Verfahren zur Herstellung von 1-Phenyl-^-amino-cvelohex-2-en·
1-carbonsaureestern und ihren Salzen
Gegenstand der Erfindung· sind neue l-Phenyl-4~arnino-cyclohex-2-en-l-carbonsäureester
der allgemeinen Formel I
worin R und R2 plelch oder verschieden sind und H, Alkyl,
Hydroxyalkyl, Alkenyl, vor^jgsveise mit 1-4 C-Atomen,
Aralkyl, vorzugsweise mit 1 - 10 C-Atomen, bedeuten, die
miteinander verbunden sein können,"wobei der entstehende
Heterocyclen noch durch die OH-Gruppe substituiert sein

oder ein Stickstoffatom erfolgt, das seinerseits ein

2- und 4-Stellung substituiert sein kann, Acyl, vorzugsweise Alkanoyl mit 1-4 C-Atomen und Aroyl, vorzugsweise mit
1- 10 C-Atomen, oder Aralkyl, vorzugsweise mit 1 - 10 C-Atomen,
trägt und R-, einen Alkylrest mit 1-4 C-Atomen
darstellt, sowie deren Salze und nuaternäre Ammoniumverbindungen,
und ein Verfahren zu ihrer Herstellung.
Es wurde überraschenderweise gefunden, daß sich Cyclohexene der
allgemeinen Formel II

worin Y eine nukleophil austauschbare Gruppe wie Cl-, Br-, J-, CH5SO2-O-, P-CH-J-C6H4-SO2-O-, C6H5SO2-O-, P-Br-C6H4SO2-O-,
P-NO2-C6H4-SO2-O-, P-NO2-C6H4-CO-O- u.a. bedeutet,mit Aminen
der allgemeinen Formel H-NR.R2 in glatter Reaktion zu den Amino·
cyclohexenen der allgemeinen Formel I umsetzen.
Im allgemeinen führt man die Umsetzung so durch, daß man die Amine — ■

HNR1R2 auf die Verbindungen II in einem organischen Lösungsmittel,
vorzugsweise in Dioxan, Dimethylformamid, Dimethylsufloxid,
ßthanol oder Acetonitril, gelegentlich aber auch in Abwesenheit eines Lösungsmittels in Gegenwart oder Abwesenheit einer HiIfsbase
bei Temperaturen zwischen 10 und l60°C einwirken läßt.
Nach der Erfindung .-ist es ferner möglich, eine Verbindung der
allgemeinen Formel I nach an sich bekannten Methoden in das Säureadditionssalz
umzuwandeln, z. B. durch Behandeln mit einer physiologisch akzeptablen anorganischen oder organischen Säure wie
Chlorwasserstoffsäure, Phosphorsäure, Schwefelsäure oder Weinsäure, Fumarsäure, Ascorbinsäure, Oxalsäure u. a. m. Es ist auch
möglich, eine Verbindung der allgemeinen Formel I nach üblichen Methoden, z. B. durch Behandlung mit einem Alkylhalogenid am
Stickstoff zu quaternisieren.
Gemäß der Erfindung können die Verbindungen der allgemeinen Formel
II auf verschiedenen VJ eg en hergestellt werden.
a) Man kann ein Cyclohexen-Derivat der allgemeinen Formel III

(R3 hat die vorstehend angegebene Bedeutung) in grundsätzlich
bekannter Weise mittels Säurehalogeniden wie PCl5, POCl3, PBr3,
POBr3, SOCl2, CH3COBr u. a. rn. oder mittels Halogenwasserstoff~
säuren umsetzen. Diese"Umsetzung führt man vorteilhafter Weise
in einem inerten Lösungsmittel wie Benzol, Chloroform oder Tetrachlorkohlenstoff durch; die Umsetzung mit Halogenwascer- ·
stoffsäuren kann jedoch auch in wäßriger oder alkoholischer
Phase oder in Eisessig mit Erfolg durchgeführt werden.
b) Man kann ein Cyclohexen-Derivat der allgemeinen Formel III in
grundsätzlich bekannter V/eise mit einem SulfonsäurechloriO... wie

Die als Ausgangsmaterial verwendeten l-Phenyl-2-acetoxy-cyclohex-3-en-l-carbonsäureester
der allgemeinen Formel IV

werden erfindungsgemäß durch eine EinV^opfreaktion der Komponenten
Crotonaldehyd, Essigsaureanhj^drid 'fcnd Atropasäureester in Gercen-Viart
von wasserfreiem Natriumacetat hergestellt. Im Einzelnen Acht
man dabei so vor, daß man den Atropasaureester und den Crotonalciehyd
in eine Suspension des Natriuinacctats in Easigsäureanhyd:.nid,

v/elehes hierbei zugleich die Rolle eines Lösungsmittels spielt,
einträgt und das Gemisch p-8 Stdn., vorzugsweise 6 Stan, zum
Sieden erhitzt.
Die nach der Erfindung erhaltenen Verbindungen besitzen hervorragende
analgetische und neuroleptische Eigenschaften,, v.robei das
gleichzeitige Vorliegen beider Eigenschaften in ein or Substanz als überraschend und neuartig zu bezeichnen ist. Als besonders
wirksam haben sich dabei solche Verbindungen der allgemeinen For-·
mel I erv/iesen, worin R1 und R2 unmittelbar" oder über ein Stickstoffatom
zu einem Ring verbunden sind. Auch ausgeprägte spasmolytisehe
Aktivität, meist im Vereine mit analgetischen und/oder sedativen Komponenten wird in zahlreichen Vertretern"*der erfindungsgemäßes
Produkte gefunden.
Die neuen Substanzen lassen sich zu allen für pharmazeutische
Zwecke üblichen Zubereitungsfromen verarbeiten. Zum Beispiel kann man daraus Dragees, Tabletten, Emulsionen, ^njektionslBsungen und

Hauptmenge des Äthanols wird i. Vak. abgezogen, der Rückstand in Wasser eingegossen und die wäßrige Phase mit Äthylacetat extrahiert.
Nach dem Trocknen wird das Lösungsmittel entfernt und der Rückstand destilliert. Kpn -, 118-2O°C. Ausbeute 60 g.

500C tropfen. Man rührt 2 Tage bei 200C, wäscht dann mit
viel Wasser und etwas verd. Natronlauge und trockent die Benzolphase
über CaCl2. Der Rückstand der Benzolphase \nr>d destilliert.
Kp0 01 119-121°C. Ausbeute 41 g.

HCl-Gas beladen und 2 Stdn. unter Rückfluß erhitzt. Es wird abdestilliert
und unmittelbar im Feinvakuum fraktioniert. Kpn ,
1300C, Ausbeute I69 g. Gaschromatographisch einheitlich.
IR-spektroskopisch identisch mit dem nach A hergestellten Produkt.
20 g dieser Verbindung werden analog Methode A mit Dimethylamin
in Dimethylformamid umgesetzt. Man erhält nach der dort beschriebenen
Aufarbeitung 8 g des Hydrochlorides der Titelverbindung als
Viertelhydrat vom Schmp. 3/57-90C."
Methode G-
5 g l-Phenyl-2-hydroxy-cyclohex-3-en-l-earbonsäureäthylcKter werden
in 10 ml Pyridin gelöst und bei 5°C mit 2,5 g frisch destilliertem
Methansulfonsäurfiohlorid tropfenweise unter Schütteln
versetzt. Man läßt den Ansatz j5 Tage bei 50C stehen, gibt ihn
dann in 100 ml 2N HCl ein und extrahiert mit Benzol. Der Benzolrückstand
bildet ein nicht destillierbares öl, das dünnschichtchromatographisch
einheitlich ist und sich IR-spektroskopisch als Mesylat des Cyclohexenols erweist. 2 g des Mesylates werden mit
10 ecm 1Obiger Dimethylamin-LÖsung in Dimethylformamid aufgenommen,
24 Stdn. bei 200C und 2 Stdn. bei 9O0C gehalten. Anschließend
wird in verd. HCl eingetragen und nach Entfernung der Neutral
stoffe mit 2IsT NaOH alkalisiert. Das Hydrochlorid der Beispiel-Verbindung
wird, wie beschrieben, nach dem Aufnehmen der abgeschiedenen Base in Kthylacetat gefällt. Ausbeute 0,5 g vom Schmp.

30 Min. u. Rückfluß erhitzt. Man destilliert im Rotationsverdampfer bis 100°C/20 Torr alles ab, nimmt den Rückstand in
100 ecm DMSO auf und versetzt mit 30 g Piperidin. Der Ansatz

wird 3 Stdn. auf 1000C erhitzt, sodann in Eiswasser eingetragen
und mit.2Ή CHl auf pH 3 gestellt. Neutralprodukte
werden mit Toluol entfernt. Die wäßrige Phase wird mit Ammoniak alkalisiert und das abgeschiedene öl in Äther
aufgenommen. Aus der getrockneten Atherphase fällt man das
Hydrochlorid mittels HCl-Gas. Ausbeute 20,2 g vom Schmp.

Claims (1)
- Patentansprücheί\Λ Verfahren zur Herstellung von 1-Phenyl-4-amino-cyclohex-2-en-l-carbonsäureestern, ihren Salzen und nuaternären Ammoniumverbindungen der allgemeinen Formel Iworin R. und R? gleich oder verschieden sind und FI, Alkyl, Hydroxyalkyl, Alkenyl, vorzugsweise mit 1-4 C-Atomen, Aralkyl, vorzugsweise mit 1-10 C-Atomen, bedeuten, die miteinander verbunden sein können, wobei der entstehende Heterocyclus noch durch die OH-Gruppe substituiert sein kann und wobei die Verbindung gegebenenfalls über ein Sauerstoff- oder ein Stickstoffatom erfolgt, das seinerseits ein FI, Alkyl mit 1 - "4 C-Atomen, Aryl, das durch Chlor,vorzugsweise in j5- oder 4-Stellung, und durch Methoxy, vorzugsweise in 2- und 4-Steilung, substituiert sein kann, Acyl, vorzugs\\reise Alkanoyl mit 1-4 C-Atomen und Aroyl, vorzugsweise mit 1-10 C-Atomen, oder Aralkyl, vorzugsweise mit 1 - 10 C-Atomen, trägt und R^, einen Alkylrest mit 1-4 C-Atomen darstellt, dadurch gekennzeichnet, daß man Cyclohexene der allgemeinen Formel II209837/1188IIworin Y eine nukleophil austauschbare Gruppe wie Cl-, Br-, J-, CH5SO2-O-, P-CH5-C6H4-SO2-O-, C6H5SO2-O-, P-Br-C6H4SO2-O-, P-NO2-C6H11-SO2-O-, P-NO2-C6H11-CO-O- u.a. bedeutet, mit Aminen der allgemeinen Formel H-NR-R2, worin R, und R die oben genannte Bedeutung besitzen, in einem organischen Lösungsmittel, vorzugsweise in Dioxan, Dimethylformamid, Dimethylsulfoxid, Äthanol oder Acetonitril, gegebenenfalls aber auch in Abwesenheit eines Lösungsmittels in Gegenwart oder Abwesenheit einer Hilfsbase bei Temperaturen zwischen 10 und 160 °C einwirken läßt und die so erhaltenen Basen nach an sich bekann ter Weise durch Behandeln mit einer physiologisch akzeptablen anorganischen oder organischen Säure in das zugehörige Additi onssalz oder durch Behandlung mit einem vorzugsweise niederen Alky!halogenid in das Quadern!sierungsprodukt überführt.209837/11882. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Verbindungen der allgemeinen Formel II, (für Y = Halogen) in grundsätzlich bekannter Weise durch Umsetzung eines Cyclohexen-Derivates der allgemeinen Formel IIIIIIC-O-R,Il °worin R5 die vorstehend angegebene Bedeutung hat, mit Säurehalogeniden, wie PCl5, POCl3, PBr3, POBr3, SOCl2, CH3COBr oder mit Halogenwasserstoffsäuren, vorteilhafter Vieise in einem inerten Lösungsmittel wie Benzol, Chloroform oder Tetrachlorkohlenstoff, im Falle der Halogenwasserstoffsäuren auch in wäßriger doer alkoholischer Phase oder in Eisessig darstellt.2. Verfahren nach Anspruch 1, dadurch gekennzeichnet,, daß man die Verbindungen der allgemeinen Formel II, (für Y = Sulfonsäureester) in an sich bekannter Weise durch Umsetzung eines Cyclohexen-Derivates der allgemeinen Formel III mit einem Sulfonsäurechlorid, wie Methansulfonsäurechlorid, p-Toluolsulfonsäureehlorid, p-Nitrobenzolsulfonsäurechlorid und a. m. in Gegenwart einer Base, vorzugsweise in Gegenv/art von Pyridin unter Verwendung oder in Abwesenheit eines Lösungsmittels darstellt.3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man die Verbindungen der allgemeinen Formel II (für Y = Halogen) durch Umsetzung der Verbindungen der allgemeinen Formel IVIVCH0-C-O209837/1188 /21- er ~mit wäßrig-alkoholischer Halogenwasserstoffsäure oder mit ThiQiiyihalogeniden bei 50-80°C darstellt.4. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man die Verbindungen der allgemeinen Formel H3 (für Y = J^ durch Reaktion der l-Phenyl^-dialkylamino-cyclohex-^-en-l-carbonsäur e·- ester mit mindestens 2 Äquivalenten Alkyljodid in einem inerten Lösungsmittel bei Temperaturen zwischen 15 und 800C unmittelbar darstellt.5. Akute Toxizitä'tVersuchstiere waren männliche Mäuse (NMRI) mit einem Kth pergewicht von 18 bis 23 g. Die Tiere wurden vor Versuchsbeginn 24 Stunden nüchtern gehalten, Wasser stand ad libitum zur Verfügung. Zu jeder Dosierungsgruppe gehörten 6 Tiere. Die Dosis wurde mit dem Faktor 1,5 bis 2,0 gesteigert. Das verabreichte Flüssigkeitsvolumon betrug bei intragastraler Applikation 2 ml, bei subeutaner 1 ml/100 g Körpergewicht. Die Beobachtung dauerte 48 Stunden.209837/11882· Phenyl-p-chinon-VersucheZu jeder Dosierungsgruppe gehörten 12 männliche Mäxise im Gewicht von 18 bis 23 g. Phenyl-p-chinon, 1,25 ml der 0,02 %igen Losung pro ICO g Körpergewicht, wurde 15 Minuten nach Applikation der zu untersuchenden Substanzen intraperitoneal verabreicht. Als geschützt wurden nur solche Tiere gewertet, bei denen in den folgenden 20 Minuten keine typische Schiuerzreaktion auftrat. Der Versuch wurde verworfen, wenn nicht mindestens 10 von 12 Tieren der Kontrollgruppe reagierten.3. Heizplatten-VersucheAls Versuchstiere verwendeten wir männliche Mäuse mit einem Körpergewicht von 18 bis 23*g. Die Tiere wurden auf eine konstant auf 55 C gehaltene Kupferplatte gesetzt und die Zeit bis zum Lecken der Vorderpfoten geinessen. ,. Nach 3 Ausgangsmessungen wurden die Substanzen verabreicht. Danach erfolgten die Messungen 2 Stunden lang im Abstand von 30 Minuten. Zu jeder Dosierungsgruppe gehörten 12 Tiere. Das verabreichte Flüssigkeitsvolumen entsprach dem unter 1. angegebenen.4. Fotokäfig-TestsVersuchstiere waren ebenfalls männliche weisse Mäuse im Gewicht von 18 bis 23 g. Die Spontanmotitilität wird in einem Glaskasten (25 χ 50 cm),durch.dessen Querachse in gleiehmässigen Abständen drei Infrarotstrahle laufen, gemessen. Als Ausdruck der Spontanmotilität wird die Anzahl der Unterbrechungen der209837/11S8«tr -Infrarotstrahle innerhalb 30 Minuten gewertet. Zu jeder Versuchsgruppe gehörten 5 Tiere. Die Substanzen wurden entweder intragastral mit der Schlundsonde oder sübcutan verabreicht.5. Körpertemperatursenkende WirkungVersuchstiere waren männliche Ratten (SIV) im Gewicht von 80 bis 120 g. Die Körpertemperatur wurde im Rahmen eines Verhalt ens tests ni'cch Irwin ermittelt, in dem je 5 Ratten nach Beobachtung des Normalverhaltens und nach Messen der Nornialtemperatur mit den Substanzen behandelt werden und nach 30, 60 uua 120 Minuten abermals einem bestimmten Versuchsprogi-amns unterzogen werden. Die in der Tabelle aufgeführten Werte entsprechen der Differenz der Ausgangskörpertemperatur und der Temperatur 2 Stunden nach Substanzapplikation. Die Körpertemperatur wurde rektal mit einem Thermometer der Firma Ellab ermittelt.6. Antihistaminisehe WirkungDie Versuche wurden am isolierten Mcerschweinchenileum durchgeführt. Wir benutzten die von Magnus beschriebene Versuchsanordnung. Der Inhalt des Organbades, Krebs- oder Tyrodelösung, betrug 25 ml,die Temperatur 37°C. Zur Erzeugung der Spasmen verwendeten wir Histamin in Konzentrationen von 4 χ Bei den in der Tabelle angegebenen Konzentrationen wurde der Ilistaminspasmus gänzlich gelöst.209837/11887. Adrenolytischc WirkungVersuchstiere waren männliche und weibliche Hunde im Gewicht von 4 bis 9 kg in Nembutal-Narkose. Der arterielle und venöse Druck wurden mit 11x3 Ic von Stathamelementen, das Atemvolumen mit einem Spiroraeter nach Gildemeistcr registriert. Hs wurde geprüft, inwieweit die Blutdruckreaktionen von Adrenalin, Noradrenalin und Phenylephrin ( 1 bis 2 y/kg iv. ) durch steigende Dosen der erfindungsgemaToen Substanzen beeinflusst werden. Eine schwache bis massige Reduzierung der blutdruckerhühenden Wirkung der genannten Substanzen wird mit +, eine-^deutliche Reduzierung mit ++ und eine Umkehr des Adrenalin-Effektes mit -H-+ bewertet.8. LokaIanaesthetische Wirkung ,Verstichstiere waren männliche und weiblich Kaninchen mit einem Körpergewicht von 1,8 bis 3 kg. Die Substanzen wurden in Konzentrationen von 0,1 bis 2 % in den Bindehautsack appliziert (2 Tropfen pro Auge). Im Abstand von 4 Minuten wurde mit einer Borste, die bei einer Belastung von 250 mg abbiegt, der Corncaroflex ausgelöst. Maximal wurden 100 Schläge appliziert. Der Versuch wurde so lange fortgeführt, bis die Ausgangswerte wieder erreicht waren. Mit + werden lokalanaesthetischo Effekte der 1 und 2 %igon Lösung, mit ++ solche der 0,5 und mit +++ die der 0,1 %igen Lösung bezeichnet.37/1188 .Akute Toxizität
