Die vorliegende Erfindung betrifft Acylderivate der allgemeinen Formel
H NH-CO-CHF2
CH0-SO0 ff \) C-C-CH0OR

worin R ein Wasserstoffatom oder einen
Glycylrest darstellt,
und Säureadditionssalze derjenigen Verbindungen, worin
R einen Glycylrest darstellt.
• Da die Derivate der Formel I und deren Salze zwei asymmetrische
Kohlenstoffatome enthalten, können sie in vier
stereoisomeren Formen oder als Racemate vorliegen.

Bevorzugte Verbindungen der Formel I sind das
Drthreo-l-[(p-Methylsulfonyl)~phenyl]-2-(2,2-difluoracetamido)·
1,3-propandiol (Fluorthiamphenicöl) und der entsprechende
^-.Glycinester.
Die Verbindungen der Formel I können in an sich bekannter Weise dadurch hergestellt werden, dass man die

oder ein Säureadditionssalz hiervon mit Difluoressigsäure oder einem funktionellen Derivat hiervon umsetzt, dass man,
gegebenenfalls, eine so erhaltene Verbindung der Formel I, worin R ein Wasserstoffatom bedeutet, zum entsprechenden
Glycinester verestert und dass man, gegebenenfalls, einen so erhaltenen Ester in ein Säureadditionssalz überführt.
Als reaktionsfähige funktionelle Derivate der Difluoressigsäure kommen z.B. Ester, z.B. Methyl- oder Aethylester;
Halogenide, nämlich Chloride, Bromide und Fluoride; Azide, Anhydride, besonders gemischte Anhydride mit stärkeren Säuren;
Amide, z.B. Imidazolide, und dergleichen, in Betracht.
Die Umsetzung einer Verbindung der Formel II mit Difluoressigsäure
oder einem reaktionsfähigen funktionellen Derivat davon kann in üblicher Weise durchgeführt werden. So kann man
z.B. die freie Difluoressigsäure mit einer Verbindung der Formel II mittels eines Kondensationsmittels, beispielsweise

eines Carbodiimide, wie Dicyclohexylcarbodiimid, in einem
inerten Lösungsmittel, wie Essigester, Acetonitril, Dioxan, Chloroform, Methylenchlorid, Benzol, Dimethylformamid kondensieren.
Anstelle von Carbodiimiden lassen sich als Kondensat!ons mittel
auch Oxazoliumsalze, z.B. N-Aethyl-5-phenyl-isoxazolium-3'-sulfonat,
verwenden.
In einer anderen AusfUhrungsform setzt man ein Salz einer
Verbindung der Formel II, z.B. das Hydrochlorid, mit einem reaktionsfähigen
funktioneilen Derivat der Difluoressigsäure in
einem Lösungsmittel, zweckmassig einem polaren Lösungsmittel, beispielsweise einem niederen Alkanol, wie z.B.. Methanol, um.
Die Umsetzung einer Verbindung der Formel II mit Di-.fluoressigsäure
oder einem funktioneilen Derivat davon kann zweckmassig bei Temperaturen zwischen etwa Raumtemperatur und
Rückflusstemperatur des Reaktionsgemisches durchgeführt werden.
Die Ueberführung einer Verbindung der Formel I, worin
R ein Wasserstoff.atom darstellt, in den entsprechenden Ester* worin R einen Glycylrest darstellt, kann in an sich bekannter
Weise durchgeführt, werden. So kann man beispielsweise das
freie Fluorthiamphenicol der Formel I mit Glycin, dessen Aminogruppe
in geschützter Form vorliegt, z.B. mit N-t-Butyloxycarbonyl-glycin,
in Gegenwart eines Kondensationsmittels, beispielsweise eines Carbodilmids, wie· Dicyclohexylearbodiimid,
in einem inerten Lösungsmittel, wie z.B. Acetonitril, .bei einer Temperatur zwischen etwa +50C und -1O°C, beispielsweise.bei etwa
O0C, umsetzen und anschliessend die Schutzgruppe durch Behandlung
mit einer verdünnten Säure, z.B. mit einem Gemisch von Salzsäure und Eisessig, abspalten.

Die Verbindungen der Formel I, worin R einen Glycylrest
darstellt, sind Basen und können mittels anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure,
Phosphorsäure und dergleichen und mit organischen Säuren, wie Essigsäure, Weinsäure, Maleinsäure, .Fumarsäure, Zitronensäure,
Oxalsäure, Toluolsulfonsäure und dergleichen in Säureadditionssalze
Übergeführt werden.
Optisch aktive Verbindungen der Formel I können sowohl ausgehend von dem entsprechenden Antipoden der Formel II als
auch durch Racematspaltung hergestellt werden. So kann man beispielsweise das Racemat der Formel I, worin R den Glycylrest
darstellt, mit einer optisch aktiven Säure, z.B. einem Antipoden der Camphersulfonsäure, Weinsäure oder dergleichen auftrennen.
Die Verbindung der Formel II ist eine bekannte Verbindung und kann in an sich bekannter Weise hergestellt werden.
Die Verbindungen der Formel I und deren Säureadditionssalze, insbesondere in der D-threo-Form, sind anti-
biotisch wirksam. Sie besitzen ein breites Wirkungsspektrum gegen grampositive und gramnegative Mikroorganismen,
insbesondere gegen Proteus und Pseudomonas aeruginosa. Die Verbindungen der Formel I und die pharma-.
zeutisch verwertbaren Salze davon können als Therapeutika und Desinfektionsmittel verwendet werden. Für den Erwachsenen
kommen Einzeldosierungen von ca. 1 g in Betracht. Während sich Fluorthiamphenicöl besonders für die orale Applikation
eignet, kommt dessen ^-Glycinester-hydrochlorid aufgrund seiner ausgezeichneten Wasserlöslichkeit für die parenterale
Applikation in Frage. Letzteres wird im Organismus zu freiem Fluorthiamphenicol hydrolytisch metabolisiert.
Die akute Toxizität (IiDc0 in mg/kg) des Fluorthiamphenicols
und dessen ^-Glycinester-hydrochlorids an der Maus bei intravenöser, subkutaner und oraler Verabreichung sowie

die Wirksamkeit (CD^0 in mg/kg) dieser beiden Substanzen
gegen Pseudomonas aeruginosa bei oraler Verabreichung an der Maus werden aus nachstehender Tabelle ersichtlich:

Pharmazeutische Präparate können die Verbindungen der Formel I und deren Salze in Mischung mit einem für die enteraie,
insbesondere orale; oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen
inerten Trägermaterial, wie z.B. Wasser, Gelatine, Gummi
arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche
OeIe, .Polyalkylenglykole, Vaseline, usw. enthalten.
Die pharmazeutischen Präparate können in fester Form* z.B.
als Tabletten, Dragees, Suppositorien, Kapseln; in halbfester Form, z.B. als Salben; oder in flüssiger Form, z.B» als
Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenen-, falls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe,
wie Konservierungs-, Stabilisi'erungs-, Netz- oder
Emulgiermittel, Salze zur Veränderung des osmotlschen Druckes
oder Puffer. . ·

Herstellung von D-threo-l-[(p-Methylsulfonyl)phenyl]-2-(2,2-difluoraoetamido)-lj3-PPopandiol
- 5*64 g D-threo-l-[(p-Methylsulfonyl)phenyl]-2-amino-1,3-propandiol-hydrochlorid
werden in 100 ml Methanol mittels 2,76 ml Triäthylamin in Lösung gebracht und mit 5*35 g
Difluoressigsäure-methylester versetzt. Das Reaktionsgemisch wird 10 Minuten unter Rückfluss erhitzt .und danach unter
vermindertem Druck eingedampft. Der ölige Rückstand wird in der minimalen Menge Wasser gelöst und zweimal mit Aethylacetat
extrahiert. Die vereinigten. Aethylacetatextrakte werden über
Magnesiumsulfat getrocknet und unter vermindertem. Druck eingeengt,
wobei die gewünschte Verbindung in reinem Zustand auskristallisiert. Das farblose Kristallisat wird abgenutscht,
mit Diäthyläther und mit Petroläther gewaschen und bei 300C unter vermindertem Druck getrocknet. Ausbeute 4,15 g
Schmelzpunkt 152-154°C, (a3^° = + l4,2° (c = 1,610 in Methanol)
Herstellung von D-threo-1-[(p-Methylsulfonyl)phenyl]- 2" (2t2-a±fl\iora.Qetasi\iao) -1 ,^■"
chlorid
9,7 g D-threo-1-[(p-Methylsulfonyl)phenyl]-2-(2,2-difluoracetamido)-l,3-propandiol,
5,25 g N-t-Butyloxycarbonyl-glycin
und 2,4 g Pyridin werden in 150 ml Acetonitril gelöst. Diese
Lösung wird bei 00C mit einer Lösung von 6,15 g Dicyclohexyl-carbodiimid
in 50 ml Acetonitril versetzt. Das Reaktionsgemisch wird 1/2 Stunde bei 00C und danach 20 Stunden
bei 200C stehen gelassen. Nach Abfiltrieren des Dicyclohexylharnstoffes
wird das Piltrat unter vermindertem Druck eingedampft.
Der resultierende Rückstand wird in Aethylacetat

gelöst und unter Eiszusatz nacheinander mit IN Schwefelsäure
und mit 10^-iger Natriumhydrogenearbonat-Lösung gewaschen.
Die Aethylacetatlösung wird dann über Magnesiumsulfat getrocknet, und unter vermindertem Druck eingedampft. Der als Rückstand
verbleibende helle Schaum (12,5 g) wird an einer Kieselgel-Säule mit Aethylacetat als Elutionsrnittel ehromatographiert,
wobei 4,05 g D-threo-l-[(p-Methylsulfonyl)-phenyl]-2-(2,2-difluoracetamido-l,3-propandiol~3-(N-t-butyloxycarbonyl)-glycinester
in reiner Form als farbloser Schaum erhalten werden.
4,0 g D-threo-l-[(p-Methylsulfonyl)phenyl]-2-(2,2-difluoracetamido)-l,3-propandiol-3-(Nrt-butyloxycarbonyl)-glycinester
werden in 150 ml einer 1,4N Salzsäure-Eisessig-Lösung gelöst und J50 Minuten stehen gelassen.' Das Reaktionsgemisch wird unter vermindertem Druck eingeengt und mit Diäthyläther
versetzt, wobei die gewünschte Substanz ausfällt. Diese wird dann aus Aethanol-Diäthyläther umgefällt, abgenutscht, mit Aether
gewaschen und bei 200C unter vermindertem Druck getrocknet.
Man erhält 5*25 g amorphe Reinsubstanz.als farbloses Pulver.
Schmelzpunkt oberhalb 12<
(c = 0,800 in Methanol).
(c = 0,800 in Methanol).
PO Schmelzpunkt oberhalb 1200C (Zersetzung). Γα]J = + 15,1°
Es wird in üblicher Weise eine Gelatine-Steckkapsel folgenden Inhaltes hergestellt:

Es wird in Üblicher Weise ein Lyophilisat folgender
Zusammensetzung hergestellt:
Fluorthiarnphenicol-^'-glycinester- l4;5Q mg
hydrochlorid
p-Hydroxybenzoesäure-methylester 2 mg
p-Hydroxybenzoesäure-propylester 0,2 mg
Dieses Lyophilisat wird in eine Ampulle abgefüllt. Eine derartige Ampulle enthält die lyophilisierten Wirkstoffe für
5,3 ml spritzfertiger Lösung. Zur Herstellung von 5,3 ml
spritzfertiger Lösung werden dem Lyophilisat 4 ml destilliertes Wasser zugesetzt.

Claims (1)
- Patentansprüche1. Verfahren zur Herstellung von Acylderivaten der allgemeinen FormelH NH-CO-CHF2CH-SO-V/ XS C-C-CH OR•5 ^ \ / I I 2OHHworin R ein Wasserstoffatom oder einen Glycylrest darstellt,und von Säureadditionssalzen derjenigen Verbindungen, worin R einen Glycylrest darstellt, dadurch gekennzeichnet, dass man die Verbindung der FormelH NH2CH0-SO0 { x) C-C-CHJDH nOHHoder ein Säureadditionssalz hiervon.mit Diflupressigsäure1 oder einem funktioneilen Derivat hiervon umsetzt, dass man, gegebenenfalls, eine so erhaltene Verbindung der Formel I^ worin R ein Wasserstoffatom bedeutet, zum entsprechenden Glycinester verestert und dass man, gegebenenfalls, einen so erhaltenen Ester in ein Säureadditionssalz überführt.■ 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Ausgangsmaterial der Formel II D-threo-1-[ (p-509822/1034 .Methylsulfonyl)-phenyl]-2-amino-l,3-propandiol-hydrochlorid verwendet.j5. Verfahren zur Herstellung von pharmazeutischen Präparaten, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen FormelH NH-CO-CHF2CH-SO-/ y C-C-CH2OROHHworin R ein Wasserstoffatom oder einen Glycylrest darstellt,oder, falls R einen Glycylrest darstellt·, ein pharmazeutisch verwertbares Säureadditionssalz davon als wirksamen Bestandteil^ mit zur therapeutischen Verabreichung geeigneten, nicht-toxischen, inerten, an. sich in solchen Präparaten Üblichen, festen und/oder flüssigen Trägern und/oder Excipientien vermischt.4. Pharmazeutische Präparate, gekennzeichnet durch einen Gehalt an einer Verbindung· der allgemeinen FormelH NH-CO-CHF2CH0-SO0 ff \) C-C-CH^OROHH509822/103worin R ein Wasserstoffatom oder einen Glycylrest darstellt,■ .oder, falls R einen Glycylrest darstellt, einem pharmazeutisch verwertbaren Säureadditionssalζ davon;zusammen mit einem pharmazeutischen Trägermaterial.· Racemische oder optisch aktive Acylderivate der allgemeinen FormelH NH-CO-CHF2C-C-CH OROHHworin R ein Wasserstoffatom oder einen Glycylrest darstellt,und, falls R einen Glycylrest darstellt, Säureadditionssalze hiervon.6.
