Abstract

本发明的林蛙抗菌肽凝胶剂及其制备方法属抗菌镇痛药物及其制备领域。其原料和辅料按质量百分比用量为:林蛙抗菌肽10~40%;羧甲基纤维素钠1~5%;甘油5~20%;十二烷基磺酸钠0.1~0.4%;尼泊金乙酯0~1%;余量为水。制备方法是用辅料经高温灭菌制得透明凝胶基质;再添加林蛙抗菌肽。本发明的产品主要用于由铜绿色假单孢菌引起的各类皮肤疾病和烧伤烫伤感染,具有抑菌活性高,毒副作用小,稳定性和耐寒耐热性好;处方无配伍禁忌,给药方便,释药快,与皮肤粘附效果极佳,无刺激性,不污染衣物;不易产生细菌的耐药性等优点。

Description

林蛙抗菌肽凝胶剂及其制备方法
技术领域
本发明属一种抗菌镇痛药物及其制备领域,特别涉及一种林蛙抗菌肽药物和制成林蛙抗菌肽凝胶剂的方法。
背景技术
抗菌肽(Antimicrobial peptides,AMPs)是生物体内部防御系统的一个重要组成部分。抗菌肽具有抗革兰氏阳性菌、革兰氏阴性菌、真菌、包被病毒、原生生物和耐药菌的能力,目前已被广泛应用于食品保鲜和医药领域。
两栖类动物生存环境复杂多样,它们的皮肤在维持它们生存、开拓和适应广阔栖息地中起着重要作用。为了适应生存环境,两栖类动物的皮肤中分布了种类繁多、功能复杂的活性物质,是一个巨大的活性生物分子资源库和药物资源库,1994年Jacob L.在文章中指出某些蛙类皮肤产生的抗菌组分能杀死各种肿瘤细胞,其ED50是LD50的十分之一,使用安全性非常高。
皮肤感染和烧伤烫伤感染多由铜绿假单孢菌(绿脓杆菌)引起的,目前主要采用几种抗生素联合使用来抑制铜绿假单孢菌。从林蛙皮中提取的抗菌肽对铜绿假单孢菌有很好的杀菌抑菌作用,而且对其他革兰氏阳性菌、革兰氏阴性菌和真菌以及临床多重耐药菌也有一定的杀菌抑菌作用。林蛙抗菌肽除杀菌抑菌作用外,还具有一定程度的镇痛作用。
跟本发明相近的背景技术是公开号为CN1216768A的中国专利,该专利公开了林蛙抗菌肽的制备方法。该专利尚未将林蛙抗菌肽制作成药物。
发明内容
本发明要解决的技术问题是,将林蛙抗菌肽制成凝胶剂药品,并给出该药品的制作方法,达到处方无配伍禁忌,给药方便,释药快;制作工艺简便,制剂质量稳定的目的。
本发明的林蛙抗菌肽凝胶剂,其原料和辅料及按质量百分比用量如下:
林蛙抗菌肽为10~40%;
羧甲基纤维素钠为1~5%;
甘油为5~20%;
十二烷基磺酸钠(SDS)为0.1~0.4%;
羟基酯类防腐剂为0~1%;
余量为水。
羧甲基纤维素钠为水性凝胶基质,易于涂布和洗除,无油腻感,并能吸收组织渗出液,不妨碍皮肤正常生理功能,药物释放快;亦可用卡伯姆替代羧甲基纤维素钠做凝胶基质。加入的SDS能乳化皮肤表面脂质,改善药物在角质层分配,可以促进药物的渗透性。而甘油则作为保湿剂,防止凝胶变硬,其他保湿剂如丙二醇、山梨醇等也可用于本制剂的。羟基酯类防腐剂最好使用尼泊金乙酯,也可以用山梨酸等防腐剂代替。余量的水可以是蒸馏水或注射用水。
本发明的林蛙抗菌肽凝胶剂,其具有如下特征。
(1)外观性:淡黄色凝胶;(2)pH值:3.5-5;(3)稳定性:在酸性条件、温度121℃、压强1.01×105Pa下20分钟仍不失活;(4)抑菌活性:采用管碟法进行抗菌活性测定,对临床多重耐药的铜绿色假单胞菌和金黄色葡萄球菌均有抑制作用。
本发明的林蛙抗菌肽凝胶剂通过抑制细菌生长繁殖、控制皮肤或伤口感染抗炎、镇痛,促进创面愈合。非感染伤口可将本发明直接涂抹一层在患处,感染伤口或烧伤需处理后用药,涂抹后在患处形成薄膜,起到对患处的保护作用。
本发明的林蛙抗菌肽凝胶剂的制备方法是,按用量比例称取原料、辅料,再按如下步骤进行配制:
(1)将羧甲基纤维素钠加适量水溶胀,加入甘油研磨润湿;再加入十二烷基磺酸钠(SDS)研磨制得透明凝胶基质;
(2)另取羟基酯类防腐剂加入适量水中,加热至40~50℃使之全部溶解,加入到凝胶基质中;再添加水至足量搅拌均匀,高温灭菌;
(3)将林蛙抗菌肽加入到灭菌后的凝胶基质中,搅拌均匀,即得林蛙抗菌肽凝胶剂。
本发明的林蛙抗菌肽凝胶剂主要用于由铜绿假单孢菌引起的各类皮肤疾病和烧伤烫伤感染有较好的治疗作用,对有其他病菌引起的感染也有一定的疗效。
本发明的有益效果在于,处方工艺简便,制剂质量稳定,处方无配伍禁忌,给药方便,释药快,无油腻性,易涂展,与皮肤粘附效果极佳,涂于皮肤后形成透明薄膜,对皮肤及粘膜无刺激性,能与水溶液混合,可用水冲洗,并能吸收组织渗出液,有利于分泌物的排除,治疗后药物易洗除,不污染衣物。本发明与抗生素类药物相比具有毒副作用小、耐寒耐热性好,不易产生细菌的耐药性及使用方便等优点,易于临床推广。
具体实施方式
本发明以分子量5000~20000的抗菌肽为原料,具体实施步骤如下,但并不意味着对本发明范围的限制。
实施例1林蛙抗菌肽用量对抑菌活性的影响
选用羧甲基纤维素钠10克、甘油64克,十二烷基磺酸钠(SDS)0.8克,尼泊金乙酯2.0克。先将羧甲基纤维素钠加适量注射用水溶胀,加入甘油研磨润湿,再加入SDS研磨制得透明凝胶基质;另取尼泊金乙酯加入适量注射用水中,加热至40-50℃使之全部溶解,加入到凝胶基质中,再添加水至足量搅拌均匀,高温灭菌,制得的凝胶基质分四份待用;将林蛙抗菌肽10克、15克、30克、40克分别加入到四份凝胶基质中,搅拌均匀,配制成林蛙抗菌肽按质量占约10%、15%、30%、40%的林蛙抗菌肽凝胶剂。
采用管碟法进行铜绿假单孢菌抗菌活性测定,结果表明:随着林蛙抗菌肽用量的增加,抗菌活性也在增强;但是用量超过30%抗菌活性的增强已经缓慢。
林蛙抗菌肽凝胶剂的原料、辅料比较好的用量如下:
抗菌肽为20~25%;
羧甲基纤维素钠为2~4%;
甘油为13~17%;
SDS为0.2~0.3%;
尼泊金乙酯为0.1~0.6%;
余量为水。
实施例2
1、林蛙抗菌肽凝胶剂的原料、辅料最佳用量如下:
(1)羧甲基纤维素钠为3%;
(2)甘油为15%;
(3)SDS为0.25%;
(4)尼泊金乙酯为0.2%;
(5)抗菌肽为22%;
(6)余量为注射用水。
2、抑菌圈试验
(1)菌种
铜绿假单孢菌Pseudomonas aeruginosa ATCC27853
大肠埃希氏菌Escherichia coli ATCC25922
金黄色葡萄球菌Staphylocouus aureus ATCC25923
临床多重耐药性的铜绿假单孢菌(主要耐药名称哌拉西林、氧氟沙星、头孢噻肟、新诺明、三代头孢、哌酮)
临床多重耐药性的金黄色葡萄球菌(主要耐药名称青霉素、红霉素、氧氟沙星、甲氧西林)
(2)方法
将上述各菌种培养至对数生长期,用液体培养基稀释105-106CFU/ml,用无菌棉蘸取稀释菌液均匀涂于无菌平皿中的固体培养基上。将无菌牛津杯(10×7.8×6mm)置于平皿中,每个平皿中放四个牛津杯,中心牛津杯为阴性对照,周围三个放林蛙抗菌肽凝胶剂,每个牛津杯中放加入药品量为100μg,盖上陶瓦盖后,置37℃培养16~24小时,用游标卡尺量抑菌圈直径,每一实验重复3~5次。

    3、稳定性试验
将本发明的林蛙抗菌肽凝胶剂分别在室温、37℃、50℃下依次放置3个月、2个月、1个月,每周用上述方法做抑菌试验,菌种为临床多重耐药性的铜绿假单孢菌。

Claims (4)

1、一种林蛙抗菌肽喷凝胶剂,其特征在于,由原料和辅料及按质量百分比的用量组成是,林蛙抗菌肽为10~40%;羧甲基纤维素钠为1~5%;甘油为5~20%;十二烷基磺酸钠为0.1~0.4%;羟基酯类防腐剂为0~1%;余量为水。
2、按照权利要求1所述的林蛙抗菌肽凝胶剂,其特征在于,所说的原料和辅料按质量百分比是,林蛙抗菌肽为20~25%;羧甲基纤维素钠为2~4%;甘油为13~17%;十二烷基磺酸钠为0.2~0.3%;羟基酯类防腐剂为0~0.6%;余量为水。
3、一种权利要求1的林蛙抗菌肽凝胶剂的制备方法,其特征在于,按用量比例称取原料、辅料,再按如下步骤进行配制:
(1)将羧甲基纤维素钠加适量水溶胀,加入甘油研磨润湿;再加入十二烷基磺酸钠,研磨制得透明凝胶基质;
(2)另取羟基酯类防腐剂加入适量水中,加热至40~50℃使之全部溶解,加入到凝胶基质中;再添加水至足量搅拌均匀,高温灭菌;
(3)将林蛙抗菌肽加入到灭菌后的凝胶基质中,搅拌均匀,即得林蛙抗菌肽凝胶剂。
4、一种权利要求1的林蛙抗菌肽凝胶剂的应用,其特征在于,用于由铜绿假单孢菌引起的皮肤疾病和烧伤烫伤感染。
