背景技术
由于各种内部和外部因素,人类的皮肤随年龄增长表现其衰退的功能。根据皮肤衰老的内部特点,形成身体的组分的活性和生物合成由于细胞的降低活性而显示下降的趋势,并且活性氧类攻击细胞组分,从而导致其在结构和功能方面的破坏作用。根据皮肤衰老的外部特点,如紫外线的围绕皮肤的外因导致皮肤干燥、降低皮肤弹性、以及色素沉积。尤其地,与紫外线相关的强刺激促使皮肤内侧组分的破坏。典型地,活性氧类的增高含量是皮肤内细胞破坏、和促进胶原酶生物合成的原因,从而导致胶原和弹性蛋白的降解。

发明内容
本发明人对研制用于改善皮肤皱纹的新型活性组分已经进行了深入的调查,并已发现,野桐(Mallotus japonicus)提取物对引起皮肤皱纹的弹性蛋白酶的活性和胶原的生物合成施加有效的作用,并且,在改善皮肤皱纹方面,显示临床上极优的功效。

野桐为一种落叶乔木,其属于大戟科,主要生长在韩国的南部。尽管其高度有时达到10米,但是,其典型地显示灌木外貌。野桐的树皮已通常被用作胃溃疡、胃炎和肝虚(anhepatia)的治疗药物。
此处使用的术语“野桐提取物”指从野桐的各种器官和部位(如叶、花、根、茎、枝、树皮和果实);优选从根、茎、枝和树皮;更优选从根、枝和树皮;最优选从树皮获得的物质。
在优选实施例中,使用各种提取溶剂可以获得野桐提取物,如提取溶剂为(a)水;(b)含有1~4个碳原子的无水或含水的低级醇(甲醇、乙醇、丙醇、丁醇等);(c)低级醇和水的混合物;(d)丙酮;(e)乙酸乙酯;(f)氯仿;(g)乙酸丁酯和(h)1,3-丁二醇。用于获得野桐提取物的提取溶剂的适当量为比野桐的干重高1~20(优选1~10)倍。
根据示例方法,如下所示可以获得提取物:用蒸馏水洗涤并干燥的野桐被剪成小块,并且加入1~10倍量的提取溶剂。在用冷凝器防止活性组分挥发的条件下,将浸在溶剂中的野桐在40~100℃下加热3~20小时以提取。可选择地,将浸在溶剂中的野桐在4~40℃下加热1~15天以提取,并且,所得产物在旋转真空蒸发器中彻底干燥。当使用1,3-丁二醇作为提取溶剂时,如上所述,在低温下进行提取,然后,调节提取物的减少的干重至1%(w/v)。
此外,其它常规溶剂可以用于制备野桐提取物,并且与上述溶剂显示相同的效果,对于本领域普通技术人员是显然的。
根据本发明的野桐的提取物包括通过公知的方法进行额外纯化的提取物,以及通过提取获得的提取物。例如,可以理解,本发明的提取物包括使用各种额外的纯化方法获得的活性片段,如使用规定的分子量截取值的超滤和各种色谱法(根据大小、电荷、疏水性和亲和性而设计的用于纯化)。
根据本发明的野桐提取物可以通过如冻干法和喷雾干燥的附加的方法被粉碎。
根据本发明的优选实施例,基于化妆品组合物的总重,化妆品组合物中的野桐提取物的有效量为0.0001~20wt%、更优选0.01~10wt%、再更优选0.1~5wt%、最优选1~3wt%。如果野桐提取物的量低于0.0001wt%,则改善皮肤皱纹的效果可能是微不足道的;当野桐提取物的量超过20wt%时,极可能发生一些如皮肤刺激和制剂不稳定的副作用。
此外,除野桐提取物外,本发明的化妆品组合物可以包含辅剂、以及载体。辅剂的非限制的例子包括防腐剂、抗氧化剂、稳定剂、增溶剂、维生素、色素、香味改良剂、或这些组分的混合物。
本发明的化妆品组合物可以以多种形式被制剂,其非限制的例子包括溶液、悬浮液、乳液、浆糊、软膏、凝胶、乳霜、洗剂、粉末、皂剂、含表面活性剂的清洁剂、油剂、粉底霜、乳液底霜、蜡剂底霜和喷雾剂。详细地,可以以皮肤软化剂(皮肤洗剂)、收敛洗液、营养乳液(乳液)、营养面霜、按摩霜(message cream)、香精、眼霜、清洁霜、清洁泡沫、清洁水和面膜、喷雾剂和粉末的形式提供本发明的化妆品组合物。
本发明的化妆品组合物中包含的化妆品可接受的载体可以根据制剂的类型变化。例如,软膏剂、糊膏剂、面霜剂和凝胶剂可以包含动物和植物脂肪、蜡、石蜡、淀粉、黄芪胶、纤维素衍生物、聚乙二醇、硅酮、膨润土、甲硅烷、滑石粉、氧化锌、或这些组分的混合物。
在粉末或喷雾制剂中,其可以包含乳糖、滑石粉、甲硅烷、氢氧化铝、硅酸钙、聚酰胺粉末和这些组分的混合物。喷雾剂可以额外地包含常规的推进剂,例如,氯氟代烃、丙烷/丁烷或二甲醚。
溶液和乳液制剂可以包含溶剂、增溶剂和乳化剂,如,水、乙醇、异丙醇、碳酸乙酯、乙酸乙酯、苯甲醇、苯甲酸苄酯、丙二醇、1,3-丁二醇、油类,具体地,包含棉籽油、花生油、玉米胚芽油、橄榄油、蓖麻油、和芝麻油、甘油脂肪酸酯、聚乙二醇、山梨聚糖的脂肪酸酯或这些组分的混合物。
悬浮液制剂可以包含液体稀释剂,例如,水、乙醇、丙二醇;悬浮剂,例如,乙氧化异硬脂醇类、聚氧化乙烯山梨醇酯和聚氧化乙烯山梨聚糖酯;微结晶纤维素;偏氢氧化铝(aluminum metahydroxide);膨润土;琼脂和黄芪胶;或这些组分的混合物。
具有表面活性剂的清洁组合物制剂可以包含脂肪醇硫酸酯、脂肪醇醚硫酸酯、磺基琥珀酸单酯、isothinate、咪唑啉衍生物、甲基牛磺酸酯、甘草酸酯(sarcocinate)、脂肪酸酰胺醚硫酸酯、烷胺甜菜碱(alkyl amido betain)、脂肪醇、脂肪酸甘油酯、脂肪酸二乙醇胺、植物油、羊毛脂衍生物、乙氧化甘油脂肪酸酯或这些组分的混合物。
本发明的化妆品组合物具有改善皮肤皱纹的新用途。本发明的组合物显示在分子水平对弹性蛋白酶活性的抑制作用、以及对胶原生物合成的促进作用、以及作为结果的极优的改善皮肤皱纹的功效。在以下所述的实施例中证明该作用和功效。应该理解,皮肤皱纹的改善涵盖皮肤保护的通常应用(如,防止皮肤皱纹、去除皮肤皱纹和防止皮肤衰老)。
具体实施方式
以下特定的实施例旨在解释本发明,而不应理解为限制由附加权利要求所限定的本发明的范围。
实施例1
将200g用蒸馏水洗涤并干燥的野桐树皮放入1.2L水中,在装有冷凝器的提取器中、在70~90℃下加热5小时,通过300目的滤布过滤、并保持在5~10℃下放置7~10天熟化,并且通过Whattman No.5滤纸过滤。过滤物在65℃的旋转真空蒸发器中干燥,得到5.9g的野桐干粉。
实施例2
在15~35℃下,将200g用蒸馏水洗涤并干燥的野桐树皮在1.2L的水中浸泡5天,通过300目的滤布过滤,并且通过Whattman No.5滤纸再过滤,然后在旋转真空干燥器中干燥,以浓缩至2倍的浓度。加入0.6L 100%的乙醇,在5~10℃下保持放置7~10天熟化,并且通过Whattman No.5滤纸过滤。过滤物在65℃的旋转真空蒸发器中干燥,得到5.8g的野桐干粉。
实施例3
在4~40℃下,将200g用蒸馏水洗涤并干燥的野桐树皮在1.2L的水中浸泡5天,通过300目的滤布过滤,在5~10℃下保持放置7~10天熟化,并且,通过Whattman No.5滤纸过滤。过滤物在65℃的旋转真空蒸发器中干燥,得到5.4g的野桐干粉。
实施例4~21
使用表1中所述的提取溶剂,进行与以上实施例3相同的方法,以制备野桐提取物。结果如表1所示。

将200g用蒸馏水洗涤并干燥的野桐树皮放入1.2L的10%乙醇中,在装有冷凝器的提取器中加热5小时,通过300目的滤布过滤、并在5~10℃下保持放置7~10天熟化,并且通过Whattman No.5滤纸过滤。然后,过滤物在65℃的旋转真空蒸发器中干燥,得到6.2g的野桐干粉。

试验实施例1:对弹性蛋白酶活性的抑制作用
如下测试实施例1~32中制备的野桐提取物对弹性蛋白酶活性的抑制作用。
以100μg/ml的量使用各野桐提取物。向60μl的缓冲液(用0.267M HCL调节至pH8.0的0.267M Tris)中加入20μl的底物溶液(8.8mM弹性蛋白酶底物Succ-Ala-Ala-Ala-p-硝基酰替苯胺标准溶液,Sigma),然后,加入100μl的野桐提取物,并且加入20μl的酶溶液(10μg/ml的猪胰腺弹性蛋白酶标准样品,Sigma),以在25℃下反应15分钟。在410nm下测量吸收值。对照组包含代替野桐提取物的蒸馏水。根据以下等式计算抗弹性蛋白酶活性的抑制率:抗弹性蛋白酶活性的抑制率(%)={(测试组的吸收值-对照组的吸收值)/对照组的吸收值}×100

试验实施例2:分析对胶原生物合成的作用
从韩国细胞系研究基金会(Korean Cell Line ResearchFoundation)获得的人正常成纤维细胞被接种于含有DMEM的96-孔微板中,并且在37℃下孵育24小时。孵育后,用含100ppm野桐提取物的无DMEM的新鲜血清代替微板中的培养基,然后,再孵育24小时。对照组不含野桐提取物。孵育后,使用购自Takara Co.(日本)的试剂盒测量由各孔的上清液中获得前胶原型IC-肽(PICP),并且,将测量值转换为ng/ml,其表示新生成的胶原的量。结果总结于表4中。

试验实施例3:本发明的化妆品组合物对改善皮肤皱纹的功效
通过实际使用估测化妆品组合物对改善皮肤皱纹的功效。使用制剂4中所述的含有2%野桐提取物的营养霜,并且用等量的蒸馏水代替野桐提取物,作为该试验的对照。最初,被随机分为两组,每组20位年龄为30~40岁的妇女被给予制剂4的营养霜或其对照霜。在眼缘部位每日早、晚两次施用,坚持2个月。通过与对照组比较观察而评估皮肤皱纹的改善功效。结果总结在以下表12中。

Claims (6)
1、一种用于抑制弹性蛋白酶活性的组合物,其包括作为活性组分的野桐(Mallotus japonicus)提取物。
2、一种用于促进胶原生物合成的组合物,其包含作为活性组分的野桐提取物。
3、一种用于改善皮肤皱纹的化妆品组合物,其包含:(a)作为活性组分的野桐提取物;和(b)化妆品可接受的载体。
4、根据权利要求3的化妆品组合物,其中,所述野桐提取物源自野桐树皮。
5、根据权利要求3的化妆品组合物,其中,基于组合物的总重量,野桐提取物的含量为0.0001~20wt%。
6、根据权利要求3的化妆品组合物,其中,所述组合物为选自溶液、悬浮液、乳液、糊剂、软膏、凝胶、霜剂、洗剂、粉末、皂剂、含表面活性剂的清洁剂、油剂、粉底霜、乳液底霜、蜡剂底霜和喷雾剂之一的形式。
