细胞穿透肽(Cell-penetrating-peptides,CPPs)是一种少于30个氨基酸的多肽,作为药物输送载体正在进入人们的视野。CPPs是一类具有强渗透性的小分子,可以将多肽,蛋白质,核酸等大分子携带到细胞内,这些肽能够进入细胞而不产生细胞溶解作用。此外,它们有效地绕过BBB中的P-糖蛋白(P-gp),因此为外源性物质进入细胞甚至BBB开辟了新的途径。近年来,越来越多的研究表明,使用CPPs将药物分子传递到大脑是一种有效的策略。CPPs通过连接核酸,蛋白,多肽,小分子药物等,将其传递进入细胞,这种连接可以是共价的也可以是非共价的(Curr Pharm Biotechnol.2012Sep 13(12):2417-26,Int J MolSci.2022Aug 12;23(16):9038)。
SUL水溶性差,t1/2为8~9h,半衰期短,口服生物利用度差(20%~30%),且存在肠道P-gp的外排,不易穿透BBB,进入脑脊液中很少。SUL副作用较少,主要集中在体重增加和血清中泌乳素释放增多两个方面。血清中泌乳素释放增加的主要原因是SUL虽然具有拮抗多巴胺受体的作用,但是并不能特异性作用于中枢的多巴胺受体,当其作用于外周,如脑垂体的D2受体时,多巴胺作为催乳素释放抑制因子的一种,不能结合D2受体发挥抑制泌乳素释放的作用,导致垂体乳状细胞释放催乳素增多,就会引起血清中泌乳素水平的升高,造成高泌乳素血症。
发明内容
发明目的:本发明的目的是提供一种增强舒必利抗抑郁效果、减少副作用的细胞穿膜肽偶联舒必利前药体系;本发明的另一目的是提供一种细胞穿膜肽偶联舒必利前药体系的制备方法;本发明的另一目的是提供一种细胞穿膜肽偶联舒必利前药体系的应用。
技术方案:本发明的一种细胞穿膜肽偶联舒必利前药体系,所述细胞穿膜肽偶联舒必利前药体系由细胞穿透肽和舒必利偶联所得,所述偶联为通过酰胺键共价连接。
上述方案中,将细胞穿透肽与舒必利通过酰胺键共价连接,实现让更多的舒必利进入大脑,减少舒必利对外周D2受体的拮抗,主要作用于中枢的D2受体,不仅可以增强舒必利的抗抑郁效果,还能抑制泌乳素的过多释放,减少血清中泌乳素的浓度,降低舒必利带来的副反应。
优选的,所述细胞穿膜肽偶联舒必利前药体系包括如通式(Ι)所示的化合物VPALR-SUL:
上述方案中,细胞穿透肽为CPPs VPALR(Val-Pro-Ala-Leu-Arg),CPPs VPALR(Val-Pro-Ala-Leu-Arg)与SUL通过酰胺键共价连接得到VPALR-SUL。
另一方面,本发明提供一种细胞穿膜肽偶联舒必利前药体系的制备方法,所述化合物VPALR-SUL的制备为:Fmoc-VPALR多肽的羧基与舒必利的氨基反应连接得到VPALR-SUL;所述Fmoc-VPALR多肽中,VPALR为Val-Pro-Ala-Leu-Arg;Fmoc为氨基保护基,其用于减少Val-Pro-Ala-Leu-Arg的自身环化;Fmoc-VPALR的化学结构式如下所示:
优选地,所述Fmoc-VPALR多肽通过固相合成的方法制备。
作为上述方案的进一步改进,所述Fmoc-VPALR多肽的制备包括以下步骤:
(1)取氯三苯甲基氯树脂加入固相合成管中,然后加入二氯甲烷活化一段时间,得到活化后的氯三苯甲基氯树脂;
(2)再向上述固相合成管中加入Fmoc-Arg(Mtr)-OH、N,N-二异丙基乙胺以及助溶剂,反应一段时间后,封闭氯三苯甲基氯树脂的活化位点,然后再加入哌啶/DMF溶液,以切去Fmoc保护基团;
(3)再向固相合成管中加入Fmoc-Leu-OH、连接剂和助溶剂,反应一段时间,然后按照上述方法依次加入Fmoc-Ala-OH,Fmoc-Pro-OH,Fmoc-Val-OH,并保留最后一个氨基酸的Fmoc;反应结束后,得到Fmoc-VPALR多肽。
作为上述方案的进一步改进,具体的,所述化合物VPALR-SUL的制备包括以下步骤:
(1)有机溶剂中分别加入Fmoc-VPALR多肽、舒必利、连接剂和催化剂,避光搅拌反应一段时间,得到第一产物;
(2)第一产物中加入萃取剂,以去除有机溶剂,得到第二产物;第二产物中再加入哌啶/DMF溶液进行反应,以切去第二产物中的Fmoc保护基团;得到化合物VPALR-SUL。
优选地,步骤(1)中,Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶的摩尔比为1:1:2:2~1:2:2:2。更优选地,Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶的摩尔比为1:1:2:2。
优选地,所述连接剂为1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐,所述催化剂为4-二甲氨基吡啶。
优选地,步骤(1)中,有机溶剂为二甲基甲酰胺;由于SUL不溶于二氯甲烷,所以采用了二甲基甲酰胺(DMF)作为反应溶剂,优选的,步骤(2)中,萃取剂为0~4℃的乙酸乙酯。
上述技术方案中,将Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶溶解于适量DMF中,在磁力搅拌器上搅拌24h。由于DMF沸点高,通常需要油泵抽走DMF,过程繁琐,且DMF不易被完全除净。所以,采用了液液萃取的方法除掉残留的DMF。选用了乙酸乙酯或者乙醚进行试验,因为乙醚和乙酸乙酯都易挥发,不易残留,且DMF溶于两者,化合物不溶于两者。分别用两种萃取剂进行萃取,发现乙酸乙酯萃取后,能将实验中剩下的催化剂4-DMAP萃取出来,提高了本发明化合物的纯度,乙醚容易发生爆炸,萃取过程中不能使用离心机离心除去上清液,所以选用了乙酸乙酯。低温有助于化合物的析出,所以,最终选取0~4℃的乙酸乙酯作为萃取溶剂。
优选地,步骤(1)中,所述避光搅拌反应的时间为24~48h。
上述技术方案中,对搅拌时间进行了优化,分别搅拌了24h,36h,48h,得到的化合物分别进行产率的计算和纯度分析,发现经过24h反应后,VPALR-SUL的产率最高,纯度最高。
本发明的技术关键点在于:CPPs VPALR与SUL的共价连接。SUL存在磺胺基团,而合成的VPALR,R的侧链存在羧基,为了减少VPALR自身环化,最后一个氨基酸的氨基保护基Fmoc未切除,将两者的羧基和氨基连接,形成酰胺键,构成VPALR-SUL。
另一方面,本发明提供一种细胞穿膜肽偶联舒必利前药体系在制备抗抑郁症药物中的应用。
本发明化合物与SUL相比,对脂多糖(Lipopolysaccharide,LPS)造模的炎症抑郁小鼠具有更好的治疗作用。在体内药效试验中,长期腹腔给药7天后,VPALR-SUL组小鼠悬尾实验和旷场实验结果均表现出比SUL组更明显的药效,例如悬尾实验中VPALR-SUL组小鼠的不动时间缩短,旷场实验小鼠的总路程增加,并且更趋向于向中央区活动。ELISA法分别测定小鼠脑组织中炎症因子IL-1β,TNF-a,IL-10的浓度,化学法分别测定MDA浓度,caspase-3和TNOS的活性等生化指标,实验结果表明VPALR-SUL组小鼠脑组织中炎症因子浓度降低,TNOS和caspase-3的活力减弱,再一次验证了VPALR-SUL可以缓解LPS造模小鼠的神经炎症。同时,在腹腔给药7天后,采用ELISA的方法测定了两组小鼠血清中的泌乳素浓度,发现VPALR-SUL组小鼠血清中的泌乳素浓度确实低于SUL组,这个实验结果再一次验证了VPALR-SUL可降低SUL带来的副反应。
有益效果:与现有技术相比,本发明具有如下显著优点:本发明化合物与SUL相比,对脂多糖(Lipopolysaccharide,LPS)造模的炎症抑郁小鼠具有更好的治疗作用。实验结果表明VPALR-SUL组小鼠脑组织中炎症因子浓度降低,TNOS和caspase-3的活力减弱。

下面结合附图对本发明的技术方案作进一步说明。本发明实施例提供一种细胞穿膜肽偶联舒必利前药体系,其将CPPs VPALR(Val-Pro-Ala-Leu-Arg)与SUL通过酰胺键共价连接,实现让更多的SUL进入大脑,减少SUL对外周D2受体的拮抗的目的。其中所述细胞穿膜肽偶联舒必利前药体系包括如通式(Ι)所示的化合物VPALR-SUL:

下面以具体实施例进行详细说明。
实施例1Fmoc-VPALR的合成
本实施例提供了一种Fmoc-VPALR的合成方法,其使用固相合成的方法合成Fmoc-VPALR多肽,具体步骤如下:
(1)称取约1g 2-氯三苯甲基氯树脂(1.0~1.2mmol/g)加入固相合成管中,加入20mL二氯甲烷(DCM),活化30min后过滤,DCM洗涤3次,每次摇摆2min。
(2)向上述固相合成管中加入Fmoc-Arg(Mtr)-OH,N,N-二异丙基乙胺(DIPEA),和20mL DCM。作为反应中的助溶剂,DIPEA是氨基酸摩尔量的1/3倍,摇摆2h抽干,DCM洗涤3次,每次约2min。加入DCM:甲醇:DIPEA(80:15:5)混合液15mL封闭树脂活化位点,摇摆15min后抽干,再加入上述混合液15mL,摇摆15min后抽干,用DMF洗涤5次,每次摇摆约2min。加入20%哌啶/DMF(V/V)溶液20mL,摇摆5min抽干后,再加入该溶液20mL摇摆30min,切去Fmoc保护基团,方便下一个氨基酸的连接,加入适量DMF洗涤5次,每次摇摆约2min。
(3)加入摩尔量是第一个氨基酸2倍的Fmoc-Leu-OH,连接剂为O-苯并三氮唑-N,N,N',N'-四甲基脲四氟硼酸(TBTU)和1-羟基苯并三唑(HOBT),助溶剂为DIPEA,用10mL DMF溶解后,加至固相合成管中,再加入10mL DMF,摇摆3h后,分别用DCM和DMF洗涤5次,每次摇摆2min。按照上述方法分别加入Fmoc-Ala-OH,Fmoc-Pro-OH,Fmoc-Val-OH,保留最后一个氨基酸的Fmoc。最后一个氨基酸反应结束后,加入99% TFA搅拌2h,过滤留取滤液,再用新鲜的TFA冲洗树脂2~3次,收集滤液旋蒸浓缩。加入0~4℃乙醚分散得固体,超声15min后静置2h。过滤得固体,乙醚淋洗3次后,沉淀物加入适量重蒸水,冷冻干燥得Fmoc-VPALR白色产物,产率约95.6%,纯度约98.9%。

实施例2化合物VPALR-SUL的合成
本实施例提供一种化合物VPALR-SUL的合成方法,包括以下步骤:
(1)向25mL DMF中加入Fmoc-VPALR多肽(20.23mg,2mM),舒必利(7mg,2mM),1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐(EDC-HCL,8mg,4mM),4-二甲氨基吡啶(4-DMAP,5mg,4mM),避光搅拌24h。
(2)加入0~4℃乙酸乙酯萃取,反复萃取3次,加入水冻干得白色粉末。再加入25mL20%哌啶/DMF溶液,避光室温搅拌1h,0~4℃乙醚萃取3次,静置,冷冻干燥的白色粉末,产率约为60.2%,纯度约94.9%。
图4显示了ESI-MS:C40 H67N11O9S,calc.MW=877.6,obsvd.[M+H]+=878.6.1H NMR(500MHz,DMSO-d6)(图5)。
实施例3
本实施例提供一种化合物VPALR-SUL的合成方法,其中与实施例2的区别在于,步骤(1)中避光搅拌时间不同,本实施例避光搅拌36h,具体的,所述合成方法包括以下步骤:
(1)向25mL DMF中加入Fmoc-VPALR多肽(20.23mg,2mM),舒必利(7mg,2mM),1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐(EDC-HCL,8mg,4mM),4-二甲氨基吡啶(4-DMAP,5mg,4mM),避光搅拌36h。
(2)加入0~4℃乙酸乙酯萃取,反复萃取3次,加入水冻干得白色粉末。再加入25mL20%哌啶/DMF溶液,避光室温搅拌1h,0~4℃乙醚萃取3次,静置,冷冻干燥的白色粉末,产率约为45.3%,纯度约60.5%。
实施例4
本实施例提供一种化合物VPALR-SUL的合成方法,其中与实施例2的区别在于,步骤(1)中避光搅拌时间不同,本实施例避光搅拌48h,具体的,所述合成方法包括以下步骤:
(1)向25mL DMF中加入Fmoc-VPALR多肽(20.23mg,2mM),舒必利(7mg,2mM),1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐(EDC-HCL,8mg,4mM),4-二甲氨基吡啶(4-DMAP,5mg,4mM),避光搅拌48h。
(2)加入0~4℃乙酸乙酯萃取,反复萃取3次,加入水冻干得白色粉末。再加入25mL20%哌啶/DMF溶液,避光室温搅拌1h,0~4℃乙醚萃取3次,静置,冷冻干燥的白色粉末,产率约为49.4%,纯度约39.8%。
通过实施例2-4的比较发现,在相同条件下,避光搅拌的时间控制在24h时,VPALR-SUL的产率最高,纯度最高。
实施例5
本实施例提供一种化合物VPALR-SUL的合成方法,其中与实施例2的区别在于,步骤(1)中Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶的加入量不同,本实施例中Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶的摩尔比为1:2:2:2。具体的,所述合成方法包以下步骤:
(1)向25mL DMF中加入Fmoc-VPALR多肽(20.23mg,2mM),舒必利(7mg/14mg,2/4mM),1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐(EDC-HCL,8mg,4mM),4-二甲氨基吡啶(4-DMAP,5mg,4mM),避光搅拌48h。
(2)当摩尔比为1:1:2:2时,加入0~4℃乙酸乙酯萃取,反复萃取3次,加入水冻干得白色粉末。再加入25mL 20%哌啶/DMF溶液,避光室温搅拌1h,0~4℃乙醚萃取3次,静置,冷冻干燥的白色粉末,产率约为60.2%,纯度约94.9%。当摩尔比为1:2:2:2时,产率约为50.6%,纯度约39.9%。
对比例1
本对比例提供一种化合物VPALR-SUL的合成方法,其中与实施例2的区别在于,步骤(2)中使用的萃取剂不同,本对比例使用的萃取剂为乙醚,具体步骤如下:
(1)向25mL DMF中加入Fmoc-VPALR多肽(20.23mg,2mM),舒必利(7mg,2mM),1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐(EDC-HCL,8mg,4mM),4-二甲氨基吡啶(4-DMAP,5mg,4mM),避光搅拌24h。
(2)加入乙醚萃取,反复萃取3次,加入水冻干得白色粉末。再加入25mL20%哌啶/DMF溶液,避光室温搅拌1h,0~4℃乙醚萃取3次,静置,冷冻干燥的白色粉末,产率约为60.2%,纯度约58.4%。
在以上萃取的过程中发现,乙醚容易发生爆炸,萃取过程中不能使用离心机离心除去上清液,因此,与实施例2相比,对比例1的纯度不高。
实施例6体内抗抑郁药效实验:
为了验证实施例2制得VPALR-SUL的抗抑郁效果,进行体内抗抑郁药效实验,具体步骤如下:
ICR小鼠(18~22g)均购自江苏南京华创生物科技有限公司,在持续温度(24±1℃)、相对湿度(50±10%)和12h昼夜周期下,每天给予充足的食物和水。动物实验按照美国国立卫生研究院《实验动物护理和使用指南》(NIH出版物第8023号,1978年修订)的指导方针进行,并得到中国药科大学动物伦理委员会的批准(受理号:2023-06-003)。ICR小鼠分成4组,对照组,模型组,SUL组,VPALR-SUL组。SUL组和VPALR-SUL组分别腹腔注射30mg/kg SUL和80mg/kg VPALR-SUL7天,最后一天给药1h后,腹腔注射0.8mg/kg的LPS造模,6h后,进行旷场实验和悬尾实验,24h后,小鼠摘眼球取血,取脑组织。根据ELISA试剂盒的说明书进行操作,ELISA方法测定脑组织中TNF-a,IL-1β,IL-10三种炎症因子的浓度,化学法测定脑组织中caspase-3活性,MDA浓度和T-NOS活性,BCA测定组织蛋白浓度。取约0.1g组织,加入900μL的PH7.2~7.4冰冷的PBS溶液,匀浆,12000rpm,4℃离心10min,取上清液,备用。
实验结果表明,SUL组和VPALR-SUL组小鼠的活动轨迹走向中央区,并且VPALR-SUL组的小鼠在中央区活动的轨迹明显比SUL组更长,说明了VPALR-SUL组小鼠活动能力较SUL组更强(图6A),VPALR-SUL组小鼠活动总路程长度和平均速度较SUL组小鼠有所增加(图6B,C)。从挣扎时间来看,VPALR-SUL组的挣扎时间与SUL组相比,延长的更多。同样的,在不动时间的实验结果中,VPALR-SUL组的不动时间显著缩短,说明了VPALR-SUL使小鼠的绝望状态得到了更好的缓解(图7)。VPALR-SUL组脑组织中IL-1β,TNF-a炎症因子的浓度相比于SUL组显著降低(图8A,B),说明了VPALR-SUL让LPS引起的炎症得到了更好的缓解,减轻了小鼠的抑郁状态。TNOS活性测定实验结果表明(图8E),VPALR-SUL组的TNOS活性降低的更多,说明了氮化应激产物一氧化氮的减少。VPALR-SUL组与SUL组相比,小鼠脑内的caspase-3活力明显下降,说明了VPALR-SUL组降低了神经细胞发生凋亡的可能(图8D)。VPALR-SUL组小鼠血清中泌乳素浓度比SUL组也有所降低(图9)。

7.根据权利要求6所述的细胞穿膜肽偶联舒必利前药体系的制备方法,其特征在于,步骤(1)中,Fmoc-VPALR多肽、舒必利、1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐和4-二甲氨基吡啶的摩尔比为1:1:2:2~1:2:2:2。
8.根据权利要求6所述的细胞穿膜肽偶联舒必利前药体系的制备方法,其特征在于,步骤(1)中,所述连接剂为1-(3-二甲氨基丙基-3-乙基碳二亚胺盐酸盐;和/或,所述催化剂为4-二甲氨基吡啶;和/或,有机溶剂为二甲基甲酰胺;和/或,步骤(2)中,萃取剂为0~4℃的乙酸乙酯。
9.根据权利要求6所述的细胞穿膜肽偶联舒必利前药体系的制备方法,其特征在于,步骤(1)中,所述避光搅拌反应的时间为24-48h。
10.一种根据权利要求1-2任一所述的细胞穿膜肽偶联舒必利前药体系在制备抗抑郁症药物中的应用。
