Abstract

背景
人体中或皮肤上存在各种类型的酶类,包括来自细菌的酶类、来自饮食中的酶类以及机体产生的酶类。
已知这些酶类中一部分具有不希望存在的副反应。例如,机体排出物中存在脂酶,已知这些脂酶可引起皮炎或皮疹,这是如尿布、失禁者用品、卫生毛巾、运动裤等一次性吸湿用品的使用者经常遇到的问题。
过去数十年来尿布和皮肤护理产品的生产商们已经开发出各种各样的有助于减少尿布疹(或皮疹)产生的产品。因此注意力已主要集中在减少皮肤与机体排出物的接触。这可以通过如在尿布中引入有吸湿能力或有较强吸湿能力的物质而实现。因此而减少与皮肤接触的水量。
脂酶还与脂肪酸的产生有关,其中部分脂肪酸通过机体排出物排出体外。这些脂肪酸可引起体臭。
EP 0117632-B涉及含有脂酶抑制剂,优选含锌组分,并含有载体物质的一次性用品。US 3,091,241涉及甘油三醋酸酯在阴道塞中抑制脂酶酶活性的应用。US 3,961,486指示可用己二酸降低脂酶活性并减少皮疹。
已知各种酶如脂酶的活性依赖于被称之为金属辅助因子的特定金属离子如铁离子和铜离子的存在。有建议用EDTA络合剂与金属辅助因子反应,以降低酶活性(如NL 7506962中所述)。
但发现EDTA不是金属辅助因子中金属离子的有效络合剂,尤其是在当皮肤上或组合物中存在其它金属离子时更是如此。
本发明的发明者们发现,某些络合剂,尤其是乙二胺二琥珀酸(EDDS),可通过与包括如铜或铁等金属离子的金属辅助因子发生相互作用而有效地使酶失活或受到抑制,尤其是在当存在其它离子如钙离子时。由于钙离子在机体排出物中或在皮肤表面或毛发上通常浓度很高,因而本发明的络合剂在应用于人体或动物或应用于他们的皮肤或毛发上时,比已知的络合剂如EDTA更有效。
因此,例如,已发现本发明组合物中络合剂的应用可十分有效地减缓或预防皮炎或体臭,即使组合物中或皮肤上存在钙离子时也是如此,又例如,发现即使这些组合物中这类络合剂的含量非常少时也能按要求有效预防或减轻皮炎或体臭。
发明概述
本发明提供用于降低酶活性的包含乙二胺二琥珀酸或其盐(EDDS)的组合物,优选其中所述的酶为与人体或动物体接触的酶类,尤其是存在于排出物中的酶类。
本发明还提供可用于降低酶活性(优选降低与人体或动物体接触的酶的酶活性)的含有络合剂的组合物,并且在使用时还含有钙离子以及选自Cu、Fe、Zn、Ni和Co的一种或多种金属离子(本文中称为“选定金属离子(theselected metalions)”),此时-log10CT等于或大于A或B的最小值,其中A为-log10(LT-MT),B为

发明详述
本发明的组合物通过抑制酶或使酶失活而降低各种酶类的酶活性,其中这些酶需要金属辅助因子,尤其是含铁、铜、钴、锌或镍金属离子的辅助因子。
这些酶类可以来自细菌、真菌或藻类,也可以是人体或动物体产生的酶,或者是来自饮食中的酶。
本发明的组合物尤其对酯酶类,包括脂酶,具有十分有效的抑制或使之失活的作用。
所述组合物可用于需要降低这些酶类活性的任何应用中。特别地,所述组合物用于降低与人体或动物体,特别是与皮肤接触的酶类的酶活性。
根据本发明的一个优选方面,使用时组合物中还存在钙离子源。这里是指钙离子可在含EDDS的组合物应用之前或应用时存在,钙离子例如通过将组合物应用于含有钙离子的皮肤上而引入组合物中。
在本发明的一个较优选方面中,所述组合物用于降低排出物中存在的酶类的酶活性,尤其是包括脂酶的酯酶类的酶活性。
当钙离子存在于组合物中和/或钙离子与人体或动物体接触时,组合物特别有用。
在本发明的优选实施方案中,包括络合剂的组合物可用于治疗因酶类而引起的酶性皮炎或形成的体臭,尤其是用于降低脂酶的酶活性。
所述组合物可以是美容组合物,优选为喷洒剂、霜膏(cream)、泡沫剂(foam)、乳剂(lotion)、凝胶剂、油剂、软膏剂或粉剂或片剂形式,优选为油包水乳液形式。
尤其优选组合物包含在除臭用品或吸湿用品如尿布、湿擦拭物中,如本文所述。
“治疗”在本文中是指改善人体或动物体中由酶活性所引起的感染状况。因此在本发明的一个优选方面包括,使酶引起的体臭减缓或至少是稳定;在本发明的另一优选方面,使酶引起的酶性皮炎或皮疹减轻或至少是稳定。
本发明中用于降低酶活性或在治疗中应用的组合物的量,根据待处理状况的具体部位、待处理状况的严重程度、治疗预计所需时间、具体使用者对每种组合物的特异性敏感性、使用者的状况、同时给予的治疗、使用者出现的其它情况而不同。
对于本发明,优选施用最小抑制浓度的含络合剂的组合物(优选局部应用于),用作皮肤上存在的特定金属离子的络合剂,这些金属离子是酶类达到它们的酶性能所需的,组合物的用量和形式使得能够抑制所存在的酶类的活性,尤其在钙离子存在时。
络合剂或组合物对降低酯酶类的酶活性特别有用,并因此可用于抑制酯酶如脂酶或脂解酶或使它们失活。它们的主要活性是水解以酯形式出现的脂肪(如人体皮肤中的甘油酯),并由此产生能引起刺激和产生体臭的脂肪酸和甘油。由于这组酶类在植物、霉菌、细菌、奶、奶制品,以及几乎所有动物组织中广泛存在,并且由于人类脂酶存在于胰排出物中,因而它们几乎总是出现在人体或动物体的体内或体表。
这些脂酶的活性可引起几乎所有皮疹或特别是尿布疹,通过脂酶在皮肤自身上的消化变性作用以及通过破坏脂质皮肤组分而产生刺激,破坏了感染部位的皮肤屏障功能。这种对皮肤整体性的破坏使机体排出物(尤其是尿和粪便)中可能本身不具有刺激性的其它组分穿过受损的皮肤。此时通常无害的组分就可能变得具有刺激性。
本发明组合物的高度优选的附加组分可以是杀细菌剂或杀真菌剂,和/或其它酶抑制剂。高度优选的是可引入阳离子有机化合物,如阳离子表面活性剂。
pH测量
本文中所用的pH可通过任何已知的计算或测量水溶液的pH的方法确定。
离子强度测量
离子强度(Ⅰ)可通过以下等式确定:

其中c是可溶离子(i)的分子浓度,z是可溶离子(i)的电荷。
络合剂
本发明的组合物中所用络合剂高度优选为N,N’-乙二胺二琥珀酸或其盐(EDDS)。
已知(S,S)EDDS异构体比(R,R)EDDS异构体更易发生生物降解。因此,根据本发明水性组合物的应用,理想的是只使用一种EDDS异构体。可优选在水性组合物中使用异构体的外消旋混合物,原因例如为外消旋混合物较便宜。
本发明的组合物中引入络合剂的确切量取决于组合物的特性以及应用方式。但通常极少量的络合剂就足以达到所要求的预防或降低皮炎的作用。络合剂的通常用量可以占组合物重量的0.001%-30%、更优选0.005%-10%、最优选0.01%-5%。
制备组合物的方法
组合物可用本领域制备美容组合物或药物的任何已知方法制备。具体方法将取决于组合物的特性。络合剂可以以它的酸或盐形式加入组合物中,或先与美容组合物或药物中常用的其它组分组合,或先分散或溶解在水或油或油包水乳液中后再加入到组合物中。
组合物的使用方法
本发明的组合物可以通过领域内已知的任何方法施用于病人或使用者,但优选组合物应用于皮肤或毛发,这样可以与酶类接触或接近。
组合物也可以首先应用于用品如擦拭物或卫生纸上,这些用品然后再应用于皮肤。
当组合物用于治疗体臭时,组合物可优选为除臭剂组合物形式、液体、凝胶、霜膏或粉剂形式,包含在棒或喷洒剂中。
吸湿用品
本发明的组合物可包含在吸湿用品中,优选吸湿用品为一次性吸湿用品。由此特别优选的吸湿用品为擦拭物或尿布。尿布优选在其顶片上包含所述组合物。
在本文中所用的术语“吸湿用品”指吸收和容纳机体排出物的用品,更具体地,指紧贴或靠近穿戴者的身体放置以吸收和容纳机体排出的各种排出物的用品。术语“一次性”在本文中用以描述不打算通过洗涤或以其它方式恢复或再次用作吸湿用品的吸湿用品(即它们将在使用过一次后丢弃,优选以环保的方式回收、混合制肥或处理)。
一次性吸湿用品的结构对实施本发明并不是关键。
通常,将组合物掺入吸湿用品或尿布中,特别地,用量为足以使治疗或对酶活性的降低或抑制达到要求,优选通过频繁使用达到目的。
一次性吸湿用品优选以一定的量包含本发明的组合物,从而使得其中络合剂占该用品重量的0.01%-30%,更优选0.01%-10%,最优选0.05%-5%。
附加组分
本发明的组合物可包含附加组分。包括何种组分以及其含量为多少取决于组合物的特点及其应用。因此,例如乳剂通常包含的附加组分不同于粉剂。
优选组合物中包含一种或多种可降低皮炎的其它组分、或有助于使皮肤痊愈的化合物,如维生素类(维生素E)和可的松类,以及能软化皮肤的化合物如凡士林、甘油、三甘醇、羊毛脂、石蜡和制药以及美容品生产中广泛应用的其它类聚合物,对此本文中也有描述。
根据对本发明的组合物的不同应用,优选的附加组分可以是一种或多种增效助剂或分散剂。可优选含有晶体生长抑制剂,最好是除了分散剂以外还有晶体生长抑制剂。
适于作为晶体生长抑制剂或增效助剂的水溶性磷酸盐的适宜实例有碱金属三聚磷酸盐,焦磷酸钠、钾和铵,焦磷酸钠和钾和铵,正磷酸钠和钾,聚合程度为约6-21的聚偏/磷酸钠以及肌醇六磷酸的盐类。
本领域内已知的任何增效助剂或分散剂物质均可应用。特别有用的增效助剂或分散剂可以是单体、寡聚和聚羧酸盐组分,聚合组分,含硼酸盐的组分以及含磷酸盐的组分和含硅酸盐和硅铝酸盐的组分。
适宜的聚羧酸盐或多羧酸可以是琥珀酸、丙二酸、(亚乙二氧基)二乙酰乙酸、马来酸、二乙醇酸、酒石酸、羟基丙二酸和富马酸;柠檬酸、aconitrate、柠康酸盐以及琥珀酸衍生物如英国专利第1,379,241号中所述羧甲氧基琥珀酸盐,英国专利第1,389,732号中所述乳氧基琥珀酸盐,荷兰专利申请第7205873号中所述氨基琥珀酸盐,以及英国专利第1,387,447号中所述氧基聚羧酸盐物质如2-氧杂-1,1,3-丙烷三羧酸盐;英国专利第1,261,829号中所述氧基琥珀酸氢盐,1,1,2,2-乙烷四羧酸盐,1,1,3,3-丙烷四羧酸盐和1,1,2,3-丙烷四羧酸盐;英国专利第1,398,421和1,398,422号以及美国专利第3,936,448号中所述磺基琥珀酸盐衍生物,英国专利第1,439,000号中所述磺化的热解柠檬酸盐;
聚合组分包括水溶性有机均聚型或共聚型聚羧酸或它们的盐类,其中聚羧酸包含至少两个相差不超过两个碳原子的羧基基团。后一类聚合物在GB-A-1,596,756中有述。这样的盐类实例有分子量1,000-5,000的聚丙烯酸盐以及它们与马来酐的共聚物,这类共聚物的分子量为2,000-100,000,尤其40,000-80,000。
聚氨基组分在本文中十分有用,包括由天冬氨酸衍生的那些,如EP-A-305282、EP-A-305283和EP-A-351629中公开的那些。
含有选自马来酸、丙烯酸、聚天冬氨酸和乙烯醇单体的三元共聚物,特别是那些平均分子量为5,000-10,000的那些也适用于本文。
适于掺入本文组合物中的其它聚合组分包括纤维素衍生物如甲基纤维素、羧甲基纤维素、羟丙基甲基纤维素和羟乙基纤维素。
其它有用的聚合组分还包括聚乙二醇,尤其分子量为1,000-10,000的那些,更优选分子量为2,000-8,000,最优选分子量为约4,000。
其它优选成分为非晶形或结晶型,优选层化的,硅酸盐或硅铝酸盐物质或增效助剂。适宜的硅铝酸盐沸石具有单元式为Naz[(AlO2)z(SiO2)y].xH2O,其中z和y至少为6;z∶y的摩尔比为1.0-0.5,x至少为5,优选7.5-276,更优选10-264。硅铝酸盐物质均为水合形式并优选结晶型,含10%-28%的结合水,更优选含18%-22%的结合水。
乳剂、霜膏、油剂、泡沫剂、软膏剂、凝胶剂、粉剂、片剂等
本发明的组合物可应用于任何适当的目的。尤其,本组合物适于局部施用于皮肤或毛发上。具体地,皮肤护理型组合物可以是霜膏、喷洒剂、乳剂、凝胶剂等形式。优选本文中的美容组合物为连续水相中有一或多种油相的水包油乳液形式,每种油相包含单一油性组分或多种油性组分的混溶型或均质型混合物,但所述不同油相包含相互区别的不同物质或物质的组合。本发明组合物中油相组分的总含量优选为约0.1%至约60%,更优选约1%至约30%,最优选约1%至约10%。
本发明的组合物优选包括作为,或者全部或部分油相,或者上述油相的第一含硅氧烷相,其中包括交联的聚有机硅氧烷聚合物和硅氧烷油,其中组合物包括0.1%至约20%,优选约0.5%至约10%,更优选约0.5%至约5%重量的交联的硅氧烷和硅氧烷油的混合物,基于组合物的重量。
本发明的组合物优选还包括第二含非交联的硅氧烷的相。在优选的实施方案中,第二含硅氧烷相在组合物中的存在量是约0.1%至约20%重量,特别是约0.1%至约10%重量。
用于此处的第二含硅氧烷相中的合适的硅氧烷流体包含不溶于水的硅氧烷和非挥发性的聚烷基和聚芳基硅氧烷胶和流体、挥发性的环状和线性聚烷基硅氧烷、聚烷氧基化的硅氧烷、氨基和季铵改性的硅氧烷,以及它们的混合物。
在优选的实施方案中,第二含硅氧烷相包括硅氧烷胶或包含硅氧烷胶的硅氧烷的混合物。本文中,术语“硅氧烷胶(silicone gum)”指的是重均分子量超过约200,000并且优选为约200,000至约400,000的高分子量硅氧烷基流体。硅氧烷油的分子量通常低于约200,000。通常,硅氧烷胶在25℃时的粘度超过约1,000,000mm2s-1。硅氧烷胶包含聚二甲基硅氧烷,这在Petrarch和其它包括1979年5月1日授予Spitzer等人的US-A-4152416中以及Noll,Walter的Chemistry and Technology of Silicones (NeW York:Academic Press1968)中有描述。描述硅氧烷胶的还有General Electric Silicone Rubber ProductData Sheets SE 30,SE 33,SE 54和SE 76。
在优选的实施方案中,另一种即第三油相在组合物中的存在量为约0.1%至约15%,更优选约1%至约10%,基于组合物的重量。该第三油相可以或者是单独的相或者可以与第一或第二油相中的一个或者两者形成一相。优选第三油相为单独的相。
第三油相优选包括非硅氧烷有机油,如选自矿物油、植物油和动物油、脂肪和蜡、脂肪酸酯、脂肪醇、脂肪酸及其混合物的天然或合成油,这些成分可用于提供软化美容的性能。
合适的用于本发明的第一油相组分包括,例如,任选地羟基取代的C8-C50不饱和脂肪酸及其酯、蜂蜡、饱和和不饱和脂肪醇如二十二烷醇和十六醇、烃如矿物油、矿脂和角鲨烷、脂肪酸脱水山梨醇酯(见1976年10月26日授予Seiden的US-A-3988255)、羊毛脂和羊毛脂衍生物、动物和植物甘油三酯如杏仁油、花生油、麦芽油、亚麻子油、霍霍巴油、杏核油(oil of apricotpits)、胡桃油、棕榈油、阿月浑子油、芝麻籽油、油菜籽油、杜松油、玉米油、桃核油(peach pit oil)、罂粟籽油、松油、蓖麻油、大豆油、鳄梨油、红花油、椰子油、榛子油、橄榄油、葡萄籽油、牛油树脂、娑罗双树树脂(shoreabutter)和向日葵籽油以及二聚体和三聚体酸的C1-C24酯如二异丙基二聚体酯、二异硬脂基苹果酸酯、二异硬脂酰二聚体酯和三异硬脂基三聚体酯。上述中,特别优选矿物油、矿脂、不饱和脂肪酸及其酯以及它们的混合物。
两亲表面活性剂
本文中组合物的另一优选组分是能在产品中或在当产品在室温或升温条件下应用于皮肤上时形成近晶型易溶晶体(smectic lyotropic crystals)的有机两亲表面活性剂。优选所述两亲表面活性剂能在约20℃-约40℃的温度下形成液晶。优选所述两亲表面活性剂能形成近晶型易溶液晶。一旦将所述产品施用在皮肤上以后,皮肤表面或角质层上不再能鉴别出液晶。优选两亲表面活性剂占约0.1%-约20%,优选约0.1%-约10%的重量。
适用于本文的有机两亲表面活性剂包括那些重均HLB(亲水亲油平衡值)在约2-12,优选约4-8的范围内的表面活性剂。
本文所用的优选有机两亲表面活性剂具有饱和或不饱和的、支链的或线性的、具有约12-约30个碳原子的长亲脂链,如油基、羊毛脂、十四烷、十六烷、异硬脂基、月桂基、椰子基、硬脂基或烷基苯基的链。当形成液晶相的两亲物质的亲水基团是非离子基团时,可使用聚氧乙烯、聚甘油、多元醇酯、氧烷基化或非氧烷基化的,例如聚氧烷基化的山梨醇或糖酯。当形成液晶相的两亲表面活性剂的亲水基团是离子基团时,有利的是使用卵磷脂中的磷脂酰胆碱残基作为亲水基团。
组合物还可包括两性、阴离子型、非离子型和阳离子型表面活性剂。
在本发明的护肤组合物中也可以加入各种任选组分如非包藏性补湿剂、保湿剂、胶凝剂、中和剂、香料、着色剂和表面活性剂。
本文的组合物可包括保湿剂。适用于本文的保湿剂包括山梨醇、丙二醇、丁二醇、己二醇、葡萄糖的乙氧基化衍生物、己三醇、甘油、甘氨酸、透明质酸、精氨酸、Ajidew(NaPCA)、水溶性聚甘油基异丁烯酸酯润滑剂和泛醇。本文中优选保湿剂是甘油(有时称为glycerol或glycerin)。从化学角度看,甘油是1,2,3-丙三醇,而且是一种商业产品。该物质的一个最大来源是工业化生产肥皂。从强化补湿的角度看,本发明组合物中尤其优选甘油。本文中还优选使用丁二醇。从强化补湿的角度看,尤其优选甘油和尿素的组合物。
在本发明的组合物中,保湿剂在组合物中的优选存在量为约0.1%-约20%,更优选约1%-约15%,并尤其优选约5%-约15%重量。
本发明的组合物还可包括亲水胶凝剂,含量优选为约0.01%-约10%,更优选为约0.02%-约2%,并尤其优选为约0.02%-约0.5%。胶凝剂优选粘度(1%水溶液,20℃,Brookfield RVT)为至少约4,000 mPa.s,更优选至少约10,000 mPa.s,并尤其优选至少50,000 mPa.s。
合适的亲水胶凝剂通常被描述为水溶性或胶态水溶性聚合物,并包括纤维素醚(例如羟乙基纤维素、甲基纤维素、羟丙基甲基纤维素)、聚乙烯吡咯烷酮、聚乙烯醇、瓜尔胶、羟丙基瓜尔胶和黄原胶。
但本文优选的亲水胶凝剂是B.F.Goodrich Company以聚羰乙烯树脂(Carbopol resin)的商标名出售的丙烯酸/丙烯酸乙酯共聚物以及羧乙烯聚合物。这些树脂基本上由丙烯酸的胶态水溶性聚链烯基聚醚交联聚合物与0.75%-2.00%的交联剂如聚烯丙基蔗糖或聚烯丙基季戊四醇交联组成。实例包括Carbopol 934、Carbopo1 940、Carbopol 950、Carbopol 954、Carbopol 980、Carbopol 951和Carbopol 981。Carbopol 934是与约1%蔗糖聚烯丙基醚(平均每个蔗糖分子有约5.8个烯丙基)交联的丙烯酸的水溶性聚合物。最优选的聚合物是Carbopol 954。同样适用于本文的是丙烯酸具有两亲性的疏水改性的交联聚合物,商品名为Carbopol 1382、Carbopol 1342和Pemulen TR-1(CTFA Designation:Acrylates/10-30 Alkyl AcrylateCrosspolymer)。聚链烯基聚醚交联的丙烯酸聚合物和疏水改性的交联丙烯酸聚合物的组合也适用于本文并被本发明优选使用。本文中胶凝剂在正常温度和升温条件下均能十分有效地提供极佳稳定性。
适用于中和本文中含酸性基团的亲水胶凝剂的中和剂包括氢氧化钠、氢氧化钾、氢氧化铵、单乙醇胺、二乙醇胺和三乙醇胺。
其它任选的物质包括溶角质剂/脱屑剂如水杨酸;蛋白质和多肽及其衍生物;水溶性或可溶性防腐剂,优选含量为约0.1%-约5%,如Germall 115,羟基苯甲酸的甲酯、乙酯、丙酯和丁酯,苯甲醇,EDTA,Euxyl(RTM)K400,溴硝丙二醇(2-溴-2-硝基丙烷-1,3-二醇)和苯氧基丙醇;抗细菌物质如Irgasan(RTM)和苯氧基乙醇(优选含量为约0.1%-约5%);可溶性或胶溶性补湿剂如透明质酸以及淀粉接枝的聚丙烯酸钠如Sanwet(RTM)IM-1000、IM-1500和IM-2500(Celanese Superabsorbent Materials,Portsmith,VA,USA有售,且USA-A-4,076,663中已述);维生素类如维生素A、维生素C、维生素E和维生素K;α和β-羟基酸;芦荟;鞘氨醇和植物鞘氨醇,胆固醇;皮肤美白剂;N-乙酰半胱氨酸;着色剂;香料和香料增溶剂以及附加的表面活性剂/乳化剂如脂肪醇乙氧基化物,乙氧基化的多元醇脂肪酸酯,其中的多元醇可选自甘油、丙二醇、乙二醇、山梨醇、脱水山梨醇、聚丙二醇、葡萄糖和蔗糖。实例包括甘油单羟基硬脂酸酯和乙氧基化的硬脂醇(每摩尔醇平均10-200摩尔氧化乙烯)和PEG-6辛酸/癸酸甘油酯。
本发明的组合物还可包含安全而有效量的维生素如维生素E、B,优选维生素B3维生素或维生素A类。本发明的组合物优选包含约0.01%-约50%,更优选约0.1%-约10%,还更优选约0.5%-约10%,进一步优选约1%-5%,最优选约2%-5%的维生素。
组合物的pH优选为约4-约9,更优选约6-约8.0。
组合物的平衡剂是水或适于皮肤局部应用的水性载体。组合物的含水量通常为约30%-约98.89%,优选约50%-约95%,尤其优选约60%-约90%。
本发明的组合物优选形式是应用于皮肤上为免洗产品的保湿霜或液。
Claims (16)
1.一种用于降低酶活性的组合物,其包含乙二胺二琥珀酸或其盐(EDDS)络合剂。
2.一种用于降低酶活性的水性组合物,其包含络合剂,而且其在使用时还含有钙离子以及选自Cu、Fe、Zn、Ni、Co的一种或多种金属离子,此时-log10CT等于或大于A或B的最小值,其中A为-log10(LT-MT),B为

12.一种吸湿用品,优选为擦拭物或尿布,包含前述任一项权利要求的组合物。
13.包含前述任一项权利要求的组合物的除臭剂。
14.前述任一项权利要求的组合物在降低酶活性,优选与人体或动物体接触的酶的酶活性中的应用。
15.权利要求1-13中任一项的组合物在酶性皮炎治疗中的应用。
16.权利要求1-13中任一项的组合物在体臭治疗中的应用。
