Verfahren zur Herstellung neuer im Pyrazolkern substituierter Pyrazolo-pyrimidine Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von im Pyrazolkern N-un- substituierten, 4-unsubstituierten 3 - NHz - 6 - R1 Pyrazolo[3,4-d]pyrimidinen, worin R1 eine freie oder substituierte Aminogruppe bedeutet.
Als substituierte Aminogruppen kommen hierbei in erster Linie aliphatisch substituierte Amino- gruppen, wie z. B. Mono- oder Diniederalkylamino- gruppen oder Morpholino-, Piperidino-, Pyrrolidino- oder Piperazinogruppen in Frage.
Die neuen Pyrazolo[3,4-d]pyrimidine besitzen wertvolle Eigenschaften. Sie sind Antimetaboliten von Purin. Ferner zeigen sie tumorhemmende, diure- tische, coronarerweiternde, zentralhemmende und antirheumatische Wirkungen. Weiter sind sie wirk sam gegen Virusinfektionen, z. B. bei Infektionen durch Ektromelie-Virus, sowie gegen Schistosomen. Sie sollen als Heilmittel oder als Zwischenprodukte verwendet werden.
In dieser Beziehung ganz besonders wertvoll sind die erfindungsgemäss erhaltenen Verbindungen der Formel

B. eine niedere Dialkyl- aminogruppe, wie die Dimethylamino- oder Diäthyl- aminogruppe, oder eine Morpholino-, Piperidino-, Pyrrolidino- oder Piperazinogruppe bedeutet, und vor allem 3-Amino-6-dimethylamino-pyrazolo[3,4-d]- pyrimidin. Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, dass man 6-unsubstituierte 2-Ri Pyrimidine,
die in 4-Stellung einen sich bei der Reaktion abspaltenden Rest und in 5-Stellung eine Nitril-, Thioamid- oder Amidin- gruppe aufweisen mit Hydrazin direkt oder stufen weise umsetzt.
Als sich bei der Reaktion abspaltende Reste in 4-Stellung der als Ausgangsstoffe zur Anwendung gelangenden Pyrimidine kommen insbesondere freie oder verätherte Mercaptogruppen, z. B. die Methyl- mercaptogruppe, freie oder substituierte Amino- gruppen, z. B. die Methylaminogruppe, oder vor zugsweise Halogenatome, wie Chlor oder Brom, in Frage.
Die genannte Reaktion kann in an sich bekann ter Weise in An- oder Abwesenheit von Verdün- nungs- oder Kondensationsmitteln, gegebenenfalls bei erhöhter Temperatur, im offenen oder geschlossenen Gefäss unter Druck durchgeführt werden.
Die als Ausgangsstoffe verwendeten Pyrimidine können z. B. durch Kondensation von geeignet substi tuierten Amidinen mit eine Carbonylgruppe tragenden Malonsäuren, gegebenenfalls in Form ihrer funktio nellen Säuren undloder Carbonylderivate und gege benenfalls geeignete Abwandlung von Substituenten in den erhaltenen Pyrimidinen hergestellt werden. So lässt sich z.
B. ein S - Alkyl - isothioharnstoff mit a-Äthoxymethylen - a - cyanessigsäure-äthylester zum 2-Alkylmercapto-4-oxy-5-cyan-pyrimidin umsetzen, in dem die Substituenten in üblicher und geeigneter Weise abgewandelt werden können, um die gewünsch ten Ausgangsprodukte zu erhalten. Solche Reak tionen sind in der Schweizer Patentschrift Nr. 376115 beschrieben.
Nach einer bevorzugten Ausführungsform des vorliegenden Verfahrens geht man von dem aus 2 - Methylmereapto - 4 - oxy-5 -cyan-pyrimidin durch Umsetzung mit Dimethylamin und Behandlung mit Phosphoroxychlorid erhaltenen 2-Dimethylamino-4- chlor-5-cyan-pyrinridin aus, setzt es mit Hydrazin um und gelangt so zum 3-Amino-6-dimethylamino- pyrazolo[3,4-d]pyrimidin.
Die neuen Verbindungen bilden Salze mit an organischen oder organischen Säuren. Als salz bildende Säuren kommen beispielsweise in Frage: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure;
aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein- oder Brenztraubensäure;
Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl- oder p-Amino-salicylsäure; Methansulfon-, Äthansulfon-, Oxyäthansulfon-, Äthylensulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure;
Methionin, Tryptophan, Lysin oder Arginin. Die oben beschriebenen Pyrazolo-pyriniidine, ihre Salze oder entsprechende Gemische können z. B. in Form pharmazeutischer Präparate Verwendung fin den.
In den nachfolgenden Beispielen sind die Tempe raturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 8,3 g 2-Dimethylamino-4-chlor-5-cyan-pyrimidin werden mit einer Lösung von 4,6g Hydrazinhydrat in 100 cm3 Äthanol versetzt. Die Lösung wird wäh rend 1 Stunde zum Sieden erhitzt. Man nutscht nach Erkalten vom ausgefallenen kristallinen Pro dukt ab, bringt dieses in alkoholische Salzsäure, er hitzt erneut während 1 Stunde und lässt erkalten. Das ausgefallene Produkt wird abgenutscht und aus Äthanol umkristallisiert.

Das als Ausgangsstoff verwendete 2-Dimethyl- amino-4-chlor-5-cyan-pyrimidin wird wie folgt er halten: 11,2 g Kaliumhydroxyd und 70 cm3 Methanol werden auf 0 abgekühlt und dann mit einer Lösung von 44 g S-Methylisothioharnstoff in 200 cm3 Me- thanol versetzt.
Man nutscht vom ausgefallenen Ka- liumjodid ab, versetzt das Filtrat mit 33g Äthoxy- methylen-cyanessigester, wobei die Temperatur zwi schen 8-12 gehalten wird, filtrierc aus dem gelb ausgefallenen Niederschlag ab und wäscht mit kaltem Methanol und Äther nach. Der so erhaltene S-Methyl- isothioureido-methylencyanessigester schmilzt bei 128 bis 129 .
2,1 g S-Methyl-isothioureido-methylencyanessig- ester werden mit 20 cm-' 0,5n Natronlauge versetzt und während 10 Minuten auf 50 erhitzt (Farb- umschlag der Lösung von gelb in weiss). Man nutscht sofort von wenig Ungelöstem ab, steilt das Filtrat mit In Salzsäure auf pH 1 und nutscht von dem ausgefallenen Niederschlag ab. Letzterer wird aus viel siedendem Äthanol umkristallisiert.
