对多巴胺D2受体和5-羟色胺重摄取 位点有高亲和力的8‘4-‘3-(5-氟-1H-吲哚 -3-基)丙基!-1-哌嗪基!-2-甲基 -2H-1,4-苯并噁嗪-3(4H)-酮甲磺酸盐
本发明涉及式(I)的新颖苯基哌嗪衍生物:

专利申请号PCT/EP00/08090(尚未公布)涉及一群新颖苯基哌嗪衍生物。这群化合物对于多巴胺D2受体及5-羟色胺重摄取位置显示高亲和力。这样的组合对于治疗精神分裂症以及其他精神性疾病十分有用,能够更完全地治疗疾病所有征状(如阳性征状与阴性征状)。
此类化合物显示出多巴胺D2受体拮抗剂的活性,因为它们潜在性地抑制阿朴吗啡在小鼠身上所引发的攀爬行为。该化合物亦显示抑制5-羟色胺重摄取的活性,因为它们加强5-HTP在小鼠身上诱导的行为。
此类化合物在-些对临床上适用的抗精神病药(如条件式的回避反应;Van Der Heyden & Bradford,Behav.Brain Res.,1988,31:61-67)与抗抑郁剂或抗焦虑剂(如抑制压力所引起的发声;van derPoel等,Psychopharmacology,1989,97:147-148)敏感的治疗模型上有活性。
与临床上适用的抗多巴胺D2受体拮抗剂相比,此类化合物有较低的倾向去诱发在齿类动物身上的强直性昏厥,且这样就可能会比现存的抗精神病药剂诱发较少锥体外的副作用。
此类化合物所具有的对5-羟色胺重摄取的抑制活性可能是其在对于抗抑郁剂或抗焦虑剂敏感的行为模式中具有疗效的原因。
此类化合物可用来治疗因多巴胺能系统或5-羟色胺能系统被打乱所引起的中枢神经系统疾病或病变,例如攻击、焦虑症、孤独症、眩晕、抑郁、认知或记忆障碍、帕金森病及特别是精神分裂症与其他精神性疾病。
现在发现上式甲磺酸盐类与其游离碱相比(如EP 99202710.2之第89号化合物),具特别佳的性质。
此甲磺酸盐化合物比其游离碱状态更易溶于水,因此有较好的生物利用率。
此化合物具有手性中心;其外消旋混合物和单个的对映异构体皆属于此发明之范畴。
此化合物可利用例如液体或是固体载体材料等辅助物质,经由适当的方法制成适合给药的形式。
此化合物的游离碱可以按EP 99202710.2中所提方法制备。
游离碱可以根据已知的盐类形成方法转换成甲磺酸盐类。
本发明可利用下列实施例来加以举例说明。
实施例
取2.0克(4.7毫摩尔)的游离碱(可由EP 99202710.2中所提方法制备)(第89号化合物)悬浮于40毫升的甲醇中。将此悬浮液加热至60℃,在约2分钟内加入0.45克(4.7毫摩尔)甲磺酸溶于10毫升甲醇中的溶液。可得一澄清溶液。在60℃下搅拌5分钟后开始结晶。将此溶液在60分钟内慢慢冷却至20℃,并在此温度下搅拌30分钟。进一步在60分钟内冷却到摄氏0度并搅拌90分钟。所得固体物质利用过滤方式加以分离,并以5毫升的甲醇冲洗并在减压、50℃干燥一晚。可得白色甲磺酸盐,产率为2.17克(88%)。
