Als Substituenten in 2-Stellung kommen vor allem niedere Alkylreste, wie Methyl, Äthyl, Propyl, Isopropyl, Isobutyl, Isoamyl, Cycloalkylreste, wie Cyclopentyl oder Cyclohexyl, Hydroxyniederalkyl- reste, wie ss-Hydroxyäthyl, Halogennsederalkylreste, wie ss-Chloräthyl oder Arylreste,
wie Phenyl, Halogen phenyl, Alkylphenyl oder Alkoxyphenyl, in Frage.
Der organische Rest in 6-Stellung ist z. B. ein niederer Alkylrest.
Die genannten Verbindungen können leicht N- alkyliert werden.
Niedere Alkylreste als N-Substituenten im Pyri- midinkern sind z. B. Methyl und Äthyl.
Die geschilderten Verbindungen zeichnen sich besonders durch diuretische., koronarerweiternde und auch den Metabolismus von Purin beeinflussende Wirkung aus und können als Heilmittel Verwen dung finden.
Das erfindungsgemässe Verfahren besteht darin, dass man Verbindungen der Formeln

Wenn erwünscht, können in erhaltenen 2 - substituierten 4 - Oxo - 4,5 - dihydro- pyrazolo[3,4-d]pyrimidinen oder ihren tautomeren Formen substituierbare Pyrimidinringstickstoffatome alkyliert werden.
Zur Umsetzung mit den Aminopyrazolen verwen det man in erster Linie funktionelle Derivate der Ameisensäure, z. B. Formamid, oder niedere Fett säuren in Form ihrer Anhydride, Ester oder Halo genide. Dabei verwendet man im Falle der stickstoff freien Carbonsäure-derivate Pyrazol-4-carbonsäuren in Form ihrer eine Aminogruppe aufweisenden Säurederivate, wie des Amids. Bei solchen Umsetzun gen können zunächst offenkettige Zwischenprodukte entstehen.
Die Kondensation der Aminopyrazole zu den Pyrazolo-pyrimidinen erfolgt vorzugsweise bei Tempe raturen über 100 , gegebenenfalls in Anwesenheit von Verdünnungs- und/oder Kondensationsmitteln im offenen oder geschlossenen Gefäss. Die allfällige nachträgliche Alkylierung erfolgt z. B. in üblicher Weise.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie saure Oxygruppen, so kön nen Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Cha- rakter bilden Salze mit anorganischen oder organi schen Säuren.
Als salzbildende Säuren kommen bei spielsweise in Frage: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicylische, aromatische oder hetero- cyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal , Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-,
Dioxymalein- oder Brenztraubensäure; Phenylessig , Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl- oder p-Aminosalicylsäure; Methansulfon-, Äthansulfon-, Oxyäthansulfon-, Äthylensulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure;
- Methionin, Tryptophan, Lysin oder Arginin. Die neuen pharmakologisch wertvollen Verbin dungen, ihre Salze oder entsprechende Gemische können z. B. in Form pharmazeutischer Präparate Verwendung finden. Diese enthalten z.
B. die ge nannten Verbindungen in Mischung mit einem für die enterale, parenterale oder topicale Applikation. geeigneten pharmazeutischen organischen oder an organischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den beschriebenen Verbindungen nicht reagieren, wie z. B.
Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanz liche Öle, Wasser, Benzylalkohole, Gummit, Poly- alkylenglykole, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw.
oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel. Sie kön nen auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Präparate werden z. B. nach üblicher Methode gewonnen.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden hergestellt werden. Die Temperaturen sind in den nachfolgenden Beispielen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 19,7 g 1-Isopropyl-3-amino-4-carbäthoxy-pyrazol werden in 50 cm3 Formamid während 5 Stunden in: einem Bade von 200-2l0 erhitzt. Nach dem Erkal ten nutscht man von dem ausgefallenen kristallinen Niederschlag ab und kristallisiert diesen zur Reini- gung aus siedendem Äthylalkohol. 2-Isopropyl-4- oxy-pyrazolo[3,4-d]pyrimidin der Formel

Das Ausgangsmaterial kann wie folgt erhalten werden: Eine Lösung von 11,6g Ni Isopropyl-N2- acetyl- hydrazin und 17g Äthoxymethylen-cyanessigester in 250 em3 Äthanol wird während 12 Stunden am Rückfluss gekocht.
Man dampft dann vom Äthanol im Vakuum ab, versetzt den /3-(N; Acetyl-Ni iso- propyl-hydrazino)-a-cyan-arylsäureäthylester enthal tenden öligen Rückstand mit 150 cm3 8n alkoholi scher Salzsäure und kocht während 2 Stunden am Rückfluss. Hierauf dampft man erneut im Vakuum ein, nimmt den Rückstand in 2n wässriger Salzsäure auf, nutscht von wenig Ungelöstem ab und stellt mit Natronlauge auf pH = 8-9.
Nun wird mit Chloro form ausgezogen und der Chloroformrückstand aus Cyclohexan umkristallisiert. Man erhält so 1-Iso- propyl-3-amino-4-carbäthoxy-pyrazol der Formel

B. wie folgt er halten: Eine Lösung von 65g Nl-Methyl-N2 benzyliden- hydrazin und 85g Äthoxymethylen-cyanessigester in 500 cm s Benzol wird 10 Stunden unter Rückfluss gekocht. Es entsteht ein Niederschlag, den man ab filtriert und aus Äthanol umkristallisiert. Man erhält den ss-(N#,-Benzyliden-Ni methyl-hydrazino)-a-cyan- acrylsäureäthylester in schwach gelben Kristallen vom F. 155-156 .
80 g ss - N. - Benzyliden-N,- methyl-hydrazino)-a- cyan-acrylsäureäthylester werden 2 Stunden mit 10n alkoholischer Salzsäure unter Rückfluss gekocht. Das Lösungsmittel entfernt man hierauf durch Destilla tion im Vakuum. Der Rückstand wird in 200 cm-' 2n Salzsäure aufgenommen und die saure Lösung mit Äther ausgeschüttelt. Nach Abtrennung der wässrigen Schicht wird diese durch Zugabe von 2n Natronlauge alkalisch gemacht. Die ausgeschiedene Base extrahiert man durch mehrmaliges Ausschüt teln mit Äther.
Der nach dem Abtrennen, Trocknen und Eindampfen des Ätherextraktes verbleibende Rückstand wird bei 130 /0,01 mm Hg destilliert. Es wird so 1-Methyl-3-amino-4-carbäthoxy-pyrazol der Formel

29 g dieses Esters werden mit 11 cm3 konz. Salzsäure in 100 cm3 Äthanol 30 Minuten unter Rückfluss gekocht. Das Lösungsmittel wird hierauf eingedampft, und der Rückstand wird mit Äther ausgewaschen. Es verbleibt eine kristalline Masse, die nach Zugabe von 2n Natronlauge mit Äther extrahiert wird. Nach dem Trocknen und Eindamp fen des Lösungsmittels bleibt ein Öl zurück, das nach einigem Stehen kristallisiert.
