Abstract
Αποκαλύπτονται νέα ανάλογα καλύπτρας RNA (RNA cap) που περιέχουν μία ή περισσότερες ομάδες φωσφοροθειοϊκών. Τα ανάλογα περιέχουν επίσης τροποποιήσεις στην θέση 2'-Ο 7-μεθυλογουανοσίνης οι οποίες αποτρέπουν την ενσωμάτωση τους με αντίστροφο προσανατολισμό κατά την διάρκεια σύνθεσης mRNA in vitro και γι' αυτό τα ανάλογα είναι "αντι-αντίστροφα ανάλογα καλύπτρας" (ARCAs: anti-reverse cap analogs). Η τροποποίηση ARCA εξασφαλίζει την ακριβή τοποθέτηση του ατόμου S εντός των ενεργών θέσεων πρωτεϊνών δεσμευόμενων με καλύπτρα τόσο στο σύστημα μετάφρασης όσο και στο σύστημα αφαίρεσης καλύπτρας. Τα νέα ανάλογα S-ARCA είναι ανθεκτικά σε ένζυμα αφαίρεσης καλύπτρας in vivo. Μερικά S-ARCAs έχουν υψηλότερη συγγένεια για eIF4E απ' ότι τα αντίστοιχα ανάλογα που δεν περιέχουν ομάδα φωσφοροθειοϊκού. Όταν mRNAs που περιέχουν τα διάφορα S-ARCAs εισάγονται σε καλλιεργημένα κύτταρα, μερικά μεταφράζονται έως και πέντε φορές αποδοτικότερα από mRNAs που συντίθενται με το συμβατικό ανάλογο m7GpppG.
