Abstract
一种鹿角盘免疫活性肽,其特征在于:所述免疫活性肽的氨基酸序列为Phe‑Met‑Val‑Glu‑Ala‑Thr‑Lys(FMVEATK)、Leu‑Phe‑Gly‑Glu‑Lys(LFGEK)、Leu‑Leu‑Glu‑Pro‑Thr‑Leu‑Lys(LLEPTLK)、Val‑Leu‑Leu‑Arg(VLLR)和/或Leu‑Asp‑Phe‑Asp‑Glu‑Phe‑Leu‑Lys(LDFDEFLK)。本发明制备的鹿角盘免疫活性肽能有效促进RAW 264.7巨噬细胞与小鼠脾淋巴细胞的生长和增殖,且能刺激RAW264.7巨噬细胞发挥吞噬作用以及提高释放NO的能力,具有促进RAW264.7细胞iNOS、TNF‑α、IL‑1β和IL‑6的表达,同时还具有耐高温和耐酸碱的稳定特性,并且经过胃蛋白酶和胰蛋白酶消化后活性肽仍能保持稳定性,保持其序列的完整性。

技术领域
本发明涉及免疫活性肽技术领域,具体涉及一种鹿角盘免疫活性肽及其制备方法和应用。
背景技术
许多研究证实小肽可以完整的被机体吸收从而发挥自身的生理功能并有较低的免疫原性,免疫活性肽与一般的大分子蛋白药物相比显现出更强的优势。因人体仅能被动的接受增强免疫力的大分子药物,当停止服用大分子免疫增强药物时,人体的免疫能力就会随之下降;但免疫活性肽具有主动性,人体不会对免疫活性肽产生依赖性,因此免疫活性肽的相关研究日益增加。
免疫活性肽可以对生物体免疫系统起到调节作用,其作用机制有刺激机体淋巴细胞增殖,使巨噬细胞吞噬功能提高,增强机体对抗病原体的能力,从而使机体发病率下降,总之免疫活性肽是生命活动中必须的生物活性肽。免疫活性肽的分子结构复杂程度存在不同,这种多肽在体内大多含量较低,它是一种生命信号传递物质,其行使生物学功能的方式有多种如内分泌、旁分泌、神经分泌等,沟通各类细胞间的相互联系。生物活性肽与蛋白质相比,具有稳性性强、分子量小等特点,对人体而言更易消化吸收从而发挥特定的生物活性。一般而言,蛋白质空间构象的微小改变会造成其性质及其功能的改变。
鹿角盘是鹿科动物断茸或锯茸后第二年春季形成的骨化物质,其状如盘又有“鹿托盘”之称,又名“珍珠盘”、“龙角盘”。鹿角盘药理活性包括免疫调节、抗炎镇痛、抑菌、抗疲劳、抗骨质疏松等。鹿角盘的蛋白质含量极高,氨基酸组成丰富,其中包含人体必需的氨基酸及矿物质元素(如磷、钙、钠、镁、钾、铁、锰、锌、钡、锶等),鹿角盘含有蛋白质及多肽、无机元素、孕酮、雌二醇等多种物质其中蛋白质类物质高达46.30%。研究发现鹿角盘药理活性的多样性与其蛋白类物质的丰富性有着密切的联系,这为鹿角盘活性肽的制备及其生物活性的研究提供了丰富的蛋白质资源。
发明内容
本发明目的在于提供一种鹿角盘免疫活性肽,可有效促进RAW264.7巨噬细胞与小鼠脾淋巴细胞的生长和增殖,同时还具有耐高温和耐酸碱的稳定特性,并且经过胃蛋白酶和胰蛋白酶消化后仍能保持稳定性,保持其序列完整性。
本发明另一目的在于提供和上述鹿角盘免疫活性肽的制备方法。
本发明第三个目的在于提供上述鹿角盘免疫活性肽的应用。
本发明目的通过如下技术方案实现:
一种鹿角盘免疫活性肽,其特征在于:所述免疫活性肽的氨基酸序列为Phe-Met-Val-Glu-Ala-Thr-Lys(FMVEATK)、Leu-Phe-Gly-Glu-Lys(LFGEK)、Leu-Leu-Glu-Pro-Thr-Leu-Lys(LLEPTLK)、Val-Leu-Leu-Arg(VLLR)和/或Leu-Asp-Phe-Asp-Glu-Phe-Leu-Lys(LDFDEFLK)。
优选的,所述免疫活性肽的氨基酸序列为Leu-Phe-Gly-Glu-Lys(LFGEK)。
上述鹿角盘免疫活性肽的制备方法,其特征在于:以花马杂交鹿角盘为原料提取总蛋白,通过酶解制备鹿角盘活性总肽,依次经过Sephadex G-25凝胶层析、Sephadex G-15凝胶层析以及反向高效液相色谱(RP-HPLC)三步分离纯化,获得上述鹿角盘免疫活性肽。
进一步,所述提取总蛋白是将花马杂交鹿角盘粉末与NaOH溶液混合,磁力搅拌2h后,进行离心处理,取上清液,调节pH为4.6,静置30min,再次离心,取沉淀洗涤、干燥得鹿角盘总蛋白。
进一步,上述花马杂交鹿角盘粉末与NaOH溶液的料液比为1g:15mL,NaOH溶液的浓度为0.1mol/L。
进一步,上述离心转速为6000rpm。
进一步,所述酶解是以鹿角盘总蛋白作为底物,加入胰蛋白酶进行水解,其中底物浓度为3wt.%,pH为6.5-7,胰蛋白酶的添加量为2500-3500U/g,水解温度为40-45℃,水解时间为2-3h。
优选的,底物浓度为3wt.%,pH为6.8,胰蛋白酶的添加量为2358U/g,水解温度为41℃,水解时间为2.42h。
进一步,所述Sephadex G-25凝胶层析分离是将鹿角盘活性总肽溶于去离子水中,形成浓度为20mg/mL的溶液,过0.45μm滤器后得上样样品,将上样样品泵入层析柱,流速为1mL/min,所得产物按照相同方式进行Sephadex G-15凝胶层析分离。
进一步,所述反向高效液相色谱(RP-HPLC)的样品浓度10mg/mL,色谱柱为ZORBAX300SB-C18,流动相A为体积分数0.1%三氟乙酸的超纯水,流动相B为体积分数0.1%三氟乙酸的乙腈溶液,紫外检测波长为220nm;柱温20℃;流速0.5mL/min;洗脱梯度:0~5min,6%~15%流动相B;5~10min,15%~30%流动相B;10~25min,30%~46%流动相B,将分离得到的组分大量收集冻干。
蛋白质经酶解后混杂着多种产物,如氨基酸、多肽、盐离子等,分离提纯为了获得组分简单且活性较高的产物,此过程需去除多种杂质。因不同多肽之间可能存在性质相近的情况,如分子量、电荷、极性等,单一的分离技术很难将其完全分开,所以结合不同的分离技术才能将目的产物从成分复杂的混合物中分离开。
最具体,一种鹿角盘免疫活性肽的制备方法,其特征在于,按如下步骤进行:
步骤1:提取总蛋白
将花马杂交鹿角盘粉末与NaOH溶液混合,磁力搅拌2h后,在6000rpm下进行离心处理,取上清液,调节pH为4.6,静置30min,再次离心,取沉淀洗涤、干燥得鹿角盘总蛋白,马杂交鹿角盘粉末与NaOH溶液的料液比为1g:15mL,NaOH溶液的浓度为0.1mol/L;
步骤2:水解制备活性总肽
鹿角盘总蛋白作为底物,加入胰蛋白酶进行水解,其中底物浓度为3wt.%,pH为6.5-7,胰蛋白酶的添加量为2500-3500U/g,水解温度为40-45℃,水解时间为2-3h;
步骤3:分离纯化
(1)将鹿角盘蛋白活性肽溶于去离子水中,形成浓度为20mg/mL的溶液,过0.45μm滤器后得上样样品,将上样样品泵入层析柱,流速为1mL/min,经分离获得三种组分A1、A2和A3;
(2)经免疫活性检测,将免疫活性最优的A2按照相同方式进行Sephadex G-15凝胶层析分离,得到两种组分A2-1和A2-2;
(3)将步骤(2)或的组分进行免疫活性分析,将免疫活性最优的A2-2进行反向高效液相色谱(RP-HPLC),样品浓度10mg/mL,色谱柱为ZORBAX300SB-C18,流动相A为体积分数0.1%三氟乙酸的超纯水,流动相B为体积分数0.1%三氟乙酸的乙腈溶液,紫外检测波长为220nm;柱温20℃;流速0.5mL/min;洗脱梯度:0~5min,6%~15%流动相B;5~10min,15%~30%流动相B;10~25min,30%~46%流动相B,分离得到的组分为A2-2-1和A2-2-2,经免疫活性检测,取免疫活性更优的A2-2-1组分,收集冻干;
(4)对A2-2-1进行结构鉴定:
测序结果中含有的5条肽序列,依次为Phe-Met-Val-Glu-Ala-Thr-Lys(FMVEATK)、Leu-Phe-Gly-Glu-Lys(LFGEK)、Leu-Leu-Glu-Pro-Thr-Leu-Lys(LLEPTLK)、Val-Leu-Leu-Arg(VLLR)和Leu-Asp-Phe-Asp-Glu-Phe-Leu-Lys(LDFDEFLK)。
在酶解过程中,蛋白酶的选择是制备活性肽的关键,不同的水解蛋白酶的理化性质、切割氨基酸位点等会有所不同,所形成的酶解产物也会不同,所以功能特性也就不同。只有筛选合适的蛋白酶,再将调节酶反应条件达到最佳,提取的鹿角盘活性总肽才能具有最强的免疫活性。活性肽的免疫活性与氨基酸序列有密切关系,活性肽的N末端为碱性氨基酸或非极性氨基酸时,免疫活性较好。
上述鹿角盘免疫活性肽的应用,其特征在于:在制备促进巨噬细胞增殖产品中的应用。

本发明具有如下技术效果:
本发明制备的鹿角盘免疫活性肽能有效促进RAW 264.7巨噬细胞与小鼠脾淋巴细胞的生长和增殖,且能刺激RAW264.7巨噬细胞发挥吞噬作用以及提高释放NO的能力,具有促进RAW264.7细胞中iNOS、TNF-α、IL-1β和IL-6的表达,同时还具有耐高温和耐酸碱的稳定特性,并且经过胃蛋白酶和胰蛋白酶消化后肽仍能保持稳定性,保持其序列完整性。

实施例1
一种鹿角盘免疫活性肽的制备方法,其特征在于,按如下步骤进行:
步骤1:提取总蛋白
将花马杂交鹿角盘粉末与NaOH溶液混合,磁力搅拌2h后,在6000rpm下进行离心处理,取上清液,调节pH为4.6,静置30min,再次离心,取沉淀洗涤、干燥得鹿角盘总蛋白,马杂交鹿角盘粉末与NaOH溶液的料液比为1g:15mL,NaOH溶液的浓度为0.1mol/L;
步骤2:水解制备活性总肽
鹿角盘总蛋白作为底物,加入胰蛋白酶进行水解,其中底物浓度为3wt.%,pH为6.5-7,胰蛋白酶的添加量为2500-3500U/g,水解温度为40-45℃,水解时间为2-3h;
步骤3:分离纯化
(1)将鹿角盘蛋白活性总肽溶于去离子水中,形成浓度为20mg/mL的溶液,过0.45μm滤器后得上样样品,将上样样品泵入层析柱,流速为1mL/min,经分离获得三种组分A1、A2和A3;
(2)经免疫活性检测,将免疫活性最优的A2按照相同方式进行Sephadex G-15凝胶层析分离,得到两种组分A2-1和A2-2;
(3)将步骤(2)或的组分进行免疫活性分析,将免疫活性最优的A2-2进行反向高效液相色谱(RP-HPLC),样品浓度10mg/mL,色谱柱为ZORBAX300SB-C18,流动相A为体积分数0.1%三氟乙酸的超纯水,流动相B为体积分数0.1%三氟乙酸的乙腈溶液,紫外检测波长为220nm;柱温20℃;流速0.5mL/min;洗脱梯度:0~5min,6%~15%流动相B;5~10min,15%~30%流动相B;10~25min,30%~46%流动相B,分离得到的组分为A2-2-1和A2-2-2,经免疫活性检测,取免疫活性更优的A2-2-1组分,收集冻干;
(4)对A2-2-1进行结构鉴定:
测序结果中含有的5条肽序列,依次为Phe-Met-Val-Glu-Ala-Thr-Lys(FMVEATK)、Leu-Phe-Gly-Glu-Lys(LFGEK)、Leu-Leu-Glu-Pro-Thr-Leu-Lys(LLEPTLK)、Val-Leu-Leu-Arg(VLLR)和Leu-Asp-Phe-Asp-Glu-Phe-Leu-Lys(LDFDEFLK)。
生物活性肽相较于蛋白质更易消化吸收,且因其分子量小能在机体内保持稳定。通常,蛋白质的结构发生微小的变动时会导致蛋白质结构及性质发生改变,但大量研究证明蛋白质酶解后发生的结构上的改变是正面的,酶解产物与源蛋白相比富含更多的生物活性物质,具有更好的理化特性和加工特性。因而,本发明将提取的鹿角盘总蛋白经胰蛋白酶水解得到鹿角盘活性总肽。本发明通过碱提酸沉法提取鹿角盘总蛋白,再通过SDS-PAGE电泳结果显示在116KDa至6.5KDa之间均有总蛋白分布,BCA法检测总蛋白含量为94.6%。鹿角盘总蛋白氨基酸种类及含量分析结果显示非极性氨基酸、极性氨基酸、碱性氨基酸、酸性氨基酸分别占总氨基酸含量的34.67%、31.7%、15.02%、18.81%。且在采用胰蛋白酶水解在水解时间为2.42h,pH为6.8,加酶量为3358U\g和水解温度为41℃的最优条件下水解度达到36.78±0.53%。
通过Sephadex G-25凝胶层析分离得到3种组分,记作A1、A2、A3,对应的葡聚糖凝胶层析图谱如图1(左)所示,根据凝胶层析原理,A1为穿透峰,未进入凝胶内部就被洗脱下来了,分子量较大,所以判断组分A1应该为水解未彻底的蛋白,而A2、A3为水解得到的多肽。经过Sephadex G-25分离后,筛选得到的免疫活性最强的A2组分经Sephadex G-15分离后,得到两种组分,记作A2-1和A2-2,对应的葡聚糖凝胶层析图谱如图1(右)所示,A2-2较A2-1来说,组分洗脱时间长,保留的峰面积大,即在此范围内肽段富集最多;通过免疫活性测定,测得A2-2的免疫活性高于A2-1,因此对A2-2进行反向高效液相色谱(RP-HPLC)分离纯化,得到两种组分,记作A2-2-1和A2-2-2,对应的色谱图如图2所示,经免疫活性测定A2-2-1具有更好的免疫活性。具体免疫活性测定如下:
1.关于免疫活性的测定:
RAW264.7细胞相关免疫活性(增殖、吞噬及NO释放量)
(1)RAW264.7细胞增殖:
当RAW264.7细胞培养至对数生长期时,将细胞浓度调整为5×105个/m L,将细胞液按照每孔100μL接种于96孔板内,每组设置3个复孔。细胞贴壁培养后,弃去细胞培养基,用不同浓度的各组分(终浓度为12.5、50、100、200μg/mL)加入96孔板中,置于CO2培养箱中培养12、24、48h。培养结束后,在超净工作台中向每孔加入10μL CCK-8,继续培养2h。于450nm下检测吸光度。细胞增殖活性计算公式如下:

(2)中性红吞噬试验:
取对数生长期的RAW264.7细胞按1×105每孔的密度加入到96孔细胞培养板中,培养24h后,吸去上清液,将不同浓度的各个分级分离组分处理RAW264.7细胞,10μg/mL LPS作对照,各组设置三个复孔。培养24h后弃去上清液,分别加入100μL 0.075%中性红生理盐水溶液,37℃培养1h。吸取上清液,用PBS洗三遍,每孔加入100μL细胞裂解液,室温下放置2h,待细胞完全溶解后,在540nm处测吸光值。

(3)NO释放量检测
取对数生长期的RAW264.7细胞以每孔细胞数为1×105个接种于96孔细胞培养板中,培养24h后,将不同浓度的各个分级分离组分处理RAW264.7细胞,10μg/mL LPS作对照,各组设置三个复孔。培养24h后,首先将50μL的上清液吸取至96孔板内,其次分别加入试剂Griess ReagentⅠ和Griess ReagentⅡ50μL,最后于540nm处测定吸光值。样品上清液中的NO含量是根据NO标准曲线算出,标准曲线的计算公式为:y=0.0311x+0.6234,R2=0.9982。
2.淋巴细胞增殖:
脾脏中单细胞悬浮液的制备:在无菌条件下取出脾脏,撕下外皮并用眼科剪切成小块。将尼龙筛或细胞过滤筛放在无菌平皿内,加入少量全血和组织稀释剂(以确保所得的脾脏和细胞处于液体环境中)。将脾脏放在筛子上,用注射器柱塞或无菌镊子粉碎(尽可能控制研磨力度,保持筛子悬浮,避免直接在罐底研磨导致细胞死亡)彻底研磨后,用血液和组织稀释剂冲洗筛子,收集细胞悬液并通过筛子过滤。在96孔板中加入已调整好浓度的小鼠脾淋巴细胞悬液(100μL),然后加入各分级分离组分,每组平行重复三次。对照组用完全培养基代替,之后放入细胞培养箱进行培养(37℃,5% CO2,48h),处理结束后,再向每孔加入10μL CCK-8,继续培养2h。于450nm下检测吸光度。计算公式如下:
淋巴细胞增殖活性=A-B/C-B (公式3)
A:实验组OD;B:对照组OD;C:空白组OD
所有实验至少进行3次,通过计算平均值±标准偏差(SD)获得数据。采用单因素方差分析(One-Wayanova)和Duncan's Multiplerangetest软件,SPSS16.0分析各组间差异的显著性,P<0.05为显著,P<0.01为极显著。使用OriginPro8.0作图。
3.各分离纯化组分的免疫活性测定:
(1)水解活性总肽对RAW264.7细胞增殖活性的影响
经各蛋白酶在各自的最适条件下水解后的鹿角盘活性总肽在不同浓度下处理RAW264.7细胞12、24、48h后增殖活性的变化趋势如图3所示,图中对应的蛋白酶依次是A为碱性蛋白酶,B为木瓜蛋白酶,C为胰蛋白酶、D为中性蛋白酶。结果发现,在12、24、48h后,经中性蛋白酶和胰蛋白酶酶解得到的活性总肽处理的RAW264.7细胞,在各浓度条件下均能促进细胞增殖,胰蛋白酶的最大增殖率为1.58,中性蛋白酶的最大增殖率为1.36。碱性蛋白酶与木瓜蛋白酶酶解得到的活性总肽处理的RAW264.7细胞,均在浓度为12.5μg/mL,处理细胞24h后增殖率达最高,分别为1.16与1.22。胰蛋白酶酶解得到的酶解产物促进RAW264.7巨噬细胞的增殖能力最佳,是由于胰蛋白酶的酶切位点主要为碱性氨基酸(赖氨酸、精氨酸)的残基,使之与其他氨基酸的氨基形成的肽键,最终得到的产物是以碱性氨基酸作为羧基末端的多肽,所以胰蛋白酶酶解得到的酶解产物促进细胞增殖能力最佳。
(2)Sephadex G-25分离组分免疫活性测定
酶解得到的活性总肽经Sephadex G-25分离后获得的组分A1、A2、A3处理RAW264.7细胞12、24、48h后增殖活性的变化趋势依次对应图4中A-C,与空白对照组相比,A2组分在50μg/mL浓度下能显著刺激巨噬细胞的增殖,刺激细胞增殖能力要优于A1、A3组分。由图4中D可以看出,A1、A2、A3这三个组分在处理细胞24小时后均能不同程度的促进小鼠脾淋巴细胞的增殖,组分之间相互比较可知,A2组分对脾淋巴细胞的增殖率大于A1、A3组。由图4中E可知,即从RAW264.7巨噬细胞中性红吞噬实验看出,三个组分在处理细胞24小时后均能提高巨噬细胞的吞噬能力,组分间比较发现A2在各个浓度下提高细胞吞噬率的能力均显著高于A1和A3。由图2中F可知,A2刺激细胞产生NO的能力始终显著高于A1和A3。由上述可推断A2更容易刺激RAW264.7巨噬细胞和脾淋巴细胞发挥免疫活性。故在进行下一步分离时选用A2进行进一步的分离纯化。
(3)Sephadex G-15分离组分免疫活性测定
将上一步筛选的免疫活性最强的A2组分经进行Sephadex G-15分离得到的A2-1和A2-2组分进行免疫活性的测定,结果如图5中A、B所示,经A2-1、A2-2处理RAW264.7细胞12、24、48h后,结果发现,各组分在12.5μg/mL-200μg/mL浓度条件下可促进RAW264.7细胞生长,其中A2-2组分的增殖活性随着浓度的升高而升高,且与A2-1组分相比,增殖活性最好。如图5中C所示,经过Sephadex G-15分离得到的两个组分处理脾淋巴细胞24h后,具有不同程度的促进脾淋巴细胞的增殖,且在各浓度条件下,A2-2能显著促进脾脏淋巴细胞增殖(P<0.05)。图5中D显示,较空白对照组而言A2-1和A2-2组分在12.5-200μg/mL浓度下处理细胞24h后均能刺激巨噬细胞,提高其吞噬能力;两个组分间比较发现A2-2提高巨噬细胞的吞噬能力较强。A2-2在50μg/mL浓度下能显著刺激RAW264.7巨噬细胞的吞噬活性为1.48,作用效果显著(P<0.05)。图4-4E指出A2-1和A2-2组分在五种浓度下处理细胞24h后刺激细胞分泌NO的能力均显著高于空白对照组,而A2-2组分在各个浓度下刺激细胞分泌NO的能力均显著高于A2-1组分(p<0.05)。总体而言,A2-2组分的免疫活性要高于A2-1组分,因此接下来用A2-2组分做下一阶段的分离纯化。
(4)反相高效液相色谱免疫活性测定
经对前两步骤分离纯化筛选得到免疫活性最强的A2-2组分进行RP-HPLC分离,得到A2-2-1和A2-2-2组分。免疫活性测定结果如图6中A、B所示,组分A2-2-1和A2-2-2处理RAW264.7细胞12、24、48h,各组分的增殖活性随着处理细胞的蛋白浓度的增加而增加,但A2-2-2组分促细胞增殖能力较A2-2-1来说明显较低。如图6中C所示,经过RP-HPLC分离得到的两个组分处理脾淋巴细胞24h后,随着浓度增加,增殖能力随之增加,呈剂量依赖,且在各浓度条件下,A2-2-1比A2-2-2更能显著促进脾脏淋巴细胞增殖(P<0.05)。图4-6中D所示,A2-2-1与A2-2-2在各浓度下处理细胞24h后,与空白对照组相比能够显著刺激RAW264.7细胞吞噬活性的提升,其中A2-2-1在各个浓度下刺激巨噬细胞的吞噬活性均高于A2-2-2。图6中E为经反向高效液相分离所得组分在12.5-200μg/mL浓度下处理细胞24h后,刺激RAW264.7巨噬细胞上清液中NO分泌量的比较,结果显示,A2-2-1在各个浓度下刺激巨噬细胞分泌NO的能力均显著高于空白对照组,且显著高于A2-2-2。以上结果均说明,在鹿角盘蛋白酶解产物经过多级纯化后得到的A2-2-1组分仍具有较好的免疫调节活性。因此接下来对A2-2-1组分进行进一步的结构鉴定以及活性分析。
4.鹿角盘免疫活性肽的结构鉴定:
利用De novo从头测序法对组分A2-2-1进行序列解析,其一级质谱图如7所示,后续对一级质谱中的各信号值所代表的物质进行二级质谱,对从A2-2-1鉴定得到的肽段从亲疏水性、分子量、正负电荷等方面进行分析总结,优选测序结果可信度高的5条肽序列,依次为Phe-Met-Val-Glu-Ala-Thr-Lys(FMVEATK)、Leu-Phe-Gly-Glu-Lys(LFGEK)、Leu-Leu-Glu-Pro-Thr-Leu-Lys(LLEPTLK)、Val-Leu-Leu-Arg(VLLR)、Leu-Asp-Phe-Asp-Glu-Phe-Leu-Lys(LDFDEFLK),对应的二级质谱如图8所示。推测上述肽链其极有可能具有高免疫活性,将通过进一步的合成验证来确定。
5.合成免疫活性肽及各性能测试:
采用固相合成法分别合成上述5条肽链,再利用VarianProStar 218高效液相色谱(HPLC)仪器和Aglient Venusil MP C18反相柱纯化肽,以1mL/min的流速,用均含有0.05%TFA的蒸馏水和乙腈线性梯度洗脱肽,并在220nm进行紫外检测,然后对多肽进行Voyager-DE STR质谱(MS)分析。梯度洗脱高效液相色谱的溶剂为:溶剂A,ACN 2%,TFA 0.05%;溶剂B,ACN 90%,TFA0.05%。最后通过液相色谱-串联质谱联用(LC-ESI-MS/MS)技术测定合成肽的分子量,采用RP-HPLC对合成肽进行纯度检测,合成肽纯度>99%。依次记为:
(1)A2-2-1-1:LFGEK;
(2)A2-2-1-2:LFGEK;
(3)A2-2-1-3:LLEPTLK;
(4)A2-2-1-4:VLLR;
(5)A2-2-1-5:LDFDEFLK。
(1)合成多肽的免疫活性测定:
免疫活性测定结果表明,A2-2-1-1、A2-2-1-2、A2-2-1-3、A2-2-1-4、A2-2-1-5均能促进RAW264.7巨噬细胞不同程度的增殖,其中在处理24h时,各个肽段促进巨噬细胞增殖的能力最强。如图9中A-C所示,图A为合成肽在12h时对RAW264.7巨噬细胞增殖的影响;图B为合成肽在24h时对RAW264.7巨噬细胞增殖的影响,图C为合成肽在48h时对RAW264.7巨噬细胞增殖的影响。各合成肽之间相比,A2-2-1-1和A2-2-1-2促进RAW264.7巨噬细胞的增殖能力比其余合成肽能力强,但A2-2-1-2促增殖能力始终高于A2-2-1-1。图D为合成肽处理细胞24h后对小鼠脾淋巴细胞增殖的影响,可以看出合成肽A2-2-1-2和A2-2-1-3随着浓度的增加,促进脾淋巴细胞增殖的能力随之增强,其余各合成肽促脾淋巴细胞增殖能力不显著(P>0.05)。A2-2-1-2在浓度为100ug/mL时,促脾淋巴细胞增殖能力最强;A2-2-1-3在浓度为200μg/mL时,促脾淋巴细胞增殖能力达到最高。总而言之,在这五条合成肽中A2-2-1-2促脾淋巴细胞增殖能力最强。各合成肽促进RAW264.7巨噬细胞吞噬能力由图9中E可知,处理细胞24h后,随着浓度提高,巨噬细胞吞噬能力明显提高,但不完全呈剂量依赖性,A2-2-1-1和A2-2-1-2促吞噬能力较强,均在100ug/mL浓度下促吞噬能力达到最高。其中A2-2-1-2随着浓度的增加促吞噬能力趋近于LPS阳性对照组,说明A2-2-1-2促进RAW264.7细胞吞噬能力效果最好。再如图9中F所示,A2-2-1-1和A2-2-1-2刺激RAW264.7巨噬细胞产生NO的能力显著高于其他合成肽,其中,与空白对照组相比,A2-2-1-1浓度为100ug/mL时能够刺激巨噬细胞产生NO的表达量为17.06;A2-2-1-2刺激产生NO的表达量为30.46(P<0.01),说明A2-2-1-2刺激RAW264.7细胞产生NO的能力最强,因此,综合各方面,将A2-2-1-2组分命名为鹿角盘免疫活性肽(antler plate immunologic competence peptides,APICP)。
(2)APICP对RAW264.7巨噬细胞内TNF-α、iNOS、IL-1β、IL-6mRNA及蛋白表达水平的影响:
为了验证APICP是否可以激活巨噬细胞RAW264.7分泌细胞因子,促进体外免疫调节作用,测定了不同浓度APICP对巨噬细胞RAW264.7分泌的细胞因子mRNA及蛋白表达水平的影响。结果如图10所示,与空白对照组RAW264.7细胞比较,当APICP浓度为25μg/mL时,细胞因子TNF-α、iNOS、IL-1β、IL-6的表达量增加,呈显著性差异(P<0.05或P<0.01);当APICP浓度为100μg/mL时各个细胞因子的表达量都达到最大值,呈极显著性差异(P<0.01)。在阳性对照组LPS的刺激下,RAW264.7细胞能明显增加TNF-α、iNOS、IL-1β、IL-6的释放量,具有极显著差异(P<0.01)。这些结果表明,APICP浓度范围在25~100μg/mL时均可以提高TNF-α、iNOS、IL-1β以及IL-6的分泌量,并且呈剂量依赖性,能更加有效地刺激RAW264.7分泌细胞因子,进而发挥免疫调节作用。
免疫活性肽的功能评价主要包括体外细胞免疫和动物体内免疫这两大类。体外细胞免疫主要是通过试验样品是否能够使免疫细胞增殖、巨噬细胞吞噬能力的提高,以及免疫细胞的免疫因子表达水平的变化如IL-6、TNF-α等来体现。动物体内免疫主要包括细胞和体液免疫如对免疫器官的重量测定,以及相应细胞因子表达水平的变化。在细胞免疫功能、体液免疫功能、单核-巨噬细胞功能以及自然杀伤细胞细胞活性四个方面中,其中任意两个方面的实验结果为阳性,则可判定试验物具有增强免疫力的效果。本发明制备的鹿角盘活性免疫肽可促进免疫细胞增殖,并通过促进iNOS的分泌,从而增强杀灭病原菌的能力,进而增强先天性免疫。APICP通过促进RAW264.7细胞TNF-α、IL-1β和IL-6的分泌,增强巨噬细胞吞噬和杀伤能力,进而促进巨噬细胞向免疫应答部位移动并间接具有刺激细胞产生免疫球蛋白的功能。
(3)APICP的热稳定分析:
在活性肽的加工利用过程中可能会遇到各种高温环境,活性肽在各种温度条件下能否保持肽段的完整性尤为重要,为了评估APICP的热稳定性,通过RP-HPLC检测其在不同温度条件下APICP的状态:
将1mg的APICP用10mL的超纯水稀释成终质量浓度为100μg/mL的溶液并装于6支1.5mL离心管内,分为空白对照组和实验组。将其分别放置于4℃、20℃、40℃、60℃、80℃、100℃环境下,以4℃下的样品溶液为空白对照,待2h后将各温度下的样品溶液取出,通过RP-HPLC进行检测。检测波长:220nm,柱温:37℃,流动相A:体积分数2%乙腈+体积分数0.03%TFA+体积分数98%的超纯水,流动相B:体积分数98%的乙腈+体积分数2%的超纯水+体积分数0.05%TFA,洗脱类型:梯度洗脱,洗脱条件:0-5min,95%-90%B;5-15min,90%-45%B;15-20min,45%-30%B:20-30min,30%-95%B。洗脱流速为0.75mL/min。
获得的液相图谱如图11中A-E所示,可看出,各组峰为单一峰且出峰时间不变。经过计算各组峰面积如图11中F所示,与空白对照组相比,各条件处理后的APICP的相对峰面积无显著差异(P>0.05),表明在各个温度条件下APICP保持了肽段的完整性,并未发生改变,热稳定性较好。(积分时以小于主峰的0.05%为杂峰,根据出峰时间和主峰的峰面积判断检测样品是否发生降解和变性。)
(4)APICP的酸碱稳定性分析:
pH值是影响多肽活性的一个重要条件,为了评估APICP的酸碱稳定性,通过RP-HPLC检测其在各pH条件下APICP的状态:
将APICP分别用pH值为2、4、6、8、10的水溶液稀释成100μg/mL的浓度,37℃水浴2h。使用0.22μm孔径的滤膜将空白组和各pH值下APICP样品溶液分别过滤,随后注入RP-HPLC分析,洗脱条件与(3)中相同。根据液相图谱的峰面积大小判定APICP在不同pH值下是否发生降解。
由液相图谱如图12中A-E所示,可看出,各组峰为单一峰且出峰时间不变。经过计算各组峰面积如图12中F所示,与空白对照组相比,各条件处理后的APICP的相对峰面积无显著差异(P>0.05),表明在各个pH条件下APICP保持了肽段的完整性,并未发生改变,酸碱稳定性较好。
(5)APICP的模拟胃肠道消化稳定性分析:
活性肽进入体内后需要保证活性肽能以完整的结构到达血液,才能保留其生物学活性,所以活性肽经口服后,需要抵抗胃肠道内的胃蛋白酶、胰蛋白酶等酶的消化水解。如活性肽在该过程被降解会降低其生物学活性,甚至会失活。
因此,选取了胃肠道内的两种关键酶(胃蛋白酶和胰蛋白酶)对APICP进行体外模拟消化:
APICP用pH为2的水溶液稀释至100μg/mL,加入胃蛋白酶(1:3000),在37℃水浴中水解2h,每隔1h取样,4℃备用。2h后取出样品。然后将胃蛋白酶酶解的APICP溶液调节至pH=7,加入胰蛋白酶(1:250),37℃水浴2h,每隔1h取样,4℃保存。空白组和各实验组的APICP样品溶液分别用孔径为0.22μm的滤膜过滤,然后注入RP-HPLC分析,洗脱条件与(3)中相同,再通过RP-HPLC检测其在胃蛋白酶以及胰蛋白酶处理后APICP的状态。
液相图谱如图13中A-E所示,可以看出,各组峰为单一峰且出峰时间不变。经过计算各组峰面积如图13中F所示,与空白对照组相比,各条件处理后的APICP的相对峰面积无显著差异(P>0.05)。构效关系表明,APICP肽段C端含有的赖氨酸(Lys)可在酶解作用发生时保护肽段的完整性。稳定性分析结果表明,APICP具有耐高温和耐酸碱的特性,并且经过胃蛋白酶和胰蛋白酶消化后肽仍能保持稳定性。以上结果表明,口服APICP胃肠道消化稳定性较好,可以在胃中保持其序列完整性,为接下来的研究奠定了理论基础。
Claims (10)
1.一种鹿角盘免疫活性肽,其特征在于:所述免疫活性肽的氨基酸序列为Phe-Met-Val-Glu-Ala-Thr-Lys (FMVEATK)、Leu-Phe-Gly-Glu-Lys(LFGEK)、Leu-Leu-Glu-Pro-Thr-Leu-Lys(LLEPTLK)、Val-Leu-Leu-Arg(VLLR)和/或Leu-Asp-Phe-Asp-Glu-Phe-Leu-Lys(LDFDEFLK)。
2.如权利要求1所述的一种鹿角盘免疫活性肽,其特征在于:所述免疫活性肽的氨基酸序列为Leu-Phe-Gly-Glu-Lys(LFGEK)。
3.一种如权利要求1或2所述的鹿角盘免疫活性肽的制备方法,其特征在于:是以花马杂交鹿角盘为原料提取总蛋白,通过胰蛋白酶水解制备鹿角盘活性总肽,依次经过Sephadex G-25凝胶层析、Sephadex G-15凝胶层析以及反向高效液相色谱(RP-HPLC)三步分离纯化,获得鹿角盘免疫活性肽。
4.一种如权利要求3所述的鹿角盘免疫活性肽的制备方法,其特征在于:所述提取总蛋白是将花马杂交鹿角盘粉末与NaOH溶液混合,磁力搅拌2h后,进行离心处理,取上清液,调节pH为4.6,静置30min,再次离心,取沉淀洗涤、干燥得鹿角盘总蛋白。
5.一种如权利要求1-4任一项所述的鹿角盘免疫活性肽的制备方法,其特征在于:所述水解是以鹿角盘总蛋白作为底物,加入胰蛋白酶进行水解,其中底物浓度为3wt.%,pH为6.5-7,胰蛋白酶的添加量为2500-3500U/g,水解温度为40-45℃,水解时间为2-3h。
6.一种如权利要求5所述的鹿角盘免疫活性肽的制备方法,其特征在于:所述底物浓度为3wt.%,pH为6.8,胰蛋白酶的添加量为2358U/g,水解温度为41℃,水解时间为2.42h。
7.一种如权利要求6所述的鹿角盘免疫活性肽的制备方法,其特征在于:所述SephadexG-25凝胶层析分离是将鹿角盘活性总肽溶于去离子水中,形成浓度为20mg/mL的溶液,过0.45μm滤器后得上样样品,将上样样品泵入层析柱,流速为1mL/min,所得产物按照相同方式进行Sephadex G-15凝胶层析分离。
8.一种如权利要求7所述的鹿角盘免疫活性肽的制备方法,其特征在于:所述反向高效液相色谱(RP-HPLC)的样品浓度10 mg/mL,色谱柱为 ZORBAX 300SB-C18,流动相A为体积分数0.1%三氟乙酸的超纯水,流动相B为体积分数0.1%三氟乙酸的乙腈溶液,紫外检测波长为220 nm;柱温20 ℃;流速0.5 mL/min;洗脱梯度:0~5min,6%~15%流动相B;5~10 min,15%~30%流动相B;10~25 min,30%~46%流动相B,将分离得到的组分大量收集冻干。
9.如权利要求1所述的鹿角盘免疫活性肽的应用,其特征在于:在制备促进巨噬细胞增殖产品中的应用。
10.如权利要求1所述的鹿角盘免疫活性肽的应用,其特征在于:在制备促进淋巴细胞增殖产品中的应用。
