nach Anspruch 1 und 2, dadurch gekennzeichnet, daß man Campholenaldehyd in an sich bekannter Weise durch Wittig-Reaktion mit a-Brom-alkansäure-estern mit 3 bis 5 Kohlenstoffatomen umsetzt und die erhaltene Verbindung, gegebenenfalls nach an sich bekannter katalytischer Hydrierung der Doppelbindung in der Seitenkette, in an sich bekannter Weise zum entsprechenden Carbinol reduziert oder Campholenaldehyd in an sich bekannter Weise mittels basenkatalysierter Aldolkondensation mit aliphatischen Aldehyden mit 3 bis 5 Kohlenstoffatomen oder mit den entsprechenden substituierten Vinylalkyläthern unter BF3~Katalyse umsetzt und die auf diese Weise erhaltenen ungesättigten Aldehyde, gegebenenfalls nach an sich bekannter katalytischer Hydrierung der Doppelbindung der Seitenkette, gewünschtenfalls in an sich bekannter Weise zu den Allylalkoholen bzw. zu den gesättigten Carbinolen reduziert und gegebenenfalls in an sich bekannter Weise die Alkoholgruppe der Allylalkohole oder Carbinole mit einer Carbonsäure, insbesondere Essigsäure, verestert.909882/0273
