wobei R, und R2 Wasserstoff oder Alkylgruppen dar stellen, die geradkettig oder verzweigt sein können und deren Kette durch Heteroatorne unterbrochen sein kann.
R3 bezeichnet ein Wasserstoffatom, eine gegebenen falls durch weitere Gruppen<B>-</B> wie z. B. Halogen, Fly- droxy- oder Aminogruppen <B>-</B> substituierte Alkyl-, Cyclo- alkyl-, Aryl-, Aralkyl-, I-lydroxyalkyl-, Dialkylamino- alkylgruppe, wobei die Alkylreste der Dialkylamino- gruppe auch mit dem Stickstoffatom einen Ring bilden können,
der gegebenenfalls ein weiteres Heteroatom enthalten kann, eine Carbalkoxy-, Alkylearbalkoxy-, Carbamyl- oder Alkylearbamylgrtippe.
Xanthinderivate obiger Struktur sind bislang nach der Traube-Synthese über das 4,5-Dianünouracil herge stellt worden, das mit entsprechenden Säurederivaten acyliert und anschliessend cyclisiert wurde. Teilweise sind diese Verbindungen auch über 8-Halogenxanthin- derivate, die gleichfalls nach der Traube-Synthese mit anschliessender Halogenierung hergestellt werden, zu gänglich.
Dieser Syntheseweg besitzt mehrere Nach-teile. Ein mal sind eine relativ grosse Zahl von Verfahrensstufen erforderlich, die sich sowohl auf die Ausbeuten als auch auf die Kapazität einer gegebenen Anlage negativ aus wirken.
So muss das aus dem 4-Aminouracil durch, Nitro- sierung hergestellte 4-Amino-5-nitroso-uracü reduziert werden. Das geschieht im technischen Massstab mit Ammoniumsulfid oder mit Eisen und Salzsäure, wobei entweder grosse Mengen Schwefelwasserstoff oder Was serstoff auf treten.
Darüber hinaus sind die 4,5-Diamino-uracile sehr leicht oxydierbar und neigen unter Verfärbungen zur Bildung von Nebenprodukten. Die Verfärbungen gehen bis in die Endprodukte und lassen sich schwer entfernen. Man muss die, Diamine daher stets schnell verarbeiten und darf sie nie längere Zeit in einem alkalischen Milieu lassen.<B>[E.

Daran schliesst sich die Acylierung mit zum Teil schwer zugänglichen Säuren an und anschliessend noch die Cyclisierung. Bei der Herstellung einiger in 8-Stel- Jung mit komplizierteren Gruppen substituierter Xanthin- derivat6 wird der Syntheseweg noch länger, da erst das in 8-Stellung unsubstituierte Xanthinderivat hergestellt werden muss, das halogeniert und anschliessend mit einer weiteren Komponente zur Reaktion gebracht werden muss.
Die Xanthinsynthesen sind sehr ausgiebig bearbeitet worden, einerseits um neue Derivate herzustellen, ander seits um die Verfahren zur Herstellung von Coffein, Theophyllin und Theobromin, die in tausenden von Tonnen jährlich produziert werden, zu verbessern. Es war deshalb anzunehmen, dass alle neuen Möglichkeiten der Synthese erschöpft sind.
überraschenderweise wurde nun gefunden, dass man zu den Verbindungen der eingangs beschriebenen Struk tur auf einem wesentlich kürzeren und einfacheren Weg gelangen kann, wenn man die entsprechenden substi tuierten 4-Amino-5-nitroso-uracile der Struktur

wobei RI, R2 und R3 die obige Bedeutung besitzen, in Abwesenheit oder in Gegenwart von organischen Lö sungsmitteln durch Wasserabspaltung zu den jeweiligen Xanthin cyclisiert. Es ergibt sich folgende Reaktions gleichung.

Die Reaktion kann sowohl durch trockenes Erhit zen, als auch durch Erhitzen. in Lösungsmitteln durch geführt werden.
Beim trockenen Erhitzen variieren die Temperaturen<B>je</B> nach der Verbindung. In der Regel werden hierzu Temperaturen zwischen<B>50</B> und 200'<B>C</B> benötigt.
Für die Umsetzung in Lösung sind z. B. folgende Lösungsmittel geeignet: Dimethylformamid, Benzol, To- luol, Xylol, Alkohole, wie Äthanol, Propanol, Butanol.
Die Umsetzung in. Lösung ist leicht und einfach durchzuführen und gleicht technologisch einer Umkri stallisation der Nitrosoverbindungen, wobei jedoch Xanthinderivate entstehen. Hier zeigt sich deutlich die Vereinfachung gegenüber den bisherigen Xanthinsyn- thesen.
Das Ende der Umsetzung ist leicht an der vollstän digen Entfärbung des Reaktionsgutes zu erkennen, da die eingesetzten Nitrosoderivate gefärbt, die Endpro dukte, dagegen ungefärbt sind. Die Reaktionszeit ist von der gewähltenReaktionstemperatur abhängig, mit stei gender Temperatur wird sie stark verkürzt, so dass nur ein Erhitzen von wenigen Sekunden oder Minuten er forderlich ist. Da die Endprodukte sich nicht weiter umsetzen, wirkt sich eine zu lange Reaktionszeit nicht nachteilig aus.
Diese Reaktion weicht von allen anderen Synthese wegen grundsätzlich ab und benötigt nur einen Reak tionsschritt von der Nitrosoverbindung bis zum Xanthin, während die Traubesche Synthese hierfür<B>3</B> Stufen be nötigt, nämlich Reduktion, Acylierung und Ringschluss.
Die Ausgangsprodukte lassen sich nach bekannten Methoden in einfacher Weise bequem herstellen: <B>1.</B> Durch Umsetzung von gegebenenfalls in<B>1-</B> und/ oder 3-Stellung entsprechend substituierten 4-Chlor- uracilen mit den entsprechenden Aminen der Struktur H2N-CH2-R3 wobei R3 die oben genannte; Bedeutung besitzt. [W. Pfleiderer und K.
H. Schündehütte, <B>A.<I>612, 158</I></B> <B>(1958)]</B> und anschliessende Nitrosierung. Die benötigten 4-Chloruraeile sind leicht zugänglich und teilweise in einer Reaktionsstufe zu erhalten.<B>[G.</B> Strauss,<B>A.<I>638,</I></B> <B>205</B> (1960)1. 2. Durch Umaminierung von gegebenenfalls in 1- Stellung und/oder 3-Stellung entsprechend substituierten 4-Anuno-uraeilen mit den entsprechenden Amin-hydro- chloriden.
H2N-CH2-R3 <B>-</B> HCI <B>[C.</B> W. V#hitehead und<B>J. J.</B> Traverso, <B>J.</B> Am. Chem. Soc. <B>82, 3971 (1960)]</B> und anschliessende Nitrosierung.
Die Nitrosierungen können nach den üblichen Methoden, z. B. in Wasser mit Natriumnitrit oder in Alkohol mit Amylnitrit <B>je</B> nach den Eigenschaften der Nitrosoderivate, ausgeführt werden.
Voraussetzung für den in dieser Erfindung beschrie benen Ringschluss zu Xanthinderivaten ist das Vorhan densein einer Methyl- bzw. Methylengruppen in a-Stel- lung zur 4-Aminogruppe.
Wenn die in a-Stellung zur 4-Aminogruppe stehende Methylengruppe, durch andere, zusätzlich im Rest R3 stehende Gruppen aktiviert wird, tritt<B>je</B> nach Stärke der Aktivierung der Ringschluss zum Xanthinderivat in sol chen Fällen bereits während der Nitrosierung in statu nascendi,<B>d.</B> h. bei<U>Zimmert</U> mperatur oder darunter ein. So tritt z.
B. bei der Nitrosierung des 1,3-Dimethyl- uracüyl-4-anünoessigsäureät'hylesters zuerst eine Violett- färbung auf (Bildung des Nitrosoderivates), die im wei teren Verlauf immer heller wird und schliesslich zur Abscheidung des weissen 8-Carbäthoxytheophyllins führt. Dasselbe kann bei der Nitrosierung des 1,3-Dimethyl-4- benzylaminouracils beobachtet werden, wo das 8-Phe- nyltheophyllin erhalten wird.
In einigen Fällen, z. B. bei der Nitrosierung von 1,3- Dimethyl-4-isobtitylamino-uraeil, erhält man ein Ge misch von roten und fast weissen Kristallen. Es ist hier ebenfalls schon in der Nitrosierungslösung eine teilweise Wasserabspaltung unter Xanthinbildung eingetreten.
Sofern die Nitrosierung in alkoholischer Lösung durchgeführt wurde, kann leicht durch Erhitzen und Einengen der Nitrosierungslösung unter anderem im Vakuum die Umsetzung zum Xanthinderivat vollendet werden. Man kann aber auch das erhaltene Gemisch, bestehend aus dem 5-Nitrosoderivat und dem gewünsch ten Xanthinderivat, isolieren und anschliessend trocken erhitzen oder durch Erhitzen in einem organischen Lö- sungsmittel die Umsetzung zum Xanthinderivat zu Ende führen.
Man kann ferner die ausgegebenenfalls in<B>1-</B> und/ oder 3-Stellung entsprechend substituierten 4-Amino-5- nitroso-uracilen durch Umaminierung analog der von <B>C.</B> W. Whitehead und<B>J.
J.</B> Traverso, <B>J.</B> Am. Chem. Soe. <B>82, 3971 (1960)</B> beschriebenen Arbeitsweise erhält lichen 4-Amino-5-nitroso-uracilderivate der Struktur 11 in statu naseendi durch Wasserabspaltung zu den ent sprechenden Xanthinderivaten der Struktur<B>1</B> cyclisieren.
Durch das erfindungsgemässe Verfahren lassen sich nicht nur komplizierte in 8-Stellung substituierte Deri vate herstellen, sondern auch die in 8-Stellung unsub- stituierten Grundkörper. Wird auf diese Weise z. B. Theophyllin hergestellt, so kann man daraus in an sich bekannter Weise sehr vorteilhaft Coffein gewinnen.
Die Struktur dieser Verbindungen wurde durch die Elementaranalyse und die<B>-</B> UV-Spektren festgelegt. Ausserdem ist ein grosser Teil der Verbindungen auf anderem Wege über die klassische Traubesche Synthese zugänglich und in der Literatur beschrieben, so. dass auch die Schmelzpunkte zur Identifizierung herangezogen werden konnten. Der Strukturbeweis ist wichtig, da der Reaktionsverlauf nicht vorauszusehen war und auch andere Umsetzungssetzungen möglich erschienen.
<I>Beispiel<B>1</B></I> 4,2<B>g</B> 1,3-Dimethyl-4-methylaminouracil werden in <B>25</B> cm3 Äthanol gelöst und langsam mit 12 cm3 Iso- amylnitrit versetzt. Zur vollständigen Nitrosierung wer den<B>3</B> Tropfen konzentrierte HCI zugegeben. Man lässt nach etwa<B>30</B> Minuten lang rühren und kühlt die Mi schung dann in Eis. Die rote 5-Nitrosoverbindung wird abgesaugt, mit Alkohol ausgewaschen und getrocknet.
Smp. 260-2621 <B>C.</B> (Entfärbung bei<B>153' C</B> und Um wandlung in das Xanthinderivat.) Die rote Nitrosover- bindung wird mit etwa<B>50</B> cm3 Xylol bis zur Entfärbung unter Rückfluss gekocht (l/4 Stunde). Nach dem<B>Abküh-</B> len wird das Theophyllin abgesaugt und getrocknet. Ausbeute:<B>95-98 %,</B> Smp. <B>273-274' C.</B> (Nach<B>J.</B> H.

<I>Beispiel 2</I> 4,5<B>g</B> 1,3-Dimethyl-4-äthylaminouracil werden in <B>35</B> em# Äthanol gelöst und langsam 12 cm3 Isoamyl- nitrit zugetropft. Zur vollständigen Nitrosierung werden <B>3</B> Tropfen konzentrierte HCI zugegeben. Man lässt noch etwa<B>30</B> Minuten lang rühren und stellt über Nacht in den Kühlschrank.
Der hellrote und mit weissen Kristal len durchsetzte Niederschlag wird abgesaugt und mit Alkohol ausgewaschen. Smp.: 270-315' <B>C,</B> Entfärbung bei<B>135' C</B> und Umwandlung in Xanthinderivat. Die Rohsubstanz (Nitrosoverbindung und Xanthinderivat) wird wenige Minuten bis zur vollständigen Entfärbung in Butanol (20 cm3) erhitzt. Ausbeute: 94<B>%,</B> 1,3,8-Tri- methylxanthin. Nach Umkristallisation aus Wasser Smp.: <B>3290 C.</B> (Nach<B>J.</B> H.

R3 denotes a hydrogen atom, if necessary by further groups <B> - </B> such as. B. halogen, fly droxy or amino groups <B> - </B> substituted alkyl, cycloalkyl, aryl, aralkyl, I-hydroxyalkyl, dialkylaminoalkyl groups, the alkyl radicals of the dialkylamino group also can form a ring with the nitrogen atom,
which may optionally contain a further heteroatom, a carbalkoxy, alkylene alkoxy, carbamyl or alkylearbamyl group.
Xanthine derivatives of the above structure have so far been produced after the grape synthesis via 4,5-dianünouracil, which has been acylated with corresponding acid derivatives and then cyclized. Some of these compounds are also accessible via 8-haloxanthine derivatives, which are also produced after the grape synthesis with subsequent halogenation.
This synthetic route has several disadvantages. Once a relatively large number of process steps are required, which have a negative effect on both the yields and the capacity of a given plant.
For example, the 4-amino-5-nitroso-uracil produced from 4-aminouracil by nitro- sation must be reduced. This is done on an industrial scale with ammonium sulfide or with iron and hydrochloric acid, with either large amounts of hydrogen sulfide or hydrogen occurring.
In addition, the 4,5-diamino-uracils are very easily oxidized and tend to discolour and form by-products. The discoloration extends into the end products and is difficult to remove. The diamines must therefore always be processed quickly and must never be left in an alkaline environment for a long time. <B> [E.

This is followed by acylation with acids, some of which are difficult to access, and then cyclization. In the manufacture of some xanthine derivatives6 substituted in the 8th position with complex groups, the synthetic route becomes even longer, since the xanthine derivative unsubstituted in the 8th position has to be manufactured, which has to be halogenated and then reacted with a further component.
The xanthine syntheses have been worked on very extensively, on the one hand to produce new derivatives, on the other hand to improve the processes for the production of caffeine, theophylline and theobromine, which are produced in thousands of tons annually. It was therefore to be assumed that all new possibilities of synthesis have been exhausted.
