Abstract
The invention discloses a plant essential oil skin disinfectant and a preparation method thereof, wherein the preparation method comprises the steps of sequentially adding elsholtzia oil, zedoary turmeric oil, geranium oil, citronella oil, eucalyptus oil, strawberry seed oil, valerian oil and peppermint oil into ethanol, uniformly stirring, adding tween 20 and 1, 2-propanediol under stirring, stirring until liquid is clear, adding modified hydroxypropyl-beta-cyclodextrin, rose oil and bitter orange flower oil, stirring to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring and standing, clarifying the solution, filtering and split charging to obtain the plant essential oil skin disinfectant; the disinfectant adopts modified hydroxypropyl-beta-cyclodextrin, tween-20 and 1, 2-propylene glycol as solubilizers, and the hydroxypropyl-beta-cyclodextrin has good solubilization effect and inclusion stability, has solubilization effect on plant essential oil, enables the essential oil to more easily exert antibacterial effect, and can be used for daily disinfection and sterilization of skin tissues, mosquito repelling and aromatic refreshment.
Description
Technical Field
The invention relates to the field of skin disinfectant, in particular to a plant essential oil skin disinfectant and a preparation method thereof.
Background
The disinfectant is a preparation for killing pathogenic microorganisms on a transmission medium to achieve harmless requirements, can kill the pathogenic microorganisms outside a human body, has the effects of strong sterilization capability, low cost and easy obtainment, and is widely applied to daily life.
The sterilizing liquid in the market at present has various types, and common sterilizing liquid containing iodine (iodophor and the like), alcohols (ethanol and isopropanol), phenols (parachlorometaxylenol, drop dew and Weilu), strong oxidants (peracetic acid and 84 sterilizing liquid), quaternary amine salts (benzalkonium chloride, benzalkonium bromide, organic guanidine) and the like. Although the types of disinfectant in the market are various at present, ideal disinfectant is difficult to seek, and the disinfectant has advantages and disadvantages: iodine-containing disinfection solution such as iodophor can effectively kill various pathogens such as bacteria, spores and fungi, but is only effective on a very small part of DNA viruses, and in addition, the iodophor is not suitable for daily hand disinfection due to the darker color, and is commonly used for disinfecting skin mucous membranes in medical environments. The phenolic disinfectant is a broad-spectrum mildew-proof antibacterial agent, has the effect of killing most bacterial propagules and fungi, and has no inactivation effect on viruses. The strong oxidant disinfectant has strong corrosiveness to metal, clothes, leather and skin mucous membrane, so that the application range is limited, and the hand disinfectant is not suitable for hand skin disinfection. The quaternary amine salt disinfectant belongs to a dual-function disinfectant, can kill bacteria and fungi and inactivate viruses, and has great advantages compared with the disinfectants. However, the quaternary ammonium salt disinfectant needs to be washed by water after being disinfected, and hands are wet and cold in winter, so that the quaternary ammonium salt disinfectant is very inconvenient. All of the above-mentioned sterilizing solutions have unsatisfactory places.
Essential oils are a general term for volatile oily liquid substances with fragrance and (or fragrance) extracted from leaves, flowers, seeds, fruits, roots, barks, resins, wood cores and the like of plants. The plant essential oil has better antibacterial and antiviral effects, has the advantages of high safety, wide sterilization spectrum and the like, is widely applied to the industries of medicine, food health care and cosmetics, and is widely favored by the market.
The Chinese patent document CN201110113813.8 discloses a plant essential oil sterilizing disinfectant which is prepared from the following raw materials in percentage by weight: eucalyptus oil 0.3%, tea tree oil 0.4%, mugwort oil 0.3%, rosemary oil 0.5%, bergamot oil 0.5%, lemon oil 0.5%, lavender oil 0.7%, clove oil 0.2%, thyme oil 0.4%, and the balance of diluent. Compared with the traditional skin bactericide and the bactericide of sanitary appliances, the disinfectant is prepared from brand-new pure natural plant essential oil and has the characteristics of natural environment protection, good sterilization and disinfection effects, comfortable smell, no harm to human bodies and benefit to physical and mental health of people.
However, the plant essential oil disinfectant on the market at present has the advantages that the disinfection effect is to be improved, the color stability is not high, the color is easy to change after being placed for a period of time in an open mode, the appearance and the color of the product are deteriorated, and the use experience of consumers is affected.
Disclosure of Invention
In order to solve the defects in the prior art, the invention aims to provide the plant essential oil skin disinfectant which can be used for daily disinfection and sterilization of skin tissues and the like, mosquito repelling, aromatic refreshment and has high color stability and no color change caused by metal ions.
In order to achieve the above purpose, the present invention adopts the following technical scheme:
the plant essential oil skin disinfectant is prepared from the following components in parts by weight: 6-10 g/L of elsholtzia oil, 3-6 g/L of zedoary turmeric oil, 1-3 g/L of geranium oil, 1-3 g/L of citronella oil, 1-3 g/L of eucalyptus oil, 3-6 g/L of strawberry seed oil, 2-4 g/L of valerian oil, 1-3 g/L of peppermint oil, 1-2 g/L of rose oil, 1-3 g/L of bitter orange flower oil, 20-70 g/L of tween, 50-70 g/L of 1, 2-propanediol, 5-20 g/L of modified hydroxypropyl-beta-cyclodextrin, 240-280 g/L of ethanol and the balance of deionized water.
Preferably, each liter of skin disinfectant is prepared from the following components in parts by weight: 8g/L of elsholtzia oil, 5g/L of zedoary turmeric oil, 2g/L of geranium oil, 2g/L of citronella oil, 2g/L of eucalyptus oil, 4g/L of strawberry seed oil, 3g/L of valerian oil, 2g/L of peppermint oil, 1g/L of rose oil, 2g/L of bitter orange flower oil, 60g/L of tween 20, 60g/L of 1, 2-propylene glycol, 10g/L of modified hydroxypropyl-beta-cyclodextrin, 250g/L of ethanol and the balance of deionized water.
Preferably, the preparation method of the modified hydroxypropyl-beta-cyclodextrin comprises the following steps:
dissolving cystine in NaOH solution, adding hydroxypropyl-beta-cyclodextrin, adding epichlorohydrin after complete dissolution, stirring for reaction, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin.

Elsholtzia oil: can inhibit the activity of bacteria, viruses, anaerobic bacteria and acidogenic enzymes, and has the effects of accelerating the volatilization of sweat, removing oil and sweat;
zedoary turmeric oil: can be used for treating blood qi and heart pain, food stagnation, abdominal distention and pain, amenorrhea due to blood stagnation, dysmenorrhea, abdominal mass, and traumatic injury;

Compared with the prior art, the invention has the following beneficial effects:
1) The invention provides a plant essential oil skin disinfectant, which adopts modified hydroxypropyl-beta-cyclodextrin, tween 20 and 1, 2-propanediol as solubilizers, and has synergistic solubilization effect, thus increasing the solubility of essential oil small molecules and ensuring the stability of a disinfectant system; the water solubility of the hydroxypropyl-beta-cyclodextrin is greatly increased compared with that of beta-cyclodextrin, meanwhile, the cavity structure of the beta-cyclodextrin is maintained, the hydroxypropyl-beta-cyclodextrin has good solubilization effect and inclusion stability, the solubilization effect on plant essential oil is achieved, the solubility of the essential oil is increased, the essential oil can exert the antibacterial effect more easily, and the prepared disinfectant can be used for daily disinfection and sterilization of skin tissues and the like, mosquito repelling and aromatic refreshment.
2) The invention provides a preparation method of strawberry seed oil, which is characterized in that mould is added for fermentation pretreatment before extracting essential oil from fresh strawberry seeds, aspergillus niger grows in crushed strawberry seeds to generate polysaccharide hydrolase such as cellulase, pectase, xylanase and the like, and the cell wall structure is destroyed, so that the essential oil is released from cells, and the extraction rate of the essential oil is improved; in the process of extracting essential oil by ethanol, microwave treatment is carried out simultaneously, and microwave energy penetrates through ethanol medium to reach the inside of strawberry seed cells, so that the temperature inside the cells rises rapidly, the internal pressure exceeds the bearing capacity of cell walls to break, active ingredients in plant cells flow out, are captured and dissolved by ethanol under the condition of lower temperature, and the extraction efficiency is improved; compared with the conventional method without fermentation pretreatment and microwave treatment, the extraction yield of the strawberry seed oil is effectively improved.
3) The invention provides modified hydroxypropyl-beta-cyclodextrin, which is prepared by grafting cystine onto hydroxypropyl-beta-cyclodextrin under alkaline conditions by taking hydroxypropyl-beta-cyclodextrin as a raw material and epoxy chloropropane as a cross-linking agent, wherein carboxyl, amino and disulfide bonds are introduced into the modified hydroxypropyl-beta-cyclodextrin, so that the modified hydroxypropyl-beta-cyclodextrin has hydrophobicity and complexation simultaneously, can have inclusion effect with small molecules of hydrophobic plant essential oil, plays an excellent solubilization effect, can also have coordination effect with metal ions, prevents the essential oil from influencing the color and appearance of a product due to the color change of the metal ions, and plays a role in stabilizing essential oil compound components and resisting oxidation.

Of course, the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
Unless otherwise specified, both chemical reagents and materials in the present invention are purchased through a market route or synthesized from raw materials purchased through a market route.
Aspergillus niger was GIM3.546.
The invention will be further illustrated by the following examples.
Example 1
The preparation method of the strawberry seed oil comprises the following steps:
drying strawberry seeds, crushing, sieving with a 100-mesh sieve to obtain strawberry seed powder, adding deionized water to enable the water content to be 50wt%, adding molasses and aspergillus niger which are 10% of the weight of the strawberry seed powder, and intermittently ventilating and fermenting for 12 days at 35 ℃ to obtain strawberry seed fermented products; mixing the strawberry seed ferment with 95wt% ethanol solution, soaking at normal temperature for 12h, performing interval microwave treatment with 800W microwave for 60s, stopping for 5min, taking supernatant, removing solvent by rotary evaporation, and centrifuging at 10000r/min for 10min to obtain strawberry seed oil.
Example 2
A preparation method of a plant essential oil skin disinfectant comprises the following steps:
(1) Dissolving 12g of cystine in 1mol/L NaOH solution, adding 9g of hydroxypropyl-beta-cyclodextrin, adding 12g of epichlorohydrin after complete dissolution, magnetically stirring at 80 ℃ and 800r/min for reaction for 3 hours, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin;
(2) Sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, uniformly stirring, adding 60g of tween 20 and 60g of 1, 2-propanediol under stirring, stirring until liquid is clear, adding 15g of modified hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring for 60min, standing for 18h, and obtaining the plant essential oil skin disinfectant after solution is clear, filtering and split charging.
Example 3
A preparation method of a plant essential oil skin disinfectant comprises the following steps:
(1) Dissolving 10g of cystine in 1mol/L NaOH solution, then adding 9g of hydroxypropyl-beta-cyclodextrin, adding 8g of epichlorohydrin after complete dissolution, magnetically stirring at 60 ℃ and 500r/min for reaction for 1h, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin;
(2) Sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, uniformly stirring, adding 60g of tween 20 and 60g of 1, 2-propanediol under stirring, stirring until liquid is clear, adding 15g of modified hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring for 60min, standing for 18h, and obtaining the plant essential oil skin disinfectant after solution is clear, filtering and split charging.
Example 4
A preparation method of a plant essential oil skin disinfectant comprises the following steps:
(1) Dissolving 11g of cystine in 1mol/L NaOH solution, then adding 9g of hydroxypropyl-beta-cyclodextrin, adding 9g of epichlorohydrin after complete dissolution, magnetically stirring at 7 ℃ and 600r/min for reaction for 2 hours, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin;
(2) Sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, uniformly stirring, adding 60g of tween 20 and 60g of 1, 2-propanediol under stirring, stirring until liquid is clear, adding 15g of modified hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring for 60min, standing for 18h, and obtaining the plant essential oil skin disinfectant after solution is clear, filtering and split charging.
Example 5
A preparation method of a plant essential oil skin disinfectant comprises the following steps:
(1) Dissolving 11g of cystine in 1mol/L NaOH solution, then adding 9g of hydroxypropyl-beta-cyclodextrin, adding 11g of epichlorohydrin after complete dissolution, magnetically stirring at 70 ℃ and 700r/min for reaction for 2.5 hours, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin;
(2) Sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, uniformly stirring, adding 60g of tween 20 and 60g of 1, 2-propanediol under stirring, stirring until liquid is clear, adding 15g of modified hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring for 60min, standing for 18h, and obtaining the plant essential oil skin disinfectant after solution is clear, filtering and split charging.
Comparative example 1
A preparation method of skin disinfectant comprises the following steps:
sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, uniformly stirring, adding 60g of tween 20 and 60g of 1, 2-propanediol under stirring, stirring until liquid is clarified, adding 15g of hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water until the total amount is 1000mL, stirring for 60min, standing for 18h, and obtaining the skin disinfectant after solution is clarified, filtering and split charging.
Comparative example 2
A preparation method of skin disinfectant comprises the following steps:
sequentially adding 10g of elsholtzia oil, 5g of zedoary turmeric oil, 2g of geranium oil, 2g of citronella oil, 3g of eucalyptus oil, 4g of strawberry seed oil, 3g of valerian oil and 2g of peppermint oil into 250g of 95wt% ethanol, stirring until the liquid is clear, adding 15g of modified hydroxypropyl-beta-cyclodextrin, 2g of rose oil and 2g of bitter orange flower oil, stirring for 60min to obtain emulsion, adding deionized water to the total amount of 1000mL, stirring for 60min, standing for 18h, clarifying the solution, filtering and packaging to obtain the skin disinfectant.
Comparative example 3
The preparation method of the strawberry seed oil comprises the following steps:
drying strawberry seeds, crushing and sieving with a 100-mesh sieve to obtain strawberry seed powder; mixing strawberry seed powder with 95wt% ethanol solution, soaking at normal temperature for 12h, performing interval microwave treatment with 800W microwave for 60s, stopping for 5min, taking supernatant, removing solvent by rotary evaporation, and centrifuging at 10000r/min for 10min to obtain strawberry seed oil.
Comparative example 4
The preparation method of the strawberry seed oil comprises the following steps:

The plant essential oil skin disinfectant is characterized by comprising the following components in parts by weight per liter: 6-10 g/L of elsholtzia oil, 3-6 g/L of zedoary turmeric oil, 1-3 g/L of geranium oil, 1-3 g/L of citronella oil, 1-3 g/L of eucalyptus oil, 3-6 g/L of strawberry seed oil, 2-4 g/L of valerian oil, 1-3 g/L of peppermint oil, 1-2 g/L of rose oil, 1-3 g/L of bitter orange flower oil, 20-70 g/L of tween, 50-70 g/L of 1, 2-propanediol, 5-20 g/L of modified hydroxypropyl-beta-cyclodextrin, 240-280 g/L of ethanol and the balance of deionized water.

The plant essential oil skin disinfectant is characterized by comprising the following components in parts by weight per liter: 8g/L of elsholtzia oil, 5g/L of zedoary turmeric oil, 2g/L of geranium oil, 2g/L of citronella oil, 2g/L of eucalyptus oil, 4g/L of strawberry seed oil, 3g/L of valerian oil, 2g/L of peppermint oil, 1g/L of rose oil, 2g/L of bitter orange flower oil, 60g/L of tween 20, 60g/L of 1, 2-propylene glycol, 10g/L of modified hydroxypropyl-beta-cyclodextrin, 250g/L of ethanol and the balance of deionized water.
3. The skin disinfectant according to claim 1, characterized in that the preparation method of the modified hydroxypropyl-beta-cyclodextrin is as follows:
dissolving cystine in NaOH solution, adding hydroxypropyl-beta-cyclodextrin, adding epichlorohydrin after complete dissolution, stirring for reaction, adding hydrochloric acid to adjust the pH of the reaction solution to be neutral, precipitating with DMF, carrying out suction filtration, dissolving the obtained solid with distilled water, precipitating with DMF, carrying out suction filtration, soaking the solid with DMF to remove unreacted hydroxypropyl-beta-cyclodextrin, filtering, freezing and drying to obtain modified hydroxypropyl-beta-cyclodextrin.
