Abstract
本发明公开了一种具有多种免疫调节因子的增强型溶瘤病毒及其制备方法和应用,所述增强型溶瘤病毒基因组中包含了(i)第一核酸序列,所述第一核酸序列编码PD‑L1单链抗体;(ii)第二核酸序列,所述第二核酸序列编码CD40L分子;(iii)第三核酸序列,所述第三核酸序列编码IL2mimic分子,所述IL2mimic优选是IL‑2受体的多肽激动剂。
Description
技术领域
本发明涉及生物医药领域,具体涉及一种含有多个免疫调节因子的增强型溶瘤病毒和所述增强型溶瘤病毒的制备方法以及在抗肿瘤方面的应用。
背景技术
溶瘤病毒(OVs)是经过工程改造或可选择性在癌细胞中繁殖并杀死肿瘤细胞的一类病毒。研究人员根据病毒遗传物质不同把溶瘤病毒分为两大类:DNA溶瘤病毒和RNA溶瘤病毒。其中DNA溶瘤病毒主要包括:腺病毒、单纯疱疹病毒、细小病毒以及痘病毒。RNA溶瘤病毒主要包括:呼肠孤病毒、柯萨奇病毒、脊髓灰质炎病毒、塞内加古病毒、麻疹病毒、新城疫病毒、水疱性口炎病毒、马拉巴病毒以及寨卡病毒等。目前,为了评估这些溶瘤病毒在不同恶性肿瘤中的有效性和安全性,多项临床实验正在开展,其中以溶瘤腺病毒和溶瘤疱疹病毒研究最为广泛。作为生物制剂疗法,OVs可以局部或全身给药,因此对于原发性和转移性肿瘤部位都起作用。随着现代免疫疗法的成功,溶瘤病毒目前被认为是对癌症患者的潜在治疗选择,这些患者在接受免疫检查点抑制剂治疗后无反应或无法实现持久反应,OVs提供了一个可供选择的其他方面的治疗平台。
对于肿瘤细胞的直接杀伤和诱导全身性抗肿瘤免疫反应,是溶瘤病毒抑制肿瘤生长的主要作用机制。病毒感染肿瘤细胞后会产生内质网应激和基因毒性应激来启动抗病毒反应,这种反应导致活性氧的上调和抗病毒细胞因子的表达,活性氧可以作用并激活抗原提呈细胞。感染的肿瘤细胞可以释放危险信号分子,招募并刺激免疫细胞(如抗原呈递细胞和自然杀伤细胞),溶瘤病毒感染细胞也可释放子代病毒和肿瘤相关抗原。激活后的抗原提呈细胞可以迁移到淋巴结组织,将包括肿瘤相关抗原在内的加工抗原传递给CD8+T和CD4+T细胞,诱导机体产生特异性的抗肿瘤的免疫应答。
肿瘤微环境(tumor microenvironment,TME)由多类细胞,各种化学因子、细胞因子,酶类以及胞外基质形成。TME与肿瘤的发生、发展及转移息息相关。例如,髓系来源的抑制性细胞(MDSCs)是一种多样的骨髓祖细胞,它可以产生精氨酸酶1(Arg1)促进肿瘤细胞生长并抑制免疫细胞功能。肿瘤细胞表面高表达细胞程序性死亡受体配体(PD-L1),与肿瘤浸润的T细胞表面PD-1结合,诱导T细胞的耗竭和凋亡。
CD40配体(CD40L)也称作CD154,肿瘤坏死因子相关激活蛋白,是一种32kDa~39kDa的II型跨膜蛋白。CD40L是CD40的主要配体,,主要由活化的CD4+T淋巴细胞和血小板表达。CD40/CD40L信号通路在机体有着广泛的调节作用,包括细胞增殖,自身免疫病,T、B细胞活化与增殖等。目前一些观点认为针对CD40/CD40L信号通路,对于连接天然免疫和适应性免疫,有效激活T细胞抗肿瘤功能具有重要作用。
IL2被认为是T细胞活化和扩增重要的细胞因子,在肿瘤的临床治疗上大剂量的IL-2被使用,可促进CD8+T肿瘤杀伤性细胞的增殖,但是也存在着一些弊端,例如毒副作用及对肿瘤组织的调节性T细胞Treg的诱导分化。Treg可通过分泌IL-10和TGF-β,抑制T细胞及抗原提呈细胞,抑制机体的抗肿瘤免疫反应。
为了更好的改善肿瘤局部免疫抑制的微环境,我们在溶瘤病毒载体水疱性口炎病毒VSV基因组中插入CD40L-PD-L1单链抗体(scFv)-IL2mimic序列,期望获得更好的肿瘤治疗的效果。
发明内容
本发明旨在针对现有溶瘤病毒在治疗癌症中相关技术问题,提出一种新型的PD-L1单链抗体和一种增强型溶瘤病毒,该增强型溶瘤病毒能够分泌新型的PD-L1单链抗体抑制CD8+T杀伤性细胞耗竭,同时能够分泌CD40L和IL2mimic,改善肿瘤局部免疫抑制的微环境,提升肿瘤杀伤效果,提高肿瘤患者生存时间。
在本发明的第一方面,本发明提出一种PD-L1单链抗体,所述PD-L1单链抗体包含重链抗体可变区和轻链抗体可变区;所述重链抗体可变区具有SEQ ID NO:5所示的氨基酸序列,所述轻链抗体可变区具有SEQ ID NO:6所示的氨基酸序列;
所述重链抗体可变区与所述轻链抗体可变区通过铰链序列连接,且所述重链抗体可变区位于所述轻链抗体可变区的N端;
所述铰链序列具有与SEQ ID NO:15所示的氨基酸序列。
优选地,所述PD-L1单链抗体还包含分泌信号肽;
所述PD-L1单链抗体与SEQ ID NO:7所示的氨基酸序列具有至少90%序列同一性。
在本发明的第二方面,本发明提出一种用于治疗癌症的药物组合物,所述药物组合物包括上述的PD-L1单链抗体。
优选地,所述癌症是肺癌、黑色素瘤、头颈部癌症、肝癌、脑癌、结直肠癌、膀胱癌、乳腺癌、卵巢癌、子宫癌、官颈癌、淋巴癌、胃癌、食道癌、肾癌、前列腺癌、胰腺癌或白血病等血液癌。
在本发明的第三方面,本发明提出一种增强型溶瘤病毒,所述溶瘤病毒基因组中包括:
(i)第一核酸序列,所述第一核酸序列编码PD-L1单链抗体;
(ii)第二核酸序列,所述第二核酸序列编码CD40L分子;
(iii)第三核酸序列,所述第三核酸序列编码IL2mimic分子,所述IL2mimic优选是IL-2受体的多肽激动剂。

本发明提供的增强型溶瘤病毒,能够大量分泌PD-L1单链抗体,CD40L分子以及IL2mimic分子,能够有效的杀伤肿瘤细胞的同时,改善肿瘤免疫抑制的微环境,提高治疗癌症的效果,延长治疗对象的存活时间。
本发明提供的PD-L1单链抗体为发明人新设计的单链抗体,具有更好的肿瘤杀伤效果,另外能在溶瘤病毒中实现大量的分泌,实现更好治疗癌症的效果。

本发明中所使用的的“治疗”是指改善或治愈对象的疾病(例如,癌症)。在一些实施例中,治疗可以包括减轻、延迟、缓解癌症的症状的严重程度(例如,肿瘤体积减小)、降低癌症的症状的出现频率(例如疼痛等)、延长患有癌症的对象的生存时间、存活率增加、下降癌细胞存活能力或杀灭癌细胞等。
本发明中所使用的“有效剂量”是指足以提供有用的或以其他方式减少有害的无益事件的组合物的量(例如足以治疗疾病的剂量)。在本申请中,组合物包括溶瘤病毒。包含在有效剂量的组合物内的溶瘤病毒的量取决于一种多种因素,包括但不限于治疗目的、对象的体重、性别、年龄和一般健康状况、施用途径、施用时间以及待治疗疾病的性质。
本发明中所使用的“VSV病毒”是指水疱性口炎病毒(vesicular stomatitisvirus,VSV),属于弹状病毒科,是水疱病毒属的单股负链RNA病毒。基因组不分节段,长约11kb。从3′端→5′端依次排列着N、P、M、G、L共5个不重叠的基因,分别编码核(N)蛋白、磷酸(P)蛋白、基质(M)蛋白、糖(G)蛋白、及RNA聚合酶(L)蛋白5种不同的主要蛋白。
本发明中所使用的“重组VSV病毒”是指经过改造的水疱性口炎病毒,其与野生型水疱性口炎病毒相比,包含人工引入的外源表达基因。应当理解的是,本发明重组VSV病毒不受限于其生产方式。例如,本发明的重组VSV病毒可通过反向遗传系统产生,也可通过培养感染所述重组VSV病毒的细胞而制备。
本发明中所使用的病毒载体,指的是基于病毒基因组而构建获得的、能够携带外源核苷酸序列的一种核酸运载工具。通常而言,病毒载体能够在合适的宿主细胞中自我复制和/或表达其所包含的基因(内源的和外源的)。病毒载体可包含完整的野生型病毒的基因组,或者经突变或修饰的病毒基因组。然而,出于安全性考虑,病毒载体一般优选包含经突变或修饰的病毒基因组。本发明的病毒载体衍生自VSV的病毒基因组,因此,本发明的病毒载体也可被称为VSV病毒载体。
本发明所使用的“异源”是指病毒中非天然存在的核酸(或由病毒非天然产生的蛋白质)。例如,相对于VSV,编码人CD40L的核酸或编码PD-L1单链抗体的核酸是“异源”的。
本发明所使用的“免疫检查点抑制剂”或“检查点抑制剂”是指完全或部分地减少、抑制、干扰或调节一种或多种检查点蛋白的分子。检查点蛋白调节T细胞激活或功能。许多检查点蛋白是已知的,例如CTLA-4及其配体CD80和CD86;以及PD-1及其配体PD-L1和PD-L2。这些蛋白质负责T细胞应答的共抑制相互作用。免疫检查点蛋白调节和维持自身耐受性以及生理免疫应答的持续时间和幅度。免疫检查点抑制剂包括抗体或可以衍生自抗体。
本发明所使用的程序性细胞死亡蛋白1(PD-1)是在T细胞、B细胞、单核细胞、自然杀伤细胞表面诱导表达的含有288个氨基酸的细胞表面共抑制性受体。它由位于人2号染色体长臂37区3带的PDCD1基因编码,是一个55kDa的跨膜蛋白。其与肿瘤细胞表面分子PD-L1结合后,抑制T细胞活化,阻碍免疫应答反应,诱导抗肿瘤特异性T细胞耗竭,甚至凋亡,进而抑制机体的抗肿瘤免疫应答。
本发明所使用的IL2mimic是IL-2受体的多肽激动剂,所述多肽激动剂多肽序列有100个左右的氨基酸,可以高强度结合IL2Rβ、γc,但与CD25(IL2Rα识别受体)、CD215(IL15Rα识别受体)无结合,与天然的IL2比较,IL2mimic热稳定性更好、活性更高、免疫原性更低、安全窗口更大,且对抑制性Treg细胞的诱导能力较弱。
本发明所使用的“hCD40L”是指人源的CD40L分子;“mCD40L”是指鼠源的CD40L分子。
本发明所使用的多顺反子蛋白接头序列为P2A,来源于猪捷申病毒(Porcineteschovims)(氨基酸序列如SEQ ID NO:16所示,核苷酸序列如SEQ ID NO:11或SEQ ID NO:17所示),该蛋白序列可以被细胞内蛋白酶识别并切割,使得多顺反子蛋白断裂成为单个蛋白,可用于以任何取向连接两个蛋白分子。
需要说明的是,任何病毒都可以用于产生本发明的重组溶瘤病毒。通常,可以对病毒进行修饰,例如以调节其复制(例如,相对于在健康细胞中,优先在肿瘤细胞中复制)、其被宿主的免疫系统检测的能力,以及包括外源核酸。在一些较佳的实施方式中,溶瘤病毒可以是水疱性口炎病毒,其属于弹状病毒科,水疱病毒属。在一些较佳的实施方式中,溶瘤病毒可以是经人工改造的溶瘤病毒。例如,在野生型VSV病毒中去M基因突变的溶瘤病毒,从而使其安全性更好,即,获得的溶瘤病毒为无致病性/非神经毒性且是溶瘤性的。本发明的重组溶瘤病毒还可包含增强外源核酸翻译(例如哺乳动物翻译)的序列。例如,已知KOZAK序列增强哺乳动物翻译。因此,在一些实施例中,溶瘤病毒包含Kozak序列。
下面将通过实施例来进一步说明本公开,但是本公开并不因此而受到任何限制。
除非特别说明,本公开所用到的试剂、培养基等试验材料均为市售商品。
下述实施例中的VSV病毒载体委托上海晶朗生物科技有限公司全基因合成。所述VSV病毒载体系统包含WHp001(其序列如SEQ ID NO:1所示,图谱如图1所示)、WHp002(其序列如SEQ ID NO:2所示,图谱如图2所示)、WHp003(其序列如SEQ ID NO:3所示,图谱如图3所示)和WHp004(其序列如SEQ ID NO:4所示,图谱如图4所示)四个质粒,其中质粒WHp001编码了VSV全长基因组,在WHp001质粒中编码VSV基因组G和L基因之间,包含了hCD40L-PDL1-IL2mimic序列,WHp002编码了VSV N蛋白,WHp003编码了VSV P蛋白,WHp004编码了VSV L蛋白,上述质粒中均含有T7 RNA聚合酶启动子。
下述实施例中的表达T7聚合酶质粒pT7 poly购自上海林渊生物科技有限公司(货号:65974)。
下述实施例中293T细胞购自购自中国科学院典藏细胞库,使用含10%的胎牛血清(Sigma)的DMEM培养基(Gibco)(含1.5mM L-Glutamine,100U/ml penicillin,100μg/mlStreptomycin),在37℃、5%CO2饱和湿度培养箱中进行常规培养。
下述实施例中VSV病毒载体系统包装方法:先在T25细胞培养瓶(Coming)中铺设4.0×105个293T细胞,待细胞过夜贴壁后第二天,用JetPEI@细胞转染试剂(Polyplus,货号:101000053)共转染VSV包装四质粒WHp001(2.5ug),WHp002(1.0ug),WHp003(1.25ug),WHp004(0.4ug)和表达T7聚合酶质粒pT7 poly(3.0ug)。在转染后6小时,细胞去除转染培养基,更换新鲜培养基。转染48小时后,收集细胞培养上清,利用培养上清,重新感染新的培养瓶中293T细胞进行扩增病毒,得到VSV病毒。
实施例1修饰的PD-L1scFv单链抗体在溶瘤病毒中的功能验证
1.1PDL1单链抗体序列设计和质粒构建:
通过NCBI查询,获得PD-L1重链抗体可变区序列(MP575191),其氨基酸序列如SEQID NO:5所示;轻链抗体可变区序列(MQ234473),其氨基酸序列如SEQ ID NO:6所示;发明人在此基础上优化设计了PD-L1单链抗体序列,包括加上分泌信号肽,轻链和重链位置互换,轻链和重链之间增加铰链连接序列(氨基酸序列如SEQ ID NO:15所示),得到优化后的PD-L1scFv单链抗体序列,其示意图如图5所示,其氨基酸序列如SEQ ID NO:7所示,以此来增强单链抗体的功能和胞外分泌。通过全基因合成PD-L1scFv序列,克隆到WHp001质粒的Xho I/NheI位点,插入到WH001质粒中编码VSV基因组G和L基因之间,得到单独表达PD-LlscFv单链抗体的VSV全长基因组编码质粒WHp032(图谱如图6所示)。
1.2表达PD-LlscFv单链抗体VSV重组病毒的制备:
在T25细胞培养瓶中铺设4.0×105个293T细胞,待细胞过夜贴壁后第二天,用PEI细胞转染试剂共转染VSV包装四质粒WHp032(2.5ug),WHp002(1.0ug),WHp003(1.25ug),WHp004(0.4ug)和表达T7聚合酶质粒pT7poly(3.0ug)。在转染后6小时,细胞去除转染培养基,更换新鲜培养基。转染48小时后,收集细胞培养上清,利用培养上清,重新感染新的培养瓶中293T细胞进行扩增病毒,得到重组的表达PD-LlscFv单链抗体的VSV重组病毒WHV-006。
1.3VSV重组病毒WHV-006表达的PD-L1单链抗体功能鉴定
使用human PD-L1 ELISA试剂盒(Abcam,ab277712)对VSV重组病毒WHV-006感染细胞表达的PDL 1单链抗体进行功能鉴定。检测流程如图7所示,检测样品中如果含有PD-L1抗原,在孵育情况下,会和试剂盒自带的两个anti-PD-L1抗体结合,从而形成三明治结构,试剂盒自带的一个anti-PD-L1抗体偶联HRP辣根过氧化物酶,在加入TMB显色底物后能呈现颜色反应,通过与标准样品PD-L1的颜色比较,从而来标定检测样品中PD-L1含量(图7A)。如果除了试剂盒自带的anti-PD-L1抗体,样品中含有其他PD-L1的抗体,那么样品中含有的PD-L1抗体必然会干扰PD-L1抗原和试剂盒自带anti-PD-L1抗体结合,导致最后加入TMB底物之后,显色反应减弱(图7B)。
取25uLWHV-006感染细胞的上清或者细胞裂解液,和含有11.1pg的PD-L1标准样品混合,阳性对照为50uL含有11.1pg的PD-L1样品,结果如图8所示,检测上清中含有细胞培养上清或者细胞裂解液,和阳性样品黄色比较,细胞感染上清和细胞裂解液颜色反应微弱,吸光值分别为0.23和0.41,而阳性对照吸光值为1.75,以上结果说明重组病毒WHV-006能够分泌表达PD-L1单链抗体,并且分泌的PD-L1单链抗体能够与试剂盒中的PD-L1抗原结合,干扰ELISA试剂盒中最后的显色反应。
实施例2VSV重组病毒WHV-001的构建
2.1含有hCD40L-PD-L1scFv-IL2mimic外源基因重组VSV病毒载体构建
在VSV病毒基因组中插入hCD40L基因片段(核苷酸序列如SEQ ID NO:8所示,氨基酸序列如SEQ ID NO:18所示)、PD-L1scFv单链抗体的基因片段(核苷酸序列如SEQ ID NO:9所示,氨基酸序列如SEQ ID NO:7所示)和IL2mimic的基因片段(核苷酸序列如SEQ ID NO:10所示,氨基酸序列如SEQ ID NO:19所示),上述三个基因片段之间通过P2A序列(SEQ IDNO:11)进行连接,通过P2A蛋白酶切位点,hCD40L-PD-L 1scFv-IL2mimic编码的多聚蛋白在细胞被可以被剪切为各自独立蛋白hCD40L、PD-L1scFv单链抗体和IL2mimic;在CD40L基因片段前有KOZAK序列GCCACC(SEQ ID NO:14),增强蛋白起始翻译效率。上述三个基因片段经过改造修饰后得到的序列为hCD40L-PD-L 1scFv-IL2mimic,其核苷酸序列如SEQ ID NO:12所示,其氨基酸序列如SEQ ID NO:20所示。将hCD40L-PD-L1scFv-IL2mimic插入到VSV基因组G和L基因之间,两侧含有Xho I/NheI位点,得到含有hCD40L-PD-L1scFv-IL2mimic外源基因重组VSV病毒载体WHp001(图1),由上海晶朗生物科技有限公司通过全基因合成方式合成。
2.2含有hCD40L-PD-L1scFv-IL2mimic序列的VSV重组病毒制备
在T25细胞培养瓶中铺设4.0×105293T细胞,待细胞过夜贴壁后第二天,用PEI细胞转染试剂共转染VSV包装四质粒WHp001(2.5ug),WHp002(1.0ug),WHp003(1.25ug),WHp004(0.4ug)和表达T7聚合酶质粒pT7poly(3.0ug)。在转染后6小时,细胞去除转染培养基,更换新鲜培养基。转染48小时后,收集细胞培养上清,利用培养上清,重新感染新的培养瓶中293T细胞进行扩增病毒,得到重组的表达外源三基因的VSV重组病毒WHV-001。
图9显示使用上述制备得到的重组病毒WHV-001感染Vero细胞(购自购自中国科学院典藏细胞库)48小时产生病变,说明重组病毒WHV-001已被成功制备,并具备侵染能力。
实施例3重组病毒WHV-001中hCD40L、PD-L1scFv和IL2mimic的表达鉴定.
使用实施例2中制备的重组病毒WHV-001感染293T细胞,分别收集细胞上清和细胞裂解液,用于hCD40L、PD-L1scFv和IL2mimic的表达鉴定
(1)hCD40L功能鉴定
用hCD40L ELISA试剂盒(Abcam,ab99991)检测上清和细胞裂解液中CD40L的含量,结果如图10所示,检测上清原液OD值过大,稀释10倍之后,测得OD为1.47,对应曲线hCD40L含量为4924pg/mL,重组病毒WHV-001感染细胞上清中表达的CD40L的含量为49.24ug/mL。
(2)PD-L1功能鉴定
取25uL细胞培养上清和细胞裂解液,和含有11.1pg的PD-L1标准样品混合,阳性对照为50uL含有11.1pg的PDL1样品,如图19所示,通过颜色反应发现,检测上清中含有细胞培养上清或者细胞裂解液,和阳性样品黄色比较,能完全封闭试剂盒自带抗体和PD-L1的结合,最后显色反应微弱,OD读值0.0577;检测上清稀释10倍,也能较好地封闭PD-L1抗原,和阳性对照比较,呈现弱微的黄色,OD值为0.1876;说明重组病毒WHV-001能够分泌表达PD-L1单链抗体,并且分泌的PD-L1能够很好的与PD-L1结合。
(3)IL2mimic功能鉴定
本实施例使用小鼠IL2mimic单克隆抗体检测IL2mimic的相对含量,检测步骤如下:
①包板:将样品包被在96孔酶标板Costar 9018(Coming,CLS9018)的96孔板中,每个样品做3个复孔,每孔100μL,密封于4℃孵育过夜。
②使用PBS-T洗3-5次后,用ELISA稀释液室温封闭1h。
③使用PBS-T洗3-5次后,加入小鼠IL2mimic的单抗(委托苏州优诺科生物有限公司制备)1∶1000稀释于diluent室温孵育2h。
④使用PBS-T洗3-5次后,加入goat anti-mouse IgG-HRP,室温孵育1h。
⑤加入TMB,37℃避光孵育10min,加入stop solution,OD450读值。
检测结果如图21所示,在WHV-001感染细胞的上清液中,IL2 mimic的含量达到304ng/mL,说明重组病毒WHV-001能够分泌表达IL2mimic,并且分泌的IL2mimic能够很好的与IL2mimic的单抗结合。
实施例4重组病毒WHV-001抑制肿瘤细胞效果验证
为了确定重组病毒WH-001能够感染肿瘤细胞,并诱导细胞死亡。
在6孔细胞培养板中接种3×105细胞/孔,分别接种了小鼠非小细胞肺癌细胞LLC(中国科学院典藏细胞库),小鼠乳腺癌细胞4T1(中国科学院典藏细胞库)和小鼠结直肠癌细胞MC38(湖南丰晖生物科技有限公司),以MOI=1感染复数的重组病毒WHV-001感染细胞,并且在感染后48小时收集细胞,利用CCK8试剂盒(苏州新赛美生物科技有限公司)对细胞活力进行检测。细胞杀伤效率的计算=[(B-A)/(B-C)]×100%。结果如图11所示,重组病毒WHV-001对不同肿瘤细胞的肿瘤均有杀伤能力,对于LLC细胞,病毒感染后48小时能100%的杀死感染细胞。
实施例5VSV重组病毒WHV-010的制备
由于重组病毒WHV-001插入的外源基因中的CD40L是人来源的CD40L(hCD40L)的分子,为了避免在小鼠肿瘤模型中hCD40L和小鼠CD40不结合,发明人构建了WHV-010病毒,在WHV-010基因组位置中,hCD 40L被小鼠的CD40L(mCD40L)替换,得到新的mCD40L-PDL1scFv-IL2mimic片段,上述mCD40L序列如SEQ ID NO:13所示。委托上海晶朗生物科技有限公司合成mCD40L-PDL1scFv-IL2mimic片段,通过NheI/xhoI酶切位点,替换了质粒WHp001上面hCD40L-PDL1scFv-IL2mimic片段,得到质粒WHp026,其图谱如图12所示。
如实施例2,在T25细胞培养瓶中铺设4.0×105个293T细胞,待细胞过夜贴壁后第二天,用PEI细胞转染试剂共转染VSV包装四质粒WHp026(2.5ug),WHp002(1.0ug),WHp003(1.25ug),WHp004(0.4ug)和表达T7聚合酶质粒pT7 poly(3.0ug)。在转染后6小时,细胞去除转染培养基,更换新鲜培养基。转染48小时后,收集细胞培养上清,利用培养上清,重新感染新的培养瓶中293T细胞进行扩增病毒,得到重组的表达mCD40L、PD-L1scFv和IL2mimic的VSV重组病毒WHV-010。取25uL重组病毒WHV-010侵染的细胞培养上清和细胞裂解液,和含有11.1pg的PD-L1标准样品混合,阳性对照为50uL含有11.1pg的PD-L1标准样品,结果如图20所示,检测上清中含有细胞培养上清或者细胞裂解液,和阳性样品黄色比较,没有稀释或者稀释10倍,能完全封闭试剂盒自带抗体和PD-L1的结合,最后显色反应微弱,OD读值只有0.0587和0.0591;即使检测上清稀释100倍和1000倍,也还是能较好地封闭PD-L1抗原,和阳性对照比较,呈现弱微的黄色,以上结果说明重组病毒WHV-010能够分泌表达PD-L1单链抗体,并且分泌的PD-L1能够在很低的含量下与PD-L1结合。
实施例6重组病毒WHV-010抑制乳腺癌效果验证
按每只Balb/c小鼠(6周龄,雌性,购自北京维通利华公司)在皮下接种5.0×105个乳腺癌细胞4T1细胞。接种后第7天,小鼠肿瘤体积达到40-50mm3,荷瘤小鼠随机分为3组(n=10),分别为PBS组、WHV-010组和WHab-001组,其中PBS组每只荷瘤小鼠瘤内注射100uLPBS,每两天注射一次,共注射3次;WHV-010组每只荷瘤小鼠瘤内注射1.0×107PFU的WHV-010病毒每两天注射一次,共注射3次;WHab-001组每只荷瘤小鼠瘤内注射50ug PD-1抗体(Bioxcell,货号:be0146-25mg),每周注射一次,共注射3次。每两天测量肿瘤体积,连续观察记录肿瘤体积的变化情况,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。
在乳腺癌小鼠肿瘤模型中,PBS组肿瘤生长迅速,明显快于WHab-001单抗组和WHV-010病毒治疗组(图13),WHV-010病毒治疗组的小鼠肿瘤生长被明显的抑制,同时好于PD-1单抗治疗的效果,说明本发明的溶瘤病毒WHV-010治疗效果优于市面目前常用的免疫制剂PD-1抗体疗法。相对应地,治疗小鼠的生存曲线(图14)也显示溶瘤病毒WHV-010治疗组的小鼠存活时间与PBS组及WHab-001单抗组比较有了显著的延长。
实施例7重组病毒WHV-010抑制结肠直肠癌效果验证
6-8周龄C57BL/6J小鼠(6周龄,雌性,购自北京维通利华公司)右侧皮下接种小鼠结直肠癌细胞MC-38,每只小鼠接种7.0×105(100uL)MC-38细胞。接种后第8天,小鼠肿瘤体积达到40-50mm3,荷瘤小鼠随机分为3组(n=10),分别为PBS组、WHV-010组和WHab-001组。其中PBS组每只荷瘤小鼠瘤内注射100uL PBS,每两天注射一次,共注射3次;WHV-010组每只荷瘤小鼠瘤内注射1.0×107PFU的WHV-010病毒每两天注射一次,共注射3次;WHab-001组每只荷瘤小鼠瘤内注射50ug PD-1抗体(Bioxcell,货号:be0146-25mg),每周注射一次,共注射3次。每两天测量肿瘤体积,连续观察记录肿瘤体积的变化情况,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。
WHV-010治疗组可以非常明显的抑制小鼠结直肠肿瘤的生长(图15),在接种后31天,PBS未治疗组肿瘤平均大小为1200mm3,而WHV-010治疗组为174mm3,WHab-001单抗治疗组为426mm3;对于小鼠生存率进行考察(图16),PBS未治疗组小鼠42天全部死亡,而WHV-010治疗组存活率为70%(n=10),WHab-001单抗治疗组存活率为10%(n=10),由上可知含有多种免疫调节增强因子的VSV溶瘤病毒在小鼠节直肠癌有非常显著的治疗效果,与目前免疫疗法PD-1抗体比较,疗效显著。
实施例8重组病毒WHV-010抑制肺癌治疗效果验证
6-8周龄C57BL/6J小鼠右侧皮下接种小鼠非小细胞肺癌细胞LLC,每只小鼠接种5.0×105(100uL)LLC细胞。接种后第8天,小鼠肿瘤体积达到40-50mm3,荷瘤小鼠随机分为3组(n=7),分别为PBS组、WHV-006和WHV-010组,分别瘤内注射100uL PBS、1×107PFU WHV-006和WHV-010病毒,每两天注射一次,共注射3次。每两天测量一次肿瘤体积,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。
WHV-010治疗可以非常显著的抑制小鼠肺癌细胞的生长(图17),在接种后28天,PBS未治疗组肿瘤平均大小为1813mm3,而WHV-010治疗组为196mm3,表达PD-L1单链抗体的WHV-006治疗组为674mm3,对于小鼠生存率进行考察(图18),PBS未治疗组小鼠36天全部死亡,而WHV-010治疗组存活率为57.14%(n=7),WHV-006治疗组为28.57%(n=7),由上可知重组病毒WHV-010和WHV-006对肺癌均具有非常显著的治疗效果。
实施例9重组病毒WHV-001抑制结直肠癌治疗效果验证
6-8周龄Balb/c小鼠右侧皮下接种小鼠结直肠癌细胞CT26,每只小鼠接种5.0×105(100uL)CT26细胞。接种后第8~10天,小鼠肿瘤体积达到50mm3,荷瘤小鼠随机分为3组(n=8),分别为PBS组和WHV-001组,分别瘤内注射100uL PBS、1×108PFU WHV-001病毒,每两天注射一次,共注射3次。每两天测量一次肿瘤体积,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。WHV-001治疗可以非常显著的抑制小鼠结直肠癌细胞的生长(图22),在接种后28天,PBS未治疗组肿瘤平均大小为800mm3,而WHV-001治疗组为160mm3,PBS未治疗组小鼠31天全部死亡,而WHV-001治疗组存活率为62.5%(5/8)。
同时我们也用小鼠结直肠细胞MC-38在C57BL/6J小鼠建立皮下荷瘤模型。每只小鼠接种5.0×105(100uL)MC-38细胞。接种后第8~10天,小鼠肿瘤体积达到50mm3,荷瘤小鼠随机分为3组(n=8),分别为PBS组和WHV-001组,分别瘤内注射100uL PBS、1×108PFU WHV-001病毒,每两天注射一次,共注射3次。每两天测量一次肿瘤体积,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。WHV-001治疗可以非常显著的抑制小鼠结直肠癌细胞的生长(图23A),在接种后28天,PBS未治疗组肿瘤平均大小为1780mm3,而WHV-001治疗组为172mm3,PBS未治疗组小鼠30天全部死亡,而WHV-001治疗组在45天全部死亡,死亡时间明显延迟(图23B)。
由上可知溶瘤病毒WHV-001对结直肠癌具有非常显著的治疗效果。
实施例10重组病毒WHV-001抑制乳腺癌治疗效果验证
6-8周龄Balb/c小鼠右侧皮下接种小鼠结乳腺癌细胞4T1,每只小鼠接种5.0×105(100uL)4T1细胞。接种后第8~10天,小鼠肿瘤体积达到50mm3,荷瘤小鼠随机分为2组(n=8),分别为PBS组和WHV-010组,分别瘤内注射100uL PBS、1×107PFU WHV-006和WHV-010病毒,每两天注射一次,共注射3次。每两天测量一次肿瘤体积,并统计小鼠生存率。小鼠肿瘤体积计算公式:体积=短径2×长径×0.5。WHV-001治疗可以非常显著的抑制小鼠乳腺癌细胞的生长(图24),在接种后32天,PBS未治疗组肿瘤平均大小为934mm3,而WHV-001治疗组为488mm3。由上可知溶瘤病毒WHV-001006对乳腺癌的生长具有非常显著的抑制效果。
Claims (8)
1.一种增强型溶瘤病毒,其特征在于,所述溶瘤病毒基因组中包括:
(i)第一核酸序列,所述第一核酸序列编码PD-L1单链抗体;
(ii)第二核酸序列,所述第二核酸序列编码CD40L分子;
(iii)第三核酸序列,所述第三核酸序列编码IL2mimic分子,所述IL2mimic是IL-2受体的多肽激动剂;
所述PD-L1单链抗体包含重链抗体可变区和轻链抗体可变区;
所述重链抗体可变区为SEQ ID NO:5所示的氨基酸序列,所述轻链抗体可变区为SEQID NO:6所示的氨基酸序列;
所述重链抗体可变区与所述轻链抗体可变区通过铰链序列连接,且所述重链抗体可变区位于所述轻链抗体可变区的N端;所述铰链序列为SEQ ID NO:15所示的氨基酸序列;
所述PD-L1单链抗体还包含分泌信号肽,所述第一核酸序列编码的PD-L1单链抗体为SEQ ID NO:7所示的氨基酸序列;
所述第二核酸序列编码的CD40L分子为SEQ ID NO:18所示的氨基酸序列;
所述第三核酸序列编码的IL2mimic分子为SEQ ID NO:19所示的氨基酸序列;
所述溶瘤病毒是水疱性口炎病毒。
2.根据权利要求1中所述的增强型溶瘤病毒,其特征在于,所述第一核酸序列、第二核酸序列和第三核酸序列通过编码接头元件的核酸序列连接;所述接头元件可以是被蛋白酶识别的短肽序列,所述接头元件是P2A序列,所述P2A序列编码的氨基酸序列为SEQ ID NO:16所示的氨基酸序列。
3.根据权利要求2所述的增强型溶瘤病毒,其特征在于,所述第一核酸序列、第二核酸序列和第三核酸序列位于所述溶瘤病毒基因组的G和L基因之间。
4.根据权利要求3所述的增强型溶瘤病毒,其特征在于,所述第一核酸序列、第二核酸序列、第三核酸序列和P2A序列按照以下顺序存在:第二核酸序列-P2A序列-第一核酸序列-P2A序列-第三核酸序列。
5.根据权利要求1-4中任一项所述的增强型溶瘤病毒,其特征在于,所述溶瘤病毒基因组中进一步包含增强哺乳动物翻译的核酸,所述增强哺乳动物翻译的核酸是Kozak序列,所述Kozak序列为SEQ ID NO:14所示的核苷酸序列。
6.一种用于治疗癌症的药物组合物,其特征在于,所述药物组合物包括权利要求1-5中任一项所述的溶瘤病毒,所述癌症是结直肠癌。
7.一种如权利要求1-5任一项所述的溶瘤病毒在制备抗癌症药物中的应用;所述癌症是结直肠癌。
8.一种试剂盒,其特征在于,所述试剂盒包括权利要求1-5中任一项所述的溶瘤病毒。
