Verfahren zur Herstellung neuer Pyrimido-pyrimidine
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Aminogruppen enthaltenden Pyrimido [4,5-d] pyrimidinen mit dem Kern der Formel

Für die Aminogruppe in 7-Stellung der neuen Verbindungen kommt in erster Linie eine mono-oder disubstituierte Aminogruppe in Frage, wobei die Substituenten aus der aromatischen, heterocyclischen, araliphatischen, heterocyclylaliphatischen, cycloaliphatischen oder aliphatischen Reihe gewählt sein können. Solche Substituenten sind besonders niedere Alkyl-, Cycloalkyl-oder Alkylenreste, die auch in den Kohlenstoffketten durch Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff, unterbrochen sein können.
So kann die Aminogruppe z. B. eine niedere Monooder Dialkylamino-, tert.-Aminoalkylamino-, Pyrrol- idino-, Piperidino-, Morpholino-oder Piperazinogruppe sein. Die Aminogruppe kann auch anorganisch substituiert sein und z. B. eine Hydrazinogruppe dar- stellen.
Die neuen Verbindungen enthalten in mindestens einer der Stellungen 2,4 oder 5 freie oder substituierte Aminogruppen. In zwei der genannten Stellungen kön- nen sie auch freie oder verätherte Hydroxylgruppen oder niedere Alkylreste aufweisen.
Als verätherte Hydroxygruppen kommen vor allem niedere Alkoxygruppen, wie Methoxy und als substituierte Aminogruppen in erster Linie die oben beschriebenen in Betracht.
Die neuen Verbindungen können auch an den Ringstickstoffatomen Substituenten aufweisen, und zwar vorzugsweise niedere aliphatische Reste, wie niedere Alkyl-oder Aminoalkylreste, z. B. Methyl.
Die neuen Verbindungen besitzen wertvolle Eigenschaften. Sie sind Antimetaboliten von Purin. Ferner zeigen sie insbesondere tumorhemmende, aber auch diuretische, coronarerweiternde, zentrale und antirheumatische Wirkungen. Sie sollen als Heilmittel oder als Zwischenprodukte verwendet werden.
In dieser Hinsicht ganz besonders wertvoll sind Verbindungen der Formel

worin Rt eine freie Aminogruppe und R2 eine niedere Dialkylamino-oder Alkylenaminogruppe, wie die Dimethylaminogruppe, bedeuten, beispielsweise das 4 Amino-7-dimethylamino-pyrimido- [4,5-d] pyrimidin, sowie ihre Salze.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, dass man 7 Amino-pyrimido [4,5-d] pyrimidine, die in mindestens einer der Stellungen 2,4 und 5 eine freie oder ver ätherte Mercaptogruppe oder ein Halogenatom aufweisen, mit Ammoniak oder Aminen umsetzt.
Die Ausgangsstoffe können nach an sich bekannten Methoden gewonnen werden.
Die genannten Reaktionen können in An-oder Abwesenhe, it von Verdünnungsmitteln, gegebenenfalls bei erhöhter Temperatur, im offenen oder, wenn erwünscht, im geschlossenen Gefäss, gegebenenfalls unter Druck durchgeführt werden.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie freie Oxygruppen, so können Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Charakter bilden Salze mit anorganischen oder organischen Säuren.
Als salzbildende Säuren kommen beispielsweise in Frage : Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure ; aliphatische, alicyclische, aromatische oder heterocyclische Carbon-oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein-oder Brenztraubensäure ; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl-oder p-Aminosalicylsäure ; Methansulfon-, Athansulfon-, Oxyäthansulfon-, Äthylensulfonsäure ; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure ; Methionin, Tryptophan, Lysin oder Arginin.
Die oben beschriebenen Pyrimidopyrimidine, ihre Salze oder entsprechende Gemische können wie gesagt als Arzneimittel Verwendung finden.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
11 g 4-Methylmercapto-7-dimethylamino-pyr imido- [4, 5-d] pyrimidin werden zusammen mit 80 cm- flüssigem Dimethylamin im verschlossenen Rohr wäh- rend 8 Stunden auf 100 bis 125 erhitzt. Nach Abdampfen des überschüssigen Dimethylamins wird der Rückstand in 100 cm3 2-n. Salzsäure aufgenommen.
Nach Abfiltrieren von wenig Ungelöstem stellt man mit 10-n. Natronlauge auf pH = 10 und versetzt mit festem Atzkali. Hierbei fällt ein 01 aus, das bald kristallin erstarrt. Man nimmt dieses in Chloroform auf und stellt aus dem Chloroformrückstand auf folgende Weise direkt das Hydrochlorid der erhaltenen Base her :
Die Base wird in äthanolischer Salzsäure heiss gelöst und das entstandene Hydrochlorid durch Zusatz von Essigester und Ather wieder ausgefällt. Nach Kristallisation aus Athanol erhält man 4,7-Bis dimethylamino-pyrimido- [4, 5-d] pyrimidin-hydrochlorid der Formel

Das als Ausgangsstoff verwendete 4-Methylmer- capto-7 [4, 5-d] pyrimidin kann wie folgt erhalten werden :
Die auf 0 abgekühlten Lösungen von 11,2 g Kaliumhydroxyd in 70 cm3 Methanol und von 44 g S-Methylisothioharnstoff-hydrojodid in 200 cm3 Methanol werden zusammengegeben. Man versetzt dann bei einer Temperatur von 8 bis 12 mit 25 g Athoxymethylen-malonitril, lässt während 2 Stunden bei Zimmertemperatur weiterrühren und nutscht von dem ausgefallenen Niederschlag ab. 10 g 2-Dimethylamino-4-amino-5-cyan-pyrimidin werden mit 200 cm3 2-n. Natronlauge und 200 cm3 Äthanol während 3 Stunden am Riickfluss gekocht.
Man lässt erkalten und nutscht dann von dem aus gefallenen 2-Dimethylamino-4-amino-5-carbamyl-pyr- imidin der Formel

18,1 g 2-Dimethylamino-4-amino-5-carbamylpyrimidin werden mit 40 g Formamid während 4 Stunden erhitzt. Man lässt erkalten, nutscht von dem ausgefallenen Produkt ab, löst in 300 cm3 2-n.

9 g 4-Hydroxy-7-dimethylamino-pyrimido [4,5-d]pyrimidin und 12g Phosphorpentasulfid werden in 200 cma Pyridin gekocht, und es wird während 6 Stunden in einem Bade von 130 erhitzt. Man giesst dann die heisse Pyridinlösung auf 1500 cma Wasser, wonach beim Stehen ein brauner Niederschlag ausfällt. Man löst letzteren in verdünnter Natronlauge, behandelt mit Tierkohle, fällt mittels verdünnter Salzsäure und erhält nach Umkristallisation aus viel Methanol 4-Mercapto-7-dimethylamino-pyrimido [4,5 d]-pyrimidin der Formel

Beispiel 2
7 g 4-Methylmercapto-7-dimethylamino-pyrimido [4,5-d] pyrimidin werden mit 10 cm3 Piperidin in 40 cm3 Athanol während 8 Stunden unter Rückfluss gekocht. Anschliessend dampft man die Lösung im Vakuum zur Trockene ein und gibt zum Rückstand 3 cm3 10-n. alkoholische Salzsäure. Auf Zugabe von Ather tritt Kristallisation ein. Man erhält so das 4-Piperidino-7-dimethylamino-pyrimido [4,5-d]-pyr imidin-hydrochlorid der Formel

In analoger Weise kann man durch Umsetzung mit Ammoniak das 4-Amino-7-dimethylamino-pyr- imido [4,5-d] pyrimidin-hydrochlorid vom F. 330 bis 332 (Zersetzung) erhalten.

For the amino group in the 7-position of the new compounds, a mono- or disubstituted amino group is primarily suitable, it being possible for the substituents to be chosen from the aromatic, heterocyclic, araliphatic, heterocyclylaliphatic, cycloaliphatic or aliphatic series. Such substituents are particularly lower alkyl, cycloalkyl or alkylene radicals, which can also be interrupted in the carbon chains by heteroatoms such as oxygen, sulfur or nitrogen.
So the amino group can e.g. B. be a lower mono or dialkylamino, tert-aminoalkylamino, pyrrolidino, piperidino, morpholino or piperazino group. The amino group can also be inorganically substituted and z. B. represent a hydrazino group.
The new compounds contain free or substituted amino groups in at least one of the positions 2, 4 or 5. In two of the positions mentioned, they can also have free or etherified hydroxyl groups or lower alkyl radicals.
Lower alkoxy groups, such as methoxy, are particularly suitable as etherified hydroxyl groups, and those described above are primarily suitable as substituted amino groups.
The new compounds can also have substituents on the ring nitrogen atoms, preferably lower aliphatic radicals, such as lower alkyl or aminoalkyl radicals, e.g. B. methyl.
The new compounds have valuable properties. They are antimetabolites of purine. Furthermore, they show in particular tumor-inhibiting, but also diuretic, coronary-expanding, central and anti-rheumatic effects. They are intended to be used as remedies or as intermediates.

The process according to the invention for the preparation of the new compounds consists in that 7 aminopyrimido [4,5-d] pyrimidines, which have a free or etherified mercapto group or a halogen atom in at least one of the positions 2,4 and 5, with ammonia or amines.
The starting materials can be obtained by methods known per se.
The reactions mentioned can be carried out in the presence or absence of diluents, if appropriate at elevated temperature, in an open or, if desired, in a closed vessel, if appropriate under pressure.
Depending on the substituents present in the process products, various salts can be prepared. If they have free oxy groups, metal salts can be obtained, e.g. B. by dissolving in alkaline solutions. Compounds of a basic character form salts with inorganic or organic acids.
Examples of salt-forming acids are: hydrohalic acids, sulfuric acids, phosphoric acids, nitric acid, perchloric acid; aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids, such as formic, acetic, propionic, oxalic, amber, glycolic, milk, apple, wine, lemon, ascorbic, oxymalein, dioxymalein - or pyruvic acid; Phenylacetic, benzoic, p-aminobenzoic, anthranil, p-oxybenzoic, salicylic or p-aminosalicylic acid; Methanesulfonic, athanesulfonic, oxyethanesulfonic, ethylene sulfonic acid; Toluenesulfonic, naphthalenesulfonic or sulfanilic acid; Methionine, tryptophan, lysine or arginine.
The above-described pyrimidopyrimidines, their salts or mixtures thereof can, as mentioned, be used as medicaments.
In the following examples, the temperatures are given in degrees Celsius.

11 g of 4-methylmercapto-7-dimethylamino-pyrimido- [4,5-d] pyrimidine are heated to 100 to 125 for 8 hours together with 80 cm liquid dimethylamine in a closed tube. After evaporation of the excess dimethylamine, the residue is in 100 cm3 2-n. Hydrochloric acid added.
After filtering off a little undissolved matter, 10-n. Sodium hydroxide solution to pH = 10 and mixed with solid caustic potash. Here an 01 precipitates, which soon solidifies in crystalline form.
