Abstract
The invention discloses a traditional Chinese medicine composition for treating salpingitis of laying hens and a preparation method thereof. The invention is a pure traditional Chinese medicine preparation, has obvious treatment effect, reasonable formula, synergistic effect among the raw material components, no medicine residue, no drug resistance, no palatability problem, simple preparation process, low cost and the like. The traditional Chinese medicine composition has the effects of detumescence, clearing damp, clearing heat and detoxicating, soothing liver, strengthening spleen, nourishing yin, tonifying kidney and the like, and can effectively treat salpingitis of laying hens, quickly restore laying rate of the laying hens, improve egg quality and reduce poor egg yield.
Description
Technical Field
The invention belongs to the field of traditional Chinese medicines, and in particular relates to a traditional Chinese medicine composition for treating salpingitis of laying hens and a preparation method thereof.
Background
Salpingitis of laying hens is one of the frequently occurring diseases in the chicken raising industry, and is frequently generated in the peak period of laying hen. Laying hen salpingitis is clinically characterized by hemorrhagic salpingitis, abnormal egg production, and often discharge of yellow and white purulent secretions around cloaca, etc., and often causes peritonitis and oophoritis. The pathogenesis of salpingitis of laying hens is many and complex, and serious economic loss is caused for the chicken raising industry.
The main causes of salpingitis of laying hens are as follows: the sanitary condition of the raising chicken house is too bad, and the cloaca is polluted by bacteria such as escherichia coli, salmonella pullorum, paratyphoid bacillus and the like and invades the oviduct; overmuch animal feed is fed, the egg is produced too much or double-yolk eggs are produced, and sometimes the eggshells are broken in the oviducts to damage the oviducts; the feed lacks trace elements such as vitamin A, D, E; other factors; such as being stressed, misused with the drug, etc. When the oviduct is inflamed, the laying rate and the quality of eggs are firstly affected, the laying rate is reduced (10% -30%) and the number of inferior eggs is increased, secondary infection of other organs, even systemic infection, and even more serious, the death number of the chickens is increased. The sick chicken is mainly characterized by uneasiness, difficulty in lying down, and the laid eggs and eggshells often have blood stains, and the feathers around cloaca are polluted. The sectioning and inspection of the sick chicken can be seen; the abdominal cavity of the sick chicken has obvious lesions, the wall of the oviduct becomes thin, the follicular part shows swelling, water filling, bleeding, necrosis and mucosa adhesion, the follicular shrinkage is irregular, and broken yolk liquid and the like are arranged in the abdominal cavity.
According to the basic theory of Chinese veterinarian, the laying hen is difficult to clear due to exogenous pathogenic factors, excessive strain of egg production and the like, so that five endogenous pathogenic factors, damp-heat cementation and blood stasis are difficult to remove, qi and blood circulation are unsmooth, viscera function is disordered, and the damage can not be cleared, and new growth is caused, so that damp stagnation, blood stasis, qi stagnation, cold coagulation and deficiency of vital qi occur, so that the disease of salpingitis is aggravated due to the invasion of exogenous pathogenic factors such as dampness and the like. The recipe takes the effects of detumescence, clearing away heat and toxic material, soothing liver, strengthening spleen, nourishing yin, tonifying kidney and the like as the direction.
At present, most of the laying hen salpingitis diseases are treated by using antibiotic medicines, firstly, the antibiotics are easy to generate drug resistance, and the treatment is ineffective or easy to recur; secondly, a large amount of antibiotics can generate side effects and drug residues, and harmful residues can directly influence human health. The dialectical treatment of the traditional Chinese medicine has better treatment advantage, and the Chinese herbal medicine preparation has the characteristics of nature, no public hazard, green environmental protection, no drug resistance and no residue. The action plan of reducing resistance, forbidding resistance and product no-resistance of the breeding industry is accelerated. The application of traditional Chinese medicine in the breeding industry is indispensable, and the traditional Chinese medicine preparation is also a trend to replace antibiotic products in the breeding industry.
Disclosure of Invention
In order to solve the defects of the prior art and meet clinical requirements, the invention provides the traditional Chinese medicine composition for treating the salpingitis of the laying hens, which has the advantages of obvious treatment effect, reasonable formula, synergistic effect among the raw material components, no medicine residue, no medicine resistance and no palatability, simple preparation process and low cost, and the preparation method thereof.

the Chinese medicinal composition is prepared from, by weight, 20-30 parts of Chinese medicinal herbs, 10-20 parts of peltate yam herb, 8-12 parts of amaranthus auriculatus, 6-12 parts of large fruit euonymus alatus, 20-30 parts of azulene root, 10-20 parts of gloriope gloriosa, 6-12 parts of Chinese angelica, 6-12 parts of glossy privet fruit and 10-20 parts of camphol.
Preferably, the traditional Chinese medicine composition is prepared from the following traditional Chinese medicinal materials, by weight, 23-27 parts of selvedge, 12-18 parts of peltate yam herb, 9-11 parts of amaranthus auriculatus, 8-10 parts of euonymus alatus, 23-27 parts of azulene root, 12-18 parts of abacus root, 8-10 parts of Chinese angelica, 8-10 parts of glossy privet fruit and 12-18 parts of campanumoea.
More preferably, the traditional Chinese medicine composition is prepared from the following traditional Chinese medicinal materials in parts by weight, 20 parts of large tile, 20 parts of stinky tree, 10 parts of clematis chinensis, 10 parts of white flowers, 10 parts of glossy privet roots, 8 parts of green tangerine peel, 6 parts of liquorice, 12 parts of platycodon grandiflorum, 8 parts of lily and 8 parts of bighead atractylodes rhizome.
More preferably, the traditional Chinese medicine composition is characterized in that: the Chinese medicinal composition is granule, oral liquid, decoction, powder, pill or tablet.

The preparation for preparing the traditional Chinese medicine composition comprises the following preparation methods:
firstly, weighing the raw medicinal materials according to the parts by weight: 20-30 parts of well grass, 10-20 parts of shield fruit grass, 8-12 parts of amaranthus auriculata, 6-12 parts of large fruit euonymus alatus, 20-30 parts of azulene root, 10-20 parts of gloriosa albo-marginata root, 6-12 parts of Chinese angelica, 6-12 parts of glossy privet fruit and 10-20 parts of campanumaea acutifera; cleaning, oven drying, cutting, and mixing;
secondly, adding water which is 8-10 times of the total weight of all the medicinal materials, and soaking the sieved raw medicinal materials in the first step for 0.5-1h; then decocting with slow fire for 1.5h after boiling;
thirdly, filtering the traditional Chinese medicine in the second step, and adding water which is 7-9 times of the total weight of the medicinal materials into filter residues for decoction for 1h;
fourth, filtering the traditional Chinese medicine in the third step, and adding water which is 6-8 times of the total weight of the medicinal materials for decoction for 1h;
fifthly, merging the three decocted liquid medicines, standing for 24 hours, concentrating under reduced pressure until the relative density is 1.12-1.16, and cooling for standby;
and sixthly, slowly adding 70% ethanol with the amount of 2-2.5 times of the weight of the liquid medicine into the traditional Chinese medicine fluid extract obtained in the fifth step, standing for 24-48h, uniformly stirring, hermetically refrigerating for 24-48h, filtering, recovering the ethanol of the filtrate, and concentrating to the relative density of 1.33-1.36.
Seventh, the traditional Chinese medicine fluid extract obtained in the sixth step is spray-dried, granulated, sterilized and sub-packaged to obtain the traditional Chinese medicine compound preparation.
The invention also provides application of the traditional Chinese medicine composition in treating salpingitis of laying hens.
The medicinal properties and effects of the medicinal materials in the traditional Chinese medicine composition are as follows:
radix seu herba Physalis Pubescentis; taste: bitter; and (5) cooling. Channel tropism: a core; the large intestine channel. Clear away heat and toxic materials, and relieve swelling. Can be used for treating carbuncle, furuncle, bacillary dysentery, and enteritis.
Grass on the well side; taste: octyl; slightly bitter; and (5) cooling. Channel tropism: liver; large intestine; the bladder meridian. Clearing heat and promoting diuresis; promoting blood circulation and removing swelling. Primary diarrhea; diarrhea; edema; hepatitis; cholecystitis; throat obstruction; urinary system infection; carbuncle, swelling and sore; rheumatalgia; swelling and pain from injuries from falls.
Inverted zaolone; taste: octyl; bitter; and (5) cooling. Channel tropism: a lung; the liver meridian. Pain relieving, antiinflammatory, menstruation regulating. It is mainly used for treating furuncle, sore, and menoxenia.
azulene root; taste: sweet; flat. Channel tropism: liver; spleen meridian. Clear away damp-heat and detumescence toxin. It is mainly used for treating jaundice, damp arthralgia, heat stranguria, dysentery, swelling toxin, scrofula, etc.
Amaranthus auriculatus (L.) Spreng; taste: sweet in taste; light; the sex is flat. Channel tropism: spleen invigorating; the bladder meridian. Invigorating spleen and promoting diuresis; promoting qi circulation and removing blood stasis. Anorexia due to spleen deficiency; chest fullness and stuffiness; acute and chronic cystitis; women's leukorrhagia; traumatic injury, stasis and swelling and pain.
Celastrus angulatus; taste: sweet; slightly bitter; flat. Channel tropism: liver; spleen; kidney meridian. Kidney-tonifying and waist-strengthening; remove blood stasis and promote diuresis. Lumbago due to kidney deficiency; restlessness of fetal movement; chronic nephritis; lochia after childbirth is not complete; traumatic fracture; rheumatalgia; and (5) taking off.
Root of glochidion; taste: bitter and flat. Channel tropism: liver; the large intestine channel. Clear heat and promote diuresis, activate blood and detoxify. It is indicated for dysentery, malaria, jaundice, turbid urine, rheumatalgia, metrorrhagia, leukorrhagia, carbuncle, scrofula, etc.
Chinese angelica root; sex taste: sweet, pungent and warm. Channel tropism: it enters liver, heart and spleen meridians. Replenishing blood, regulating menstruation, relieving pain, moistening dryness and smoothing intestine. Treating menoxenia, amenorrhea, abdominal pain, abdominal mass, metrorrhagia; headache due to blood deficiency, dizziness, flaccid paralysis; difficulty in treating constipation, and severe red dysentery; carbuncles, sores, and the like.
Fructus Ligustri Lucidi; taste: bitter, sweet and flat. Channel tropism: liver; kidney meridian. Nourishing liver and kidney, improving eyesight and blackening hair. Can be used for treating soreness of waist and knees, clearing heat and improving eyesight.
The invention has the beneficial effects that: in the recipe, the selvedge and the azulene root are taken as monarch drugs, and the two drugs together have the effects of detumescence and clearing damp and heat. The campanumaea pilosula, the peltate yam and the root of the glabrous greenbrier rhizome are taken as ministerial drugs, and the three drugs act together to play roles of clearing heat, promoting diuresis, activating blood, removing toxicity, diminishing inflammation, relieving pain, and enhancing the detumescence and dampness removal of the monarch drug. The large fruit euonymus has the effects of tonifying kidney, strengthening waist, removing blood stasis and promoting diuresis; the amaranthus auriculatus can invigorate spleen, promote diuresis, promote qi circulation and remove blood stasis; dang Gui can enrich and harmonize blood, regulate menstruation and relieve pain, moisten dryness and smooth intestines; the glossy privet fruit can nourish liver and kidney, clear heat and improve eyesight, and the large fruit euonymus alatus, the amaranthus auriculata, the Chinese angelica and the glossy privet fruit are used as adjuvant drugs together to achieve the effects of tonifying liver and spleen, nourishing yin and tonifying kidney. Example 1
A traditional Chinese medicine composition for treating salpingitis of laying hens is prepared from the following raw materials in parts by weight: 28 parts of well grass, 13 parts of shield fruit grass, 9 parts of amaranthus auriculata, 10 parts of large fruit euonymus, 20 parts of azulene root, 12 parts of abacus root, 8 parts of Chinese angelica, 8 parts of glossy privet fruit and 15 parts of campanumoea.
The traditional Chinese medicine composition for treating salpingitis of laying hens is prepared according to the following steps:
firstly, weighing the raw materials in parts by weight: 28 parts of well grass, 13 parts of peltate yam herb, 9 parts of amaranthus auriculatus, 10 parts of large fruit euonymus alatus, 20 parts of azulene root, 12 parts of abacus root, 8 parts of Chinese angelica, 8 parts of glossy privet fruit and 15 parts of campanumaea pilosula; cleaning, oven drying, cutting, and mixing;

fifthly, merging the three decocted liquid medicines, standing for 24 hours, concentrating under reduced pressure until the relative density is 1.12-1.16, and cooling for standby;
and sixthly, slowly adding 70% ethanol by weight which is 2.5 times of the weight of the liquid medicine into the traditional Chinese medicine fluid extract obtained in the fifth step, standing for 24 hours, uniformly stirring, hermetically refrigerating for 24 hours, filtering, recovering the ethanol of the filtrate, and concentrating to the relative density of 1.33-1.36.
Seventh, the traditional Chinese medicine fluid extract obtained in the sixth step is spray-dried, prepared into granules, sterilized and packaged to obtain the traditional Chinese medicine compound preparation.
Example 2

sixthly, slowly adding 70% ethanol in the Chinese medicinal fluid extract in the fifth step, wherein the amount of the ethanol is 2.5 times of the weight of the liquid medicine, standing for 24 hours, uniformly stirring, hermetically refrigerating for 24 hours, filtering, and recovering the ethanol from the filtrate to obtain refined liquid; then adding water for dissolving, heating to completely dissolve, standing and refrigerating for more than 12 hours, filtering, concentrating the liquid medicine until the crude drug is 1g/mL, packaging, and sterilizing to obtain the oral liquid.

Can be used for treating jaundice due to damp-heat, swelling and itching of yin, leukorrhagia, strengthening middle-jiao, eczema and itching, conjunctival congestion, deafness, hypochondriac pain, bitter taste, and convulsion. The present example is another comparative example of a traditional Chinese medicine prescription for treating salpingitis of laying hens, in which the root of gloriosa is replaced by Ligusticum wallichii, and the difference between the present comparative example and example 1 is that the raw materials (in parts by weight): 15 parts of well grass, 15 parts of peltate yam, 15 parts of amaranthus auriculatus, 15 parts of large fruit euonymus, 15 parts of azulene root, 15 parts of ligusticum wallichii, 15 parts of Chinese angelica, 15 parts of glossy privet fruit and 15 parts of campholly.
Ligusticum wallichii; taste: temperature is high; pungent. Channel tropism: liver, gallbladder and pericardium channels. Promoting blood circulation, activating qi-flowing, dispelling pathogenic wind and relieving pain. Can be used for soothing nerves, relieving headache, abdominal pain, stabbing pain in chest and hypochondrium, swelling and pain from traumatic injury, headache, and rheumatalgia.

This example is another comparative example of a traditional Chinese medicine formulation for treating salpingitis of laying hens, which is different from example 1 in the raw materials (in parts by weight) without adding gloriosa, angelica and glossy privet fruit: 25 parts of well grass, 15 parts of peltate yam herb, 10 parts of amaranthus auriculatus, 10 parts of large fruit euonymus alatus, 25 parts of azulene root and 15 parts of campholone.
The preparation method is described in example 1.
Comparative example 7
This example is another comparative example of a traditional Chinese medicine prescription for treating salpingitis of laying hens, in which azulene root is replaced with rheum officinale, and the difference between this comparative example and example 1 is that the raw materials (in parts by weight): 28 parts of well grass, 13 parts of peltate yam herb, 9 parts of amaranthus auriculatus, 10 parts of large fruit euonymus, 20 parts of rheum officinale, 12 parts of radix polygoni multiflori, 8 parts of Chinese angelica, 8 parts of glossy privet fruit and 15 parts of campholly root.
Radix et rhizoma Rhei; taste: bitter and cold. Channel tropism: stomach, liver and large intestine channels. Purging excessive heat, removing accumulation and stagnation, and promoting blood stasis. The main indications are: constipation due to excessive heat, food stagnation, abdominal pain, acute appendicitis, acute infectious hepatitis, amenorrhea due to blood stasis, carbuncle, swelling and ulcer.

Test example 1 the effect verification of the Chinese medicinal composition of the invention on salpingitis of artificially infected laying hens
1. Test materials
1.1 test animals
The 27-week-old sea orchid brown layer chicken is purchased from Beijing Fuhao experimental animal cultivation center.
1.2 Test reagent
Phenol (formulated as a 15% phenol solution) was purchased from Shanghai Meilin Biochemical technologies Co.
10% amoxicillin soluble powder, purchased from North China pharmaceutical group animal health products Limited company.
2. Test method
2.1 Test group and administration
2.2 Test method
Except for the blank control group, the equivalent normal saline is injected, and the other groups of laying hens are injected to the position of 2-3cm of the oviduct according to 1mL of phenol reagent per feather.
Starting from the 1 st day of molding, the blank control group is not dosed and is fed normally; the other groups are continuously administered for 7 days according to the medicine mixture or mixed water, wherein the western medicine control group is amoxicillin; test groups 1-7 were dosed as examples 1-7, respectively, in the present invention; comparative groups 1-7 were dosed as comparative examples 1-7, respectively, in the present invention. The method comprises the steps of feeding test laying hens normally for 2 times a day, observing the egg yield and abnormal egg conditions of the test laying hens, wherein inferior eggs comprise blood spot eggs, fecal eggs, soft preserved eggs, sand preserved eggs, broken eggs and the like, and observing and recording the mental condition, appetite, morbidity and cutting and examining of dead laying hens.
Test treatment criteria; the cure is that the mental state and feeding of the sick chickens are recovered to be normal, the egg yield is close to that of a blank control group, and the inferior egg yield is reduced or not. The effective method is that the appetite and the feeding of the sick chickens are recovered to be normal, the egg yield is improved, and the inferior egg yield is gradually reduced. The invalidation means that after the administration, the mental state and the feeding condition of the sick chickens are not recovered, the egg yield is not improved, and the inferior egg yield is not reduced.
3. Test results
Table 1 tests of the Effect of each group of drugs on salpingitis of artificially infected laying hens
As can be seen from the test data in Table 1, the cure effective rates of the test groups 1 to 7 are higher than those of the western medicine control group; the laying rate of the test group 3 (88.1%) is higher than that of the western medicine control group (86.9%) and other test groups; inferior egg rates of test group 3 (12.2%), test group 6 (12.2%) and test group 4 (13.1%) were lower than western control group (13.7%); test groups 1-7 did not die during the test period, while the control groups had different degrees of layer death during the test period, the anatomical test results were shown as C1 and C2 in FIG. 1, and C3 in FIG. 1 was a blank control group layer profile. The test results show that the traditional Chinese medicine composition prepared by the invention can effectively treat salpingitis of laying hens, improve laying rate, improve egg quality and reduce death rate of the laying hens.
Test example 2 the effect verification of the Chinese medicinal composition of the invention in clinic for treating salpingitis of laying hens
The clinical test chicken flock is 6000 feathers of 350-day-old laying hens and is cultivated by a certain raiser in the Pinggu area of Beijing city. The laying hen of the house is listless, inappetence, camping, belly swelling, fecal matter attached to feathers around cloaca, the egg yield of the whole house is continuously reduced, the egg shells are provided with blood marks, soft shell eggs, sand shell eggs and broken shell eggs, and the laying hen of the house is judged to have salpingitis. The average laying hen of the house is divided into 6 groups of 1000 laying hens, and the blank control group is normally fed without drug administration; the western medicine control group is given with amoxicillin 0.3% mixed material and continuously administered for 7 days; the medicines prepared in example 3 and example 6 of the present invention were used in test group 1 and test group 2, respectively, and the medicines prepared in comparative example 1 and comparative example 6 of the present invention were used in comparative group 1 and comparative group 2, respectively, and were continuously administered for 7 days in accordance with a 0.4% mix.

As can be seen from the test data in table 2, the egg laying rates of the test group 1 and the test group 2 are higher than those of the western medicine control group and the blank control group; the inferior egg rate and the death rate of the test group 1 and the test group 2 are lower than those of the western medicine control group, the contrast group and the blank control group.
Proved by clinical verification results, the traditional Chinese medicine composition prepared by the invention can effectively improve the laying rate, improve the egg quality and reduce the death rate of the laying hens, and the traditional Chinese medicine composition prepared by the invention can effectively treat salpingitis diseases of the laying hens.
According to the traditional Chinese medicine composition disclosed by the invention, the compatibility of medicines is reasonable, the pharmacological effects of the medicines are fully exerted according to the syndrome differentiation and treatment principle of a traditional Chinese medicine veterinarian, and the combination of the medicines can achieve the effects of synergistic effect, and the traditional Chinese medicine composition has the effects of detumescence, dampness elimination, heat clearing, detoxification, liver soothing, spleen strengthening, yin nourishing, kidney tonifying and the like. The traditional Chinese medicine compound preparation formulation has no influence on the drug effect, and test results show that the powder and the granules are effective in preventing and treating salpingitis of laying hens.
Claims (7)
1. The traditional Chinese medicine composition for treating salpingitis of laying hens is characterized by comprising the following traditional Chinese medicinal materials in parts by weight: 20-30 parts of well grass, 10-20 parts of shield fruit grass, 8-12 parts of amaranthus auriculata, 6-12 parts of large fruit euonymus alatus, 20-30 parts of azulene root, 10-20 parts of glorious gloriosa root, 6-12 parts of Chinese angelica, 6-12 parts of glossy privet fruit and 10-20 parts of campanumaea acum.
2. The traditional Chinese medicine composition for treating salpingitis of laying hens according to claim 1 is characterized by comprising the following traditional Chinese medicinal materials in parts by weight: 23-27 parts of well grass, 12-18 parts of shield fruit grass, 9-11 parts of amaranthus auriculata, 8-10 parts of large fruit euonymus alatus, 23-27 parts of azulene root, 12-18 parts of abacus root, 8-10 parts of Chinese angelica, 8-10 parts of glossy privet fruit and 12-18 parts of campanumoea.

The traditional Chinese medicine composition for treating salpingitis of laying hens according to claim 2 is characterized by comprising the following traditional Chinese medicinal materials in parts by weight: 25 parts of well grass, 15 parts of peltate yam herb, 10 parts of amaranthus auriculatus, 10 parts of large fruit euonymus alatus, 25 parts of azulene root, 15 parts of gloriosa root, 10 parts of angelica sinensis, 10 parts of glossy privet fruit and 15 parts of campanumoea.
4. The traditional Chinese medicine composition for treating salpingitis of laying hens according to claim 1, 2 or 3, further comprising pharmaceutically acceptable auxiliary materials.
5. The traditional Chinese medicine composition for treating salpingitis of laying hens according to claim 4, wherein the traditional Chinese medicine composition is: granule, oral liquid, decoction, powder, pill or tablet.
6. The traditional Chinese medicine composition according to claim 5, wherein the dosage form of the traditional Chinese medicine composition is a granule.
7. A method for preparing the traditional Chinese medicine composition according to claim 1, which is characterized in that the method comprises the following steps:
firstly, weighing the raw medicinal materials according to the parts by weight: 20-30 parts of well grass, 10-20 parts of shield fruit grass, 8-12 parts of amaranthus auriculata, 6-12 parts of large fruit euonymus alatus, 20-30 parts of azulene root, 10-20 parts of gloriosa albo-marginata root, 6-12 parts of Chinese angelica, 6-12 parts of glossy privet fruit and 10-20 parts of campanumaea acutifera; cleaning, oven drying, cutting, and mixing;
secondly, adding water which is 8-10 times of the total weight of all the medicinal materials, and soaking the sieved raw medicinal materials in the first step for 0.5-1h; then decocting with slow fire for 1.5h after boiling;
thirdly, filtering the traditional Chinese medicine obtained in the second step, and adding water which is 7-9 times of the total weight of the medicinal materials into filter residues for decoction for 1h;
fourth, filtering the traditional Chinese medicine obtained in the third step, and adding water which is 6-8 times of the total weight of the medicinal materials to decoct for 1h;
fifthly, merging the three decocted liquid medicines, standing for 24 hours, concentrating under reduced pressure until the relative density is 1.12-1.16, and cooling for standby to obtain traditional Chinese medicine fluid extract;
step six, slowly adding 70% ethanol with the amount of 2-2.5 times of the weight of the liquid medicine into the traditional Chinese medicine fluid extract obtained in the step five, standing for 24-48h, uniformly stirring, hermetically refrigerating for 24-48h, filtering, recovering the ethanol of the filtrate, and concentrating to obtain the traditional Chinese medicine fluid extract with the relative density of 1.33-1.36;
seventh, spray drying, granulating, sterilizing and sub-packaging the traditional Chinese medicine fluid extract obtained in the sixth step.
