Abstract
本发明公开一种壳聚糖‑聚乙烯醇SOD酶水凝胶敷料,该水凝胶敷料包括背衬层、粘结剂层和水凝胶层,其特点在于所述水凝胶层由壳聚糖‑聚乙烯醇水凝胶、SOD酶溶液、明胶和琼脂组成,其中,SOD酶溶液与壳聚糖‑聚乙烯醇水凝胶的体积比为1:1~1:2,明胶与壳聚糖‑聚乙烯醇水凝胶的质量体积比为1~3:10g/ml,琼脂与壳聚糖‑聚乙烯醇水凝胶的质量体积比为0.5~1.5:10g/ml。与现有水凝胶敷料相比,本发明的壳聚糖‑聚乙烯醇SOD酶水凝胶敷料具有生物相容性好,抗过敏性好,伤口愈合快的优势。

技术领域
本发明属于外科医用敷料领域,具体涉及一种水凝胶敷料。
背景技术
近年来研究发现,壳聚糖及其衍生物具有良好的抗菌活性,壳聚糖及其衍生物以不同的形式在抗菌领域的研究较为广泛。而SOD酶则是对外伤有很好的促进愈合的功效,将两者结合起来对一些较严重的外伤及手术缝合口可以加快愈合的作用。
发明内容
本发明的目的在于提供一种可以加快伤口愈合的壳聚糖-聚乙烯醇SOD酶水凝胶敷料。
本发明实现上述目的所采用的技术方案如下:
壳聚糖-聚乙烯醇SOD酶水凝胶敷料,包括背衬层、粘结剂层和水凝胶层,其特点在于:所述水凝胶层由壳聚糖-聚乙烯醇水凝胶、SOD酶溶液、明胶和琼脂组成,其中,SOD酶溶液与壳聚糖-聚乙烯醇水凝胶的体积比为1:1~1:2,明胶与壳聚糖-聚乙烯醇水凝胶的质量体积比为1~3:10g/ml,琼脂与壳聚糖-聚乙烯醇水凝胶的质量体积比为0.5~1.5:10g/ml。
优选地,所述粘结剂层选自马来胶、α-氰基丙烯酸酯和水乳胶。
通过对比对皮肤接触的舒适度和过敏度,发现α-氰基丙烯酸酯生物相容性好,抗过敏性好。
优选地,所述水乳胶为羧基丁腈胶乳。
水凝胶层中加入的明胶和琼脂可以使敷料贴合在皮肤表面时更舒适柔和。
优选地,所述粘结剂层由马来胶30~50vt%、α-氰基丙烯酸酯40~60vt%和水乳胶10~20vt%组成。
优选地,SOD酶溶液的浓度为5~15mg/L。
优选地,壳聚糖-聚乙烯醇水凝胶是以聚乙烯醇和壳聚糖为主要原料,京尼平为交联剂,通过循环冷冻-解冻法制备得到。
更优选地,聚乙烯醇与壳聚糖的质量比为1~3:1,最优选地为2:1。
优选地,壳聚糖与水的质量体积比10~30g:100ml,更优选地为20g:100ml。
有益效果
与现有技术相比,本发明的壳聚糖-聚乙烯醇SOD酶水凝胶敷料具有生物相容性好,抗过敏性好,伤口愈合快的优势。

具体实施方式
以下结合实施例对本发明的技术方案做进一步详细说明。
原料:
水乳胶采用羧基丁腈胶乳;
壳聚糖-聚乙烯醇水凝胶是以聚乙烯醇(PVA)和壳聚糖(CS)为主要原料,京尼平为交联剂,通过循环冷冻-解冻法制备得到,具体制备过程如下:
将聚乙烯醇80g,壳聚糖40g,加入200ml的沸水中,使其变成胶状物质,冷却到25℃时,加入京尼平1mg,使其交联结合。放入-20℃冷冻24h,室温解冻4h,接着冷冻24h,室温解冻4h,反复4次,即得到壳聚糖-聚乙烯醇水凝胶。
抗菌实验
在4个营养琼脂板上分别加入水10ml,壳聚糖-聚乙烯醇水凝胶10ml,SOD酶溶液10ml(10mg/L),壳聚糖-聚乙烯醇水凝胶5ml+SOD酶溶液5ml(10mg/L)。在37℃的培养箱中,72h后观察菌落数。实验结果如下:
1)只加水观察的菌落数为15个
2)只加壳聚糖-聚乙烯醇水凝胶观察的菌落数为8个
3)只加SOD酶溶液观察的菌落数为6个
4)壳聚糖-聚乙烯醇水凝胶+SOD酶溶液观察的菌落数为2个。
结果表明,壳聚糖-聚乙烯醇水凝胶与SOD酶结合有抗菌协同作用。
实施例1
壳聚糖-聚乙烯醇SOD酶水凝胶敷料的结构如图1-2所示,依次包括背衬层、粘结剂层和水凝胶层。
背衬层采用聚氨酯薄膜或无纺布。
粘结剂层由马来胶8ml,α-氰基丙烯酸酯10ml,水乳胶3ml组成,将各原料混合均匀后涂布于背衬层上。
水凝胶层由壳聚糖-聚乙烯醇水凝胶10ml,SOD酶溶液(10mg/L)5ml,明胶2g,琼脂1g组成,将各原料混合均匀后涂布于粘结剂层(贴合皮肤面)上。
对比例1
水凝胶层由壳聚糖-聚乙烯醇水凝胶15ml,明胶2g,琼脂1g组成,其它与实施例1相同。
伤口愈合实验
在3只大鼠的腿部制造相似的伤口,一只自然愈合,其它分别贴实施例1及对比例1的水凝胶敷料,观察大鼠的伤口愈合情况,重复实验3次。实验结果如表1所示:

实验结果表明,本发明的壳聚糖-聚乙烯醇SOD酶水凝胶敷料可以有效加快伤口的愈合。

1.壳聚糖-聚乙烯醇SOD酶水凝胶敷料,包括背衬层、粘结剂层和水凝胶层,其特征在于:所述水凝胶层由壳聚糖-聚乙烯醇水凝胶、SOD酶溶液、明胶和琼脂组成,其中,SOD酶溶液与壳聚糖-聚乙烯醇水凝胶的体积比为1:1~1:2,明胶与壳聚糖-聚乙烯醇水凝胶的质量体积比为1~3:10g/mL,琼脂与壳聚糖-聚乙烯醇水凝胶的质量体积比为0.5~1.5:10g/mL。
2.根据权利要求1所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:所述粘结剂层选自马来胶、α-氰基丙烯酸酯和水乳胶。
3.根据权利要求2所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:所述粘结剂层由马来胶30~50vt%、α-氰基丙烯酸酯40~60vt%和水乳胶10~20vt%组成。
4.根据权利要求2或3所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:所述水乳胶为羧基丁腈胶乳。
5.根据权利要求1所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:SOD酶溶液的浓度为5~15mg/L。
6.根据权利要求1所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:壳聚糖-聚乙烯醇水凝胶是以聚乙烯醇和壳聚糖为主要原料,京尼平为交联剂,水为溶剂,通过循环冷冻-解冻法制备得到。
7.根据权利要求6所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:聚乙烯醇与壳聚糖的质量比为1~3:1。
8.根据权利要求7所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:聚乙烯醇与壳聚糖的质量比为2:1。
9.根据权利要求6所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:壳聚糖与水的质量体积比10~30g:100mL。
10.根据权利要求9所述的壳聚糖-聚乙烯醇SOD酶水凝胶敷料,其特征在于:壳聚糖与水的质量体积比为20g:100mL。
