Abstract
本发明提供了以PLLP基因为靶点的分子在制备肿瘤疾病治疗药物中的应用,以及PLLP基因作为肿瘤疾病诊断试剂盒中诊断标志物的应用。本发明通过对缺氧环境下的血管内皮细胞进行过表达PLLP基因的处理后,血管内皮细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力显著降低,并显著了提高血管内皮细胞的凋亡。

技术领域
本发明公开了一种以基因为靶点的分子的制药学应用。
背景技术
缺氧是肿瘤微环境的一个重要特质,血液的供应不足会导致肿瘤常在缺氧的环境下生存。研究发现肿瘤细胞通过各种机制诱导缺氧,例如高新陈代谢速率和高氧消耗,从而引起内皮功能障碍或由于对血管的各种作用而破坏氧气的输送,制造慢性缺氧环境,激活低氧诱导因子HIF(Hypoxia-inducible factors)信号通路,加速肿瘤的生长,提高肿瘤的侵袭性,促使肿瘤发生转移。与正常代谢的细胞不同,肿瘤细胞能够适应缺氧的环境,并在低氧的水平下达到血管生成、增殖和侵袭的目的,最终促进形成更具侵袭性的肿瘤表型。缺氧导致肿瘤出现缺氧耐受细胞,该细胞能够促进肿瘤的局部侵袭、转移和复发。故靶向缺氧成为治疗癌症的重要策略之一,寻找与缺氧相关的关键基因,对于改善患者的预后具有重要意义。内皮细胞作为血管的重要构成部分,参与了氧气和营养物质的转移与血管的运动调节,在肿瘤环境中,缺氧可以刺激肿瘤细胞过度释放生长因子、细胞因子和乳酸等物质,以促进肿瘤血管的生成,促进内皮细胞的代谢。血管内皮细胞在新生血管形成中起重要作用,内皮细胞的增殖和迁移可形成毛细血管网络,为新生血管形成提供条件。相比于常氧条件下,缺氧条件下肿瘤组织就会像出芽一样不断的生成新生的血管,细胞因子的释放也会增加,肿瘤细胞的增殖、侵袭与粘附也会增加。因此,肿瘤血管就成为研究抗肿瘤药物的一个重要靶点,而作为肿瘤血管的重要组成细胞,内皮细胞对于抗肿瘤血管的生成又显得尤为重要。
当癌细胞处于缺氧状态,癌症的治疗就会变得更加困难,主要原因在于癌细胞能够通过改变自身生物学过程适应缺氧环境,并对标准治疗方法产生抵抗。该项目就沿着这个思路逆向思维,阻止这个过程,即研发抗血管生成药物,切管血管生长,阻断肿瘤的养料供应,从而杀死肿瘤。运用单链向导RNA(sgRNA)引导Cas9蛋白产生切割,而后使得被切割的基因通过非同源末端接合或同源介导修复来进行修复,此时非同源末端接合会引起碱基的插入或删除,最后达到了对目的基因的定点敲除。使用到的CRISPR-gRNA 全人类基因敲除文库则是将全人类基因组的gRNA囊括其中,将此文库转入细胞后就能通过将全人类基因组gRNA转入,再通过外界不同条件的压力筛选,以达到定点敲除该gRNA对应基因的目的,因此,gRNA文库是药物筛选或靶向筛选特定通路的理想工具,gRNA文库的建立将在功能基因筛选、疾病机制研究及药物研发等方面发挥重要的功能。
CRISPR-gRNA 全人类基因敲除(GeCKO)将全人类基因组的gRNA囊括其中,将此文库转入细胞后就能通过将全人类基因组gRNA转入,再通过外界不同条件的压力筛选,以达到定点敲除该gRNA对应基因的目的。本发明的目的就是使用此文库,对人血管内皮细胞进行大规模感染,在全基因组范围内筛选血管内皮细胞与缺氧相关的基因,进而提供与缺氧相关的肿瘤治疗的靶点,并提供该靶点在制备肿瘤治疗药物的中的应用。
发明内容
基于上述目的,本发明首先提供了一种以PLLP基因为靶点的分子在制备肿瘤疾病治疗药物中的应用。
本发明通过CRISPR-gRNA 全人类基因敲除筛选出血管内皮细胞缺氧相关的膜蛋白相关基因PLLP(Plasmolipin)基因,发现在缺氧条件下PLLP的敲低表达可以显著提高低氧条件下血管内皮细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力,并抑制血管内皮细胞的凋亡,而过表达PLLP基因则显著降低低氧条件下血管内皮细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力,并提高血管内皮细胞的凋亡,证实了PLLP基因显著影响缺氧环境下的细胞的生长状态,而缺氧是肿瘤微环境的一个重要特质,血液的供应不足会导致肿瘤常在缺氧的环境下生存,这是很多实体肿瘤的一种标志,例如胶质瘤、乳腺癌、肝细胞癌、胰腺癌、胃癌、结直肠癌(CRC)和前列腺癌等肿瘤。因此本发明提供了以PLLP基因为靶点的分子在制备肿瘤疾病治疗药物中的应用。
在一个优选的实施方案中,所述以PLLP基因为靶点的分子为促进PLLP表达的分子。
基于本发明的研究,过表达PLLP基因可以显著降低低氧条件下血管内皮细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力,并提高血管内皮细胞的凋亡,因此,促进PLLP表达的分子可以作为肿瘤治疗药物应用,以在低氧环境下为肿瘤细胞提供促进PLLP表达的分子从而达到抑制肿瘤细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力,并提高肿瘤细胞的凋亡。
在一个更为优选的实施方案中,所述促进PLLP表达的分子为含有PLLP编码基因的载体。
在一个尤为优选的实施方案中,所述载体为慢病毒载体。
在本发明的一个具体实施方案中,所述PLLP编码基因的序列如SEQ ID NO.1所示。
本发明构建了过表达PLLP编码基因的慢病毒载体,所述载体转染HUVEC细胞后可以显著抑制细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力,并促进了细胞凋亡,显示了所述过表达PLLP编码基因的慢病毒载体可以作为肿瘤疾病治疗药物的应用前景。
在本发明的一个具体实施方案中,所述肿瘤疾病治疗药物为抑制肿瘤细胞增殖和/或促进其凋亡的药物。本发明的实施例显示,过表达PLLP编码基因可以显著抑制细胞增殖和/或促进其凋亡。
在本发明的一个具体实施方案中,所述肿瘤疾病治疗药物为抑制肿瘤细胞迁移和/或侵袭的药物。本发明的实施例显示,过表达PLLP编码基因可以显著抑制细胞迁移和/或侵袭。
在本发明的一个具体实施方案中,所述肿瘤疾病治疗药物为抑制肿瘤细胞紧密连接能力的药物。本发明的实施例显示,过表达PLLP编码基因可以显著抑制细胞迁移和/或侵袭。
在本发明的一个具体实施方案中,,所述肿瘤疾病治疗药物为抑制细胞致血管形成能力的药物。本发明的实施例显示,过表达PLLP编码基因可以显著抑制血管形成能力。
其次,本发明提供了PLLP的编码基因作为肿瘤疾病诊断试剂盒中诊断标志物的应用,所述试剂盒中含有检测待检测样本中的所述PLLP的编码基因的含量或者表达量的试剂。
基于本发明的研究,PLLP的编码基因与缺氧环境下细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力高度相关,即,在缺氧环境下,PLLP基因的转录量和表达量显著提高,同时,细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力也显著提高,凋亡下降,而缺氧也是肿瘤细胞生长的环境,细胞的活力、增殖率、细胞迁移和/或侵袭能力、细胞紧密连接能力,血管形成能力显著提高,凋亡下降也是肿瘤细胞的生长特点,因此,鉴于PLLP基因与肿瘤细胞生长的相关性,本发明提供了PLLP基因可以作为肿瘤疾病诊断试剂盒中诊断标志物的应用。本发明提供的应用可以针对临床患者进行PLLP基因的含量或者表达量的检测从而筛选、检测肿瘤疾病或者评价肿瘤治疗的预后。
在本发明的一个优选实施方案中,所述应用为PLLP基因检测试剂盒,所述基因检测试剂盒含有序列分别如SEQ ID NO.2和SEQ ID NO.3所示的用于PCR扩增的上游引物和下游引物。
在另一个优选的技术方案中,所述应用为PLLP表达物,即PLLP蛋白质检测试剂盒,所述蛋白质检测试剂盒为蛋白质免疫检测试剂盒。在本领域许多的免疫检测试剂盒,例如,Westernblot、免疫荧光、ELISA等均可以通过特异性结合PLLP蛋白的抗体实施对PLLP蛋白的检测。
本发明通过CRISPR-gRNA 全人类基因敲除(GeCKO)文库,对人血管内皮细胞感染,筛选到一种膜蛋白相关基因Plasmolipin(PLLP)。通过在常氧与低氧条件下检测PLLP在细胞增殖、迁移、侵袭、活力以细胞连接、血管生成等一系列与肿瘤发生发展相关的指标,发现PLLP在低氧条件下可以显著影响血管内皮细胞的上述特性,而在常氧条件下对细胞没有任何影响,由于缺氧是肿瘤微环境的一个显著特征,是很多实体肿瘤的一种标志,在整个癌症发展过程中,缺氧可能产生不同的影响。特别是它可以刺激血管生成,致使癌细胞恶性增殖、抑制细胞凋亡、免疫逃避等;最终导致更加恶性和致命的癌症,例如胶质瘤、乳腺癌、肝细胞癌、胰腺癌、胃癌、结直肠癌(CRC)和前列腺癌等肿瘤,因此,PLLP有望成为一类新型用于上述缺氧类型的癌症治疗的药物。

具体实施方式
下面结合具体实施例来进一步描述本发明,本发明的优点和特点将会随着描述而更为清楚。但这些实施例仅是范例性的,并不对本发明的权利要求所限定的保护范围构成任何限制。
实施例1. 稳定表达Cas9细胞系的构建
1. 实验材料及来源:人脐静脉血管内皮细胞 (货号:CM-H261 上海盖宁生物科技有限公司),慢病毒包装的质粒载体Leti-Cas9-puro (上海吉凯基因科技有限公司,货号为:7768-1,表达CAS9蛋白),CRISPR-Pool™ KOUT 文库(吉凯基因),lenti CRISPR v2(Addgene:货号#52961)。
2. Cas9慢病毒转染单克隆细胞
1)在37℃、5%CO2条件下使用ECM培养基正常培养HUVEC细胞;
2)在5cm细胞皿中接种细胞,待细胞生长密度达50%时进行Cas9慢病毒(Leti-Cas9-puro)转染;
3)向2mL细胞培养基中加入10μL的慢病毒和40μL的病毒转染增强剂,于37℃、5%CO2条件下培养细胞6h;
4)更换新鲜ECM培养基,于37℃、5%CO2条件下培养细胞12h。
3. 压力筛选
1)更换含puromycin浓度为2μg/mL的ECM培养基,于37℃、5%CO2条件下继续培养进行压力筛选;
2)观察细胞,并及时更换含puromycin的新鲜培养基;
3)观察到有细胞存活并正常生长,使用克隆环挑选出存活的单克隆细胞,并进行扩大培养,获得稳定表达Cas9的HUVEC单克隆细胞。
4. PCR检测Cas9在基因组水平表达情况
(1) 转染Cas9单克隆细胞的基因组提取,采用QIAGEN试剂盒对挑选出的八个单克隆细胞的基因组进行提取,提取到的基因组可置于-20℃留存;
(2) PCR扩增
PCR反应条件:首先95 ℃,3 min;然后94 ℃,30 s;60 ℃,30 s;72 ℃,25 s;共28个循环,然后7 ℃保存。
扩增基因组的PCR的引物如表1所示:

将扩增后产物进行凝胶电泳,使用浓度为1%的琼脂凝胶,上样量为10μL;在核酸凝胶显影仪进行显影。
人脐静脉血管内皮细胞(HUVEC细胞)感染包装Cas9表达质粒的慢病毒后通过加压筛选获得了8个生长状态良好的单细胞克隆,提取基因组后利用PCR检测Cas9的表达情况,结果如图1所示,8个单克隆细胞的Cas9在基因组水平均有表达。
5. 反转录PCR检测Cas9在单克隆细胞转录水平情况
(1)使用QIAGEN RNeasy Mini Kit试剂盒对挑选出的八个单克隆细胞的RNA进行提取;测量提取的RNA浓度,提取到的RNA可置于-80℃留存;
(2)反转录反应程序:1. 25 ℃10 min;2. 37 ℃2 h;3. 85 ℃5 min;4. 4 ℃。反转录后得到cDNA,再将cDNA进行PCR扩增,扩增程序、电泳检测程序同前述PCR扩增,引物同表1。
利用RT-PCR检测了上述8个单克隆细胞Cas9在转录水平的表达情况。结果如图2所示,所有单克隆细胞的Cas9在转录水平均有表达。
6. Western-blot检测转染Cas9单克隆细胞蛋白的表达
1)细胞蛋白裂混合解液冰上裂解30min;
2)在冰上将细胞碎片和裂解混合液移入EP管中低温高速离心机中4℃,12000´g离心5min;
3)取上清BCA试剂盒进行蛋白质定量;
4)蛋白质上样量为40μg进行SDS-凝胶电泳,浓缩胶部分设置电压80V,分离胶部分设置电压120V;
5)于冰上干式转膜PVDF 1h,设置电压100V;
6)使用 5% 的封闭液进行PVDF膜的封闭,
7)孵育好一抗后,取出将其放入 1×洗膜液,洗五次,每次 5 min;
8)择对应的二抗,稀释成相应比例,将膜放入对应的二抗,孵育 1 h;
9)孵育好二抗后,取出放入1×洗膜液,洗五次,每次 5 min;
10)显影液混合液覆盖住PVDF膜,覆盖上一层孵育膜,放入显影仪进行显影。
利用Western-blot技术检测了上述8个单克隆细胞CAS9在蛋白水平的表达情况。结果如图3所示,单克隆细胞除单克隆3号和单克隆6号细胞外CAS9在蛋白水平均有表达。
实施例2. CRISPR-gRNA文库转染稳定表达Cas9 HUVEC细胞后筛选与低氧条件下存活相关基因
本实施例使用CRISPR-gRNA 全人类基因敲除(CRISPR-Pool™ KOUT 文库)文库将全人类基因组的gRNA通过慢病毒包装的质粒载体转入和大规模感染人脐静脉血管内皮细胞(HUVEC细胞),通过外界不同条件的压力筛选,以达到定点敲除该gRNA对应基因的目的,进而在全基因组范围内筛选血管内皮细胞与缺氧相关的基因。
1. 慢病毒包装的CRISPR-gRNA文库转染稳定表达Cas9的 HUVEC细胞
在低氧环境(37℃、5% CO2、1% O2)和常规常氧(37℃、5%CO2)培养条件下共同培养HUVEC细胞和稳定表达CAS9的HUVEC细胞(Cas9细胞),并在低氧下观察低氧24h和48h的细胞生长状态。如图4所示,两个细胞株在低氧下生长状态均变差,随着培养时间的延长,细胞出现大量死亡。
用慢病毒包装的CRISPR-gRNA全人类基因组文库感染稳定表达CAS9的单克隆细胞,并在低氧条件下培养,筛选出在低氧下能正常生长的单克隆细胞。具体步骤如下:
(1)根据CRISPR-gRNA全人类基因组文库慢病毒中所包含的gRNA数量来培养所需细胞数,A文库含63950个sgRNA,B文库含56869个sgRNA,使用15cm细胞培养皿进行细胞的培养,共培养30皿细胞;
(2)根据文库慢病毒的病毒滴度,向10mL细胞培养基中加入10μL的慢病毒A文库和40μL的病毒转染增强剂,于37℃、5%CO2、1%O2低氧环境下培养细胞6h;
(3)更换新鲜ECM培养基,于37℃、5% CO2、1% O2低氧环境下培养细胞12h;
(4)更换含puromycin浓度为2 μg/mL的ECM培养基,于低氧环境下继续培养进行压力筛选;
(5)观察细胞并及时更换含puromycin的新鲜培养基;
(6)观察到有细胞存活并正常生长,使用克隆环挑选出存活的单克隆细胞,并进行扩大培养,结果筛选到三个细胞克隆。
2. 低氧相关基因的筛选
(1)对上述单克隆细胞进行基因组的提取;利用文库通用引物sgRNA-F以及sgRNA-R扩增。

三个克隆都扩增得到300bp的片段(图5),将扩增获得的DNA片段进行切胶回收,进行TA克隆后利用测序引物M13进行测序,只得到两条gRNA序列,序列分别为TCCCGTCGAAAGTTAGCACG和CAGTCTACAAGTGCATTAAG(图6),将测序结果与质粒序列进行比对,可以得到转入的gRNA序列,再根据与数据库的比对得到gRNA所对应的基因;与人类基因组数据库比对这两条gRNA分别且唯一对应的基因为PLLP和WEE2。
(2)根据比对得到的gRNA和基因再次进行基因组PLLP基因的特异性扩增,所使用的扩增序列如表3所示:

扩增后得到的产物进行高通量测序,测序结果经过和PLLP基因组序列(Homosapiens plasmolipin (PLLP), mRNA NM_015993.3)比对,得到gRNA所引导Cas9发挥作用的位置,并判断产生作用的碱基发生突变是否为有义突变。
将上述PLLP的gRNA构建到Lenti-V2表达质粒上,委托合元生物进行慢病病毒包装,滴度为1×108。将上述慢病毒感染HUVEC细胞,进行药物筛选挑取单克隆。将上述对已验证转入PLLP gRNA的单克隆细胞再次进行验证,提取基因组后,在gRNA附近设计引物PLLP-F以及PLLP-R,对提取到单克隆细胞的基因组进行扩增,扩增片段进行TA克隆后进行测序。通过测序检测gRNA引导Cas9产生切割作用是否为有效切割。因此PLLP的gRNA确定转入细胞并在基因组水平产生作用,测序结果如图7所示,PLLP-gRNA引导Cas9产生切割的区域是编码区,且产生以下突变类型(1)从突变密码子可以看出是由丝氨酸突变为天冬氨酸(2)插入(3)缺失(图7)。
实施例3. 敲低PLLP的HUVEC细胞的制备
1.根据实施例2的方法,将靶向PLLP的gRNA构建到Lenti-V2表达质粒上进行慢病病毒包装,滴度为1×108。将上述慢病毒感染HUVEC细胞进行药物筛选,获得PLLP被敲低的HUVEC细胞。
2.利用qRT-PCR检测敲低PLLP的HUVEC细胞的PLLP表达量
(1)提取上述已转入PLLP gRNA单克隆细胞的总RNA以及全蛋白;
(2)反转录PCR:25℃10min, 37℃120min,85℃5min,4℃保存。
(3)qRT-PCR:预变性 95℃3min ,变性95℃ 10s,退火和延伸60℃ 30s,循环40次。
每个样品设置3个复孔,以GAPDH作为内参基因。qPCR反应结束后,每个样品中目的基因三个复孔的平均Ct值作为最终结果。各目的基因的相对表达量计算公式为2^-△Ct,其中△Ct= Ct(目的基因)- Ct(内参基因)。

如图8所示,利用PLLP gRNA可以显著敲低PLLP基因的表达(图8中PLLP gRNA,P<0.0001),使用以上细胞进行功能以及表型的验证。
3. Western-blot检测敲低PLLP的HUVEC细胞的的PLLP蛋白表达水平
为了进一步检测PLLP gRNA的功能,利用PLLP抗体(abcam公司,ab236668)进行Western-blot检测转入PLLP-gRNA的单克隆细胞的PLLP蛋白表达水平,结果如图9所示,所有转入PLLP-gRNA的单克隆细胞PLLP表达水平显著敲低,说明PLLP gRNA在细胞内产生有效切割,导致PLLP的蛋白水平显著下调。由于6号克隆的PLLP蛋白表达水平几乎为0,选取6号克隆进行后续的功能实验。
实施例4. PLLP过表达HUVEC细胞的构建及检测
1. PLLP过表达慢病毒结构信息和构建方法(苏州吉玛基因有限公司)
(1)制备编码慢病毒颗粒的重组病毒质粒PLLP-过表达质粒(质粒图谱如图10所示)及其三种辅助包装原件载体质粒(PG-P1-VSVG,PG-P2-PEV和PG-P3-PRE),其中,PLLP的编码基因如SEQ ID NO.1 所示,对照质粒为慢病毒过表达质粒载体的空载体,分别进行高纯度无内毒素抽提。
(2)用该公司的转染试剂RNAi-Mate共转染293T细胞,转染6h后更换为完全培养基。
(3)培养72h后,收集富含慢病毒颗粒的细胞上清液,对其浓缩,得到高浓度的慢病毒浓缩液,用于感染目的细胞。
2. 利用慢病毒感染细胞构建过表达PLLP的HUVEC细胞
(1)在37℃、5%CO2条件下,使用ECM培养基培养HUVEC细胞;
(2)在六孔板中接种细胞,待细胞生长密度达50%时进行PLLP过表达慢病毒和对照慢病毒转染;
(3)向2mL细胞培养基中加入20μL的慢病毒和2μL的病毒转染增强剂,于37℃、5%CO2条件下培养细胞6h;
(4)弃去含病毒的培养液,更换新鲜ECM培养基,于37℃、5%CO2条件下培养细胞12h;
(5)更换含2μg/mL puromycin的ECM培养基,于37℃、5%CO2条件下继续培养细胞;
(6)观察细胞,并及时更换含puromycin的新鲜培养基。
3. 利用qRT-PCR检测过表达PLLP的HUVEC细胞的PLLP表达量
方法同实施例3。结果如图8所示,利用PLLP过表达的慢病毒感染细胞后HUVEC细胞的PLLP表达量较对照细胞显著升高,上调89.25倍(图8中PLLP过表达,P<0.001),使用以上细胞进行功能以及表型的验证。
4. Western-blot检测过表达PLLP的HUVEC细胞的的PLLP蛋白表达水平
方法同实施例3。结果如图11所示,相较于对照细胞,PLLP的蛋白表达水平显著升高PLLP表达量较对照细胞显著升高。
实施例5. PLLP基因敲低和过表达对细胞功能的影响
1. 细胞增殖检测
为了检测PLLP基因对细胞增殖的影响,我们分设了常氧和低氧两个平行条件,利用EdU方法检测敲除PLLP以及过表达PLLP细胞的增殖率。
(1)实验步骤
1)使用BeyoClickTMEdU-594细胞增殖检测试剂盒进行细胞增殖的检测;
2)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞分别在常氧下、低氧下在EdU小室处理24h;
3)将1×EdU 工作液和细胞培养基以1:1000的比例配置,将小室中的培养基更换为含EdU工作液的细胞培养基混合液,在37℃、5%O2条件下孵育2h;
4)去除培养基,加1mL含4%多聚甲醛的PBS固定液,室温固定15min;
5)去除固定液,加1mL PBS洗涤细胞,洗3次,每次5min;
6)去除洗涤液,加1mL含0.3%Triton X-100的PBS通透液,室温孵育15min;
7)去除通透液,加1mL PBS洗涤细胞,洗2次,每次5min;
8)配置Click反应液混合液,混合液含Click Reaction Buffer 1.72mL,CuSO2 80μL,Azide-526 4μL,Click Additive Solution 200μL,混合液共2mL;
9)去除洗涤液,加入1mL Click反应混合液,轻轻摇晃培养板以确保反应混合物可以覆盖样品;
10)室温避光孵育30min;
11)去Click反应混合液,加1mL PBS洗涤细胞,洗3次,每次5min;
12)滴加一滴DAPI试剂进行染色,盖上盖玻片,在荧光显微镜下拍照。
(2)结果
在常氧条件下,PLLP敲除及过表达细胞与对照细胞相比,细胞增殖率没有显著性变化,但是在低氧条件下,PLLP敲除细胞的增殖率为对照细胞的2.98倍(P<0.001,如图12所示),而PLLP过表达的细胞几乎完全死亡,无法统计增殖率(图12中未显示),上述结果说明,在常氧条件下,PLLP不影响细胞的增殖,而在低氧条件下则影响细胞的增殖,尤其是低氧条件下过表达PLLP会导致细胞的死亡。
2. 细胞活力检测
Cell Titer-Glo(CTG)是基于ATP检测的快速细胞活力检测法,是公认的高灵敏度发光检测法,是细胞活力检测的金标准。为了检测PLLP基因对HUVEC细胞活力的影响,我们利用CTG发光法检测了PLLP敲除和过表达HUVEC细胞的活力。
(1)具体步骤
1)使用CellTiter-Glo®Luminescent Cell Viability Assay试剂盒进行细胞活力的检测;
2)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞分别以2×104的细胞密度于常氧下、低氧下在96孔板培养,培养时间为0h,6h,12h,18h,24h,每个细胞设置3个复孔,每个孔加入100μL培养基进行培养,并设置单培养基的空白组Blank;
3)根据培养时间设置的不同,分别在培养0h、6h、12h、18h、24h后加入100μLCellTiter-Glo®Reagent;
4)室温摇床摇晃2min;
5)室温孵育10min;
6)分光光度计检测全波长的发光;
7)结果计算=实验组检测到全波长值-Blank组全波长发光值。
(2)结果
以全波长的发光度作为细胞活力高低的标准,在常氧条件下敲除PLLP和过表达PLLP的细胞与对照细胞相比,细胞活力没有显著性变化,但是在低氧条件下,PLLP敲除细胞的活力为对照细胞的1.66倍,明显高于对照细胞(P<0.001,如图13所示,PLLP-gRNA),而PLLP过表达细胞的活力为对照细胞的0.48倍,明显低于对照细胞(P<0.001,如图14所示),上述结果说明,在常氧条件下,PLLP与细胞活力无关,而在低氧条件下,PLLP可以显著影响细胞的活力。
3. 细胞迁移力检测
为了检测PLLP基因对细胞的迁移情况,我们分别在常氧和低氧条件下,对HUVEC细胞、Cas9细胞和PLLP敲除细胞进行划痕实验和Transwell迁移实验。
(1)划痕实验
1)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞同时进行培养;
2)以2×104的细胞密度于常氧下、低氧下在6孔板过夜培养,使用200μL枪头在6孔板竖直划下一条线;
3)于0h、12h、24h进行拍照;
4)通过Image J计算划痕的面积。
(2)Transwell迁移实验
1)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞同时进行过夜培养;
2)以2×104的细胞密度于常氧下、低氧下接种于8.0μm孔径Transwell小室的上室中,使用200μL无血清培养基接种,下室加入600μL有血清正常培养基;
3)培养12h后,取出小室,弃去培养基,在PBS涮洗3次;
4)将小室下室置于95%乙醇中进行细胞的固定,室温下固定30min;
5)取出小室,在PBS涮洗3次;
6)将小室下室置于0.1%结晶紫中进行细胞的染色,室温下染色30min;
7)取出小室,在PBS涮洗3次;
8)棉签擦去上室未迁移的细胞,在显微镜下拍照计数。
(3)结果
从划痕实验的结果发现,将划痕的愈合面积作为迁移速率高低的参照,在常氧条件下敲除PLLP或过表达PLLP对细胞的迁移无影响,但是在低氧条件下,PLLP敲除细胞的迁移速率为对照细胞的2.56倍,显著高于对照细胞(P<0.001,如图15所示),PLLP过表达细胞的迁移率明显为对照细胞的54%,显著低于对照细胞(P<0.001,如图16所示)。从Transwell迁移实验的结果同样能够得出这一结论(如图17-18所示)。上述结果说明PLLP在常氧条件下与细胞迁移没有相关性,而在低氧条件下会显著影响细胞的迁移。
4. 细胞侵袭力检测
为验证PLLP基因对HUVEC细胞侵袭的影响,设定了常氧和低氧两个条件,进行transwell侵袭实验。
(1)具体步骤
1)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞同时进行过夜培养;
2)提前一天于4℃预冷所需枪头、培养基、PBS和Transwell小室,将Matrigel胶从-20℃取出置于4℃融化;
3)将Matrigel胶使用无血清培养基以1:10的比例稀释,于冰上操作;
4)在小室的上室中央加入100μL稀释后的Matrigel胶,于37℃放置5h使胶凝固;
5)取出小室,小心吸去上室未凝固的胶液;
6)以2×104的细胞密度于常氧下、低氧下接种于小室的上室中,使用200μL无血清培养基接种,下室加入600μL有血清正常培养基;
7)培养24h后,取出小室,弃去培养基,在PBS涮洗3次;
8)将小室下室置于95%乙醇中进行细胞的固定,室温下固定30min;
9)取出小室,在PBS涮洗3次;
10)将小室下室置于0.1%结晶紫中进行细胞的染色,室温下染色30min;
11)取出小室,在PBS涮洗3次;
12)棉签擦去上室未侵袭的细胞,在显微镜下拍照计数。
(2)结果
结果发现,在常氧条件敲除PLLP或过表达PLLP对细胞的侵袭无影响,但是在低氧条件下,敲除PLLP的HUVEC细胞侵袭能力为对照细胞的1.52倍,明显高于对照细胞(P<0.001,如图19所示),而过表达PLLP的细胞侵袭能力为对照细胞的45%,明显低于对照细胞(P<0.001,如图20所示)。该结果说明,在常氧条件下,PLLP对细胞侵袭没有影响,当时在低氧条件下PLLP可以显著影响细胞的侵袭能力。
5. 细胞电阻率检测
内皮细胞的一个重要特征就是紧密连接,为探索 PLLP 在低氧条件下对细胞紧密连,我们通过测定细胞膜间电阻来反映细胞紧密连接程度,在常氧与低氧下分别培养细胞24 h、48 h、72h和 96 h,并在这些时间点检测细胞膜电阻率。
(1)具体步骤
1)将细胞接种到 4μm 孔径的小室中,以 2ⅹ104 的密度进行共培养,并将细胞培养 在上室和下室的相同培养基中;
2)分别在 24h、48h、72h 和 96h 用 Millicell®ERS-2(美国 Millipore)测量电池电阻率;
3)测量值为电阻值,称为 R。使用以下公式计算单位面积电阻:单位面积电阻=Rⅹ有效膜面积。
(2)结果
敲除 PLLP 以及过表达PLLP后HUVEC细胞在常氧下的电阻率与对照细胞相比没有显著性差异,说明在常氧条件下PLLP不影响细胞的紧密连接;而在低氧条件下,PLLP 敲除细胞的细胞膜电阻率为对照细胞的1.68倍,较对照细胞显著升高(P<0.001,如图21所示),而过表达PLLP的细胞的细胞膜电阻率为对照细胞的50%,较对照细胞显著降低(P<0.001,如图22所示),说明在低氧条件下PLLP敲除后细胞紧密连接的能力增强,而过表达PLLP后细胞紧密连接能力显著降低。
6. 细胞凋亡实验
为验证PLLP基因敲除以及过表达后HUVEC细胞在常氧和低氧下的凋亡情况,我们分设了常氧和低氧两个条件,利用流式细胞仪检测对照细胞、PLLP敲除细胞以及PLLP过表达细胞的凋亡情况。
(1)具体步骤:
1)使用Annexin V-FITC细胞凋亡检测试剂盒进行凋亡实验;
2)将HUVEC细胞、Cas9细胞、PLLP-gRNA细胞分别在常氧和低氧下同时培养0h、24h、48h和72h;
3)分别在0h、24h、48h和72h将细胞培养基吸取置于EP管中,把细胞消化下来,与原培养基一起在室温下1500rpm离心5min;
4)去上清,加入1mL PBS重悬沉淀,室温下1500rpm离心5min;
5)去上清,加入200μL FITC结合液重悬沉淀;
6)加入10μL PI试剂;
7)加入5μL FITC试剂;
8)在冰上避光放置10min;
9)在Thermo流式细胞仪上进行凋亡的检测;
10)使用Flowjo进行数据和图片的分析;
(2)结果
在常氧条件下HUVEC细胞敲除PLLP或过表达PLLP后与对照细胞相比较,细胞的凋亡没有显著性差异;在低氧条件下,对照细胞的细胞凋亡率相对于常氧条件下显著上升(P<0.001),但PLLP敲除细胞的凋亡率为对照细胞的62%,显著低于对照细胞(P<0.001,如图23所示),且与常氧条件下的细胞凋亡率没有显著性差异,说明在低氧条件下,敲除PLLP后能够抑制细胞凋亡。PLLP过表达时,HUVEC细胞在常氧条件细胞凋亡率与对照细胞无显著仙差异,但是在低氧条件下,PLLP过表达细胞的凋亡率为对照细胞的2.32倍,显著高于对照细胞(P<0.001,如图24所示),表明PLLP在缺氧条件下可以加剧细胞凋亡的发生。
7. 细胞成管实验
为了验证PLLP敲除以及过表达后的HUVEC细胞在常氧和低氧下对血管形成的影响,我们分设了常氧与低氧两个条件,通过统计分支数、成网数、节点数、支线总长度和成网总面积数考察血管形成能力。
(1)具体步骤
1)准备将胶铺到24孔板中,将2个孔板提前放到-20℃预冷低温处理,将新鲜的无菌EP管放到-20℃预冷低温处理,将分装的底胶于-4℃冻融。
2)进行铺胶,每孔300μL底胶稀释液,用无血清空白DMEM培养基以1:1的比例稀释。
3)取出预冷的EP管、底胶置于冰盒上,取出预冷的枪头,用DMEM空白培养基以1:1的比例稀释底胶,混匀,取出2个预冷的24孔板,迅速加入底胶稀释液,每孔300μL,垂直加入孔中央,慢慢摇晃孔板使得底胶铺满孔底,有大泡泡,拿针筒戳破,小泡泡敲击板低把气泡震出。
4)将铺好的孔板放到37℃培养箱中凝固1h。
5)将培养的细胞用胰酶消化下来,分别用含10%FBS的DMEM培养基重悬 ,细胞计数仪计数,在24孔板中每孔加入300μL的细胞悬液,分别加两个板子,保持9×104个细胞/孔。垂直加入细胞悬液,然后晃匀,四面敲击孔板,尽量让细胞能均匀铺在孔板。两个培养板分别在常氧下和低氧下培养。在37℃培养箱中孵育5h,可见管腔形成,显微镜下观察拍照。
(2)结果
如图25和图26结果所示,敲除PLLP细胞在常氧条件下的分支总长度以及网格总面积为对照细胞的1.28倍(P<0.05)和1.42倍(P<0.05),高于对照细胞,其余指标与对照细胞相比无显著性差异,但是在缺氧条件下,敲除PLLP细胞的成管节点数、网格数、血管分支数、网格总面积分别为对照细胞的2.36倍(P<0.001)、2.94倍(P<0.001)、1.90倍(P<0.001)和7.89倍(P<0.001),说明敲除PLLP细胞在低氧条件下其成管能力显著高于对照细胞的成管能力。而过表达PLLP时HUVEC 细胞在常氧条件下网格数、分支数以及网格总面积分别为对照细胞的82%(P<0.05)、1.12倍(P<0.05)和85%倍(P<0.05),但在低氧条件下,除了血管分支数为对照细胞的86%(P<0.05)外,成管的节点数、网格数、分支总长度以及网格总面积分别为对照细胞的44%(P<0.001)、22%(P<0.001)、32%(P<0.001)和5%,较对照细胞显著下降。以上结果说明,PLLP在常氧条件下不影响血管形成能力,但是在低氧条件下,可以影响血管形成能力,PLLP的过表达会导致血管形成能力缺失。

1.以PLLP基因为靶点的分子在制备肿瘤疾病治疗药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述以PLLP基因为靶点的分子为促进PLLP表达的分子。
3.根据权利要求2所述的应用,其特征在于,所述促进PLLP表达的分子为含有PLLP编码基因的载体。
4.根据权利要求3所述的应用,其特征在于,所述载体为慢病毒载体。
5. 根据权利要求4所述的应用,其特征在于,所述PLLP编码基因的序列如SEQ ID NO.1所示。
6.根据权利要求1-5任一所述的应用,其特征在于,所述肿瘤疾病治疗药物为抑制肿瘤细胞增殖和/或促进其凋亡的药物。
7.根据权利要求1-5任一所述的应用,其特征在于,所述肿瘤疾病治疗药物为抑制肿瘤细胞迁移和/或侵袭的药物。
8.根据权利要求1-5任一所述的应用,其特征在于,所述肿瘤疾病治疗药物为抑制肿瘤细胞紧密连接能力的药物。
