技术领域
本发明属于功能微生物筛选与应用技术领域,具体涉及一种耐盐芽孢杆菌SW207及其应用。
背景技术
公开该背景技术部分的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
在禽畜养殖业中,通常使用抗生素来促进禽畜的生长和预防腹泻。然而,过度使用抗生素会导致禽畜的肠道病原菌耐药性和药物残留等问题。研究人员致力于研究能够改善禽畜的生长性能和免疫功能而又无任何副作用的抗生素替代品。益生菌具有促进营养物质的消化吸收、提高免疫力、维持肠道菌群平衡、保护肠粘膜屏障等优势,可以成为替代抗生素的替代品。
近年来,芽孢杆菌作为一种潜在的益生菌被广泛使用,其具有很强的耐受性。研究发现,芽孢杆菌对多种动物病原具有拮抗作用,包括大肠杆菌、金黄色葡萄球菌和沙门氏菌。其中一些芽孢杆菌也通过产生细菌素有效地抑制肉类中李斯特菌的生长。此外,芽孢杆菌还被用于家禽、猪和水产养殖业的生长促进剂,通过参与体内一些特定的新陈代谢和生产短链脂肪酸,可以改善宿主的代谢能力。耐盐芽孢杆菌是芽孢杆菌科的一个种属,目前主要是从植物中分离得到,是一种高效抗菌的生防菌,但现有技术中未出现从动物体内分离出耐盐芽孢杆菌,也未将耐盐芽孢杆菌应用于生猪养殖生产中。
发明内容
为了克服上述问题,本发明提供了一种Bacillus halotoleransSW207及其应用。本发明从具有优良抗病功能的松雷猪肠道中分离了一株可以有效拮抗产肠毒素大肠杆菌的Bacillus halotoleransSW207,经研究发现Bacillus halotoleransSW207菌株对断奶仔猪的细菌性腹泻、生产性能和免疫功能具有较好的作用效果。
为实现上述技术目的,本发明采用如下技术方案:
本发明的第一个方面,提供了一种耐盐芽孢杆菌,命名为Bacillus halotoleransSW207,于2022年12月02日保存于武汉市中国典型培养物保藏中心,保藏编号为CCTCC NO:M 20221856。
所述Bacillus halotoleransSW207的16S rDNA序列见SEQ ID NO.1。
在本发明中,所述Bacillus halotoleransSW207的形态学特征为:单菌落形态呈近圆形,浅黄色,不透明;培养初期菌落表面光滑,边缘整齐,后期表面有褶皱,边缘略不整齐,四周呈云雾状扩散;经革兰氏染色表明:所述Bacillus halotoleransSW207为革兰氏阳性菌,杆状,菌体长1.2~2.1μm,宽0.5~0.7μm,芽孢呈椭圆形,近端生,孢囊稍膨大,长0.5~1.2μm,宽0.2~0.4μm。
本发明的Bacillus halotoleransSW207具有强耐酸和耐胆盐能力,菌株在pH4.0条件下处理180min以及0.1%浓度胆盐处理120min过程中能始终维持85%以上的存活率。
本发明的Bacillus halotoleransSW207具有一定的产蛋白酶、淀粉酶和纤维素酶能力。菌株在1%接种量的条件下接种在蛋白酶菌株筛选培养基,阳性对照水解圈直径为1.8~2.1cm,Bacillus halotoleransSW207菌株水解圈直径为1.9~2.1cm;菌株在1%接种量的条件下接种在淀粉水解培养基,阳性对照水解圈直径为2.5~2.7cm,Bacillus halotoleransSW207菌株水解圈直径为2.4~2.6cm;通过可见分光光度法在540nm的波长下测定纤维素酶活性,产纤维素酶量为1.5U/104cell。
本发明的Bacillus halotoleransSW207具有较少的耐药基因,在22种常见种抗生素的药敏试验中,仅对青霉素和林可霉素不敏感,安全性较高。
本发明的Bacillus halotoleransSW207具有较广谱的抑菌作用,体外抑菌试验显示菌株Bacillus halotoleransSW207对产肠毒素大肠杆菌、产气荚膜梭菌、胸膜肺炎放线杆菌和肺炎克雷伯氏菌均有抑菌作用,并具有良好的菌体表面性质;次级代谢产物分析菌株Bacillus halotoleransSW207产生7种新型抗菌肽和3种未知抗菌肽,具有抗多种病原的能力。
细菌全基因组分析显示,本发明的Bacillus halotoleransSW207大小为4,163,876bp,GC百分比为48.31%,共编码基因4,389个,总长度为3,691,962bp,平均长度为841bp;含有tRNA基因86个,5S、16S、23S rRNA各10个,基因岛13个,前噬菌体16个和CRISPR位点1个;菌株在碳水化合物代谢、氨基酸代谢、辅助因子和维生素的代谢、能量代谢等通路上有丰富的基因注释信息,具有促进动物生产、提高耐粗饲能力的潜力。
本发明的Bacillus halotoleransSW207在保护猪小肠上皮细胞(IPEC-J2)受产肠毒素大肠杆菌(ETEC K88)感染试验中,发现菌株Bacillus halotoleransSW207首先竞争性地获得IPEC-J2细胞上的结合位点,与IPEC-J2细胞黏附来抑制ETEC K88生长。进一步与TLR2和TLR4相互作用,诱导组织产生IL-10和GM-CSF,降低炎性细胞因子表达(TNF-α、IL-1β、IL-6、IL-8、IL-18)、提高HDPs和紧密连接蛋白(ZO-1、Claudin-1和Occludin)的表达量,增强肠道上皮的屏障功能,调节免疫系统,抑制ETEC K88感染IPEC-J2细胞。
本发明的Bacillus halotoleransSW207在防治断奶仔猪腹泻、促进断奶仔猪生长及提高免疫功能中的应用中发现,添加Bacillus halotoleransSW207可以提高断奶仔猪生长性能,降低断奶仔猪料肉比和腹泻率,提高断奶仔猪的免疫功能。
本发明的第二个方面,提供了一种仔猪益生菌制剂,其活性成分为Bacillus halotoleransSW207 CCTCC NO:M 20221856。
本发明的第三个方面,提供了一种动物饲料添加剂,其活性成分为Bacillus halotoleransSW207 CCTCC NO:M 20221856。
在一种或多种实施方式中,所述动物饲料添加剂中Bacillus halotoleransSW207CCTCC NO:M 20221856的含量为1×109~5×109CFU/kg。

松雷猪也称野梅香猪,是通过我国特有的杂交内三元猪,具备能吃草、无污染、肉质佳、营养高等优点,在生猪品种、养殖方式、饲料结构、抗阻病疫、猪肉品质、经济效益六个方面具有重大突破。松雷猪能在零下28℃的寒冷环境正常生产,并具有抗病和耐粗纤维的能力,可以比其他猪种更少接触抗生素。
本申请从具有优良抗病功能和耐粗饲能力的松雷猪肠道中分离出一株可以有效拮抗产肠毒素大肠杆菌的Bacillus halotoleransSW207。经研究发现Bacillus halotoleransSW207菌株对断奶仔猪的细菌性腹泻、生产性能和免疫功能具有较好的作用效果,因此该菌株可作为一种绿色、安全的益生菌应用到生猪生产中。
为了使得本领域技术人员能够更加清楚地了解本发明的技术方案,以下将结合具体的实施例详细说明本发明的技术方案。
实施例1
Bacillus halotoleransSW207的分离鉴定
取180日龄的松雷猪盲肠食糜0.3g,按照1:10的比例,用无菌PBS溶液进行稀释,后用无菌PBS溶液按照10倍梯度稀释,选取1×104、1×105和1×106三个稀释梯度,分别吸取100μL的菌悬液涂布于BHI固体培养基上,37.5℃倒置恒温培养16~24h。挑取固体平板上不同颜色和形态的菌落进行传代,直至获得纯化的单菌落,接种于BHI液体培养基中增菌,在37.5℃的恒温摇床中220rpm培养1~2d,每个稀释梯度和每个菌落至少做3个平板。所述BHI固体培养基(1L)含:脑心浸粉17.5g、D-葡萄糖2.0 g、蛋白胨10.0 g、氯化钠5.0 g、磷酸氢二钠2.5 g、琼脂15.0 g。
分离Bacillus halotoleransSW207的菌液通过DNA提取试剂盒提取了DNA,并扩增了16S rRNA基因,送于通用生物(安徽)股份有限公司进行Sanger测序,序列如SEQ ID No.1所示。16S rDNA序列在EzbioCloud中进行序列相似性比对得到结果,菌株SW207与耐盐芽孢杆菌有99.79%的序列相似度。如图1所示,纯化的Bacillus halotoleransSW207在BHI培养基上的生长照片(图1中A),Bacillus halotoleransSW207革兰氏染色照片(图1中B),扫描电子显微镜(SEM)照片(图1中C)和透射电子显微镜(TEM)照片(图1中D),观察其形态学特征。Bacillus halotoleransSW207的形态学特征为:单菌落形态呈近圆形,浅黄色,不透明;培养初期菌落表面光滑,边缘整齐,后期表面有褶皱,边缘略不整齐,四周呈云雾状扩散;经革兰氏染色表明:所述Bacillus halotoleransSW207为革兰氏阳性菌,杆状,菌体长1.2~2.1μm,宽0.5~0.7μm,芽孢呈椭圆形,近端生,孢囊稍膨大,长0.5~1.2μm,宽0.2~0.4μm。综上鉴定菌株SW207为耐盐芽孢杆菌。
实施例2
Bacillus halotoleransSW207的益生特性评价
(1)采用比浊法测定Bacillus halotoleransSW207的生长曲线,将Bacillus halotoleransSW207在固体培养基活化后,接种于液体培养基(BHI)过夜培养12h,将菌株培养液按2%(w/v)的接菌量重新接种于液体培养基(BHI);所述Bacillus halotoleransSW207接种的液体培养基(BHI)(1L)含:脑心浸粉17.5 g、D-葡萄糖2.0 g、蛋白胨10.0 g、氯化钠5.0 g、磷酸氢二钠2.5 g,加入蒸馏水1000ml,调整pH为6.8~7.2,121℃灭菌15min;将混合均匀后的Bacillus halotoleransSW207放入37.5℃摇床中培养,间隔一定时间,即2h、4h、6h、8h、10h、12h、16h、24h、48h后从摇床上取其中一管菌液,吸取200μL菌液测量其600nm处的吸光值,将测量结果用R-4.2.2绘制成细菌生长曲线,其结果如图2中A所示。
(2)耐酸试验:将BHI液体培养基的pH分别调整为2,3,4,5,7,且进行高压蒸汽灭菌,备用。将过夜培养12h的菌液按10%(w/v)的接菌量分别接种于各pH的BHI液体培养基中,以pH为7的培养基为对照,37.5℃恒温摇床培养12h,吸取各管中菌液200μL测定其OD600,计算菌株Bacillus halotoleransSW207在不同pH下的存活率,存活率=(试验组OD600÷对照组OD600)×100%,试验结果如图2中B图所示。
(3)耐胆盐试验:分别配置胆盐浓度为0,0.1%,0.2%,0.3%,0.4%,0.5%的BHI液体培养基,且进行高压蒸汽灭菌,备用。将过夜培养12h的菌液按10%(w/v)的接菌量分别接种于各胆盐浓度的BHI液体培养基中,以未加胆盐的培养基作为对照组,37.5℃恒温摇床培养12h,吸取各管中菌液200μL测定其OD600,计算菌株Bacillus halotoleransSW207在不同胆盐浓度下的存活率,存活率=(试验组OD600÷对照组OD600)×100%,试验结果如图2中C图所示。
(4)产酶试验:配置接种菌液,将过夜培养12h的菌液用PBS调整菌液浓度约106CFU/mL,4℃冷藏备用。
淀粉酶试验:吸取上述接种菌液2μL,接种到淀粉酶筛选培养基上,倒置于37℃恒温培养箱培养24h后,加2mL碘液染色,用直尺测量各菌苔直径和菌苔四周透明圈的直径,测量三次取平均值,透明圈直径与菌苔直径的比值代表菌株产淀粉酶能力的大小,试验结果如图2中D图所示。
蛋白酶试验:吸取上述接种菌液2μL,接种到蛋白酶筛选培养基上,倒置于37℃恒温培养箱培养24h后,用直尺测量各菌苔直径和菌苔四周透明圈的直径,测量三次取平均值,透明圈直径与菌苔直径的比值代表菌株产蛋白酶能力的大小,试验结果如图2中D图所示,。
纤维素酶试验:基于北京索莱宝生物科技公司的纤维素酶(CL)活性检测试剂盒(BC2540)方法,通过可见分光光度法检测菌株Bacillus halotoleransSW207的产纤维素酶能力,试验结果如图2中D图的柱状图所示。
(5)抗生素敏感性试验:采用药敏纸片琼脂扩散法测定菌株Bacillus halotoleransSW207对抗生素的敏感性。将活化后的菌株过夜培养12h,用无菌一次性涂布棒将菌株培养液分别均匀的涂抹在BHI平板上,在超净台中自然干燥后,将22种药敏纸片均匀的贴于BHI平板表面,用镊子轻压药敏纸片使其不易脱落,37℃培养24h后,测量各药敏片的抑菌圈直径,测量三次取平均值,试验结果如图2中E图所示。
(6)体外抑菌试验:采用琼脂扩散法检验菌株Bacillus halotoleransSW207对4种致病菌的抗菌能力。病原指示菌分别为产肠毒素大肠杆菌、肺炎克雷伯菌、产气荚膜梭菌和胸膜肺炎放线杆菌。将以上病原菌在LB和TSB 液体培养基中活化三代恢复活性后,按2%(w/v)的接菌量重新接入TSB液体培养基,在37℃摇床培养12h后,用无菌一次性涂布棒将病原菌菌液均匀涂抹在TSB平板上,将培养基在超净台中晾至自然干燥,用9mm直径的打孔器在TSB平板上打孔,用灭菌过的镊子将多出的琼脂块夹出,将培养皿在酒精灯外焰上加热3~5s进行封底。将菌株Bacillus halotoleransSW207接种于BHI液体培养基培养12h,将菌液离心、弃掉上清液,然后用PBS重悬,洗涤2次后,调节菌液浓度约1×108CFU/mL,用移液枪吸取100μL菌液加入孔内,其中一个孔加入无菌的PBS作为空白对照,静置至菌液完全吸收进培养基后,再放入37℃恒温培养箱中培养24h,测量各孔透明抑菌圈的直径,直径越大,抑菌能力越强,试验结果如图2中G图所示。
(7)菌体表面性质测定:
菌体表面疏水率测定,菌体悬液制备:将菌株Bacillus halotoleransSW207接种于液体培养基培养24h,将菌株培养液离心后弃上清,并用PBS将菌液浓度调整为OD600=0.6,设为A0。将调整后的菌悬液分别与等体积的氯仿和二甲苯混合,在混合仪上混合5min,室温下静置60min。液面分层后吸取水相200μL,以PBS为空白对照,测量其600nm处的吸光值(A1)。根据公式计算细菌的表面疏水率:菌体表面疏水率(%)=(1-A1÷A0)×100,试验结果如图2中F图所示。
自凝集试验:菌体悬液摇匀后用移液器吸取2mL OD600=0.6的菌液于4mL的EP管中,静置10h,至菌体沉淀后,吸取200μL上清液测量其600nm处的吸光值(A2)。根据公式计算菌株的自凝集率:自凝集率(%)=(1-A2÷A0)×100,试验结果如图2中F图所示。
共凝集实验:检验菌株Bacillus halotoleransSW207与产肠毒素大肠杆菌、产气荚膜梭菌和胸膜肺炎放线杆菌的共凝集效果。分别取2mL的菌株Bacillus halotoleransSW207和指示菌菌液于10mL的EP管中,在涡旋混匀仪上混匀3min,静置10h,至菌体沉淀后,吸取200μL上清液测量其600nm处的吸光值(A3)。根据公式计算菌株于指示菌的共凝集率:共凝集率(%)=(1-A3÷A0)×100,试验结果如图2中F图所示。
试验结果如下:
如图2所示,菌株在PH4.0条件下处理180min以及0.1%浓度胆盐处理120min的过程中始终能维持85%以上的存活率。益生菌需要进入胃肠道中定殖才能发挥作用,必须耐受胃液的低pH值才能存活,而食物消化吸收的部位主要在小肠,而胆汁由肝脏排入小肠,胆汁中的胆盐对细菌生长有抑制作用,因此候选益生菌必须有一定的耐胆盐能力。而本发明菌株Bacillus halotoleransSW207具有强耐受高胆盐和胃酸的能力,具备在猪肠道定植的基础;菌株Bacillus halotoleransSW207还具备产蛋白酶、淀粉酶和纤维素酶能力,可帮助宿主消化吸收,尤其是由于其自身消化系统不成熟,内源性酶的产生可能不足的断奶仔猪。
如果益生菌中存在耐药基因,可能会对肠道正常菌群和致病菌发生耐药基因的转移,引起菌群紊乱以及疾病发生,本发明菌株Bacillus halotoleransSW207在23种常见种抗生素的药敏试验中,仅对青霉素和林可霉素不敏感,安全性较高。
益生菌对病原菌的抑菌能力是提高宿主免疫力以及抗病的首要条件之一,体外抑菌试验显示菌株Bacillus halotoleransSW207对产肠毒素大肠杆菌(抑菌圈直径:17±1.0mm)、产气荚膜梭菌(抑菌圈直径:13±1.0mm)、胸膜肺炎放线杆菌(抑菌圈直径:16±0.5mm)和肺炎克雷伯氏菌(抑菌圈直径:13±0.5mm)均有抑菌作用。
益生菌的黏附能力对肠道定植十分重要,菌株的自凝集性和菌体表面疏水性与黏附能力呈正相关,菌体的自凝集性越强,菌体能在肠道中更好的聚集,形成更大的生物量,而疏水性越强,自凝集性越强。益生菌的共凝集性反映了益生菌的抗菌能力,通过与致病菌的共凝集作用,可以抑制致病菌对肠道的黏附定殖。本发明菌株Bacillus halotoleransSW207具有较强的自凝集性和疏水性,对3种病原菌的共凝集性也较强,表明菌株Bacillus halotoleransSW207有较强的黏附能力和抑制病原菌黏附的能力。
因此,菌株Bacillus halotoleransSW207具有优良的生物学特性和抑菌作用,安全性高,可在实际生产中推广应用。
实施例3
菌株Bacillus halotoleransSW207全基因组分析
(1)细菌全基因组测序:测序委托北京诺禾致源科技股份有限公司完成。质检合格的菌株DNA样品经Covaris gTUBE剪切成小片段DNA,受损的DNA经修复后用DNA黏合酶连接接头,使用AMpure PB磁珠对DNA片段进行纯化,并构建SMRT Bell文库。Buffer溶解,经片段筛BluePipin后分成固定大小片段,再次使用AMpure PB磁珠进行纯化。二次得到的文库使用Qubit进行浓度测定,Agilent 2 100对文库质量质检。采用第3代测序仪PacBioRS II对DNA进行测序;其结果如图3中A所示。
(2)生物信息学分析:过滤除掉低质量的reads后的clean data进行基因组组装,包含菌株的基因组基因情况的序列文件进行评价;分析菌株基因组成分,涉及编码基因(http:∥topaz.gatech.edu/GeneMark/)、重复序列及滚环(http:∥topaz.gatech.edu/GeneMark/)、tRNA(http:∥lowelab.ucsc.edu/tRNAscan-SE/)等基因成分的分析;然后对编码基因进行功能注释,包含的数据库有KEGG(https:∥www.genome.jp/kegg/)、CAZy数据库(http://www.cazy.org/)和NCBI数据库(https:∥www.ncbi.nlm.nih.gov/)。从基因组和基因两层面分别比较样品与参考基因组的差异;呈现基因组组分分析结果和功能注释结果,其结果如图3中B所示。
(3)次级代谢产物分析:通过antiSMASH(v6.1.1)进行快速菌株Bacillus halotoleransSW207全基因组识别,基于规则聚类检测,通过核心生物合成酶来注释和分析45种不同类型的次级代谢产物生物合成途径,其结果如图3中C所示。
试验结果:如图3显示,Bacillus halotoleransSW207全基因组大小为4,163,876bp,GC%为43.81%。共编码基因4,389个,总长度为3,691,962bp,平均长度为841bp。含有tRNA基因86个,5S rRNA 、16S rRNA和23S rRNA各10个,基因岛13个,前噬菌体16个和1个CRISPR位点;菌株Bacillus halotoleransSW207在碳水化合物代谢途径、氨基酸代谢途径、辅助因子和维生素的代谢途径和能量代谢等途径上有着丰富的基因注释信息,具有较强的代谢碳水化合物能力,可应用于促生产、提高耐粗饲能力的产品中;次级代谢产物分析菌株Bacillus halotoleransSW207产生7种新型抗菌肽和3种未知抗菌肽,包括多烯类(Bacillaene)、丰原素(Fengycin)、儿茶酚型嗜铁素(Bacillibactin)、枯草芽孢杆菌素(Subtilosin A)、抗菌二肽溶杆菌素(Bacilysin)和表面活性肽(Surfactin)等等,研究显示这些抗菌肽具有具有抗病毒、抗肿瘤、抗细菌、抗真菌、抗支原体、抗炎等多种作用,展示出极大的临床应用潜力,是新型的抗菌肽。因此,菌株Bacillus halotoleransSW207具有较大的畜牧业和药业应用潜力。
实施例4
菌株在产肠毒素大肠杆菌感染猪小肠上皮细胞中的保护作用
细菌和细胞的培养:将菌株Bacillus halotoleransSW207在固体培养基活化后,接种于液体培养基过夜培养12h,将菌株培养液按2%(w/v)的接菌量重新接种于液体培养基,37.5℃培养24h,6000rpm离心取菌体,用PBS缓冲液重悬洗涤2次,然后重悬于PBS缓冲液中待用;猪小肠上皮细胞IPEC-J2细胞按1×106细胞/孔接种于6孔细胞培养板中,加入DMEM/F12完全培养基,放置5%CO2,37℃细胞培养箱中培养,每隔一天换DMEM/F12完全培养基一次,至100%细胞融合时进行试验。
菌株Bacillus halotoleransSW207保护IPEC-J2细胞免受ETEC诱导的细胞毒性作用:
试验三株菌:1、菌株Bacillus halotoleransSW207;2、枯草芽孢杆菌(B. subtilis);3、嗜黏蛋白阿克曼氏菌(A. muciniphila),对比本发明菌株BacillushalotoleransSW207与其他两种益生菌的效果,并进行统计分析、评价。试验共分为四组:1、NT Control组;2、Bacillus halotoleransSW207组;3、B. subtilis组;A. muciniphila组。每个组进行三种处理:1、无处理;2、共孵育:益生菌(108CFU/mL)和ETEC(108CFU/mL)共同在IPEC-J2细胞中孵育,37℃的CO2培养箱中培养4h;3、预孵育:益生菌(108CFU/mL)首先在IPEC-J2细胞中孵育,37℃的CO2培养箱中培养3h。然后加入ETEC(108CFU/mL)进行刺激,37℃的CO2培养箱中培养4h。使用Cytation 5 Cell Imaging Multi-Mode Reader放大20倍,在明场模式下使用100毫米刻度。除去培养基后,用PBS洗涤细胞两次并用0.25%胰蛋白酶-EDTA收集细胞。通过使用具有TC20自动细胞计数器的标准台盼蓝测定来计数活细胞的数量,其试验结果如图4中A所示。
对于菌株Bacillus halotoleransSW207对ETEC感染细胞的黏附作用,采用108CFU/mL的Bacillus halotoleransSW207和ETEC在37℃与IPEC-J2细胞共培养3h。粘附的细菌在LB培养基中连续稀释10倍并铺在LB琼脂板上。好氧培养24h后计数Bacillus halotoleransSW207和ETEC菌落数。Bacillus halotoleransSW207和ETEC分别在BHI和LB琼脂平板中计数;对于IPEC-J2细胞系中的一氧化氮产量测量:通过使用Griess试剂系统(Promega, Madison, WI)测量不同处理诱导的一氧化氮产生,该系统测量亚硝酸盐作为NO的主要分解产物,结果表示为产生的亚硝酸盐浓度(mM),其结果如图4中B、C所示。
细胞增殖测定:IPEC-J2细胞在37℃下用108CFU/mL的ETEC刺激2小时。然后去除旧培养基,将细胞与含有Bacillus halotoleransSW207的108CFU/mL 的新鲜培养基在37℃下再孵育6小时。使用CCK-8试剂盒测量细胞增殖,测量450nm处的吸光度来计数增殖中的代谢活性细胞,其结果如图4中D所示。
实时荧光定量PCR:用108CFU/mL的ETEC和108CFU/mL的Bacillus halotoleransSW207感染IPEC-J2细胞,并在37℃培养箱中孵育4小时。收集细胞,并使用Norgen总RNA纯化试剂盒从细胞中分离总RNA;用NanoDrop8000分光光度计评估RNA产量和质量;使用iScript Reverse Transcription Supermix试剂盒将提取的RNA样品逆转录为互补DNA;使用Bio-Rad CFX Connect实时系统进行定量实时PCR;使用Primer-BLAST工具设计引物,并由吉林省库美生物科技有限公司合成;引物信息列在表1中。实时定量PCR,95℃进行激活酶2分钟;95℃,变性5s;然后在60℃,退火20秒,反应重复40个循环。通过qPCR计算每个引物组的效率,只选择效率在90~110%之间的引物组进行下游基因表达分析。通过琼脂糖凝胶电泳验证每个引物组的产物大小。一式三份评估样品。通过将靶基因的循环阈值,CT值标准化为管家基因编码的CT值来分析相对mRNA表达GAPDH。结果表示为使用2-ΔΔCt法方法计算的倍数变化,其结果如图4中E、F所示。

试验结果:
如图4所示,为分析Bacillus halotoleransSW207在产肠毒素大肠杆菌感染猪小肠上皮细胞中的保护效果,通过与2株益生菌(枯草芽孢杆菌和嗜黏蛋白阿克曼菌)来共同比较分析。结果表明,三株新型益生菌均不影响IPEC-J2细胞的活力,产肠毒素大肠杆菌在孵育4h后可显著(P<0.001)引起IPEC-J2细胞死亡。但当IPEC-J2细胞与Bacillus halotoleransSW207共培养或预孵育后,产肠毒素大肠杆菌引起细胞的损伤显著减弱,达到了阴性对照的水平。但IPEC-J2细胞与其余两株益生菌共培养或预孵育后无效果,细胞损伤严重。因此,Bacillus halotoleransSW207可以保护IPEC-J2细胞由ETEC诱导的细胞毒性和损伤,菌株Bacillus halotoleransSW207具有优良的保护猪小肠上皮细胞免受产肠毒素大肠杆菌侵袭的能力。
为研究Bacillus halotoleransSW207对ETEC感染细胞的黏附作用,采用相同浓度的Bacillus halotoleransSW207和ETEC在37℃与IPEC-J2细胞共培养3h。结果显示,Bacillus halotoleransSW207比ETEC在单独培养时有着更高的黏附能力。共培养时,Bacillus halotoleransSW207显著降低了ETEC与IPEC-J2细胞的黏附(P<0.001)。对废培养基中的ETEC活性检测发现,与单独ETEC感染的IPEC-J2细胞相比,共培养的ETEC数量显著减少(P<0.001)。因此,Bacillus halotoleransSW207可能通过竞争性地获得IPEC-J2细胞上的结合位点,与IPEC-J2细胞黏附来抑制ETEC生长。
为分析Bacillus halotoleransSW207在ETEC感染细胞中产生NO的作用,通过共培养试验发现ETEC组NO水平显著高于对照组和Bacillus halotoleransSW207组(P<0.001)。Bacillus halotoleransSW207与ETEC共培养时,Bacillus halotoleransSW207显著降低了感染细胞中NO的含量,但仍高于未处理的对照细胞(P<0.001)。因此,Bacillus halotoleransSW207可能通过调节细胞内NO产生来发挥其对IPEC-J2细胞的保护作用。
为研究Bacillus halotoleransSW207能否减弱ETEC对细胞增殖的影响。采用ETEC与IPEC-J2细胞预孵育2h,并在8h后通过检测CCK-8发现细胞增殖显著下降,ETEC孵育的细胞增殖代谢活性明显低于对照细胞。当Bacillus halotoleransSW207与ETEC共培养8h后,细胞的代谢活性有显著提升(P<0.01)。这表明SW207能够恢复细胞增殖,并减弱ETEC对IPEC-J2细胞增殖的影响。
为验证Bacillus halotoleransSW207能否减轻ETEC对IPEC-J2细胞的炎症反应,检测了肿瘤坏死因子-α(TNF-α)、白介素6(IL-6)、白介素8(IL-8)、白介素1β(IL-1β)、白介素18(IL-18)、粒细胞巨噬细胞集落刺激因子(GM-CSF)和白介素10(IL-10)的相对表达。结果显示,Bacillus halotoleransSW207组与对照组并无差异。但ETEC组显著增加了TNF-α、IL-6、IL-8、IL-1β和IL-18的mRNAs水平(P<0.01)。在Bacillus halotoleransSW207与ETEC共培养后,上述炎性细胞因子水平显著下降(P<0.01),而GM-CSF和IL-10的mRNA表达显著提高(P<0.05)。
宿主防御肽(HDPs)是动物机体先天免疫系统的重要组成部分,分析了BD3、MUC1和PG-1在ETEC感染中的表达量。在Bacillus halotoleransSW207组和ETEC组中,PG1的mRNA表达均有增加的趋势,但差异不显著。Bacillus halotoleransSW207组显著增加了MUC1、BD3和PG1的mRNA表达(P<0.05,图4F)。在ETEC与Bacillus halotoleransSW207共培养后,PG-1的mRNA水平无明显变化,MUC1和BD3的mRNA水平显著上升(P<0.01)。因此,Bacillus halotoleransSW207通过减少或维持炎症和HDPs基因的动态平衡来调节模式识别受体的表达。
ETEC会导致宿主的肠道屏障功能失调,为探索Bacillus halotoleransSW207是否可以减轻ETEC导致的肠屏障功能下降,检测了Claudin-1、Occludin和Zona Occludens-1的mRNAs表达量。结果显示,Bacillus halotoleransSW207组没有改变肠道屏障功能基因水平,但ETEC显著降低细胞中Claudin-1、Occludin和ZO-1的mRNAs表达(P<0.01)。在共培养后,肠道屏障基因没有出现和ETEC组一样显著的下降趋势(图4中F)。因此,在ETEC感染的IPEC-J2细胞中加入Bacillus halotoleransSW207可能通过提高Occludin、Claudin-1和ZO-1的mRNA表达来增强肠屏障功能。
实施例5
Bacillus halotoleransSW207在防治断奶仔猪腹泻、促进断奶仔猪生长及提高免疫功能中的应用
试验日粮及试验设计:将12头28日龄松雷猪的断奶仔猪随机分为4组(对照组、Bacillus halotoleransSW207组、ETEC组和Bacillus halotoleransSW207+ETEC组)。所有猪都饲喂全价日粮:玉米一豆粕一乳制品型日粮配方参考NRC(2012)饲喂标准,自由饮水。Bacillus halotoleransSW207组和Bacillus halotoleransSW207+ETEC组在全价日粮基础上添加5×109CFU/kg的Bacillus halotoleransSW207。ETEC组和Bacillus halotoleransSW207+ETEC组在试验第14天开始添加1×108CFU/kg的产肠毒素大肠杆菌(ETEC K88,菌株购自中国兽医药品监察所菌种保藏中心),连续灌喂病原菌3天,而后评价观察96个小时。试验为期共21天,每天记录每只仔猪的生长情况。
影响断奶仔猪的生长性能:试验开始后每周记录采食量,每周进行称重。每天观察腹泻数量。根据以上数据计算:日采食量=总采食量/采食天数;日增重=(结束体重-起始体重)/生长天数;饲料转化率=阶段日采食量旧增重;腹泻率=腹泻总头数/记录天数/存栏数,其结果如图5中A所示。
肠道内容物消化酶活性:试验于21天后将所有猪屠宰,使用线绳在空肠,回肠、结肠的肠结处结扎,剪断结扎处后,立即将肠道内容物放至干净的塑料烧杯中,混匀后分装至无菌无酶的冻存管中,标记好后立即放入-80℃冰箱中保存。取回肠内容物测定纤维素酶、淀粉酶和蛋白酶活性水平。所有酶活性测定采用江苏酶免实业有限公司的ELISA试剂盒,方法按相应试剂盒说明书执行,其结果如图5中B所示。
血清生化及抗氧化指标:血液样品经3000rpm转速离心20min分离出血清。血清生化指标:丙氨酸氨基转移酶、血清白蛋白、谷丙转氨酶、血尿素氮、碱性磷酸酶、总胆固醇、天门冬氨酸氨基转移酶、血清总蛋白,血清生化指标检测于南京建成生物工程研究所;血清氧化应激指标,:丙二醛、过氧化氢、过氧化氢酶、总抗氧化能力、总超氧化物歧化酶、髓过氧化物酶、谷胱甘肽过氧化物酶、谷胱甘肽—S转移酶,试剂盒采购于南京建成生物工程研究所,方法按相应试剂盒说明书执行,其结果如图5中C、E所示。
血清免疫球蛋白、内毒素、D-乳酸和肠道sIgA:血液样品经3000rpm转速离心20min分离出血清。采用酶联免疫吸附(ELISA)试剂盒与酶标仪测定血清免疫球蛋白A(IgA)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)含量,ELISA试剂盒采购于南京建成生物工程研究所;检测血清内毒素和D-乳酸的含量,所用试剂盒采购于江苏宝莱生物科技有限公司;用载玻片轻挂起十二指肠、空肠和回肠的肠黏膜,检测分泌型免疫球蛋白A(sIgA)的含量。所用试剂盒采购于江苏酶免实业有限公司,方法按相应试剂盒说明书执行,其结果如图5中D所示。
回肠炎性因子水平:测定通过qPCR检测回肠中肿瘤坏死因-α(TNF-α)、白细胞介素-1β(IL-1β),白细胞介素-6(IL-6)、白细胞介素-8(IL-8)和白细胞介素-10(IL-10)的含量。首先用TRIzoI reagent提取回肠组织的总RNA,然后通过反转录试剂盒PrimeScript TMRT reagent Kit with gDNA Eraser对总RNA进行反转录合成cDNA。通过qRT-PCR反应试剂和Bio-Rad CFX384仪器检测黏液蛋白的表达量,扩增程序为:首先95℃ 30s,然后 95℃5s,60℃ 15s和72℃ 15s,进行40个循环。相应测定基因的引物序列如表1。其结果如图5中E所示。
回肠紧密连接蛋白基因、TLRs蛋白基因表达量:首先用TRIzoI reagent提取回肠组织的总RNA,然后通过反转录试剂盒PrimeScript TM RT reagent Kit with gDNAEraser对总RNA进行反转录合成cDNA。通过qRT-PCR反应试剂和Bio-Rad CFX384仪器检测黏液蛋白的表达量,扩增程序为:首先95℃ 30s,然后95℃ 5s,60℃ 15s和72℃ 15s,进行40个循环。相应测定基因的引物序列如表1。其结果如图5中F所示。
回肠H&E染色和PAS染色:用生理盐水清洗十二指肠、空肠和回肠,去除血液及其他内容物,然后取组织(3~5cm)并用4%多聚甲醛固定24 h,用70%、80%、90%、95%乙醇分别脱水2h,2h,1h和1h,再次用无水乙醇进行二次脱水1h,经二甲苯透明后,将组织浸入液体蜡中进行包埋切片,切片厚度约为5μm,然后将切片附着在载玻片上,在37℃环境下干燥12h。H&E染色、干燥、中性树脂固定、最后用盖玻片封片;PAS染色:首先从4%多聚甲醛溶液中取出仔猪十二指肠、空肠和回肠组织,洗净后用石蜡进行包埋。用切片机将包埋的组织切成约5μm厚的切片,然后用碘酸雪夫氏periodic acid schiff(PAS)染色试剂盒对肠道组织进行染色,具体步骤参照PAS染色试剂盒的说明书。肠道杯状细胞可被染成品红色,用光学显微镜在100倍和200倍的视野下对肠道组织进行拍照,,其结果如图6A、B所示。
试验结果:
与对照组相比,Bacillus halotoleransSW207组的仔猪体重在14d时显著升高(P<0.01)、平均日增重在14d内显著增加(P<0.001)、饲料转化率显著降低(P<0.001),同时Bacillus halotoleransSW207能显著降低断奶仔猪腹泻(P<0.05);大部分物质在仔猪的小肠中被消化和吸收,这里也存在各种消化酶。因此,评估了TL106对回肠内容物的肠道消化酶活性的影响,4组间纤维素酶活、淀粉酶活性和蛋白酶活性均有显著性差异。与对照组和疾病组相比,Bacillus halotoleransSW207组能显著提高小肠的三种酶活(P<0.01),促进了肠道营养物质的消化和吸收,最终实现促进断奶仔猪的生长;通过检测断奶仔猪的生化指标,不仅可以研究Bacillus halotoleransSW207对仔猪生理功能的影响,同时还可以确定Bacillus halotoleransSW207对仔猪是否有负面影响。
本研究发现饲喂Bacillus halotoleransSW207显著提高了断奶仔猪由ETEC K88引起的血清氧化应激下调,包括:过氧化氢酶(CAT)、总抗氧化能力(T-AOC)和总超氧化物歧化酶(T-SOD);显著降低了由ETEC K88引起的血清氧化应激上调。包括:丙二醛(MDA)、过氧化氢(H2O2)、髓过氧化物酶(MPO)和谷胱甘肽过氧化物酶(GSH-PX)。Bacillus halotoleransSW207缓解了由断奶和病原导致的氧化应激,并能提高断奶仔猪的抗氧化能力。
免疫球蛋白(Ig)是由浆细胞合成分泌的一组具有抗体活性的球蛋白,饲喂Bacillus halotoleransSW207显著提高了血清中IgG的含量,并显著提高了由ETEC K88引起的血清IgG和IgM的下降。sIgA为局部粘膜系统分泌,是粘膜免疫的主要成分,本研究发现Bacillus halotoleransSW207显著提高了十二指肠中sIgA的含量,并提高了由ETEC K88引起的十二指肠、空肠和回肠的sIgA下降(P<0.05);此外,断奶仔猪免疫Bacillus halotoleransSW207可以显著降低由ETEC K88引起的血清内毒素和D-乳酸升高(P<0.05),进一步维持了肠道上皮屏障功能;在免疫Bacillus halotoleransSW207后,断奶仔猪回肠的炎性因子水平菌显著降低,包括TNF-α(P<0.05)、IL-1β(P<0.05)、IL-6(P<0.01)、IL-8(P<0.05)和TLRs蛋白基因,同时显著提高了IL-10(P<0.01)的表达量,无论是对于健康还是腹泻仔猪均有良好的免疫作用,降低了促炎因子的表达,提高了抑炎因子的表达。
如图6所示,H&E染色发现Bacillus halotoleransSW207可显著增加攻毒后十二指肠、空肠和回肠的绒毛高度与隐窝深度的比值,改善仔猪的肠道粘膜组织的完整性。PAS染色发现,Bacillus halotoleransSW207可显著增加攻毒后十二指肠、空肠和回肠的肠道杯状细胞(可分泌黏液蛋白)的数量。上述结果表明,来自松雷猪的Bacillus halotoleransSW207能够降低断奶仔猪的腹泻率,增强了免疫力。通过增加肠道消化酶活力,降低肠道炎性因子表达,进而影响肠道内环境稳定、免疫调节和营养代谢,最终可观察到Bacillus halotoleransSW207对断奶猪生长性能的改善。
