Die vorliegende Erfindung betrifft neue 1,4-Dihydropyridin-Zuckerderivate,
mehrere Verfahren zu ihrer Herstellung sowie ihre Verwendung als Arzneimittel, insbesondere als
kreislaufbeeinflussende Mittel.
Es ist bereits bekannt geworden, daß man 2,6-Dimethyl-4-phenyl-1,4-dihydropyridin-3,5-dicarbonsäure-diäthylester
erhält, wenn man Benzaldehyd mit ß-Amino-crotonsäureäthylester und Äcetessigsäureäthylester umsetzt (Literaturstelle:

Weiterhin ist bekannt, daß bestimmte 1,^Dihydropyridine
interessante pharmakologische Eigenschaften aufweisen (F.Bossert, W.Vater, Die Naturwissenschaften 58, 578 (1971) ).
Die Erfindung betrifft neue 1^-Dihydropyridin-Zucker-Derivate
der Formel I,
in welcher
R für einen Arylrest steht, der gegebenenfalls 1 bis 3 gleiche oder verschiedene Substituenten aus der Gruppe
Phenyl, Alkyl, Alkenyl, Alkinyl, Alkoxy, Alkenoxy, Alkinoxy, Halogen, Trifluormethyl, Trifluormethoxy,
Hydroxy, Amino, Alkylamino, Nitro, Cyano, Azido, Carbal koxy, Carbonamido, Carboxy, Sulfonamido oder SO -Alkyl
(m = 0 bis 2) enthält,

Die neuen erfindungsgemäßen 1,4-Dihydropyridin-Derivate besitzen
wertvolle pharmakologische Eigenschaften. Auf Grund ihrer Kreislaufbeeinflussenden Wirkung können sie als
antihypertensive Mittel, als Vasodilatatoren sowie als
Coronartherapeutika Verwendung finden und sind somit als
Bereicherung der Pharmazie anzusehen.

Je nach der Art der verwendeten Ausgangsstoffe kann die
Synthese der erfindungsgemäßen Verbindungen durch folgende Formelschemata wiedergegeben werden, wobei 2-(1,4-Dihydrο-5-methoxycarbonyl-2,6-dimethyl-4-o-nitrophenyl-3-pyridylcarbonyloxy)-äthyl-ß-D-glucopyranosid und 3-0-(l,4-Dihydro-5-methoxycarbonyl-2,6-dimethyl-4-m-nitrophenyl-3-pyridylcarbonyl)-D-glucose als Beispiele gewählt seien.
Synthese der erfindungsgemäßen Verbindungen durch folgende Formelschemata wiedergegeben werden, wobei 2-(1,4-Dihydrο-5-methoxycarbonyl-2,6-dimethyl-4-o-nitrophenyl-3-pyridylcarbonyloxy)-äthyl-ß-D-glucopyranosid und 3-0-(l,4-Dihydro-5-methoxycarbonyl-2,6-dimethyl-4-m-nitrophenyl-3-pyridylcarbonyl)-D-glucose als Beispiele gewählt seien.

Die als Ausgangsstoffe verwendeten Verbindungen der Formel IV sind bisher nicht bekannt, können aber nach literaturbekannten
Verfahren zur Darstellung von ß-Ketosäureestern hergestellt werden (Vgl. z. B. Houben-Weyl, Methoden der organischen Chemie
VII/4, 230 ff (1968) )

Die Reaktion mit Diketen wird bevorzugt ohne Lösungsmittel bei Temperaturen zwischen 80 und 14O°C und Normaldruck
durchgeführt.
Neu sind auch die Enaminoverbindungen der Formel V. Sie können durch Wasserabspaltung aus ß-Ketoestern der Formel IV
und primären Aminen auf literaturbekanntem Wege hergestellt werden : (Vgl. A.C. Cope, J. Amer. Chem. Soc. 67, 1017 (1945))

Die erfindungsgemäß verwendbaren Amine der Formel IV sind bereits bekannt. Als Beispiele seien genannt :
Ammoniak, Methylamin, n-Propylamin, Isopropylamin, n-Butylamin,
sec-Butylamin, Isobutylamin, ß-Methoxyäthylamin,
Benzylamin, % Anilin.
Zur Herstellung der erfindungsgemäßen Verbindungen nach den
Verfahrensvarianten A) bis F) kommen als Verdünnungsmittel Wasser und inerte organische Lösungsmittel in Frage. Hierzu
gehören vorzugsweise Alkohole wie Aethanol, Methanol, Isopropanol,
Aether wie Dioxan, Diäthyläther, Tetrahydrofuran, Glykolmonomethyläther, Glykoldimethyläther oder Eisessig,
Dimethylformamid, Dimethylsulfoxid, Acetonitril, Pyridin
und Hexamethylphosphorsäuretrxamxd.
Die Reaktionstemperaturen können in einem größeren Bereich variiert werden. Im allgemeinen arbeitet man zwischen 200C
und 1500C, vorzugsweise bei der Siedetemperatur des jeweiligen
Lösungsmittels.
Die Umsetzung kann bei Normaldruck aber auch bei erhöhtem Druck durchgeführt werden. Im allgemeinen arbeitet man unter
Normaldruck.

Bei der Durchführung des erfindungsgemäßen Verfahren werden die an der Reaktion beteiligten Stoffe im allgemeinen in
etwa äquimolaren Mengen eingesetzt.
Bei den Verfahrensvarianten C) und D) wird das Amin der
Formel VIII vorzugsweise im Ueberschuß eingesetzt.
Gegebenenfalls, vorhandene Schutzgruppen der Zuckerreste lassen
sich nach an sich bekannten Verfahren entfernen. Beispielsweise werden Acylschutzgruppen vorzugsweise durch Umesterung unter
basischen Bedingungen (z.B. mit katalytischen Mengen NaOCH., in Methanol; Zemplen-Methode) bei Raumtemperatur entfernt, Acetal-
oder Ketalschutzgruppen durch Hydrolyse in Wasser/Alkohol- oder Wasser/Dioxangemischen unter sauren Bedingungen bei
Temperaturen zwischen 60° und 1000C
(Vgl. z. B. Whistler, Wolfram, Methoden in Carbohydrate Chemistry II, S. 215 ff u. S. 318 ff)
Eine Reinigung der rohen Wirkstoffe erfolgt vorzugsweise mit Hilfe chromatographischer Methoden. Im Falle der Verbindungen
der Formel I mit ungeschützten Zuckerresten wird
eine Säulenchromatographie an Cellulosepulver oder an Kieselgel
bevorzugt. Als Fließmittel werden Butanol bzw. Butanol/Wassergemische
oder Chloroform/Äthanolgemische bevorzugt verwendet.

* Die neuen Verbindungen sind als Arzneimittel verwendbare
Substanzen. Sie haben ein breites und vielseitiges phannakologisches
Wirkungsspektrum.
Im einzelnen konnten im Tierexperiment folgende Hauptwirkungen
nachgewiesen werden :
1) Die Verbindungen bewirken bei parenteraler, oraler und perlingualer Zugabe eine deutliche und langanhaltende
Erweiterung dir Coronargefäße.

Diese Wirkung auf die Coronargefäße wird durch einen
gleichzeitigen Nitritähnlichen herzentlastenden Effektverstärkt.
Sie beeinflussen bzw. verändern den Herzstoffwechsel im Sinne einer Energieersparnis.
2) Die Erregbarkeit des Reizbildungs- und Erregungsleitungssystems
innerhalb des Herzens wird herabgesetzt, so daß eine in therapeutischen Dosen nachweisbare Antiflimmerwirkung
resultiert.
3) Der Tonus der glatten Muskulatur der Gefäße wird unter
der Wirkung der Verbindungen stark vermindert. Diese gefäßspasmolytische Wirkung kann im gesamten Gefäßsystem
stattfinden, oder sich mehr oder weniger isoliert in umschriebenen Gefäßgebieten (wie z.B. dem Zentralnervensystem)
manifestieren.
4) Die Verbindungen senken den Blutdruck von normotonen
und hypertonen Tieren und können somit als antihypertensive Mittv?l verwendet werden.
5) Die Verbindungen haben stark muskulär-spasmolytische Wirkung, dies an der glatten Muskulatur des Magens, Darmtra'stes,
des Urogenitaltraktes und des Respirationssystems deutlich werden.
Die erfindungsgemäßen Verbindungen eignen sich aufgrund dieser
Eigenschaften besonders zur Prophylaxe -und Therapie der akuten und chronischen ischämischen Herzkrankheit im weitesten Sinne,
zur Therapie' des Hochdrucks sowie zur Behandlung von cerebralen und peripheren Durchblutungsstörungen.

Die neuen Wirkstoffe können in bekannter Weise in die üblichen Formulierungen übergeführt werden, wie Tabletten,
Kapseln, Dragees, Pillen, Granulate, Aerosole, Sirupe, Emulsionen, Suspensionen und Lösungen, unter Verwendung
inerter, nichttoxischer pharmazeutisch geeigneter Trägerstoffe oder Lösungsmittel. Hierbei soll die therapeutisch
wirksame Verbindung jeweils in einer Konzentration von etwa 0,5 ibs 90 Gewichtsprozent der'Gesamtmischung vorhanden
sein, d.h. in Mengen, die ausreichend sind, um den angegebenen Dosierungsspielraum zu erreichen.
Die Formulierungen werden beispielsweise hergestellt durch Verstrecken der Wirkstoffe mit Lösungsmitteln und/oder
Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z.B. im Fall der
Benutzung von Wasser als Verdünnungsmittel gegebenenfalls organische Lösungsmittel als Hilfslösungsmittel verwendet
werden können.
Als Hilfsstoffe seien beispielhaft aufgeführt :
Wasser, nichttoxische organische Lösungsmittel, wie Paraffine (z.B. Erdölfraktionen), pflanzliche OeIe (z.B. Erdnuß-/
Sesamöl),'Alkohole (z.B. Aethylalkohol, Glycerin), Glykole
(z.B. Propylenglykol, Polyäthylenglykol), feste Trägerstoffe, wie z.B. natürliche Gesteinsmehle, (z.B. Kaoline, Tonerden,
Talkum, Kreide), synthetische Gesteinsmehle (z.B. hochdisperse Kieselsäure, Silikate), Zucker (z.B. Roh-, Milch-
und Traubenzucker), Emulgiermittel, wie nichtionogene und anionische Emulgatoren (z.B. Polyoxyäthylen-Fettsäure-Ester,
Polyoxyäthy.Len-Fett-alkohol-Aether, Alkylsulfonate und

Arylsulfonate), Dispergiermittel (z.B. Lignin, Sulfitablaugen, Methylcellulose, Stärke und Polyvinylpyrrolidon)
und Gleitmittel (z.B. Magenesiumstearat, Talkum, Stearinsäure und Natriumlaurylsulfat).
Die Applikation erfolgt in üblicher Weise, vorzugsweise oral oder parenteral, insbesondere perlingual oder intravenös.
Im Falle der oralen Anwendung können Tabletten selbstverständlich außer den gena;inten Trägerstoffen auch Zusätze,
wie Natriumeitrat, Calciumcarbonat und Dicalciumphosphat zusammen mit verschiedenen Zuschlagsstoffen, wie Stärke,
vorzugsweise Kartoffelstärke, Gelatine und dergleichen enthalten. Weiterhin können Gleitmittel, wie Magnesiumstearat,
Natriumlaurylsulfat und Talkum zum Tablettieren mitverwendet werden. Im Falle wäßriger Suspensionen und/oder
Elixieren, die für orale Anwendungen gedacht sind, können die Wirkstoffe außer mit den obengenannten Hilfstoffen mit
verschiedenen Geschmacksaufbesserern oder Farbstoffen versetzt werden.
Für den Fall der parenteralen Anwendung können Lösungen der Wirkstoffe unter Verwendung geeigneter flüssiger Träger»
materialien eingesetzt werden.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei intravenöser Applikation Mengen von etwa 0, Ol bis 15 mg/kg
vorzugsweise etwa 0, 05 bis 1 mg/kg Körpergewicht pro Tag zur Erzielung wirksamer Ergebnisse zu verabreichen,
lind bei oraler Applikation beträgt die Dosierung etwa 0, 05 bis 10 mg/kg, vorzugsweise 0, 1 bis 5 mg/kg Körpergewicht
pro Tag.

Trotzdem kann es gegebenenfalls erforderlich, sein, von den
genannten Mengen abzuweichen, und zwar in Abhängigkeit vom Körpergewicht des Versuchstieres bzw. der Art des Applikationsweges,
aber auch auf Grund der Tierart und deren individuellem Verhalten gegenüber dem Medikament bzw. der
Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall zu welchem die Verabreichung erfolgt. So kann es in
einigen Fällen ausreichend sein, mit weniger als der vorgenannten Mindestmenge auszukommen, während in anderen
Fällen die genannte obere Grenze überschritten werden muß. Im Falle der Applikation größerer Mengen kann es empfehlenswert
sein, diese in mehrere Einzelgaben über den Tag zu verteilen. Für die Applikation in der Humanmedizin ist der
gleiche Dosierungsspielraum vorgesehen. Sinngemäß gelten hierbei auch die obigen Ausführungen-

2,4 g (5m Mol) (2-Acetonylcarbonyloxyäthyl)-2,3,4,6-tetra-0-acetyl-ß-D-glucopyranosid,
0,75 g (5 m Mol) m-Nitrobenzaldehyd und 0,72 g (5 m Mol) ß-Aminocrotonsäureisopropylester
werden in 10 ml Aethanol 24 Stunden zum Sieden erhitzt. Anschließend wird das Lösungsmittel am Rotationsverdampfer
entfernt.
Der Rückstand wird in 100 ml einer 0,l#igen Lösung von
Natriummethanolat in Methanol aufgenommen und 12 Stunden bei Raumtemperatur aufbewahrt.
Anschließend verdünnt man mit 30 ml Wasser, neutralisiert mit schwach saurem Ionenaustauscher (H+-Form), filtriert vom
Ionenaustauscher ab und bringt die Lösung am Rotationsverdampfer zur Trockne. Der Rückstand wird in wenig Butanol,
das mit Wasser, -gesättigt ist, gelöst und auf eine mit Cellulose (Avicel; Merck, Darmstadt) gefüllte Säule (Länge : 50 cm;
0 : 2,5 cm)-aufgetragen.
Als Fließmittel wird wassergesättigtes n-Butanol verwendet.
Es werden Fraktionen von jeweils 200 Tropfen gesammelt,, Die eineelnen Fraktionen werden nach dem Ergebnis einer dtlnnschichtchromatographischen
Prüfung auf Kieselgelplatten (Fließmittel : CHCl3/Aethanol,, 2 : 1) zusammengefaßt.

Zu 23 g (59 m Mol) (2-Hydroxyäthyl)-2,3,4,6-tetra-0-acetylß-D-giucopyranosid
gibt man 0,5 ml Triäthylamin und erwärmt auf 800C. Zu dieser Mischung tropft man unter Feuchtigkeitsausschluß und Rühren 5 ml (65 m Mol) Diketen. Dabei steigt die
Temperatur bis auf 1100C an. Kan erwärmt noch eine Stunde auf
1000C und versetzt den Rückstand nach dem Abkühlen mit Petroläther,
wobei das Reaktionsprodukt auskristallisiert. Ausbeute : 25,5 g.
Nach Umkristallisation aus Isopropanol schmilzt die Verbindung
bei 78-800C.

1,6 g (13,9 m Mol) ß-Aminocrotonsäuremethylester und 2,1 g (13,9 m Mol) m-Nitrobenzaldehyd werden in 30 ml
Aethanol 24 Stunden unter Rückfluß erhitzt.
Anschließend wird das Lösungsmittel am Rotationsverdampfer entfernt.
Das Rohprodukt wird durch Chromatographie auf eina^mit Al2O3' gefülltenSäule
gereinigt. Als Fließmittel dient Chloroform.
Die einzelnen Fraktionen werden dünnschichtchromatographisch auf Kieselgelplatten geprüft- (Fließmittel : Chloroform/Essigester
2:1; Rf-Wert : 0,5-0,6).
Ausbeute : 5,5 g.
Das nicht kristalline Produkt ist ein Gemisch zweier diastereomerer
Verbindungen. Eine teilweise Auftrennung der Diastereomeren erfolgt an der Al2O3-Säule.
Eine bessere Auftrennung wird durch Craig-Verteilung erreicht.

5,5 g des Diastereomerengemisches aus Beispiel 3 werden in 35 ml Aethanol, 25 ml H2O und 1 ml konz. Schwefelsäure 20
Minuten auf 600C erwärmt. Nach dem Abkühlen verdünnt man mit
40 ml Wasser. Den ausgefallenen Niederschlag saugt man ab,
wäscht ihn mit Wasser neutral und nimmt ihn in Chloroform auf. Die Chloroformlösung wird mit Na2SO4 getrocknet und
am Rotationsverdampfer zur Trockne gebracht. Der Rückstand wird in wenig Chloroform gelöst auf eine mit neutralem
Al2O3 beschickte Säule (Länge : 25 cm; 0 2,5 cm) aufgetragen.
Zuerst wird mit Chloroform, dann mit Chloroform/Aethanol 20 : 1, schließlich mit Chloroform/Aethanol 10 : 1 eluiert.
Die einzelnen Fraktionen werden durch Dünnschichtchromatographie auf Kieselgelplatten (Laufmittel : CHCl3/MeOH 10 : 1, RF-Wert:
0,4-0,5) geprüft.
Ausbeute : 3,5 g.
Das nicht kristalline Produkt ist ein Gemisch diastereoisomerer
Verbindungen. Eine teilweise Auftrennung der Diastereomeren erfolgt an der Al2O3-Säule, wenn die Säule mit nur sehr wenig
Substanz beladen und sehr kleine Fraktionen aufgefangen werden.
Eine Auftrennung der Diastereomeren erfolgt auch durch Kristallisation aus Dioxan/Wasser 1:1.
Das aus diesem Lösungsmittelgemisch auskristallisierende Diastereomere besitzt einen Festpunkt von 220-2210C.

2,8 g des Diastereomerengemisches aus Beispiel 3 werden in 15 ml 2 η H2SO^ xind 15 ml Dioxan 30 Minuten unter Rückfluß erwärmt.
Das Reaktionsgemisch wird mit Wasser verdünnt und mit BaCO, neutralisiert.
Es wird vom Niederschlag abgesaugt und das Filtrat am Rotationsverdampfer zur Trockne gebracht. Der Rückstand wird
auf eine mit Kieselgel 60 (Merck, Darmstadt) gefüllte Säule mit CHCl^/Äthanol 5:1 eluiert. Die einzelnen Fraktionen werden dünnschichtchromatographisch
auf Kieselgelplatten geprüft (Laufmittel: CHCWMeOH 10:1 oder CHCl^/MeOH 3:1,
Rf-Wert: 0,1 bzw. 0,6).
Rf-Wert: 0,1 bzw. 0,6).
Ausbeute: 1,1 g.

Zu 39 g (150 m Mol) !^^,ö-Di-O-isopropyliden-a-D-glucofuranose
gibt man 1,2 ml Triäthylamin und erwärmt auf 1000C. Man tropft I4r4 ml 94#iges Diketen (175 m Mol) zu, wobei die
Temperatur rasch auf 130°C ansteigt. Man rührt noch 15 Minuten bei 1000C nach. Anschließend wirddas Reaktionsgemisch mit
Petroläther versetzt und gut durchgerührt. Der Petroläther wird abdekantiert und die Reinigungsoperation mehrfach wiederholt.
Zum Schluß wird restlicher Petroläther am Rotationsverdampfer entfernt. Die Dünnschichtchromatographie auf
Kieselgelplatten mit CHCl3ZCH3COOCaH5. 2 : 1 als Lauf mittel
zeigt ein einheitliches Reaktionsprodukt. (Rf-Wert : 0,63).
Ausbeute : 49 g (nicht kristallin).

Ύ -Pyridylgruppe oder durch eine Aminogruppe substituiert
sein kann, wobei diQ.se Aminogruppe gegebenenfalls entweder Wasserstoff und einen Substituenten oder zwei gleiche
oder verschiedene Substituenten aus der Gruppe Alkyl, Alkoxyalkyl,
Aryl und Aralkyl trägt und wobei diese Substituenten gegebenenfalls mit dem Stickstoffatom einen 5 bis 7
gliedrigen Ring bilden, der als weiteres Heteroatom ein Sauerstoff oder Schwefelatom enthalten kann,
oder
für einen Arylrest steht, der gegebenenfalls 1 bis 3 gleiche oder verschiedene Substituenten aus der Gruppe Alkyl, Alkoxy,
Halogen, Cyano, Tr if luormethyl, Trifluormethoxy, Ainino, Alkylamino
oder Nitro enthält,
R2und I^ gleich oder verschieden sind und für Wasserstoff,
einen geradkettigen oder verzweigten Alkylrest, einen Arylrest oder einen Aralkylrest stehen.

wobei X für eine Alkylenkette aus 2 bis 4 Kohlenstoffatomen steht, an die Carbonylgruppe des 1,4-Dihydropyridinrests
gebunden ist, wobei die Bindung zum Zucker über alle Hydroxylgruppen der Zuckereinheit erfolgen
kann und sowohl eine Esterbindung als auch eine glykosidische Bindung sein kann.
4) Arzneimittel enthaltend ein kreislaufbeeinflußendes 1,4-Dihydropyridin-Zuckerderivat
gemäß Anspruch 1.
5) Verfahren zur Herstellung von Arzneimitteln,dadurch gekennzeichnet,
daß man 1,4-Dihydropyridin-Zuckerderivate gemäß Anspruch 1 mit Inerten, nichtioxischen pharmazeutisch
geeigneten Trägerstoffen vermischt.
