Abstract
本发明涉及一种甲壳素包埋或包胶胶囊,其含有为甲壳素或其盐或其衍生物的甲壳素内容物和包埋或包胶所述甲壳素内容物的食用胶。
Description
技术领域
本发明涉及一种甲壳素包埋或包胶胶囊,其含有为甲壳素或其盐或其衍生物的甲壳素内容物和包埋或包胶所述内容物的食用胶。
背景技术
甲壳素(chitosan)是由(1→4)连结2-乙酰胺-2-脱氧-β-D-吡喃葡萄糖基残基((1→4)-linked 2-acetamido-2-deoxy-β-D-glucopyranosyl residue)所组成的多糖,其是经由使几丁质(chitin)完全或部分去乙酰化而得。
商业上可购得具各种分子量(如10-1,000kDa)的甲壳素产品。商业产品的甲壳素内容物通常具有70%至90%的去乙酰化程度。甲壳素可广泛应用于化妆品、食品、营养补充品和医药。例如,Kanauchi等人指出甲壳素可阻碍脂质吸收(Kanauchi等人,Biosci.Biotechnol.Biochem.,59(5),786-790(1995))。甲壳素几乎无法被消化,且具有溶解和吸附油脂与胆固醇的能力。因此,甲壳素可用以减轻体重和降低胆固醇。
甲壳素在食品和医药上主要是以如片剂或胶囊形式的无水状态加以应用。传统的甲壳素或经修饰的甲壳素具有有限的溶解度,水保留性低而生物利用度有限。美国第6,638,918号专利揭示一种经修饰的甲壳素,其具有较佳的溶解度和生物利用度。
甲壳素在酸性水溶液中溶解时带有正电荷特性,因而可能难以与溶液中其它成份相容而导致稳定性的问题。甲壳素的正电荷也在口中造成强烈味觉干涩味,因而限制含甲壳素食品和医药的形态与应用。美国第6,458,938号揭示一种甲壳素与聚丙二醇的共轭物,其具有改进溶解度以及与其它化合物的相容性。
然而,迄今并无任何现有技术揭示不具有甲壳素涩味的甲壳素产品且仍然保持其吸附油脂及其它可应用的所需功能,或取得所述产品的方法。
发明内容
发明概述
本发明涉及一种甲壳素包埋或包胶胶囊,其含有为甲壳素或其盐或其衍生物的甲壳素内容物和包埋或包胶所述内容物的食用胶。
发明详细说明
本发明惊人地发现,将甲壳素以食用胶包胶或包埋后可解决上述甲壳素问题,即涩味、在水溶液中的低溶解度、低稳定度及与其它化合物的低相容性。
本发明提供一种甲壳素包埋或包胶胶囊,其含有为甲壳素或其盐或其衍生物的甲壳素内容物和包埋或包胶所述内容物的食用胶。
根据本发明,用语“甲壳素包埋胶囊”是指甲壳素内容物与食用胶一起混合或以食用胶包埋所形成的胶囊。“甲壳素包胶胶囊”是指以食用胶包覆或包胶甲壳素内容物所形成的胶囊。
甲壳素内容物
任何形式的甲壳素都可用于本发明。例如,甲壳素可通过将如虾、蟹、墨鱼等甲壳动物、昆虫来源或真菌来源的生物质,经多糖几丁质去乙酰化处理及分离、纯化所得产物后制得。具有不同程度的去乙酰化(游离氨基数目)、纯度、分子量分布和粘度的商业产品也可用于本发明。
根据本发明,甲壳素盐可为由甲壳素与有机或无机酸反应所形成的盐。甲壳素有机酸盐的实例包括但不限于,甲壳素甲酸盐、甲壳素乙酸盐、甲壳素丙酸盐、甲壳素氯乙酸盐、甲壳素羟乙酸盐、甲壳素丁酸盐、甲壳素异丁酸盐、甲壳素丙烯酸盐、甲壳素柠檬酸盐、甲壳素酒石酸盐及其混合物。甲壳素无机酸盐的实例包括但不限于,甲壳素氢氯酸盐、甲壳素氢溴酸盐、甲壳素磷酸盐及其混合物。
适用于本发明的甲壳素衍生物包括但不限于,中或长链的N-烷基或N-烷酰基甲壳素。在此,用语“中链N-烷基或N-烷酰基”表示含有8至13个碳原子的N-烷基或N-烷酰基。在此,用语“长链N-烷基或N-烷酰基”表示含有14至18个碳原子的N-烷基或N-烷酰基。
Deuchi等人指出甲壳素关于油脂消化和吸收的效果随其去乙酰化程度的增加而提高(Deuchi,K.等人,1995,Biosci.Biotechnol.Biochem.,59(5),781-785)。根据本发明的一具体实施例,本发明胶囊的甲壳素内容物具有75%以上的去乙酰化程度,较佳为90%以上,而最佳为95%以上。
根据本发明的另一具体实施例,本发明胶囊的甲壳素内容物中的甲壳素部分分子量为1,000至1,000,000,较佳为30,000至300,000,而最佳为100,000至200,000。
根据本发明的又一具体实施例,本发明胶囊的甲壳素内容物的平均颗粒尺寸小于40至50筛目,较佳为小于60至100筛目,最佳为小于100至150筛目。
根据本发明的又一具体实施例,本发明胶囊的甲壳素内容物的甲壳素部分包括以包埋食用胶溶液的重量计为0.5%至30%,较佳为5%至15%。
食用胶
任何可逆或不可逆的食用胶都可用于本发明以包埋所述甲壳素内容物。适用于本发明的食用胶的实例包括但不限于,刺槐豆胶(locust bean gum)、洋菜胶、海藻酸钠、蒟蒻胶、瓜尔豆胶(guar gum)、阿拉伯胶(gum arabic)、鹿角菜胶(carrageenan)、黄原胶(xanthangum)、果胶(pectin)、黄蓍胶(tragacanth gum)、明胶(gelatin)或其混合物。
因应多价金属离子(如Ca2+)的离子交换作用或酸碱(pH)值或温度的变化,含有糖苷(glycoside)的食用胶会通过交联(cross-linking)反应形成不溶性胶体。根据本发明,酸性状态或含有金属离子形态的食用胶也可用于本发明。此外,与上述食用胶具有相同特性的其他合成或改进材料也可用于本发明。
根据本发明的一具体实施例,所述食用胶较佳为海藻酸钠、瓜尔豆胶、阿拉伯胶、鹿角菜胶、黄原胶、果胶和黄蓍胶或其混合物。更佳地,所述食用胶为海藻酸钠、瓜尔豆胶、阿拉伯胶和鹿角菜胶或其混合物。最佳地,所述食用胶为海藻酸钠。海藻酸钠不具毒性,其是由食用海藻抽出物加工制得,且在食品工业上被广泛应用作为增粘剂和稳定剂。其也为仿生食品的主要成份。此外,海藻酸钠与钙离子反应后形成的胶体钙-海藻酸盐(Ca-alginate)具有极高的热、酸稳定性和肠道溶解性。
根据本发明的一具体实施例,所述食用胶的比例以本发明的胶囊的湿重计为0.5%至30.0%,较佳为1.0%至2.0%。
根据本发明的另一具体实施例,所述甲壳素包埋或包胶胶囊含有海藻酸钠和明胶或蒟蒻胶。所述明胶或蒟蒻胶在被添加的情况下为0.3%至6%(以食用胶重量计)。
其它成份
为增加价值、多样性、不同味觉和口感,本发明的甲壳素包埋或包胶胶囊可依喜好包含促进健康的成份和/或一般食品添加剂,例如,调味剂、甜味料、着色剂,或其他可调整或改善口感的添加剂及其混合物。
任何可促进健康的成份都可与甲壳素一起包埋或包胶于胶囊中。这些成份的实例包括但不限于,红曲、蓝藻、绿藻、绿茶、蓝莓、蘑菇、灵芝、冬虫夏草、樟芝、人参、乳酸菌、双叉杆菌、纤维素及其混合物。所述成份可以提取物干燥粉末、液态或半液态(膏状)的形式添加,添加量可介于0.1-10%(以胶囊湿重计)之间。
用于本发明的调味剂可为任何天然或合成的调味剂,其提供果香、花香或坚果类香料,其包括但不限于苹果、草莓、柠檬、柳橙、葡萄、樱桃、水蜜桃、可可、茶香、玫瑰、茉莉、桂花、杏仁等类的香味。
用于本发明的着色剂可为任何天然或合成的着色剂。适用于本发明的着色剂实例包括但不限于黄槴子、甜菜红、茄红素、紫苏色素、黄色4号和红色6号等类。
用于本发明的甜味剂可为任何天然或合成甜味剂,包括但不限于蔗糖、果糖、葡萄糖、菊糖和阿斯巴甜(Aspartame)等。
甲壳素包埋或包胶胶囊的制备
可使用常规凝胶方法以适合的食用胶将本发明的甲壳素胶囊的甲壳素内容物包埋或包胶。根据本发明的一具体实施例,本发明的胶囊是以混合甲壳素与食用胶,再将所述所得混合物依所述食用胶凝胶的方式加以凝结而制得。例如,如海藻酸钠的具凝胶特性的高分子材料和制造含所述材料胶囊的大规模制备方法是揭示于美国第4,692,284号专利与第5,472,648号专利中。
根据本发明的另一具体实施例,本发明的胶囊是通过用食用胶包胶甲壳素内容物而制得。
甲壳素包埋胶囊的实用性
根据本发明,以食用胶包埋或包胶甲壳素可有效解决存在于液态食品中的甲壳素的涩味。根据本发明,以食用胶包埋或包胶甲壳素也可将甲壳素在液体食品中的量大幅提高至以重量计为20%,而可以不产生令人不快的味觉或不导致甲壳素溶解后的粘度或稳定性问题。本发明的甲壳素-包埋或包胶胶囊可突破基于甲壳素本身特性的应用限制,并可应用在如果冻、布丁和糖果甜点的高含水量食物产品中。本发明的甲壳素-包埋或包胶胶囊可避免甲壳素以粉末或溶液状态添加所造成的如涩味、不良口感、外观和稳定性问题,其也可增加食物产品中甲壳素内容物的含量。此外,本发明的胶囊可包含如促进健康成份、调味剂、着色剂等添加物从而增加含甲壳素产品的价值、变化性、顺服性和方便性。

具体实施方式
实例1甲壳素-海藻酸钠胶囊的制备
本实施例说明本发明的甲壳素包埋胶囊的一般制备方式。
先行配制1%海藻酸钠溶液。在强力搅拌下使海藻酸钠完全溶解后,再加入20克甲壳素粉末(Premix,FG95 ChitoClear),使其与200毫升1%海藻酸钠溶液混合均匀。将所得混合物以内径0.8毫米的注射针头吸入50毫升注射针筒。甲壳素-海藻酸钠混合物胶囊是通过将所述混合物推压出针筒而注入1.5%氯化钙溶液中制得。通过钙离子的离子交换作用而硬化形成平均直径为2.0毫米的胶囊。甲壳素-海藻酸钠胶囊可于氯化钙溶液中放置30分钟,接着以蒸馏水冲洗。所得胶囊即可直接用于饮料或其他食品。或者,所述甲壳素-海藻酸钠胶囊可经干燥脱水后制成干燥状态产品,待使用前再加以复水处理。
实例2红曲-甲壳素胶囊的制备
本实施例说明制造含红曲与甲壳素的胶囊的方法。
胶囊是以近似实施例1描述的方法制得,除20克甲壳素外,另外伴随搅拌将2克红曲干燥粉末加入200毫升1%海藻酸钠溶液中以制得含有甲壳素和红曲的海藻酸钠溶液。以近似实例1描述的方法将所述溶液吸入针筒而后推压滴入1.5%的氯化钙溶液中以硬化成型。在经过冲洗干燥后即制得含有红曲和甲壳素的胶囊。
根据近似于本实施例所描述的方法,其他添加物如绿藻粉、绿茶粉、调味剂、着色剂和甜味料等均可加入本发明胶囊中。添加物的量可视需要做调整。大体而言,选择性添加物的量的范围在海藻酸钠溶液重量的0.1%至10%之间。
实例3甲壳素包埋胶囊的口感品评
如实施例1所制备获得的甲壳素包埋胶囊经口感品评测试比较其与未经包埋甲壳素粉末在口中的涩味感。将未经包埋的甲壳素粉末与甲壳素包埋胶囊加入运动饮料、果汁或酸奶中,将各产品随机提供给五位品评员品评。而有关于涩味感的分数如下:无味-1分,很弱-2分,弱-3分,中-4分,稍强-5分,强-6分,非常强-7分。甲壳素包埋胶囊与未经包埋的甲壳素粉末的平均分数分别为1.4分和6.8分。
所述测试结果显示甲壳素-海藻酸钠胶囊可有效降低甲壳素所产生的涩味感。
Claims (21)
1.一种甲壳素包埋或包胶胶囊,其含有为甲壳素或其盐或其衍生物的甲壳素内容物和包埋所述甲壳素内容物的食用胶。
2.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物具有75%或以上的去乙酰化程度。
3.如权利要求2所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物具有90%或以上的去乙酰化程度。
4.如权利要求3所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物具有95%或以上的去乙酰化程度。
5.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物中甲壳素部分的分子量为1000至1,000,000。
6.如权利要求5所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物中甲壳素部分的分子量为30,000至300,000。
7.如权利要求6所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物中甲壳素部分的分子量为100,000至200,000。
8.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物的平均颗粒尺寸的范围为40至50筛目。
9.如权利要求8所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物的平均颗粒尺寸的范围为60至100筛目。
10.如权利要求9所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物的平均颗粒尺寸的范围为100至150筛目。
11.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物的含量以所述胶囊湿重计为0.5%至30%。
12.如权利要求11所述的甲壳素包埋或包胶胶囊,其中所述甲壳素内容物的含量以所述胶囊湿重计为5%至15%。
13.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素盐是选自下列群组:甲壳素氢氯酸盐、甲壳素氢溴酸盐、甲壳素磷酸盐及其混合物的无机盐,或选自下列群组:甲壳素甲酸盐、甲壳素乙酸盐、甲壳素丙酸盐、甲壳素氯乙酸盐、甲壳素羟乙酸盐、甲壳素丁酸盐、甲壳素异丁酸盐、甲壳素丙烯酸盐、甲壳素柠檬酸盐、甲壳素酒石酸盐及其混合物的有机盐。
14.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述甲壳素衍生物为C8-C18 N-烷基或C8-C18 N-烷酰基甲壳素。
15.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述食用胶是选自下列群组:刺槐豆胶、洋菜胶、海藻酸钠、蒟蒻胶、瓜尔豆胶、阿拉伯胶、鹿角菜胶、黄原胶、果胶、黄蓍胶、明胶或其混合物。
16.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述食用胶为海藻酸钠。
17.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述食用胶的含量以所述胶囊湿重计为0.5%至5.0%。
18.如权利要求18所述的甲壳素包埋或包胶胶囊,其中所述食用胶的含量以所述胶囊湿重计为1.0%至2.0%。
19.如权利要求1所述的甲壳素包埋或包胶胶囊,其中所述食用胶另包括以食用胶重量计为0.3%至6%的明胶或蒟蒻胶。
20.如权利要求1所述的甲壳素包埋或包胶胶囊,其另包括促进健康的成份或食品添加剂。
21.如权利要求1所述的甲壳素包埋或包胶胶囊,其可有效减少甲壳素在口中的涩味。
