背景技术
肿瘤是机体正常细胞恶变的产物,其特点是不断增殖并在体内转移。因此肿瘤细胞在免疫学上的突出特点是出现某些在同类正常细胞中看不到的新的抗原标志。
自然杀伤细胞(natural killer cell,NK)是机体重要的免疫细胞,不仅与抗肿瘤、抗病毒感染和免疫调节有关,而且在某些情况下参与超敏反应和自身免疫性疾病的发生,能够识别靶细胞、杀伤介质。由于NK细胞具有部分T细胞分化抗原,如80~90%NK细胞CD2+,20~30%NK细胞CD3+(表达CD3ζ链),30%NK细胞CD8+(α/α)和75~90%NK细胞CD38+,而且NK细胞具有IL-2中亲和性受体,在IL-2刺激下可发生增殖反应,活化NK细胞可产生IFN-γ;因此一般认为NK细胞与T细胞在发育上关系更为密切。IL-2、IL-12、IFN-α、TNF-α以及白细胞调节素(leukoregulin,LR)对NK细胞的活化和分化有正调节作用,体外培养时加入上述细胞因子可明显提高NK的杀伤活性。
γ干扰素(IFN-γ)主要由T细胞、NK细胞和NKT细胞产生。IFN-γ与其细胞表面受体IFN-γRα结合诱导IFN-γRα二聚,从而形成与IFN-γRβ组装的位点。当IFN-γRα/IFN-γRβ异构化后,其细胞内janus家族激酶JAK1和JAK2分别二聚并磷酸化。这种磷酸化创造了信号转导和转录激活蛋白(STAT),主要是STAT1的结合位点。IFN-γ直接通过诱导凋亡或促进非凋亡细胞死亡,以及间接地通过使肿瘤细胞易受诱导凋亡的免疫应答或化疗来发挥其杀伤作用。虽然IFN-γ/STAT1途径诱导肿瘤细胞凋亡,但激活STAT1也能抑制肿瘤细胞的生长并建立休眠状态。
白细胞介素-2(IL-2)是趋化因子家族的一种细胞因子。它是由多细胞来源(主要由活化T细胞产生),又具有多向性作用的细胞因子(主要促进淋巴细胞生长、增殖、分化);对机体的免疫应答和抗病毒感染等有重要作用,能刺激已被特异性抗原或致丝裂因数启动的T细胞增殖;能活化T细胞,促进细胞因子产生;刺激NK细胞增殖,增强NK杀伤活性及产生细胞因子,诱导LAK细胞产生;促进B细胞增殖和分泌抗体;激活巨噬细胞。
白细胞介素-4(IL-4)是II型辅助T细胞(Th2细胞)分泌的细胞因子。白细胞介素-4的生物作用,包括刺激活化B细胞和T细胞增殖、CD4+ T细胞分化成II型辅助T细胞;它也在调节体液免疫和适应性免疫中起关键作用。内源性IL-4作用于CD4+ T细胞,诱导并稳定其Th2状态。IL-4还下调CD8+ T细胞中CD8,穿孔素和颗粒酶A,B和C的表达,并使这些细胞具有较弱的细胞溶解表型。此外,肿瘤细胞通常具有增加的IL-4R表达。IL-4与其在肿瘤细胞上的受体的结合增加了抗凋亡分子(例如Bcl-xL和cFLIP)的水平。内源性IL-4的这些多重作用共同导致免疫偏离和肿瘤扩大。
白细胞介素10(IL-10)是一种多功能负性调节因子,主要由辅助性T细胞2(Th2)、活化的B细胞、单核细胞、巨噬细胞产生。IL-10是某些肿瘤细胞的生长因子。大量研究已证实许多恶性肿瘤组织或细胞均可产生IL-10,如恶性黑色素瘤、结直肠癌、卵巢癌、肺癌、脑胶质瘤等。IL-10还可能与肿瘤转移有关。
因此,提供一种CMU-Pb-L5在制备提高肿瘤患者免疫力药物中的应用是本领域技术人员亟需解决的问题。
发明内容
有鉴于此,本发明提供了一种CMU-Pb-L5在制备提高肿瘤患者免疫力药物中的应用。
为了实现上述目的,本发明采用如下技术方案:
一种CMU-Pb-L5,其保藏编号为CGMCC No.25653,已保藏于中国微生物菌种保藏管理委员会普通微生物中心,简称CGMCC,地址北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,保藏日期为2022年09月05日,分类命名为副干酪乳杆菌Lactobacillusparacasei。
CMU-Pb-L5(副干酪乳杆菌,Lactobacillus paracasei)从健康人体粪便分离得到,具有较强生物被膜形成能力,能耐受胃肠液,在肠道内发挥作用,且摄入安全,不会对小鼠产生毒害作用,同时可显著抑制小鼠结肠炎模型中促炎细胞因子的表达,进而调控Th17/Tregs细胞平衡,改善结肠炎小鼠的症状和病理损伤。
进一步,所述的一种CMU-Pb-L5在制备提高肿瘤患者免疫力药物中的应用。
进一步,所述肿瘤为结直肠癌。
进一步,所述CMU-Pb-L5为菌悬液。
经由上述的技术方案可知,与现有技术相比,本发明公开提供了一种CMU-Pb-L5在制备提高肿瘤患者免疫力药物中的应用,有益效果如下:
CMU-Pb-L5对小鼠血清细胞因子的影响:与非干预对照组相比,CMU-Pb-L5干预组IFN-γ和IL-2含量显著增加(P<0.01),IL-4和IL-10含量显著降低(P<0.05),炎症相关细胞因子IL-6、IL-1β、TNF-α和IL-17A含量显著降低(P<0.01)。

具体实施方式
下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
5周龄SPF级雄性Balb/c小鼠(体重约18-20g),购自南方医科大学实验动物中心。
统计学方法:采用GraphPad 9.5软件进行统计学分析,计量资料用均值±标准误差(Mean±SEM)表示,两组间差异用独立样本Student’s t检验,多组间差异用ANOVA进行检验,P值小于0.05为差异有统计学意义。
实施例1
(1)菌株复苏及菌落形态观察
从-80℃冰箱内取出实验室分离的CMU-Pb-L5,于37℃恒温水浴锅快速震荡;待菌株融化后,用一次性接种环取适量菌液接种到MRS琼脂平板中,置于37℃厌氧工作站中(O2浓度为0%,湿度60%)倒置培养24h,接着挑取平板上的单个菌落,用四区划线法将其转种至新的MRS琼脂平板上,37℃厌氧工作站中培养48h,观察菌落形态;随后挑取单菌落,均匀涂布于滴有无菌生理盐水的玻片上,待玻片干燥后,过酒精灯外焰进行固定,玻片冷却后,滴加龙胆紫染料染色10s,之后水洗甩干。接着滴加碘液染色10s,之后水洗,甩干。随后用脱色液95%酒精(购自珠海贝索生物技术有限公司)脱色10s,之后水洗,甩干水分。最后滴加沙黄溶液(主要成分为品红、沙黄和乙醇组成,购自珠海贝索生物技术有限公司),复染10s,之后水洗,用滤纸吸干,置于x1000油镜下观察。结果主要说明:革兰染色结果显示CMU-Pb-L5为菌体紫色、杆状、单个或成对短链排列的革兰氏阳性菌。
(2)CMU-Pb-L5菌悬液的制备
用灭菌的枪头或牙签挑取平板上CMU-Pb-L5单菌落,置于装有1mL MRS培养液的2mL的微型离心管里,混匀并做好标记,将离心管置于37℃恒温摇床培养18h。取100μL培养18h的CMU-Pb-L5菌液转移至另一个含900μL MRS培养液的微型离心管中培养18h,重复此操作两次以活化菌株。活化后的CMU-Pb-L5以2%(v/v)比例加到无菌MRS肉汤培养基中,37℃120r/min恒温培养18h后,4000r/min离心5min,用PBS洗3次后,再用无菌生理盐水调整菌液浓度,用超微量核酸蛋白测定仪测定菌液OD600,调整菌液浓度为1.0×109CFU/mL。
实施例2
不同细胞在肿瘤环境中产生的细胞因子在癌症发病过程中具有重要作用。在感染、炎症和免疫等情况下释放的细胞因子可以起到抑制癌症发展和恶化的作用。大量的临床研究发现,癌症患者的细胞因子有以下几种,包括IL-2、IL-6、IL-8、IL-10、IL-12、IL-18、TNF-α、TGF-β、IFN-γ、巨噬细胞迁移抑制因子(MIF)和CXCR4等。
IL-2主要来源于CD4+T细胞,为抗原刺激T淋巴细胞的生长因子,在适应性免疫中负责抗原识别后的T细胞克隆扩张。IL-2是对免疫系统具有多效性的重要细胞因子之一。癌症的进展或预后不良与IL-2浓度降低或可溶性IL-2受体浓度增加有关。
IL-6由许多肿瘤产生,是一种增殖能力非常强的细胞因子,在恶性肿瘤的发病过程中起重要作用。高浓度的IL-6在癌症中被发现,且往往表达失调。众多报道表明,IL-6在多种类型的肿瘤中过度表达。肿瘤微环境中IL-6水平高,说明炎症与癌症之间的联系密切。IL-6通过调节癌症相关标志物和多种信号通路,影响细胞凋亡、生存、增殖、血管生成、侵袭性和转移等,促进肿瘤的发生。
IL-10具有高度的免疫抑制作用,在肿瘤细胞中常高表达,所以大多数人类癌症中循环血清中IL-10的浓度高。
IFN-γ具有一系列的生物学功能:抗病毒作用,抑制肿瘤细胞生长,促进B细胞产生抗体。此外,IFNγ活化巨噬细胞,增强NK细胞的细胞毒作用,刺激T细胞的细胞毒作用。
1)CT26小鼠结直肠癌模型建立以及菌株干预
(1)菌株预干预:18只SPF级雄性Balb/c小鼠喂养一周后,分为以下3组:非干预对照组(PBS,Control组)、CMU-Pb-L5干预组和5-氟尿嘧啶阳性药物对照组(5-Fu组),每组6只。每日给CMU-Pb-L5干预组小鼠灌胃200μL CMU-Pb-L5(1×109CFU/mL),非干预对照组和5-Fu组小鼠灌胃相同体积的PBS溶液。
(2)小鼠皮下接种CT26鼠源性结肠癌细胞
①CT26细胞培养:CT26细胞用完全DMEM培养基(含10%FBS,1%双抗)复苏后,在10cm细胞培养皿按常规细胞培养方法进行扩大培养。待生长至对数生长期,消化离心细胞,用含基质胶的PBS溶液调整细胞浓度为5×106个/mL。接种前注意观察CT26细胞的生长情况,保证接种细胞活力旺盛。
②CMU-Pb-L5干预两周后,非干预对照组、CMU-Pb-L5和5-Fu组于每只小鼠右腋下部位单次皮下注射100μL CT26细胞悬液(5×105个细胞),注射前常规消毒注射部位皮肤。接种结束后,每日查看肿瘤的生长状况,待其长至2~5mm时,CMU-Pb-L5干预组每日继续灌胃200μL CMU-Pb-L5(1×109CFU/mL)菌液,连续干预20d;5-Fu组25mg/kg剂量腹腔注射给药,每3d一次。(5-Fu用生理盐水配置,现配现用),非干预对照组每日灌胃等体积的PBS溶液。
实验结束后,收集小鼠粪便,采用眼球取血法收集各组小鼠血液。
实施例3液相芯片法检测血清细胞因子表达情况
(1)样品与标准品准备
①血清10000rpm离心10min,用样本稀释液将上清进行4倍稀释,取50μL稀释好的溶液上样检测。
②500μL标准品稀释液稀释标准品,混匀后,按照图1梯度稀释标准品。
(2)芯片检测操作
①样品孵育:用配套稀释液稀释微珠后以1400rpm,振荡30s;随后50μL/孔加入96孔板中,洗板机洗涤3次;在96孔板中加入50μL标准品、样品和Blank,封口膜封住,以850rpm室温避光振荡孵育30min。
②抗体检测:弃去样品,使用洗板机洗涤3次;使用Antibody Diluent按说明书要求稀释Detection Antibody;25μL/孔加入稀释好溶液,封口膜封住,以850rpm室温避光振荡孵育30min。
③显色:弃去检测抗体,洗涤3次;用稀释液稀释Streptavidin-PE;50μL/孔加入稀释好的Streptavidin-PE,贴上封口膜,封口膜封住,以850rpm室温避光振荡孵育10min;随后洗涤3次;125μL/孔加入Assay Buffer,封口膜封住,以850rpm室温避光振荡孵育2min;送入已校正的Bio-Plex机器中读值。
1)CMU-Pb-L5对小鼠血清免疫相关细胞因子的影响:
干扰素γ(IFN-γ)是与抗肿瘤免疫相关的细胞因子,其具有抑制肿瘤细胞生长、促进肿瘤细胞凋亡和增强细胞毒性T细胞杀伤能力的作用,其在抗肿瘤免疫方面发挥着核心作用,CMU-Pb-L5干预组IFN-γ的含量显著高于其余各组(P<0.01)(图2A),说明CMU-Pb-L5干预后可提高荷瘤小鼠体内IFN-γ含量,从而发挥抗肿瘤免疫作用。IL-2在CD4+T细胞分化过程中关键作用,可促进CD8+T细胞和NK细胞活性,在体内发挥抗肿瘤作用。与非干预对照组相比,CMU-Pb-L5干预组IL-2含量显著上升,差异具有统计学意义(P<0.01)(图2B)。肿瘤微环境中存在免疫抑制性细胞因子,这些细胞因子具有免疫抑制能力,可促进癌细胞的存活和增殖,最终导致肿瘤发生发展,IL-10和IL-4已被证实为有助于肿瘤生长和转移的免疫抑制性细胞因子。与非干预对照组相比,CMU-Pb-L5干预组IL-4水平显著降低(P<0.05)(图2C),IL-10水平显著降低(P<0.05)(图2D)。
2)CMU-Pb-L5对小鼠血清炎性细胞因子的影响
与非干预对照组相比,CMU-Pb-L5干预组炎性细胞因子IL-6、TNF-α、IL-1β和IL-17A含量显著下降(P<0.05)(图3)。
