Abstract
本申请提供了一种含胶原蛋白的修复材料的制备方法,所述制备方法包括将鱼皮胶原蛋白溶于热水,加入植物多酚以及冷却的步骤。本申请的方法所制备的材料在凝胶强度、保水性能和功能之间实现了良好的平衡,适用于医用敷料和化妆品等多个领域,其中的胶原蛋白提供了良好的保水和承载性能;儿茶素等植物多酚以及硫酸锌不仅改善了凝胶性能,还能提供抗氧化、抑菌效果和修复效果。
Description
技术领域
本申请属于蛋白领域和皮肤修复领域,具体地,本申请提供了一种含胶原蛋白的修复材料的制备及其应用。
背景技术
胶原蛋白是多种动物细胞外基质的组成成分,由于其具有保水、润滑、屏障等多种功能,在医疗、化妆品、食品等领域中有广泛的应用。不同来源的胶原蛋白性质差异明显,鱼皮胶原蛋白来源丰富(鱼食品加工常见废料),提取前处理步骤简单,保水性能佳,是胶原蛋白的常见来源。
将胶原蛋白制备成凝胶形式用作敷料、化妆品如面膜等非常方便,但不同来源的胶原蛋白形成的凝胶在强度、保水性能等方面各有特点;此外,为丰富产品功能,往往需要在凝胶中加入其他功能成分,不同与其他承载成分之间的相互作用也各不相同。
目前此类产品的种类和功能还有待丰富。
发明内容
为改善胶原蛋白凝胶的性能并赋予其其更多的功能,我们在凝胶中含有代表性植物多酚的多种植物提取物,绿茶提取物(儿茶素)、葡萄籽提取物(原花青素)、石榴皮提取物(鞣花酸)以及多种盐成分,寻求凝胶强度、保水性能和功能之间的平衡。
一方面,本申请提供了一种含胶原蛋白的修复材料的制备方法,所述制备方法包括将胶原蛋白溶于热水,加入植物多酚以及冷却的步骤;所述修复材料为凝胶形式。
进一步地,所述胶原蛋白为鱼皮胶原蛋白。
进一步地,所述胶原蛋白为鲫鱼皮胶原蛋白。
进一步地,所述制备方法包括配制1-4%w/v的胶原蛋白溶液,将pH调至6;加热至30-50℃,保持20-50分钟;加入植物多酚4-6%和盐0.005%-0.01%,混合均匀;置于1-8℃冰箱中冷却4-12小时。
进一步地,所述植物多酚选自绿茶提取物、葡萄籽提取物、石榴皮提取物中的一种;所述盐选自硫酸锌或硫酸铜。
进一步地,所述植物多酚选自儿茶素、原花青素、鞣花酸;所述盐为硫酸锌。
进一步地,所述制备方法为配制3%w/v的胶原蛋白溶液,将pH调至6;加热至35℃,保持30分钟;加入儿茶素4%和硫酸锌0.01%,混合均匀;置于4℃冰箱中冷却8小时。
另一方面,本申请提供了根据上述方法制备的修复材料。
进一步地,所述修复材料中还包含选自除儿茶素、硫酸锌之外的抑菌剂、除儿茶素之外的抗氧化剂、酪氨酸激酶抑制剂、生长因子的成分。
上述抑菌剂、抗氧化剂、酪氨酸激酶抑制剂、生长因子本领域技术人员可以根据相关规定选用允许的种类和用量,并根据其耐热性能选择加入时机。例如,对于对温度不敏感的成分,可以选择随胶原加入或者在加热后随植物多酚和盐一起加入,对于对热敏感的成分,可以在冷却至一定温度时加入。本领域技术人员也可以常规验证这些成分对凝胶性能的影响程度。
另一方面,本申请提供了上述修复材料在制备修复皮肤的化妆品,如面膜或皮肤修复凝胶或医用敷料中的用途。
本申请材料中胶原蛋白提供了良好的保水和承载性能;儿茶素等植物多酚以及硫酸锌不仅改善了凝胶性能,还能提供抗氧化、抑菌效果和修复效果。本申请的修复材料不仅可以单独使用,也可用于承载其他美白和修复成分。

具体实施方式
实施例1 主要原料和实验方法
原料:
儿茶素,绿茶提取物,纯度98%(西安嘉和生物技术有限公司)。
原花青素,葡萄籽提取,纯度94%(广州泽力医药科技有限公司)。
鞣花酸,石榴皮提取,纯度92%(西安嘉和生物技术有限公司)。
鲫鱼鱼皮,购自江苏某养殖场(同品种、基本同规格:200-300g),宰杀后取鱼皮。
硫酸钠、硫酸铜、硫酸钾、硫酸锌购自江苏科伦多食品配料有限公司和苏州智翔精细化工有限公司。
鲫鱼鱼皮胶原蛋白的提取:
按照现有技术中的记载,鲫鱼鱼皮胶原蛋白的提取方法为:
(1)将鱼皮清洗后剪碎,按照料液比1:5(m:v)加入0.5M的醋酸溶液中,匀浆;
(2)4℃下浸泡提取48小时,过滤取滤液;
(3)0.05M的Tris-HCl缓冲液(pH 8.0)将滤液透析12小时;
(4)离心,冷冻干燥即得鲫鱼鱼皮胶原蛋白。
凝胶强度的检测:
使用凝胶强度计(海博,HBS-NJ01,1厘米探头)检测凝胶强度。测试时:统一配制3%(w/v)的胶原蛋白溶液,pH调至6,加热至30℃,保持30分钟,置于4℃冰箱中冷却8小时后测试凝胶强度(三次取平均)。
保水性的检测:
通过离心法测定凝胶的保水性能,取一定量的凝胶在4℃下10000g离心10分钟,测量离心前离心管重量,离心后倾倒出液体后离心管的重量,以离心后离心管的重量与离心前离心管重量的比值作为保水性指标(%,三份取平均)。
实施例2 加入植物多酚对凝胶强度的影响
依照实施例1“凝胶强度的检测”部分的凝胶配制方法,在加热至35℃,保持30分钟后加入一定量的儿茶素、原花青素、鞣花酸(按照胶原溶液的体积计算w/v百分数)。结果如图1所示,三种植物多酚的加入对于凝胶强度均有正面影响,低用量(1-2%)时对凝胶强度的影响不显著,在4%-5%时则对凝胶强度有明显的增强效果。
实施例3 加入植物多酚对凝胶保水性能的影响
按照实施例1中“保水性的检测”部分所述的方法检测实施例2中所制备凝胶的保水性能,结果如图2所示,凝胶的保水性能基本与其凝胶强度成反比,随着植物多酚用量的增加而降低,相比鞣花酸和原花青素,加入儿茶素后导致的凝胶保水性下降幅度较低。
实施例4加入金属离子
按照申请人的预期,植物多酚中的羟基与胶原中氢键的连接是导致凝胶结构和性能变化的主要原因。申请人尝试加入金属离子来改变胶原自聚集性能以尽量保持凝胶的保水性。实验以加入5%儿茶素的凝胶为基础进行,在加入儿茶素的同时加入0.005%、0.01%、0.02%的硫酸钠、硫酸铜、硫酸钾、硫酸锌(以无水硫酸盐计算)。产品的保水性和凝胶强度如下表1所示:

结果表明,加入各种金属离子对于凝胶强度和保水性能的影响不一。出于效果和安全性的考量,我们选择硫酸锌作为添加剂(目前的研究表明,硫酸锌外用有防腐抑菌以及促进组织再生的效果且安全性良好,是食品、药品和化妆品中允许添加的原料)。
Claims (7)
1.一种含胶原蛋白的修复材料的制备方法,所述制备方法包括将胶原蛋白溶于热水,加入植物多酚以及冷却的步骤;所述修复材料为凝胶形式;所述胶原蛋白为鱼皮胶原蛋白;所述制备方法包括配制1-4%w/v的胶原蛋白溶液,将pH调至4-7;加热至30-50℃,保持20-50分钟;加入植物多酚4-6%和盐0.005%-0.01%,混合均匀;置于1-8℃冰箱中冷却4-12小时。
2.根据权利要求1所述的制备方法,其中所述胶原蛋白为鲫鱼皮胶原蛋白。
3.根据权利要求1所述的制备方法,其中所述植物多酚选自绿茶提取物、葡萄籽提取物、石榴皮提取物中的一种;所述盐选自硫酸锌或硫酸铜。
4.根据权利要求3所述的制备方法,其中所述植物多酚选自儿茶素、原花青素、鞣花酸中的一种;所述盐为硫酸锌。
5.根据权利要求1所述的制备方法,其中所述制备方法为配制3%w/v的胶原蛋白溶液,将pH调至6;加热至35℃,保持30分钟;加入儿茶素4%和硫酸锌0.01%,混合均匀;置于4℃冰箱中冷却8小时。
6.使用根据权利要求1-5任一项所述的制备方法制备得到的含胶原蛋白的修复材料。
7.根据权利要求6所述的修复材料在制备修复皮肤的化妆品或医用敷料中的用途。
