神经保护旨在防止神经元死亡、再生神经元网络和改善脑功能障碍,抑制神经炎症和氧化应激反应是进行神经保护干预主要措施。神经炎症几乎是所有脑部病变的开始,炎症发生过程中伴随着如氧化应激、线粒体障碍、内质网应激等许多细胞功能的改变,从而脑损伤并导致各种脑部疾病的发生和发展。此外,机体内自由基的产生和抗氧化防御系统之间的不平衡会导致氧化应激现象,主要表现为超氧化物(ROS)和一氧化氮(NO)的过量形成。大脑由于耗氧量大和缺乏能够被氧化的脂质,容易发生氧化应激现象,导致神经元因坏死或细胞凋亡而死亡,而清除ROS可以减少神经炎症的产生。因此,抗炎和抗氧化是有前途的神经保护方法。
近年来,一些分子量很小的神经保护肽逐渐走进了人们的视线,它们可以自由的通过血脑屏障,通过静脉、皮下注射等给药途径即可以进入中枢神经系统,并具有很强的神经营养作用。神经保护肽的来源一般分为4种:(1)天然未加工的食品;(2)微生物、动物或植物蛋白的酶水解物;(3)微生物、动物或植物蛋白的微生物发酵物;(4)体外直接合成,包括化学法和酶法。闲又的神经保护肽以酶解来源为主,且研究主要集中在功效验证,而新的神经保护肽序列和功效还有待进一步拓展探索。
发明内容
本发明的目的在于提供一种具有抗炎、抗氧化和神经保护作用的细胞穿透肽。
本发明提供了一种神经保护多肽,它是如下任意一种序列的多肽或其衍生物:YLYLR、WHWPYAF、WPPFK。
本发明还提供了上述神经保护多肽在制备抗炎药物中的用途。
进一步地,上述抗炎药物是抑制神经炎症的药物。
本发明还提供了上述神经保护多肽在制备抗氧化药物中的用途。
进一步地,上述抗氧化药物是抑制神经氧化应激损伤的药物。
本发明还提供了上述神经保护多肽在制备神经保护药物中的用途。
进一步地,上述神经保护药物是防治神经系统疾病的药物。
更进一步地,上述神经系统疾病包括癫痫或阿尔兹海默症。
本发明还提供了一种药物,它是以上述的神经保护多肽为活性成分,加上要学上可接受的辅料制备而成的药物。
本发明的有益效果:本发明提供的多肽具有良好的抗炎、抗氧化作用,毒性低,具有神经炎症和抑制神经氧化应激的活性,在制备神经保护的药物中具有很好的应用前景。
显然,根据本发明的上述内容,按照本领域的普通技术知识和惯用手段,在不脱离本发明上述基本技术思想前提下,还可以做出其它多种形式的修改、替换或变更。
以下通过实施例形式的具体实施方式,对本发明的上述内容再作进一步的详细说明。但不应将此理解为本发明上述主题的范围仅限于以下的实例。凡基于本发明上述内容所实现的技术均属于本发明的范围。

具体实施方式
本发明所用原料与设备均为已知产品,通过购买市售产品所得。
本发明多肽WHWPYAF、YLYLR、WPPFK通过本领域公知的固相合成方法进行合成,纯度大于95%。
实施例1、本发明多肽对BV2细胞和LPS诱导的BV2细胞的影响
1、实验方法
1.1本发明多肽对BV2细胞活力的影响
细胞种板:将生长良好的BV2细胞经0.25% Trypsin胰酶消化30s,含10%FBS的RPMI-1640完全培养基终止消化。离心(1000rpm,5min)重悬制备密度为1×105个/ml的单细胞悬液。96孔板每孔接种100μL,接种密度为1×104个/孔,共接种3块96孔板,每块板接种3×9个孔,在周围孔均加入适量PBS缓冲溶液以降低边缘效应的影响。
分组设置:
(1)板一,空白对照组(只含培养基)、正常组、多肽WHWPYAF浓度梯度组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(2)板二,空白对照组(只含培养基)、正常组、多肽YLYLR浓度梯度组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(3)板三,空白对照组(只含培养基)、正常组、多肽WPPFK浓度梯度组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
药物配制:WHWPYAF,YLYLR,WPPFK多肽各50mg,分别加入0.438mL、0.573mL、0.618mL的DMSO溶液,统一配制成浓度为120nM的多肽母液,储存于4℃冰箱。给药前,用1640完全培养基将母液稀释成相应的给药浓度。
细胞活力检测:种板孵育待细胞贴壁后(约24h左右)吸除的上清液,空白组和正常组添加100μL的RPMI-1640完全培养基,其余实验组添加100μL不同浓度的多肽溶液。孵育24h后,每孔加入CCK-8检测液10μL,细胞培养箱孵育1h后检测450nm波长下的吸光度,记录数据;以正常组细胞活力作为对照,计算相应细胞活力,计算公式如下:细胞活力=(多肽实验组吸光度-空白对照组吸光度)/(正常组吸光度-空白对照组吸光度)×100%。
1.2本发明多肽对LPS诱导BV2细胞NO释放量的影响
细胞种板:参照1.1方法,将BV2细胞接种于3个96孔板中。
分组设置:(1)板一,培养基空白对照组、正常组、LPS组(1μg/mL)、LPS(1μg/mL)+多肽WHWPYAF实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(2)板二,培养基空白对照组、正常组、LPS组(1μg/mL)、LPS(1μg/mL)+多肽YLYLR实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(3)板三,培养基空白对照组、正常组、LPS组(1μg/mL)、LPS(LPS 1μg/mL)+多肽WPPFK实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
药物配制:(1)LPS粉末用无菌PBS缓冲液配制成1mg/mL的母液,储存于4℃。给药前用含2%血清的1640培养基稀释成1μg/mL的给药浓度。
(2)用含1μg/mL LPS的RPMI-1640基础培养基(不含血清)作为稀释液,按一定比例稀释多肽母液,配置成相应给药浓度。
(3)Griess试剂A液:对氨基苯磺酸0.05g,10%稀醋酸定容至15mL。B液:α-萘胺0.01g,蒸馏水2mL,10%稀醋酸定容至15mL。临用前将A液与B液按体积比1:1混匀配制得到Griess试剂。
细胞上清液NO检测:种板孵育24h,细胞贴壁后吸除上清液,更换至无血清基础1640培养基培养饿血培养12h之后,按照分组设置换为相应的含药培养基,空白组换为100μL 1640基础培养基,多肽与LPS共同处理孵育。药物处理24h后,吸取50μL细胞培养上清液与50μL新鲜配置的Griess试剂混匀,96孔板中室温孵育10min,检测560nm波长下的吸光度,记录数据。
1.3本发明多肽对LPS诱导的BV2细胞IL-1β、TNF-α、IL-6mRNA的影响
参照1.1方法,将BV2细胞(1×106个/孔)接种在六孔板中,次日,将MHWPYAF、WPPFK和YLYLR分别与LPS(1μg/mL)共同孵育培养24h后采取柱式提取法提取细胞总RNA,逆转录之后利用qPCR的方法对细胞中IL-1β、TNF-α、IL-6mRNA含量进行测定。以GAPDH作为内参基因采用2-ΔΔCT法计算炎症因子的相对mRNA表达量。
1.4免疫荧光检测本发明多肽对LPS诱导的BV2细胞中NF-κB核转位
将BV2细胞(1×106个/皿)接种在共聚焦小皿上,每个小皿加1mL完全培养基培养24h,贴壁之后加入LPS(1μg/mL)和60μM的WHWPYAF、YLYLR、WPPFK共同孵育24h后进行免疫荧光染色。
2、实验结果
2.1本发明多肽对BV2细胞的毒性
如图1所示,多肽MHWPYAF、WPPFK和YPYPL在浓度120μM以下对BV2细胞均没有明显的细胞毒性。因此,选择15,30,60μM的治疗浓度检测多肽对LPS诱导的BV2细胞产生NO含量的影响。
2.2本发明多肽对LPS诱导的BV2细胞NO产生的影响
如图2所示,在LPS诱导的BV2中,所有肽均表现出抑制NO生成的作用,且结果呈剂量依赖性。其中MHWPYAF和WPPFK在60μM时表现出显著的抑制作用(P<0.001),因此后续的实验选择60μM作为多肽的治疗浓度,测定多肽的抗炎指标。
2.3本发明多肽对LPS诱导的BV2细胞IL-1β、IL-6、TNF-α转录的影响
如图3所示,与空白对照组比较,LPS诱导的BV2细胞IL-1β、IL-6、TNF-α的mRNA含量显著上升(P<0.001),说明LPS刺激了BV2细胞的炎症反应。与模型组比较,本发明多肽均能抑制IL-1β、IL-6、TNF-αmRNA含量的上升,其中特别是多肽MHWPYAF和WPPFK在60μM时能显著抑制IL-1β、IL-6、TNF-αmRNA含量的上升(P<0.001)。
2.4本发明多肽对LPS诱导的BV2细胞NF-κB核转位的影响
如图4所示,BV2空白对照组细胞NF-κB蛋白主要在细胞质中表达,细胞核中几乎不表达。与空白对照组比较,BV2细胞经LPS刺激24h后,观察到NF-κB发生了核转位,胞体变大,呈阿米巴样分化。多肽MHWPYAF、WPPFK和YLYLR(给药浓度均为60μM)各组不同程度抑制了NF-κB的核转位现象,其中MHWPYAF和WPPFK的抑制效果尤其明显。
目前,使用LPS诱导的小鼠BV2小胶质细胞NO释放的体外研究模型被用于大量化合物的抗神经炎症活性评价。在L-精氨酸向L-瓜氨酸的转化过程中,一氧化氮合酶(NOS)催化产生NO。在大脑内,神经胶质细胞会产生NO以应对炎症,而过量的NO会加剧神经炎症,导致神经元死亡和组织损伤。由此产生的神经元死亡进一步刺激了NOS的表达,产生更多的NO,从而使组织损伤循环持续下去。因此,NO是治疗神经炎症疾病的合理目标。此外,活化的小胶质细胞释放促炎细胞因子IL-1β,IL-6,TNF-α导致神经元的损伤,其转录主要由NF-κB蛋白发生磷酸化入核参与调控。因此NF-κB磷酸化入核是炎症发生的重要条件。
而根据上述实验结果,可以证明本发明多肽干预之后的LPS诱导的BV2细胞的NO产量,炎症因子IL-1β,IL-6,TNF-αmRNA水平,NF-κB的核转位情况均有改善,因此,本发明多肽表现出抑制神经炎症活性。
实施例2、本发明多肽对PC12细胞和H2O2诱导的PC12细胞的影响
1、实验方法
1.1本发明多肽对PC12细胞活力的影响
细胞种板:将生长良好的PC12细胞经0.25% Trypsin胰酶消化30s,含10%FBS的RPMI-1640完全培养基终止消化。1000rpm离心5min,重悬制备密度为1×105个/ml的单细胞悬液。96孔板每孔接种100μL,接种密度为1×104个/孔,共接种3块96孔板,每块板接种3×9个孔,在周围孔均加入适量PBS缓冲溶液以降低边缘效应的影响。
分组设置:同实施例1。
药物配制:同实施例1。
细胞活力检测:同实施例1。
1.2H2O2诱导PC12细胞损伤模型的建立
细胞种板:同1.1。
分组设置:空白组(不含细胞)、正常组、H2O2刺激组(60、90、120、150μM),每组设置3个复孔。
药物配制:30% H2O2溶液使用RPMI 1640完全培养基稀释至浓度分别为60、80、100、120、150μM。每次临用前现配现用。
细胞活性检测:同实施例1。
1.3本发明多肽对H2O2诱导PC12细胞的活性影响
细胞种板:同1.1。
分组设置:(1)板一,空白组(不含细胞)、正常组、模型组(H2O2,100μM)、H2O2(100μM)+多肽WHWPYAF实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(2)板二,空白组(不含细胞)、正常组、模型组(H2O2,100μM)、H2O2(100μM)+多肽YLYLR实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
(3)板三,空白组(不含细胞)、正常组、模型组(H2O2,100μM)、H2O2(100μM)+多肽WPPFK实验组(3.75、7.5、15、30、60、120μM),每组设3个复孔。
药物配制:多肽药物配制同实施例1,H2O2配制同1.2。
细胞活性检测:同实施例1。
1.4本发明多肽对H2O2诱导PC12细胞ROS表达的影响
将PC12细胞以1×105个/孔的密度接种于6孔板上。PC12细胞分别用MHWPYAF、WPPFK和YPYPL在60μM条件下预处理12h,然后在100μM过氧化氢的存在下再培养12h。根据试剂盒的说明,处理后的细胞用10μM DCFH-DA探针预孵育30min,然后用PBS洗涤两次,使用流式细胞仪测量荧光。
2、实验结果
2.1本发明多肽对PC12细胞的毒性
如图5所示,在3.75μM-60μM浓度下,多肽MHWPYAF、WPPFK和YLYLR对PC12细胞均没有明显的细胞毒性。因此,选择15,30,60μM的治疗浓度检测多肽MHWPYAF、WPPFK和YPYPL对H2O2诱导的PC12细胞活力的影响。
2.2本发明多肽对H2O2诱导PC12细胞的活性影响
结果如图6所示,100μM H2O2对PC12细胞增殖的抑制率在40%左右(P<0.001)。与模型组比较,多肽预处理后各组均表现出一定的对H2O2诱导的PC12细胞的保护作用,其中特别是MHWPYAF和WPPFK具有显著的抑制H2O2对PC12活力造成的损伤(P<0.001),且结果呈剂量依赖性,但YLYLR对H2O2诱导的PC12在30μM和60μM时,保护作用相差不明显,因此后续的实验选择60μM作为多肽的治疗浓度。
2.3本发明多肽对H2O2诱导的PC12细胞内ROS含量的影响
从图7可以看出,与正常组细胞相比,当PC12细胞暴露于H2O2溶液中12h后,PC12细胞内荧光强度增加,表明产生的ROS含量升高(P<0.001);而与对照组相比,多肽MHWPYAF、WPPFK和YLYLR(60μM)预处理可显著降低H2O2诱导的PC12细胞内ROS的累积(P<0.001)。
目前,H2O2诱导的PC12模型常用的氧化应激造成的神经损伤模型,在癫痫,阿尔兹海默症,中风中的药效评价中发挥重要作用。H2O2干预造成PC12细胞增殖活性下降是细胞发生氧化应激损伤的最直观表现。此外,神经损伤相关的炎症、细胞焦亡、线粒体功能障碍、兴奋性毒性、铁代谢和坏死性凋亡等都与细胞内ROS的增多密切相关,ROS的产量是评价细胞氧化应激水平的核心指标之一。
而根据上述实验结果,可以证明本发明多肽干预之后的H2O2干预PC12细胞活力和ROS含量均有改善,因此,本发明多肽表现出抑制神经氧化应激活性。
综上,本发明提供了一种多肽,具有良好的抗炎、抗氧化作用,毒性低、具有神经炎症和抑制神经氧化应激的活性,在制备神经保护的药物中具有很好的应用前景。

1.一种神经保护多肽,其特征在于,它是如下任意一种序列的多肽或其衍生物:YLYLR、WHWPYAF、WPPFK。
2.权利要求1所述的神经保护多肽在制备抗炎药物中的用途。
3.如权利要求3所述的用途,其特征在于,所述抗炎药物是抑制神经炎症的药物。
4.权利要求1所述的神经保护多肽在制备抗氧化药物中的用途。
5.如权利要求4所述的用途,其特征在于,所述抗氧化药物是抑制神经氧化应激损伤的药物。
6.权利要求1所述的神经保护多肽在制备神经保护药物中的用途。
7.如权利要求6所述的用途,其特征在于,所述神经保护药物是防治神经系统疾病的药物。
8.如权利要求7所述的用途,其特征在于,所述神经系统疾病包括癫痫或阿尔兹海默症。
9.一种药物,其特征在于,它是以权利要求1所述的神经保护多肽为活性成分,加上要学上可接受的辅料制备而成的药物。
