Description

血管生成对于实体肿瘤的生长是非常重要的。没有血管生成,限制了氧和其他营养物可以传送的距离,结果肿瘤生长就被限制在微小的尺寸。实体肿瘤可以产生血管生成因子,如血管内皮生长因子(VEGF)和碱性成纤维细胞生长因子(bFGF),这些已经在早期的开发中指出。VEGF被认为在肿瘤新血管形成的引发中起着非常重要的作用,同时碱性成纤维细胞生长因子(bFGF)被认为在保持血管生成响应中是非常重要的。这是一个明确的证据,即实际上是在血管生成启动的激活之后,靶向血管生成可以进一步降低肿瘤生长。进一步和不曾预料的肿瘤血管生成抑制的抑制作用是宿主肿瘤免疫响应的再激活,该响应作为非-血管生成内皮更容易被刺激以增加其对淋巴细胞的粘合性,增加淋巴细胞肿瘤的渗透。

2-甲氧基雌二醇

D’Amato(WO95/04535)公开了2-甲氧基雌二醇和某些类似物在治疗与血管生成有关的疾病中的用途。Stewart(PCT/AU97/00286)公开了治疗以气管炎症为特征的疾病的2-甲氧基雌二醇和类似物。但是,2-甲氧基雌二醇在实际使用中遇到了明显的限制:在治疗范围内具有剂量-限制毒性。在水溶液中具有高的不溶性并因此难以服用。
本发明的目的是克服或至少减轻一个或多个现有技术的困难或不足。
一方面,本发明提供雌二醇化合物的轭合前药,优选地是与生物活性修饰剂共轭的2-甲氧基雌二醇或功能类似物或其衍生物。
优选地,生物活性修饰剂也具有作为掩蔽和/或寻靶剂的功能。
生物活性修饰优选是碳水化合物或肽部分。
“功能活性”是指2-甲氧基雌二醇的类似物或衍生物具有一个或多个2-甲氧基雌二醇的生物活性。2-甲氧基雌二醇的生物活性包括,但不限于:内皮细胞增生的抑制;平滑肌细胞增生的抑制;肿瘤细胞增生的抑制;微管功能的抑制;白细胞激活的抑制。这些功能活性类似物或衍生物的实例包括2-乙氧基雌二醇,2-羟基雌二醇和其他在2位修饰的类似物,2-甲氧基雌二醇-3甲醚,4-甲氧基雌二醇,和其他B环扩环为7-员环的类似物。也可参见WO95/04535,其全部公开插入此作为参考。
优选地,轭合前药包括与糖部分共轭的2-甲氧基雌二醇或其功能活性类似物或衍生物,优选的糖部分是作为在所治疗的疾病中可能存在的,或以增加的水平出现的酶的底物。这样,化合物具有中央控制(servo-controlled)的生物利用度,其中酶水平是药物(雌二醇化合物例如2-甲氧基雌二醇或其功能活性类似物或衍生物)从前药中释放速率的决定性因素。这样就可以使药物释放局部化,例如在实体瘤部位。
更优选地,糖部分是5-员或6-员糖。具体优选的糖部分是葡萄糖醛酸化物,其是葡萄糖醛酸酶的底物,该酶是与肿瘤相关的酶,但也可能出现在炎症响应中。其它特别优选的糖部分是作为半乳糖甙酶底物的半乳糖甙。肿瘤和炎症疾病与β-半乳糖甙酶和其它溶酶体水解酶的高度表达有关。可以用与疾病的范围和活性成比例的速度将前药转化为活性剂。该新药的靶向机制导致系统毒性的降低。雌二醇化合物例如2-甲氧基雌二醇或其类似物的葡萄糖醛酸化物或半乳糖甙轭合物的用途也可以给化合物带来水溶性并因此增加制备所需给药途径的任何制剂的容易程度。

将雌二醇化合物例如2-甲氧基雌二醇或其功能活性类似物或衍生物结合到疾病-依赖的活性前药中可以显著地改善该抗癌和抗炎剂的效力和选择性。疾病活性的两个特征被前药承载基,例如直接或通过一个连接部分与母体分子连接的半乳糖甙基利用。β-半乳糖甙酶在衰老的内皮细胞和巨噬细胞中是不可调节的,这不同于单核细胞。内皮衰老在活跃的血管生成(已经经过了多个数量倍增的局部内皮细胞)面积中是可以预料的并且在此具有显著的巨噬细胞渗透。这两个条件在风湿性关节炎中的慢性炎症和实体瘤的发展中均满足。内皮细胞增生到新血管瘤将显示更高的衰老细胞比例,这将表达高的与衰老有关的β-半乳糖甙酶。该表达,除由白血球水解酶导致之外,还用于2-甲氧基雌二醇-17β吡喃半乳糖甙的局部转化。已知巨噬细胞在溶酶体β-半乳糖甙酶在由细胞分裂和巨噬细胞刺激引起的进一步分化时获得高度表达。因此,巨噬细胞依赖的前药激活也可以出现。2-甲氧基雌二醇本身导致β-半乳糖甙酶在内皮细胞中表达的增加,因此2-甲氧基-3,17β-雌二醇-β-D-吡喃半乳糖甙用内皮细胞保温会在前药的激活中产生阳性反应。
糖蛋白和肽部分与雌二醇化合物可以在任何适宜的位置轭合。优选地,糖,蛋白质和肽部分与雌二醇化合物例如2-甲氧基雌二醇或其类似物在3和/或17位轭合。
在本发明这一方面特别优选的形式中,轭合前药是17β吡喃半乳糖甙-2-甲氧基雌二醇或其功能性类似物或衍生物。现已发现2-甲氧基雌二醇的半乳糖甙轭合物比母体化合物2-甲氧基雌二醇具有改进的水溶性和低的细胞毒性。
糖可以直接与雌二醇化合物如2-甲氧基雌二醇或其类似物轭合,或者两者可以通过可裂解的连接轭合。可裂解的连接可以是任意适宜的类型,例如碳酸酯或氨基甲酸酯。本领域技术人员可以容易地确定适宜的连接。例如使用连接有利于前药在血浆中的稳定。

典型的单位剂量形式包括1mg至500mg本发明化合物。
本发明方法可以用于预防,减轻或治疗炎症疾病,具体的疾病是以慢性或急性气管炎为特征的,例如哮喘,或慢性炎症疾病,例如风湿性关节炎。本发明方法也可以用于治疗任何与增强的血管生成或加速的细胞分裂有关的疾病,特别是癌症,例如实体瘤。

产物可以通过对本领域普通技术人员显而易见的适宜技术分离。过滤,闪式色谱和制备HPLC可以认为是适宜的。
本发明参考下列实施例进行更全面的说明。但是,可以理解,下列描述只是用于举例说明并不以任何方式限制上述的本发明概念。
实施例1:2-甲氧基-17β-雌二醇-17-D-吡喃葡萄糖醛酸(见图1)
β-雌二醇二苄基醚(2)
氮气下将β-雌二醇(20.00g,73.4mmol)溶解在300mL无水DMF中冷却至0℃。连续搅拌下分批加入氢化钠(10.4g,260mmol)。在0℃继续搅拌15分钟并同时滴加苄基溴(40.0mL,336mmol)然后加入碘化四丁基铵(2.0g,5.4mmol)。然后将反应液在室温继续搅拌72小时。将反应混合物冷却至0℃并用100mL50%的乙醇水溶液淬灭过量的氢化钠。将反应混合物用3M的HCl溶液酸化。然后将全部的反应混合物真空蒸发至约300mL体积并在饱和氯化钠溶液(200mL)存在下用3×150ml的乙酸乙酯进行萃取。将合并的萃取液用水(200mL),饱和氯化钠溶液(200mL)洗涤,用无水硫酸钠干燥并真空蒸发得到粘稠有机油状物。用石油醚研磨得到奶白色固体(26.3g,收率76.8%)。产物的TLC分析显示为一个点。
2-溴-β-雌二醇二苄基醚(3)
在氮气下将搅拌的β-雌二醇二苄基醚(18.60g,41.1mmol)在120mL无水THF/180mL乙酸中的溶液冷却至-10℃以下。在30分钟内和保证温度在-10℃以下向其中滴加溴(2.53mL,49.4mmol)。在-10℃继续搅拌30分钟然后将反应加热至室温并继续搅拌2.5小时。将反应混合物倾倒入冰/水混合物(400mL)中并用3×200mL的二氯甲烷进行萃取,然后将合并的有机层用水(200mL),饱和碳酸氢钠溶液(200mL),10%硫代硫酸钠溶液(200mL)和水(200mL)洗涤,并用无水硫酸钠干燥。将蒸发后得到结块(sluggy)奶白色固体用乙酸乙酯重结晶得到奶白色结晶固体(13.21g,60.5%)。TLC分析显示为一个点。
2-羟基-β-雌二醇二苄基醚(4)
在氮气下将搅拌的2-溴-β-雌二醇二苄基醚(10.00g,18.8mmol)在200mL无水THF中的溶液冷却至-78℃,并在保证反应温度保持在-70℃以下向其中滴加1.7M叔丁基锂的戊烷(24.35mL,41.4mmol)溶液。将反应混合物在该温度继续搅拌15分钟并在此时滴加MoOPH试剂(9.08g,20.7mmol)在无水THF中的悬浮液并保证反应温度在-70℃以下,在此温度下继续搅拌1小时,在将反应加热至0℃之前并进一步搅拌1小时。将反应混合物通过滴加水淬灭,然后通过3×150mL二氯甲烷萃取。将合并的有机层用50%水/50%饱和氯化钠溶液(150mL),饱和碳酸氢钠(150mL)溶液和饱和氯化钠溶液(150mL)洗涤,然后用无水硫酸钠干燥。蒸发后得到的粗品为深绿色粘稠油状物。TLC分析显示为不纯的产品,将其通过闪式色谱纯化得到5.56g(63.2%)纯品。
2-甲氧基-β-雌二醇二苄基醚(5)
在氮气下将无水碳酸钾(12.03g,87.0mmol)加入搅拌的2-羟基-β-雌二醇二苄基醚(4.08g,8.7mmol)的无水DMF(250mL)溶液中。将反应混合物室温搅拌15分钟后滴加碘甲烷(8.13g,130.5mmol),然后加入碘化四丁基铵(0.48g,1.3mmol)作为相转移催化剂。室温下继续搅拌90小时,然后在50%水/50%饱和氯化钠溶液(300mL)存在下用乙酸乙酯萃取(3×200mL)。将合并的萃取液用水(200mL),1M氢氧化钠溶液(2×200mL)和饱和氯化钠溶液(200mL)洗涤,然后用无水硫酸钠干燥。蒸发后得到的浅黄色油状物(3.93g,94.3%)静置后固化。TLC分析显示仅为单一成分。
2-甲氧基-β-雌二醇(6)
将2-甲氧基-β-雌二醇二苄基醚(1.41g,2.9mmol)溶解在乙酸乙酯(30mL)中并用1MHCl的乙醚溶液(3mL)酸化。加入10%的钯炭(777mg,0.73mmol)并将反应置于帕尔氢化仪中并且在开始的氢气压力65psi下氢化48小时。通过塞里塑料垫过滤除去催化剂并用二氯甲烷(3×30mL)洗涤。将滤液蒸发后得到浅黄色油状物,静置后固化。TLC分析显示为两个产物,通过闪式色谱纯化(乙酸乙酯∶石油醚3∶7v/v和1∶1v/v)得到产物(6)为奶白色固体(560mg,63.8%)和乙酸酯副产物(7)(230mg,22.9%)为白色固体。
另一部分(6)从2-甲氧基-17β-乙酰氧基-雌二醇(7)的水解得到。将2-甲氧基-17β-乙酰氧基-雌二醇(7)(1.39g,4.0mmol)溶解在THF(25mL)中并在氮气下搅拌,向其中滴加0.4M氢氧化锂(22.2mL,8.9mmol)水溶液并在室温连续搅拌23小时。通过TLC可知此时出现了将少量乙酸酯产物,加入另一部分氢氧化锂(5.0mL,2.0mmol)溶液。将反应混合物在室温再连续搅拌1小时,然后用1MHCl溶液将其酸化至pH3。用3×25mL的二氯甲烷萃取后将合并的萃取液用水(25mL)和饱和氯化钠溶液(25mL)洗涤。用无水硫酸钠干燥并蒸发得到黄色结晶固体(1.20g,98.3%)。产物通过闪式色谱分离(乙酸乙酯∶石油醚3∶7v/v和1∶1v/v)得到产物(6),为奶白色固体(921mg,75.5%)和乙酸酯起始原料(7)(110mg,7.9%)为白色固体。

在机械搅拌下通过迪安-斯达克装置保证反应的完全干燥,将2-甲氧基-β-雌二醇(500mg,1.65mmol)的甲苯(25mL)溶液和碳酸钙(177mg,4.95mmol)回流至大约一半的体积。在其中滴加1α-溴-2,3,4-三-O-乙酰基葡萄糖醛酸酯(397mg,4.95mmol)的无水甲苯(25mL)溶液,通过迪安-斯达克气水阀连续收集直至反应混合物体积的为约25mL。继续回流总共5小时,通过塞里塑料过滤并用甲苯洗涤除去催化剂。蒸发得到1.06g米色泡沫,将其开始用闪式色谱然后用制备TLC纯化得到282mg(27.6%)产物。
2-甲氧基-17β-雌二醇-17-D-吡喃葡萄糖醛酸(9)
搅拌下在9(245mg,0.40mmol)在15mL绝对乙醇中的溶液中加入5M氢氧化钠溶液(0.5mL)。将反应液室温搅拌1.5小时,然后用0.2M硫酸调节pH至3。蒸发后将少量粗混合物样品(12mg)通过制备HPLC纯化得到7mg白色洗净的固体结晶产物(9)。
实施例2:2-甲氧基-3,17β-雌二醇-β-D-吡喃半乳糖甙
开始将3,17β-雌二醇进行双保护为3,17β-二苄基醚,是通过用氢化钠去质子并随后与苄基溴在DMF中在碘化四丁基铵存在下反应而进行的。处理后以高收率得到双保护产物并不需要进一步纯化。3,17β-雌二醇二苄基醚在2-位进行溴化,在THF/乙酸混合物中进行,具有高的区域选择性(>80%)。用乙酸乙酯重结晶以高收率得到纯的2-溴-3,17β-雌二醇二苄基醚,然后在低温下用叔丁基锂进行卤素-锂交换。将锂中间体与MoOPH试剂(五氧化钼配合物)反应,然后进行还原处理高收率地得到2-羟基-3,17β-雌二醇二苄基醚。在此时需要色谱纯化,但相对简单的是给出产物杂质信息。
定量的甲基化可以通过将酚用碳酸钾去质子然后在DMF中用碘甲烷烷基化而进行。2-甲氧基二苄基醚保护的甾醇的氢解可以在正氢压力下在10%钯炭存在下进行。所得到的产物2-甲氧基3,17β-雌二醇为白色或浅黄色固体。该方法结合了Cushman等1997的相似合成方法,但是是具有改进的收率和更直接和更可靠的2-羟基化方法。
小心选择氢化的条件,允许选择性地形成各种不同的产物。可以生产2-甲氧基雌二醇的17β-单苄基醚,也可以生产17β-乙酸酯。根据随后的反应条件,和所需的羟基保护程度,这是最有利的。2-甲氧基雌二醇的3-羟基功能基的保护可以选择性地通过在温和的反应条件下形成乙酸酯而进行,或通过标准方法再用苄基保护为3-单苄基醚。
通过改进的Koniga-Knorr法,将这些化合物与2,3,4,6-四-O-乙酰基-α-吡喃半乳糖基溴在碳酸钙存在下缩合,随后进行相应的3-(β-D)或17(β-D)糖的脱保护过程(Blackwell等1993)。2,3,4,6-四-O-乙酰基-α-吡喃半乳糖基溴也可以用于该反应步骤中得到相应的3或17β-(β-吡喃葡萄糖基)取代的甾醇(Paulsen,1982)。
实施例3
各种2-甲氧基雌二醇(1)类似物和衍生物(见图2)的合成
3-乙酰基2-甲氧基雌二醇(2)
在氮气氛下将2-甲氧基雌二醇(105mg,0.347mmol)溶解在无水吡啶(6.0mL)中并冷却至-40℃(乙腈/CO2浴),在10分钟内滴加乙酸酐(3.0mL)并将反应在低温搅拌165分钟,加入50mL1M的HCl并使反应液至室温。用乙酸乙酯(3×50mL)萃取然后将合并的有机萃取液随后用3MHCl水溶液,水(50mL)和饱和NaCl水溶液(50mL)洗涤。将有机层用硫酸钠干燥然后过滤并将溶剂真空蒸发得到黏稠油状物。用闪式色谱纯化(1∶1乙酸乙酯/石油醚)得到标题化合物为白色固体(87mg,71%)。

葡萄糖醛酸甲酯(3)
在氮气氛下将新制备的碳酸钙(73.7mg,0.428mmol)加入3-乙酰基-2-甲氧基雌二醇(2)(36.8mg,0.107mmol)的无水甲苯(40mL)溶液中。将悬浮液加热回流直至在迪安-斯达克气水阀中收集的溶剂体积达到20mL。此时,将1-溴-1-去氧-2,3,4-三-O-乙酰基-a-D-葡萄糖醛酸甲酯(127.4mg,0.321mmol)的甲苯(20mL)溶液以与连续蒸馏到迪安-斯达克气水阀中相同的速度滴加到反应液中。反应60分钟后,将另一部分1-溴-1-去氧-2,3,4-三-O-乙酰基-a-D-葡萄糖醛酸甲酯(84.8mg,0.214mmol)加入甲苯(10mL)中并进一步蒸馏10mL溶剂。然后将反应液回流27小时之后冷却至室温。通过塞里塑料垫过滤除去固体并用甲苯彻底洗涤。将合并的滤液和洗涤液连续用1M的HCl水溶液(30mL),水(30mL)和饱和NaCl水溶液(30mL)洗涤。将有机层用硫酸钠干燥然后过滤并蒸发至干,得到133mg黄褐色油状物。用闪式色谱部分纯化(SiO2,0.5∶5∶94.5,三乙基胺/乙醚/二氯甲烷)得到略微不纯的标题化合物(67.0mg)。

6-氧-2-甲氧基雌二醇(8)
在氮气氛和搅拌下将无水碳酸钾(20mg,0.145mmol)加入3,17-二乙酰基-6-氧-2-甲氧基雌二醇(7)(34mg,0.085mmol)的8.0mL甲醇溶液中。加入水(3.0mL)并将反应液室温搅拌16小时。此时,将反应液的pH通过滴加1M的HCl水溶液调节至pH4。用乙酸乙酯(3×10mL)萃取然后将合并的萃取液用水(10mL)和饱和NaCl水溶液(10mL)洗涤,然后用硫酸钠干燥并过滤。真空蒸发溶剂得到奶白色固体,通过硅胶闪式色谱纯化(3∶2乙酸乙酯∶石油醚)得到26.4mg(98.3%)为白色结晶固体的标题化合物。

实施例4
2-甲氧基-雌二醇-17β-D-葡萄糖醛酸化物对由葡萄糖醛酸酶代谢的敏感性
将2-MEG用纯的大肠杆菌衍生的β-葡萄糖醛酸酶在台罗德式缓冲液中保温16小时(大鼠肾匀浆5.5小时)。在该保温结束后,通过将100μl反应混合物用200μl乙腈(ACN)然后通过加入5μl2M的HCl进行萃取,在1500g(室温)进行涡流和离心。然后将酶样品保温液或质量对照保温液在至少10分钟内急冷至-20℃,然后除去120μl上层(ACN)相并将其加入120μl蒸馏水中。将100μl该萃取液注射入Hewlett Packard Series 1050HPLC的C18分析柱中(Alltech Hypersil ODS 3μM;4.6×7.6mm),代谢物用Hewlett Packard 1046A荧光检测仪检测(激发270nm,发射,320nm),代谢物在40∶60的CAN:缓冲液的流动相中分离(缓冲液包含20mMNaH2PO4,0.02%磷酸,pH3.1-3.2),流速0.7ml/min最初的4分钟为线性梯度在4.5分钟为ACN:缓冲液(76∶24)的流动相,保持分离的剩余时间(10分钟)。
成功地定量回收质量对照的标准品。2-MEG的剩余时间为4.35min且2-MEO的剩余时间为9.8min。2-MEO的荧光约5倍于2-MEG(图3)。这证明2-MEG到2-MEO的可评估的转化用大肠杆菌β-葡萄糖醛酸酶(~20%转化)和大鼠肾匀浆(~6%)分别出现在16小时和5.5小时的保温期间。
实施例5
2-甲氧基-雌二醇-17β-D-葡萄糖醛酸化物对气管间质细胞数量和DNA合成的调节
人气管成纤维细胞是从Melbourne Epidemiological Survey of ChidhoodAsthma(MESCA)(Oswald等,1994)跟踪调查的自愿者中得到的,这些自愿者是开始在1965年7岁时招募的并且现在已经42岁了。将提供约2mm3的上皮和粘膜组织样品的自愿者进行支气管活体组织检查。将这些样品解剖除去上皮并置于塑料培养盘中。并根据上述方法(Zhang等,1996)通过外植体培养6周以上使成纤维细胞成长。将细胞在6个细胞盘中以亚融合密度接种并在72小时后通过除去血清而终止。除去血清24小时后,将细胞暴露在凝血酶(0.3U/ml)和不含血清而含有胰岛素,转铁球蛋白和硒的培养基中。通过用胰蛋白酶(0.5%w/v)保温10分钟后收集细胞。或直至制备单个细胞悬浮液。用血细胞计数器测定细胞的数量。
在5%FCS和2-MEO(10微M)存在下将气管平滑肌细胞在培养基中保持72小时快速地进行可识别的形状变化(图4),而细胞在相同的保温期间和在相同的MEG浓度下却未检测到该变化,说明MEG缺乏母体分子的这种活性。
将从肺移植接受者中切除的气管中获得平滑肌细胞并按上述方法培养(Fernandes等1999)。按照与人气管成纤维细胞中所述相同设计的实验测定细胞的数量,但是增加1%(v/v)胎牛血清(FCS)刺激的评价。此外,在加入凝血酶后24和28小时之间测定[3H]-胸腺嘧啶核苷结合以确定DNA的合成(Femandes等1999)。
在人气管成纤维细胞凝血酶-刺激增加细胞数量中,减少到2-MEO对照的响应的12±8%,但是2-MEG没有明显的作用(表1)。在人气管平滑肌响应中,凝血酶和FCS的降低程度均是2-MEO大于2-MEG。此外,2-MEO使凝血酶刺激的DNA合成降低,但是未受2-MEG的影响(表1)。
将A549细胞保持在含有5%胎牛血清的DMEM中并进行不同步的增殖。通过在加入2-甲氧基雌二醇,6-氧-2-甲氧基雌二醇和2-甲氧基-雌二醇-17β-D-葡萄糖醛酸化物后24小时后加入[3H]-胸腺嘧啶核苷测定DNA的合成。2-甲氧基雌二醇和6-氧-2-甲氧基雌二醇均在10微摩尔显著地减少DNA的合成,而2-甲氧基-雌二醇-17β-D-葡萄糖醛酸化物则没有作用(图5)。
这些数据显示2-MEG与母体分子2-MEO相比活性低或没有活性。
实施例6
大鼠子宫胞质雌激素受体结合
大鼠子宫胞质是标准的雌激素受体制剂(Markavevich等19xx)。从4个Sprague Dawley大鼠采集子宫,在10ml/g湿重的缓冲液(pH7.4,0.002M二硫苏糖醇,0.25M蔗糖)中通过3×10s爆发在超Turrax匀浆器上匀浆。将所得匀浆在4℃以1000xg离心5分钟以除去核和细胞碎片。将上清液用BeckmanJA离心机在4℃以35,000xg离心30分钟。将上清液用于结合试验并用相同的缓冲液进行。检测0.2nM[3H]-雌二醇(150Ci/mmol)的置换,通过200μl子宫胞质用50μl放射活性的雌二醇和50μl含有在0.003nM至10nM范围的雌二醇(没有放射标记),2-MEO在0.003至10μM,2-MEG在0.003至10μM的缓冲液保温而进行。测定在保温中的总结合,其中加入缓冲液以替代转移剂和10μM雌二醇用于定义非特异性结合。
每个替换产生置换但在10μM2-MEO有一个不完全响应,其中logIC50值表示2-MEG对雌激素受体亲和性只有2-MEO的1/50(图6)。这证明在降低间质细胞增殖具有约100nM的IC50的浓度范围内,2-MEO对雌激素受体具有显著的亲和性,尽管其亲和性比其前体雌二醇(IC50为3nM)低很多
实施例7
2-MEO和MEG的相对溶解性
辛醇:水分配系数是衡量化合物相对于水和脂的溶解性的尺度。对于2-MEO,该系数是在大量混合以保证获得平衡的水溶液中通过在Ex=250nm,Em=325nm的荧光光谱测定的。用Ex=240nm,Em=352nm的荧光光谱测定的辛醇/水分配系数对于2-甲氧基雌二醇是9.05(logP=0.957),对于MEG是0.372(logP=-0.429)。因此,葡萄糖醛酸化物的水溶性约是2-MEO本身的25倍。

12.根据权利要求11的化合物,其中R1至R4分别选自羟基,C1-6烷氧基,亚氨基,C3-8亚氨基环烷基,酮,亚肼基,氨基甲酸酯,或糖,蛋白质或肽残基。
13.根据权利要求10的化合物,
其中R1是一个或多个羟基,烷氧基,酰氧基或碳水化合物,蛋白质或肽残基;
R3包括烷基;且
R4是一个或多个羟基,烷氧基,O-保护基或碳水化合物,蛋白质或肽残基。
14.根据权利要求13的化合物:
其中R1包括甲氧基。
15.根据权利要求14的化合物,其中:
R3包括一个或多个H,酮,或亚肼基。
16.根据权利要求11的化合物,其中:
R1,R2或R4包括具有结构式II的糖残基:其中Z是空格或者是一个键;
m是0或1;且
R5至R12各自独立地选自H,OH,氨基,卤素,COOR13,CH2OR13,CHOH-COOR13,PO3H2,CH2-PO3H2或CHOH-PO3H224,CH2COOR13,OCOR13;
其中R13是H或任选取代的C1至C6烷基,C1-6链烯基,C3-8环烷基,C1-6链烷酰基,C1-6烷氧基,芳基,氨基,酰基,C1-6酰氧基氨基烷基,亚氨基,C1-6亚氨基烷基,酮,亚肼基,或羧基。
18.根据权利要求10的轭合前药,其中肽残基具有结构式III:
-Z-(ABC)m(DF)n(GHJK)pIII其中Z是空格或者是一个键
m,n和p分别是0,1,2的整数
ABCDFGHJK可以相同或不同,分别为氨基酸。
19.根据权利要求18的轭合前药,其中肽是,或包括序列-Ala-Phe-Lys或-Val-Leu-Lys。
20.根据权利要求10的化合物,选自:
3-乙酰基2-甲氧基雌二醇
3-乙酰基6-氧-2-甲氧基雌二醇
3,17-二乙酰基-2-甲氧基雌二醇
17-乙酰基-2-甲氧基雌二醇
3,17-二乙酰基-6-氧-2-甲氧基雌二醇
3,17-二乙酰基-2-甲氧基雌二醇-6-腙
2-甲氧基雌二醇-6-腙
2-甲氧基雌二醇-17β-单二烯醚
2-甲氧基雌二醇-17β-乙酸酯
2-甲氧基-17β-雌二醇-17-D-三乙酰基吡喃葡萄糖醛酸(glucopyranosiduronic acid)甲酯
2-甲氧基-17β-雌二醇-17-D-吡喃葡萄糖醛酸2,3,17β-雌二醇-17-β-D-吡喃半乳糖甙-2-甲醚,或2,3,17β-雌二醇-3-β-D-吡喃半乳糖甙-2-甲醚。
各R1,R2,R3和R4是独立地选自一个或多个下列的取代基:氢,羟基,卤素,任选取代的C1-6烷基,C1-6链烯基,C1-6炔基,C3-8环烷基,C1-6链烷酰基,C1-6烷氧基,芳基,氨基,酰基,C1-6酰氧基,氨基烷基,亚胺基,C1-6亚胺基烷基,C1-C3亚胺基环烷基,酮,醛,亚肼基,肟基,羟亚氨基,C1-6烷氧基亚胺基,C3-8环烷氧基亚胺基或羧基,或O-保护基,或碳水化合物,肽或蛋白质残基,或其衍生物或药用盐或体内可水解酯,酰胺碳酸酯,或其氨基甲酸酯,
条件是至少有一个R1至R4存在并且包括碳水化合物,肽或蛋白质残基。
22.根据权利要求21的轭合前药:
其中
R1是一个或多个羟基,烷氧基,酰氧基或碳水化合物,蛋白质或肽残基;
R3包括烷基;和
R4是一个或多个羟基,烷氧基,O-保护基或碳水化合物,蛋白质或肽残基。
23.根据权利要求22的轭合前药,其中R1包括甲氧基。
25.根据权利要求21的轭合前药,其中肽残基具有结构式III:
-Z-(ABC)m(DF)n(GHJK)pIII其中Z是空格或者是一个键
m,n和p分别是0,1,2的整数
ABCDFGHJK可以相同或不同,分别为氨基酸。
26.药物组合物,包括:
与生物活性修饰剂共轭的雌二醇化合物的轭合前药;和
其药用载体,稀释剂或赋形剂。
27.根据权利要求26的药物组合物,其中雌二醇化合物是2-甲氧基雌二醇或其功能活性类似物或衍生物。
各R1,R2,R3和R4是独立地选自一个或多个下列的取代基:氢,羟基,卤素,任选取代的C1-6烷基,C1-6链烯基,C1-6炔基,C3-8环烷基,C1-6链烷酰基,C1-6烷氧基,芳基,氨基,酰基,C1-6酰氧基,氨基烷基,亚胺基,C1-6亚胺基烷基,C1-C3亚胺基环烷基,酮,醛,亚肼基,肟基,羟亚氨基,C1-6烷氧基亚胺基,C3-8环烷氧基亚胺基或羧基,或O-保护基,或碳水化合物,肽或蛋白质残基,或其衍生物或药用盐或体内可水解酯,酰胺碳酸酯,或其氨基甲酸酯,
条件是至少有一个R1至R4存在并且包括碳水化合物,肽或蛋白质残基。
29.预防或治疗与血管生成或细胞分裂数量加速或增加,或炎症有关的疾病的方法,所述方法包括给需要这种预防或治疗的患者服用有效量的与生物活性修饰剂共轭的雌二醇化合物的轭合前药。
30.根据权利要求29的方法,其中雌二醇化合物是2-甲氧基雌二醇或其功能活性类似物或衍生物。
31.根据权利要求30的方法,其中轭合前药具有下列结构式:其中m,n,p和q分别为0,1,2或3的整数,
各R1,R2,R3和R4是独立地选自一个或多个下列的取代基:氢,羟基,卤素,任选取代的C1-6烷基,C1-6链烯基,C1-6炔基,C3-8环烷基,C1-6链烷酰基,C1-6烷氧基,芳基,氨基,酰基,C1-6酰氧基,氨基烷基,亚胺基,C1-6亚胺基烷基,C1-C3亚胺基环烷基,酮,醛,亚肼基,肟基,羟亚氨基,C1-6烷氧基亚胺基,C3-8环烷氧基亚胺基或羧基,或O-保护基,或碳水化合物,肽或蛋白质残基,或其衍生物或药用盐或体内可水解酯,酰胺碳酸酯,或其氨基甲酸酯,
条件是至少有一个R1至R4存在并且包括碳水化合物,肽或蛋白质残基。
32.制备式I化合物的方法,其中该方法包括:
L-Z-(ABC)m(DF)n(GHJK)p VIII
其中除式IV外,R1,R2,R3,R4,ABCDFGHJK和m,n,p和q定义如权利要求10和18,
q是0,1或2;
在式V中,m是0,1或2且
在式VI中,n是0,1或2
R5至R12定义如权利要求16,
L是离去基,
Z是空格或者是一个键;
其中如果需要,保护任何一个功能基;且
(b)出去任何保护基;
(c)任选地,在所述反应之前或之后将式III,V或VI的任何化合物转化为式IV,V或VI的其它化合物;
(d)任选地形成药用盐或体内可水解酯,酰胺,碳酸酯或氨基甲酸酯。
33.根据权利要求32的方法,其中反应是在催化剂存在下进行的。
34.轭合前药,或式I化合物,基本上如前文参考任何一个实施例所描述的。
