ein Verfahren zur Herstellung von 1,4-Benzodiazepinen der allgemeinen Formel
in der R, Wasserstoff, Halogen, einen niederen Alkylrest oder eine Nitrogruppe,

Die Endprodukte des erfindungsgemäßen Verfahrens sind wertvolle Arzneimittel,
die als Psychopharmaka mit Vorwiegend sedierenden und tranquillisierenden Eigenschaften

Verbindungen der allgemeinen Formel I lassen sich nach verschiedenen
Verfahren mit einer mehr oder weniger großen Anzahl von Verfahrensstufen und sehr
unterschiedlichen Ausbeuten herstellen. Als Ausgangsmaterial dienen entsprechend
substituierte 2-Aminobenzophenone. So werden gemäß DDR-Patentschrift 40 185 2-Aminobenzophenons
mit al-Aminocarbonsäuren, ihren Estern und entsprechenden Derivaten durch vielstündiges
Erhitzen (im allgemeinen 15 Stunden und länger) in Pyridin zur Umsetzung gebracht.
Diese Reaktion ist gleichzeitig mit einer starken Dunkelfärbung sowohl der lReaktionsmischung
als
auch der gebildeten 1,4-Benzodiazepine verbunden. Diese Endprodukte
sind offensichtlich so stark verunreinigt' daß sie zum Beispiel entsprechend der
DDR-Patentschrift 42 687 durch Überführung in ein Säureadditionssalz der Salpeter-oder
Schwefelsäure und anschließende Zersetzung desselben mit Basen und nachfolgende
Umkristallisation einem besonderen Reinigungsprozeß unterworfen werden müssen.Compounds of general formula I can be according to various

Die Herstellung von 1,4-Benzodiazepinen hach der DDR- Patentschrift
40 855 ist zwar eine Einstufenreaktion (reduktive Entfernung des Sauerstoffatoms
am Stickstoffatom in 4-Stellung), jedoch umfaßt die Herstellung dieser als Vorstufe
benötigten 1,4-Benzodiazepin-2-on-4-oxide weitere 3 bis 4 Reaktionsstufen, da man.
entweder zunächst aus dem entsprechenden 2-Aminobenzo,. phenon das zugehörige x-Oxim
herstellen, dieses weiter in ein Chinazolin-3-Oxid überführen und dieses schließlich
einer Ringerweiterung gemäß,DDR-Patentschrift 40 184 unterwerfen muß. Man kann auch
über das entsprechende 2-Aminobenzophenon-ß-oxim gehen, dieses mit Ohloracetylchlorid
acylieren und das Acylierungsprodukt nach DDR Patentschrift 47 413 durch-Alkali
zum 1,4-Benzodiazepin-2-ön-4-oxid zyklisieren.The production of 1,4-benzodiazepines according to the GDR patent

Die DDR-Patentschrift 40 50$- sieht als Vorstufen die Herstellung
von 2-Carbobenzoxyglycylamido-benzophenonen vor, die, nach saurer Abspaltung des
Garbobenzoxy-Restes, zum Siebenring zyklisiert werden Die Gewinnung der 2-Carbobenzoxyglycylamidobenzophenone
als Vorprodukte ist ebenfalls ein vielstufiger ProzeB# der vor allem den Nachteil
in sich birgt, relativ teure Ausgangsmaterialien (Glykokoll, Benzylalkohol) und
Zwischen- bzw. Hilfsprodukte (N,N9-Dicyclohexylcarbodiimid) zu bedürfen. In der
DDR-Patentschrift 41 256 ist zur Herstellung von 1,4-Benzodiazepinen die Umsetzung
von 2-Halogenacetamido-benzophenonen mit Ammoniak vorgesehene wobei - wie den Beispielen
der Patentschrift zu entnehmen ist - offensichtlich nur die 2-Bromacetamidobenz®phenone
eine hinreichende Reaktionsfähigkeit mit Ammoniak besitzen, so daß zur Herstellung
der Vorstufen das relativ teure Bromacetylbromid erforderlich ist.
Die
holländische Patentanmeldung-Nr. 6 500 446 schließlich beschreibt als Methode zur
Gewinnung von 1,4-Benzodiazepinen die Umsetzung von 2-(Phthalylglycylamido)-benzophenonen
mit Hydrazin. Auch hier sind die Vorprodukte nur mit Hilfe von Glykokoll und die
Endprodukte nur mittels des relativ kostspieligen Hydrazins zugänglich. Die vorliegende
Erfindung stellt sich die Aufgabe, die 1,4-Benzodiazepine der allgemeinen Formel
I aus billigen und leicht zugänglichen Vor- bzw.-Zwischenprodukten in guten Ausbeuten
und hoher Reinheit herzustellen. Es wurde überraschenderweise gefunden, daB man
leicht und in. guten Ausbeuten die Verbindungen der allgemeinen Formel I erhalten
kann, wenn man ein 2-(Arylsulfonyll;lykoloylamino)-benzophenon der allgemeinen Formel
worin die Reste R,1 und R2 die gleiche Bedeutung wie oben besitzen .und R3 gleich
Wasserstoff ist oder eineu niederen Alkylrest darstellt, in Anwesenheit eines inerten
organischen Lösungsmittels mit Ammoniak behandelto Die 1,4-Benzodiazepine der allgemeinen
Formel I kann man, sofern R,1 und R2 = Wasserstoff bedeuten, durch anschließende
Alkylierung und/oder Nitrierung in an-sich bekannter Weise in entsprechend substituierte
Derivate der allgemeinen Formel I umwandelli. Dieser Weg ist aber nicht erforderlich,
da es günstiger ist, din bereits die gewünschten Reste R, und R2 tragendes Benzophenonderivat
der allgemeinen Formel II einzusetzen.
Zur Umsetzung von Verbindungen
der. allgemeinen Formel II mit Ammoniak sind als organische Lösungsmittel u.a.-geeignets
niedere aliphatische Alkoholen Dioxan, Dialkylformamide, Acetonitriln Tetrahydrofuran
und Benzol' Zweckmäßigerweise führt man die Umsetzung mit Ammoniak bei Temperaturen
durch, die zwischen Raumtemperatur und + 120 0C liegen. Hierbei wird Ammoniak entweder
als Gas eingeleitet oder auch als wäßrige,''konzentrierte Lösung zugegeben. Die
Ausbeuten dieser Ringschlußstnfe liegen bei dem erfindungsgemäßen Verfahren, insbesondere-in
Abhängigkeit von dem Substituentren R,1 und R2, zwischen 50 % und 80 %, wobei die
anfallenden Produkte bereits sehr rein sind. Das Verfahren, entsprechend der Erfindung,
ist in mehrerer Hinsicht überraschend. Einmal mußte damit_gerechnet werden, daß
sich bei Reaktion von Arylsulfonsäureestern der allgemeinen Formel II mit Ammoniak
mehrere Reaktionsprodukte bilden (vgl. Houben-Weyl, Methoden der organischen Chemie,
Georg Thieme Verlag 1957, Bd. 11/19 S. 217), so daß hohe Ausbeuten an Benzodiazepinen
der allgemeinen Formel I nicht zu erwarten waren. Überraschend ist weiterhin, daß'die
2-(Arylsulfonylglykoloylamino)-benzophenone der allgemeinen Formel II unter den
o. g. Bedingungen, d.h. selbst in Gegenwart von Wasser, in einem
ohne daß Zwischenstufen isolierbar $ind, in hoher Ausbeute in die 1,4-Benzodiazepine
der allgemeinen Formel I übergehen, da hierzu die Abspaltung von 1 Mol Wasser erforderlich
ist. Die als Ausgangsmaterial verwendeten 2-(Arylsulfonylglykoloylamino)-benzophenone
der allgemeinen Formal II lassen sich sehr bequem und in hohen Ausbeuten aus entsprechenden
Arylsulfonylglykolsäuren bzw. -säurehalogeniden und entsprechend substituierten
2-Aminobenzophenonen gemäß der btit. Patentschrift Nr. 1 021 382 herstellen.
Die
erwähnten Arylsulfonylglykolsäuren wiederum sind sehr leicht und in guten Ausbeuten
aus billigen Rohstoffen zugänglich, indem man Arylsulfochloride entweder mit Glykolsäure
(hergestellt aus Monochloressigsäure und konzentrierter wäßriger Natronlauge) oder
mit Glykolsäurenitril (hergestellt aus wäßriger Formaldehydlösung und Natriumcyanid)
zur Umsetzung bringt. Im letzten Fall muB das zunächst gebildete Arylsulfonylglykolsäurenitril
einer anschließenden Hydrolyse, am besten mittels Schwefelsäure, unterworfen werden
(vgl. J. Lichtenberger, Oh. Faure, Bull. Soc. chim. France, 1948, So 995 ff). Beispiel
1
25 g 2-(0-p-Toluolsulfonyl-glykolöylamino)-5-chlorbenzophenon
werden mit 500 ml Dioxan und 250 ml konzentriertem wäßrigen
Ammoniak zum Sieden erhitzt und in die siedende Mischung unter
Rühren Ammoniakgas@eingeleitet. Nach etwa 15 Minuten ist die
Reaktionsmischung klar. Man leitet noch weitere 6 Stunden unter
Kochen Ammoniakgas ein. AnschlieBend.engt man im Vakuum zur
Trockne ein, versetzt den Rückstand mit 300 ml Aceton
und erhitzt
30 Minuten zum Sieden. Vom ungelöst bleibenden Ammoniumsalz
der
p-Toluolsulfonsäure (9,5 g 90 r% der Theorie, Fp; 325 - 329
o C)
wird heiß abfiltriert und däs Filtrat auf 150 ml eingeengt.
Nach dem Erkalten scheidet sich das 7-Chlor-5-pher@yl-1,3-di-
hydro-2H-1,4-benzodiazepinr-2an in fast weißen Kristallen ab.
Ausbeute 9,2 g, entsprechend 60 yb der Theorie, Fp: 217 °C.
Das aas Ausgangsmaterial verwendete p-Toluoisulfonylglykoloylderivat wird wie folgt
hergestellt: 62 g p--Toluolsulfonylglykolsäure werden mit 60 ml Thionylchlorid übergossen
und das Gemisch auf dem Dampfbad erhitzt, bis sich alles gelöst hat und kein Gas
mehr entweicht. Überschüssiges Thionylchlorid wird im Vakuum entfernt und der dunkle,
nicht kristallisierende
Rückstand in 50 ml Aceton gelöst. Diese
Lösung des Säurechlorids gibt man rasch zu einer Lösung von 55 g 2=Amino-5-chlorbenzophenon
in 180-ml Aceton. Das Gemisch erwärmt sich fast bis zum Sieden. Man läßt 2 Stunden
nachrühren und kocht dann 30.Minuten unter Rückfluh, Durch Zugabe von 200 ml Wasser
wird die nach dem Erkalten vorhandene Kristallmenge noch vermehrt. Man saugt ab
und wäscht mit Methanol. Nach Umkristallisation aus Alkohol oder besser aus Eisessig
erhält.man 69,1 g (6_5-% der Theorie) schwach gelblicher Nadeln vom Fp. 150 - 152
°C. _ Beispiel 2 5 g 2-(0-Benzolsulfonyl-glykoloylamino)-5-chlorbenzophenon werden
in einem Gemisch aus 50 ml Dioxan und 50 ml konzentriertem wäßrigen Ammoniak 2 Stunden
unter Rückiluß gekocht. Die Reak-
tionsmischuhg wird nach etwa 10 Minuten klar. Man dampft im
Vakuum zur Trockne, kocht den Rückstand 30 Minuten mit 50 ml
Aceton aus und saugt heiß vom unlöslichen Ammonium salz der
Benzolsulfonsäure ab (64 ro der Theorie, Fp. 270 - 280 °c).
Das Filtrat wird im Vakuum auf 25 ml eingeengt, das sich beim
`Abkühlen bildende Kristallisat abgesaugt und mit wenig Aceton
gewaschen. Ausbeute 2,3 g (?3 % der Theorie). Nach Umkristalli-
sation aus Butanol liegt der Fp. bei 215 00. Das gebildete
?-Chlor-5-phenyl-1,3-dihydro-2F1-1,4-benzodiazepin-2-on ist
na-
hezu weiß.
Das als Vorstufe benötigte 2-(0-Benzolsulfonyl-glykoloylamino)-.
5-chlorbenzophenon stellt man entweder analog der entsprechen-
den p-Toluolsulfonylverbindung (siehe Beispiel 1) aus 32,4
g
Benzolsulfonylglykolsäure durch Umsetzung mit Thionylchlorid
(55 m.) und anschließender Reaktion des intermediär gebildeten
Benzolssulfoa;ylglykolsäurechlorida mit 2-Amino-5-chlorbenzophe-
non (23,1 g) in Ac®ton oder Benzol als Lösungsmittel
her. Es
ist auch möglich, die Acylierung des 2 Amino-5-chlorbenzophenons
durch die Benzolsulfonylglykolsäure nach dem Garbodiimid-Ver-
fahren durchzuführen. Zu diesem Zwecke werden 4,3 g Benzol-
aulfoa@ylglykolsäure und 4,6 g 2-Amino-5-chlorbenzophenon in
100 ml Methylenchlorid gelöst und zu dem Gemisch unter Rühren bei
0 - 5 °C innerhalb von 30 Minuten in 4 Portionen 4,5 g N,N#-Dicyclohexylcarbodiimid
gegeben. Bei der gleichen Temperatur läßt man 6 Stunden nachreagieren und dann über
Nacht bei Raumtemperatur stehen Es werden 5 ml Eisessig hinzugefügt, 30 Minutengerührt
und vom nunmehr vollständig ausgeschiedenen N,N'-Dicyclohexylharnstoff abgesaugt.
Das Filtrat wird in Vakuum zur Trockne gebracht, der Rückstand mit wenig Alkohol
aufgenommen, abgesaugt und mit Alkohol gewaschen. Man erhält gelbliche Kristalle
vom Fp. 135 - 13? °C. Die Ausbeute ist fast quantitativ. Beispiel 3 10 g
2-(0-p-Toluolsulfonyl4-glykoloylamino)-benzophenon werden zusammen mit 100 ml Dioxan
und 100 ml konzentriertem wäßrigen Ammoniak 2 Stunden unter Rückfluß gekocht. Nach
etwa 10 Minuten entsteht eine klare, gelbe Lösung. Man läßt erkalten, dampft im
Vakuum zur Trockne, gibt zum Rückstand ?5 ml Aceton und Kocht 30 Minuten. Das in
der Siedehitze ungelöst bleibende Ammoniumsalz der p-Tolnolsulfonsäure wird durch
Filtration abgetrennt und das Filtrat Weitgehend eingeengt. Die Ausbeute an rohem
5 Phenyl-1' 3-dihydro-2äi-1,4-benzodiazepin-2-on ist quantitativ. Nach Umkristallisation
aus Aceton erhält man weiße Kristalle vom Fp. 18'I oC. Beispiel 4 10 g 2-(0-p-Toluolsulfonyl-glykoloylamino)-benzophenon
erhitzt man in 150 ml n-Butanol unter Einleiten von gasförmigem Ammoniak zum Sieden.
Nach etwa-10 Minuten ist die Reaktionsmischung klar. Man kocht uhd begast insgesamt
3 Stunden. Nach dem Erkalten saugt man das ausgefallene Ammoniumsalz der p-Toluolsulfonsäure
(4,3 g 93 der Theorie, Fp. 318r 336 00) ab, entfernt das Lösungsmittel im. Vakuum,
gibt zum Rückstand etwas Aceton und saugt den feinkristallinen Niederschlag ab.
Ausbeute 2,6 g (" 46 9b der Theorie) 9 Pp. 182 0 0.
Das
als Ausgangsmaterial für Beispiel-3 und 4 benötigte acylierte 2-Aminobenzophenon
wird wie folgt hergestellt: Aus 34,5-g p-Tolüolsulfonylglykölsäure und 40 ml Thionylchlorid
stellt man durch Kochen unter Rückfluß (1 Stunde) p-Toluolsulfonylglykolsäurechlorid
her. Restliches Thior@ylchlorid wird im Vakuumentfernt und der Rückstand in 40 ml
ACceton aufgenommen. Die Lösung des Säurechlorids wird zu einer Lösung von 19'7
g 2-Aminobenzophenon in 100 ml Aceton unter Rühren rasch zugegeben. Man kocht 30
Minuten unter Rückfluß, wobei Ohlorwasserstoffgas entweicht. Nach dem Abkühlen wird
mit reichlich Wasser verdünnt, die Flüssigkeit mit konzentrierter Natronlauge schwach
alkalisiert und die heilbraunen Kristalle abgesaugt. Man wäscht mit Wasser, trocknet
und kristallisiert aus Alkohol um. Ausbeute 25 g (61 % der Theorie), weiße Stäbchen
vom Fp. 115 - 117 00. Beispiel 5
20 g 2-(0-p-.Tciluolsulfonyl-glykoloylamino)-5-methyl-
benzophenon werden mit 200 ml Dioxan und 100 ml wäßrigem,
konzentriertem Ammoniak 4 Stunden unter Rückfluß zum Sieden
erhitzt. Anschließend dampft man im Vakuum zur Trockne und
kocht den Rückstand erschöpfend mit Aceton aus. Man saugt
vom ungelöst bleibenden Ammonium salz der p-Toluolsulfonsäure
ab und gewinnt aus dem Filtrat durch Einengen 9,6 g (" 81
der Theorie) ? Methyl-5-phenyl-1.93-dihyd.ro-2H-1,4-benzodiaze-
pin-2-on als Rohprodukt. Nach Umkristallisieren aus Aceton
liegt der Pp. bei 206 - 208 0C.
Das als Vorstufe eingesetzte 2-(0-p-Toluolsulfonyl-glyko-
loylamino)-5 methylbenzophenon stellt man-her, indem man
87 g pe-tToluolsulfonylglykolsäure durch Kochen mit 90 ml
Thionylchlorid in das entsprechende Säurechlorid überf@.ihrt,
Nach Abdampfen des überschüssigen Thionylchloride löst man
den Rückstand in 100 ml Benzol und gibt die Lösung zu 59 g
-2-Amino-5-methylbenzophenon in 300 ml Benzol.-Man rührt
30 Minuten bei Raumtemperatur und erhitzt dann nochmals die
gleiche Zeit zum Sieden. Das Benzol wird im Vakuumentfernt und
der Rückstand nach Zugabe von 150 ml Alkohol abgesaugt. Man kristallisiert aus .800
ml Alkohol um und erhält 100 g (85 % der Theorie) an schwach gelblichen Kristallen
vom Fp. 123-12500. Beispiel 6
50 g 2-(0-p-Toluolsulfonyl-glykoloyl-N-methylamino)-5-
chlorbenzophenon werden in 250 ml Acetonitril gelöst und
bei Raumtemperatur 8 Stunden lang unter Rähren mit Ammoniak-
gas behandelt. Dabei scheidet sich in zunehmender Menge das
Ammonium salz der p-Toluolsulfonsäure ab. Man läßt anschließend
über Nacht stehen, gibt 600 ml Wasser hinzu und schüttelt mehr-
fach mit Benzol aus. Die organische Phase wird zweimal mit
Wasser gewaschen, getrocknet und das Lösungsmittel abgedampft.
Der alsbald, nach Zugabe von etwas Isopropanol, kristallin
werdende Mickstand wird abgesaugt und mit Isopropanol und
Benzin gewaschen. Man erhält 19'3 g 62 % der Theorie) von
fast weißem 7-Ohlor-1 9 3-dihyäro-l-methyl-5-phenyl-2H-1,4-
benzodiazepin.-2-on mit einem Fp. von 125 = 127 o0.
Durch Um-
kristallisieren aus Isopropanol kann das Produkt noch wenig
aufgehellt worden*
Das als Ausgangsmaterial benötigte p-eoluolsulfonyl-glykoloyl-
derivat wird aus p-Toluolsulfonylglykolsäurechlorid (Herstel-
lugg aus 101,6 g p-Toluolsulfonylglykolsäure und 102 ml
Thionylchl6rid, vgl. Beispiel 1), das in 200 ml Benzol gelöst
wird, und 98,4 g 2-Methylamino-5-chlorbenzophenon, gelöst in
480 ml Benzol; hergestellte Die Lösungen der Komponentenwer-
den zusammengegeben und 2 Ständen unter Rückfluß gekocht. Man
dampft das Benzol in Vakuum ab und versetzt den Räckstand mit
200 ml Alkohol.. Zur Kristallisation 1613t man einige Stunden
3.m
Kühlschrank stehen. Man saugt ab, wäscht gut mit Alkohol und
trocknet. Die Ausbeute an hellgrauem Rohprodukt beträgt 171,g
(93 % der Theorie) q Fpe 110 - 114 00. Zur Weiterverarbeitung
ist eise Reinigung durch Umkristallisation nicht erforderlich.
Beispiel 7
10 g 2-(0-p-Toluolsulfonyl-glykoloylamino)-5-nitrobenzo--
phenon werden in 200 ml Dioxanbei 50 0C Badtemperatur ge-
löst und in die Lösung bei der gleichen Temperatur 16 Stun-
den lang Ammoniakgas eingeleitet. Nach jeweils 8stündiger Ein-
leitungszeit bleibt das.Reaktionsgemisch über Nacht bei Raum-
temperatur, mit Ammoniakgas gesättigt, stehen. Man verteilt
dann zwischen 150 9n1 Essigsäureäthylester und 250 ml Wasser,
schüttelt die wäßrige Phase nochmals mit 50 ml Essigester aus
und wäscht den Essigesterextrakt zweimal mit je 100 ml Wasser.
Nach Trocknung über Natriumsulfat wird der Essigester im Vakuum
abgedampft, der Rückstand mit 50 ml Benzol versetzt und er-
schöpfend mit 1 n Natronlauge extrahiert. Die wäßrig-alkalische
Phase wird vom Benzol abgetrennt und mit verdünnter Salzsäure
auf pH 7 gebracht. Das abgeschiedene rohe 7-Nitro-5-phenyl-
1,3-dihydro-2H-1,4-benzodiazepin-2-on wiegt nach absaugen,
waschen und trocknen 3,4 g ('--` 55-# der Theorie).
Nach Umkristal-
lisation aus Alkohol oder Isöpropänol liegt der Fp. bei 226
00.
Das als Vorstufe eingesetzte Toluolsulfonylderivat erhält man
aus p-Toluolsulfor@ylglykolsäurechlorid (hergestellt aus 50,6
g
p-Toluolsulfonylglykolsäure und 60 ml Thionylchlorid nach Beispiel
1) und 2-Amino-5-nitrobenzophenon (48,4 g) durch zweistündiges
Kochen in 380 ml Benzol. Man läßt erkalten, saugt ab und
wäscht mit Alkohol. Als Ausbeute erhält man 81 g ('-_` 89 %
der
Theorie) stäbahenförmiger Kristalle vom Fp. 17o - 181 °c:.
Beispiel 8
