技术领域
本发明属于医药技术领域,具体涉及一种塑料组合包装碳酸氢钠林格注射液制备方法。
背景技术
碳酸氢钠林格注射液为复方电解质注射液,用于在循环血液量以及组织间液减少时(如围手术期等情况),作为细胞外液的补充调节剂,纠正代谢性酸中毒。碳酸氢钠林格注射液能快速、有效地改善代谢性酸中毒,在维持酸碱平衡及电解质平衡上具有明显的效果。
碳酸氢钠林格注射液中的成分包括氯化钠、氯化钾、氯化钙、氯化镁、碳酸氢钠、枸橼酸钠和枸橼酸,枸橼酸对本品的PH值无明显影响,生产过程中需要通过添加二氧化碳来调节产品的PH值,产品过滤后灌装于直立式无微漏输液袋内,灭菌后外覆聚酯高阻隔层/粘合层/聚丙烯基材层高阻隔膜,能够保证产品的质量均一、稳定;但是上述生产方法中灭菌后需要充入二氧化碳,增加了成本,并且步骤增多,降低了生产效率。
发明内容
本发明旨在提供一种塑料组合包装碳酸氢钠林格注射液制备方法,解决现有碳酸氢钠林格注射液在生产时灭菌后需要充入二氧化碳,增加了成本的技术问题。
为解决上述技术问题,本发明采用以下技术方案:
一种塑料组合包装碳酸氢钠林格注射液制备方法,包括以下步骤:
(1)向稀配罐内注入处方量75%~85%的注射用水;
(2)待注射用水的水温降至40±5℃时,开启搅拌,然后依次加入处方量的枸橼酸、枸橼酸钠和氯化钠,搅拌至溶解;
(3)待上述步骤所投原料完全溶解后,再加入氯化钾、氯化钙和氯化镁,搅拌至溶解;
(4)上述原料全部溶解后加入处方量碳酸氢钠,再补加注射用水至全量,开启稀配罐自循环,循环搅拌10分钟;
(5)接着开启大循环和二氧化碳充气阀门,二氧化碳流速≤3.5 Nm/s,二氧化碳调节配置液pH值至6.4±0.2,当药液pH值达到6.4时,降低二氧化碳流速至1Nm/s以下,维持充气;
(6)循环20分钟取样进行中间产品检测,中间产品检测合格后进行过滤,然后灌装于直立式无微漏输液袋内;
(7)进行灭菌,温度在121℃,时间15min,F0≥15,出柜温度不超过60℃;
(8)灭菌后,在内外袋之间放入规格1ml二氧化碳指示剂小包,使用热封聚酯高阻隔层/粘合层/聚丙烯基材层高阻隔膜。
与现有技术相比,本发明的有益效果是:本发明碳酸氢钠林格注射液通过在配料中加入微量的枸橼酸,有效降低了调节注射液的碳酸氢根的消耗;通过加入微量的二氧化碳,抑制了添加枸橼酸的副作用;使得产品在灭菌前后pH值变化在可控范围内。同时,通过将注射液的包装更换为直立式无微漏输液袋,并在外部套设阻隔袋,内外袋之间放入二氧化碳指示剂小包,使注射液在有效期内pH值保持在合格范围内;提高了产品的稳定性。
