worin R,; Methyl oder Äthyl bedeutet, R3 für ein Wasserstoffatom oder einen niederen Alkylrest steht und R, einen Oxaalkyl-, Cycloalkyl- oder Cyclo- alkylalkylrest oder einen:
Aralkyl- oder HeterocYclyl- alkylrest oder einen Alkylrest mit mehr als zwei Kohlenstoffatomen bedeutet, oder ihrer Salze.
In den neuen Verbindungen kommen als Alkyl- reste insbesondere niedere Alkylreste, wie Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Pen- tyl-(1)-, Pentyl-(2)-, Pentyl-(3)-, 2 Methylbutyl-(3)- oder Hexylreste, und als Oxa-al'kylreste z.
B. 3-Oxa- pentyl-(5)- oder 3-Oxa-@heptyl-(6)-reste in Betracht.
Cycloalkyl- oder Cycloalkylreste sind beispiels weise Cyclopentyl- oder Cyclohexylreste, oder Cy- clopentyl- oder Cyciohexyl-metiiyl-, -äthyl- oder -pro- pylreste.
Als Aralkylreste kommen insbesondere Phenyl- alkylreste, wie 1- oder 2-Phenyläthyl- oder Phenyl- methylreste, in Frage, in denen die aromatischen Kerne Substituenten tragen können, wie niedere Al kylreste oder freie oder substituierte Hydroxy-, Amino- oder Mercaptogruppen, Halogenatome, Tri- fluormethyl- oder Nitrogruppen.
In den genannten substituierten Hydroxy-, Mercapto- oder Amino- gru.ppen sind die Substituenten insbesondere solche der oben genannten Art, vor allem niedere Alkyl- Teste, so dass es sich z.
B. um Mefthoxy-, Äthoxy-, Propoxy- oder B.utoxygruppen, entsprechende Alkyl- mercaptogruppen, A.lkylendioxydgruppen, wie Me- thylendioxydgruppen, Mono- oder Dialkylamino- gruppen, wie Mono- oder Dknethyl-, -äthyl-, "pro pyl-, -butyl- oder zpentylaminogruppen handelt. Als Halogenatome sind vor allem Fluor-, Chlor- oder Bromatome zu nennen.
Die Aralkylreste können mehrere Arylreste enthalten, wie z. B. im Diphenyl- methylrest.
Als Heterocyclylalkylreste seien beispielsweise Pyridylmethyl-, Thenyl- oder Furfurylreste, die im heterocyclischen Rest wie oben für ,die Arylreste an gegeben, substituiert sein können, genannt.
Die neuen Verbindungen und ihre Salze besitzen wertvolle pharmakologische Eigenschaften. Insbeson dere sind sie coronarerweiternd wirksam. Die neuen Verbindungen können somit .als Heilmittel, .insbeson dere bei Durchblutungsstörungen des Herzmuskels, aber auch als Zwisdhenprodukte zur Herstellung sol cher Heilmittel dienen.
Besonders wertvoll als coronarerweiternde Mit tel sind Verbindungen der Formel

und ihre tautomeren Formen und Salze davon, wor in R3 für Wasserstoff oder Niederalkyl steht, Rs Methyl oder Äthyl darstellt und R1 einen Cyclo- alkylrest, wie Cyclopentyl oder Cyclohexyl,
oder vor allem einen Alkylrest mit mindestens 3 Kohlenstoff- atomen, wie Isopropyl, Butyl-(2), Pentyl-(2) oder (3), oder einen Oxaalkylrest, wie 3-Oxalpentyl-(5) oder 3-Oxa-heptyl-(6) darstellt.
Zu nennen ist besonders das 1-Isopropyl-4-hy- droxy-6-äthyl"pyrazolo[3,4-d]pyrimidin und seine Salze.
Das erfindungsgemässe Verfahren zur Herstel lung der neuen Verbindungen ist dadurch gekenn zeichnet, dass man eine 2-R1-3-amino-5 R3-pyrazol- 4-carbonsäure oder ein eine Oxogruppe enthaltendes funktionelles Säurederivat davon, wie einen Ester, z.
B. einen Alkylester, ein Halogenid oder dies Amid mit einer Carbonsäure der Formel R6-COOH oder einem reaktionsfähigen, funktionellem Säurederivat davon, wie :einem Ester, z.
B. einem Alkylester, An hydri.d, Halogenid, Iminoäther, dem Amid, Amidin, einem entsprechenden Schwefelderivat, oder dem Nitril, mit der Massgabe umsetzt, das mindestens eine der Carboxylgruppen stickstoffhaltig funktionell abgewandelt ist.
Die Reaktion kann z. B. so erfolgen, dass man eine 2-R1-3-Amino-5 R,3-pyrazol-4-carbons:äure oder einen Ester oder ein Halogenid .davon mit der Car bonsäure der Formel R6- COOH in Form ihres Amids, Thioamids, Nitrils, oder Amidin:s oder ein 2-R1-3-Amino-5-R3-ipyrazol-4-carbonsäureamid mit der Carbonsäure :
der Formel Ra-COOH, ihrem An hydrid, oder einem Ester, Halogenid, Iminoäther, Thioiminoäther, oder ihrem Amid, Thioamid, Ami- din oder Nitrit umsetzt.
Man kann aber auch so vorgehen, dass man zu nächst die Aminogruppe der 3-Amino-2 R1-5 R pyrazol-4-carbonsäure oder ihrer Derivate mit der Carbonsäure der Formel RE-COOH oder ihren De rivaten, vor allem .den Anhydriden oder Halogeniden, acyliert und in einer 2. Reaktionsstufe, den Ring schliesst.
Die Kondensation der Aminopyrazole zu den Pyrazolopyrimidinen erfolgt vorzugsweise bei erhöh ter Temperatur gegebenenfalls in Anwesenheit von Verdünnungs- und/oder Kondensationsmitteln im offenen oder geschlossenen Gefäss. überraschender weise wurde dabei gefunden, dass .die Umsetzung sehr vorteilhaft unter Verwendung der 2 R1-3- Amino-5-R3-pyrazol-4,carbonsäureester, z.
B. der Alkylester, und eines Nitrits :der Formel R6- CN vor genommen wird, wobei zweckmässig ein Konden sationsmittel, z. B. ein Alkalimetall, wie Natrium verwendet werden kann. Hierbei ist auch die Ver wendung von Verdünnungsmitteln, wie Benzol, To luol, Xylol oder Äthern, vorteilhaft.
Die erhaltenen 4 Hydroxy-pyrazolopyrimidine können in üblicher Weise in ihre Salze mit Basen, z. B. in ihre Metallsalze, wie Alkalimetallsalze, um gewandelt werden, z. B. durch Lösen in Alkalilaugen. Die Salze ihrerseits lassen sich in die freien. Hydroxy- verhindungen umwandeln, zweckmässig durch Be handlung mit Säuren.
Die neuen, pharmakologisch wertvollen Verbin dungen, ihre Salze oder entsprechende Gemische können z. B. in Form pharmazeutischer Präparate Verwendung finden. Diese enthalten die genannten Verbindungen in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeu tischen organischen oder anorganischen Träger material.
Die verfahrengemäss erhaltenen. Endstoffe sind aber auch wertvolle Zwischenprodukte, z. B, für die Herstellung der im Schweizer Patent Nr. 390264 be schriebenen 4-Aminoverbindungen.
Sofern die beim erfindungsgemässen Verfahren verwendeten Ausgangsstoffe neu sind, lassen sie sich nach an siclh bekannten Methoden herstellen.
Als Ausgangsstoffe werden gemäss der vorlie genden Erfindung vorzugsweise diejeni gen verwen det, die zu den eingangs als besonders wertvoll ge schilderten Endstoffen führen.
In den nachfolgenden Beispielen sind die Tem peraturen in Celsiusgraden .angegeben.
<I>Beispiel 1</I> In eine Lösung von 9,9 g 2-Isopropyl-3-amino-4- carbäthoxy-pyrazol in 100g Propionitril werden un ter Kühlung mit Wasser 2,3 g Natrium fein zerklei nert eingetragen. Sobald die exotherme Reaktion be endet ist, erhitzt man während 4 Stunden unter Rüh ren auf 100-110 , versetzt nach dem Erkalten mit 100 Cm2 Alkohol und dampft im Vakuum zur Trockne ein. Der Rückstand wird in 150 cm3 2-n.
Natronlauge aufgenommen, die alkalische Lösung wird zur Abtrennung von Ungelöstem mit Chloro form ausgeschüttelt und dann mit 6-n. Salzsäure auf pH-6 gestellt, wonach ein schmieriges Produkt aus fällt. Dieses wird aus Alkohol umkristallisiert.
Man hält so 1-Isopropyl-4@hydroxy-6-äthyl-pyrazolo[3,4- d]pyrimidin der Formel

in farblosen Kristallen vom F. 180-182 . <I>Beispiel 2</I> Zu einer Lösung von 9,9 g 2-Isoprapyl-3-amino- 4-carbäthoxy-pyrazol in 50 cm3 Acetonitril gibt man 2,3 g Natrium unter Eiskühlung. Die Temperatur darf während der Reaktion nicht über 30o steigen.
Sobald die exotherme Reaktion beendet ist, erhitzt man während 4 Stunden auf 90-95e. Das Reaktions gemisch wird dann abgekühlt und mit 100 cm\' A.thanol versetzt. Man dampft im Vakuum zur Trockne ein, gibt zum Rückstand l50 cm3 2-n. Na tronlauge und extrahiert das überschüssige Aceto- nitril mit Chloroform. Die wässerige Phase wird durch Zugabe von 5-n.
Salzsäure auf pH 3-4 ge- stellt, wobei nach längerem Stehen ein fester Nieder schlag ausfällt, den man abfiltriert und aus Äthanol umkristallisiert. Man & hält das 1-Isopropyl-4 hy- d.roxy-6-methyl-pyrazolo[3,4-d]pyrimidin der Formel
