Abstract
El objetivo de la invención es un proceso para la producción devirus en cultivos celulares con una alta producción y bajos niveles de contaminación por proteínas celulares. El virus es apropiado para ser usado en la preparación de lotes de semilla y vacunas. Como se usa aquí, el término "virus" significa virus inactivado, virus atenuado y virus recombinante, incluyendo virus quimérico. El proceso comprende las etapas de : (a) preparar un cultivo de células que es permisivo al virus y aceptable como un sustrato para producción de vacunas; (b) suspender las células en un medio adecuado seguido por un crecimiento celular en bajas densidades; c) incubar el cultivo de células entre 30 a 40° C durante un período de tiempo apropiado; d) remover el medio desde el cultivo de células de la etapa (c) e inocularlo con virus de semilla; (e) incubar el cultivo celular de la etapa (d) entre 25 a 40° C durante un período de tiempo apropiado; (f) remover el medio seguido por un lavado de las células una o varias veces y reemplazar el medio; (g) incubar el cultivo celular de la etapa (f) entre 25 a 40° C durante un período de tiempo apropiado; (h) recolectar total o parcialmente el sobrenadante del cultivo que contiene el virus con o sin la adición del estabilizador, (i) opcionalmente, llevar a cabo múltiples recolecciones del medio que contiene virus, a cualquier intervalo deseado, por medio de reemplazar el medio removido y reencubar el cultivo durante un período apropiado; (j) opcionalmente, remover los desechos de células y células completas del virus recolectado; (k) opcionalmente, inactivación del virus y; (l) almacenar el virus a -45° C o menos.
