在优选的实施方案中,通过自甲醇代谢中涉及的基因衍生的启动子来控制甲基营养酵母中HBsAg的表达。充分描述过的启动子是MOX启动子,DAS和FMD启动子(Ledeboer,A.M.等,核酸研究(Nucl.Acids Res.)(1985),13,9,3063-3082;Janowics,Z.A.等,核酸研究(Nucl.Acide Res.)(1995),13,9,2043-3062,EP299108)。为了实现所述启动子控制下的最佳表达,首先让酵母细胞在全合成营养培养基中生长。加入甲醇诱导,使得细胞悬浮液中含有大约1%的甲醇。关于甲基营养酵母的培养和上述三种启动子的诱导条件的详细情报可以例如参见Gelissen,G.,Murooka/lmanaka(编著)工业和农业应用的重组微生物(Recombinant microbes for industrial and agriculturalapplications),Marcel Dekker,NY1993,787-796和Weydemann,U.等,应用微生物.生物技术(Appl.Microbiol.Biotechnol.)(1995),44,377-385。
完成发酵过程后,通过切向流过滤从培养基成分分离细胞。通过用破碎缓冲液适当稀释来调节期望的细胞密度。
对于破碎,优选以50-150克/升细胞干重的细胞密度使用酵母细胞。精确的细胞密度取决于各重组甲基营养酵母细胞物种。特别地,70-120克/升细胞干重的细胞密度是优选的。如果根据本发明使用的重组甲基营养酵母细胞的细胞密度太低,则该方法变得费时且不经济。酵母细胞细胞密度过高时,破碎变得越来越效率低,对冷却能力的要求提高。
根据本发明使用的高压匀浆器正如导论中所描述的是常规高压匀浆器,其可以用于细胞破碎。特别优选的是使用Nanojet LAB30PL型或相当型的高压匀浆器。在细胞破碎过程中,匀浆器装置中的压力是1.0×108-2.0×108帕。1.2×108-1.6×108帕的压力是优选的。特别优选的压力是1.5×108-1.6×108帕。
酵母细胞的起始温度优选是2-15℃,特别优选4-8℃。通常以细胞悬浮液形式存在的酵母细胞可以在搅拌下冷却到期望的温度。优选冷却高压匀浆器,使得在产物出口处观察到的低出口温度例如是2-15℃,优选3-13℃,特别优选5-10℃。
在高压匀浆器中,细胞通常经几次循环(通过)破碎。发现共3-8次循环(通过)对于最佳产物收率就足够了。对于回收重组HBsAg的细胞破碎优选3-6次,特别优选4次循环(周期)。
细胞破碎过程完全后,用该方法制备的产物优选使进行进一步分离和纯化步骤。所述附加的处理步骤是将提取物中的重组HBsAg进行进一步纯化和富集的常规方法。例如,可以进行一种或几种下面的分离和纯化步骤,如以指出的顺序:
用聚乙二醇沉淀细胞碎屑,分离PEG上清液,在硅石基质上吸附,分离硅石基质,从硅石基质解离产物,分离硅石基质上清液,离子交换层析,通过超滤浓缩离子交换集液(ion exchanger pool),氯化铯中的密度梯度分离,大小排阻层析和无菌过滤(最后的含水物质(bulk))。
下面的实施例将解释本发明。
实施例1
通过高压破碎细胞破碎多形汉逊氏酵母回收HBsAg
1.高压匀浆器
下面说明的装置用于高压破碎方法:
Nanojet Lab30 PL:
破碎原理: 高压
体积流速: (100±20)毫升/分钟,不可调节
活塞泵输入压力: 4×105-7×105帕
匀浆装置压力: 1.2×108-1.6×108帕
匀浆阀门: 碳化钨
冷却: 管式热交换器
细胞干重: 70克/升
密封剂: EPDM
高压密封: Buna N
材料: 1-45-71钢
2.微生物及其培养
在全合成营养培养基中培养表达HBsAg的多形汉逊氏酵母菌株,例如Janowicz等,酵母(Yeast),7:431-443,1991中描述的菌株。在发酵过程中,控制pH,通气量和温度。发酵过程中,响应溶解的氧的浓度间歇性地向发酵罐培养基补充作为底物的甘油使得细胞密度最大大约90克/升细胞干重。在细胞密度大约90克/升细胞干重和约46小时发酵时间时,通过18-26小时内加入甲醇一次,在多形汉逊氏酵母培养基中诱导HBsAg表达。通过在干重天平上蒸发直至恒重从培养物等份测定细胞干重。最后加入后,使培养物再生长2-9小时。接着,通过交叉流过滤从发酵培养基成分大量分离细胞。破碎之前将细胞缓冲。缓冲使用20mM磷酸盐缓冲液和2mM Na2-乙二胺四乙酸二水合物(Triplex III)。向最后的悬浮液加入每克细胞干重75毫克洗涤剂Tween2O。破碎多形汉逊氏酵母时,以大约70克/升细胞干重细胞密度使用发酵罐培养肉汤。
3.细胞破碎
搅拌下将缓冲的细胞冷却到4-8℃。破碎装置上的起始温度调节到8±5℃。在一个分开的冷却搅拌容器中收集细胞提取物。
通过活塞冲程体积和每分钟的冲程次数测定高压匀浆器中体积流速并且不能各个调节。作为原则,在每分钟100毫升细胞悬浮液的流速下进行破碎。将细胞共4次循环破碎。第4次循环完成后,用80mM PMSF贮存溶液调节到2mM终浓度。
细胞破碎后,可以以下面指出的次序或者以不同的次序通过下面的分离和纯化步骤来纯化产物:
·用聚乙二醇沉淀细胞碎屑
·分离PEG上清液
·在硅石基质上吸附
·分离硅石基质
·从硅石基质解离产物
·分离硅石基质上清液
·离子交换层析
·通过超滤浓缩离子交换器集液
·氯化铯中的密度梯度分离
·大小排阻层析
·无菌过滤(最后的含水物质)
为了研究高压破碎的效率,通过Lowry方法测定总的蛋白质含量,并且通过免疫学方法(“ELISA”)测定细胞提取物中HBsAg浓度。
以10升发酵规格进行Nanojet高压破碎。为了保证可比性,使HBsAg的产率与细胞破碎之前各细胞量相关联。
4.结论
4.1测试Nanojet高压破碎

冷却: 双层夹套和转子冷却
细胞干重: 100-120克/升
体积流速: (200-300)毫升/分钟
密封材料: 乙烯丙烯
Dyno Mill KDL Spezial:
破碎原理: 湿磨
有效研磨腔体积: 600ml
搅拌盘: 4,聚氨基甲酸乙酯
研磨体的填入: 有效研磨体积的80-85%
转速: 6.8m/s
同轴环间距: 0.1mm
冷却: 双套夹套
细胞干重: 70克/升
体积流速: 100毫升/分钟
密封材料: Viton
使用直径0.45-0.55mm的玻璃球进行湿磨。
2.微生物及其培养
使用和实施例1相同的材料,但是发酵培养肉汤的细胞密度是70-120克/升细胞干重。
3.细胞破碎
根据实施例1所述进行前处理和后处理。
Dyno Mill KDL Spezial中湿磨
通过体积流速为100毫升/分钟的蠕动泵向卧式破碎室连续加入含有70克/升细胞的细胞悬浮液。同时,为了蛋白酶抑制,加入80mM PMSF溶液,在立即混合下1/40的细胞悬浮液流向破碎室的分开的入口部分。在一个循环内破碎细胞悬浮液。
Fryma Coball Mill MS18内湿磨
这里也是通过蠕动泵连续向竖式破碎室加入细胞悬浮液。但是,该实施例中的细胞密度是100-120克/升,体积流速设定在200-300毫升/分钟。与Dyno Mill KDL Spezial相对照,蛋白酶抑制剂贮存液(80mMPMSF)不在球磨机出口之前加入。细胞悬浮液在一个循环内被破碎。终细胞提取物中的PMSF终浓度是2mM。
以50升规模进行使用Fryma Coball Mill MS18进行破碎试验。在使用Dyno Mill KDL Spezial进行的湿研磨中使用10升发酵规模。为了保证可比性,使HBsAg的产率与细胞破碎之前各细胞量相关联。

实施例2
湿磨和高压破碎之间的比较
1.Nanojet高压破碎和Dyno Mill玻璃球破碎的破碎效率
在与Dyno Mill玻璃球研磨机中细胞破碎比较时,从相同的起始物每克干物质释放出高四倍的产物的量。而同时总的蛋白质量只增加了2倍。结果,从相同的起始物释放出了更大量的产物(HBsAg)。在得到最后的含水物质的所有随后的处理步骤之后,高压破碎方法中最初的产物产率保持增长,产物的质量相当于标准方法(湿磨)的质量。这可以利用所描述的Lowry和ELISA的组合来证明。
2.Nanojet高压破碎和Fryma Coball Mill玻璃球破碎的破碎效率
从各三次通过的平均值来看,与玻璃球破碎相比,Nanojet高压破碎每克干物质释放出高2.9倍的产物的量。细胞破碎更高的产物量得到高2.6倍的量的最后的含水物质中纯化的产物。根据下面的分析证明证实,两种破碎方法的产物的质量处于相同规格。

4.结论
上述分析提供了与现有技术作出的设想相反的证据,高压破碎对于酵母细胞,至少对于甲基营养型酵母细胞相当有利。通过高压破碎,产率可以提高2.5-4倍。
Claims (7)
1.一种回收重组乙肝表面抗原的方法,其中使用高压匀浆器破碎能表达乙肝表面抗原的重组甲基营养酵母细胞,并且从得到的细胞碎屑回收乙肝表面抗原。
2.根据权利要求1的方法,特征在于所述重组甲基营养酵母细胞是汉逊氏酵母属的种的酵母细胞。
3.根据权利要求1或2的方法,特征在于所述酵母细胞在50-150克/升细胞干重的细胞密度下用于破碎。
4.根据权利要求1的方法,特征在于在高压匀浆器的匀浆装置中的1×108-2×108帕的压力下破碎酵母细胞。
5.根据权利要求1的方法,特征在于产物出口处的温度是2℃至15℃。
6.根据权利要求1的方法,特征在于细胞破碎3-6次循环。
7.根据权利要求1的方法,特征在于细胞破碎之后,使重组乙肝表面抗原进行进一步的分离和纯化步骤。
