<B>Verfahren zur Herstellung neuer</B> 4-Mereapto-pyrazolo [3,4-dlpyrimidine Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 2-substituierter 4-Mercapto-6-R- pyrazolo [3,4-d] pyrimidine, worin R für Wasserstoff, einen niederen Alkylrest oder eine freie oder ver- ätherte Mercaptogruppe steht.
Der Substituent in 2-Stellung ist besonders ein Alkyl-, wie ein niederer Alkylrest, ein Cycloalkylrest oder ein Hydroxynieder- alkylrest, z. B. ein Methyl-, Äthyl-, Propyl-, Isopro- pyl-, Isobutyl-, Isoamyl-, Cyclopentyl- oder Cyclo- hexylrest oder ein ss-Oxy-äthylrest.
Die neuen Verbindungen, ihre 4-Thioäther und ent sprechende Salze besitzen wertvolle pharmakologische Eigenschaften. So zeigen sie eine koffeinartige Wir kung. Ferner besitzen sie tumorhemmende und auch antibakterielle und antimykotische Eigenschaften. Die neuen Verbindungen sollen daher als Heilmittel Ver wendung finden. Sie sind aber auch wertvolle Zwi schenprodukte für die Herstellung von Heilmitteln. So lassen sich in den neuen Verbindungen freie oder verätherte Mercaptogruppen gegen Aminogruppen austauschen.
Das erfindungsgemässe Verfahren besteht darin, dass man in 2-substituierten 4-Oxy-6-R"-pyrazolo- [3,4-d]pyrimidinen, worin R" dasselbe wie R' oder die Oxygruppe bedeutet, die Oxygruppen durch Um setzung mit einer Schwefel abgebenden Verbindung, z. B. Phosphorpentasulfid, gegen freie Mercaptogrup- pen austauscht. Die freien Mercaptogruppen können anschliessend veräthert werden.
Die Ausgangsstoffe lassen sich dadurch gewinnen, dass man 1-substitu- ierte 3-Amino-pyrazol-4-carbonsäureester oder -amide mit Carbonsäuren in Form ihrer funktionellen Deri vate oder der freien Säuren unter der Massgabe um setzt, dass in den Reaktionspartnern wenigstens eine der beiden Carboxylgruppen eine zum Ringschluss geeignete Aminogruppe enthält. Zur Umsetzung mit den Aminopyrazolen verwen det man in erster Linie funktionelle Derivate der Ameisensäure, z.
B. Formamid, oder der Kohlen säure, wie Harnstoff, Thiohamstoff, Halogenkohlen säureester, Kohlensäuredihalogenide, z. B. Phosgen, Cyanate, z. B. Alkalicyanate, oder Thiocyanate, oder niedere Fettsäuren in Form ihrer Anhydride, Ester oder Halogenide. Dabei verwendet man im Falle der stickstofffreien Carbonsäurederivate die Pyrazol- 4-carbonsäure in Form des Amids.
Die Kondensation der Aminopyrazole zu den Pyrazolo-pyrimidinen erfolgt vorzugsweise bei Tem peraturen über 100 C, gegebenenfalls in Anwesen heit von Verdünnungs- und/oder Kondensationsmit- teln im offenen oder geschlossenen Gefäss.
Die zur Herstellung der Ausgangsstoffe benötigten 1-substituierten 3-Amino-pyrazol-carbonsäureester oder -amide können dadurch gewonnen werden, dass man a-Cyano-ss-oxopropionsäuren, gegebenen falls in Form ihrer funktionellen reaktionsfähigen Säure- und/oder Oxoderivate mit N-monosubstitu- ierten Hydrazinen, die am N'-Stickstoffatom einen durch Hydrolyse oder Alkoholyse abspaltbaren Rest tragen, kondensiert,
die erhaltenen offenkettigen Pro dukte mit hydrolysierenden oder alkoholysierenden Mitteln behandelt und zugleich oder nachträglich den Pyrazolring schliesst und, falls der Rest in 4- Stellung keine veresterte oder amidierte Carboxyl- gruppe ist, diesen Rest in die veresterte oder amidierte Carboxylgruppe überführt.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie saure Mercaptogruppen, so können Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Charakter bilden Salze mit anorganischen oder orga- nischen Säuren.
Als salzbildende Säuren kommen beispielsweise in Frage: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Per chlorsäure; aliphatische, alicychsche, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Gly- kol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-,
Dioxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Amino-benzoe-, Anthranil-, p-Oxy-benzoe-, Salicyl- oder p-Amino-salicylsäure; Methansulfon-, Äthansulfon-, Oxyäthansulfon- oder Äthylensulfonsäure; Toluolsulfon-, Naphthalinsulfon- säuren oder Sulfanilsäure;
Methionin, Tryptophan, Lysin oder Arginin.
Die neuen, pharmakologisch wertvollen Verbin dungen, ihre Salze oder entsprechende Gemische können in Form pharmazeutischer Präparate Ver wendung fänden. Diese enthalten die genannten Ver bindungen z. B. in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeig neten pharmazeutischen organischen oder anorgani schen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den beschriebenen Verbin dungen nicht reagieren, wie z. B.
Gelatine, Milch zucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Wasser, Benzylalkohole, Gummi, Polyalkylen- glykole, Cholesterin oder andere bekannte Arznei mittelträger.
Die pharmazeutischen Präparate können als Tabletten, Dragees oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisie- rungs-, Netz- oder Emulgiermittel. Sie können auch noch andere therapeutisch wertvolle Stoffe enthal ten. Die Präparate können nach üblichen Methoden gewonnen werden.
Die Temperaturen sind in den nachfolgenden Bei spielen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 10 g 2-Isopropyl-4-hydroxy-pyrazolo[3,4-d]pyri- midin und 15 g Phosphorpentasulfid werden in <B>100</B> cm3 Pyridin gebracht und während 4 Stunden in einem Bade von 130 erhitzt. Man giesst dann die heisse Pyridinlösung auf 1000 cm3 Wasser, wonach beim Stehen ein brauner Niederschlag ausfällt.
Man löst letzteren in verdünnter Natronlauge, behandelt mit Tierkohle, fällt mittels verdünnter Salzsäure und erhält nach Umkristallisation aus Äthanol 2-Isopro- pyl-4-mercapto-pyrazolo [3,4-d] pyrimidin der Formel

Das Ausgangsmaterial kann wie folgt erhalten werden: 8 g Ni Isopropyl-NZ benzyliden-hydrazin und 8 g Äthoxymethylen-cyanessigester werden in 50 cm3 Benzol während 10 Stunden auf<B>800</B> erwärmt. Nach dem man das Lösungsmittel im Vakuum entfernt hat, wird der Rückstand aus Äthanol umkristallisiert. Man erhält den ss-(N.- Benzyliden-Nl-isopropyl-hydra- zino)-a-cyan-acrylsäureäthylester in gelben Prismen vom F. 118-120 .
4 g (3-(N2 Benzyliden-Nl isopropyl-hydrazino)-a- cyan-acrylsäureäthylester kocht man 2 Stunden mit 10n alkoholischer Salzsäure und entfernt hierauf den Alkohol durch Destillation im Vakuum. Der Rückstand wird in 200 cm3 2 n Salzsäure aufgenom men, und die Lösung wird mit Äther extrahiert. Nach dem Abtrennen der wässerigen Lösung wird diese durch Zugabe von 2 n Natronlauge alkalisch gemacht. Die ausgeschiedene Base extrahiert man mit Äther.
Nach dem Trocknen und Abdampfen des Äthers bleibt 1-Isopropyl-3-amino-4-carbäthoxypyra- zol der Formel

Das Ausgangsmaterial kann wie folgt erhalten werden: Eine Lösung von 65g Ni Methyl-N2 benzyliden- hydrazin und 85g Äthoxymethylen-cyanessigester in 500 cm3 Benzol wird 10 Stunden unter Rückfluss gekocht. Das Lösungsmittel entfernt man hierauf durch Destilla tion im Vakuum. Der Rückstand wird in 200 cm3 2 n Salzsäure aufgenommen und die saure Lösung mit Äther ausgeschüttelt. Nach Abtrennung der wäs serigen Schicht wird diese durch Zugabe von 2 n Natronlauge alkalisch gemacht. Die ausgeschiedene Base extrahiert man durch mehrmaliges Ausschüt teln mit Äther.
Der nach dem Abtrennen, Trocknen und Eindampfen des Äther-Extraktes verbleibende Rückstand wird bei 130 /0,01 mm Hg destilliert. Es wird so 1-Methyl-3-amino-4-carbäthoxy-pyrazol der Formel
