Abstract
本发明公开了一种猪脾汤口服液的制备工艺,属于口服液制备技术领域,包括以下步骤:步骤一、取健康猪的脾脏,用纯化水洗净,去膜、筋及脂肪组织,破碎匀浆,透析,上清液过滤除菌;步骤二、取山楂洗净,加纯化水50‑55℃浸泡,过滤,滤液加低聚果糖混合后灭菌;步骤三、合并上述两液;步骤四、灌封,包装,检验合格即得。通过采用健康猪脾脏透析液和山楂提取液制成双调因子口服液,口服安全方便。其具有较好的提高人体免疫力作用,提高了猪脾和山楂的综合利用价值,具较好的社会效益和经济效益。

技术领域
本发明涉及口服液制备技术领域,具体为一种猪脾汤口服液的制备工艺。
背景技术
转移因子(Transfer factor)口服液系用健康猪脾脏为原料,制成的含多肽氨基酸和多核苷酸混合物的溶液,高效安全。转移因子口服液是一种免疫制剂,主要提高人体免疫力,达到增强健康、预防疾病的目的。
山楂(Crataegus pinnatifida Bunge)蔷薇科山楂属落叶乔木植物。抗衰老作用位居群果之首。《本草图经》、《本草再新》等书籍中记录山楂有消食健胃,行气散瘀等作用,也可医治泻痢腹痛、瘀血经闭、产后瘀阻、心腹刺痛、疝气疼痛、高血脂症。

发明内容
本发明的目的在于克服上述背景技术困难,提供一种双调因子口服液配方,并且提高了猪脾脏的提取效率,使其药效和营养更佳,而提供一种猪脾汤口服液的制备工艺。
为达到上述目的,采用的技术方案为:一种猪脾汤口服液的制备工艺,包括以下步骤:
步骤一、取山楂洗净,破碎后加纯化水,温度50℃-55℃浸泡4小时,过滤,提取液加入山楂3倍质量的低聚果糖,混合,灭菌;
步骤二、取猪脾脏,洗净,去膜、筋及脂肪组织,破碎匀浆后得到猪脾浆,猪脾浆在温度4-10℃进行透析过滤,收集透析液,灭菌过滤,收集滤液,备用;
步骤三、将步骤一和步骤二收集的液体等比例混合均匀,静置沉淀,过滤取上清液;
步骤四、将步骤三混合液灌封,包装,检验合格即得。
进一步的,将步骤一中过滤后的山楂渣,加食醋、白酒和适量水搅拌,发酵7-10天,然后过滤获取发酵滤液;再将发酵滤液添加到步骤二的猪脾浆中混合后,4-10℃保存15-24小时,再进行透析过滤。
进一步的,所述步骤一中,山楂和纯化水的质量比为1:8。
进一步的,所述山楂渣、食醋和白酒重量比为28-32:4-6:2-4。

通过采用本技术方案,一方面使山楂渣得到有效的再利用,增加了猪脾口服液产品有效的营养成分,另一方面更进一步的对提高猪脾脏多肽的提取率起到了重要作用。本制备工艺通过采用健康猪脾脏透析液和山楂提取液制成的系双调因子口服液,兼具了猪脾脏和山楂药用保健功能。其利用猪脾脏透析液提取的能有效提高人体免疫力的纯生物活性因子,将其中有利于人体吸收的多肽以及多种微量元素,配以具有促进脂肪消化、降血脂等多种功能的山楂水解提取液配制而获得成品。本发明的口服液提高了猪脾和山楂的综合利用价值,具较好的社会效益和经济效益。

实施例一:
如图1所示,一种猪脾汤口服液的制备工艺,包括以下步骤:
步骤一、取山楂洗净,破碎后加纯化水,温度50℃-55℃浸泡4小时,过滤,提取液加入山楂3倍质量的低聚果糖,混合,灭菌。山楂的主要成分为黄酮类及有机酸类化合物。山楂提取物能促进胃中消化酶的分泌,促进消化。所含脂肪酶可促进脂肪的分解消化;含有多种有机酸、维生素C可提高胃蛋白酶活性,促进蛋白的消化。低聚果糖是一种天然活性物质。甜度为蔗糖的0.3-0.6倍的新型甜味剂。在山楂提取液中加入一定比例的低聚果糖可起到调味和调节肠道菌群,增殖双歧杆菌,促进钙的吸收,调节血脂,免疫调节等保健功能。
步骤二、取猪脾脏,洗净,去膜、筋及脂肪组织,破碎匀浆后得到猪脾浆,猪脾浆在温度4-10℃进行加水透析过滤,收集透析液,灭菌过滤,收集滤液,备用。转移因子(Transfer factor)是从动物脾脏提取的具有免疫活性的T淋巴细胞所释放的一种能够转移免疫致敏信息的因子,能够增强动物免疫功能,提高抗感染能力。对于活性多肽的提取,由于多肽来源于蛋白质,故要先使带有活性的蛋白质进行溶解。将猪脾脏进行破碎匀浆,即进行了细胞破碎和固相分离,其为释放猪脾脏内的蛋白质和肽类进入溶液创造了条件。因而采用在温度4-10℃进行加水透析过滤。此温度范围有助于保持多肽物质的生物活性。
步骤三、将步骤一和步骤二收集的液体等比例混合均匀,静置沉淀,过滤取上清液。静置沉淀溶液可形成上清液,因为物质的静置可导致溶液中的各种成分根据颗粒的大小和密度分离出来。可使用只允许液体通过的过滤器滤除固体沉淀物而获取上清液。
步骤四、将步骤三混合液灌封,包装,检验合格即得。口服液是将原材料用水或其他溶剂,采用适当的方法提取,经浓缩制成的内服液体剂型。其特点是:能浸出原材料中的多种有效成分。吸收快,显效迅速。能大批量生产,免去临用煎药的麻烦,应用方便。服用量减小,便于携带、保存和服用。成品经灭菌处理,密封包装,质量稳定,不易变质。
优选的,所述步骤一中,山楂和纯化水的质量比为1:8。
优选的,所述步骤二中,猪脾脏和水的比例为1:10。
现有技术中转移因子口服液临床用于免疫缺陷患者,如细菌性或霉菌性感染、病毒性带状疱疹、乙型肝炎、麻疹、流行性腮腺炎。转移因子口服液是以健康动物脾脏为原料,采用现代生物分离技术提取的纯生物活性因子(转移因子)。山楂含多种有机酸。山楂味酸,助消化,口服后增强胃液酸度,提高胃蛋白酶活性,促进蛋白质的消化。山楂中含脂肪酶,能促进脂肪的消化。山楂含有维生素C等成分,口服可增进食欲。降血脂、抗动脉粥样硬化。山楂黄酮、水解产物增加缺血心肌营养性血流量,其中以山楂水解产物作用最强。山楂在增加冠状动脉血流量的同时,还能降低心肌耗氧量,提高氧利用率。
通过测试对比分析,结果表明本实施例通过采用健康猪脾脏透析液和山楂提取液制成的系双调因子口服液,其兼具了猪脾脏和山楂药用保健功能。其利用猪脾脏透析液提取纯生物活性因子,将其中有利于人体吸收的多肽以及多种微量元素,配以具有促进脂肪消化、降血脂等多种功能的山楂水解提取液配制而获得成品。它富含人体必需的活性多肽、钙、铁、锌、氨基酸、维生素等活性营养素。它通过直接补充和促进饮食中的微量元素的吸收利用的间接补充相结合,全面平衡补充人体营养,取得进一步优化的药用保健效果。
实施例二:
如图1所示,一种猪脾汤口服液的制备工艺,在实施例一的基础上优选的,将步骤一中过滤后的山楂渣,加食醋、白酒和适量水搅拌,发酵7-10天,然后过滤获取发酵滤液;再将发酵滤液添加到步骤二的猪脾浆中混合后,4-10℃保存15-24小时,再进行透析过滤。优选的,所述发酵采用复合乳酸菌,包含双歧杆菌、肠膜明串珠菌和植物乳杆菌,比例1:1:1,接种菌落总数2×107CFU/mL。
将步骤一中煎煮过的山楂渣粉碎,加食醋、白酒和适量水。采用复合乳酸菌发酵工艺,双歧杆菌、肠膜明串珠菌、植物乳杆菌1:1:1,接种菌落总数2×107CFU/mL,搅拌均匀,料堆水泽不外流,手握可滴水即可。发酵7-10天,然后过滤获取发酵滤液。过滤时可适当加压。添加的所述食醋为普通家用食醋,白酒为普通饮用白酒,皆系食品。本实施例中优选的采用食醋酸度为5%,白酒为52度。食醋的主要成分是醋酸,添加白酒可起到助发酵、消毒、杀菌的作用。白酒除自身带有酸性之外,在醋酸菌的作用下会生成醋酸,对快速形成乳酸菌生长条件有帮助。由于食醋和白酒对山楂渣发酵产生了作用。
对于活性多肽的提取,将猪脾脏进行破碎匀浆,即进行了细胞破碎和固相分离。其后就要将破碎的细胞置于溶剂中,使被分离的蛋白质或肽类生物大分子充分释放到溶剂中。脾脏的匀浆和溶解是转移因子提取的关键步骤,可使细胞内的小分子多肽和多核苷酸能完全释放出来。并应保持猪脾脏原来的天然状态,不丢失其生物活性。进一步的,本实施例中发酵滤液添加到猪脾浆中混合后可获得酸性溶液。该酸性溶液条件一方面能保证提取物的活性。同时,在酸性溶液中细胞易发生破膜作用,有利于增加溶液在胞内多肽物质的含量,从而增加多肽物质的提取率。本实施例采用将发酵滤液添加到步骤二的猪脾浆中混合后,4-10℃保存15-24小时,以适宜的环境温度保持猪脾脏的活性状态。
优选的,所述山楂渣、食醋和白酒的重量比为28-32:4-6:2-4。本实施例中优选的采用食醋酸度为5%,白酒为52度。对经过细胞破碎的猪脾脏中的活性多肽的提取,需要先使带有活性的多肽蛋白质进行溶解。即是需要将破碎的细胞置于含有适当溶剂中,使被分离的活性多肽类生物大分子充分释放到溶剂中。经测试对比,采用将发酵滤液添加到步骤二的猪脾浆中混合后,4-10℃保存15-24小时后,猪脾浆中的多肽成分提高了15-20%。
优选的,所述步骤二的发酵温度45-55℃,容器敞口,保持通风,遮光。温度是微生物发酵的重要条件。经测试验证,在上述其它条件相同下,采用本实施例所述步骤二发酵温度45-55℃,容器敞口,保持通风,遮光,可以保证山楂渣的有效发酵以获得所需的有效成分,保证后续猪脾脏多肽提取效率。经测试验证,光照对该发酵的微生物和产生成分有影响,遮光发酵相对于强光环境效果好。
要使被提取物由固相转入液相,经过一定的保藏时间以尽量增大溶剂对被提取物的溶解度,以求获取猪脾脏中多肽的更高的提取率。溶剂中所含的物质种类和其含量、酸的浓度等,对于猪脾脏中多肽提取物的溶解度和稳定性具有显著的影响。要取得对多肽的更大的提取率,就需要对这些相关因素进行调节和控制。通过对本实施例步骤二的发酵滤液进行检测,显示发酵滤液中通过微生物代谢还产生了一定量的蛋白酶。该蛋白酶可进一步分解环境中的蛋白质从而获得小分子的多肽,进一步提高对蛋白质的水解度,从而进一步提高多肽的提取率。经对比测试,采用本实施例的山楂发酵滤液与破碎匀浆的猪脾脏混合搅拌均匀冷藏15-24小时,提取的猪脾脏多肽提高15-20%。相较猪脾脏经过洗净,去膜、筋及脂肪组织,破碎匀浆后直接进行透析,收集透析的提取技术,猪脾脏多肽提取率有显著提高。
本实施例的技术方案一方面使山楂渣得到有效的再利用,另一方面通过山楂渣发酵滤液提高了猪脾脏多肽的提取率。通过对比分析,结果表明本实施例通过采用健康猪脾脏透析液和山楂水解提取液制成的系双调因子口服液。通过对山楂渣的发酵一方面使山楂渣得到有效的再利用,增加了猪脾口服液产品有效的营养成分。另一方面发酵滤液中含有的有益成分对猪脾脏多肽的提取率的提高起到了重要促进作用。采用发酵滤液中的微生物通过代谢可产生一定量的蛋白酶,该蛋白酶可进一步分解环境中的蛋白质从而获得更多小分子的多肽,进一步提高对蛋白质的水解度,从而又进一步促使多肽的提取率的增加。本实施例制取的口服液提高了猪脾和山楂的综合利用价值,具较好的社会效益和经济效益。

Claims (7)
1.一种猪脾汤口服液的制备工艺,其特征是,包括以下步骤:
步骤一、取山楂洗净,破碎后加纯化水,温度50℃-55℃浸泡4小时,过滤,提取液加入山楂3倍质量的低聚果糖,混合,灭菌,备用;
步骤二、取猪脾脏,洗净,去膜、筋及脂肪组织,破碎匀浆后得到猪脾浆,猪脾浆在温度4-10℃进行透析过滤,收集透析液,灭菌过滤,收集滤液,备用;
步骤三、将步骤一和步骤二收集的液体等比例混合均匀,静置沉淀,过滤取上清液;
步骤四、将步骤三混合液灌封,包装,检验合格即得。
2.根据权利要求1所述的一种猪脾汤口服液的制备工艺,其特征是:
将步骤一中过滤后的山楂渣,加食醋、白酒和适量水搅拌,发酵7-10天,然后过滤获取发酵滤液;再将发酵滤液添加到步骤二的猪脾浆中混合后,4-10℃保存15-24小时,再进行透析过滤。
3.根据权利要求1所述的一种猪脾汤口服液的制备工艺,其特征是:所述步骤一中,山楂和纯化水的质量比为1:8。
4.根据权利要求2所述的一种猪脾汤口服液的制备工艺,其特征是:所述山楂渣、食醋和白酒的重量比为28-32:4-6:2-4。
5.根据权利要求2所述的一种猪脾汤口服液的制备工艺,其特征是:所述发酵温度45-55℃,容器敞口,保持通风,遮光。
6.根据权利要求1所述的一种猪脾汤口服液的制备工艺,其特征是:所述步骤二中,猪脾脏和水的比例为1:10。
7.根据权利要求2所述的一种猪脾汤口服液的制备工艺,其特征是:所述发酵采用复合乳酸菌,包含双歧杆菌、肠膜明串珠菌和植物乳杆菌,比例1:1:1,接种菌落总数2×107CFU/mL。
