Abstract
棉铃虫蜕皮调节转录因子cDNA及其克隆方法与重组应用,属于生物基因工程技术领域。棉铃虫蜕皮调节转录因子具有SEQ ID NO 1和SEQ ID NO.2所示的序列。本发明的克隆方法是,从棉铃虫中提取总RNA或mRNA,反转录合成cDNA;根据蜕皮调节转录因子的保守序列设计引物,通过链式聚合酶反应扩增到一个DNA片段;将扩增产物纯化,取纯化产物克隆到pGEM-T Easy载体,转化DH5α细胞,平板过夜培养。本发明棉铃虫蜕皮调节转录因子,通过基因重组技术在昆虫核型多角体病毒中进行表达,获得有杀虫活性的基因重组病毒,用于害虫防治;或者通过基因工程方法将棉铃虫蜕皮调节转录因子专入作物,获得具有抗虫能力的作物。
Description
(一)技术领域
本发明涉及生物基因工程技术领域中的克隆、表达技术,具体涉及棉铃虫蜕皮调节转录因子cDNA及克隆、表达技术。
(二)背景技术
自有机合成农药在40年代问世以来,在保证农业丰收方面起了巨大作用,但它对人类的危害也日趋明显,对环境造成严重污染、危害人类健康已成为不争的事实,因使用农药而中毒的事件时有发生。大量使用广普性杀虫剂使害虫天敌及有益生物也一同被消灭,打破了自然界的生态平衡,引起新的害虫猖獗。大量使用有机合成农药还使害虫产生抗药性,形成无药可治的害虫,目前合成新药剂的速度往往赶不上害虫产生抗性的速度。化学农药带来的种种问题已引起全世界注意,减少和合理使用化学农药、从耕作制度、作物栽培及生物防治等人手,对害虫实行综合防治势在必行,利用分子生物学和基因工程技术控制害虫是当今生物学领域的研究热点和前沿。本发明旨在克隆棉铃虫蜕皮过程中关键因子-蜕皮调节转录因子基因,将该基因重组到目前已经在田间害虫防治中使用的核型多角体病毒,构建基因重组核型多角体病毒,通过对人及环境安全的昆虫核型多角体病毒载体使棉铃虫蜕皮调节转录因子在害虫体内表达,干扰害虫的正常生长和发育,导致害虫非正常蜕皮而死亡,从而达到控制害虫的目的,减少害虫防治中对化学农药的依赖,保护人类生存环境和人类健康。
昆虫幼虫要经过多次蜕皮才能变为成虫,蜕皮过程是由体内的蜕皮激素引起的,在昆虫生长发育过程中,蜕皮激素(Ecdysteroid)的滴度呈现出周期性的变化,在蜕皮激素滴度升高时,昆虫停止取食,行动迟缓,最后蜕皮,在体内则引发一系列级联反应,蜕皮激素与蜕皮激素结合蛋白(Ecdysteroid receptor,EcR)及ultraspiracle(USP)结合成为三聚体,然后结合在DNA上,诱导产生蜕皮调节转录因子(molt regulatingtranscription factor,又称为激素结合子,Hormone Receptor,HR3)。蜕皮调节转录因子(HR3)有多种功能,它结合于DNA,启动蜕皮早期必需的蛋白酶和几丁质酶表达,使旧表皮中的蛋白质和几丁质被分解吸收,同时阻遏幼虫表皮蛋白表达,并结合蜕皮激素使蜕皮激素水平下降,随着蜕皮激素水平下降,不再产生蜕皮调节转录因子,表皮蛋白得以表达,形成新的表皮,同时,在蜕皮激素下降后,蜕壳激素(EclosionHormone)释放,使旧表皮脱落,多巴脱羧酶也在蜕皮激素下降后出现,合成表皮硬化必需的前体物一多巴胺,使表皮完成鞣化和硬化。如果人为使虫体内持续产生蜕皮调节转录因子,它可结合于DNA,启动蜕皮早期反应,同时它的持续存在又抑制表皮蛋白表达,使新表皮不能产生,蜕皮晚期基因不能表达,从而导致虫体死亡。
国外已对烟草天蛾、枞色卷蛾、蜡螟、果蝇的蜕皮调节转录因子进行了分子克隆和作用机理研究,基因库(GenBank)检索结果证明目前尚无棉铃虫蜕皮调节转录因子的研究报道。昆虫核多角体病毒具有种属特异性,不感染人及其它脊椎动物,国内外均已经用做生物杀虫剂并在大田应用,但野生型核型多角体病毒杀虫速度较慢,在自然界使用达不到理想的防虫效果。通过基因工程方法,将棉铃虫蜕皮调节转录因子基因转入核型多角体病毒,可以使害虫提前进入蜕皮,从而增加核型多角体病毒的杀虫速度。通过基因工程方法,将棉铃虫蜕皮调节转录因子基因转入作物体内,可以引起取食的害虫进入非正常的提前蜕皮导致死亡,达到控制害虫的目的。由于棉铃虫属于鳞翅目夜蛾科,因此该项发明对其它鳞翅目害虫、特别是夜蛾科害虫的防治具有参考意义。

本发明针对现有技术的不足,提供一种从棉铃虫提取的棉铃虫蜕皮调节转录因子cDNA,并提供棉铃虫蜕皮调节转录因子cDNA的克隆方法以及重组技术。

本发明的棉铃虫蜕皮调节转录因子cDNA的克隆方法如下:
(1)从棉铃虫提取总RNA或mRNA, 然后反转录合成cDNA;
(2)根据蜕皮调节转录因子的保守序列设计引物:
正向引物:5’-ATGAACAACAACCAGTTCCAC-3’
反向引物:5’-GTCCTGGACTACACCCACGGT-3’
(3)通过链式聚合酶反应扩增到一个DNA片段;
(4)将扩增产物纯化,取纯化产物克隆到pGEM-T Easy载体,转化DH5α细胞,平板过夜培养。
上述棉铃虫蜕皮调节转录因子cDNA的克隆方法具体操作如下:
从田间采集或人工饲养的棉铃虫中,采用一步法或RNA kit提取总RNA,或者用mRNAkit分离提取mRNA。
然后利用5~10微克总RNA或mRNA反转录合成cDNA。cDNA第一链合成:5~10微克总RNA或mRNA,加反应液20微升,反应液是50毫摩尔氯化钾和3毫摩尔氯化镁混合液,10毫摩尔三羟甲基氨基甲烷-盐酸缓冲液(Tris-HCl)pH8.3,1毫摩尔二硫苏糖醇(DTT),5微摩尔寡聚脱氧胸腺核苷酸(Oligod(T)17),500微摩尔脱氧核糖核苷酸混合物(dNTP),25单位RNA酶抑制剂,8单位AMV逆转录酶)42℃反应90分钟。70℃10分钟终止反应。
链式聚合酶反应(PCR)试剂与条件如下:首先将下列试剂混在一起:

链式聚合酶(PCR)反应条件为:首先94℃变性2分钟,然后进入下列循环:94℃30秒,53℃45秒,72℃45秒,共进行30个循环,最后72℃延伸10分钟。
结果扩增到一个DNA片段。将扩增产物纯化,取3μl纯化产物克隆到pGEM-T Easy载体(Promega公司产品),转化DH5α细胞(常用载体宿主细胞),平板过夜培养。挑取3个白斑,提取质粒,用于序列测定。
测序得到一个开放阅读框部分,1668bp,由此推得具556个氨基酸的一段序列,将此序列放入国际基因库中检索,与已发表的埃及伊蚊、家蚕、果蝇等动物的蜕皮调节转录因子的序列比较,结果显示一致性为70%左右,因此可以确定该基因为一种新的蜕皮调节转录因子。
上述棉铃虫蜕皮调节转录因子cDNA的重组应用,通过基因重组技术在昆虫核型多角体病毒中进行表达,获得有杀虫活性的基因重组病毒,用于害虫防治;或者通过基因工程方法将棉铃虫蜕皮调节转录因子转入作物,获得具有抗虫能力的作物。
根据棉铃虫蜕皮调节转录因子cDNA序列,设计表达引物,通过基因重组技术将该蜕皮调节转录因子重组到昆虫核型多角体病毒,产生有杀虫活性的蜕皮调节转录因子基因重组核型多角体病毒,获得具有生产价值的基因重组病毒杀虫剂;或者通过基因工程方法将棉铃虫蜕皮调节转录因子重组到作物,增加作物抗虫性,获得具有抗虫能力的作物,利于害虫的防治。
本发明克隆到棉铃虫蜕皮调节转录因子cDNA,通过在昆虫核型多角体病毒中进行表达,可获得有杀虫活性的基因重组病毒,用于害虫防治;通过基因工程方法将棉铃虫蜕皮调节转录因子转入作物,可获得具有抗虫能力的作物。
(四)具体实施方式
实施例1:棉铃虫(Helicoverpa armigera),田间采集。
1.总RNA的提取:采用现有技术一步法提取总RNA。
2.cDNA第一链合成:6微克总RNA,加反应液20微升,反应液是50毫摩尔氯化钾和3毫摩尔氯化镁混合液,10毫摩尔三羟甲基氨基甲烷-盐酸缓冲液(Tris-HCl)pH8.3,1毫摩尔二硫苏糖醇(DTT),5微摩尔寡聚脱氧胸腺核苷酸(Oligod(T)17),500微摩尔脱氧核糖核苷酸混合物(dNTP),25单位RNA酶抑制剂,8单位AMV逆转录酶)42℃反应90分钟。70℃10分钟终止反应。
3.PCR反应:链式聚合酶反应(PCR)试剂与条件:
首先将下列试剂混在一起:

PCR反应条件为:首先94℃变性2分钟,然后进入下列循环:9-4℃ 30秒,53℃45秒,72℃45秒,共进行30个循环,最后72℃延伸10分钟。
4.反应产物纯化:利用德国奎因(QIAGEN)公司产品QIAquick Gel Extraction Kit,操作步骤按产品说明书进行。
5.棉铃虫蜕皮调节转录因子cDNA克隆:取纯化产物3微升,连接于pGEM-T Easy载体(Promega公司产品)。转化到大肠杆菌DH5α菌株,在含有氨苄青霉素(100微克/毫升)、5-溴-4-氯-3-引哚-β-D-半乳糖苷(X-gal 0.2微克/毫升)和异丙基硫代半乳糖苷(IPTG0.1摩尔/毫升)的平板生长过夜,挑取3个白斑,在LB液体培养基(5毫升,含100微克/毫升安苄青霉素)中培养过夜。
6.质粒纯化:收取过夜培养菌液2毫升,离心(10000转,1分钟)收集细胞。用微量DNA纯化试剂盒(Wizard plus SV Minipreps DNA Purification System,美国普洛麦Promega公司)纯化质粒,纯化步骤按说明书进行。
7.序列测定与同源检索:取纯化质粒4微升,用载体引物T7进行全自动测序(本工作在上海生工公司完成)。将所得序列与基因库序列比较。
所得基因具有SEQ ID NO 1和SEQ ID NO.2所示的序列。
实施例2:
如实施例1所述,所不同的是棉铃虫是人工饲养的,采用RNA kit提取总RNA。然后用8微克总RNA进行cDNA第一链合成。
实施例3:
如实施例1所述,所不同的是用mRNA kit分离提取mRNA。然后用10微克mRNA进行cDNA第一链合成。质粒纯化中收取过夜培养菌液1毫升进行质粒纯化。
实施例4:
将实施例1获得的棉铃虫蜕皮调节转录因子cDNA通过现有技术基因工程方法,转入昆虫核型多角体病毒中,在昆虫细胞或昆虫体内表达,生产具有杀虫功能的基因重组病毒
实施例5:
将实施例2获得的棉铃虫蜕皮调节转录因子cDNA通过现有技术基因工程方法,转入植物体内,如棉花,增加其抗虫性,在大田中应用。
按照本发明的方法可以通过现有基因工程技术,修饰棉铃虫蜕皮调节转录因子基因并用于所述研究和生产。

(1)从棉铃虫提取总RNA或mRNA,然后反转录合成cDNA;
(2)根据蜕皮调节转录因子的保守序列设计引物:
正向引物:5’-ATGAACAACAACCAGTTCCAC-3’
反向引物:5’-GTCCTGGACTACACCCACGGT-3’
(3)通过链式聚合酶反应扩增到一个DNA片段;
(4)将扩增产物纯化,取纯化产物克隆到pGEM-T Easy载体,转化DH5α细胞,平板过夜培养。
3.如权利要求2所述棉铃虫蜕皮调节转录因子cDNA的克隆方法,其特征在于,所述步骤(1)中反转录合成cDNA是第一链合成cDNA,取5~10微克总RNA或mRNA,加反应液20微升,反应液是50毫摩尔氯化钾和3毫摩尔氯化镁混合液,10毫摩尔三羟甲基氨基甲烷-盐酸缓冲pH8.3,1毫摩尔二硫苏糖醇,5微摩尔寡聚脱氧胸腺核苷酸,500微摩尔脱氧核糖核苷酸混合物,25单位RNA酶抑制剂,8单位AMV逆转录酶,42℃反应90分钟,70℃反应10分钟终止反应。
4.如权利要求2所述棉铃虫蜕皮调节转录因子cDNA的克隆方法,其特征在于,链式聚合酶反应试剂与条件如下:
首先将下列试剂混在一起:
10×Taq DNA聚合酶缓冲液 5微升
模板cDNA 1μl
正向引物1.25μg/μl 1μl
反向引物1.25μg/μl 1μl
脱氧核苷酸混合物 4μl
Taq DNA聚合酶 0.25μl
灭菌水 37.75μl
总体积 50μtl
链式聚合酶反应条件为:首先94℃变性2分钟,然后进入下列循环:94℃30秒,53℃45秒,72℃45秒,共进行30个循环,最后72℃延伸10分钟。
5.一种权利要求1所述棉铃虫蜕皮调节转录因子cDNA的重组应用,通过基因重组技术在昆虫核型多角体病毒中进行表达,获得有杀虫活性的基因重组病毒,用于害虫防治;或者通过基因工程方法将棉铃虫蜕皮调节转录因子专入作物,获得具有抗虫能力的作物。
