Abstract
本发明涉及一种从富硒茶中提取生物活性功效成分的方法及应用,其方法包括对选择的茶叶破壁、低温浸泡萃取、分离、茶渣蒸煮、微生物发酵、煎煮、酸性消化液酶解、过滤、碱性消化液酶解、膜分离、真空喷雾干燥、得茶肽多糖螯合硒等步骤。本发明通过微生物发酵使其茶叶中的茶多糖、茶多酚、茶碱等功效成分进行生物转化,同时增加茶叶蛋白质含量5‑10%,再通过仿人胃肠消化液技术融合超微粒化纳米粉碎技术、超声波辅助技术和膜分离技术,提取分离与纯化制取茶肽多糖螯合硒生物活功能物质和富硒茶固体饮料及饲料添加剂。本发明极大地提高茶叶价值和综合效益,降低了环境污染,拉伸茶叶全产业链,创造出巨大的社会效益和经济效益。
Description
技术领域
本发明涉及生物医药食品加工技术领域,特别指一种从富硒茶中提取茶肽多糖螯合硒生物活性功效成分的方法及应用。
背景技术
富硒茶是湖南安化的特色农产品,也是具有明确功效的保健食品原料,深受国内外消费者晴睐。富硒茶的营养成分相当多,有维生素、矿物质、蛋白质、氨基酸、糖类等,尤其是含硒量达到1500-3000ug/kg,是一般茶叶含硒量的10-20倍。硒是维持人体健康必须的微量元素,参与人体生命活动全过程,而人体硒元素来源主要是膳食。但目前国内绝大多数人处于缺硒状态,这是因为土壤缺硒导致食物缺硒,植物中的硒又难以被人消化吸收,加之中国人以素食为主,只有硒元素与肽螯合在一起,才能有效地被人体消化吸收。
近年来,国内外对茶功能性成分的研究主要针对其水溶性成分对茶多糖茶多酚、咖啡碱的研究较多,而对占茶总干重为15-25%的非水溶性茶蛋白的研究目前还很少。在茶功能成分的提取加工中,大量含有茶蛋白的茶渣被废弃。在此背景下如何充分利用这些富硒茶蛋白以及研究其功效将成为富硒黑茶研究的又一热门方向。湖南安化富硒茶资源十分丰富,仅安化县黑茶年产值过100亿元,是农民的主要经济来源,但近几年由于安化黑茶产业快速发展带动了周边地区黑茶生产,出现了茶叶种植与黑茶生产过剩,甚至滞销,综合加工技术含量低、成本高、终端产品价值低,导致茶农和茶企效益差,严重制约了茶叶生产和黑茶产业健康稳步发展。
发明内容
本发明的目的是针对背景技术中存在的缺点和问题加以改进和创新,提供一种从富硒茶中提取生物活性功效成分的方法。
本发明的另一目的是提供上述活性功效成分的应用。
本发明提取生物活性功效成分的方法包括以下步骤:
A)破壁:对选择的茶叶进行破壁处理,得茶糊。
B)低温浸泡萃取:按茶原料重量加入1:5-10的纯净水,浸泡水温60-80℃,浸泡1-2h后分离,在浸泡过程中使用超声波细胞搅碎,超声波功率为300W,超声波时间25-50min。
C)分离滤液:先用80目过滤网粗滤,后用300目过滤,1200r/min离心分离得悬浮液,在2-5℃条件下静沉8-12h,取上层透明液备用,将滤渣和沉淀物合并为茶渣备用。
D)制备茶精华素:将C)步上层透明液通过60-80℃真空浓缩罐浓缩并真空喷雾干燥,再经一步制粒法制得颗粒状茶精华素。
E)制备茶肽多糖螯合硒,具体方法如下:
a)茶渣蒸煮:将C)中的茶渣放入蒸汽灌中通入饱和蒸汽,0.12pa压力,120℃蒸煮30-60min。
b)微生物接种发酵:将a)步煮后茶渣倒入发酵池内,冷却至25-40℃时接种复合微生物,混合均匀,发酵时间72-86小时。
c)煎煮:按b)步发酵料重量加入3-5倍量的纯净水,100℃煎煮60-90min,相对无菌条件冷却至25-40℃。
d)酸性消化液酶解:将c)步物料按重量加入5-10%的酸性消化液,搅拌混合均匀并使混合体的PH值达到2.5-3.5,辅助微波酶解温度45-55℃,酶解30-60min。
e)过滤与膜分离:先粗滤后膜分离,滤液备用,滤渣转入。
f)碱性消化液酶解:将e)步滤渣按重量加3-5倍量的纯净水,用NaOH调PH值10.5-11.5,加复合碱性酶2-5‰,辅助微波酶解温度45-55℃,酶解30-60min。
g)过滤与膜分离:先粗滤后膜分离,滤液备用,滤渣作饲料添加剂或有机肥用。
h)合并滤液浓缩:将e)步滤液和g)步滤液合并,真空浓缩再喷雾干燥即得茶肽多糖螯合硒。
在其中一个实施例中,所述的复合微生物种类、配比、添加量如下:1)复合微生物组合:黑曲霉菌20-30份,芽孢杆菌40-50份,酵母菌10-20份;2)复合微生物有效活菌数量:100-200亿CFU/g;3)接种量:发酵底物重量的1-3%。
在其中一个实施例中,所述的酸性消化液由以下重复份的原料制成:纯净水60-70份,10%纯度的食用级盐酸5-10份,乳酸10-20份,醋酸5-10份,以及加入上述原料总量的胃蛋白酶(1:3000)2-5‰。
在其中一个实施例中,所述的复合碱性酶配比为:胰蛋白酶(1:400)1:木瓜蛋白酶3。
在其中一个实施例中,所述的茶叶为老茶叶或低级鲜茶。
本发明方法提取的生物活性功效成分——茶肽多糖螯合硒和茶精华素应用于固体饮料中。
在其中一个实施例中,所述的固体饮料由以下原料配制而成:茶精华素700g,茶肽多糖螯合硒100g,甘露糖醇50g,L-天门冬氨酸螯合钙50g,刺五加提取物100g和维生素B620mg。该固体饮料的制备方法如下:1)调制:按所需用量取原料茶精华素、茶肽多糖螯合硒、甘露糖醇、L-天门冬氨酸螯合钙、刺五加提取物和维生素B6,参照国家固体茶饮料基本配方标准调制;2)制粒:采用一步湿法制粒风干杀菌设备完成;3)灌装:采用真空无菌灌装成2g/包的袋装产品。
本发明方法提取的生物活性功效成分——茶肽多糖螯合硒应用于膳食营养补充剂中。
在其中一个实施例中,所述的膳食营养补充剂由以下原料配制而成:茶肽多糖螯合硒800g,黄芪提取物80g,党参提取物80g和葡萄糖酸锌40g;该补充剂的制备方法如下:1)调制:按所需用量取原料茶肽多糖螯合硒、黄芪提取物、党参提取物和葡萄糖酸锌,参照国家固体茶饮料基本配方标准调制;2)制粒:采用一步湿法制粒风干杀菌设备完成;3)灌装:采用真空无菌灌装成5g/包的袋装产品。
本发明的优点及有益效果:
本发明利用丰富的富硒茶资源,尤其是利用了无法作为优质茶叶生产原料的老茶叶或低级鲜茶,通过破壁、低温浸泡萃取、酸性消化液酶解、碱性消化液酶解、膜分离技术等技术的融合,有效提取茶叶中的功效成份,避免了茶叶滞销和原料有效成份的流失,具有产品附价值高、生产成本低、竞争优势明显、安全性高的优点。
本发明采用超微粒化纳米技术破壁和真空临界点配合超声波技术萃取分离出富含(保留茶80%以上的功效成分)茶粗蛋白、多糖、茶多酚及硒等矿物元素的茶渣和富有特有风味的茶汤;茶汤经真空低温喷雾干燥成“茶精华素”即富硒茶固体饮料。通过仿制人体胃肠消化酶解液,在特定的消化条件下分解茶渣中蛋白、多糖并进行生化反应生成“茶肽多糖螯合硒”生物活性功效成分,广泛用于生物医药及功能保健食品原料和膳食营养素补充剂。本发明不仅解决了富硒茶综合深度开发技术瓶颈,极大地提高茶叶价值和综合效益,而且可以变废为宝,降低环境污染,创造出巨大的社会效益和经济效益,对科技、经济和食品业的发展具有深远的意义。
具体实施方式
为便于理解本发明,下面给出了本发明的几个实施例。但是,本发明可以以许多不同的形式来实现,并不限于本文所描述的实施例。相反地,提供这些实施例的目的是使对本发明的公开内容更加透彻全面。
除非另有定义,本文中所使用的所有的技术和科学术语与本发明的技术领域的技术人员通常理解的含义相同。说明书中所使用的术语只是为了描述具体的实施目的,不是旨在于限制本发明。
实施例1:
采用100kg鲜茶叶(老叶、修剪茶叶、茶叶末)提取茶肽多糖螯合硒生物活性功效成分,具体步骤如下:
1、破壁:采用螺杆挤压和螺杆搅切技术对茶叶进行超微粒粉碎破壁处理得100kg茶糊。
2、低温浸泡萃取:按100kg茶糊重量加入800kg纯净水,浸泡水温60-80℃,浸泡1-2h后分离。在浸泡过程中使用超声波细胞搅碎,超声波功率为300W,超声波时间25-50min。萃取茶糊中水溶性有效成分(茶多酚、茶色素、游漓氨基酸、微量元素及维生素)。
3、分离滤液:先用80目过滤网粗滤,后用300目,1200r/min离心分离得悬浮液,在2-5℃条件下静沉8-12h,取上层透明液备用即茶汤(待制茶饮料颗粒用)。滤渣和沉淀物合并约150kg用于提取分离茶肽原料。上述萃取液只获得茶叶中水溶性有效营养成分,如游漓氨基酸,15-20%的可溶性蛋白质、微量元素、维生素和茶多酚、茶色素,绝大部分保留在茶渣中。
4、浓缩:将步骤3制得的茶汤进行真空浓缩后,再经喷雾干燥得茶精华素(茶固体颗粒饮品)4.46kg。
5、蒸煮:将步骤3的茶渣和沉淀物150kg用蒸煮罐在0.12pa、120℃条件下蒸煮45min。
6、微生物发酵:将步骤5蒸煮后的茶糊冷却至25-40℃时加入复合发酵菌种2kg,搅拌混合均匀,恒温发酵72h。复合发酵菌种的种类、配比、添加量如下:1)种类配比为:黑曲霉菌20-30份,芽孢杆菌40-50份,酵母菌10-20份;2)复合微生物有效活菌数量:100-200亿CFU/g;3)接种量:发酵底物重量的1-3%。
7、煎煮:将步骤6的发酵物按重量即150kg的3倍加入450kg纯净水,100℃煎煮60min。
8、酸性条件下酶解:将步骤7煎煮后的茶糊冷却至40℃时,按重量(约600kg)加入复合酸水40kg、胃蛋白酶3kg,搅拌均匀,微波恒温54℃消化酶解60min。复合酸水的制备:纯净水60-70份、食用级盐酸(10%纯度)5-10份、乳酸10-20份和醋酸5-10份。
9、加热过滤:将步骤8酶解后的水解物加热至100℃时过滤,然后釆用小1000Da的超滤膜截留分离,滤液备用,滤渣约70kg待下次碱性条件下再次酶解。
10、碱性条件下酶解:将步骤9的滤渣加入400kg纯净水,加入50gNaOH调PH值到11.2,加入复合碱性酶4kg,微波恒温54℃消化酶解60min。复合碱性酶配比:胰蛋白酶(1:400)1:木瓜蛋白酶3。
11、加热过滤:将步骤10酶解后的水解物加热至100℃时过滤,然后釆用小1000Da的超滤膜截留分离,滤液备用,滤渣用作饲料添加剂或有机肥。
12、合并滤液浓缩:将步骤9和步骤11的滤液合并浓缩,真空喷雾干燥,得茶肽多糖螯合硒干粉13.43kg,茶功能肽(≦1000Da)含量77.64%。

二、茶肽多糖螯合硒安全性评价试验(急性毒性试验):
选用体重为18-22g的健康昆明种小鼠20只,雌雄各半。以21500mg/kg·bw剂量灌胃后,连续观察两周,记录中毒表现及死亡情况急性毒性试验:

由表3可见,以21500mg/kg·bw剂量的茶肽多糖螯合硒液给两种性别的小鼠灌胃后,观察14天,未见明显的中毒症状,也无死亡。茶肽多糖螯合硒液对雌、雄小鼠的急性经口LD50大于21500mg/kg·bw,根据GB15193.3-94中的急性毒性分级标准,该样品属无毒物。
三、茶肽多糖螯合硒免疫试验:
选择清洁级昆明种雌、雄小鼠120只,体重为18~22g,由中南大学湘雅医学院实验动物学部提供。据人体口服推荐量,设小鼠低剂量为1.65ml/kg·bw,中剂量为3.30ml/kg·bw,高剂量为9.90ml/kg·bw,分别相当于人口服推荐量的5、10、30倍,取样品加蒸馏水配制各剂量所需浓度受试液,以此液灌胃,每天灌胃一次,对照组予以等体积的蒸馏水,灌胃体积为0.2ml/kg·bw,连续30天。
1、实验方法:
1)脏器/体重比值测定:称重后处死小鼠,取出脾脏和胸腺,在电子分析天平上称重,计算脏/体比值。
2)迟发型变态反应(DTH)(足跖增厚法):
小鼠腹腔注射2%(v/v)SRBC(0.2ml/每鼠)致敏后4天,测量左后足跖厚度,然后在测量部位皮下注射20%(v/v)SRBC(20ul/每鼠),于注射后24h测量左右足跖部厚度,同一部位测量三次,取平均值,以攻击前后足跖厚度差值(足跖肿胀度)来表示DTH的程度。
3)抗体生成细胞检测(Jerne改良玻片法):
取羊血,用生理盐水洗涤3次,每次离心(2000r/min)10min,将压积SRBC用生理盐水配成2%(v/v)的细胞悬液,每鼠腹腔注射0.2ml,将免疫后4天的小鼠处死,取脾,轻轻撕碎,用Hanks液制成细胞悬液,200目筛网过滤,洗涤、离心2次,最后将细胞悬浮在5ml Hanks液中,将表层培养基加热溶解后与等量的PH7.4、2倍浓度的Hanks液混合,分装小试管,每管0.5ml,再向管内加入用SA液配制的10%SRBC50ul(v/v)、20ul脾细胞悬液,迅速混匀后倾倒于已刷薄层琼脂糖的玻片上,待琼脂糖凝固后将玻片水平扣放在玻片架上,放入二氧化碳培养箱中温育1.5h,然后用SA液稀释的补体(1:10)加入到玻片凹槽内,继续温育1.5h后计数溶血空斑数。
4)小鼠碳廊清试验:
小鼠尾静脉注射以生理盐水稀释4倍的印度墨汁,每10g体重注射0.1ml,墨汁注入后立即计时,于注入墨汁后第2、10min,分别从内眦静脉丛取血20ul,加入到2mlNa2CO3溶解中,摇匀。以Na2CO3深液作空白对照,用722型分光光度度计在60nm波长处比色测光密度值(OD)。将小鼠处死,取肝、脾,称重,计算吞噬指数。
2、实验结果:

由表4-6可见,各剂量组实验初、实验中期、实验末小鼠体重及实验期间小鼠体重增长对照组相比,差异无显著性(P>0.05)。

由表7可见,径口给予小鼠不同剂量的30天,茶肽多糖螯合硒对小鼠的脾脏/体重和胸腺/体重比值无明显影响。
3)茶肽多糖螯合硒对小鼠迟发型变态反应(DTH)的影响:

由表10可见,径口给予小鼠不同剂量的茶肽多糖螯合硒30天,高剂量组小鼠吞噬指数显著高于对照组,差异有显著性。
6)结论:
经口给予小鼠1.65、3.30、9.910ml/kg·bw剂量的茶肽多糖螯合硒30天,能明显增加小鼠的迟发型变态反应,增加小鼠的抗体生成细胞数,增加小鼠单核巨噬细胞碳廓清的能力,提示该样品能增加小鼠的细胞免疫、体液免疫功能及单核-巨噬细胞的吞噬功能,具有免疫调节作用。
本发明主要利用老茶叶、修剪茶叶及边着废茶,通过微生物发酵使其茶叶中的茶多糖、茶多酚、茶碱等功效成分进行生物转化,同时增加茶叶蛋白质含量5-10%,再通过仿人胃肠消化液技术融合超微粒化纳米粉碎技术、超声波辅助技术和膜分离技术,提取分离与纯化制取茶肽多糖螯合硒生物活功能物质和富硒茶固体饮料及饲料添加剂。因此本发明不仅解决了富硒茶综合深度开发技术瓶颈,极大地提高茶叶价值和综合效益,而且可以变废为宝,降低环境污染,拉伸茶叶全产业链,创造出巨大的社会效益和经济效益。
实施例2:
将实施例1方法制备的茶精华素和茶肽多糖螯合硒应用于抗疲劳固体饮料中:
以1000g为例,其抗疲劳固体饮料的配方为:700g实施例1制备的茶精华素,100g实施例1制备的茶肽多糖螯合硒,甘露糖醇50g,L-天门冬氨酸螯合钙50g,刺五加提取物100g,维生素B620mg。
该抗疲劳固体饮料制备具体步骤如下:参照国家固体茶饮料基本配方标准将上述原料进行调制,然后采用一步湿法制粒风干杀菌设备完成制粒,最后采用真空无菌灌装成2g/包的产品。
实施例3:
将实施例1方法制备的茶肽多糖螯合硒应用于膳食营养补充剂中:
以1000g为例,其膳食营养补充剂的配制如下:茶肽多糖螯合硒800g,黄芪提取物(黄芪多糖)80g,党参提取物(党参多糖)80g,葡萄糖酸锌40g。
该膳食营养补充剂制备具体步骤如下:参照国家固体茶饮料基本配方标准将上述原料进行调制,然后采用一步湿法制粒风干杀菌设备完成制粒,最后采用真空无菌灌装成5g/包的产品。
本发明所述的实施例仅仅是对本发明的优选实施方式进行的描述,并非对本发明构思和范围进行限定,在不脱离本发明设计思想的前提下,本领域中工程技术人员对本发明的技术方案作出的各种变型和改进,均应落入本发明的保护范围,本发明请求保护的技术内容,已经全部记载在权利要求书中。
Claims (6)
1.一种从富硒茶中提取生物活性功效成分的方法,其特征在于包括以下步骤:
A)破壁:对选择的茶叶采用螺杆挤压和螺杆搅切技术对茶叶进行超微粒粉碎破壁处理得茶糊;
B)低温浸泡萃取:按茶糊重量加入5-10倍的纯净水,浸泡水温60-80℃,浸泡1-2h后分离,在浸泡过程中使用超声波细胞搅碎,超声波功率为300W,超声波时间25-50min;
C)分离滤液:先用80目过滤网粗滤,后用300目过滤,1200r/min离心分离得悬浮液,在2-5℃条件下静沉8-12h,取上层透明液备用,将滤渣和沉淀物合并为茶渣备用;
D)制备茶精华素:将C)步上层透明液通过60-80℃真空浓缩罐浓缩并真空喷雾干燥,再经一步制粒法制得颗粒状茶精华素;
E)制备茶肽多糖螯合硒,具体方法如下:
a)茶渣蒸煮:将C)中的茶渣放入蒸汽灌中通入饱和蒸汽,0.12pa压力,120℃蒸煮30-60min;
b)微生物接种发酵:将a)步煮后茶渣倒入发酵池内,冷却至25-40℃时接种复合微生物,混合均匀,发酵时间72-86小时;
所述的复合微生物种类、配比、添加量如下:
1)复合微生物组合:黑曲霉菌20-30份,芽孢杆菌40-50份,酵母菌10-20份;
2)复合微生物有效活菌数量:100-200亿CFU/g;
3)接种量:发酵底物重量的1-3%;
c)煎煮:按b)步发酵料重量加入3-5倍量的纯净水,100℃煎煮60-90min,相对无菌条件冷却至25-40℃;
d)酸性消化液酶解:将c)步物料按重量加入5-10%的酸性消化液,搅拌混合均匀并使混合体的PH值达到2.5-3.5,辅助微波酶解温度45-55℃,酶解30-60min;
所述的酸性消化液由以下重复份的原料制成:
纯净水60-70份,10%纯度的食用级盐酸5-10份,乳酸10-20份,醋酸5-10份,以及加入上述原料总量的胃蛋白酶2-5‰;
e)过滤与膜分离:先粗滤后膜分离,滤液备用,滤渣转入;
f)碱性消化液酶解:将e)步滤渣按重量加3-5倍量的纯净水,用NaOH调PH值10.5-11.5,加复合碱性酶2-5‰,辅助微波酶解温度45-55℃,酶解30-60min;
所述的复合碱性酶配比为:胰蛋白酶1:木瓜蛋白酶3;
g)过滤与膜分离:先粗滤后膜分离,滤液备用,滤渣作饲料添加剂或有机肥用;
h)合并滤液浓缩:将e)步滤液和g)步滤液合并,真空浓缩再喷雾干燥即得茶肽多糖螯合硒。
2.根据权利要求1所述的一种从富硒茶中提取生物活性功效成分的方法,其特征在于所述的茶叶为老茶叶或低级鲜茶。
3.一种权利要求1-2所述任一方法提取的生物活性功效成分的应用,其特征在于将提取的所述茶肽多糖螯合硒和茶精华素应用于固体饮料中。
4.根据权利要求3所述的应用,其特征在于所述的固体饮料由以下原料配制而成:
茶精华素700g,茶肽多糖螯合硒100g,甘露糖醇50g,L-天门冬氨酸螯合钙50g,刺五加提取物100g和维生素B6 20mg;
制备方法如下:
1)调制:按所需用量取原料茶精华素、茶肽多糖螯合硒、甘露糖醇、L-天门冬氨酸螯合钙、刺五加提取物和维生素B6,参照国家固体茶饮料基本配方标准调制;
2)制粒:采用一步湿法制粒风干杀菌设备完成;
3)灌装:采用真空无菌灌装成2g/包的袋装产品。
5.一种权利要求1-2所述任一方法提取的生物活性功效成分的应用,其特征在于将提取的所述茶肽多糖螯合硒应用于膳食营养补充剂中。
6.根据权利要求5所述的应用,其特征在于所述的膳食营养补充剂由以下原料配制而成:
茶肽多糖螯合硒800g,黄芪提取物80g,党参提取物80g和葡萄糖酸锌40g;
制备方法如下:
1)调制:按所需用量取原料茶肽多糖螯合硒、黄芪提取物、党参提取物和葡萄糖酸锌,参照国家固体茶饮料基本配方标准调制;
2)制粒:采用一步湿法制粒风干杀菌设备完成;
3)灌装:采用真空无菌灌装成5g/包的袋装产品。
